Opis:
Stem cells have captured considerable scientific and clinic interest because of their potential to renew themselves and to differentiate into one or more adult cell types. Thus stem cells have been recognized as a potential tool for the development of innovative therapeutic strategies in different disease disorders. Stem cells can be discriminating based on their differentiated potential as totipotent, pluripotent, multipotent or unipotent cells. There are in general three types of stem cells: embryonic, fetal and adult stem cells. While embryonic stem cell therapy has a lot of ethical concerns due to their obtaining but also unlimited proliferation and uncontrolled differentiation, fetal and adult stem cells have been used in the treatment of different diseases. The bone marrow, peripheral blood and umbilical cord blood are ideal sources of adult stem cells because there are easily accessible and contain two types of stem cells: hematopoietic stem cells giving rise to all blood cell types and mesenchymal stem cells differentiating into cells of mesodermal lineage. This review describes the general characteristics of these stem cell populations and their current applications in regenerative medicine. Additionally induced pluripotent stem cells generated through the reprogramming of differentiated adult cells are described.

Cell walls are at the basis of a structural, four-dimensional framework of plant form and growth time. Recent rapid progress of cell wall research has led to the situation where the old, long-lasting juxtaposition: "living" protoplast - "dead" cell wall, had to be dropped. Various attempts of re-interpretation cast, however, some doubts over the very nature of plant cell and the status of the walls within such a cell. Following a comparison of exocellular matrices of plants and animals, their position in relation to cells and organisms is analysed. A multitude of perspectives of the biological organisation of living beings is presented with particular attention paid to the cellular and organismal theories. Basic tenets and resulting corollaries of both theories are compared, and evolutionary and developmental implications are considered. Based on these data, "The Plant Body" - an organismal concept of plants and plant cells is described.

However, they are increasingly being replaced by electric or fuel cell-equipped alternative propulsion systems. The article presents a study of fuel cell characteristics operating under both static and dynamic conditions, with a 1.2 kW fuel cell set with a voltage converter and lead-acid batteries. In the conducted tests, the fuel cell stack's maximum efficiency reached 65%. Load tests (static and dynamic) have indicated higher fuel cell efficiencies when using hybrid operation with a DC/DC converter and battery.

Opis:
Camptothecin (CPT), an alkaloid natural product, extracted from Camptotheca acuminata bark, has been reported to have potential antitumor activity in diverse cancers. MicroRNAs (MiRNAs) are a class of short, non-coding RNAs that plays a crucial role in the normal physiology by attenuating translation. Here, we showed that the CPT modulates the expression of miRNAs in hepatocellular carcinoma cells (HCC). Microarray analysis reveals that CPT modulates the expression of as many as 39 miRNAs in HCC cells (Huh7), 27 miRNAs were downregulated whereas 12 miRNAs were upregulated. miR-16 is the key miRNA upregulated by CPT and targets key prosurvival proteins (MMP-2, MMP-9 and cyclin D1). Our results demonstrate that CPT is inhibiting cell viability of HCC cells significantly when compared with the untreated cells. Wound healing and colony formation assay confirm inhibition of cell migration and clonogenic property of Huh7 cells respectively, upon the dose-dependent treatment of CPT. Furthermore, the Boyden chamber assay analysis revealed a significant inhibition of number of invasive cells in CPT treated cells with comparison to untreated Huh7 cells. Mechanistically, CPT upregulates miR-16 expression which targets MMP-2, MMP-9, cyclin D1 downregulation and subsequently upregulates the expression of E-cadherin, TIMP1, p21, and p27, thereby inhibits cell migration, invasion and clonogenic property of HCC cells. In summary, CPT treatment in Huh7 cells decreases cell viability and upregulates miR-16 expression, which results in inhibition of cell migration, invasion and clonogenic property of cells, by decreasing MMP-2, MMP-9, cyclin D1 and increasing the expression of cell cycle-regulated proteins p21 and p27.

Opis:
Manipulating particles in the blood pool with noninvasive methods has been of great interest in therapeutic delivery. Recently, it was demonstrated experimentally that red blood cells can be forced to translate and accumulate in an ultrasound field. This acoustic response of the red blood cells has been attributed to sonophores, gas pockets that are formed under the influence of a sound field in the inner-membrane leaflets of biological cells. In this paper, we propose a simpler model: that of the compressible membrane. We derive the spatio-temporal cell dynamics for a spherically symmetric single cell, whilst regarding the cell bilayer membrane as two monolayer Newtonian viscous liquids, separated by a thin gas void. When applying the newly-derived equations to a red blood cell, it is observed that the void inside the bilayer expands to multiples of its original thickness, even at clinically safe acoustic pressure amplitudes. For causing permanent cell rupture during expansion, however, the acoustic pressure amplitudes needed would have to surpass the inertial cavitation threshold by a factor 10. Given the incompressibility of the inner monolayer, the radial oscillations of a cell are governed by the same set of equations as those of a forced antibubble. Evidently, these equations must hold for liposomes under sonication, as well.

Real-time cell electronic sensing (RT-CES) based on impedance measurements is an emerging technology for analyzing the status of cells in vitro. It allows label-free, real time monitoring of the biological status of cells. The present study was designed to assess dynamic data on the cell processes during equine herpesvirus type 1 (EHV-1) infection of ED (equine dermal) cells and primary murine neuronal cell culture. We have demonstrated that the xCELLigence system with dynamic monitoring can be used as a rapid diagnostic tool both to analyze cellular behavior and to investigate the effect of viral infection.

Opis:
In the course of infections caused by pathogenic yeasts from the genus Candida, the fungal cell surface is the first line of contact with the human host. As the surface-exposed proteins are the key players in these interactions, their identification can significantly contribute to discovering the mechanisms of pathogenesis of two emerging pathogens from this genus, C. parapsilosis and C. tropicalis. Therefore, the aim of the present study was to identify the cell wall-attached proteins of these two species with the use of cell surface shaving and a shotgun proteomic approach. Different morphological forms of C. parapsilosis and C. tropicalis cells obtained after growth under various conditions were subjected to this treatment. This allowed to indicate the most abundant cell surface proteins on the basis of the normalized spectral abundance factors. In case of yeast-like forms these were, among others, proteins similar to a chitinase, glyceraldehyde-3-phosphate dehydrogenase and an inducible acid phosphatase for C. parapsilosis, and a constitutive acid phosphatase, pyruvate decarboxylase and glyceraldehyde-3-phosphate dehydrogenase for C. tropicalis. In case of pseudohyphal forms, proteins similar to a cell surface mannoprotein Mp65, chitinase and glycosylphosphatidylinositol-anchored transglycosylase Crh11 were identified at the cell surface of C. parapsilosis. The Rbt1 cell wall protein, a hyphally regulated cell wall protein and proteins from agglutinin-like sequence protein family were found as the most abundant on C. tropicalis pseudohyphae. Apart from the abovementioned proteins, several additional covalently bound and atypical cell wall proteins were also identified. These results extend the current knowledge regarding the molecular basis of virulence of these two non-albicans Candida species.

Based on computer simulation technology, the feasibility and the effectiveness of our proposed method are demonstrated by comparing our simulation results with the experimental results of the spherical silica bead and the red blood cell, respectively. Our work will provide some guidance in the experimental research for transparent phase objects.

Some studies show that cells are able to penetrate through pores that are smaller than cell size. It concerns especially Red Blood Cells but it also may concern different types of biological cells. Such penetration of small pores is a very significant problem in the filtration process, for example in micro or ultrafiltration. Deformability of cells allows them to go through the porous membrane and contaminate permeate. This paper shows how cells can penetrate small cylindrical holes and tries to assess mechanical stress in a cell during this process. A new mathematical approach to this phenomenon was presented, based on assumptions that were made during the microscopic observation of Red Blood Cell aspiration into a small capillary. The computational model concerns Red Blood Cell geometry. The mathematical model allows to obtain geometrical relation as well as mechanical stress relations.

Metal surfaces have become important over the last decade for potential surgical implants, and within this context we present here a study of the cell growth on modified gold surfaces. Gold films, deposited on glass plates and annealed with a hydrogen flame, were modified with four different carboranethiol derivatives: 1-(HS)-1,2-C2B10H11 (A), 1,2-(HS)2-1,2-C2B10H10 (B), 9,12-(HS)2-1,2-C2B10H10 (C) and 1,12-(HS)2-1,12- C2B10H10 (D). The materials engendered from these modifications were used to investigate the adhesion and growth of rat aortic smooth muscle cells cultured on these surfaces in a DMEM medium with 10% of fetal bovine serum. One day after seeding, the highest number of initially adhered cells was found on the surface of a bare gold film. However, three days after seeding, the number of cells on carboranethiol-modified gold samples B, C and D was significantly higher than the number on a bare gold film. After seven days, the number of cells on a bare gold film and on gold films modified with derivatives A, B and D was very similar, but the surface of a gold film modified with derivative C exhibited a significantly smaller number of cells. This may be explained by the exposure of the CH vertices of the carborane cluster, which are more acidic than the BH vertices exposed toward the cells in either A or B.

Aluminium foams are a new group of materials used for impact energy absorbing elements. They are light (typically 10-25% of the density of the metal they are made of) and stiff, and are frequently proposed as a light weight structural material. That is why they often are applied in automotive and transport industry solutions, for example as parts of bumpers. The methods of numerical modelling for open and closed cell aluminium foams are presented in the paper as well as closed and open cellfoam microstructure model. The numerical models of foam ideal microstructures created with shell finite elements are shown. The models were developed on the basis of Kefain tetrakaidecahedrons - structures consisting of six squares and eight hexagons. In the case of closed cell foams, the polyhedron with full walls was adopted. In the case of open cell foams the circle wholes were removed from polyhedron surfaces. Then the numerical analysis of a created models compressive test was carried out with the usage of LS Dyna computer code. The nonlinear procedures were applied. The results were analyzed in the scope of energy absorbing properties of aluminium foams.

The majority of proteins are unable to translocate into the cell interior. Hence for peptide- and protein-based therapeutics a direct intracytoplasmic delivery with the aid of transducing agents is an attractive approach. We wanted to deliver to the cell interior a putatively cytotoxic protein VPg. Protein transduction was achieved in vitro with three different commercial products. However, in our hands, delivery of various control proteins without known deleterious effects, as well as of protein VPg, always induced cell death. Finally, we used a novel transducing peptide Wr-T, which was not toxic to cultured cells, even in a quite large range of concentrations. Most importantly, control protein delivered to cells in culture did not display any toxicity while VPg protein exerted a strong cytotoxic effect. These data show that results obtained with cell-penetrating agents should be interpreted with caution.

Opis:
Chronic exposure to environmental ultraviolet radiation (UVR) plays a key role both in photocarcinogenesis and induction of accelerated skin ageing. Sailors commonly experience a significant number of cutaneous problems, related to the exposure to environmental factors due to their working conditions. Among these factors, sun exposure is able to determine both acute and chronic skin damage, mostly linked to the effects of the ultraviolet (UV) radiation on epidermal and dermal structures. We report a case of sailor with cutis rhomboidalis nuchae, solar elastosis, actinic keratosis (AK), numerous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) of the head and trunk.

The most important feature of 3G mobile cellular network is introduction of voice data integrated service under multilayered cell environment to support overflow traffic of lower layered cells by upper ones. This paper deals with traffic model of three layered cells, i.e., micro cell, macro cell and satellite cell. Here a new call admission control is introduced for three layered cell of 3G mobile cellular network. State transition chain is designed for theoretical analysis of above mentioned traffic. Blocking probability of data call, new voice call and handover failure of voice call, probability of utilization of micro cell channel, macro cell channel and satellite cell channel are analyzed against different traffic parameters and yield logical results.

Cancer cells are characterized by an aberrant increase in protein N-glycosylation and by disruption of E-cadherin-mediated adherens junctions. However, the relationship between alterations in N-glycosylation process and loss of E-cadherin adhesion in cancer remains unclear. The mechanisms of altered expression of adhesive glycoproteins in cancer cells have not been fully elucidated. Thus, the aim of this study was to examine the expression of E-cadherin and sialyl Lewisa/x, NeuAcα2-3Gal, NeuAcα2-6Gal/GalNAc structures in the normal renal tissue and intermediate and cancerous tissues from patients with clear cell RCC. Moreover, we attempted to correlate the E-cadherin expression with some specific sugar residues of renal cancer tissue glycoproteins. The expression of E-cadherin was analysed using ELISA test and immunoblotting. Oligosaccharide structures and sialylation level were detected with ELISA test using specific biotinylated lectins or antibodies. A significant decrease of E-cadherin expression as well as a significant increase in sialylated oligosaccharides level in intermediate zone and renal cancer tissue in comparison to normal renal tissue are reported. Significant decrease in expression of cadherins and increase in sialylation of oligosaccharide structures in renal cancer tissue in comparison to normal renal tissue, and in renal cancer tissue in comparison to intermediate zone of renal tissue, are important for the future research concerning detection and quantification of cadherins and sialylated oligosaccharide structures in urine and cells of urinary sediment as possible non-invasive marker of early RCC.

Opis:
Cardiac Radiofrequency (RF) ablation is a commonly used clinical procedure for treating many cardiac arrhythmias. However, the efficacy of RF ablation may be limited by two factors: small ventricular lesions and impedance rise, leading to coagulum formation and desiccation of tissue. In this paper, a high frequency (HF) energy ablation system operating at 27.12 MHz based on an automated load matching system was developed. A HF energy matched probe associated to the automated impedance matching device ensures optimal transfer of the energy to the load. The aim of this study was to evaluate this energy for catheter ablation of the atrioventricular junction. In vivo studies were performed using 10 sheep to characterize the lesions created with the impedance matching system. No cardiac perforation was noted. No thrombus was observed at the catheter tip. Acute lesions ranged from 3 to 45 mm in diameter (mean šSD = 10.3š10) and from 1 to 15 mm in depth (6.7š3.9), exhibiting a close relationship between HF delivered power level and lesion size. Catheter ablation using HF current is feasible and appears effective in producing a stable AV block when applied at the AV junction and large myocardial lesions at ventricular sites.

One of the main parameters that affect the solar cell performance is cell temperature; the solar cell output decreases with the increase of temperature. Therefore, it is important to select the proper solar cell technology that performs better at a specified location considering its average temperatures. In addition, the solar cell performance is directly reflected on the overall economics of the project. This paper is proposed to evaluate the variations in the performance of different solar cell technologies related to the temperature in Amman, Jordan. Field data of weather station and three PV systems (Poly-crystalline, Mono-crystalline and Thin-film) of identical design parameters were collected from Test Field Project at Applied Science Private University, Shafa Badran, Amman, Jordan. These data were analysed in the following way. estimated specific energy yield (kWh/kWp) for the three different PV systems was calculated depending on the measured value of solar irradiance and technical specifications of the installed solar panels and inverters, then the actual energy yield at different temperatures over one year was compared with the estimated value, so the deviations could be determined and actual temperature coefficients for energy yield could be calculated, knowing that the three PV Systems have identical design parameters (tilt angle, azimuth angle, type and dimensions of mounting structure and inverter size) and same cleaning method and schedule. It was found that the thin-film solar panels are less affected by temperature with temperature coefficient of -0.0984%, and -0.109%, -0.124% for Mono-crystalline and Poly-crystalline respectively. These results can be implemented in the preliminary design steps, specifically in the selection of the solar cell technology to be installed in a specific location.

The aim of this contribution is to propose a new averaged model of dynamic problems for thin linear-elastic cylindrical shells having a periodic structure along one direction tangent to the shell midsurface. In contrast with the known homogenized models, the proposed one makes it possible to describe the effect of the periodicity cell size on the global dynamic shell behavior (a length-scale effect). In order to derive governing equations with constant or slowly varying coefficients, the known tolerance averaging procedure is applied. The comparison between the proposed model and the model without the length-scale effect as well as the known length-scale model for cylindrical shells with the periodic structure in both directions tangent to the shell midsurface is present.
Celem pracy jest wyprowadzenie uśrednionego modelu służącego do analizy dynamiki cienkich liniowo-sprężystych powłok walcowych mających periodyczną strukturę w jednym kierunku stycznym do powierzchni środkowej powłoki.

The completely new strategy of pseudophysiological transcomplementary (transcytoplasmic) activation (PP-TCA) of nuclear-transferred oocytes, which had been derived from pWAPhGH-GFPBsd transfected foetal fibroblast cells, was recently applied to the somatic cell cloning of pigs. It resulted in the considerable enhancing not only the cleavage activity of cultured cloned embryos, but also their morula and blastocyst formation rates as compared to the use of standard simultaneous fusion and electrical activation of reconstituted oocytes (77% vs. 57%, 63% vs. 46% and 40% vs. 27%, respectively). Altogether, the use of cytosolic components descended from heterologous (rabbit) zygotes as the agents for stimulation of porcine clonal cytoplasmic hybrids (cybrids) turned out to be reliable and feasible strategy for the generation of transgenic blastocysts by somatic cell nuclear transfer (SCNT). Furthermore, to our knowledge, no previous study has reported the preimplantation developmental outcome of transgenic nuclear-transferred pig embryos following the PP-TCA that was developed and optimised in our laboratory.

Opis:
The protozoan Toxoplasma gondii, described by Nicolle and Manceaux in 1908, is a ubiquitous and cosmopolitan parasite that infects a wide range of mammal and bird species with high prevalence. The biological success of T. gondii is associated with the formation of a specific relationship between the parasite and host cells leading to the establishment of a latent, chronic infection. During primary infection, acquired mostly by the oral route, the quickly multiplying tachyzoites disseminate through the body crossing several structural-functional barriers as bloodbrain or blood-retina, then they transform into dormant bradyzoites which, enclosed in tissue cysts, occupy preferentially the brain, skeletal muscle and eye. Although T. gondii is able to infect all kinds of nucleated cells, it uses strictly defined host cells, dependent on the life-cycle phase and infection stage. The article discusses selected aspects of the parasite passing via the host body barriers as well as particular role of dendritic cells and skeletal muscle cells, used by the parasite as an very effective vehicle to disseminate throughout the host body or the site of long-term T. gondii persistence, respectively.

During lateral root (LR) development a coordinate sequence of cell divisions, accompanied by a change of the organ form takes place. Both the order of anatomical events and morphological features may vary for individual primordia. At early stages of LR primordia development oblique division walls are inserted in cells that are symmetrically located on both sides of the axis of the developing LR primordium, and thereby allow for the protrusion of the LR. We hypothesize that both oblique cell wall insertion and continuous changes in primordium form could be a consequence of a local change in stress distribution in the region of the LR initiation.

Monitoring of cellular viability is a key part of toxicological assays in vitro. On-line monitoring of metabolic activity would be particularly useful for evaluation of responses to potential therapeutic compounds. Current assays are mostly based on fluorescent dyes and optical detection methods. These methods offer high sensitivity and specificity, however are not suitable for long-term on-line observations. Electrochemical methods can be an alternative for current protocols. Electrochemical detection is low cost and label-free, therefore suitable for long-term cell culture monitoring. In this work investigations on human cancer cells viability will be presented. Cells were cultured as two-dimensional monolayer or three-dimensional spheroids. Different cell culture media were examined. Potentiometric detection was used for continuous monitoring of cell culture as well as end-point investigations. Different growth phases were identified using applied method. Finally, response to an anticancer drug was successfully observed.

Opis:
The paper describes the immunological response in the matter of percentage of T cells (receptor CD5+) and subpopulations (Th with receptor CD4+, Tc/Ts with receptor CD8+, T with receptor CD25+) and B cells with receptor CD19+, as well as the percentage of apoptotic granulocytes and lymphocytes, in rabbits experimentally infected with the Hagenow strain of the RHD virus. The material chosen for the experiment is special, as among all strains of RHD virus, there are only two strains which carry the variable haemagglutination capacity of red cells. The results of the study show that the Hagenow strain gives an untypical picture of T and B lymphocytes, whereas the results in inducing apoptosis seems to corespond with previous data, confirming the inclusion of apoptosis from 4 h p.i. and the intensity of the phenomenon being higher in granulocytes.

Opis:
The aim of this contribution is to propose a new averaged nonasymptotic model of stationary stability problems for thin linear-elastic cylindrical shells reinforced by stiffeners which are periodically, densely spaced along one direction tangent to the shell midsurface. As a tool of modelling we shall apply the tolerance averaging technique. The resulting equations have constant coefficients in the periodicity direction. Moreover, in contrast with models obtained by the asymptotic homogenization technique, the proposed one makes it possible to describe the effect of the periodicity cell size on the global shell stability (a length-scale effect). It will be shown that this effect plays an important role in the shell stability analysis and cannot be neglected.
W pracy zaproponowano nowy uśredniony nieasymptotyczny model służący do analizy stateczności cienkich liniowo-sprężystych powłok walcowych, periodycznie, gęsto użebrowanych w jednym kierunku stycznym do powierzchni środkowej. Wyprowadzony model porównano z modelem bez efektu skali i pokazano, że wpływ długości okresu periodyczności odgrywa znaczącą rolę w zgadnieniach stateczności periodycznie, gęsto użebrowanych powłok walcowych.

Squamous cell carcinoma of the pinna occurs primarily in older males who are exposed to sunlight for a prolonged period. Most squamous cell carcinomas of the pinna are well differentiated and present as a painless mass or a sessile nodule. We describe an interesting case of an adult woman with a non-healing wound located on the right pinna which was treated conservatively, and was later diagnosed as squamous cell carcinoma.

Opis:
An extensively used fluoroquinolone antibiotic ciprofloxacin shows a broad-spectrum antibacterial activity against both gram-positive and gram-negative strains. It works mainly by the inhibition of DNA gyrase and topoisomerase IV which results in impaired DNA replication leading towards microbial cell death. Eryptosis is an alternative term used for suicidal erythrocyte death. In current study, eryptotic effect of ciprofloxacin was investigated by exposing erythrocytes for 48 hours to the different concentrations (45-90µM) of ciprofloxacin. The experimental work related to the investigation of eryptosis was done by cell size measurement and confirmation of calcium role in membrane blebbing. As a possible mechanism of eryptosis, oxidative stress induced by ciprofloxacin was determined by catalase, glutathion peroxidase and superoxide dismutase activities measurement. Similarly, necrotic effect of ciprofloxacin was also illustrated by hemolysis measurement. Results of our study revealed that the therapeutical doses of ciprofloxacin may induce oxidative stress by significant decrease in superoxide dismutase, catalase and glutathione peroxidase activities as well as induce eryptosis, featured by erythrocytes membrane blebbing and hemolysis.

Opis:
Expression as well as properties of integrins are altered upon transformation. Cell adhesion regulated by integrins is modulated by glycosylation, one of the most frequent biochemical alteration associated with tumorogenesis. Characterisation of carbohydrate moieties of α3β1 integrin on the cultured human bladder carcinoma (T-24, Hu456, HCV 29T) and human normal ureter and bladder epithelium (HCV 29, Hu609) cell lines was carried out after an electrophoresis and blotting, followed by immunochemical identification of α3 and β1 integrin chains and analysis of their carbohydrates moieties using highly specific digoxigenin-labelled lectins. In all the studied cell lines α3β1 integrin was glycosylated although in general each subunit differently. Basic structures recognized in β1 subunit were tri- or tetraantennary complex type glycans in some cases sialylated (T-24, HCV 29, HCV 29T) and fucosylated (Hu609, HCV 29T). Positive reaction with Phaseolus vulgaris agglutinin and Datura stramonium agglutinin suggesting the presence of β1-6 branched N-linked oligosaccharides was found in cancerous cell lines (T-24, Hu456) as well as in normal bladder epithelium cells (Hu609). High mannose type glycan was found only in β1 subunit from Hu456 transitional cell cancer line. On the other hand α3 subunit was much less glycosylated except the invasive cancer cell line T-24 where high mannose as well as sialylated tri- or tetraantennary complex type glycans were detected. This observation suggests that changes in glycosylation profile attributed to invasive phenotype are rather associated with α3 not β1 subunit.

Opis:
Obtaining a suitable experimental cellular model is a major problem for neuroscience studies. Neuroblastoma cell lines have been often applied in studies related to pathological disorders of nervous system. However, in the search for an ideal model, these cells must be differentiated to cancel their tumor character. The subsequent reactions that are caused by differentiation are not always indifferent to the same model. We evaluated the effect of two well known substances, used for SH-N-SK cell line differentiation, retinoic acid (RA) and phorbol-12-myristate-13-acetate (PMA), on the induction of pro-inflammatory and pro-oxidative reactions in these cells. Cells differentiated with PMA were able to produce significantly higher amounts of pro-inflammatory cytokines whereas the release of nitric oxide radicals was similar to that in undifferentiated cells. On the contrary, in RA-differentiated cells no significant changes in cytokine production were observed and the nitric oxide release was decreased. Additionally, the RA-differentiated neuronal model was more sensible to lipopolysaccharide stimulation, producing pro-inflammatory cytokines abundantly. These results suggest that RA-differentiated SH-N-SK cells provide a more suitable experimental model for the study of molecular and cellular mechanisms of the inflammation and oxidative stress in neuronal cells.

Opis:
Background. Chronic autoimmune inflammation in arterial wall may lead to atherosclerosis progression. Objective. The aim of this study was to investigate the association between Treg, Th17 and B1a cell blood frequencies as well as IgM autoantibodies to oxLDL and the abundance of carotid atherosclerosis. Material and methods. 18 patients with increased IMT (intima-media thickness) and 65 patients with different severity of carotid atherosclerotic plaques were included. Treg, Th17 and B1a cell blood frequencies were assessed via direct immunofluorescence staining and flow cytometry, oxLDL as well as IgM autoantibodies to oxLDL were measured with commercial kits. Results. We observed higher values of Treg in patients without carotid atherosclerosis. Patients with intact carotid arteries as compared to patients with mild atherosclerotic plaques had decreased Th1 levels. OxLDL IgM levels were higher in patients with intact carotid arteries. Patients who received statin treatment had higher levels of Treg. Immunophenotyping of B cells revealed two cases of monoclonal B-cell lymphocytosis and 1 case of B-CLL (B-cell chronic lymphocytic leukemia) in elderly patients with intact carotid arteries. Conclusion. We hypothesize that certain parameters of cell immunity may hamper atherosclerosis while protecting from lymphoproliferative disorders.

Alterations in the genetic apparatus of mice bone marrow cells and testicles caused by Hymenolepis nana, Ascaris suum and Toxocara canis metabolites have been investigated by means of micronuclear test application. The activity of spermatogenesis has been investigated at experimental hymenolepidosis, migrating ascariasis and toxocarosis with the use of ³H-timidine. Helminths metabolites have been established to exert a mutagenie effect on somatic cells of bone marrow, spermatogonies and also on the generative cells (spermatides) of helminths in invaded mice. The concurrent increase in micronucleus number in erythrocytes, spermatogonies and in spermatides (to a lesser degree) of invaded mice has been revealed. The decrease in spermatogenesis activity has been established in experimental hymenolepidosis, migrating ascariasis and toxocarosis in invaded mice.

The aim of the study was to determine whether treatment with recommended doses of meloxicam or flunixin had an effect on the apoptosis of peripheral blood T lymphocytes in calves. The study was carried out on 4-5 months old calves (n = 24, 8 per group). Experimental animals were injected subcutaneously with a single dose of 0.5 mg . kg-1 of meloxicam or intravenously with 3 doses of 2.2 mg . kg-1 day-1 of flunixin. The non-treatment animals served as control. Blood samples were taken at day 0 and at days 1, 2, 3, 5, 7 and 14 after the first NSAIDs injection. Apoptosis was determined by flow cytometry using Annexin V-PE/7-AAD staining. The kinetic analysis of apoptosis in the total lymphocyte population, as well as in the CD4+ and CD8+ subsets did not reveal significant differences in the frequency of early apoptotic cells between control and experimental groups throughout the period studied. Although, 24 h after administration of the first dose of NSAIDs, late-stage apoptosis/necrosis was significantly increased in the total lymphocyte population (the meloxicam group), as well as in the CD4+ (the meloxicam group and the flunixin group) and CD8+ (the flunixin group) subsets of T cells. However, this disturbance was transient, relatively poorly expressed and, thus, unlikely to be of clinical significance. Our results indicate that the use of meloxicam or flunixin in accordance with the recommended dosage regimen in cattle do not have a clinically significant influence on apoptosis of peripheral blood T cells.

Opis:
Lab-on-a-chip systems are promising tools in the field of cell engineering. Microfluidic systems are integrated microlaboratories consisting of many microstructures such as microchannels and microchambers, which can be used for cell analysis and cell culture. Appropriately designed geometry of the chip allows to mimic in vivo conditions. Microsystems enables continuous culture medium perfusion. During cell culture, regulation of the flow rate of medium is possible, which allows to control conditions of the cultivation. In this paper we present a review of microfluidics systems which are used in cell engineering. We describe methods of microsystems fabrication, parameters which influence cell proliferation in microscale and examples of microsystems for cell analysis and cell culturing. Microfluidic systems for maintaining cell culture are mainly fabricated of poly(dimethylsiloxane) (PDMS) and glass, non-toxic materials for cells. The most commonly used method for fabrication of PDMS microsystems is photolithography and replica molding techniques. Cell culture in microsystems can be carried out in two ways: as a two-dimensional (2D) cell culture and three-dimensional (3D) cell culture. In two-dimensional culture cells grow as a monolayer on a flat surface of microchambers or microchannels. Microsystems for two-dimensional cell culture are widely described in the literature. They are mainly used for: (i) cell proliferation after exposure to external stimuli, (ii) testing the activity of cytotoxic drugs, (iii) interactions and cell migration and (iv) the evaluation of procedures applicable in tumor therapy e. g. photodynamic therapy. However, two-dimensional cell culture do not mimic fully in vivo conditions. In living organisms cells grow spatially creating three-dimensional structures like tissues. Therefore, nowadays microsystems for 3D cell culture are being developed intensively. Three-dimensional cell culture in microfluidic systems can be achieved in three ways: by the design of suitable geometry and topography of microchannels, by the use of hydrogels or by spheroids formation. Three-dimensional cell culture in microfluidic systems are much better experimental in vitro models than cell culture in traditional culture vessels. It is the main reason why microsystems should be still improved, as to become widely used research tools in cellular engineering.

Nineteen canine lymphomas were included in this study. Tumors were classified according to the updated Kiel classification adapted for canine lymphomas by Fournel-Fleury et al. Immunoglobulin light chains (κ and λ) and IgM and IgG expression were determined by immunohistochemical method. In all examined cases neoplastic cells were positive for one of the immunoglobulin light chains. Expression of λ light chains and κ light chains was observed in 18/19 and 1/19 tumors, respectively. In the majority of neoplastic cells in each examined specimen this reaction had a membranous pattern (sκ/sλ). In all examined cases the presence of immunoglobulin light chains was also observed in the cytoplasm of some neoplastic cells (cκ/cλ). These cells were usally rare and never constituted a dominant population. The expression of immunoglobulin was found in 13/19 cases. Most lymphomas were sIgM positive (11/13 cases). In one case expression of IgG was found, and in another lymphoma two populations of neoplastic cells with different expression of examined immunoglobulins (cells with IgM+ and IgG+ phenotypes) were observed. The reaction also had a membranous pattern. The cells containing cytoplasmic immunoglobulins were rare, and in most cases were of the same type as the surface immunoglobulins. Our study has confirmed that canine lymphomas are a monoclonal proliferation of B-cells usually expressing immunoglobulin λ light chains and that the vast majority of tumors deriving from B-cells express IgM. Our study also indicates a possibility of occurence of biclonal lymphomas in canine species.

Microfluidic devices, such as lab-on-a-chip systems, are highly advantageous for cell engineering and cell based assays. It is a particularly useful approach for development of the in vitro cellular systems mimicking the in vivo environment. In this paper, a novel lab-on-a-chip device for three-dimensional human cell culture and anticancer drug testing is presented. Cells were cultured as Multicellular Tumor Spheroids (MCTS) — the best cancer tumor model developed so far. Diff erent designs were tested and novel technique of microfabrication in poly(dimethylsiloxane) was developed. MCTS were cultured in a system of polymeric microwells, with the network of microfluidic channels for culture medium flow. Design included optimal shear stress and proper nutrients supply for cultured cells. Final design provided MCTS culture for four weeks with the homeostasis-like state achievement, which is characteristic for the in vivo situation.

These particles in the form of microplates or polyhedral microcrystals were deposited on titanium plates from ethanol solutions, dried and annealed with a hydrogen flame. Some samples were additionally modified by polyethylene imine. The materials engendered from these modifications were used to investigate the adhesion and growth of human osteoblast-like MG-63 cells on these surfaces in the DMEM medium with 10% of fetal bovine serum. One day after seeding, the highest number of initially adhered cells was found on the surfaces modified by both types of gold microparticles. This trend was the same three and seven days after seeding. The numbers of cells on pure Ti and Ti modified only with gold particles were significantly higher than on samples which were modified with polyethylene imine. The cell spreading areas projected on the materials were significantly larger in cells on the samples with polyethylene imine modification. However, the shape of these cells was mostly rounded or star-like with thin and long protrusions, while on the materials without polyethylene imine, it was mostly polygonal. The cell proliferation activity was estimated from XTT test, based on the activity of mitochondrial enzymes. This test showed that the proliferation activities of osteoblast-like MG-63 cells of the 3rd and 7th days of the experiment were more pronounced on the samples modified only by gold microparticles. Immunofluorescence showed that the focal adhesion plaques containing vinculin and the fibers containing β-actin were most apparent, more numerous and more brightly stained in cells on Ti modified by gold microplates and gold polyhedral microcrystals, especially in comparison with the corresponding samples modified with polyethylene imine (Fig. 1). Thus, it can be concluded that the modification of titanium samples by both types of gold microparticles enhanced the adhesion and growth of MG 63 cells.

Opis:
Future wireless communication networks will be largely characterized by small cell deployments, typically on the order of 200 meters of radius/cell, at most. Meanwhile, recent studies show that base stations (BS) account for about 80 to 95 % of the total network power. This simply implies that more energy will be consumed in the future wireless network since small cell means massive deployment of BS. This phenomenon makes energy-efficient (EE) control a central issue of critical consideration in the design of future wireless networks. This paper proposes and investigates (the performance of) two different energy-saving approaches namely, adaptive-sleep sectorization (AS), adaptive hybrid partitioning schemes (AH) for small cellular networks using smart antenna technique. We formulated a generic base-model for the above-mentioned schemes and applied the spatial Poisson process to reduce the system complexity and to improve flexibility in the beam angle reconfiguration of the adaptive antenna, also known as a smart antenna (SA). The SA uses the scalable algorithms to track active users in different segments/sectors of the microcell, making the proposed schemes capable of targeting specific users or groups of users in periods of sparse traffic, and capable of performing optimally when the network is highly congested. The capabilities of the proposed smart/adaptive antenna approaches can be easily adapted and integrated into the massive MIMO for future deployment. Rigorous numerical analysis at different orders of sectorization shows that among the proposed schemes, the AH strategy outperforms the AS in terms of energy saving by about 52 %. Generally, the proposed schemes have demonstrated the ability to significantly increase the power consumption efficiency of micro base stations for future generation cellular systems, over the traditional design methodologies.

In The present study, the aims was to investigate and compare the effect of insulin and embryonic stem cells (ESC) loaded nanoparticle formulations (NPs) on pancreatic-beta-TC-cell regeneration. Characterization studies of NPs were performed. Permeability of insulin and the effect of ESC on pancreatic beta cells were investigated. by the determination of Insulin or glucose levels were determined and histologic investigations were also performed. ESC encapsulation efficiency was calculated by western blot analysis. The particle sizes of insulin and ESC-loaded-NPs were determined as 0.665±0.202 µm and 0.650 ±0.310 µm. The mean zeta potentials of insulin and ESC-loaded-NPs nanoparticles were found as 6.88±0.729 mV, 5.13±0.631 mV. The polydispersity index of insulin and ESC nanoparticles were 0.660±0.175, 0.620±0.205 respectively. Encapsulation efficiency of insulin and ESC-loaded-NPs were found to be 50±1.53% and 51%. Insulin release from nanoparticles was found to be 72.8% over 48h. The gGlucose concentrations wasere decreased to 201 and 202.7 mg/dl from 250 mg/dl in streptozocin (STZ) induced diabetic mice group after insulin and ESC-loaded-NPs administration. Insulin and ESC-loaded-NPs improved the blood insulin levels in all experimental groups. These NPs may be used for repairing of pancreatic cells. Healing or some degree of regeneration was observed when insulin and ESC-loaded-NPs were administered to the mice ip. ESC-loaded-NPs can be a potential source for cell replacement therapy in the treatment of diabetes.

Renal cell carcinoma accounts for about 2-3% of all malignancies. It is characterized by a high frequency of metastases, which are most often observed in the lungs, bones, liver, brain and regional lymph nodes. Literature highlights the rare occurrence of renal cancer metastases to the head and neck, even though it is the third most common cancer that metastasizes to the head and neck. Below we present a case report of metastases of clear cell carcinoma from a kidney to the organs of the head and neck. The analysis was carried out retrospectively based on medical records of a patient hospitalized at the Department of Otorhinolaryngology, Head and Neck Surgery of the Medical University of Warsaw in the years 2009-2019. We describe an 81-year-old man after left-sided nephrectomy due to clear cell carcinoma, with numerous distant metastases, who was diagnosed with metastases to the left submandibular salivary gland 8 years after primary surgery, followed by a diagnosis of metastasis to the right submandibular salivary gland 3 years later. The patient underwent 2 radical procedures to remove the submandibular salivary glands. Due to the presence of lung metastases, treatment with a tyrosine kinase inhibitor was also implemented.

Opis:
Metacaspases, cysteine proteases belonging to the peptidase C14 family, are suspected of being involved in the programmed cell death of plants, although their sequences and substrate specificity differ from those of animal caspases. At present, the knowledge on the metacaspase reaction mechanism is based only on biochemical data and homology models constructed on caspase templates. Here we propose a novel template for metacaspase modeling and demonstrate important advantages in comparison to the conventionally used caspase templates. We also point out the connection between plant and bacterial metacaspases, underlining the prokaryotic roots of Programmed Cell Death (PCD).

We have previously shown that this cell is a new solution for photonics [1, 2]. This method involves physical and mathematical models of the phenomena relevant to that observed in these devices photorefractive-like effect. Based on the presented method was established hybrid cell simulator. This simulator enables the design of cells of this type and allows the study as designed by the user device changes its optical properties under the control field and lighting. In this paper we describe the main assumptions of the mathematical models used in the described method. In addition, we present algorithms in order to obtain a numerical solution proposed models.

Opis:
Background. Current studies have indicated many environmental factors, such as pesticides, that cause immune system disorders through inducing changes in humoral and cellular responses thereby increasing the risk of contracting infectious diseases and cancer. The literature suggests that low exposures to certain organophosphorus pesticides stimulate the immune system, whilst high exposures result in decreased function. Precise mechanisms for the fall in immunocompetence are often unclear, however it can be predicted that the intimate interaction between the nervous and immune systems can potentially lead to toxicity. Objectives. To determine the effects of organophosphorus pesticide, chlorpyrifos that is often used in Poland, on selected immunological responses, such as immune-competent cell proportions formed experimentally in-vivo by cells of Wistar rats during subchronic exposures after 45 and 90 days. Materials and Methods. The test was carried out on ten male and ten female Wistar rats in each of three test groups, who received 3 chlorpyrifos doses for 90 days intragastrically, according to OECD guidelines (No. 401). Two control groups were given olive oil. After completion, the animals were deeply anaesthetised by a mixture of ketamine (Vetaketam) and xylazine (Vetaxym). Immuno-competent cells were profiled by a commercial monoclonal antibody method. In order to measure the dynamics of any changes, the aforementioned immunological responses were investigated after 45 days using the same procedures for obtaining the relevant biological test material. Results. Test animals exposed to chlorpyrifos had altered number of white bood cells which were either increased or decreased relative to controls after 45 and 90 days for all exposure levels used. Conclusions. The study demonstrated changes in white-blood cell (lymphocyte) response profiles, reflecting an immunomodulation although such changes were equivocal, where both suppression and stimulation were observed.
Wprowadzenie. Materiał i metody. Badanie wykonano na samcach (10 szt.) i samicach (10 szt.) szczurów rasy Wistar. Zwierzętom z grupy badanej podawano dożołądkowo chloropiryfos przez 90 dni, zgodnie z zaleceniami OECD (Guideline No 401). Grupa kontrolna otrzymywała oliwę z oliwek. Po zakończeniu eksperymentu zwierzęta poddawano głębokiej narkozie przy użyciu mieszaniny: ketaminy (Vetaketam) i ksylazyny (Vetaxym). W celu określenia dynamiki ewentualnych zmian zbadano wyżej wymienione parametry immunologiczne po 45 dniach doświadczenia przy zachowaniu tych samych procedur uzyskania materiału badawczego. Wyniki. Wnioski. Odnotowane w wyniku przeprowadzonych badań zmiany w profilu białokrwinkowym świadczące o działaniu immunomodulującym były niejednoznaczne przyjmując formę zarówno immunosupresji jak i immunostymulacji.

Opis:
Multicrystalline and monocrystalline silicon solar cells are the most popular for commercial applications. Numerous material parameters could affect solar cell performance. The application of antireflective coatings of amorphous silicon based alloys can increase device efficiency. Such coatings due to tunable energy gap and refractive index and non-expensive fabrication method - Plasma Enhanced Chemical Vapour Deposition at 13.56 MHz are good candidates for large scale application. The authors presented the influence of material properties of bulk cell on solar cell efficiency by the use of computer PC-ID simulation program, which solves the fully coupled nonlinear equations for the quasi-one-dimensional transport of electrons and holes in crystalline devices, with emphasis on photovoltaic devices. The temperature, series and shunt resistance, recombination velocity and wafer thickness have the influence on current--voltage photo-characteristics of solar cells and their efficiency. Also the influence of thickness, reflective coefficient and refractive index of antireflective coatings on solar cells performance was examined. The optimal parameters of efficient solar cell were determined.

Opis:
Recently, significant attention has been paid to the possibility of thwarting cancer progression by inhibition of neoangiogenesis (formation of new blood vessels) in growing tumors. Although general mechanisms of angiogenesis have been elucidated, virtually nothing is known about the effects of low doses of ionizing radiation on pro-angiogenic properties of endothelial cells. In the present study, we evaluated the effects of a low (0.2 Gy), intermediate (1 Gy), and high (4 Gy) doses of X-rays on a few angiogenesis-related parameters of isolated murine endothelial cells. We show here that 24 to 48 hours after irradiation with 0.2 Gy the cell proliferation was inhibited to a similar extent as after the exposure to 1 Gy. Also, adhesion of the 0.2 Gy-irradiated cells to both gelatin and MatrigelŽ was inhibited 24 hours post-exposure, whereas irradiation with 1 or 4 Gy resulted in the increased adhesion of the cells to these substrata. Similar effects were observed during the "wound" migration assay. Finally, 24 hours after exposure of the cells to 0.2 Gy of X-rays, the surface expression of the â3 integrin subunit was down-regulated, whereas irradiations with 1 and 4 Gy of X-rays resulted in the significantly elevated expression of this subunit. These results indicate that proliferating endothelial cells are sensitive in vitro to relatively low doses of ionizing radiation

open cell foam structure
Opis:
Metal and polyurethane foams exhibit interesting properties. They are light, have good acoustic and/or magnetic isolation as well as ability to absorb the vibration and impact energy. They are used for sandwich panels, impact absorbers (i.e. as elements of the buffer constructions in rail vehicles), fillers ofconstruction parts, bodies of vehicles (i.e. floating combat vehicles) and for dividing walls of vessels and others. Speciflcally made open cell foams demonstrate auxetic properties and the shape memory effect. Such materials are very good for seats in aircrafts which may protect pilots and passengers during crashes and limit heavy backbone injuries. Foams are used for filtering purposes. Foams or their in combination with different types of fillers (i.e. elastomers) or the ceramic reinforcement may be used in impact energy absorbing panels for military aim (protection against an explosion shock wave and splinters). In the paper aluminium open-cell foam structures were investigated for their energy absorption ability. For this purpose a series of numerical 3D models were applied. Geometry of the models was based on Kehin's polyhedrons. Tests of uniaxial compression were simulated with the use of LS-Dyna computer code. Complex contact phenomena were considered. Preliminary test were performed with the use of single foam cell models. Further simulations were conducted using 3x3x3 cell models. The results were analyzed as force/time characteristics.

Opis:
Langerhans cell histiocytosis is a tumor-like condition characterized by idiopathic proliferation of Langerhans cells. The disease may involve the skeleton as well as other organs systems. Bone involvement may be solitary or multifocal. Unifocal osseous Langerhans cell histiocytosis may involve virtually any bone, with the calvarium being most frequently involved. Plain radiography, computed tomography and magnetic resonance imaging are the most used techniques for detection and characterization of the lesion. The use of ultrasound is less known, although it may be a valuable technique in detection and follow-up of superficially located lesions such as calvarial lesions. This case report describes an 8-year-old girl, in whom the lesion was initially detected by ultrasound. Furthermore, ultrasound was used to evaluate spontaneous resolution of the lesion. The knowledge of ultrasound characteristics may be important to avoid unnecessary radiation and gadolinium administration, particularly in a pediatric population.

Opis:
A body of evidence, linking inositide-specific phospholipase C (PI-PLC) to the nucleus, is quite extensive. The main isoform in the nucleus is PI-PLCβ1, whose activity is up-regulated in response to insulin-like growth factor-1 (IGF-1) or insulin stimulation. Whilst at the plasma membrane this PI-PLC is activated and regulated by Gαq/α11 and Gβg subunits, there is yet no evidence that qα/α11 is present within the nuclear compartment, neither GTP-γ-S nor AlF4 can stimulate PI-PLCβ1 activity in isolated nuclei. Here we review the evidence that upon occupancy of type 1 IGF receptor there is translocation to the nucleus of phosphorylated mitogen-activated protein kinase (MAPK) which phosphorylates nuclear PI-PLCβ1 and triggers its signalling, hinting at a separate pathway of regulation depending on the subcellular location of PI-PLCβ1. The difference in the regulation of the activity of PI-PLCβ1mirrors the evidence that nuclear and cytoplasmatic inositides can differ markedly in their signalling capability. Indeed, we do know that agonists which affect nuclear inositol lipid cycle at the nucleus do not stimulate the one at the plasma membrane.

Opis:
Integrin subunits present on human bladder cells displayed heterogeneous functional specificity in adhesion to extracellular matrix proteins (ECM). The non-malignant cell line (HCV29) showed significantly higher adhesion efficiency to collagen IV, laminin (LN) and fibronectin (FN) than cancer (T24, Hu456) and v-raf transfected (BC3726) cell lines. Specific antibodies to the α2, a5 and β1 integrin subunits inhibited adhesion of the non-malignant cells, indicating these integrin participation in the adhesion to ECM proteins. In contrast, adhesion of cancer cells was not inhibited by specific antibodies to the β1 integrin subunit. Antibodies to α3 integrin increased adhesion of cancer cells to collagen, LN and FN, but also of the HCV29 line with colagen. It seems that α3 subunit plays a major role in modulation of other integrin receptors especially in cancer cells. Differences in adhesion to ECM proteins between the non-malignant and cancer cell lines in response to Gal and Fuc were not evident, except for the v-raf transfected cell line which showed a distinct about 6-fold increased adhesion to LN on addition of both saccharides. N-Acetylneuraminic acid inhibited adhesion of all cell lines to LN and FN irrespective of their malignancy.

Changes in the expression of integrins and cadherins might contribute to the progression, invasion and metastasis of transitional cell cancer of the bladder and of melanomas. The expression of α5 (P < 0.001), α2 and β1 (P < 0.05 - P < 0.001) integrin subunits in melanoma cells from noncutaneous metastatic sites (WM9, A375) were significantly increased as compared to cutaneous primary tumor (WM35) and metastatic (WM239) cell lines. These differences might be ascribed to the invasive character of melanoma cells and their metastasis to the noncutaneous locations. The significantly heterogeneous expression of β1 integrin subunit in two malignant bladder cancer cell lines (T24 and Hu456) and nonsignificant differences in the expression of α2, α3, and α5 subunits between malignant and non-malignant human bladder cell lines do not allow an unanimous conclusion on the role of these intergrin subunits in the progression of transitional cancer of bladder. The adhesion molecule, expressed in all studied melanoma and bladder cell lines, that reacted with anti-Pan cadherin monoclonal antibodies was identified as N-cadherin except in the HCV29 non-malignant ureter cell line. However, neither this nor any other bladder or melanoma cell line expressed E-cadherin. The obtained results imply that the replacement of E-cadherin by N-cadherin accompanied by a simultaneous increase in expression of a2, a3 and a5 integrin subunits clearly indicates an increase of invasiveness of melanoma and, to a lesser extent, of transitional cell cancer of bladder. High expression of N-cadherin and a5 integrin subunit seems to be associated with the most invasive melanoma phenotype.

PCR identified the monoclonal nature of the T-cells. The hematological study was normal. Following biopsy, the conjunctival tumor diminished until disappear. Conclusions: Mucosal CD30-positive T-cell lymphoproliferation could be ascribed to a reactive process but it is necessary to rule out a primary conjunctival tumor or the first manifestation of an anaplastic large cell lymphoma.

Objective: Results of second-line everolimus treatment in clear cell renal carcinoma. Material and methods: Clinical data of 32 patients of the Clinic of Systemic and Metastatic Malignancies, Centre of Oncology, Maria Sklodowska-Curie Memorial Institute (Krakow Branch) treated with second-line everolimus between 2011 and 2016 were analysed. Results: Median progression-free survival was 7 months. Stable disease was observed in 24 cases. Adverse events occurred in 59% of all patients, and the majority were mild and moderate (G1 and G2) according to CTCAE. The most frequent adverse events included anaemia, stomatitis, rash and fatigue. Conclusion: Molecularly targeted drugs offer the only effective and available therapy in Poland, which can be administered in patients with advanced renal cell carcinoma. Second-line everolimus is a valuable therapeutic option in the above mentioned group (following progression on first-line TKI therapy), especially when it is necessary to avoid the accumulation of similar adverse events after TKI treatment (different toxicity profiles).

Since 1991, the Foundation for Cardiac Surgery Development in Zabrze has been implementing research on the artificial heart. In 1995, an artificial ventricle, POLVAD, was implanted to a patient, and in 1998, the prototype of a clinical controller, POLPDU-401, was created. A further development of the studies on an implantable artificial heart requires an integrated approach and an application of advanced methods of materials sciences in order to develop new materials suitable for the contact with blood, as well as to apply a multilateral biomedical diagnostics in hydrodynamic conditions. The estimation of the cell–material interaction plays an important role in the biomaterial design. An analysis of the influence of the carbon content in titanium nitride on the biological and biophysical properties of biomaterial coatings was studied. The cell-material reactions were considered in dynamic and static conditions. Three groups of materials were under examinations – titanium nitride (TiN), as well as titanium carbonitride with a low and high carbon content – of which the best properties were observed for TiN. We found a strong influence of the stoichiometry of TiN (atomic ratio of Ti/N) on the biocompatibility. A non-stoichiometric TiN could have a negative influence on the surrounding tissue.

In today’s manufacturing systems, especially in Industry 4.0, highly autonomous production cells play an important role. To reach this goal of autonomy, different technologies like industrial robots, machine tools, and automated guided vehicles (AGV) are deployed simultaneously which creates numerous challenges on various automation levels. One of those challenges regards the scheduling of all applied resources and their corresponding tasks. Combining data from a real production environment and Constraint Programming (CP-SAT), we provide a cascaded scheduling approach that plans production orders for machine tools to minimize makespan and tool changeover time while enabling the corresponding robot for robot-collaborated processes. Simultaneously, AGVs provide all production cells with the necessary material and tools. Hereby, magazine capacity for raw material as well as finished parts and tool service life are taken into account.

Cell number, density and volume of white and gray matter in brain structures are not constant values. Cellular alterations in brain areas might coincide with neurological and psychiatric pathologies as well as with changes in brain functionality during selection experiments, pharmacological treatment or aging. Several softwares were created to facilitate quantitative analysis of brain tissues, however results obtained from these softwares require multiple manual settings making the computing process complex and time-consuming. This study attempts to establish half automated software for fast, ergonomic and an accurate analysis of cellular density, cell number and cellular surface in morphologically different brain areas: cerebral cortex, pond and cerebellum. Images of brain sections of bank voles stained with standard cresyl-violet technique (Nissl staining), were analyzed in designed software. Results were compared with other commercially available tools regarding number of steps to be done by user and number of parameters possible to measure.

The subject of the study are alumina foams produced by gelcasting method. The results of micro-computed tomography of the foam samples are used to create the numerical model reconstructing the real structure of the foam skeleton as well as the simplified periodic open-cell structure models. The aim of the paper is to present a new idea of the energy-based assessment of failure strength under uniaxial compression of real alumina foams of various porosity with use of the periodic structure model of the same porosity. Considering two kinds of cellular structures: the periodic one, for instance of fcc type, and the random structure of real alumina foam it is possible to justify the hypothesis, computationally and experimentally, that the same elastic energy density cumulated in the both structures of the same porosity allows to determine the close values of fracture strength under compression. Application of finite element computations for the analysis of deformation and failure processes in real ceramic foams is time consuming. Therefore, the use of simplified periodic cell structure models for the assessment of elastic moduli and failure strength appears very attractive from the point of view of practical applications.

Two-dimensional gel electrophoresis was used to compare protein expression profiles between various stages of Gentiana kurroo Royle somatic embryos. Seven distinct stages (I–VII) were pointed out and measured from long-term embryogenic cell suspension. Isoelectric focusing was done in the pH intervals 3–10, and the second dimension was carried out with 13% SDS-polyacrylamide gel electrophoresis. Dependent on the stage from about 400 (stage IV) to more than 700 (stage II) protein spots were totally detected. The molecular weight of abundant proteins range from 12 to 70 kDa, however, majority of proteins were located between 20–49 kDa spots on the gels. The highest difference in the number of spots appeared in the case of globular embryo (stage I) and elongated cotyledonary stage (stage VII) with differences being about 130 spots. The relevance of embryogenic cell suspension choice for proteomic analysis as well as expediency of the increasing number of particular embryo stages is discussed.

Therefore, there were difference in bee space and comb thickness in similar honeybee race among two different agroecology but preparing of new casting molds and box hives consistent to the agroecology.

Opis:
The paper presents an approach to the modelling of a cyclic battery tester and contains observations about lithium-ion (Li-ion) batteries, charging/discharging procedures, conditions and protections which must be observed during the testing process. The main goal was to create and simulate a schematic which will be capable of cyclically testing Li-ion battery cells. Regulation of the final schematic is based on cascade connections of operation amplifiers, which work as a voltage source with current-limiting functions. The power part is created by two MOSFETs connected as a half-bridge. This topology allows current to flow in both directions (from and to the battery). Final simulation is supplemented by protections such as reverse polarity protection, short circuit protection and overvoltage protection during charging. Proper operation of the whole connection is demonstrated by the simulation outputs in the final section.

Opis:
Discrete cluster microforms, or simply clusters, in gravel streams result from organization of particles found in the surface layer of the gravel bed into disconnected patches. Clusters are the outcome of feedback interaction between flow, sediment and stream planform geometry. The complexity of this interaction results in several different cluster shapes, i.e. line, rhomboid and triangular. The objective of this research is to provide a quantitative characterization of cluster shape. To achieve this, we employed a novel method based on fractal theory and used for the shape description of clusters. Our novel method utilized the cell-counting method for the estimation of the areal fractal dimension, for two major datasets, namely fabricated clusters with well-defined shapes, and clusters developed in the laboratory. The principal finding of this research is that the proposed method successfully characterized cluster shape in quantitative terms. Specifically, it was shown that the new approach could identify clusters of different shapes 84% of the time, under different arrangements. This finding is of great importance for bed pattern recognition studies of stream reaches with superimposed roughness elements such as clusters. The findings of the current work could also assist numerical modellers in the development of more representative models of flows over roughness features such as clusters and in the interpretation of results from such models.

Opis:
. A single scintillation cell (ö 54 × 74 mm) is used for the measurement of radon and thoron. The radon and thoron laden air is filtered and forced to flow at 1 dm3/min through the scintillation cell in the period of 1 10 min. The count number from alpha radiation is registered in the periods of 3 10 min and 20 30 min. Two values of detection and deposition efficiency of alpha radiation are used for radon (at air flow and air at rest in the cell) and for thoron. Measurements of radon laden air and thoron laden air showed good agreement between the reference concentration and the measured concentration, not worse than 1% for radon and not worse than 2% for thoron. Combination of radon + thoron concentration showed also a small interference ("cross talk") not worse than 1%.

Opis:
Rehabilitative modalities such as incentive spirometry and physiotherapy interventions, e.g. aerobic exercises, have been shown to produce positive outcomes in patients with sickle cell disease (SCD). Hemispheric CVA and other complications arising from SCD are amenable to physiotherapy. There have been few studies on the effectiveness of physiotherapy in the management of individuals with SCD. The present study attempts to assess the SCD-related knowledge, attitude and level of participation of physiotherapists in the Nigerian healthcare system. It also examines a possible association between the foregoing and practice settings. The study was carried out among physiotherapists in South-West Nigeria registered on professional e-platforms. The respondents filled in an adapted electronic version of a questionnaire. The IBM SPSS 20 was used for statistical analysis, and a chi-square test was used to determine an association between the levels of knowledge, practice settings, and participation among the respondents (p < 0.05). The study findings revealed that only 19% of respondents had received a specific training related to the care of SCD patients, although 65% had been involved in the treatment of at least one patient with SCD. Only 29.5% of respondents had “Good Knowledge” of SCD pathophysiology. A statistically significant association (χ2 = 75.357, p = 0.012) was found between respondents’ level of knowledge and their practice settings, i.e. a teaching hospital mostly. The physiotherapists from the Nigerian South-West,mostly those working in teaching hospitals, reported an average level of knowledge of SCD care. Since the management of sickle cell disease is of multidisciplinary character and requires an all hands-on deck approach, controlled trials should be undertaken by physiotherapy researchers regarding the effects of modalities and interventions on pro-inflammatory bio-molecules.

Cabozantinib, the latest available in Poland medication for the treatment of renal cell carcinoma, registered in this indication by the European Medicines Agency (EMA) in September 2016, has been available in several cancer centers in Poland since November 2016 as part of the expanded access program. Primary hypothyroidism is a common complication during thyrosine kinase inhibitors (TKI) treatment, although there are few reports of its occurrence during treatment with cabozantinib, which belongs to this medication group. We present a case of rapid development of clinically apparent hypothyroidism after cabozantinib treatment and report data on this complication in the group of our patients.
