Criteria used for evaluating drought tolerance of wheat cultivars demanding more time and efforts are usually not efficient and conclusive. Present study was conducted to evaluate ABA sensitivity as a criterion for evaluating drought tolerance of wheat cultivars at an early stage. Ten cultivars of wheat were subjected to drought at 3-leaf stage to select the most sensitive and two tolerant cultivars using mortality rate. Tatara was found the most susceptible whereas GA-2002 and Chakwal-50 were the cultivars with maximum drought tolerance. These cultivars were used to study sensitivity to applied absicic acid (based on germination index and relative growth inhibition rate) and physiological responses (leaf water content, chlorophyll stability index, coefficient of relative inhibition and proline accumulation) under drought stress. Highest ABAsensitivity was recorded in GA-2002 and Chakwal-50 whereas Tatara manifested minimum sensitivity. ABAsensitivity corresponded to physiological indices of drought tolerance. Results show that ABA-sensitivity is an efficient criterion that can be used to evaluate drought tolerance of wheat cultivars at early stage.

The study aimed at demonstrating intracellular proline synthesis under osmotic stress conditions in selected soil fungi on the example of Trichoderma sp. and Trichotecium roseum. Effect of the increase of sodium chloride (NaCl) salinity, in concentrations of 1 to 1000 mmol dm–3 PDA medium, on proline content in the fresh matter of mycelium cultured on medium was examined. The increase of medium salinity affects production of osmoregulatory substances in the form of proline in selected soil fungi. Its content in mycelium depends on salt (NaCl) concentration in medium as well as on species. Intracellular proline synthesis increased starting with the least salinity. As osmotic stress increased, proline content in mycelium almost quadrupled in Trichoderma sp. and quintupled in Trichotecium roseum.

Opis:
The effects of fluridone on the content and composition of fatty acids and some other properties of tomato fruits during a 14-day period of ripening were studied. Mature green tomato fruits were used for the experiments. Fluridone at a 1.0% concentration in lanolin paste was applied as a 2-3 mm stripe from the top to the base of a tomato fruit, and a stripe of lanolin was applied in the same way on the opposite side of the frui as control. After 14 days of the experiment, the treated (yellow) and untreated (red) halves of the tomato were separately freeze-dried and powdered before analysis. Determination of the profile and content of fatty acids was carried out in a micro-HPLC-MS/MS system including a 5600 QTOF mass spectrometer. Fatty acids were identified, based on retention times of available standards and the MS/MS spectra. The degree of lipid peroxidation, DPPH radical scavenging activity, proline and soluble protein content were determined using spectrophotometric methods. The use of fluridone did not cause significant changes in the content of unbound fatty acids, although there was a tendency to increased amounts of unsaturated acids. Concerning total fatty acids, the use of fluridone significantly increased the ratio of oleic acid to stearic, as well as the ratio of total unsaturated acids to saturated acids in the treated pericarp of tomato fruits. Fluridone had not effect on the content of proline, total phenolic acids, MDA and antioxidant activity, but inhibited soluble protein accumulation and enhanced the content of total flavonoids.

Soil salinity is an important abiotic stress factor that reduces both plant growth and development and decreases diversity and metabolic potential of microorganisms, particularly fungi. However, some fun- gal isolates, including mycorrhizal fungi, can increase tolerance of plants to salinity, e.g. by activation of de- fense mechanisms. Main aim of our study was: (i) to identify and characterize fungal strains isolated from fruitbodies growing at saline test sites and (ii) to determine the effect of fungal inoculation on the growth and salinity tolerance of Alnus glutinosa Gaertn. seedlings. Two ectomycorrhizal strains (Amanita muscaria OW-10 and Paxillus involutus OW-5) and a saprotrophic one (Gymnopus sp. OW-6) were isolated from fruit- bodies collected at naturally saline test sites. The plant growth promotion was observed after inoculation of plants with the ectomycorrhizal fungus – P. involutus OW-5, while the use of Gymnopus sp. OW-6, displaying the highest hydrolytic activity, decreased seedling growth parameters. Moreover, P. involutus OW-5 (char- acterized by characterized by medium salt tolerance salt tolerance) promoted growth of black alder in the presence of salt, while A. muscaria OW-10 (with the highest salt tolerance) increased fresh and dry mass of seedlings only in the absence of salt. Black alder seedlings inoculated with P. involutus OW-5 revealed higher synthesis of proline in aboveground organs compared to the control plants which suggested that accumulation of this compound can be a mechanism protecting black alder seedlings against salt stress. In conclusion, we recommend the use of P. involutus OW-5, which displayed the best plant growth promoting ability and salt tolerance, for inoculation of black alder trees used in reclamation of saline habitats

Besides their structural catalytic and diverse regulatory functions, proteins are also precursors of many important biological compounds essential for normal functioning of humans. Many of these compounds may be used as markers for identification of specific pathological states. A comprehensive knowledge about the metabolism of salivary proteins and the mechanisms of action of their metabolites allowed the development of effective treatment for many disorders. However, it should not be forgotten that in some pathological conditions, these compounds not only could be involved in the pathogenesis but also could be used as tool in the prediction of many diseases. This paper is a review of the published literature on selected salivary proteins in the context of the physiological processes of the human body and chosen chronic disorders, such as diabetes, diabetic nephropathy, mucositis, oral mycoses and caries.

Opis:
In recent years there has been a dynamic development of asymmetric synthesis. Groups of researchers, particularly the one led by Benjamin List and Carlos Barbas, carried out a number of reactions and showed the effectiveness of the use of small organic molecules such as proline as catalysts. Michael addition catalyzed with proline is a particularly interesting reaction because it can be carried out in two aminocatalytic pathways. The analysis of Michael reaction reveals potential for both forms of aminocatalysis: enamine and iminium catalysis (Scheme 1) [1–14]. Presumably Michael reaction proceeds mainly according to enamine mechanism. The use of proline in Michael reaction with imine activated acceptor is slightly effective. So far the researches have shown that the modification of proline molecule or addition of other catalyst is necessary for condensation to appear. Enamine catalysis concerns the activation of carbonyl compound in situ being a donor. There is no need for enolase anion to be created earlier [2, 15–17]. When, as a result of the reaction of a,b-unsaturated carbonyl compound with proline, Michael acceptor activation appears it means that it is enamine mechanism reaction (Scheme 1) [2, 24]. One of the first examples of direct Michael reaction proceeding through enamine transition state is the reaction of cyclopentanone with nitrostyrene (Scheme 6) [20–23]. Other examples of Michael addition of ketone with nitro olefin catalysed by proline are shown in table 2 and 3 [10, 23, 30]. Nitroketones obtained in that way are useful as precursors for different organic compounds [33], also pyrrolidines [34]. Pyrrolidines are pharmacologically active and they selectively block presynaptic dopamine receptors [34] (Scheme 7). Except for Michael intermolecular reaction, intramolecular condensation adducts were also obtained. Michael intramolecular proline-catalyzed condensation in which inactive ketones transform into α,β-unsaturated carbonyl compounds was described (Scheme 9) [35, 36]. These reactions require a stoichiometric amount of a catalyst and a long time of reaction and they give as a result a little enantiomeric excess [11, 24, 35]. In 1991, Yamaguchi and co-workers carried out malonates Michael addition to α, β-unsaturated aldehydes catalyzed by L-proline [24, 39]. The reaction proceeded according to enamine mechanism, for example dimethyl malonate was reacted with hex- 2-enal in the presence of proline to give Michael adduct in 44% yield. To improve the yield an attempt of a slight modification of a proline molecule was made transforming it into proper salt. Proline lithium salt enabled to obtain the condensation product in 93% yield (Tab. 4). Regardless of a used catalyst the products in the form of racemates were obtained. In order to improve enantioselective properties of a catalyst, Michael addition of diisopropyl malonate to cycloheptenone was carried out in chloroform in the presence of different proline salts. Optimal enantioselectivity and yield was obtained by using rubidium salt (Tab. 5–7) [40, 41]. Rubidium prolinate-catalyzed Michael additions are used in industry e.g. for enantioselective synthesis of the selective serotonine reuptake inhibitior (SSRI) (–)-paroxetine (antidepressant) (Scheme 12) [24].

Opis:
Mannich reaction occuring among ketone, aldehyde, and amine is one of the ways of a synthesis of biologically active compounds. Reactions of this type were carried out in the presence of different catalysts [3–10], however in recent years a lot of attention has been paid to enantioselective Mannich reaction catalyzed with proline. Such reactions were carried out with the use of different compounds containing carbonyl group and the most frequently used amine was p-anisidine. The advantage of the use of p-anisidine is a possibility of conducting the direct Mannich reaction (Scheme 3). In this way β-amino ketones (Tab. 1, 2, 4) [15, 18–20, 23, 24], α-hydroxy-β-amino ketones (Tab. 3) [15, 22], and β-amino alcohols (Tab. 5, 6) [25, 26] were obtained. A possibility of syntheses of β-amino sugars and α-amino acids with their derivatives (Tab. 7) [28, 29] is worth noticing. In a great number of described reactions, the products were obtained with satisfactory yield and enantiomeric excess. Taking into consideration the difficulty of a removal of p-hydroxyphenyl group which protects amine group in the resulting products, the attempts of using different amine compounds in Mannich reactions catalyzed with proline were undertaken. The use of amines blocked by tert-butoxycarbonyl group (Boc) enabled to obtain the products with high yield and ee values (Tab. 12–15) [35–38]. However in the case of the use of Boc the reaction must be carried out in an indirect way (it is necessary to prepare imine blocked by Boc earlier).

Chickpea is an important food legume cultivated in semiarid regions, where water scarcity and nutrient deficiencies negatively affect crop production. This study aimed to investigate the effect of zinc and silicon from different sources, including bulk and nanostructures, on various biochemical traits of chickpea plants grown under field conditions in Maragheh, Northwest Iran. The main experimental factor consisted of three soil moisture levels: irrigation to 90% of field capacity (FC), 60% FC, and 30% FC. The subplots were assigned for foliar application of different fertilizers: control (distilled water), zinc sulfate (ZnSO), silicon dioxide nanoparticles (SiO2 NPs), ZnSO + SiO2 NPs, and zinc-containing mesoporous silica nanoparticles (MSNPs -Zn). The results showed that although decreased soil moisture had a negative impact on several biochemical processes, foliar application of Zn and Si in both conventional bulk and nanostructure significantly affected plant antioxidant system, plasma membrane integrity, and the concentrations of photosynthetic pigments and compatible solutes. However, the most inducing effects on catalase, ascorbate peroxidase, guaiacol peroxidase, superoxide dismutase, and anthocyanin were observed with the foliar spray of MSNPs-Zn and ZnSO + SiO2 under 60% FC. Moreover, foliar spray of MSNPs -Zn alleviated the negative effects of water deficit stress on photosynthetic pigments (chlorophyll a /b and carotenoid content). Water stress significantly induced the accumulation of free proline in the leaves. Overall, the results indicated that foliar spray of MSNPs -Zn, especially under 60% FC, improved the plant’s defense system, scavenged reactive oxygen species, and enhanced the accumulation and stability of pigments, thereby mitigating the effects of drought stress.

The introduction of new genotypes of crop plants is among the most strategic research programmes, especially in arid and semi-arid regions. To study the effect of drought stress on seed yield and some physiological traits of promising lines of rapeseed at different sowing dates, an experiment was conducted for two years (2015-2017) in a semi-arid region of Iran. In this research, two conventional sowing dates were set in October 12 and November 1 (late sowing). Irrigation was carried out at two levels: normal irrigation (control) and irrigation interruption from the silique formation stage to the next stage (late-season drought stress). The genotypes included four promising lines (L1112, L1091, L1093, L1206), and a cultivar (Okapi) as a control. Results showed that delayed sowing and drought stress increased carbohydrate content and decreased seed yield, with the highest carbohydrate content and highest yield loss in L1112 and the lowest carbohydrate and lowest yield loss in the L1206 line. Among the physiological traits measured, stomatal resistance had the highest degree of correlation and the highest direct negative effect on seed yield, which declined with increasing stomatal resistance. L1112 had the highest stomatal resistance (52.76 s cm-1) in delayed sowing and drought stress conditions. Therefore, L1206 and L1112 were revealed to be resistant and sensitive lines, respectively.

Opis:
Monocultures of Norway spruce widely used for afforestation of drained peat soils often have low productivity and decline in vitality. The research aim was to elucidate: (1) imbalances in the nutrient status of soil and plants in relation to Norway spruce crown status and annual increment on drained peat soil in the sub-boreal zone; (2) the suitability of using proline accumulation as a biomarker for Norway spruce vitality and stress in nutrient imbalance conditions. The study was conducted at five forest (Oxalidosa turf. mel.) sites in Latvia, each containing trees with different crown condition status. Chemical analyses of soil and spruce needles, assessment of tree crown vitality and annual increment measurements were done. Our results revealed serious disturbances in the supply of nutrients, which is an important factor in the decline of Norway spruce in monoculture stands on drained peat soils. Deficiency of K, Fe, Cu, B, N and P was found in the current year needles for all trees, but in damaged trees (≥61% defoliation, ≥61% discoloration) Zn deficiency was also found. For 2-year-old needles, deficiency of K, P, S, Zn, and Cu was found in all trees, additionally, deficiency of Fe was found in healthy and medium damaged trees (26–60% defoliation, 21–60% discoloration), and N deficiency was found for medium damaged and damaged trees. Thus, K, Cu, B, Fe concentrations had significant negative correlations with crown status parameters – defoliation, discoloration, and density. K and B deficiency could play the most significant causal role in decreased stem increment. The results revealed that the stress amino-acid proline is a reliable biomarker, having a significant relationship with spruce crown condition parameters, annual increment and nutrient status in needles – low levels of K, Fe, S, Cu, B, N. The increase in proline content in spruce needles was pronounced in moderately damaged trees, indicating the potential for the use of proline accumulation as early stress indicator for spruce. Therefore, further studies on the identification of early stress and factors affecting nutrient uptake and accumulation in needles are particularly valuable for evaluation of stand management options before significant decline of spruce stands.

Opis:
Accumulation of proline is a defense mechanism against external stress conditions, preventing damage to the structure and function of cells and improving plant development processes, such as germination. The purpose of this study was to investigate proline treatment as a means of improving the germination and development of Norway spruce seedlings. The effect of exogenous proline has been studied in three stages of initial plant development. The collected seeds were soaked in water or 8 mM proline solution and placed on the germinators. The germination capacity and the mean germination time were determined. Seedlings with radicles >10 mm were transferred to the sand-peat substrate at a constant temperature of 20 °C. Seedlings at 3 subsequent developmental stages (S1 – germinated seeds with radicles > 3 mm; S2 – seedlings with radicles >10 mm; S3 – established seedlings grown for 90 days) were examined for the oxygen consumption rate, total antioxidant capacity, hydrogen peroxide level, malondialdehyde level and intracellular proline content. Proline treatment was conducive to lowering the levels of hydrogen peroxide and malondialdehyde at stage S1. At the subsequent stages of development, the levels of hydrogen peroxide and malondialdehyde increased, and at the S3 stage, there was also a marked increase in total antioxidant capacity. At stage S3, the seedlings of the proline treatment were characterized by a lower total mass, and the response to exogenous proline was stronger in the root tissues than in the leaves. The oxygen consumption rate was higher for the proline treatment at all stages of development. Seedlings at the analyzed stages of establishment differed in response to proline treatment. Exogenous proline had some beneficial effects during the first phase of germination by reducing the level of hydrogen peroxide and improving the condition of lipid membranes. In the subsequent stages of seedling development, in response to the same concentration of proline solution, undesirable effects, such as an increase in hydrogen peroxide levels and damage to cytoplasmic membranes, were observed. Optimal concentrations of exogenous proline should be determined prior to commercial use of proline treatment to improve plant stress tolerance.

Opis:
Seasonal changes in the levels of water-soluble carbohydrates, free proline and tolerance to freezing and water deficit were studied in leaves of cherry laurel (Prunus laurocerasus) and ivy (Hedera helix). Frost and water deficit tolerance was estimated based on the measurement of electrical conductivity of aqueous media containing leaf discs that were previously treated with frost (–7°C) or polyethylene glycol solution (PEG 600), respectively. Carbohydrate content in leaves of examined species changed differently throughout the measurement period. In laurel leaves the highest carbohydrate level was found in February, while it was lowest in June. In contrast, in ivy leaves the highest carbohydrate content was recorded during summer (June, July), while the lowest in February. However, a lack of correlation between soluble carbohydrate level and membrane injury index was shown in this study. Free proline content in leaves of investigated species was the highest in early spring, i.e. April. The lowest level of free proline in laurel leaves was found in July, September and October, but in ivy from July to February. A negative correlation between proline content and membrane injury index observed in frost and PEG treated leaves of both species suggest that this amino acid may have resulted on membrane protection.

The LlPRP2 gene coding for a proline-rich protein shows a high level of similarity to, as well as significant differences from the family of ENOD2 nodule-specific genes. Several sequence motifs with putative regulatory function were identified in the 5' and 3' noncoding regions of the LlPRP2 gene. Northern blot analysis revealed that the expression of the LlPRP2 gene begins 9 days after inoculation of yellow lupin roots with Bradyrhizobium sp. (Lupinus); the expression is restricted to symbiotic nodules and is not detected in other tissues or organs. Detailed hybridization analysis showed that, when expression is activated, the LlPRP2 transcript is modified so as to produce at least three bands and a continuous distribution of decay intermediates. The modification of the LlPRP2 transcript probably involves degradation from the 5'- and/or 3'-ends of the RNA molecules. Southern blot analysis indicates that only one gene is present in the yellow lupin genome. The presence of genes homologous to the LlPRP2 gene was confirmed for three cultivars of yellow lupin and for Lupinus angustifolius. However, LlPRP2 homologues were not detected in Lupinus albus cv. Bac, indicating that this plant may lack the ENOD2 sequence.

Androgenic embryos of 3 different carrot genotypes were cultivated on the medium containing different Cu2+ concentrations: 0.1 uM (control), l uM, 10 uM and 100 uM. Carrot sensitivity to Cu2+was evaluated on the basis of growth inhibition, changes in peroxidation of membranes and proline content. The cultivation on the medium supplemented with Cu2+ resulted in dose-dependent inhibition of the growth and organogenic ability of Narbonne carrot embryos, while in the genotype 1014 treated with 10 uM Cu2+ the significant regeneration ability was obseryed. The same Cu2+ concentration greatly increased the level of free proline in the genotype 1014 which was accompanied by relatively low TBARS content. In the genotype Feria 100 uM Cu2+ concentration triggered large increase in proline content associated with high regeneration capacity of carrot embryos.

Opis:
Plant breeders have focused much attention on polyploid trees because of their resistance for forestry. To evaluate the impact of intraspecies genome duplication on NaHCO3 stress, a series of Betula platyphylla autotetraploids and diploids were generated from the same family. The growth, proline content and proline-associated gene expression of these autotetraploid individuals were compared with those diploid trees. Autotetraploids were superior in injury index and relative growth of height and base diameter compared to diploids. The proline content was higher in autotetraploid individuals compared to diploids. Gene expression data revealed autotetraploids were generally higher expression in BpP5CS1, BpP5CS2, Bp- P5CR1, BpP5CR2, BpP5CR3 and BpOAT and were lower expression in BpProDH and BpP5CDH compared to diploid trees. These results shed light on resistance variation in birch autotetraploidization and polyploidy breeding as a new approach for genetic improvement of birch trees.

Opis:
Proline in organic synthesis is used as a small molecular organocatalyst. In a catalytic act proline, similarly to an enzyme, activates reagents, stabilizes transition state and influences an orientation of substrates [1–12]. Proline works as aldolase I (so called microaldolase I). In comparison with other amino acids it shows exceptional nucleophilicity which makes imines and enamines formation easier. In the intermolecular aldol reaction proline was used for the first time by List and co-workers (Scheme 1) [3, 9, 20]. Since then an immense progress has been observed in this field. Several aldolization reactions were performed in the presence of proline. Reactions of this type proceed between the donor (nucleophile) and the acceptor (electrophile). In aldol reaction the donors can be both ketones and aldehydes which next are condensed with ketones and aldehydes acting as electrophiles (Scheme 2–18; Tab. 1–7) [21–72]. The presence of proline ensures not only high yield of homo- and heteroaldolization but mainly enables conducting enantio- and diastereoselective synthesis. Intermolecular proline-catalyzed aldol condensation proceeds according to enamine mechanism. Anti-aldols, which make a valuable source of intermediates in the synthesis of important biologically active compounds, are mainly obtained in this reaction [35–44, 54, 58, 62, 63, 68, 69, 71].

Opis:
The present study deals with biochemical and physiological methods for assessment of the optimal nutrient supply for the growth and development of garden rocket (Eruca sativa Mill.), edible vegetable. Two nitrate (0.3; 0.6 g N dm⁻³ of medium) and three potassium doses (0.3; 0.6; 0.9 g K dm⁻³ of medium) in the form of sulphate or chloride were examined. At the higher nitrate dose and the sulphate form of potassium the intense green colour of leaves, higher content of nitrates, flavonoids, L-ascorbic acid and lower carbohydrates content correlated with elevated growth parameters, e.g. the number of leaves and partially plant fresh weight. The proline and anthocyanin contents weakly diversified the nutrient supply. Despite the lack of modification in the photosynthetic pigment concentration, the chlorophyll fluorescence parameters were significantly improved when the higher nitrate dose accompanied the sulphate form of potassium (higher values of fluorescence decrease, maximum quantum efficiency of PSII photochemistry in the dark-adapted state, photochemical quenching and lower values of the fraction of absorbed light energy not used for photochemistry). The biochemical and photosynthetic parameters corresponding to the morphological characteristics (leaf colour, number of leaves and plant fresh weight) indicated that better nutrient conditions were provided to plants under the combined fertilization of the higher nitrate dose and the sulphate form of potassium.
W badaniu zastosowano metody biochemiczne i fizjologiczne w celu optymalizacji zapotrzebowania na składniki odżywcze sprzyjające wzrostowi i rozwojowi rokietty siewnej (Eruca sativa Mill.). Zastosowano dwie dawki azotu (0,3 i 0,6 g N dm⁻³) i trzy dawki potasu (0,3; 0,6 i 0,9 g K dm⁻³) w formie siarczanu i chlorku. Po zastosowaniu wyższej dawki azotu i siarczanowej formy potasu intensywny zielony kolor liści, wyższa zawartość azotanów, flawonoidów, kwasu askorbinowego i niższa zawartość węglowodanów odpowiadały podwyższonym wartościom parametrów wzrostu, tj. większej liczbie liści i częściowo wyższemu plonowi świeżej masy liści. Zawartości proliny i antocyjanów słabo różnicowały zapotrzebowanie rośliny na składniki odżywcze. Po zastosowaniu wyższej dawki azotu i siarczanowej formy potasu parametry fluorescencji chlorofilu poprawiły się (wyższe wartości wskaźnika witalności PSII, maksymalnej fotochemicznej wydajności PSII, wygaszania fotochemicznego i niższe wartości wskaźnika energii świetlnej pochłoniętej, a nie zużytej w procesach fotochemicznych) przy jednoczesnym braku wpływu na zawartość barwników fotosyntetycznych. Zmierzone parametry biochemiczne i fotosyntetyczne odpowiadały charakterystyce morfologicznej (kolor liści, liczba liści i świeża masa liści) wskazując, że lepsze warunki żywieniowe były dostarczone roślinom w warunkach nawożenia wyższą dawką azotu i siarczanową formą potasu.

Opis:
In asymmetric synthesis of organic compounds more effective solutions are being looked for which will result in higher yield(s) of product(s) and their high enantioselectivity [1]. One of such solutions is an use of a multilevel and cheap catalyst. Proline used as a catalyst is a substance of natural origin which was synthetically obtained by Willstätter who was carrying out research on hygric acid (Scheme 1) [10]. The cells of many organisms have a suitable enzymatic system essential for proline biosynthesis [15]. So far, three proline biosynthesis pathways have been described: from glutamate (Scheme 3 and 4), ornithine (Scheme 5 and 6), and arginine (Scheme 7) [16–28]. Proline which is obtained as a result of biosynthesis or supplementation is a substrate for many proteins. Characteristic and significant content (about 23%) of this amino acid was observed in collage. In cells proline can play an important role of osmoregulator [31–35] – a protective substance regulating the activity of such enzymes as catalase and peroxidase [36]. Proline as a secondary amine shows exceptional nucleophilicity facilitating imine and enamine formation. Used as a catalyst in aldol reaction makes with substrates like imine or enamine transition state imitating the activity of naturally occurring enzymes for this type of reaction, that is aldolases. In their research Hajos and Parrish, and Eder, Sauer and Wiechert used proline in intramolecular aldol reaction obtaining proper enones (Scheme 9) [60–62]. The process of intramolecular aldol reaction was used for a separation of racemic mixture of diketones (Scheme 10) [63, 64], cyclization of ortho-substituted aromatic aldehydes and ketones (Scheme 11) [65], synthesis of cyclic diketones (Scheme 13) [68] and domino reaction to obtain substituted cyclohexanones from beta-diketones and unsaturated ketones (Scheme 14) [69].

Opis:
Salinization of soils or waters is one of the world’s most serious environmental problems in agriculture. It is necessary to determine the environmental factors under which medicinal and aromatic plants give higher yields and better quality. The problem of salinity is characterized by an excess of inorganic salts and is common in the arid and semi-arid lands, where it has been naturally formed under the prevailing climatic conditions and due to higher rates of evapotranspiration and lack of leaching water. Although more frequent in arid lands, salt-affected soils are also present in areas where salinity is caused by poor quality of irrigation water. Saline soil induces physiological and metabolic disturbances in plants, affecting development, growth, yield, and quality of plants. Plants affects adversely as a result of salinity, seed germination, survival percentage, morphological characteristics, development and yield and its components. In general, salt stress decreases the photosynthesis and respiration rate of plants. Total carbohydrate, fatty acid and protein content were adversely affected due to salinity effect, but increased the level of amino acids, particularly proline. The content of some secondary plant products is significantly higher in plants grown under salt stress than in those cultivated in normal conditions. The salinity tolerance depends on the interaction between salinity and other environmental factors.

Opis:
A hydroponic experiment has been conducted to determine the physiological and biochemical response of four new rye lines – S120, S76, OT1-3 and 541 – bred at the Department of Plant Genetics and Breeding, Agricultural University in Szczecin. Seeds were sown into soil and placed into a phytotrone chamber with controlled atmosphere and photoperiod. The relative humidity was 80%, the temperature maintained at 20oC and the intensity of photosynthetically active radiation (Phar) was 700 μmol⋅m-2⋅s-1. Seed germination occurred after about 5 days. The seeds were characterised by different germination ability: 80% for S120, 100% for S76, 82% in OT1, and just 50% for 541 line. Rye seedlings, at the 2-3 leaf stage, were placed in hydroponic units and a two-factor experiment was set up, with potassium deficiency being the first factor and rye lines the second one. The control variant consisted of seedlings placed on complete Hoagland’s medium, while potassium starvation was performed on a medium with 50% deficiency of this element. After seven days, fresh matter was weighed, proline content was measured according to the BATES method (1973) and amounts assimilation pigments were measured according to the method of ARNON et al. (1956). The results were processed statistically, performing a two-factor analysis of variance, while the significance of factors was tested using Tukey’s test at α = 0.05. The study aimed at examining selected physiological and biochemical indicators of the resistance response of four rye lines to stress induced by potassium deficiency. Application of 50% potassium deficiency in a medium induced an increase in the content of assimilation pigments in leaf fresh matter of S120 rye line. A correlation was found between proline content and that of assimilation pigments. The largest proline quantity, 27.2 μg⋅g-1 f.m., was found in 541 rye line seedlings, but its content decreased together with fresh matter yield. It was also found that the total chlorophyll content was directly proportional to the content of proline in S120, S76 and OT1-3 rye lines, whereas in the case of other pigments such a relationship existed only for S 120 and S76 rye lines.
Celem doświadczenia było zbadanie reakcji fizjologicznej i biochemicznej 4 nowych linii żyta: S120, S76, OT1-3, 541, wyhodowanych w Katedrze Genetyki i Hodowli Roślin Akademii Rolniczej w Szczecinie. Nasiona wysiano do gleby i umieszczono w fitotronie z kontrolowaną atmosferą i fotoperiodem. Wilgotność względna wynosiła 80%, temperatura 20°C, a natężenie promieniowania fotosyntetycznie czynnego Phar – 700 μmol⋅m- 2⋅s-1. Kiełkowanie nasion nastąpiło po ok. 5 dniach. Nasiona charakteryzowały się różną zdolnością kiełkowania: S120 – 80%, S76 – 100% OT1 – 82%, 541 – 50%. W hydroponikach umieszczono siewki żyta w fazie 2-3 liści i założono doświadczenie dwuczynnikowe. Po 7 dniach oznaczono świeżą masę (wagowo), zawartość proliny metodą BATESA (1973), zawartość barwników asymilacyjnych metodą ARNONA i in. (1956).

Opis:
The effect of soil drought on leaf water content, proline content, and the activity of guaiacol (GuPX) and ascorbate (APX) peroxidases as well as the level of lipid peroxidation were investigated in leaves of drought resistant red fescue (Festuca rubra) and drought sensitive perennial ryegrass (Lolium perenne). Plants were grown under glasshouse conditions in soil pot culture. 26 day-old grasses were exposed to drought by withholding irrigation for 18 days. Water content in leaves of perennial ryegrass decreased more than in red fescue throughout the experimental period. On the other hand, proline content (PC) was higher in red fescue. The activity of APX and GuPX increased in leaves of red fescue, while it did not change in perennial ryegrass. Our data demonstrate that both red fescue and perennial ryegrass were able to survive applied drought, as shown by a lack of stress-induced lipid peroxidation and hence no evidence of oxidative damage. We speculate, that the observed drought stress tolerance at cellular level was associated with the ability to accumulate proline, and to maintain high activity of APX and GuPX, resulting in protection against oxidative damage and lipid peroxidation. It seems that this mechanism works better in red fescue.

Opis:
This research was conducted to evaluate the effects of water salinity and nano-silica (NS) and humic acid (HA) on Bermuda grass. The study was carried out under greenhouse and exterior space conditions in a completely randomized design with factorial arrangements. Treatments included 4 water salinity levels (0, 5, 7 and 9 dS/m) and 4 level of NS (0, 1, 2 and 3 mm/l) as well as 4 level of HA (0, 50, 100 and 150 mg/l). Results indicated a decrease in chlorophyll content, and increase in proline, malondialdehyde (MDA), catalase (CAT), superoxide dismutase (SOD) and electrolyte leakage (EL) with increasing levels of salinity. The chlorophyll content in greenhouse and exterior space also increased with increasing levels of NS and HA, while proline in two environments, with increasing levels of NS and HA decreased. EL with increasing levels of NS fluctuated, but with increasing levels of HA, it decreased in two environments. With increasing levels of NS, CAT in both environments was eventually decreased after the oscillation, but there was no particular trend in HA levels. With increasing levels of NS and HA, MDA in the greenhouse decreased and in the exterior space – it increased. Finally, by increasing levels of NS and HA, SOD did not show any change in the greenhouse condition, but in the exterior space, the SOD was decreased.

Opis:
In pot experiments on cucumber cv. Śremski F1, the effect of short-term chilling on plants earlier treated with triacontanol (TRIA) and Asahi SL was investigated. These plants were grown in a phytotron at an air temperature of 27/22ºC (day/night), using fluorescent light with far flux density of 220 μmol x m-2 x s-1, with a photoperiod 16/8. At the 4th true leaf stage, the respective experimental series were sprayed with: 1) H2O – control, 2) TRIA 0.01, 3) TRIA 0.1, 4) TRIA 1.0 mg x dm-3, 5) Asahi SL 0.2, 6) Asahi SL 0.3%. After 24 hours one half of the plants from each experimental series was treated for a period of 3 days at a temperature of 12/6ºC, with all the other growth conditions unchanged. The obtained results have shown that short-term chilling stress caused a significant increase in electrolyte leakage, free proline content and in the activity of guaiacol peroxidase in leaves, but a decrease in chlorophyll a+b content, stomatal conductance, transpiration, photosynthesis, leaf area and in the activity of catalase in leaves. The application of TRIA or ASAHI SL on leaves in the pre-stress period reduced the values of the traits which had been increased as a result of chilling and increased those which had reduced. Generally, TRIA was most effective at a concentration of 0.1 mg x dm-3, and Asahi SL at a concentration of 0.3%.
Uzyskane wyniki wykazały, że okresowy chłód wywołał istotny wzrost stopnia wypływu elektrolitów, zawartości wolnej proliny i aktywności peroksydazy gwajakolowej w liściach, spadek zaś zawartości chlorofilu „a+b”, przewodności szparkowej, transpiracji, fotosyntezy, powierzchni liści i aktywności w nich katalazy.

Opis:
In pot experiments conducted on cucumber cv. Śremski F1, the effect was studied of short-term chilling stress on plants which had grown from seeds germinating in the solution of Asahi SL or treated with this biostimulator during the early growth period. The plants were grown in a phytotron at an air temperature of 27/22ºC (day/night), using fluorescent light with FAR flux density of 220 μmol x m-2 x s-1 and with a photoperiod of 16/8. The biostimulator was applied using the following methods: a) germination of seeds in 0.01% and 0.05% solution, b) watering of plants twice with 0.01% or 0.05% solution, c) spraying leaves with 0.3% or 0.5% solution. Plants sprayed with distilled water were the control. After 24 hours from foliar or root application of Asahi SL, one half of the plants from each experimental series was treated for a period of 3 days at a temperature of 12/6ºC, with all the other growth conditions unchanged. The obtained results show that short-term chilling stress caused a significant increase in electrolyte leakage, free proline content and in the activity of ascorbate peroxidase in leaves, but a decrease in the content of chlorophyll, its maximum fluorescence (Fm) and quantum yield (Fv/Fm), carotenoid content, stomatal conductance, transpiration, photosynthesis, leaf biomass and in the activity of catalase in leaves. Foliar or root application of Asahi SL in the pre-stress period decreased the values of the traits which increased as a result of chilling or increased those which decreased. Higher concentrations of the biostimulator solutions, applied using this method, were more effective. The application of the biostimulator during seed germination did not result in significant changes in the response of plants to chilling stress.

Opis:
This study was conducted to evaluate the effects of higher conductivity of nutrient solution created by nitrate or chloride salts of potassium and calcium on growth characteristics of pepper plants (Capsicum annuum var annuum) during four months of growth period. Two EC5 and EC8 dS/m of Hoagland nutrient solutions were prepared using various salt combinations, namely: KCl + CaCl2, KNO3 + CaNO3, and KNO3 + CaNO3 + NaCl. Hoagland nutrient solution with EC 1.8 dS/m served as a control. Higher conductivity treatments had different effects on pepper plant growth. The most significant reduction in growth parameters of plant height, shoot fresh weight, fruit yield and nutrients uptake was in plants treated with KCl + CaCl2 particularly at EC8. Application of KNO3 + CaNO3 particularly at EC5, showed no difference as compared to the control regarding many growth parameters. Application of KNO3 + CaNO3 at EC5 resulted in higher shoot fresh weight compared to the control. All salinity treatments, except from KNO3 + CaNO3 at EC5, reduced the fruit yield compared to the control. Treatments of KCl + CaCl2 and KNO3 + CaNO3 + NaCl particularly at EC8 of nutrient solution, resulted in higher leaf proline concentration, catalase and peroxidase enzymes activity compared to the control. Other conductivity treatments showed no difference in catalase or peroxidase enzymes activities. Significantly the lowest amount of leaf N, K, Mg and Ca was in KCl + CaCl2 at EC8. On the other hand, the highest leaf macronutrient concentrations were in KNO3 + CaNO3 at EC5 and/or EC8 that showed only higher leaf N and Ca values compared to the control. Leaf micronutrient concentrations were the highest in KNO3 + CaNO3 at EC5 that generally showed no difference with control plants. However, application of KCl + CaCl2 particularly at EC8 and to less extent KNO3 + CaNO3 + NaCl at EC8, reduced the leaf micronutrient concentrations. Application of KNO3 + CaNO3 at EC5 increased and KCl + CaCl2 or KNO3 + CaNO3 + NaCl at EC8 decreased the leaf Fe concentration compared to control plants.

Nasze badania dotyczą zastosowania termostatowanej mikrochromatografii cienkowarstwowej do optymalizacji procesu rozdzielania wybranych aminokwasów tryptofanu i proliny w układzie faz odwróconych (RP).
Thin-layer chromatography (TLC) and high-performance thin-layer chromatography (HPTLC) are commonly used in separation, identification, purification and quantification of different analytes present in complex biological, environmental and pharmaceutical samples. Planar chromatography can be also applied to screen analysis and fast exploration of chromatographic retention profiles using different concentrations of mobile phases components. This method does not require expensive equipment, tedious and time-consuming sample pretreatments and, what is important, allows a parallel separation of many samples at the same time. The spots or bands sprayed onto the TLC plate can be easily detected under visible and UV light (usually 254nm, 366nm) or by post-chromatographic derivatization with visualization reagents, and then digitalized using office scanners. It should be noted that the great advantage of micro-planar chromatography compared to the regular sized TLC is low consumption of the eluents (usually less than 1 mL per analytical run) as well as short analysis time, due to mobile phase migration distance less than 50 mm. Under particular cases (high temperature and low viscosity eluents like acetone, dichloro-methane or n-hexane), non-forced eluent flow micro-chromatographic analysis can be completed within less than 5 minutes. This work is focused on optimization of separation process of two amino acids including tryptophan and proline (Fig. 1) under reversed-phase micro-chromatographic conditions. Particularly, selected amino acids were chromatographed on HPTLC RP-18W (wettable with water) 25 x 50 mm cutted plates using thermostated horizontal micro-chamber and unsaturated conditions (Fig. 2). The target components were sprayed on the start line (1Μg per lane) using a semi-automatic sampler (Linomat 5, Camag). The plates were developed with n-propanol: water binary mixtures as the mobile phases. Different concentrations of alcohol ranging from 0 to 100% (v/v) with steps of 20% were applied. The temperature of chromatographic separation was set at 40oC and chromatographic runs were completed within 10-30 minutes, depending on the water concentration in the mobile phase. The amino acids were visualized by dipping the plates in ninhydrin solution (prepared for concentration 0.3% in ethanol) followed by heating at 120°C for 10 minutes. Picture acquisition was performed using a Plustek OpticPro S12 scanner with an 8-bit per channel color deep mode, 300 DPI resolution, and saved as TIFF files. Retention data were derived from the plates via densitometric scans obtained with help of ImageJ freeware (http://rsb.info.nih.gov/ij). Using the raw retention data set (RF values, retardation factor), the chromatographic profiles of amino acids were calculated with step of 2% (Fig. 3A). Based on the curves observed and relationship RM = logk, where RM and k correspond to mobility and retention factors, respectively, a simple optimization criterion expressed as a separation factor (α) was calculated (Fig. 3B). Using this plot, the best separation conditions of target components were selected taking into account the maximum value of the separation factor (αmax). Due to the equal spot shape of amino acids investigated across the whole range of the mobile phases composition, such approach involving the α values allows fast optimization of separation in planar chromatography and direct application of selected conditions into column chromatographic systems based on e.g. high-performance liquid chromatography (HPLC). It should be noted that the advantages of retention data screening using micro-TLC are very low mobile phase consumption (at level of 300 ΜL per chromatographic run), short analysis time and effective detection of UV-Vis transparent substances using simple visualization reagent.

Wstawienie liści bezpośrednio po zbiorze do pożywki standardowej spowodowało 4-krotny wzrost poziomu amoniaku i prawie 3-krotny proliny w stosunku do stanu wyjściowego.
The contents of ammonium and proline in ageing cut leaves of Hosta ‘Undulata Erromena’ affected by the standard preservative used to prolong cut flower longevity (composed of 8HQC 200 mg·dm⁻³ + sucrose 2%) and benzyladenine (BA) or gibberellic acid (GA₃) was examined. In leaves kept held in water the 2.5 fold increase in ammonium content was observed as compared to the initial level and almost a similar rise in proline content occurred in the senescing leaves. In leaves kept held in the preservative solution increase in ammonium and proline contents was 4-fold and 3-fold, respectively. BA application as a 24 hour puls treatment stopped both ammonium and proline accumulation, maintaining their contents at the initial level. BA prevented ammonium and proline accumulation more effectively than GA₃ did.

Opis:
W pracy oceniano masę 1000 ziaren, zawartość białka i skład aminokwasowy w 9 próbach jęczmienia: 8 odmian jęczmienia oplewionego i 1 ród jęczmienia nieoplewionego. Ziarno jęczmienia oplewionego cechowało się mniejszą masą 1000 ziaren i mniejszą zawartością białka w porównaniu z ziarnem nieoplewionym. Badane próby jęczmienia wykazują podobny skład aminokwasowy. Jęczmień nieoplewiony zawierał więcej kwasu glutaminowego i proliny oraz mniejsze ilości pozostałych aminokwasów w porównaniu z ziarnem oplewionym. Białka jęczmienia oplewionego cechowały się wyższą wartością odżywczą niż białka jęczmienia nieoplewionego.
Kernel weight, protein content and amino acid composition were determined in 9 grain samples of barley, including 8 covered and 1 naked types. The covered samples were lower in kernel weight and in protein content than the naked type. In all barley samples there are identical patterns of amino acid composition. The naked barley contained more glutamic acid and proline and less of most of other amino acids than covered barley. A comparison with the FAO amino acid pattern indicates the high contents of essential amino acids in covered barley. Nutritionally the proteins of covered barley were better than naked barley.
