Stem cells have captured considerable scientific and clinic interest because of their potential to renew themselves and to differentiate into one or more adult cell types. Thus stem cells have been recognized as a potential tool for the development of innovative therapeutic strategies in different disease disorders. Stem cells can be discriminating based on their differentiated potential as totipotent, pluripotent, multipotent or unipotent cells. There are in general three types of stem cells: embryonic, fetal and adult stem cells. While embryonic stem cell therapy has a lot of ethical concerns due to their obtaining but also unlimited proliferation and uncontrolled differentiation, fetal and adult stem cells have been used in the treatment of different diseases. The bone marrow, peripheral blood and umbilical cord blood are ideal sources of adult stem cells because there are easily accessible and contain two types of stem cells: hematopoietic stem cells giving rise to all blood cell types and mesenchymal stem cells differentiating into cells of mesodermal lineage. This review describes the general characteristics of these stem cell populations and their current applications in regenerative medicine. Additionally induced pluripotent stem cells generated through the reprogramming of differentiated adult cells are described.
Możliwość wspomagania regeneracji nieodwracalnie uszkodzonych tkanek i narządów za pomocą transplantacji komórek macierzystych skupia uwagę środowisk medycznych i opinii publicznej. Komórki macierzyste mają zdolność do samo-odnowy oraz do różnicowania się w dojrzałe komórki struktur całego organizmu. Na określonych etapach rozwoju osobniczego wykazują różny stopień ograniczenia potencjału do dalszego różnicowania, od komórek totipotencjalnych (zygota), poprzez pluripotencjalne (komórki zarodkowe) do multipotencjalnych lub unipotencjalnych (komórki somatyczne). Zarodkowe komórki macierzyste, z uwagi na nieograniczone zdolności namnażania i pluripotencjalność wydają się najbardziej optymalne do zastosowania w terapiach regeneracyjnych, lecz zastrzeżenia natury etyczno-moralnej pozyskiwania tych komórek, a także możliwość ich niekontrolowanej proliferacji po przeszczepieniu w organizmie biorcy skłoniły badaczy do poszukiwania alternatywnych źródeł komórek macierzystych, jakimi są tkanki dojrzałego organizmu. Badania ostatnich lat wykazały, iż w dojrzałych tkankach somatycznych znajdują się pluripotencjalne komórki macierzyste, co więcej odkrycia Takahashi i Yamanaka w roku 2006 r udokumentowały możliwość przekształcenia zróżnicowanych komórek izolowanych od osobników dorosłych w komórki pluripotencjalne. Zainicjowanie nowatorskich metod badawczych, których celem będzie opracowanie skutecznych sposobów przeszczepiania somatycznych komórek macierzystych w celach regeneracyjnych jest nadzieją w terapii wielu nieuleczalnych schorzeń.

Cell walls are at the basis of a structural, four-dimensional framework of plant form and growth time. Recent rapid progress of cell wall research has led to the situation where the old, long-lasting juxtaposition: "living" protoplast - "dead" cell wall, had to be dropped. Various attempts of re-interpretation cast, however, some doubts over the very nature of plant cell and the status of the walls within such a cell. Following a comparison of exocellular matrices of plants and animals, their position in relation to cells and organisms is analysed. A multitude of perspectives of the biological organisation of living beings is presented with particular attention paid to the cellular and organismal theories. Basic tenets and resulting corollaries of both theories are compared, and evolutionary and developmental implications are considered. Based on these data, "The Plant Body" - an organismal concept of plants and plant cells is described.

Opis:
Modern internal combustion powertrains are the main source of propulsion for on-road and non-road vehicles. However, they are increasingly being replaced by electric or fuel cell-equipped alternative propulsion systems. The article presents a study of fuel cell characteristics operating under both static and dynamic conditions, with a 1.2 kW fuel cell set with a voltage converter and lead-acid batteries. In the conducted tests, the fuel cell stack's maximum efficiency reached 65%. Load tests (static and dynamic) have indicated higher fuel cell efficiencies when using hybrid operation with a DC/DC converter and battery.

Opis:
Camptothecin (CPT), an alkaloid natural product, extracted from Camptotheca acuminata bark, has been reported to have potential antitumor activity in diverse cancers. MicroRNAs (MiRNAs) are a class of short, non-coding RNAs that plays a crucial role in the normal physiology by attenuating translation. Here, we showed that the CPT modulates the expression of miRNAs in hepatocellular carcinoma cells (HCC). Microarray analysis reveals that CPT modulates the expression of as many as 39 miRNAs in HCC cells (Huh7), 27 miRNAs were downregulated whereas 12 miRNAs were upregulated. miR-16 is the key miRNA upregulated by CPT and targets key prosurvival proteins (MMP-2, MMP-9 and cyclin D1). Our results demonstrate that CPT is inhibiting cell viability of HCC cells significantly when compared with the untreated cells. Wound healing and colony formation assay confirm inhibition of cell migration and clonogenic property of Huh7 cells respectively, upon the dose-dependent treatment of CPT. Furthermore, the Boyden chamber assay analysis revealed a significant inhibition of number of invasive cells in CPT treated cells with comparison to untreated Huh7 cells. Mechanistically, CPT upregulates miR-16 expression which targets MMP-2, MMP-9, cyclin D1 downregulation and subsequently upregulates the expression of E-cadherin, TIMP1, p21, and p27, thereby inhibits cell migration, invasion and clonogenic property of HCC cells. In summary, CPT treatment in Huh7 cells decreases cell viability and upregulates miR-16 expression, which results in inhibition of cell migration, invasion and clonogenic property of cells, by decreasing MMP-2, MMP-9, cyclin D1 and increasing the expression of cell cycle-regulated proteins p21 and p27.

The lack of enteropathy in treated mice was accompanied to affected the size of worms and the number of worms, but did not prevent the retardation of nematodes in the intestine. Fecundity of females in vitro and the number of muscle larvae were not statistically changed after treatment. Ketotifen reduced the number of mast cells and IgA plasma cells in the intestine. As inflammatory influx, epithelial damage and IgA response are related to the action of T cells, ketotifen affected T dependent reactions in the gut of T. spiralis infected mice.

Manipulating particles in the blood pool with noninvasive methods has been of great interest in therapeutic delivery. Recently, it was demonstrated experimentally that red blood cells can be forced to translate and accumulate in an ultrasound field. This acoustic response of the red blood cells has been attributed to sonophores, gas pockets that are formed under the influence of a sound field in the inner-membrane leaflets of biological cells. In this paper, we propose a simpler model: that of the compressible membrane. We derive the spatio-temporal cell dynamics for a spherically symmetric single cell, whilst regarding the cell bilayer membrane as two monolayer Newtonian viscous liquids, separated by a thin gas void. When applying the newly-derived equations to a red blood cell, it is observed that the void inside the bilayer expands to multiples of its original thickness, even at clinically safe acoustic pressure amplitudes. For causing permanent cell rupture during expansion, however, the acoustic pressure amplitudes needed would have to surpass the inertial cavitation threshold by a factor 10. Given the incompressibility of the inner monolayer, the radial oscillations of a cell are governed by the same set of equations as those of a forced antibubble. Evidently, these equations must hold for liposomes under sonication, as well.

Real-time cell electronic sensing (RT-CES) based on impedance measurements is an emerging technology for analyzing the status of cells in vitro. It allows label-free, real time monitoring of the biological status of cells. The present study was designed to assess dynamic data on the cell processes during equine herpesvirus type 1 (EHV-1) infection of ED (equine dermal) cells and primary murine neuronal cell culture. We have demonstrated that the xCELLigence system with dynamic monitoring can be used as a rapid diagnostic tool both to analyze cellular behavior and to investigate the effect of viral infection.

In the course of infections caused by pathogenic yeasts from the genus Candida, the fungal cell surface is the first line of contact with the human host. As the surface-exposed proteins are the key players in these interactions, their identification can significantly contribute to discovering the mechanisms of pathogenesis of two emerging pathogens from this genus, C. parapsilosis and C. tropicalis. Therefore, the aim of the present study was to identify the cell wall-attached proteins of these two species with the use of cell surface shaving and a shotgun proteomic approach. Different morphological forms of C. parapsilosis and C. tropicalis cells obtained after growth under various conditions were subjected to this treatment. This allowed to indicate the most abundant cell surface proteins on the basis of the normalized spectral abundance factors. In case of yeast-like forms these were, among others, proteins similar to a chitinase, glyceraldehyde-3-phosphate dehydrogenase and an inducible acid phosphatase for C. parapsilosis, and a constitutive acid phosphatase, pyruvate decarboxylase and glyceraldehyde-3-phosphate dehydrogenase for C. tropicalis. In case of pseudohyphal forms, proteins similar to a cell surface mannoprotein Mp65, chitinase and glycosylphosphatidylinositol-anchored transglycosylase Crh11 were identified at the cell surface of C. parapsilosis. The Rbt1 cell wall protein, a hyphally regulated cell wall protein and proteins from agglutinin-like sequence protein family were found as the most abundant on C. tropicalis pseudohyphae. Apart from the abovementioned proteins, several additional covalently bound and atypical cell wall proteins were also identified. These results extend the current knowledge regarding the molecular basis of virulence of these two non-albicans Candida species.

Algorithms for examination of the density and selected parameters of movement of sperm cells have been elaborated and implemented. The conducted research is a part of work on a computer system for semen analysis. The system will allow for an increase in the precision of examination thanks to an exact specification of numerical values of the chosen parameters. An additional advantage of the system is an increased time efficiency of examination. Nowadays, the basic type of examination is an estimated analysis of semen parameters by visual observation of a sample, based on a physician's subjective assessment of an image. Registration of images is impossible in visual analysis.

We present a new dual-wavelength quantitative measurement approach that can be employed for simultaneously measuring both the refractive index and the thickness of the homogenous specimen. This method is realized by dual-wavelength in-line phase-shifting digital holography, and then the phase images are obtained by using four-phase step algorithm for each wavelength separately. Based on computer simulation technology, the feasibility and the effectiveness of our proposed method are demonstrated by comparing our simulation results with the experimental results of the spherical silica bead and the red blood cell, respectively. Our work will provide some guidance in the experimental research for transparent phase objects.

Some studies show that cells are able to penetrate through pores that are smaller than cell size. It concerns especially Red Blood Cells but it also may concern different types of biological cells. Such penetration of small pores is a very significant problem in the filtration process, for example in micro or ultrafiltration. Deformability of cells allows them to go through the porous membrane and contaminate permeate. This paper shows how cells can penetrate small cylindrical holes and tries to assess mechanical stress in a cell during this process. A new mathematical approach to this phenomenon was presented, based on assumptions that were made during the microscopic observation of Red Blood Cell aspiration into a small capillary. The computational model concerns Red Blood Cell geometry. The mathematical model allows to obtain geometrical relation as well as mechanical stress relations.

Metal surfaces have become important over the last decade for potential surgical implants, and within this context we present here a study of the cell growth on modified gold surfaces. Gold films, deposited on glass plates and annealed with a hydrogen flame, were modified with four different carboranethiol derivatives: 1-(HS)-1,2-C2B10H11 (A), 1,2-(HS)2-1,2-C2B10H10 (B), 9,12-(HS)2-1,2-C2B10H10 (C) and 1,12-(HS)2-1,12- C2B10H10 (D). The materials engendered from these modifications were used to investigate the adhesion and growth of rat aortic smooth muscle cells cultured on these surfaces in a DMEM medium with 10% of fetal bovine serum. One day after seeding, the highest number of initially adhered cells was found on the surface of a bare gold film. However, three days after seeding, the number of cells on carboranethiol-modified gold samples B, C and D was significantly higher than the number on a bare gold film. After seven days, the number of cells on a bare gold film and on gold films modified with derivatives A, B and D was very similar, but the surface of a gold film modified with derivative C exhibited a significantly smaller number of cells. This may be explained by the exposure of the CH vertices of the carborane cluster, which are more acidic than the BH vertices exposed toward the cells in either A or B.

Opis:
Aluminium foams are a new group of materials used for impact energy absorbing elements. They are light (typically 10-25% of the density of the metal they are made of) and stiff, and are frequently proposed as a light weight structural material. That is why they often are applied in automotive and transport industry solutions, for example as parts of bumpers. The methods of numerical modelling for open and closed cell aluminium foams are presented in the paper as well as closed and open cellfoam microstructure model. The numerical models of foam ideal microstructures created with shell finite elements are shown. The models were developed on the basis of Kefain tetrakaidecahedrons - structures consisting of six squares and eight hexagons. In the case of closed cell foams, the polyhedron with full walls was adopted. In the case of open cell foams the circle wholes were removed from polyhedron surfaces. Then the numerical analysis of a created models compressive test was carried out with the usage of LS Dyna computer code. The nonlinear procedures were applied. The results were analyzed in the scope of energy absorbing properties of aluminium foams.

In Mexico, Solanum rostratum has been employed for the treatment of several diseases, including uterine cancer. The aim of this study was to evaluate the antioxidant and anti-carcinogenic activity of extracts of Solanum rostratum Dunal on MDA and SiHa cell lines. The methanolic, ethyl acetate, and hexane extracts of the aerial parts of Solanum rostratum were tested for phenols concentration, antioxidant activity, and anti-carcinogenic effect. The leaves extracts showed the highest content of phenols; however the flower-fruits extracts showed higher scavenging activity. On the other hand, ethyl acetate extracts exhibited the highest anti-carcinogenic effect, while methanolic extracts showed the least effect. The results of this work indicated that Solanum rostratum is a promising source of antioxidants, and the extracts exert a dose-dependent anti-carcinogenic effect on both cell lines.

The majority of proteins are unable to translocate into the cell interior. Hence for peptide- and protein-based therapeutics a direct intracytoplasmic delivery with the aid of transducing agents is an attractive approach. We wanted to deliver to the cell interior a putatively cytotoxic protein VPg. Protein transduction was achieved in vitro with three different commercial products. However, in our hands, delivery of various control proteins without known deleterious effects, as well as of protein VPg, always induced cell death. Finally, we used a novel transducing peptide Wr-T, which was not toxic to cultured cells, even in a quite large range of concentrations. Most importantly, control protein delivered to cells in culture did not display any toxicity while VPg protein exerted a strong cytotoxic effect. These data show that results obtained with cell-penetrating agents should be interpreted with caution.

Opis:
Chronic exposure to environmental ultraviolet radiation (UVR) plays a key role both in photocarcinogenesis and induction of accelerated skin ageing. Sailors commonly experience a significant number of cutaneous problems, related to the exposure to environmental factors due to their working conditions. Among these factors, sun exposure is able to determine both acute and chronic skin damage, mostly linked to the effects of the ultraviolet (UV) radiation on epidermal and dermal structures. We report a case of sailor with cutis rhomboidalis nuchae, solar elastosis, actinic keratosis (AK), numerous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) of the head and trunk.

The most important feature of 3G mobile cellular network is introduction of voice data integrated service under multilayered cell environment to support overflow traffic of lower layered cells by upper ones. This paper deals with traffic model of three layered cells, i.e., micro cell, macro cell and satellite cell. Here a new call admission control is introduced for three layered cell of 3G mobile cellular network. State transition chain is designed for theoretical analysis of above mentioned traffic. Blocking probability of data call, new voice call and handover failure of voice call, probability of utilization of micro cell channel, macro cell channel and satellite cell channel are analyzed against different traffic parameters and yield logical results.

The work contains an analysis of the theoretical potential of the electric energy yield for a photovoltaic panel made of polycrystalline silicon. Calculations were based on monthly multiannual average values of solar radiation intensity available in solar conditions of Małopolska Region. For their purposes, the researchers have chosen a source with a power reaching 235 Wp, which is available in the market and may be used in farms located in this region.

Cancer cells are characterized by an aberrant increase in protein N-glycosylation and by disruption of E-cadherin-mediated adherens junctions. However, the relationship between alterations in N-glycosylation process and loss of E-cadherin adhesion in cancer remains unclear. The mechanisms of altered expression of adhesive glycoproteins in cancer cells have not been fully elucidated. Thus, the aim of this study was to examine the expression of E-cadherin and sialyl Lewisa/x, NeuAcα2-3Gal, NeuAcα2-6Gal/GalNAc structures in the normal renal tissue and intermediate and cancerous tissues from patients with clear cell RCC. Moreover, we attempted to correlate the E-cadherin expression with some specific sugar residues of renal cancer tissue glycoproteins. The expression of E-cadherin was analysed using ELISA test and immunoblotting. Oligosaccharide structures and sialylation level were detected with ELISA test using specific biotinylated lectins or antibodies. A significant decrease of E-cadherin expression as well as a significant increase in sialylated oligosaccharides level in intermediate zone and renal cancer tissue in comparison to normal renal tissue are reported. Significant decrease in expression of cadherins and increase in sialylation of oligosaccharide structures in renal cancer tissue in comparison to normal renal tissue, and in renal cancer tissue in comparison to intermediate zone of renal tissue, are important for the future research concerning detection and quantification of cadherins and sialylated oligosaccharide structures in urine and cells of urinary sediment as possible non-invasive marker of early RCC.

Cardiac Radiofrequency (RF) ablation is a commonly used clinical procedure for treating many cardiac arrhythmias. However, the efficacy of RF ablation may be limited by two factors: small ventricular lesions and impedance rise, leading to coagulum formation and desiccation of tissue. In this paper, a high frequency (HF) energy ablation system operating at 27.12 MHz based on an automated load matching system was developed. A HF energy matched probe associated to the automated impedance matching device ensures optimal transfer of the energy to the load. The aim of this study was to evaluate this energy for catheter ablation of the atrioventricular junction. In vivo studies were performed using 10 sheep to characterize the lesions created with the impedance matching system. No cardiac perforation was noted. No thrombus was observed at the catheter tip. Acute lesions ranged from 3 to 45 mm in diameter (mean šSD = 10.3š10) and from 1 to 15 mm in depth (6.7š3.9), exhibiting a close relationship between HF delivered power level and lesion size. Catheter ablation using HF current is feasible and appears effective in producing a stable AV block when applied at the AV junction and large myocardial lesions at ventricular sites.

One of the main parameters that affect the solar cell performance is cell temperature; the solar cell output decreases with the increase of temperature. Therefore, it is important to select the proper solar cell technology that performs better at a specified location considering its average temperatures. In addition, the solar cell performance is directly reflected on the overall economics of the project. This paper is proposed to evaluate the variations in the performance of different solar cell technologies related to the temperature in Amman, Jordan. Field data of weather station and three PV systems (Poly-crystalline, Mono-crystalline and Thin-film) of identical design parameters were collected from Test Field Project at Applied Science Private University, Shafa Badran, Amman, Jordan. These data were analysed in the following way. estimated specific energy yield (kWh/kWp) for the three different PV systems was calculated depending on the measured value of solar irradiance and technical specifications of the installed solar panels and inverters, then the actual energy yield at different temperatures over one year was compared with the estimated value, so the deviations could be determined and actual temperature coefficients for energy yield could be calculated, knowing that the three PV Systems have identical design parameters (tilt angle, azimuth angle, type and dimensions of mounting structure and inverter size) and same cleaning method and schedule. It was found that the thin-film solar panels are less affected by temperature with temperature coefficient of -0.0984%, and -0.109%, -0.124% for Mono-crystalline and Poly-crystalline respectively. These results can be implemented in the preliminary design steps, specifically in the selection of the solar cell technology to be installed in a specific location.

The aim of this contribution is to propose a new averaged model of dynamic problems for thin linear-elastic cylindrical shells having a periodic structure along one direction tangent to the shell midsurface. In contrast with the known homogenized models, the proposed one makes it possible to describe the effect of the periodicity cell size on the global dynamic shell behavior (a length-scale effect). In order to derive governing equations with constant or slowly varying coefficients, the known tolerance averaging procedure is applied. The comparison between the proposed model and the model without the length-scale effect as well as the known length-scale model for cylindrical shells with the periodic structure in both directions tangent to the shell midsurface is present.
Celem pracy jest wyprowadzenie uśrednionego modelu służącego do analizy dynamiki cienkich liniowo-sprężystych powłok walcowych mających periodyczną strukturę w jednym kierunku stycznym do powierzchni środkowej powłoki. Proponowany model, w przeciwieństwie do znanych modeli zhomogenizowanych, umożliwia badanie wpływu wielkości komórki periodyczności na dynamikę powłoki walcowej (wpływ ten zwany jest efektem skali). W celu wyprowadzenia równań o stałych lub wolnozmiennych współczynnikach zastosowano znaną metodę tolerancyjnego uśredniania. Wyprowadzony model porównano z modelem dla powłoki walcowej z periodyką w dwóch kierunkach wzajemnie prostopadłych i stycznych do powierzchni środkowej powłoki oraz z modelem bez efektu skali.

The completely new strategy of pseudophysiological transcomplementary (transcytoplasmic) activation (PP-TCA) of nuclear-transferred oocytes, which had been derived from pWAPhGH-GFPBsd transfected foetal fibroblast cells, was recently applied to the somatic cell cloning of pigs. It resulted in the considerable enhancing not only the cleavage activity of cultured cloned embryos, but also their morula and blastocyst formation rates as compared to the use of standard simultaneous fusion and electrical activation of reconstituted oocytes (77% vs. 57%, 63% vs. 46% and 40% vs. 27%, respectively). Altogether, the use of cytosolic components descended from heterologous (rabbit) zygotes as the agents for stimulation of porcine clonal cytoplasmic hybrids (cybrids) turned out to be reliable and feasible strategy for the generation of transgenic blastocysts by somatic cell nuclear transfer (SCNT). Furthermore, to our knowledge, no previous study has reported the preimplantation developmental outcome of transgenic nuclear-transferred pig embryos following the PP-TCA that was developed and optimised in our laboratory.

The protozoan Toxoplasma gondii, described by Nicolle and Manceaux in 1908, is a ubiquitous and cosmopolitan parasite that infects a wide range of mammal and bird species with high prevalence. The biological success of T. gondii is associated with the formation of a specific relationship between the parasite and host cells leading to the establishment of a latent, chronic infection. During primary infection, acquired mostly by the oral route, the quickly multiplying tachyzoites disseminate through the body crossing several structural-functional barriers as bloodbrain or blood-retina, then they transform into dormant bradyzoites which, enclosed in tissue cysts, occupy preferentially the brain, skeletal muscle and eye. Although T. gondii is able to infect all kinds of nucleated cells, it uses strictly defined host cells, dependent on the life-cycle phase and infection stage. The article discusses selected aspects of the parasite passing via the host body barriers as well as particular role of dendritic cells and skeletal muscle cells, used by the parasite as an very effective vehicle to disseminate throughout the host body or the site of long-term T. gondii persistence, respectively.

Opis:
During lateral root (LR) development a coordinate sequence of cell divisions, accompanied by a change of the organ form takes place. Both the order of anatomical events and morphological features may vary for individual primordia. At early stages of LR primordia development oblique division walls are inserted in cells that are symmetrically located on both sides of the axis of the developing LR primordium, and thereby allow for the protrusion of the LR. We hypothesize that both oblique cell wall insertion and continuous changes in primordium form could be a consequence of a local change in stress distribution in the region of the LR initiation.

Opis:
Monitoring of cellular viability is a key part of toxicological assays in vitro. On-line monitoring of metabolic activity would be particularly useful for evaluation of responses to potential therapeutic compounds. Current assays are mostly based on fluorescent dyes and optical detection methods. These methods offer high sensitivity and specificity, however are not suitable for long-term on-line observations. Electrochemical methods can be an alternative for current protocols. Electrochemical detection is low cost and label-free, therefore suitable for long-term cell culture monitoring. In this work investigations on human cancer cells viability will be presented. Cells were cultured as two-dimensional monolayer or three-dimensional spheroids. Different cell culture media were examined. Potentiometric detection was used for continuous monitoring of cell culture as well as end-point investigations. Different growth phases were identified using applied method. Finally, response to an anticancer drug was successfully observed.

In this short communication we consider the extensibility properties of the cell wall. This is accomplished by a heuristically motivated equation for the expanding volume of the cell. The experimentally determined characteristic time t0 and temperature T0 are the only numbers required for evaluating the effective yielding coefficient Ф(t, T) in the respective time and temperature domains.

The paper describes the immunological response in the matter of percentage of T cells (receptor CD5+) and subpopulations (Th with receptor CD4+, Tc/Ts with receptor CD8+, T with receptor CD25+) and B cells with receptor CD19+, as well as the percentage of apoptotic granulocytes and lymphocytes, in rabbits experimentally infected with the Hagenow strain of the RHD virus. The material chosen for the experiment is special, as among all strains of RHD virus, there are only two strains which carry the variable haemagglutination capacity of red cells. The results of the study show that the Hagenow strain gives an untypical picture of T and B lymphocytes, whereas the results in inducing apoptosis seems to corespond with previous data, confirming the inclusion of apoptosis from 4 h p.i. and the intensity of the phenomenon being higher in granulocytes.

The aim of this contribution is to propose a new averaged nonasymptotic model of stationary stability problems for thin linear-elastic cylindrical shells reinforced by stiffeners which are periodically, densely spaced along one direction tangent to the shell midsurface. As a tool of modelling we shall apply the tolerance averaging technique. The resulting equations have constant coefficients in the periodicity direction. Moreover, in contrast with models obtained by the asymptotic homogenization technique, the proposed one makes it possible to describe the effect of the periodicity cell size on the global shell stability (a length-scale effect). It will be shown that this effect plays an important role in the shell stability analysis and cannot be neglected.
W pracy zaproponowano nowy uśredniony nieasymptotyczny model służący do analizy stateczności cienkich liniowo-sprężystych powłok walcowych, periodycznie, gęsto użebrowanych w jednym kierunku stycznym do powierzchni środkowej. Przy wyprowadzaniu równań modelu wykorzystano znaną metodę tolerancyjnego uśredniania, zaproponowaną przez Woźniaka i Wierzbickiego (2000). Zastosowanie tej metody do znanych równań teorii powłok Kirchhoffa-Love'a doprowadziło do modelu reprezentowanego przez równania różniczkowe cząstkowe o stałych współczynnikach w kierunku periodyczności, zależnych od długościokresu periodyczności. Oznacza to, że proponowany model, w przeciwieństwie do znanych modeli zhomogenizowanych, umożliwia badanie wpływu wielkości komórki periodyczności na wartości sił krytycznych w powłoce walcowej (wpływ ten zwany jest efektem skali). Wyprowadzony model porównano z modelem bez efektu skali i pokazano, że wpływ długości okresu periodyczności odgrywa znaczącą rolę w zgadnieniach stateczności periodycznie, gęsto użebrowanych powłok walcowych.

Squamous cell carcinoma of the pinna occurs primarily in older males who are exposed to sunlight for a prolonged period. Most squamous cell carcinomas of the pinna are well differentiated and present as a painless mass or a sessile nodule. We describe an interesting case of an adult woman with a non-healing wound located on the right pinna which was treated conservatively, and was later diagnosed as squamous cell carcinoma.
Rak kolczystokomórkowy skóry zlokalizowany w obrębie małżowiny usznej występuje głównie u mężczyzn w starszym wieku, którzy są narażeni na długotrwałe działanie promieni słonecznych. W większości przypadków nowotwór ten jest dobrze zróżnicowany i ma postać niebolesnej masy lub nieuszypułowanego guzka. W pracy opisujemy ciekawy przypadek dorosłej kobiety z niegojącą się raną prawej małżowiny usznej – pacjentkę leczono zachowawczo, zanim ustalono rozpoznanie raka kolczystokomórkowego.

An extensively used fluoroquinolone antibiotic ciprofloxacin shows a broad-spectrum antibacterial activity against both gram-positive and gram-negative strains. It works mainly by the inhibition of DNA gyrase and topoisomerase IV which results in impaired DNA replication leading towards microbial cell death. Eryptosis is an alternative term used for suicidal erythrocyte death. In current study, eryptotic effect of ciprofloxacin was investigated by exposing erythrocytes for 48 hours to the different concentrations (45-90µM) of ciprofloxacin. The experimental work related to the investigation of eryptosis was done by cell size measurement and confirmation of calcium role in membrane blebbing. As a possible mechanism of eryptosis, oxidative stress induced by ciprofloxacin was determined by catalase, glutathion peroxidase and superoxide dismutase activities measurement. Similarly, necrotic effect of ciprofloxacin was also illustrated by hemolysis measurement. Results of our study revealed that the therapeutical doses of ciprofloxacin may induce oxidative stress by significant decrease in superoxide dismutase, catalase and glutathione peroxidase activities as well as induce eryptosis, featured by erythrocytes membrane blebbing and hemolysis.

Expression as well as properties of integrins are altered upon transformation. Cell adhesion regulated by integrins is modulated by glycosylation, one of the most frequent biochemical alteration associated with tumorogenesis. Characterisation of carbohydrate moieties of α3β1 integrin on the cultured human bladder carcinoma (T-24, Hu456, HCV 29T) and human normal ureter and bladder epithelium (HCV 29, Hu609) cell lines was carried out after an electrophoresis and blotting, followed by immunochemical identification of α3 and β1 integrin chains and analysis of their carbohydrates moieties using highly specific digoxigenin-labelled lectins. In all the studied cell lines α3β1 integrin was glycosylated although in general each subunit differently. Basic structures recognized in β1 subunit were tri- or tetraantennary complex type glycans in some cases sialylated (T-24, HCV 29, HCV 29T) and fucosylated (Hu609, HCV 29T). Positive reaction with Phaseolus vulgaris agglutinin and Datura stramonium agglutinin suggesting the presence of β1-6 branched N-linked oligosaccharides was found in cancerous cell lines (T-24, Hu456) as well as in normal bladder epithelium cells (Hu609). High mannose type glycan was found only in β1 subunit from Hu456 transitional cell cancer line. On the other hand α3 subunit was much less glycosylated except the invasive cancer cell line T-24 where high mannose as well as sialylated tri- or tetraantennary complex type glycans were detected. This observation suggests that changes in glycosylation profile attributed to invasive phenotype are rather associated with α3 not β1 subunit.

Obtaining a suitable experimental cellular model is a major problem for neuroscience studies. Neuroblastoma cell lines have been often applied in studies related to pathological disorders of nervous system. However, in the search for an ideal model, these cells must be differentiated to cancel their tumor character. The subsequent reactions that are caused by differentiation are not always indifferent to the same model. We evaluated the effect of two well known substances, used for SH-N-SK cell line differentiation, retinoic acid (RA) and phorbol-12-myristate-13-acetate (PMA), on the induction of pro-inflammatory and pro-oxidative reactions in these cells. Cells differentiated with PMA were able to produce significantly higher amounts of pro-inflammatory cytokines whereas the release of nitric oxide radicals was similar to that in undifferentiated cells. On the contrary, in RA-differentiated cells no significant changes in cytokine production were observed and the nitric oxide release was decreased. Additionally, the RA-differentiated neuronal model was more sensible to lipopolysaccharide stimulation, producing pro-inflammatory cytokines abundantly. These results suggest that RA-differentiated SH-N-SK cells provide a more suitable experimental model for the study of molecular and cellular mechanisms of the inflammation and oxidative stress in neuronal cells.
