Over the last 25 years one of us (WKS) has been investigating physical properties of lipid bilayer membranes. In 1991 a group led by WKS was organized into the Laboratory of Structure and Dynamics of Biological Membranes, the effective member of which is AW. Using mainly the electron paramagnetic resonance (EPR) spin-labeling method, we obtained unexpected results, which are significant for the better understanding of the functioning of biological membranes. We have developed a new pulse EPR spin-labeling method for the detection of membrane domains and evaluation of lipid exchange rates. This review will be focused on our main results which can be summarized as follows: (1) Unsaturation of alkyl chains greatly reduces the ordering and rigidifying effects of cholesterol although the unsaturation alone gives only minor fluidizing effects, as observed by order and reorientational motion, and rather significant rigidifying effects, as observed by translational motion of probe molecules; (2) Fluid-phase model membranes and cell plasma membranes are not barriers to oxygen and nitric oxide transport; (3) Polar carotenoids can regulate membrane fluidity in a way similar to cholesterol; (4) Formation of effective hydrophobic barriers to the permeation of small polar molecules across membranes requires alkyl chain unsaturation and/or the presence of cholesterol; (5) Fluid-phase micro-immiscibility takes place in cis-unsaturated phosphatidylcholine-cholesterol membranes and induces the formation of cholesterol-rich domains; (6) In membranes containing high concentrations of transmembrane proteins a new lipid domain is formed, with lipids trapped within aggregates of proteins, in which the lipid dynamics is diminished to the level of gel-phase.

Since energy storage is a basic metabolic process, the synthesis of neutral lipids occurs in all kingdoms of life. The yeast, Saccharomyces cerevisiae, widely accepted as a model eukaryotic cell, contains two classes of neutral lipids, namely steryl esters and triacylglycerols. Triacylglycerols are synthesized through two pathways governed by the acyl-CoA diacylglycerol acyltransferase Dga1p and the phospholipid diacylglycerol acyltransferase Lro1p, respectively. Steryl esters are formed by the two steryl ester synthases Are1p and Are2p, two enzymes with overlapping function which also catalyze triacylglycerol formation, although to a minor extent. Storage of neutral lipids is tightly linked to the biogenesis of so called lipid particles. The role of this compartment in lipid homeostasis and its interplay with other organelles involved in neutral lipid dynamics, especially the endoplasmic reticulum and the plasma membrane, are subject of current investigations. In contrast to neutral lipid formation, mobilization of triacylglycerols and steryl esters in yeast are less characterized at the molecular level. Only recently, the triacylglycerol lipase Tgl3p was identified as the first yeast enzyme of this kind by function. Genes and gene products governing steryl ester mobilization still await identification. Besides biochemical properties of enzymes involved in yeast neutral lipid synthesis and degradation, regulatory aspects of these pathways and cell biological consequences of neutral lipid depletion will be discussed in this minireview.

Below the melting point temperature of lipids, artificial lipid membranes usually exist in the ordered gel phase. Above these temperatures lipid acyl chains become fluid and disordered (liquid-crystalline phase). Depending on the chemical composition of artificial membranes, phase separation may occur, leading to the formation of transient or stable membrane domains. A similar phase separation of lipids into ordered and disordered domains has been observed in natural membranes at physiological temperature range. Moreover, it has been reported that certain proteins prefer certain organization of lipids, as for example glycosylphosphatidylinositol-anchored proteins or Src family of tyrosine kinases. The aim of present review is to discuss the possibility that some lipid microdomains are induced or stabilized by lipid-binding proteins that under certain conditions, for example due to a rise of cytosolic Ca2+ or pH changes, may attach to the membrane surface, inducing clustering of lipid molecules and creation of ordered lipid microdomains. These domains may than attract other cytosolic proteins, either enzymes or regulatory proteins. It is, therefore, postulated that lipid microdomains play important roles within a cell, in signal transduction and enzymatic catalysis, and also in various pathological states, as Alzheimer's disease, anti-phosphatidylserine syndrome, or development of multidrug resistance of cancer cells.

The purpose of this work was to compare the fatty acid composition of the following reserve lipid tissues: mane, perirenal and intermuscular, and muscles: semimembranous with thigh adductor, middle gluteal, dorsal longissimus and infraspinous with serratus. Perirenal tissue contained significantly less unsaturated fatty acids (59.96%, p<0.05) compared to mane (61.43%, p<0.05) and intermuscular (61.24%, p<0.05) tissues. The tissues differed in the content of palmitic, stearic, oleopalmitic, miristic, and linolenic acids. All the tissues had a predominance (from 2.87% to 3.92%) of linolenic acid over linoleic acid, unique to animal fats. The percentage of particular fatty acids, except palmitic and linoleic acids in all muscle lipids was very similar. In contrast to reserve lipid tissues, the content of linoleic acid in muscle lipids was higher by 1.68% to 4.18% than of linolenic acid.
Celem podjętych badań było porównanie składu kwasów tłuszczowych zapasowych tkanek Puszczowych tusz końskich; grzywowej, okołonerkowej i międzymięśniowej oraz mięśni: półbłoniastego uda z przywodzicielem uda, pośladkowego środkowego, najdłuższego grzbietu, podgrzebieniowego z zębatym brzusznym. Ustalono, że tkanka okołonerkowa zawierała istotnie mniej kwasów nienasyconych (59.96%) w porównaniu do tkanek grzywowej (61.43%) międzymięśniowej (61.24%). Badane tkanki różniły się ilością kwasów palmitynowego, stearynowego, oleopalmitynowego, mirystynowego i linolenowego. Wszystkie tkanki tłuszczowe zapasowe cechowała unikalna wśród tłuszczów zwierzęcych przewaga kwasu linolenowego nad molowym wynosząca od 2.87% do 3.92%. Lipidy wszystkich wymienionych mięśni charakteryzowały się bardzo wyrównanym udziałem procentowym poszczególnych kwasów tłuszczowych, za wyjątkiem kwasów palmitynowego i linolowego. W lipidach mięśni w przeciwieństwie do tkanek zapasowych ilość kwasu linolowego była wyższa niż kwasu linolenowego d 1.68% do 4.14%.

Lipidy, szczególnie te nienasycone, są związkami niestabilnymi chemiczne i łatwo ulegają reakcjom oksydacji. Utlenione lipidy stanowią, trudną w analizie, mieszaninę różnych związków, które wpływają nie tylko na pogorszenie się jakości żywności, ale także prowadzą do niekorzystnych zmian w organizmie człowieka. W artykule przedstawiono ogólny mechanizm reakcji oksydacji lipidów oraz wskazano na znaczenie powstałych produktów w aspekcie żywności, surowców kosmetycznych oraz zdrowia człowieka. Omówiono także dostępne metody analizy produktów utleniania lipidów oraz związane z nią problemy.
Lipids, especially unsaturated ones, are unstable compounds which easily undergo oxidation reactions. Difficult to analyze, oxidized lipids are a mixture of different compounds which not only cause the deterioration of food quality, but also lead to adverse changes in the human body. This article presents a general mechanism of lipid oxidation reaction and shows the importance of the resulting products in terms of food, cosmetic raw materials and human health. The available analysis methods of lipid oxidation products and the associated problems are also discussed.

Opis:
Myeloperoxidase (MPO), an abundant enzyme in phagocytes, has been implicated in the pathogenesis of various inflammatory diseases including atherosclerosis. The major oxidant produced by MPO, hypochlorous acid (HOCl), is able to modify a great variety of biomolecules by chlorination and/or oxidation. In this paper the reactions of lipids (preferentially unsaturated fatty acids and cholesterol) with either reagent HOCl or HOCl generated by the MPO-hydrogen peroxide-chloride system are reviewed. One of the major issues has been whether the reaction of HOCl with lipids of low density lipoprotein (LDL) yields predominantly chlorohydrins or lipid hydroperoxides. Electrospray mass spectrometry provided direct evidence that chlorohydrins rather than peroxides are the major products of HOCl- or MPO-treated LDL phosphatidylcholines. Nevertheless lipid peroxidation is a possible alternative reaction of HOCl with polyunsaturated fatty acids if an additional radical source such as pre-formed lipid hydroperoxides is available. In phospholipids carrying a primary amino group such as phosphatidylethanolamine chloramines are the preferred products compared to chlorohydrins. Cholesterol can be converted by HOCl to great variety of oxysterols besides three isomers of chlorohydrins. For the situation in vivo it appears that the type of reaction occurring between HOCl and lipids would very much depend on the circumstances, e.g. the pH and the presence of radical initiators. The biological effects of lipid chlorohydrins are not yet well understood. It has been shown that chlorohydrins of both unsaturated fatty acids as well as of cholesterol may cause lysis of target cells, possibly by disruption of membrane structures.

Opis:
Unsaturated lipids are rapidly oxidized to toxic products such as lipid hydroperoxides, especially when transition metals such as iron or copper are present. In a Fenton-type reaction Fe2+ converts lipid hydroperoxides to the very short-lived lipid alkoxyl radicals. The reaction was started upon the addition of Fe2+ to an aqueous linoleic acid hydroperoxide (LOOH) emulsion and the spin trap in the absence of oxygen. Even when high concentrations of spin traps were added to the incubation mixture, only secondary radical adducts were detected, probably due to the rapid rearrangement of the primary alkoxyl radicals. With the commercially available nitroso spin trap MNP we observed a slightly immobilized ESR spectrum with only one hydrogen splitting, indicating the trapping of a methinyl fragment of a lipid radical. With DMPO or 5-diethoxyphosphoryl-5-methyl-1-pyrroline N-oxide (DEPMPO) adducts were detected with carbon-centered lipid radical, with acyl radical, and with the hydroxyl radical. We also synthesized lipophilic derivatives of the spin trap DEPMPO in order to detect lipid radical species generated in the lipid phase. With all spin traps studied a lipid-derived carbon-centered radical was obtained in the anaerobic incubation system Fe2+/LOOH indicating the trapping of a lipid radical, possibly generated as a secondary reaction product of the primary lipid alkoxyl radical formed. Under aerobic conditions an SOD-insensitive oxygen-centered radical adduct was formed with DEPMPO and its lipophilic derivatives. The observed ESR parameters were similar to those of alkoxyl radical adducts, which were independently synthesized in model experiments using Fe3+-catalyzed nucleophilic addition of methanol or t-butanol to the respective spin trap.

The frequency of complications of dental implantation at the present stage varies from 6% to 23%, what determined the relevance of this examination. The aim of this work is to determine the predictive effectiveness of the development of inflammatory complications of dental implantation on the basis of indicators of lipid peroxidation (POL) of the oral fluid (level of malondialdehyde (MDA) and the activity level of superoxide dismutase (SOD)). We examined 49 patients 25-59 years old which had no traumas, operations and diseases requiring medical rehabilitation, inflammatory disease in the maxillofacial area and the gastrointestinal tract, as well as other factors that can affect qualitative and quantitative content of the oral fluid. These patients underwent delayed dental implantation (one implant in one segment of the jaw). The POL indices were determined twice: before operation of dental implantation and 3 days after operation. The effectiveness of the proposed method was determined in accordance to the clinical and economic studies requirements. Prognostic efficiency in accordance with the procedure of conducting clinical and economic examinations on the basis of MDA level was 78%, and based on the level of SOD activity – 76%, which classifies it as high and allow to reduce the number of complications and diagnostic mistakes including in the planning and execution of invasive procedures associated with the installation of dental implants.

The Langmuir monolayer technique and voltammetric analysis were used to investigate the properties of model lipid membranes prepared from dioleoylphosphatidylcholine (DOPC), hexadecaprenol (C80), and their mixtures. Surface pressure- molecular area isotherms, current-voltage characteristics, and membrane conductance-temperature were measured. Molecular area isobars, specific molecular areas, excess free energy of mixing, collapse pressure and collapse area were determined for lipid monolayers. Membrane conductance, activation energy of ion migration across the membrane, and membrane permeability coefficient for chloride ions were determined for lipid bilayers. Hexadecaprenol decreases the activation energy and increases membrane conductance and membrane permeability coefficient. The results of monolayer and bilayer investigations show that some electrical, transport and packing properties of lipid membranes change under the influence of hexadecaprenol. The results indicate that hexadecaprenol modulates the molecular organisation of the membrane and that the specific molecular area of polyprenol molecules depends on the relative concentration of polyprenols in membranes. We suggest that hexadecaprenol modifies lipid membranes by the formation of fluid microdomains. The results also indicate that electrical transmembrane potential can accelerate the formation of pores in lipid bilayers modified by long chain polyprenols.

Hodowla i wykorzystanie mikroglonów jest obecnie intensywnie rozwijającym się obszarem badawczym. Niektóre gatunki mikroalg, w odpowiednich warunkach, gromadzą duże ilości tłuszczu w komórkach, który ma odpowiedni profil kwasów tłuszczowych do produkcji biodiesela. Etapem kierunkującym mikroglony na akumulację lipidów jest hodowla w określonych warunkach fizykochemicznych. Celem badań było określenie wpływu zmiennego stężenia fosofru w medium hodowlanym na olejogenność mikroglonów Chlorella vulgaris oraz wyznaczenie parametrów hodowli w trybie szoku. Przeprowadzone badania potwierdziły możliwość stosowania szoku fosforowego w celu makasymalizacji syntezy lipidów przez mikroglony Chlorella vulgaris. W trakcie badań uzyskano 45,23% oleju z biomasy z hodowli z szokiem fosforowym, natomiast z biomasy pochodzącej z hodowli standardowej uzyskano o 18% oleju mniej niż z biomasy pochodzącej z hodowli z deficytem fosforu.
The cultivation and utilization of microalgae is now a intensively developing area of research. Some species of microalgae, under appropriate conditions, accumulate large amounts of lipids in the cells. This lipids have a suitable profile of fatty acids for biodiesel production. The culture of microalgae for lipids accumulation should be performed in certain physicochemical conditions. The aim of the study was to determine the effect of variable ortophophates concentrations in the culture medium for lipids accumulation of microalgae Chlorella vulgaris and to determine of parameters of the phosphoric shock in the medium. The study confirmed the possibility of the use of the phosphoric shock in the medium to maximize lipids accumulation by the microalgae Chlorella vulgaris. In the study, 45.23% of the oil was obtained from the biomass from the culture with phosphoric shock in the medium and 18% less of the oil was obtained from the biomass from the standard culture.

This study aims to analyze the lipid, carbohydrate and protein content of Chlorella pyrenoidosa after being treated with the tofu wastewater. The method used in this study was an experimental method with 4 treatments and 3 replications. The treatment was performed by administering different concentrations of the tofu wastewater to the C. pyrenoidosa. The concentrations used were 10%, 15% and 20%. Determination of these concentrations was based on the preliminary test. The main parameters observed were the lipid, carbohydrate and protein content of C. pyrenoidosa and the supporting parameters were the growth rate, doubling time and characteristics of the tofu wastewater. The study was conducted for 8 days using the batch culture method. In the exponential phase, the microalgae were harvested and then their contents were analyzed. The data obtained were analyzed using MS Office Excel 2016. The highest content of lipid, carbohydrate and protein of C. pyrenoidosa was in the treatment of 20% tofu wastewater, reaching 1.56%; 28.92%; and 28.92%, respectively. Meanwhile, the highest growth rate and the smallest doubling time in the treatment of 20% tofu wastewater accounted for 0.8264 day-1 and 0.0349 hours-1. Moreover, the rates of BOD and TSS of the tofu wastewater at the end of the study decreased. It can be concluded that administering different concentrations of the tofu wastewater can affect the lipid, carbohydrate and protein content of C. pyrenoidosa. It was suggested that further research is needed to conduct semi-continuous cultivation of C. pyrenoidosa using a higher density so that the results obtained can be optimized.

The aim of the study was to determine the effect of the addition of antioxidants on changes in contents of available lysine and methionine in raw Polish sausage. Experimental material consisted of four variants of experimental sausages. One variant consisted of a sample with no antioxidant added (control), while the following were added to the other variants: BHT (0.02%), rosemary ethanol extract (0.05%) and soy protein hydrolysate (2%). Oxidation degree of lipid using peroxide value (PV), anisidine value (AV), thiobarbituric acid reactive substances (TBARS) and the Totox index was controlled and content of available lysine and methionine were determined periodically. In order to determine the effect of the addition of antioxidants on the stability of available lysine and methionine, values of the coefficient of the slope of curve of changes in time (coefficient a/24 h) were analyzed along with the half-life period of the amino acids (TIC50). Antioxidants exhibited an inhibitory effect on the advancement of lipid autoxidation reactions and reduced quantitative losses of available lysine and methionine in analyzed sausages. Natural antioxidants exhibited a lower capacity to reduce losses of nutritive value of protein than it was found for BHT.
W pracy określono wpływu dodatku przeciwutleniaczy na stabilność dostępnej lizyny i metioniny w kiełbasie typu „polska surowa”, jako istotnych czynników determinujących wartość żywieniową produktów mięsnych. Materiałem badawczym były cztery warianty wędlin doświadczalnych. Jeden wariant stanowiła próba bez dodatku przeciwutleniacza (próba kontrolna), do pozostałych dodano odpowiednio: BHT (0,02%), etanolowy ekstrakt rozmarynu (0,05%) i hydrolizat sojowy (2%). Okresowo oznaczano liczbę nadtlenkową (PV), liczbę anizydynową (AV), zawartość substancji dających reakcję barwną z kwasem 2- tiobarbiturowym (TBARS), indeks Totox oraz zawartość dostępnej lizyny i metioniny. W celu określenia aktywności przeciwutleniającej badanych dodatków wyznaczono współczynniki kierunkowe (Wsp.a/24 h), stanowiące kąt nachylenia krzywej regresji wykreślonej z punktów pomiarowych danego wskaźnika w czasie. Wpływ przeciwutleniaczy na stabilność dostępnej lizyny i metioniny analizowano na podstawie wyznaczonego czasu połowicznego rozpadu aminokwasów (TIC50). Przeciwutleniacze wykazywały hamujący wpływ na postęp reakcji autooksydacji lipidów w badanych wędlinach oraz w istotny sposób zmniejszały straty ilościowe dostępnej lizyny i metioniny. Przeciwutleniacze naturalne wykazywały mniejszą zdolność obniżania strat wartości odżywczej białka niż BHT.

Canthaxanthin (β,β-carotene 4,4' dione) used widely as a drug or as a food and cosmetic colorant may have some undesirable effects on human health, caused mainly by the formation of crystals in the macula lutea membranes of the retina of an eye. Experiments show the exceptional molecular organization of canthaxanthin and a strong effect of this pigment on the physical properties of lipid membranes. The most striking difference between canthaxanthin and other macular pigments is that the effects of canthaxanthin at a molecular level are observed at much lower concentration of this pigment with respect to lipid (as low as 0.05 mol%). An analysis of the molecular interactions of canthaxanthin showed molecular mechanisms such as: strong van der Waals interactions between the canthaxanthin molecule and the acyl chains of lipids, restrictions to the segmental molecular motion of lipid molecules, modifications of the surface of the lipid membranes, effect on the membrane thermotropic properties and finally interactions based on the formation of the hydrogen bonds. Such interactions can lead to a destabilization of the membrane and loss of membrane compactness. In the case of the retinal vasculature, it can lead to an increase in the permeability of the retinal capillary walls and the development of retinopathy.

Paraoxonase 1 (PON1), contained in plasma high-density lipoproteins, plays an important role in the protection of plasma lipoproteins and cell membranes from oxidative damage. Previous studies indicate that human PON1 is stimulated by high NaCl concentrations. The aim of this study was to characterize in more detail the effect of salts on serum PON1. Paraoxon-hydrolyzing activity of human serum was stimulated by 81.6% following the addition of 1 M NaCl. The effect of NaCl was dose-dependent between 0.5 and 2 M. PON1 activity toward phenyl acetate was reduced by 1 M NaCl by 55.2%. Both the paraoxon- and phenyl acetate-hydrolysing activity was slightly lower in heparinized plasma than in serum, but NaCl had similar stimulatory and inhibitory effects on these activities, respectively. In rat, rabbit, and mouse, NaCl reduced PON1 activity. KCl had a similar effect on human PON1 as NaCl. Sodium nitrite also stimulated human PON1 but much less effectively than chloride salts. In contrast, sucrose, sodium acetate and sodium lactate had no significant effect. NaBr was a less effective PON1 activator than NaCl, whereas the effect of NaJ was non-significant. The activity of human PON1 toward homogentisic acid lactone and γ-decanolactone was unaltered by NaCl. These data indicate that: 1) high concentrations of chlorides stimulate human PON1 activity toward paraoxon but not other substrates, 2) PON1 is inhibited by Cl- in other mammalian species, 3) the potency of human PON1 activation by halogene salts increases with decreasing atomic mass of the halide anion.

Objectives: The aim of the study was to evaluate effects of Finnish sauna bathing on lipid profile in healthy, young men. Material and Methods: Sixteen male subjects (20–23 years) were subjected to 10 sauna bathing sessions in a Finnish sauna every 1 or 2 days. The mean sauna temperature was 90±2°C, while humidity was 5–16%. Each session consisted of three 15-minute parts and a 2-minute cool-down between them. The following measurements were taken before and after the sauna sessions: body mass, heart rate, body skinfold thickness. The percentage fat content and then, the lean body mass were calculated. Total cholesterol, triacylglycerols, lipoprotein cholesterol LDL and HDL were measured in blood samples. Results: A statistically significant decrease of total cholesterol and LDL cholesterol was observed during 3 weeks of sauna treatment and in the week afterwards. A significant decline in triacylglycerols was found directly after the 1st and 24 h directly after the 10th sauna session. After the 10th sauna session the level of HDL cholesterol remained slightly increased, but this change was not statistically significant. A decrease in blood plasma volume was found directly after the 1st and the last sauna bathing session due to perspiration. An adaptive increase in blood plasma volume was also found after the series of 10 sauna sessions. Conclusions: Ten complete sauna bathing sessions in a Finnish sauna caused a reduction in total cholesterol and LDL cholesterol fraction levels during the sessions and a gradual return of these levels to the initial level during the 1st and the 2nd week after the experiment. A small, statistically insignificant increase in HDL-C level and a transient decline in triacylglycerols were observed after those sauna sessions. The positive effect of sauna on lipid profile is similar to the effect that can be obtained through a moderate-intensity physical exercise.

Flotacja jest jedną z najefektywniejszych metod wstępnej identyfikacji oddziaływań białko-błony lipidowe. W większości przypadków wykorzystuje się w niej małe jednowarstwowe pęcherzyki lipidowe, które służą jako modele błonowe i nie wymagają dodatkowych nośników, takich jak membrany czy nanocząstki polimerowe, które są często używane w innych metodach mających na celu identyfikację oddziaływań białkolipid. W poniższej pracy prezentujemy wyniki uzyskane podczas badań oddziaływań kwasu fosfatydowego i syndapiny. Omawiamy także niektóre techniczne aspekty metody, kładąc nacisk na to jak małe zmiany w warunkach metody mogą wpłynąć na otrzymywane wyniki.
Flotation is one of the best method for preliminary identification of protein-lipid interactions. In most widely used approach it utilizes large unilamellar vesicles, that are excellent models of freestanding membranes and do not require any additional components, like solid supports or beads that are needed in other methods commonly used for protein-lipid binding studies. Here we present results obtained during our studies on phosphatidic acid - syndapin interactions and discuss some technical aspects of this method underlying how relatively small changes in the conditions can influence the results.

Lipopolysaccharides of seven Bradyrhizobium strains and three whole-cell fatty acid preparations from bacteria isolated from nodules of Sarothamnus scoparius (Common Broom) were studied for the presence of very long chain (ω-1)-hydroxy fatty acids. Several such fatty acids were identified. Among them, straight-chain as well as mono- and dimethyl branched acids with chains in the range from 26 to 34 carbon atoms were found. Pyrrolidides and 4,4-dimethyloxazoline derivatives were used to determine the branching position. Carbons at the (ω-10) and/or (ω-11) positions in alkyl chains were points of attachment of methyl groups. These data complete the structure of bradyrhizobial lipid A with important details. The obtained results can be applied in the chemotaxonomy of Bradyrhizobium.

The influence of gelatin coatings modified with fungal melanin on pork lard oxidative stability was studied. The lard was coated with gelatin coatings containing 0.1%; 0.5%; 1% of fungal melanin and control gelatin coating devoid of melanin. The peroxide values (POV), iodine values (IV) and acid values (AV) were studied after 7, 14 and 21 days storage in controlled conditions. Lard covered with modified coatings had lowered oxidative rancidity. Hence, modified coatings containing fungal melanin can be use effectively for the prevention of lard oxidation.

The glycine receptor belongs to the ligand-gated ion channel superfamily. It is a chloride conducting channel composed of four transmembrane domains. It was previously shown that the second transmembrane domain (M2) of the glycine receptor forms an ion conduction pathway throught lipid bilayers. The amino-acid sequence of the transmembrane segment M2 of the glycine receptor has a high homology to all receptors of the ligand-gated ion channel superfamily. In our report, we have used a synthetic M2 peptide. It was incorporated into a planar membrane of known lipid composition and currents induced by M2 were measured by the Black Lipid Membrane technique. When the planar lipid bilayer was composed of 75% phosphatidylethanolamine and 25% phosphatidylserine, the reversal potential measured in a 150/600 mM KCl (cis/trans) gradient was -19 mV suggesting that the examined pore was preferential to anions, PK/PCl = 0.25. In contrast, when 75% phosphatidylserine and 25% phosphatidylethanolamine was used, the reversal potential was +20 mV and the pore was preferential to cations, PK/PCl = 4.36. Single-channel currents were recorded with two predominant amplitudes corresponding to the main-conductance and sub-conductance states. Both conductance states (about 12 pS and 30 pS) were measured in a symmetric solution of 50 mM KCl. The observed single-channel properties suggest that the selectivity and conductance of the pore formed by the M2 peptide of the glycine receptor depend on the lipid composition of the planar bilayer.

Total antixodative effect is difficult to predict because the total antiradical capacity depends not only on properties of individual compounds, but also on their mutual interactions. The study examined the effect of the addition of Ginkgo biloba antioxidants to refined rapeseed oil on the stabilization of the oil and its triacylglycerols, measured by an Oxidograph accelerated oxidative stability test and the Rancimat test. The mineral composition of oil was also determined. The tested samples were observed to respond differently in terms of the induction period, which was the longest periods for ethanolic extracts, and the shortest for aqueoeus ones. The highest values of the protective factor were observed in the samples with added ethanolic extract of green leaves, while the lowest ones were noted in all aqueous extracts. In all the tested samples, values of the protection factor increased along with the growing extract concentration in a sample. Among the tested additives, butylated hydroxytoluene added to a sample of oil resulted in the longest induction period. Among the analysed microelements, iron was the prevalent one, with its concentration being significantly higher in water extracts. The least abundant was selenium. The antioxidative capacity of Ginkgo biloba extract resulted from the presence of transitional metals and polyphenols, as the presence of zinc, copper and iron affects the bioavailability of flavonoids. Negative relationships were noted between microelements in the extracts and its oxidative stability, which indicated the oxidative activity of the former. G. biloba extracts may be a new source of stabilising additives for high-fat foods. However, further reasearch I needed to confirm this assumption, particularly because food is a complex matrix, in which ingredients might interact with one another, causing higher or lower antioxidative capacity

Seed germination is the earliest process in plant development and is crucial for further plant growth and fitness. The process is regulated by various internal and external factors, including soil pollutants such as nonessential metals. In the present study, we examined in detail the impact of short-term imbibition in Cd solutions at several concentrations (5, 10, and 25 mg/L) on germination rate and physiological state of soybean seeds. The results showed that although Cd was readily absorbed by the seeds, the metal had no effect on seeds cell viability, oxidative stress intensity, or germination percentage. In contrast, imbibition in Cd solution led to slight reduction in antioxidant capacity of seeds. Seedlings grown from seeds pretreated with metal showed no differences in growth in relation to the control. Taken together, the results indicate that soybean seeds are relatively tolerant even to high Cd concentration (up to 25 mg/L).

Liposomes (LSs) are promising nanoparticles with unique properties such as controlled nanosize, large surface area, increased reactivity, and ability to undergo modification. Worldwide, licensed liposomal forms of antibiotics, hormones, antioxidants, cytostatics, ophthalmic drugs, etc., are available on the pharmaceutical market. This review focuses on the adjuvant properties of LSs in the production of vaccines (VACs). LS-VACs have the following advantages: antigens with low immunogenicity can become highly immunogenic; LSs can include both hydrophilic and hydrophobic antigens; LSs allow to achieve a prolonged specific action of antibodies; and LSs reduce the toxicity and pyrogenicity of encapsulated antigens and adjuvants. The immune response is influenced by the composition of the liposomal membrane, physicochemical characteristics of lipids, antigen localization in LSs, interaction of LSs with complement, and a number of proteins, which leads to opsonization. The major requirements for adjuvants are their ability to enhance the immune response, biodegradability, and elimination from the organism, and LSs fully meet these requirements. The effectiveness and safety of LSs as carriers in the antigen delivery system have been proven by the long-term clinical use of licensed vaccines against hepatitis A, influenza, herpes zoster, malaria, and COVID-19.

Opis:
Nanostructured lipid carriers (NLC) composed of the substances generally recognized as safe (GRAS) were obtained by using a hot high-pressure homogenization technique (HPH). The influence of the number of homogenization cycles and concentration of a decyl glucoside surfactant on the NLC properties were studied. The system's stability was assessed by macroscopic observation, light backscattering and zeta potential measurements. NLC particle size was measured using dynamic light scattering (DLS). The kinetically stable formulations were loaded with forskolin and selected for in vitro drug permeation study using the Franz cell method. Concentration of forskolin in the receptor solution (i.e. ethanol/PBS mixture) was analyzed with high performance liquid chromatography (HPLC) with UV detection. The obtained results have shown that NLC formulations could be used as effective carriers for forskolin permeation through the skin.

Our previous study showed the efficacy of lactoferrin-monophosphoryl lipid A isolated from Hafnia alvei LPS complex (LF-MPLH.a.) as an adjuvant in stimulation of humoral and cellular immune response in mice to conventional antigens and a lower pyrogenicity of the complex as compared with MPLH.a. alone. In the present investigation we demonstrated that LF-MPLH.a. complex enhanced the immunity of BALB/c mice immunized with Plesiomonas shigelloides CNCTC 138/92 bacterial vaccine, against P. shigelloides infection. The adjuvant effect was evidenced by a significant increase of the antigen-specific serum IgG, IgG2a, and IgG1 and elevation of antigen-specific serum IgA concentrations. In addition, application of the adjuvant facilitated better clearance of the bacteria in spleens and livers of infected mice when compared with MPLH.a. alone. These features of the new adjuvant may predispose it for vaccination protocols in humans.

The results presented demonstrate the influence of aromatic indolinic aminoxyls: 1,2-dihydro-2-ethyl-2-phenyl-3H-indole-3-phenylimino-1-oxyl (IA-C2) and 1,2-dihydro-2-octadecyl-2-phenyl-3H-indole-3-phenylimino-1-oxyl (IA-C18) on oxidation of lipids and proteins of cardiac sarcoplasmic reticulum membranes. We have used doxorubicin and t-butyl hydroperoxide as agents inducing oxidative stress in isolated rat cardiac sarcoplasmic reticulum membrane system. Carbonyl groups were measured as the end product of membrane protein oxidation, and thiobarbituric acid reactive substances were assessed as a marker of lipid peroxidation. Inhibition of peroxidation of certain membrane components depends on the length of acyl chain. Aminoxyl IA-C2 inhibits the lipid peroxidation process while IA-C18 is an efficient protector against protein oxidation.

Invasive alien species are non-native plant species that displace native species and pose adverse effects to environment, ecosystem, economy, and human health by diminishing the growth of native plants and by exhibiting higher stress tolerance. In our present study, four invasive alien species, namely Lantana camara, Parthenium hysterophorous, Ricinus communis, and Ageratum conyzoides, were studied from different locations. Plants growing under natural environmental conditions were sampled at random in the vicinity of Jalandhar. To gain insights into the biochemical basis of invasiveness of these plants, the samples were subjected to chemical fingerprinting by using UV-Vis, fluorescent, and Fourier transform-infrared (FT-IR) techniques under natural abiotic stress conditions (moderate and hot conditions). Indices of oxidative stress, such as malondialdehyde (MDA), were also studied. MDA levels were enhanced under hot conditions. Elevated peaks (major and minor) were observed in UV fingerprinting during adverse abiotic conditions. Fluorescent spectroscopy also validated the enhanced levels of secondary metabolites. FT-IR spectroscopy confirmed the presence of alkaloids and phenolics during stress conditions. Peaks were identified as rutin, vanillic acid, ascorbic acid, and glutathione reduced. The obtained results showed that under stressful conditions, the studied plants may produce an increased level of metabolites that might play a role in minimizing the oxidative stress faced by these plants. It was concluded that the studied plants, namely P. hysterophorus, L. camara, R. communis, and A. conyzoides, have the potential to cope with abiotic stress such as high temperature, which could be the reason for their invasiveness and vast adaptability.

Z uwagi na fakt, że lipidy stanowią bezpieczne składniki kosmetyków, w części opisowej artykułu na wstępie scharakteryzowano lipidy. W dalszym ciągu omówiono kwasy tłuszczowe, oraz lipidy proste, złożone izoprenowe. Następnie podano przegląd kosmetyków zastosowujących lipidy. W części badawczej artykułu przedstawiono wyniki ankiety przeprowadzonej z udziałem 48 respondentek w wieku 20 do 50 lat. Ankieta zawierająca 12 pytań dotyczyła stosowania kosmetyków głównie zawierających lipidy.
Consideration the fact, that lipids are safety ingredients of the cosmetics, the lipids, o on the begining of the article, in the descriptive section, were described. On the next step the fatty acids, the ordinary lipids, the complex and isoprene lipids were discused. After wards there are show the cosmetics, in which lipids were used. The results of the executed questionnaire, in which 48 participants in age from 20 to 50 years took part, are presented in the article. The questionaire contained 12 questions respecting adaptation of the cosmetics included lipids.

Natural and synthetic derivatives of benzo-γ-pyrones (i.e. flavones, chromones, and coumarins) and their synthetic analogues possess a wide range of biological properties in vitro and in vivo. In this paper we investigated the influence of two hydrazone compounds of chromones, 3-{[(2-dimethoxytiophosphoryl)-2-methylhydrazono]-methyl}-chromen-4-one (CH-3) and 2-amino-6-chloro-3-[(2-hydroxyethyl)-hydrazonomethyl]-chromen-4-one (A-12), on lipid peroxidation and bFGF concentration in the HL-60 cells. Both of the studied compounds had a significant influence on bFGF and TBARS in ranges -137.20 ~ 380.26% and -81.66 ~ -28.68%, respectively, in comparison with the control (counted as 0 %).

The objective of this study was to assess how probiotic bacteria added to diets of 60 hybrid barrows (Landrace–Yorkshire x Duroc) affected the basic chemical composition of meat, the fatty acid profile of intramuscular fat (IMF), m. longissimus lumborum (LL) and lipid quality indices, because IMF cannot be removed before consumption and this inevitably has an impact on human health. After 78 days, the pigs were divided into the control group (CT, n=30) and the experimental group (BP, n=30), which until 97 days of rearing were supplied with BioPlus YC probiotic (Bacillus licheniformis DSM 5749 and Bacillus subtilis DSM 5750). After slaughter, 12 carcasses of similar weight (90 ± 5 kg) from each group were selected for testing. Supplementation with probiotic Bacillus did not affect the basic chemical composition of meat. It was established that BioPlus YC had a significant impact on capric – C10:0 (0.08 vs. 0.13), eicosatrienoic – C20:3 n-3 (1.76 vs. 2.14) and ΣPUFA n- 3 acids (1.88 vs. 2.41), causing a reduction in their percentage, which was thereby different than in the CT group. Among lipid quality indices, IMF LL of pigs from the BP group, the thrombogenicity index (TI) was characterized by a significantly (P≤0.01) higher value than in the CT group (1.08 vs. 0.99). These results suggest that further research is needed to study differences in compounds affecting the fatty acid profile of pork, and to confirm the association of the lipid profile with the use of BioPlus YC probiotic in pig fattening.

Egg yolk and margarine are commonly applied in starchy food processing as a lipid source. Both may affect the starch properties to different extent. Therefore, the effects of adding egg yolk (0.3, 1.0, and 1.7 g/100 g of flour) and margarine (1, 3, and 5 g/100 g of flour) to banana flour on the complex formation of gelatinized starch–lipid and on paste functional properties were investigated in this study. Native banana flour was used as a control. The complexing index (CI) value increased with the increasing egg yolk or margarine content in the paste. Egg yolk lipids formed complexes with flour starch more favorably than margarine lipids. The heat-moisture treatment (HMT) of banana flour increased its ability to form a complex. Complex formation may partly affect the swelling volume (SV) and freeze-thaw stability (FTS) of pastes. The decrease in SV was smaller in the paste with egg yolk than with margarine. Egg yolk also led to a lesser syneresis and solubility than margarine. For freeze-thaw stability, the interplay roles between the reducing effect of lipid addition and the increasing effect of HMT led to an increased syneresis suggesting that HMT factor prevailed. The reducing effect of lipid addition on egg yolk complex solubility predominated the increasing impact of HMT. Lipid source did not affect pasting properties except set back viscosity. HMT-pretreatment played a major role in the alteration of pasting properties compared to the lipid content.

Objectives The aim of this study is the evaluation of the influence of repeated (5 times for 15 min) exposure to electromagnetic field (EMF) of 1800 MHz frequency on tissue lipid peroxidation (LPO) both in normal and inflammatory state, combined with analgesic treatment. Material and Methods The concentration of malondialdehyde (MDA) as the end-product of the lipid peroxidation (LPO) was estimated in blood, liver, kidneys, and brain of Wistar rats, both healthy and those with complete Freund’s adjuvant (CFA)-induced persistent paw inflammation. Results The slightly elevated levels of the MDA in blood, kidney, and brain were observed among healthy rats in electromagnetic field (EMF)-exposed groups, treated with tramadol (TRAM/EMF and exposed to the EMF). The malondialdehyde remained at the same level in the liver in all investigated groups: the control group (CON), the exposed group (EMF), treated with tramadol (TRAM) as well as exposed to and treated with tramadol (TRAM/EMF). In the group of animals treated with the complete Freund’s adjuvant (CFA) we also observed slightly increased values of the MDA in the case of the control group (CON) and the exposed groups (EMF and TRAM/EMF). The MDA values concerning kidneys remained at the same levels in the control, exposed, and not-exposed group treated with tramadol. Results for healthy rats and animals with inflammation did not differ significantly. Conclusions The electromagnetic field exposure (EMF), applied in the repeated manner together with opioid drug tramadol (TRAM), slightly enhanced lipid peroxidation level in brain, blood, and kidneys.

Introduction and objective. There is a significantly higher risk of lipid disorders occurrence, including atherogenic dyslipidemia in women after menopause than it is in general population. The aim of the work was to investigate the correlation between health behaviours and the occurrence of lipid disorders in women after menopause working in agriculture. Material and methods. The study was conducted in years 2015–2016 and included 843 post-menopausal women working in agriculture. The following were used: a questionnaire including socio-demographic data, laboratory lipid tests, inventory of health behaviours. The following were estimated: logistic regression models for serum lipids concentration versus frequency of health behaviours in the examined women. Results. Adverse lipid profile was found in over a half of post-menopausal women working in agriculture, whereas the frequency of health behaviours were estimated at the average level, although the frequency of correct eating habits and health practices was significantly lower than preventive behaviours and positive psychological attitudes. A correlation was found between the frequency of health behaviours and the occurrence of lipid disorders in women after menopause working in agriculture: more frequent health practices co-existed with the lower concentration of total cholesterol and a higher concentration of HDL-cholesterol, more frequent preventive behaviours co-existed with lower concentration of LDL-cholesterol. Women with higher concentration of triglycerides undertook pro-health practices relatively more often. Conclusions. The study revealed a high prevalence of lipid disorders in postmenopausal women working in agriculture. More effective health education programmes are necessary in the area of reduction the risk factors of CVD in the population of women working in agriculture.

EPR spectroscopy using 5-doxylstearic acid (5-SASL) and 16-doxylstearic acid (16-SASL) spin probes was used to study the fluidity of thylakoid membranes. These were isolated from wild type Synechocystis and from several mutants in genes encoding selected enzymes of the carotenoid biosynthesis pathway and/or acyl-lipid desaturases. Cyanobacteria were cultivated at 25°C and 35°C under different light regimes: photoautotrophically (PAG) and/or in light-activated heterotrophic conditions (LAHG). The relative fluidity of membranes was estimated from EPR spectra based on the empirical outermost splitting parameter in a temperature range from 15°C to 40°C. Our findings demonstrate that in native thylakoid membranes the elimination of xanthophylls decreased fluidity in the inner membrane region under optimal growth conditions (25°C) and increased it under sublethal heat stress (35°C). This indicated that the overall fluidity of native photosynthetic membranes in cyanobacteria may be influenced by the ratio of polar to non-polar carotenoid pools under different environmental conditions.

Introduction and objective. Cardiovascular diseases are the leading cause of death in Europe and worldwide. One of the most important risk factors for atherosclerosis are lipid metabolism disorders, in particular hipercholesterolaemia. The aim of the study was to determine the correlation between gut microbiota composition and atherosclerosis risk factors, so in order that it might be used as a biomarker for coronary artery disease diagnosis. Material and method. The study involved middle-aged men in eastern Poland with central obesity (n=20), subjects with atherosclerosis (n=15) and those with no cardiovascular diseases (n=5). The gut microbiota composition was determined using tag-encoded 16S rRNA gene using Illuminal MiSeq. Data were analyzed with the use of t-test. Results. Firmicutes (49.26%) and Bacteroidetes (44.46%) were the dominant Phyla in the middle-aged men in eastern Poland. Subjects with improper levels of total cholesterol were enriched in Prevotella (p=0.03) and decreased level of Clostridium (p=0.02). They also showed a falling tendency in Faecalibacterium (p=0.07). An upward trend was observed in Prevotella (p=0.07) in subjects with improper LDL-C values. Conclusions. The study showed that intestinal microbiome is likely to play a role in the pathogenesis of atherosclerosis through its role in lipid metabolism. Bacterial genera of particular importance were Prevotella, Bacteroides, Clostridium, Faecalibacterium. However, further studies involving larger groups of subjects are required to confirm these observations.

Method for evaluating lipid-binding qualities of enterosorbent was described. Mathod takes into account peculiarities of food digestion. A number of lipid binding agents was tested. The researches showed that biologically active additive to food "Chizitel" extracted via electrochemical method has the highest sorption properties towards triglycerides and fatty acids in comparison with well-known enterosorbents such as chitosan and microcrystalline cellulose (MCC) in the investigated PH-range.

Nanostructured lipid carriers (NLC) are stable colloidal formulations with notable advantages for drug delivery systems. Thanks to their physicochemical stability, biocompatibility, biodegradability and controlled drug release, they have received increasing attention for the last several years. The aim of the study was to prepare and characterize nanostructured lipid carriers (NLC). Both, the effect of the process parameters and the effect of the preemulsion composition on the NLC properties were investigated. In the work, different type of surfactants (i.e. decyl glucoside, Poloxamer188, Tween 80, sodium cholate) and their combinations were used to stabilize NLC dispersions. Moreover, several kinds of solid lipids (modified beeswax, gliceryl behenate, cetyl palmitate and berry wax) and liquid lipids (caprilic/capric triglyceride and decyl oleate) were applied. An ultrasonication method using a probe type sonicator was used to obtain NLC, and the time and energy of the process were modified throuhout. The physicochemical properties of the formulations, such as particle size, size distribution, polidispersity index were studied using the dynamic light scattering (DLS) method. The electrophoretic mobility of obtained particles was also measured, using the Zetasizer Nano ZS Malvern Instrument based on the Laser Doppler Velocimetry (LDV) technique. Knowing the value of electrophoretic mobility of particles for given conditions, the zeta potential was determined. The obtained results showed that the process parameters and the composition of the preemulsion had significant impact on the nanoparticles structure. The optimal formulations size ranged between 60 and 80 nm, and the value of their zeta potential was up to -30mV. The stability of these systems was further confirmed by macroscopic observation.

Model systems such as black lipid membranes or conventional uni- or multilamellar liposomes are commonly used to study membrane properties and structure. However, the construction and dimensions of these models excluded their direct optical microscopic observation. Since the introduction of the simple method of liposome electroformation in alternating electric field giant unilamellar vesicles (GUVs) have become an important model imitating biological membranes. Due to the average diameter of GUVs reaching up to 100 µm, they can be easily observed under a fluorescent or confocal microscope provided that the appropriate fluorescent probe was incorporated into the lipid phase during vesicle formation. GUVs can be formed from different lipid mixtures and they are stable in a wide range of physical conditions such as pH, pressure or temperature. This mini-review presents information about the methods of GUV production and their usage. Particularly, the use of GUVs in studying lipid phase separation and the appearance and behavior of lipid domains (rafts) in membranes is discussed but also other examples of GUVs use in membrane research are given. The experience of the authors in setting up the GUV-forming equipment and production of GUVs is also presented.

This work is focused on the function of the microtubule and actin networks in plasmid DNA transport during liposomal transfection. We observed strong binding of plasmid DNA-lipid complexes (lipoplexes) to both networks and directional long-range motion of these lipoplexes along the microtubules. Disruption of either of these networks led to the cessation of plasmid transport to the nucleus, a decreased mobility of plasmids, and accumulation of plasmid DNA in large aggregates at the cell periphery. Our findings show an indispensable but different role of both types of cytoskeleton, actin and microtubular, in the processes of gene delivery.

The prospect of depletion of natural energy resources on the Earth forces researchers to seek and explore new and alternative energy sources. Biomass is a composite resource that can be used in many ways leading to diversity of products. Therefore, microalgal biomass offers great potential. The main aim of this study is to find the best physical method of microalgal bio- mass pretreatment that guarantees efficient lipid extraction. These studies identifies biochemical composition of microalgal biomass as source for biodisel production. The influence of drying at dif- ferent temperatures and lyophilization was investigated. In addi- tion, wet and untreated biomass was examined. Cell disruption (sonication and microwave) techniques were used to improve lipid extraction from wet biomass. Additionally, two different extrac- tion methods were carried out to select the best method of crude oil extraction. The results of this study show that wet biomass after sonication is the most suitable for extraction. The fatty acid com- position of microalgal biomass includes linoleic acid (C18:2), palmitic acid (C16:0), oleic acid (C18:1), linolenic acid (C18:3), and stearic acid (C18:0), which play a key role in biodiesel production.

Introduction. Verapamil is a widely used cardiac drug. It belongs to the non-dihydropiridine subgroup of calcium channel blockers (CCBs), and is used mainly for the treatment of atrial dysrhythmias, hypertension or angina. In this article, we describe a case of poisoning with verapamil mixed with ethanol and review current treatment options. A 54-year-old female was admitted to the Toxicology Clinic by emergency medical services after ingesting sustained-release CCBs and ethanol in a suicidal attempt. The patient was successfully treated by administration of catecholamines and calcium, and the introduction of a high-dose insulin infusion (HDI). Conclusions. This case report emphasizes that even in severe poisoning, early and proper intervention may prevent circulatory breakdown. It also summarizes treatment options in patients after verapamil overdose. Along with decontamination, administration of catecholamines and correcting electrolyte abnormalities, the main options are HDI and lipid emulsion therapy (LET).

The symptoms of oxidative stress and antioxidative response were investigated on Norway spruce seedlings subjected to freezing desiccation conditions. Three-year-old seedlings were exposed to freezing desiccation at -3oC and -10oC for 45 days in two acclimation stages: autumn (October) and winter (January). The stress enhanced the production of reactive oxygen species (ROS): superoxide radical anion (O2.-), and hydrogen peroxide (H2O2). Concentrations of low molecular antioxidants: glutathione (GSH), ascorbic acid (AsA) and a-tocopherol declined at both low temperatures and acclimation stages. The activity of superoxide dismutase (SOD) increased with ROS production, while guaiacol peroxidase (POX) activity decreased. The freeze-induced desiccation of needles was significantly correlated with the cold hardiness (LT50), the level of low-molecular antioxidants, and POX activity, but not with SOD activity. Under extreme freezing desiccation conditions, these reactions continued, leading to the degradation of membrane phospholipids and a strong decrease in cold hardiness. The results show that membranes are the primary site of injury induced by ROS, produced under the influence of low temperature combined with dehydration. The acclimation response of Norway spruce needles to the oxidative stress generated by long-term cold and/or freezing desiccation is discussed.

Background. The lifestyle of young boys has impact on the risks of cardiovascular diseases. Objective. The aim of the study was to evaluate the effect of atherosclerosis risk factors determined by overweight and obesity and lifestyle, i.e.: eating habits, low physical activity and smoking cigarettes, on blood lipid profile of boys at the age of 16 to 18. Material and Methods. The study covered 369 boys from secondary schools. They were evaluated for the supply of dietary constituents with atherogenic and protective actions, for nutritional status acc. to Cole’s criteria, the level of physical activity, and smoking cigarettes. Lipid metabolism was determined based on criteria recommended by the American National Cholesterol Education Program (NCEP). Logistic regression analysis was conducted and risk odds ratio [OR] was determined. Results. Analyses showed the boys to be characterized by overweight (10.8%) and obesity (2.7%), and by inappropriate concentration of total cholesterol (26.5%), LDL (13.3%), HDL (21.7%) and triglycerides (41.7%). High BMI turned out to be a significant risk factor of an elevated total cholesterol concentration: [aOR]=2.27; triglycerides: [aOR]=2.35 and LDL: [aOR]=2.41. Low physical activity was found to negatively affect the concentration of LDL: [aOR]=1.88. The boys smoking cigarettes were shown to have a reduced HDL: [aOR]=1.65. The total content of fat and saturated fatty acids in diet exerted a significantly negative impact on blood lipid profile of the boys. Conclusions. The lifestyle of the young boys was demonstrated to determine the risk of cardiovascular diseases. Overweight and obesity, abdominal obesity in particular, were found to be a significant risk factor of disorders in their lipid metabolism.

Introduction. Lipid is an essential component of the cell and its organelles membrane. The uniqueness and selectivity of lipids to specific functions and asymmetry of lipid distribution in the organelle’s membrane give the cell ability of being highly qualified and specified. Aim. The paper provides a comprehensive review of membrane lipids in different tissues and organelles of the cell in norm and disease. Material and methods. The paper analyzed the present literature data on membrane lipids behavior in physiology and pathology. Analysis of the literature. The major structural and functional lipids of the cell membrane are phosphatidylcholine > phosphatidylethanolamine. The absence/deficiency or augmentation of a specific type of lipid results in serious defects and usually life-threatening with a permanent disability. The observations discussed here suggest, the lipid peroxidation severity depends on the membrane lipid composition of the cell. Some tissue cells can handle lipoperoxidation and protect themselves from the peroxidation damaging products better, while other cells cannot compensate. Therefore, some organs are highly sensitive to peroxidation and irreversible changes occur rapidly. Conclusion. To sum up, the understanding of lipid’s role in norm and disease is clinically crucial to evaluate a novel therapeutic target to treat many metabolic disorders such as metabolic syndrome and some lysosomal storage disorders via targeting specific new signaling pathways, lipid molecules, and enzymes

It was shown that in membranes containing raft domains, the macular xanthophylls lutein and zeaxanthin are not distributed uniformly, but are excluded from saturated raft domains and about ten times more concentrated in unsaturated bulk lipids. The selective accumulation of lutein and zeaxanthin in direct proximity to unsaturated lipids, which are especially susceptible to lipid peroxidation, could be very important as far as their antioxidant activity is concerned. Therefore, the protective role of lutein against lipid peroxidation was investigated in membranes made of raft-forming mixtures and in models of photoreceptor outer segment membranes and compared with their antioxidant activity in homogeneous membranes composed of unsaturated lipids. Lipid peroxidation was induced by photosensitized reactions using rose Bengal and monitored by an MDA-TBA test, an iodometric assay, and oxygen consumption (using EPR spectroscopy and the mHCTPO spin label as an oxygen probe). The results show that lutein protects unsaturated lipids more effectively in membranes made of raft-forming mixtures than in homogeneous membranes. This suggests that the selective accumulation of macular xanthophylls in the most vulnerable regions of photoreceptor membranes may play an important role in enhancing their antioxidant properties and ability to prevent age-related macular diseases (such as age-related macular degeneration (AMD)).

Infrared (IR)-A irradiation can be useful in back and musculoskeletal pain therapy. In this study joint and vertebral column pain and mobility were measured during two weeks of IR-A irradiation treatment of patients suffering from degenerative osteoarthritis of hip and knee, low back pain, or rheumatoid arthritis. Additionally, before and after IR-A treatment MDA serum levels were measured to check if MDA variations accompany changes in pain intensity and mobility. Two-hundred and seven patients were divided into verum groups getting IR-irradiation, placebo groups getting visible, but not IR irradiation, and groups getting no irradiation. In osteoarthritis significant pain reduction according to Visual Analogue Scale and mobility improvements occurred in the verum group. Even though beneficial mean value changes occurred in the placebo group, the improvements in the placebo and No Irradiation groups were without statistical significance. In low back pain, pain and mobility improvements (by 35-40%) in the verum group were found, too. A delayed (2nd week) mobility improvement in rheumatoid arthritis was seen. However, pain relief was seen immediately. In patients suffering from low back pain or rheumatoid arthritis, the pain and mobility improvements were accompanied by significant changes of MDA serum levels. However, MDA appears not a sensitive biofactor for changes of the pain intensity in degenerative osteoarthritis. Nevertheless, unaffected or lowered MDA levels during intensive IR-A therapy argue against previous reports on free radical formation upon infrared. In conclusion, rapid beneficial effects of IR-A towards musculoskeletal pain and joint mobility loss were demonstrated.

Penetration of the liposome membranes doped with vanadium complex formed in the liquid-crystalline phase from egg yolk lecithin (EYL) by the TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl) spin probes has been investigated. The penetration process was followed by 360 hours at 24◦C, using the electron spin resonance (EPR) method. The spectroscopic parameter of the partition (F) of this probe indicated that a maximum rigidity of the membrane was at 3% concentration of the vanadium complex. Computer simulations showed that the increase in the rigidity of the membrane corresponds to the closure of gaps in the surface layer of the membrane, and indicates the essential role of the membrane surface in transport processes.

Purpose:To examine lipid peroxidation, antioxidant protection and L-arginine-NO – system in patients with papulo-pustular rosacea.Materials and methods:The study included 128 women with papulo-pustular rosacea aged 40.1±0.99 years (range: 18-68 years). The patients were divided into three groups based on the severity of the symptoms: group I –patients with mild rosacea (n=42), group II – patients with moderate rosacea (n=49), group III – patients with severe rosacea (n=37). Indicators of lipid peroxidation and antioxidant protection were defined in all patients by a spectrophotometric method.Results:The first group of patients showed a significant decrease in superoxide dismutase (SOD) and ceruloplasmin when compared to the control group. A significant increase in diene conjugates (DC), malondialdehyde (MDA), liposoluble antioxidants (retinol, α-tocopherol) and decrease in SOD, catalase and ceruloplasmin were observed in the second group. Patients in the third group had similar dynamics with a worsening of lipid peroxidation.Conclusions: The changes in some parameters of lipid peroxidation and antioxidant protection were revealed in patients with papulo-pustular rosacea. The nature of these changes depends on the severity of the disease. Evaluation of the antioxidant imbalance may be informative to determine the understanding of the genesis of dermatosis and to study therapeutic strategy aimed at reducing the generation of reactive oxygen species (ROS), leading to a decrease in the capacity of the antioxidant defense.

Opis:
Effectiveness of green tea (compared to two single polyphenols) in removing ozone derived reactive oxygen species acting on dioleoylphosphatidylcholine monolayers was determined. Lipid oxidation was followed by changes in mechanical properties of the layer spread on the aqueous subphase containing various amounts of ozone in the presence and absence of polyphenolics. It was shown that the tea extract (containing 8.5 x 10-4 mg/cm3 polyphenols) is capable of inactivating 0.4 ppm ozone. The DPPH radical scavenging test set polyphenols in the order of increasing activity, consistent with their protective effect in relation to lipid oxidation, showing the highest efficacy of EGCG.

The objective of this study was to assess the dynamics of oxidative damage to cellular macromolecules such as proteins, lipids and DNA under conditions of oxidative stress triggering early stages of estrogen-dependent carcinogenesis. A rodent model of carcinogenesis was used. Syrian hamsters were sacrificed after 1, 3, 5 h and one month from the initial implantation of estradiol. Matching control groups were used. Kidneys as target organs for estradiol-mediated oxidative stress were excised and homogenized for biochemical assays. Subcellular fractions were isolated. Carbonyl groups (as a marker of protein oxidation) and lipid hydroxyperoxides were assessed. DNA was isolated and 8-oxodGuo was assessed. Electron paramagnetic resonance spectroscopy was used to confirm the results for lipid peroxidation. Exposition to estradiol in the rodent model leads to damage of macromolecules of the cell, including proteins and DNA, but not lipids. Proteins appear to be the primary target of the damage but are closely followed by DNA. It has previously been speculated that protein peroxides can increase DNA modifications. This time sequence was observed in our study. Nevertheless, the direct relation between protein and DNA damage still remains unsolved.

Amphotericin B (AmB) is a well known polyene macrolide antibiotic used to treat systemic fungal infections. Despite its toxicity AmB is still regarded as a life-saving drug. The lack of adequate knowledge of the AmB mechanism of action is a serious obstacle to efficient development of new less toxic derivatives. Complementary to various experimental approaches, computational chemistry methods were used to study AmB mechanism of action. A programme lasting for a decade, that was run by our group covered studies of: i) molecular properties of AmB and its membrane targets, ii) structure and properties of AmB membrane channels, and iii) interaction of AmB with the membrane.

There is growing evidence that proteins are early targets of reactive oxygen species, and that the altered proteins can in turn damage other biomolecules. In this study, we measured the effects of proteins on the oxidation of liposome phospholipid membranes, and the formation of protein hydroperoxides in serum and in cultured cells exposed to radiation-generated hydroxyl free radicals. Lysozyme, which did not affect liposome stability, gave 50% protection when present at 0.3 mg/ml, and virtually completely prevented lipid oxidation at 10 mg/ml. When human blood serum was irradiated, lipids were oxidized only after the destruction of ascorbate. In contrast, peroxidation of proteins proceeded immediately. Protein hydroperoxides were also generated without a lag period in hybrid mouse myeloma cells, while at the same time no lipid peroxides formed. These results are consistent with the theory that, under physiological conditions, lipid membranes are likely to be effectively protected from randomly-generated hydroxyl radicals by proteins, and that protein peroxyl radicals and hydroperoxides may constitute an important hazard to biological systems under oxidative stress.

Opis:
The effect of marine fish fat in different stages of oxidation on biochemical and morphological changes in guinea pigs was studied. Oxidized fat was found to adversely affect lipids balance in the organism and reduce the content of antioxidant vitamins (C and E).
Celem badań było wyjaśnienie wpływu tłuszczu ryb morskich o różnym stopniu utlenienia na organizm świnek morskich w warunkach spożywania aterogennej diety. Chodziło m.in. o uzyskanie odpowiedzi czy spożywanie ryb morskich zawierających tłuszcz utleniony może korzystnie wpływać na gospodarkę lipidową ustroju w warunkach hipercholesterolemii. Badano także zawartość w ustroju witamin antyoksydacyjnych. Doświadczenie przeprowadzono na świnkach morskich samcach, którym podawano przez 12 tygodni określone diety. Grupa kontrolna I otrzymywała granulowaną dietę hodowlaną LSK firmy „Bacutil", grupa kontrolna II diety j. w. z dodatkiem 0,1% cholesterolu. Grupom badanym podawano ryby morskie z dodatkiem 0,1% cholesterolu – grupie III rybę świeżą, grupie IV rybę z tłuszczem utlenionym. Ogólna zawartość tłuszczu w dietach doświadczalnych wynosiła ok. 14% kcal. Wyniki badań masy ciała i przyrostów masy ciała wykazały, że dodatek cholestrolu do diety hodowlanej miał ujemny wpływ na ustrój zwierząt, natomiast dodatek do diety ryby świeżej nie wpływał na tempo wzrostu zwierząt w porównaniu z grupą kontrolną II. Najmniejszą masę ciała i przyrosty masy ciała stwierdzono w grupie otrzymującej rybę z tłuszczem utlenionym. W grupach doświadczalnych, tj. otrzymujących badane ryby nastąpiło powiększenie masy wątroby. W tych grupach stwierdzono obniżenie wartości hemoglobiny i hematokrytu. Dodatek do diety hodowlanej cholesterolu (dieta kontrolna II) spowodował istotny wzrost cholesterolu całkowitego w surowicy krwi, natomiast w grupie zwierząt otrzymujących rybę świeżą nastąpił spadek zawartości cholesterolu w porównaniu z grupą kontrolną II a w grupie której podawano rybę z tłuszczem utlenionym - wzrost zawartości cholesterolu. Zawartość cholesterolu w LDL była najniższa w grupie kontrolnej I a najwyższa w grupie otrzymującej rybę z tłuszczem utlenionym. Zawartość cholesterolu w HDL był a najniższa w grupie kontrolnej I. W grupie otrzymującej rybę świeżą zawartość cholesterolu w HDL była wyższa niż w grupie kontrolnej I i II a najwyższa w grupie której podawano rybę z tłuszczem utlenionym. Zawartość nadtlenków lipidowych w surowicy krwi była najwyższa w grupie, której podawano rybę z tłuszczem utlenionym, najniższa w grupie kontrolnej I. Stwierdzono również, że w grupie zwierząt otrzymujących w diecie rybę z tłuszczem utlenionym obniżyła się zawartość tokoferolu i kwasu askorbinowego w surowicy krwi oraz zawartość kwasu askorbinowego w nadnerczach.

Background. Overweight and obesity are defined as abnormal or excessive fat accumulation that may impair health. Obesity is associated with many chronic diseases, including cardiovascular disease and diabetes, and recently the role of overweight and obesity in lung disease has received new interest. Chronic obstructive lung disease is the third-leading cause of death globally, and both obesity and diet appear to play roles in its pathophysiology. Cross-sectional studies have demonstrated an inverse association between obesity and the prevalence of chronic obstructive pulmonary disease (COPD). Objective. This study aims to evaluate the relationship between smoking, lipid profile and obesity (body composition changes) in a selected groups of population (30 non-smokers, 30 smokers and 60 COPD patients). Material and Methods. We evaluated fat mass, fat free mass, body mass index and lipid profile in a group of 120 randomly selected probands (60 COPD patients; 30 smokers without COPD; 30 non-smokers without COPD) to identify possible negative relationships of smoking to body composition. To the measurement of fat mass (FM) and fat free mass (FFM) was used a device Bodystat Quadscan 4000 (Bodystat Ltd, British Isles). The device works by using four-frequency bioelectrical impedance analysis. Laboratory parameters as total cholesterol (T-C), high-density cholesterol (HDL-C), low-density cholesterol (LDL-C) and triacylglycerols (TG) were investigated by automated clinical chemistry analyzer LISA 200th. The measured values were statistically processed and evaluated in a statistical program STATISTICA Cz. Version 7.1. (Kruskall-Wallis test). Results. A comparison of the mean fat mass we found statistically highly significant differences between the group of COPD patients and non-smokers (P <0.001) and insignificant differences (P ≥ 0.05) between the other groups of our experiment. A comparison of the mean fat mass values of all three groups of the experiment shows a steady increase in fat from non-smokers (17.66 ± 10.04 kg) to COPD patients with the highest mean value (25.08 ± 10.14 kg). In the group of COPD patients we recorded the lowest average value of FFM (51.76 ± 13.84 kg), in group of smokers the middle (56.06 ± 10.76 kg) and in non-smokers the highest average value of FFM (59.91 ± 9.90 kg) at relatively the same body weight in the groups. Based on calculated body mass index (BMI), we found in group of COPD patients overweight in 15 cases (25%), obesity in 7 patients (11.67%), severe obesity in 14 patients (23.3%) and morbid obesity in 2 patients (3.33%); in the group of smokers overweight in 16 cases (53.33%), obesity in 5 cases (16.6%) and severe obesity in 1 case (3.33%); in non-smokers we recording overweight in 14 cases (46.67%), obesity in 5 cases (16.67%) and severe obesity in 2 cases (6.67%). In the lipid profile of the monitored groups of probands, we observed statistically significant differences only for LDL cholesterol (LDL-C). There was a statistically significant difference (P <0.001) between the group with COPD and smokers, as well as between the group of smokers and non-smokers (P < 0.05). Conclusions. In the vast majority of patients with COPD, the lung damage that leads to COPD is caused by long-term cigarette smoking. The presence and absence of risk factors such as smoking, inappropriate lipid profile and obesity (amount of fat mass) in selected population groups were observed. Additional studies to explore both the quantitative and qualitative changes in body composition with disease process of COPD are required.

Background. The aim of this study was to investigate the possibility of using sodium selenite as a treatment for acute glyphosate poisoning using the activity of the lipid peroxidation and antioxidant defence systems as a readout for efficacy. Material and methods. Experimental glyphosate poisoning and subsequent treatment using sodium selenite was performed in albino rats (105). Glyphosate was given in doses of 50, 100 and 130 mg/kg, and sodium selenite was administered at a dose of 2 μg/kg. The blood concentrations of lipid peroxidation markers including conjugates of diene andtrienoic and malondialdehyde were determined. The endogenous glutathione (reduced form) level and activities of catalase, superoxide dismutase and glutathione peroxidase in the serum were measured. Results. Glyphosate poisoning has been found to result in a significant increase in lipid peroxidation activity. For example, malonic dialdehyde demonstrates a 2.35 times increase at a glyphosate dose of 130 mg/kg. At the experimental glyphosate poisoning dose of 100 mg/kg the measurements of superoxide dismutase and glutathione peroxidase have been found to decrease 1.58 and 2.21 times, respectively. At a dose of 130 mg/kg, those values decreased 2.51 and 4.76 times, respectively, compared to untreated controls. Conclusions. The use of sodium selenite at a dose of 2 μg/kg after poisoning of white rats with glyphosate (at doses of 50, 100 and 130 mg/kg) normalizes the lipid peroxidation and antioxidant defence activities of the body.
Wprowadzenie. Celem niniejszej pracy było zbadanie możliwości zastosowania seleninu sodu w leczeniu ostrego zatrucia glifosatem wykorzystując aktywność peroksydacji lipidów oraz antyoksydacyjnych układów ochronnych jako wskaźnik skuteczności. Materiał i metody. 105 szczurów albinosów poddano eksperymentalnemu zatruciu glifosatem i leczeniu seleninem sodu. Glifosat został zastosowany w dawkach 50, 100 i 130 mg/kg. Selenin sodu podawano w dawce 2 μg/kg. Oznaczono stężenie markerów peroksydacji lipidów we krwi: koniugatów dien i trienoicznych oraz dwualdehydu maleinowego. Zbadano poziom endogennego glutationu (w zredukowanej formie) oraz aktywność katalazy, dysmutazy ponadtlenkowej i peroksydazy glutationowej w surowicy krwi. Wyniki. Stwierdzono, że zatrucie glifosatem powoduje znaczny wzrost aktywności peroksydacji lipidów. Przykładowo, poziom dwualdehydu maleinowego wzrósł 2,35 razy w przypadku dawki glifosatu wynoszącej 130 mg/kg. Stwierdzono również, że wskaźniki dysmutazy ponadtlenkowej i peroksydazy glutationowej zmniejszają się odpowiednio 1,58 i 2,21 w eksperymentalnym zatruciu glifosatem w dawce 100 mg/kg. Zaobserwowane wartości zmniejszyły się 2,51 i 4,76 razy podczas zastosowania dawki 130 mg/kg w porównaniu z grupą kontrolną zdrowych zwierząt. Wnioski. Zastosowanie seleninu sodu w dawce 2 μg/kg, aby przeciwdziałać zatruciu glifosatem w dawkach 50, 100 i 130 mg/kg u szczurów albinosów, normalizuje peroksydację lipidów i czynności obrony antyoksydacyjnej organizmu.

Oleic acid (OA) is widely used in pathology studies of hepatocellular lipid deposition. Identifying the effects of different solvents on OA-induced liver lipid deposition would be beneficial for studies on hepatocytes. We treated BRL 3A cells with OA dissolved in different solvents. After 12 h incubation, cell viability was assessed using MTT assays. Reactive oxygen species (ROS), triglyceride (TG) and total cholesterol (TC) counts, and the expression level of glucose regulated protein (GRP78), sterol regulatory element binding protein (SREBP-1C) and fatty acid synthase (FAS) were analyzed. Water, PBS and DMSO were disadvantageous to the dissolution of OA and did not cause an OA-induced response in hepatocytes. In the alcohol+OA-treated cells, the severe ER stress, oxidative stress and cellular fat deposition were significantly increased. BSA promoted cell growth and the cells treated with 1.2% BSA+OA showed a lower grade TG and endoplasmic reticulum stress compared with KOH+OA and alcohol+OA treatments. KOH had no significant influence on BRL 3A cells viability. When treated with OA dissolved in KOH, BRL 3A cells showed a typical hepatocyte damage. KOH was considered the suitable choice for an OA solvent for BRL 3A cells in hepatic lipidosis research.

Particles generated from numerous anthropogenic and/or natural sources, such as crystalline α-Fe2O3 nanoparticles, have the potential to damage lung cells. In our study we investigated the effects of these nanoparticles (12.5 µg/ml) on lipid peroxidation and the antioxidative system in MRC-5 lung fibroblast cells following exposure for 24, 48 or 72h. Exposure to α-Fe2O3 nanoparticles increased lipid peroxidation by 81%, 189% and 110% after 24, 48 and 72h, respectively. Conversely, the reduced glutathione concentration decreased by 23.2% and 51.4% after 48 and 72h of treatment, respectively. In addition, an augmentation of the activities of superoxide dismutase, catalase, glutathione peroxidase, glutathione transferase and glutathione reductase within the interval between 48-72h was noticed. Taking into account that the reduced glutathione level decreased and the malondialdehyde level, a lipid peroxidation product, remained highly increased up to 72h of exposure, it would appear that the MRC-5 antioxidant defense mechanisms did not efficiently counteract the oxidative stress induced by exposure to hematite nanoparticles.

Opis:
Bone regeneration is a process of vital importance since fractures of long bones and large joints have a highly deleterious impact on both, individuals and society. Numerous attempts have been undertaken to alleviate this severe medical and social problem by development of novel bioactive materials, among which bioactive glass is the most attractive because of its osteoconductive and osteostimulative properties. Since lipid peroxidation is an important component of systematic stress response in patients with traumatic brain injuries and bone fractures, studies have been undertaken of the molecular mechanisms of the involvement of 4-hydroxynonenal (HNE), an end product of lipid peroxidation, in cellular growth regulation. We found that HNE generated in bone cells grown in vitro on the surfaces of bioactive glasses 45S5 and 13-93. This raises an interesting possibility of combined action of HNE and ionic bioglass dissolution products in enhanced osteogenesis probably through a mitogen-activated protein kinase (MAPK) pathway. While the proposed mechanism still has to be elucidated, the finding of HNE generation on bioglass offers a new interpretation of the osteoinducting mechanisms of bioglass and suggests the possibility of tissue engineering based on manipulations of oxidative homeostasis.

In this work was investigated the effect of pre-treatment with (PhSe)2 and (PhTe)2 on chemical seizure and 4-aminopyridine-induced lethality in mice. Additionally, lipid peroxidation levels of whole brain after treatment with 4-aminopyridine and effect of pre-treatment with (PhSe)2 and (PhTe)2 on these levels were investigated. Mice were pre-treated with (PhSe)2 or (PhTe)2 (50, 100, or 150 µmol/kg) 30 min before 4-aminopyridine (12 mg/kg) administration. The treatment with 4-aminopyridine caused a significant incidence of seizures (clonic, tonic) and death. Pre-treatment with (PhSe)2 and (PhTe)2 significantly increased the latency for clonic and tonic seizures, and prevented 4-aminopyridine-induced death. Significantly, the pre-treatment with (PhSe)2 or (PhTe)2 increased the latency for clonic seizures in a dose-dependent manner. Additionally, a significant increase was observed in the brain lipid peroxidation level after treatment with 4-aminopyridine, which was significantly inhibited by pre-treatment with 150 µmol/kg (PhSe)2 or (PhTe)2. These results demonstrate that (PhSe)2 and (PhTe)2 counteract the harmful effects of 4-aminopyridine. It is possible that this effect results from modulation of the redox state of N-methyl-d-aspartate receptors and/or of Ca2+ channel activity with subsequent alteration in neurotransmitter release. Importantly, this study provides evidence for anticonvulsant and antioxidant properties of (PhSe)2 and (PhTe)2, which indicates a neuroprotective activity of these compounds.

The effect of extrusion on the chemical changes and physico-chemical properties of proteins and starch of buckwheat flour and its mixture with milk proteins was examined. It was found that the temperature of the process has an influence on the formatiron of starch-protein and starch-lipid complexes. The solubility and swelling power of starch was also changed.
Ekstrudowanie mąki gryczanej lub jej mieszaniny z białkami mleka powoduje wiele zmian chemicznych i właściwości fizykochemicznych białek i skrobi. Nie stwierdza się obniżenia zawartości azotu ogólnego w produktach ekstrudowanych, natomiast obserwuje się wyraźne zwiększenie zawartości związków azotowych nierozpuszczalnych. Kolejnym dowodem zmian właściwości fizykochemicznych białek jest zmniejszenie się pola histerezy wyznaczonego w wyniku miareczkowania kwasem i zasadą zawiesiny białek produktu ekstrudowanego z mąki gryczanej. W przypadku zastosowania jako surowca mieszaniny mąki gryczanej z 25% dodatkiem białek mleka pola histerezy miareczkowania zwiększa się. Również wyniki rozdziału chromatograficznego na żelu Sephadex białek produktów ekstrudowanych wskazują na termolabilność białek gryki i możliwość ich agregacji z białkami mleka. Skrobia badanych produktów ekstrudowanych charakteryzuje się zmienionym składem chemicznym i właściwościami fizykochemicznymi w porównaniu ze skrobią ziarna gryki. W składzie chemicznym skrobi produktów ekstrudowanych zwiększa się zawartość związków azotowych, tłuszczowców wolnych i związanych oraz popiołu. W,skazuje to na możliwość tworzenia się kompleksów skrobiowa-białkowych i skrobiowo-tłuszczowych, które mają duże znaczenie w tworzeniu struktury i jej trwałości. W zakresie właściwości fizykochemicznych skrobi stwierdza się wysoki jej stopień kleikowania, zwiększanie się rozpuszczalności a obniżanie zdolności pęcznienia.

Problem toksyczności tlenu i jego wpływu na układ nerwowy jest zagadnieniem ważnym wobec stosowania tlenu i mieszanin oddechowych w praktyce nurkowej., oraz w świetle uderzającego synergizmu działania tlenu i promieniowania jonizującego. Badania poziomu nadtlenków lipidów wykonywano w mózgu szczurów. Zwierzęta poddawano wyczerpującemu wysiłkowi w komorze ciśnieniowej pod działaniem tlenu przy ciśnieniu od 0-3 atn w okresie 2560 minut. Równolegle przeprowadzano badania na zwierzętach znajdujących się w spoczynku. Po wypreparowaniu mózg homogenizowano i oznaczono nadtlenki lipidów metodą Wollmana z TBA. U zwierząt obciążonych wysiłkiem fizycznym w podanych przedziałach czasu, nie stwierdzono odchyleń w poziomie nadtlenków lipidów, w porównaniu z grupą kontrolną. Wzrost poziomu nadtlenków lipidów stwierdzono u szczurów u których wystąpiły objawy zatrucia tlenowego. Na podstawie powyższych wyników autorzy przypuszczają, że wzrost nadtlenków lipidów mózgu w przypadkach hiperbarii tlenowej należy uznać za daleko posunięty szkodliwy efekt działania tlenu, następujący po uszkodzeniu enzymatycznym oraz zachwianiu obrony antyoksydatywnej komórek. Przy niskich nadciśnieniach tlenu nie stwierdzono odchyleń w poziomie nadtlenków lipidów, w porównaniu z grupą kontrolną.
The problem of oxygen toxicity, and its effect on the nervous system, is an important topic with regard to the application of oxygen and breathing mixes in the pursuit of diving, as well as in the light of the striking synergy between the effects of oxygen and ionising radiation. Studies on the level of lipid peroxides were performed on rat brains. The animals were subjected to exhaustive physical strain in a pressure chamber and oxygen at the pressure between 0-3 atm for the period of 25-60 minutes. Parallel research was conducted on rested animals. Following the dissection, the brain was homogenised and the levels of lipid peroxides were determined using the Wollman method with TBA. In animals subjected to physical effort over the specified time, no deviations in the levels of lipid peroxides were observed in comparison to the control group. An increase in lipid peroxide level was noted in rats manifesting oxygen toxicity symptoms. On the basis of the above findings, the authors presume that the growth of lipid peroxides in the brain in cases subjected to hyperbaric oxygenation should be recognised as a far-reaching harmful effect of oxygen, occurring after enzymatic damage and the violation of cellular antioxidant protection. At low oxygen overpressures, no deviations in the levels of lipid peroxides were noted as compared to the control group.

Electrolyte efflux, chlorophyll, malondialdehyde content and enzymatic activities (catalase, peroxidase) in cucumber cotyledons as well as stability of model lipid membranes in the presence of some new, acyclic and cyclic, aminophosphonates were studied. They also differed in the substituents at the phosphorus and nitrogen atoms. It was found that the effectiveness of the compounds to influence mentioned parameters depended on their lipophilicity and structural features. The most active compounds were these with isopropyl substituents at the P atom unless their hydrocarbon chains at the N atom were not too long. Butyl substituents at phosphorus significantly decreased aminophosphonates efficiency. Similar effect was observed when trans-type hydrocarbon chain was replaced with a short-branched one. It also seems that cyclic compounds with hexane rings were weaker modifiers of model and biological membranes than these with pentane rings. Results show that aminophosphonates interact with plasma membranes and presumably induce oxidative stress.
Badano wpływ nowych cyklicznych i acyklicznych aminofosfonianów na zawartość chlorofilu, dialdehydu malonowego oraz aktywność enzymów (katalaza, peroksydaza) w liścieniach ogórka. Określono efekt działania związków na stabilność modelowych błon lipidowych i wypływ elektrolitów z tkanki roślinnej. Badane aminofosfoniany różniły się ponadto podstawnikami przy atomie fosforu i azotu. Stwierdzono, że efektywność działania związków zależała od ich lipofilowości i kształtu cząsteczki. Najbardziej aktywne aminofosfoniany zawierały podstawnik izopropylowy przy atomie fosforu oraz niezbyt długi łańcuch węglowodorowy przy atomie azotu. Podstawnik butylowy przy atomie fosforu istotnie obniżał efektywność działania aminofosfonianów. Wydaje się, że cykliczne związki z pierścieniem cykloheksanowym w słabszym stopniu wpływają na membrany biologiczne i modelowe w porównaniu do związków zawierających cyklopentan. Wyniki badań wskazują, że aminofosfoniany oddziałują na membrany plazmatyczne i przypuszczalnie indukują stres oksydacyjny.

Objectives The increasing number of devices emitting electromagnetic radiation (EMR) in people’s everyday life attracted the attention of researchers because of possible adverse effects of this factor on living organisms. One of the EMR effect may be peroxidation of lipid membranes formed as a result of free radical process. The article presents the results of in vitro studies aimed at identifying changes in malondialdehyde (MDA) concentration – a marker of lipid peroxidation and antioxidant role of vitamin A during the exposure of blood platelets to electromagnetic radiation generated by liquid-crystal-display (LCD) monitors. Material and Methods Electromagnetic radiation emitted by LCD monitors is characterized by parameters: 1 kHz frequency and 220 V/m intensity (15 cm from display screen). The time of exposure was 30 and 60 min. The study was conducted on porcine blood platelets. The samples were divided into 6 groups: unexposed to radiation, unexposed + vitamin A, exposed for 30 min, exposed for 30 min + vitamin A, exposed for 60 min, exposed for 60 min + vitamin A. Results The MDA concentration in blood platelets increases significantly as compared to control values after 60 min of exposure to EMR. A significant decrease in MDA concentration after the addition of vitamin A was noticed. In the blood samples exposed to EMR for 30 and 60 min the MDA concentration was significantly increased by addition of vitamin A. Conclusions The results show the possibly negative effect of electromagnetic radiation on the cellular membrane structure manifested by changes in malondialdehyde concentration and indicate a possible protective role of vitamin A in this process. Objectives: Cardiovascular — coronary heart disease, causing heart attack and heart failure is a leading cause of disability and death worldwide. The most important risk factors for CVD are well-established and physical activity has long been considered the cornerstone of interventions and has shown extremely important in reducing the burden of this disease. The aim of this study was to examine the association between activity and inactivity and non-lipid CVD risk factors in healthy military recruits. Materials and Methods: Selected hormones and variables such as testosterone, free testosterone, Vitamin D, homocysteine, folic acid, with the inflammatory cytokines such as hsCRP, TNF-α and IL-6 were assessed in two groups (active = 50, inactive = 50), randomly matched by a questionnaire. Relationships between some measured variables and CVD risk factors were determined using simple correlation procedures. Results: Physically active subjects had signifi cantly lower results of homocysteine (14.3±1.9 vs. 16.5±3.7, p < 0.000) and interleukin-6 (2.9.±5.3 vs. 6±8.2, p < 0.025) and tended to have a nearly signifi cantly higher free testosterone level (13.1±7.1 vs. 11±5.3, p = 0.08). Signifi cant correlations were found between BMI, WC, tHcy, and the corresponding risk factors; and also between hsCRP and IL-6, as well as TT and FT. Conclusions: Reducing the risk factors of developing CVD events such as homocysteine and interleukin-6, together with increasing free testosterone and signifi cantly preventing its progression, clearly serve to underscore the benefi cial properties of physical activity and to promote its effectiveness as a support means for healthier lifestyles in the community and, particularly, among military personnel.

Oleje roślinne tłoczone na zimno należą do najbardziej stabilnych przechowalniczo, jednak również podlegają procesom obniżającym ich jakość podczas przechowywania. Kinetyka tych zmian uzależniona jest od wielu czynników, między innymi od temperatury przechowywania. Ostatnio obserwuje się wzrost podaży oraz urozmaicenie asortymentu olejów roślinnych w nowe, dotąd niewystępujące na polskim rynku produkty. Takimi olejami są na przykład oleje wytwarzane z orzechów. Ze względu na to, że najczęstszą przyczyną pogorszenia jakości olejów są procesy utleniania, przeprowadzono badania procesów utleniania podczas przechowywania trzech olejów z orzechów w temperaturach 4oC i 20oC. Materiał badawczy przechowywano 6 tygodni. W wyniku przeprowadzonych badań stwierdzono, iż w temperaturze 4oC zmiany zachodziły wolniej, natomiast najbardziej podatny na zmiany oksydacyjne był olej z orzechów włoskich.
Vegetable oils cold pressed are among the most stable during storage, but also subjected to the processes of lowering their quality during storage. The kinetics of these changes depend on many factors, including temperature storage. Recently, there is an increase in supply and more diversified range of vegetable oils in new, previously not present on the Polish market products. Such oils are, for example, oils prepared from the nuts. Due to the fact that the most common cause of deterioration of oils is oxidation, studies on oxidation during storage of three nut oils at temperatures 4oC and 20oC were carried out. The research material was stored 6 weeks. The results showed that at 4oC changes occurred more slowly, while the most susceptible to oxidative changes was walnut oil.

Alternatywnym sposobem zwiększenia biernej przepuszczalności błon biologicznych dla cząsteczek biologicznie aktywnych, w tym leków, jest elektroporacja. Pod wpływem pola elektrycznego o odpowiednich parametrach w fazie lipidowej błon biologicznych pojawiają się pory. Podatność błony na elektroporację zależy od wielu czynników, w tym otoczenia komórki oraz od stanu lipidów tworzących błony. Wyniki badań doświadczalnych oraz teoretycznych dotyczące wpływu wybranych produktów utlenienia lipidów na podatność błony na elektroporację nie prowadzą do jednoznacznych konkluzji. Celem niniejszej pracy jest zbadanie wpływu wszystkich produktów utlenienia lecytyny jajecznej obecnych w utlenionej lecytynie na przebieg elektroporacji. Produkty utlenienia lecytyny zbadano techniką spektroskopii podczerwieni. Pomiary wartości parametrów elektrycznych i elektroporacyjnych utworzonych błon przeprowadzono za pomocą potencjostatu-galwanostatu w układzie 4-elektrodowym. Porównanie wyników badań z użyciem błon utworzonych z lecytyny utlenionej oraz nieutlenionej wykazało spadek wartości pojemności właściwej, wzrost rezystancji oraz stabilności błony zawierającej produkty utlenionej lecytyny. Uzyskane wyniki badań mogą być przydatne w optymalizacji protokołów elektroporacyjnych u pacjentów szczególnie narażonych na czynniki utleniające np. większą ekspozycję na UV.
An alternative way of increasing the passive permeability of biological membranes to biologically active molecules, including drugs, is electroporation. Electric field of the relevant parameters applied to biological membranes creates pores in lipid part of membranes. Membrane susceptibility to electroporation depends on many factors: external environment of the cell, as well as a condition of membrane lipids. The results of experimental and theoretical research on influence of selected products of lipid oxidation on membrane susceptibility to electroporation do not lead to clear conclusions. The aim of this study is to examine the impact of all the products of oxidation of egg lecithin on the electroporation process. The oxidation of lecithin was examined by infrared spectroscopy. Measurements of the electrical membrane characteristics and parameters of electroporation were performed using a potentiostat-galvanostat in a four-electrode system. Comparison of the results obtained in experiments performed on oxidized and unoxidized lecithin have revealed a decline in the value of the specific capacity, an increase in the resistance, and stability of the membrane containing the oxidized lipids. These results may be useful in optimizing protocols of electroporation for patients, particularly those exposed to oxidizing agents, e.g. exposure to UV.

Skeletal and craniocerebral injuries simulating and their combinations within the papillary layer of a kidney is leading to a significant shift of antioxidant-prooxidant ratio with prevailing Pro-oxidant mechanisms. These mechanisms are enlarging from the first to the seventh day of the posttraumatic period and significantly higher against the background of combined cranio-skeletal injuries on the 3rd and on the 7th day of post-traumatic period.

Phosphatidic acid (PA) is the simplest glycerophospholipid naturally occurring in living organisms, and even though its content among other cellular lipids is minor, it is drawing more and more attention due to its multiple biological functions. PA is a precursor for other phospholipids, acts as a lipid second messenger and, due to its structural properties, is also a modulator of membrane shape. Although much is known about interaction of PA with its effectors, the molecular mechanisms remain unresolved to a large degree. Throughout many of the well-characterized PA cellular sensors, no conserved binding domain can be recognized. Moreover, not much is known about the cellular dynamics of PA and how it is distributed among subcellular compartments. Remarkably, PA can play distinct roles within each of these compartments. For example, in the nucleus it behaves as a mitogen, influencing gene expression regulation, and in the Golgi membrane it plays a role in membrane trafficking. Here, we discuss how a biophysical experimental approach enabled PA behavior to be described in the context of a lipid bilayer and to what extent various physicochemical conditions may modulate the functional properties of this lipid. Understanding these aspects would help to unravel specific mechanisms of PA-driven membrane transformations and protein recruitment and thus would lead to a clearer picture of the biological role of PA.

Electrochemical impedance spectroscopy was used for the study of two-component lipid membranes. Phosphatidylcholine and ceramide were to be investigated, since they play an important biochemical role in cell membranes. The research on biolipid interaction was focused on quantitative description of processes that take part in a bilayer. Assumed models of interaction between amphiphilic molecules and the equilibria that take place there were described by mathematical equations for the studied system. The possibility of complex formation for two-component system forming bilayers was assumed that could explain the deviation from additivity rule. Equilibria were described by mathematical equations that were further verified experimentally. The determined values of parameters (stability constant, molecular area of complex, capacitance and conductance of the lipid membranes formed from molecules and complexes) were used for calculation of model curves. The comparison of model curves and experimental points verified the assumed model.

Background The photocopying occupation contributes to various health issues in workers. Elevated levels of oxidative stress have been found to be associated with respiratory problems in occupational workers. The intention of the current research is to evaluate the association between the pulmonary function parameters, a biomarker of oxidative stress, and the photocopying occupation in Lahore, Pakistan. Material and Methods The study was performed on a group of photocopying operators (N = 100) and a control group (N = 100). A structured questionnaire was developed to gather the demographic attributes. Lung function testing was done with the spirometer. The blood lipid peroxides level, as thiobarbituric acid reactive substances (TBARS), was assessed spectrophotometrically. Results A statistically significant reduction was found in the pulmonary function parameters as compared to the control group. In the study group, the prevalence of respiratory symptoms, including cough (31%), sneezing (18%), wheezing (9%) and breathing issues (19%), was high as compared to the control group. A significantly higher plasma TBARS concentration was observed in the photocopying workers. A negative correlation prevailed between FVC (−0.081), FVC% predicted (−0.038) and FEV1 (−0.043), and the duration of exposure. Conclusions The study showed that the occupational exposure could have a significant effect on the normal pulmonary function and the oxidative potential of well-being. The profound emissions in the vicinity of photocopying centers may be responsible for the variations in the pulmonary function parameters and oxidative stress. Med Pr. 2019;70(4):403–10

Objective: The aim of the present work was to establish whether or not prescribed medication is a usable risk indicator for work‑related ischaemic heart disease (IHD), in Denmark. Material and Methods: Weighted Spearman rank correlation coefficients (rho) were used to evaluate the agreement between Standardised Hazard Ratios (SHR) for hospital treatment or death due to IHD and SHR for purchase of prescriptions for medicine that may prevent IHD from (re)occurring, among socio-occupational and economic activities groups in Denmark. The SHR were based on a 10-year prospective follow-up of 2 million people in Danish national registers 1996–2005. Results: We found approximately 7 times more cases of medicine usage (N = 411 651) than we did for hospital treatment or death (N = 55 684). The correlations between the 2 types of SHR were strong (rho = 0.94 for the socio-occupational groups; rho = 0.74 for the economic activities groups). We observed, however, one markedly contradictive result; the industrial group entitled ‘general practitioner, dentists etc.’ was associated both with significantly high rates of medicine usage (SHR = 1.15, 95% CI: 1.12–1.19) and significantly low rates of hospital treatment or death due to IHD (SHR = 0.80, 95% CI: 0.71–0.91). Conclusion: Apart from a few caveats, the strong correlations obtained in the present study signify that purchase of a prescription for IHD-related medication is a usable risk indicator for IHD in the working population of Denmark. The usage of medicine data in addition to or instead of the use of death or hospital data in epidemiological studies on work-related IHD risk will bring about a tremendous increase in statistical power.

Synthetic antioxidants, such as butylated hydroxytoluene (BHT), butylated hydroxyanisole (BHA), and tert-butylhydroquinone (TBHQ), have been widely used for many years to stabilise lipids in foods. Nonetheless, their effect on the bioavailability and metabolism of lipids stabilised with them still remains unknown. To investigate this issue, in vitro digestion of a model high-fat product (20 g fat/100 g) without antioxidants and supplemented with BHT, BHA and TBHQ (100 mg/kg) was conducted, followed by studies using the Caco-2 and Hep G2 cell lines. BHT, BHA and TBHQ increased intestinal absorption of triacylglycerols and modified the structure of chylomicrons (CM). The addition of BHT and TBHQ inhibited apolipoprotein-IV (apoA-IV) synthesis. At the same time, smaller chylomicrons were secreted, but their amount was greater than in the model product without antioxidants. In contrast, BHA activated apoA-IV synthesis, resulting in the formation of fewer but very large CMs. Of concern was the significant decrease in apoA-IV in cells upon the use of BHT and TBHQ, which can lead to obesity due to the lack of the sensation of feeling full after eating high-fat foods stabilised with these antioxidants. Furthermore, the efficiency of CM remnant (CMR) uptake by hepatocytes was inversely correlated with their size. The large CMRs generated for samples with BHA were not absorbed by hepatocytes, which can lead to atherosclerosis. The results of our in vitro study shed new light on the role of synthetic phenolic antioxidants used in food technology as potential obesity and atherosclerosis triggers and suggested the need for further research that will clearly separate antioxidants that promote the development of these diseases from those, which do not do that.

The lipid profiles of the oyster Crassostrea gigas cultured in Khanh Hoa coast at three locations and harvested in different months throughout the year were compared. Seasonal and locational changes in total lipid (TL) content, phospholipid content, free fatty acid content, fatty acid (FA) composition, and thrombogenicity index were found. The total lipid content and n-3/n-6 ratio in oysters from Ninh Hoa were higher compared to those from Cam Lam and Van Ninh. For oyster from all three farming areas, the total lipid content was the lowest during the two spawning seasons (May and September). Lipids from C. gigas were separated into neutral and polar lipid fractions using silica gel column chromatography. The amount of neutral lipids recovered from the oyster muscle was significantly higher than that of the polar lipids. The FA composition showed that C. gigas was abundant in saturated fatty acids (30.89–39.16 g/100 g TL), followed by polyunsaturated fatty acids (28.13–35.88 g/100 g TL), and monounsaturated fatty acids (19.32–23.75 g/100 g TL). The dominant polyunsaturated fatty acids of oysters from the three farming areas were eicosapentaenoic acid (9.09–13.77 g/100 g TL) and docosahexaenoic acid (6.71–16.47 g/100 g TL). Based on the present findings, it can be concluded that the Pacific oyster C. gigas cultured in Khanh Hoa, Vietnam is a promising source of highly nutritious exploitable lipids.

Introduction and aim. Garlic is one of the most popular traditional medicinal herbs which has a number of desirable health benefits. The study was designed to depict the improvement of serum biochemical parameters as well as the histomorphological recovery potential of garlic aqueous extract in hypercholesterolemic mice. Material and methods. A total of thirty Swiss albino mice weighing 24±5g and aged 5 weeks were randomly divided into three groups. Group A: supplied standard mice pellet and water; Group B: standard mice pellet + hypercholesterolemic diet (cow brain: 2 g/kg b.w.t.); and Group C: standard mice pellet + hypercholesterolemic diet (cow brain: 2 g/kg b.w.t.) + garlic extract (25 ml/kg b.w.t.). After four weeks of experimental tenure, samples (blood, liver, and kidney) were collected from each group of mice for serum biochemical analysis and histomorphological study. Results. Compared with hypercholesterolemic mice, total cholesterol (TC), triglyceride (TG) concentration, and low-density lipoprotein (LDL) levels significantly decreased respectively by 7%, 20% and 48% along with high-density lipoprotein (HDL) levels significantly increased by 47% in garlic extract supplemented group. Based on the histological evaluation in the liver sample of group C, both portal and central veins were normal, and fat droplets were not found in the hepatocytes which were found in the liver of group B. On the other hand, unchanged renal cortex, glomerulus, Bowman’s space, and kidney tubules were seen in group C. Conclusion. Therefore, the above findings of the present research would assist to provide affirmation about the cholesterol-decreasing and cellular restoration potentiality of garlic aqueous extract.

Consumers around the world are choosing sustainable and unprocessed foods. Roe deer meat, due to the natural origin, is a source of organic and healthy meat. The selection of suitable storage conditions and times plays an important role in a deterioration of the meat's functional and nutritional values. The knowledge about oxidation processes in roe deer meat stored and packed deploying different methods is limited. The main aim of this study was therefore to investigate the effect of storage method on the physicochemical properties of musculus longissimus thoracis et lumborum (LTL), musculus biceps femoris (BF), and musculus vastus lateralis (VL) of roe deer. The muscles were stored either dry (DRY-AGED), vacuum-packed (VAC), or packed under modified atmosphere (MAP) for 21 days. The quality of roe deer meat was assessed based on chemical composition, technological properties, pH values, water activity, colour, and oxidation processes of proteins and lipids. Roe deer meat had high protein (216.5–228.6 g/kg) and low fat content (17.1–25.8 g/kg). Both DRY-AGED and VAC contributed to improving meat tenderness during storage, while the Warner–Bratzler shear force of the MAP muscles increased. The high-oxygen conditions during MAP storage strongly induced the oxidation processes; an average increase of 1263% for thiobarbituric acid reactive substance (TBARS) level and 155% for protein carbonyl content on day 21. Vacuum packaging and dry-ageing are recommended methods for storing roe deer meat. The high oxygen atmosphere negatively affected the quality of this game species. It carries the risk of increased oxidation of proteins and lipids which may promote the formation of potentially detrimental compounds.

The release of influenza RNA inside the host cell occurs through the fusion of two membranes, the viral envelope and that of the cellular endosome. The fusion is mediated by the influenza hemagglutinin protein (HA), in particular by the fusion peptide (HAfp) located in the N-terminal fragment of HA2 subunit. This protein fragment anchors in the internal endosomal membrane, whereas the C-terminal HA2 part comprises a transmembrane domain (TMD) embedded in the viral envelope. A drop of pH in the endosome acts as the main trigger for HA2 large conformational change that leads to anchoring of the fusion peptide, close contact of the membranes and the subsequent fusion. Throughout the years the major research effort was focused on a 20-aminoacid fragment (HAfp1-20), shown by NMR to adopt a 'boomerang'-like structure. However, recent studies showed that extending HAfp1-20 by three highly conserved residues W21-Y22-G23 leads to formation of a unique, tight helical hairpin structure. This review summarizes recently discovered structural aspects of influenza fusion peptides and their relations with the membrane fusion mechanism.

Geranylgeranoic acid (GGA) and 2,3-dihydrogeranylgeranoic acid (2,3-diGGA) are geranylgeraniol-derived metabolites (Kodaira et al. (2002) J Biochem 132: 327-334). In the present study, we examined the effects of these acids on HL-60 cells. The cells were differentiated into neutrophils by GGA stimulation like retinoic acid stimulation. In the case of cells stimulated with 2,3-diGGA, neutrophils were not detected, but the formation of lipid droplets was induced. On the other hand, when the cells were cultured in the presence of 0.1% FBS instead of 10% FBS, apoptotic cells were induced not only by GGA stimulation but also with 2,3-diGGA. In the latter case, when the cells were cultured in the co-presence of a caspase-3 inhibitor (Ac-DMQD-CHO), the lipid droplets formation was observed in the cells. These results suggest that GGA and 2,3-diGGA are extremely different from each other with respect to their effects on HL-60 cells.

Introduction and objective. Metabolic disorders are associated with a number of negative health consequences. Therefore, the aim of this review was to define the relationships between overweight and obesity, lipid metabolism disorders, hypertension, disorders of carbohydrate metabolism, and cognitive functions. Abbreviated description of the state of knowledge. Metabolic disorders may be linked to the reduction of cognitive functions, such as memory, abstract reasoning, verbal fluency, attention and psychomotor speed. Obesity may lead to changes in the hippocampus, one of the main structures involved in learning and memory. The evidence reviewed suggests that obesity, by inducing inflammation, leads to the dysfunction of brain structures such as the frontal cortex and corpus callosum involved in plasticity processes and cognitive functions. In addition, obesity has been found to be associated with abnormalities in the white matter and glial cells in the brain, which may also cause reduction in processing speed. It is possible that there is a non-linear relationship between lipids and cognitive function moderated by age and the pro- inflammatory process. Disturbances in carbohydrate metabolism can also lead to cognitive impairment, such as: memory, verbal fluency or abstract reasoning. Conclusions. Metabolic disorders are associated with a higher risk of developing cognitive impairment. This association can be moderated by additional factors: lifestyle, oxidative stress, pro-inflammatory processes, genetic factors, age and education.

Introduction. A decreased serum high density lipoprotein-cholesterol (HDL-C) is a strong predictor of cardiovascular risk. However, total HDL is a very dynamic, changeable fraction, and does not perform the function of atherosclerosis markers. In the presented study, the pattern of serum lipids, including HDL-C subclasses (HDL2- and HDL3-cholesterol), in a middleaged Polish Lower Silesia population was defined. Materials and method. A group of 746 males and 1,298 females, aged 35–70, were investigated. All subjects were participants in the PURE study. Mean serum lipid levels were determined for groups selected on the basis of gender, age, cigarette smoking, drinking alcohol and place of residence (urban/rural area). The data were analyzed using STATISTICA 6.0 PL. Results. In multiple linear regression models, age was the most important independent and consistent predictor of total cholesterol (TC) and LDL cholesterol (LDL-C). The prevalence of low HDL-C (threshold 40 mg/dL in males, 50 mg/dL in females) was 16.5% for males and 22.6% for females. This gender-conditioned difference in the prevalence of low HDL-C was greater in rural (20.0% vs. 30.9%, respectively, in males and females) in comparison to urban (14.4% vs. 17.1%) areas. The lipid pattern was significantly worse in rural than in urban females. Female rural inhabitants showed higher triglycerides (TG) and lower HDL cholesterol (total and contained in subclasses HDL2 and HDL3). Simultaneously, a higher BMI, higher percent of smokers and drinkers and lower age of smoking female rural inhabitants in comparison to urban females were estimated. In the total population, cigarette smoking or drinking alcohol were associated with significant increases in TC, LDL-C and TG, also with decreased HDL-C (smoking) or HDL2-C (drinking). Two-way analysis of variance showed the existence of interaction between these risk factors in their influence on HDL-C and HDL3-C. Conclusion. In the middle-aged population of the Lower Silesian region in Poland the place of residence (urban/rural area) had a significant impact on the lipid pattern. This pattern is more atherogenic in rural women than in urban women.

The present study was designed to evaluate the oxidative stress-related parameters in alloxan-induced diabetes in rabbits. After 3, 6, 12 and 24 weeks of hyperglycaemia the enzymatic and non-enzymatic factors were measured in heart tissue of diabetic and control groups. Superoxide dismutase and glutathione peroxidase activities and the contents of total sulfhydryl compounds significantly increased at all time intervals. Catalase activity increased initially (after 3 and 6 weeks), decreased after 12 weeks and increased again at the 24th week of the experiment. Glutathione reductase activity increased initially (at 3rd week), decreased below control level after 6 and 12 weeks, then increased again. Ascorbic acid concentration decreased after 3 and 6 weeks, and increased at the 12th and 24th weeks. The level of lipid peroxidation products was reduced after 3, 6 and 12 weeks of the experiment. After 24 weeks it was significantly elevated. These data suggest that hyperglycaemia induces oxidative stress in the heart but the defense mechanisms in the heart tissue are fairly efficacious against oxidative injury.

Two natural flavonoids, quercetin and isorhamnetin 3-O-acylglucosides, were examined for their inhibitory influence on the in vitro production and release of reactive oxygen species in polymorphonuclear neutrophils (PMNs). The generation of superoxide radical, hydrogen peroxide and hypochlorous acid were measured by, respectively, cytochrome c reduction, dichlorofluorescin oxidation and taurine chlorination. Membrane lipid oxidation was studied by the thiobarbituric acid method in mouse spleen microsomes. The addition of flavonoids at the concentration range 1-100 μM inhibited PMNs oxidative metabolism and lipid peroxidation in a dose-dependent manner. The results suggest that these flavonoids suppress the oxidative burst of PMNs and protect membranes against lipid peroxidation.

Numerous results on membrane lipid composition from different regions of autopsied Alzheimer's disease brains in comparison with corresponding fractions isolated from control brains revealed significant differences in serine- and ethanolamine-containing glycerophospholipid as well as in glycosphingolipid content. Changes in membrane lipid composition are frequently accompanied by alterations in membrane fluidity, hydrophobic mismatch, lipid signaling pathways, transient formation and disappearance of lipid microdomains, changes in membrane permeability to cations and variations of other membrane properties. In this review we focus on possible implications of altered membrane composition on β-amyloid precursor protein (APP) and on proteolysis of APP leading eventually to the formation of neurotoxic β-amyloid (Aβ) peptides, the major proteinaceous component of extracellular senile plaques, directly involved in Alzheimer's disease pathogenesis.

Purpose: Despite people suffering from several forms of ill health, constant exposure to toxic wastes and chronic diseases as a result of mining, there is a tragic gap in the availability of ‘scientific’ studies and data on the health hazards of mining in India. This study was proposed to understand better the relationship between occupational exposure to coal, blood lipid profiles and red blood cell (RBC) functional status of coal workers. Materials and methods: Blood samples were obtained from coal miners (n=32) of an underground mine in West Bengal. Blood lipid profiles and RBC functional status were determined. Students’ t-test and Pearson correlation analyses were completed to analyze the data. Results: Compared to the control subjects, significantly higher levels of cholesterol (p<0.01), triglycerides (p<0.01), LDL (p<0.001), and VLDL (p<0.001) were observed in coal miners. HDL, Hb, Na+-K+ATPase and SOD activity were significantly (p<0.001) lower in coal miners, whereas MDA levels (p<0.001) and osmotic fragility in coal miners were increased significantly (p<0.01). Conclusions: Our study indicates that elevated MDA and antioxidant insufficiency caused disruption in the structural integrity of erythrocyte, which may be a pathophysiological mechanism in the progression of disease in coal miners. Also, cardiovascular disease risk factors were more prevalent in the coal miners.

In the present paper, cold plasma-activated and chitosan-coated polyetheretherketone (PEEK) was covered with thin films of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine, cholesterol, cyclosporine A, and their mixtures using the Langmuir-Blodgett technique. The thermodynamic function, i.e., surface free energy, of those systems was determined based on the contact angle hysteresis (CAH) approach. This parameter seems to be essential in determination of cell adhesion to polymeric materials and molecular interactions with living tissues. The obtained results show that the wettability and surface free energy of PEEK can be changed depending on the composition of the coating
