Opis:
Synthetic and natural polymers blends represent a new brand of materials with application in wound healing, scaffolds or drug delivery systems. Polycaprolactone/gelatin (PCL/Gt) blends were analyzed in terms of their miscibility. The PCL structure was investigated as a function of Gt content. Changes in the PCL spherulitic structure with Gt content were investigated by a polarizing-interference microscope. The analysis of the glass transition temperature (Tg) of both components as a function of PCL/Gt ratio by differential scanning calorimetry indicates that the system of polycaprolactone/gelatin belongs to a type of s.c. compatible system, being intermediate between miscible and immiscible systems. There is possibility of very limited miscibility of both components. Supplementary wide angle X-ray scattering results are presented.

This research aimed to characterize bioplastic packaging made from modified tapioca flour so that it has the best thickness, transparency, tensile strength, elasticity and biodegradability values. This research was conducted at the Laboratory of Fisheries Products Processing, Faculty of Fisheries and Marine Sciences, Padjadjaran University, Jatinangor, starting from February-April 2019. Bioplastic were tested namely bioplastic in four treatments and three repeat with the addition of chitosan (K) and gelatin (G) concentrations, namely treatment A (2% K, 5% G), B (3% K, 10% G), C ( 4% K, 15% G), and D (4% K, 20% G). The data obtained were analyzed using a comparative descriptive method. The parameters observed included thickness, transparency, tensile strength, percent elongation, and biodegradability. Thickness testing with measurement methods at 5 different spots using a digital screw micrometer. Transparency testing was done by the method of inserting a sample in the spectrophotometer and the absorbance value was determined. Tensile strength and elongation tests were carried out by the withdrawal method at both ends of the sample using the Instron tool. Testing the value of biodegradability was carried out using the soil burial test method. The best bioplastic packaging produced is using the addition of 2% chitosan and 5% gelatin concentration which is seen from the thickness parameter values of 0,072 mm, transparency value of bioplastic 2,09, the tensile strength of 19,05 MPa, percent elongation 28,333% and can be degraded by soil after 14 days about 99,84% based on Japanesse Industrial Standard (JIS).

Opis:
An organic solvent and surfactant stable α-amylase was obtained from soybean seeds. The direct and indirect effect of various organic solvents (non-polar, polar protic, and polar aprotic) and surfactants on the activity and stability of free enzyme was determined. The enzyme showed a very high catalytic efficiency and stabilization against most of the organic solvents and surfactants tested, except for few. Those organic solvents and surfactants (like chloroform, dimethyl formamide, n-butanol, and Tween 20), which caused an inhibition in enzyme activity, were used to study their effects on immobilized enzyme. The inhibitory effect was found to be decreased in immobilized enzyme as compared to free enzyme indicating that immobilization imparted stability to the enzyme. Moreover, the possibility of reuse of the enzyme in the presence of the organic solvents and surfactants was increased upon immobilization. The stability of soybean α-amylase towards organic solvents and surfactants shows that it is a potential candidate for use in organic-solvent biocatalysis as well as in detergent industries.

The influence of gelatin coatings modified with fungal melanin on pork lard oxidative stability was studied. The lard was coated with gelatin coatings containing 0.1%; 0.5%; 1% of fungal melanin and control gelatin coating devoid of melanin. The peroxide values (POV), iodine values (IV) and acid values (AV) were studied after 7, 14 and 21 days storage in controlled conditions. Lard covered with modified coatings had lowered oxidative rancidity. Hence, modified coatings containing fungal melanin can be use effectively for the prevention of lard oxidation.

Opis:
The present study aimed to prepare hydrogel based on polyvinyl alcohol (PVA) and gelatin (Ge) and characterization of PVA/Ge hydrogel for their potential use as a sustained drug delivery system. Polyvinyl Alcohol (PVA) and-Gelatin (Ge) were cross-linked using glutaraldehyde (GA) as a crosslinking agent and hydrochloric acid (HCl) as a catalyst. Different feed polymer ratio and crosslinking agent concentration were used to prepare a series of PVA/Ge hydrogels. The obtained PVA/Ge hydrogels were investigated for dynamic and equilibrium swelling studies. The effect of polymers ratio, degree of crosslinking and pH of the medium on swelling of PVA/Ge hydrogels was investigated. Furthermore, the values of diffusion coeficient (D), volume fraction, polymer-solvent interaction parameter, molecular weight between crosslink and crosslink density were calculated. For swelling studies, 0.05M USP phosphate buffer solutions of different pH (1.2, 5.5, 6.5 and 7.5) were used. For the drug release study, ciprofloxacin HCl was loaded into selected samples as a model drug. The release of drug from these samples was performed for 12 hours in USP phosphate buffers of pH 1.2, 5.5 and 7.5. The release data from these samples were fitted into various kinetic models like zero order, first order, Higuchi and Peppas models to investigate the release mechanism. It was found that by varying the composition of PVA/Ge hydrogel and GA concentration, a significant difference was observed in drug release kinetics. FTIR spectroscopy and X-ray diffraction were used for the characterization of hydrogels. PVA/Ge hydrogel showed sustained release of the model drug at various pH values suggesting its potential use as a sustained drug delivery system.

Foils were prepared from potato starch (S), furcellaran (F) and gelatin (G) (S/F/G foils) using glycerol as a plasticizer. Their mechanical properties, aqueous solubility, water content, water uptake, enzymatic hydrolysis, and thermal (DSC) properties were determined. The 0.16 mm thick foil has ~ 80 MPa mechanical resistance with an elongation at break of 27.8 %. The aqueous solubility and water uptake of the S/F/G foils reached 35 % and 145 %, respectively. The S/F/G foils were susceptible to hydrolysis with polysaccharide enzymes such as glucoamylase and Viscozyme L (a blend of arabanase, cellulase, β-glucanase, hemicellulase and xylanase), as well as trypsin, a proteinase.
Zbadano kinetykę reakcji hydrolizy enzymatycznej folii S/F/G w obecności enzymów polisacharydowych: glukoamylazy i Viscozyme L (mieszanina enzymów arabinazy, celulazy, β-glukanazy, hemicelulazy, ksylanazy) oraz enzymu białkowego trypsyny.

Opis:
Fungal melanin was used to prepare gelatin-based composite films. The films of various melanin concentrations (0.1%, 0.5% and 1% w/w) were prepared using a solution casting method. The mechanical, antioxidant, antimicrobial, water vapor, oxygen and UV-Vis barrier properties, as well as any available surface polyphenolics were studied. FT-IR and Raman spectroscopy studies were carried out to analyse the chemical composition of the resulting films. The hydrophobocity, solubility, colour response, optical properties and opacity were also determined. The results of this study showed that the modification of gelatin with fungal melanin had no influence on mechanical and water vapor barrier properties, oxygen barrier properties were improved. The UV-Vis barrier properties of modified films were significantly improved. Modified gelatin films showed good antioxidant activity but were inactive against microorganisms. The modification with melanin caused changes in colour values, decreasing lightness and increasing the redness and yellowness of the films. Based on the results of this study, fungal melanin has good potential to be utilised as a value-added modifier that can improve the properties of gelatin films.

Opis:
The Fricke xylenol orange (FX) gel system is a chemical dosimeter characterized by good sensitivity, linear dose response, tissue equivalence, no toxicity, easy preparation, reproducibility and low cost. Thanks to the presence of the gelatinous matrix, the system is particularly suitable to perform reliable 3D mapping of the absorbed dose spatial distribution via magnetic resonance imaging (MRI) or optical techniques. The aim of this work is to study in a systematic way the influence of the pre-irradiation storage procedure upon sensitivity, dose response stability and lifetime of use of a FX gel system made with gelatin from porcine skin subjected to homogeneous irradiation. For this purpose, different pre-irradiation storage procedures, in terms of temperature and duration of each storage step, were investigated. In order to evaluate the dose response stability, the optical analyses of the samples were performed up to 6 hours after irradiation. Moreover, the samples were irradiated at time intervals of 24 hours for up to 7 days after preparation in order to evaluate the system lifetime of use. Regardless of their thermal and temporal life, the samples show linear dose responses in the investigated dose range (3–24 Gy) and an increase of sensitivity with the time elapsed between preparation and irradiation. Among the three pre-irradiation storage procedures considered here, a procedure that provides the best dose response stability and lifetime of use was identifi ed and recommended for further use. The analyzed dosimetric system possesses good properties that make it promising for medical application, particularly concerning the evaluation of pre-treatment plan quality assurance within the conformational external beam radiotherapy.
