Stem cells have captured considerable scientific and clinic interest because of their potential to renew themselves and to differentiate into one or more adult cell types. Thus stem cells have been recognized as a potential tool for the development of innovative therapeutic strategies in different disease disorders. Stem cells can be discriminating based on their differentiated potential as totipotent, pluripotent, multipotent or unipotent cells. There are in general three types of stem cells: embryonic, fetal and adult stem cells. While embryonic stem cell therapy has a lot of ethical concerns due to their obtaining but also unlimited proliferation and uncontrolled differentiation, fetal and adult stem cells have been used in the treatment of different diseases. The bone marrow, peripheral blood and umbilical cord blood are ideal sources of adult stem cells because there are easily accessible and contain two types of stem cells: hematopoietic stem cells giving rise to all blood cell types and mesenchymal stem cells differentiating into cells of mesodermal lineage. This review describes the general characteristics of these stem cell populations and their current applications in regenerative medicine. Additionally induced pluripotent stem cells generated through the reprogramming of differentiated adult cells are described.
Możliwość wspomagania regeneracji nieodwracalnie uszkodzonych tkanek i narządów za pomocą transplantacji komórek macierzystych skupia uwagę środowisk medycznych i opinii publicznej. Komórki macierzyste mają zdolność do samo-odnowy oraz do różnicowania się w dojrzałe komórki struktur całego organizmu. Na określonych etapach rozwoju osobniczego wykazują różny stopień ograniczenia potencjału do dalszego różnicowania, od komórek totipotencjalnych (zygota), poprzez pluripotencjalne (komórki zarodkowe) do multipotencjalnych lub unipotencjalnych (komórki somatyczne). Zarodkowe komórki macierzyste, z uwagi na nieograniczone zdolności namnażania i pluripotencjalność wydają się najbardziej optymalne do zastosowania w terapiach regeneracyjnych, lecz zastrzeżenia natury etyczno-moralnej pozyskiwania tych komórek, a także możliwość ich niekontrolowanej proliferacji po przeszczepieniu w organizmie biorcy skłoniły badaczy do poszukiwania alternatywnych źródeł komórek macierzystych, jakimi są tkanki dojrzałego organizmu. Badania ostatnich lat wykazały, iż w dojrzałych tkankach somatycznych znajdują się pluripotencjalne komórki macierzyste, co więcej odkrycia Takahashi i Yamanaka w roku 2006 r udokumentowały możliwość przekształcenia zróżnicowanych komórek izolowanych od osobników dorosłych w komórki pluripotencjalne. Zainicjowanie nowatorskich metod badawczych, których celem będzie opracowanie skutecznych sposobów przeszczepiania somatycznych komórek macierzystych w celach regeneracyjnych jest nadzieją w terapii wielu nieuleczalnych schorzeń.

Cell walls are at the basis of a structural, four-dimensional framework of plant form and growth time. Recent rapid progress of cell wall research has led to the situation where the old, long-lasting juxtaposition: "living" protoplast - "dead" cell wall, had to be dropped. Various attempts of re-interpretation cast, however, some doubts over the very nature of plant cell and the status of the walls within such a cell. Following a comparison of exocellular matrices of plants and animals, their position in relation to cells and organisms is analysed. A multitude of perspectives of the biological organisation of living beings is presented with particular attention paid to the cellular and organismal theories. Basic tenets and resulting corollaries of both theories are compared, and evolutionary and developmental implications are considered. Based on these data, "The Plant Body" - an organismal concept of plants and plant cells is described.

Opis:
Modern internal combustion powertrains are the main source of propulsion for on-road and non-road vehicles. However, they are increasingly being replaced by electric or fuel cell-equipped alternative propulsion systems. The article presents a study of fuel cell characteristics operating under both static and dynamic conditions, with a 1.2 kW fuel cell set with a voltage converter and lead-acid batteries. In the conducted tests, the fuel cell stack's maximum efficiency reached 65%. Load tests (static and dynamic) have indicated higher fuel cell efficiencies when using hybrid operation with a DC/DC converter and battery.

Opis:
Camptothecin (CPT), an alkaloid natural product, extracted from Camptotheca acuminata bark, has been reported to have potential antitumor activity in diverse cancers. MicroRNAs (MiRNAs) are a class of short, non-coding RNAs that plays a crucial role in the normal physiology by attenuating translation. Here, we showed that the CPT modulates the expression of miRNAs in hepatocellular carcinoma cells (HCC). Microarray analysis reveals that CPT modulates the expression of as many as 39 miRNAs in HCC cells (Huh7), 27 miRNAs were downregulated whereas 12 miRNAs were upregulated. miR-16 is the key miRNA upregulated by CPT and targets key prosurvival proteins (MMP-2, MMP-9 and cyclin D1). Our results demonstrate that CPT is inhibiting cell viability of HCC cells significantly when compared with the untreated cells. Wound healing and colony formation assay confirm inhibition of cell migration and clonogenic property of Huh7 cells respectively, upon the dose-dependent treatment of CPT. Furthermore, the Boyden chamber assay analysis revealed a significant inhibition of number of invasive cells in CPT treated cells with comparison to untreated Huh7 cells. Mechanistically, CPT upregulates miR-16 expression which targets MMP-2, MMP-9, cyclin D1 downregulation and subsequently upregulates the expression of E-cadherin, TIMP1, p21, and p27, thereby inhibits cell migration, invasion and clonogenic property of HCC cells. In summary, CPT treatment in Huh7 cells decreases cell viability and upregulates miR-16 expression, which results in inhibition of cell migration, invasion and clonogenic property of cells, by decreasing MMP-2, MMP-9, cyclin D1 and increasing the expression of cell cycle-regulated proteins p21 and p27.

The lack of enteropathy in treated mice was accompanied to affected the size of worms and the number of worms, but did not prevent the retardation of nematodes in the intestine. Fecundity of females in vitro and the number of muscle larvae were not statistically changed after treatment. Ketotifen reduced the number of mast cells and IgA plasma cells in the intestine. As inflammatory influx, epithelial damage and IgA response are related to the action of T cells, ketotifen affected T dependent reactions in the gut of T. spiralis infected mice.

Manipulating particles in the blood pool with noninvasive methods has been of great interest in therapeutic delivery. Recently, it was demonstrated experimentally that red blood cells can be forced to translate and accumulate in an ultrasound field. This acoustic response of the red blood cells has been attributed to sonophores, gas pockets that are formed under the influence of a sound field in the inner-membrane leaflets of biological cells. In this paper, we propose a simpler model: that of the compressible membrane. We derive the spatio-temporal cell dynamics for a spherically symmetric single cell, whilst regarding the cell bilayer membrane as two monolayer Newtonian viscous liquids, separated by a thin gas void. When applying the newly-derived equations to a red blood cell, it is observed that the void inside the bilayer expands to multiples of its original thickness, even at clinically safe acoustic pressure amplitudes. For causing permanent cell rupture during expansion, however, the acoustic pressure amplitudes needed would have to surpass the inertial cavitation threshold by a factor 10. Given the incompressibility of the inner monolayer, the radial oscillations of a cell are governed by the same set of equations as those of a forced antibubble. Evidently, these equations must hold for liposomes under sonication, as well.

Real-time cell electronic sensing (RT-CES) based on impedance measurements is an emerging technology for analyzing the status of cells in vitro. It allows label-free, real time monitoring of the biological status of cells. The present study was designed to assess dynamic data on the cell processes during equine herpesvirus type 1 (EHV-1) infection of ED (equine dermal) cells and primary murine neuronal cell culture. We have demonstrated that the xCELLigence system with dynamic monitoring can be used as a rapid diagnostic tool both to analyze cellular behavior and to investigate the effect of viral infection.

In the course of infections caused by pathogenic yeasts from the genus Candida, the fungal cell surface is the first line of contact with the human host. As the surface-exposed proteins are the key players in these interactions, their identification can significantly contribute to discovering the mechanisms of pathogenesis of two emerging pathogens from this genus, C. parapsilosis and C. tropicalis. Therefore, the aim of the present study was to identify the cell wall-attached proteins of these two species with the use of cell surface shaving and a shotgun proteomic approach. Different morphological forms of C. parapsilosis and C. tropicalis cells obtained after growth under various conditions were subjected to this treatment. This allowed to indicate the most abundant cell surface proteins on the basis of the normalized spectral abundance factors. In case of yeast-like forms these were, among others, proteins similar to a chitinase, glyceraldehyde-3-phosphate dehydrogenase and an inducible acid phosphatase for C. parapsilosis, and a constitutive acid phosphatase, pyruvate decarboxylase and glyceraldehyde-3-phosphate dehydrogenase for C. tropicalis. In case of pseudohyphal forms, proteins similar to a cell surface mannoprotein Mp65, chitinase and glycosylphosphatidylinositol-anchored transglycosylase Crh11 were identified at the cell surface of C. parapsilosis. The Rbt1 cell wall protein, a hyphally regulated cell wall protein and proteins from agglutinin-like sequence protein family were found as the most abundant on C. tropicalis pseudohyphae. Apart from the abovementioned proteins, several additional covalently bound and atypical cell wall proteins were also identified. These results extend the current knowledge regarding the molecular basis of virulence of these two non-albicans Candida species.

Algorithms for examination of the density and selected parameters of movement of sperm cells have been elaborated and implemented. The conducted research is a part of work on a computer system for semen analysis. The system will allow for an increase in the precision of examination thanks to an exact specification of numerical values of the chosen parameters. An additional advantage of the system is an increased time efficiency of examination. Nowadays, the basic type of examination is an estimated analysis of semen parameters by visual observation of a sample, based on a physician's subjective assessment of an image. Registration of images is impossible in visual analysis.

We present a new dual-wavelength quantitative measurement approach that can be employed for simultaneously measuring both the refractive index and the thickness of the homogenous specimen. This method is realized by dual-wavelength in-line phase-shifting digital holography, and then the phase images are obtained by using four-phase step algorithm for each wavelength separately. Based on computer simulation technology, the feasibility and the effectiveness of our proposed method are demonstrated by comparing our simulation results with the experimental results of the spherical silica bead and the red blood cell, respectively. Our work will provide some guidance in the experimental research for transparent phase objects.

Some studies show that cells are able to penetrate through pores that are smaller than cell size. It concerns especially Red Blood Cells but it also may concern different types of biological cells. Such penetration of small pores is a very significant problem in the filtration process, for example in micro or ultrafiltration. Deformability of cells allows them to go through the porous membrane and contaminate permeate. This paper shows how cells can penetrate small cylindrical holes and tries to assess mechanical stress in a cell during this process. A new mathematical approach to this phenomenon was presented, based on assumptions that were made during the microscopic observation of Red Blood Cell aspiration into a small capillary. The computational model concerns Red Blood Cell geometry. The mathematical model allows to obtain geometrical relation as well as mechanical stress relations.

Metal surfaces have become important over the last decade for potential surgical implants, and within this context we present here a study of the cell growth on modified gold surfaces. Gold films, deposited on glass plates and annealed with a hydrogen flame, were modified with four different carboranethiol derivatives: 1-(HS)-1,2-C2B10H11 (A), 1,2-(HS)2-1,2-C2B10H10 (B), 9,12-(HS)2-1,2-C2B10H10 (C) and 1,12-(HS)2-1,12- C2B10H10 (D). The materials engendered from these modifications were used to investigate the adhesion and growth of rat aortic smooth muscle cells cultured on these surfaces in a DMEM medium with 10% of fetal bovine serum. One day after seeding, the highest number of initially adhered cells was found on the surface of a bare gold film. However, three days after seeding, the number of cells on carboranethiol-modified gold samples B, C and D was significantly higher than the number on a bare gold film. After seven days, the number of cells on a bare gold film and on gold films modified with derivatives A, B and D was very similar, but the surface of a gold film modified with derivative C exhibited a significantly smaller number of cells. This may be explained by the exposure of the CH vertices of the carborane cluster, which are more acidic than the BH vertices exposed toward the cells in either A or B.

Opis:
Aluminium foams are a new group of materials used for impact energy absorbing elements. They are light (typically 10-25% of the density of the metal they are made of) and stiff, and are frequently proposed as a light weight structural material. That is why they often are applied in automotive and transport industry solutions, for example as parts of bumpers. The methods of numerical modelling for open and closed cell aluminium foams are presented in the paper as well as closed and open cellfoam microstructure model. The numerical models of foam ideal microstructures created with shell finite elements are shown. The models were developed on the basis of Kefain tetrakaidecahedrons - structures consisting of six squares and eight hexagons. In the case of closed cell foams, the polyhedron with full walls was adopted. In the case of open cell foams the circle wholes were removed from polyhedron surfaces. Then the numerical analysis of a created models compressive test was carried out with the usage of LS Dyna computer code. The nonlinear procedures were applied. The results were analyzed in the scope of energy absorbing properties of aluminium foams.
