It is not always clear that some equations affected by complicated factors can, actually, be interpreted as a ratio of two polynomials of first degree and so that they can be, in general, represented by rectangular hyperbolas. In this paper we present an easy procedure to rearrange those equations into Michaelis-Menten-type equations and so to make the aspects of these rectangular hyperbolas more clear, particularly for researchers familiar with general biochemistry. As an example, the method is applied to transform the classical rate equation of the Cleland×s Ordered Uni Bi enzyme mechanism.

Opis:
A combined analysis of enzyme inhibition and activation is presented, based on a rapid equilibrium model assumption in which one molecule of enzyme binds one molecule of substrate (S) and/or one molecule of a modifier X. The modifier acts as activator (essential or non-essential), as inhibitor (total or partial), or has no effect on the reaction rate (v), depending on the values of the equilibrium constants, the rate constants of the limiting velocity steps, and the concentration of substrate ([S]). Different possibilities have been analyzed from an equation written to emphasize that v = Ł([X]) is, in general and at a fixed [S], a hyperbolic function. Formulas for Su (the value of [S], different from zero, at which v is unaffected by the modifier) and vsu (v at that particular [S]) were deduced. In Lineweaver-Burk plots, the straight lines related to different [X] generally cross in a point (P) with coordinates (Su, vsu). In certain cases, point P is located in the first quadrant which implies that X acts as activator, as inhibitor, or has no effect, depending on [S]. Furthermore, we discuss: (1) the apparent Vmax and Km displayed by the enzyme in different situations; (2) the degree of effect (inhibition or activation) observed at different concentrations of substrate and modifier; (3) the concept of Ke, a parameter that depends on the concentration of substrate and helps to evaluate the effect of the modifier: it equals the value of [X] at which the increase or decrease in the reaction rate is half of that achieved at saturating [X]. Equations were deduced for the general case and for particular situations, and used to obtain computer-drawn graphs that are presented and discussed. Formulas for apparent Vmax, Km and Ke have been written in a way making it evident that these parameters can be expressed as pondered means.

Introduction. The standard porcine-derived pancreatic enzyme replacement therapy (PERT) is a lifesaving treatment for patients with diseases causing exocrine pancreatic insufficiency (EPI). An attempt to replace PERT with microbial enzymes were undertaken. The aim was to highlight whether the mode of application, mixed with food or applied directly to the stomach, of pancreatic-like enzymes of microbial origin (PLEM) can affect their activity along the gastrointestinal tract. Materials and method. The activity of amylase, lipase and proteinase in the stomach, duodenum and ileum were tested in EPI pigs (n=6) after supplementation of PLEM, either orally – before and during feed consumption – or via the stomach – before and during feed consumption. Healthy pigs not treated with PLEM (n=3) served as controls. Activity of the enzymes measured in the chyme were obtained together with the digesta pH. Activity of the enzymatic residues in the stool samples was also checked. Results. The highest pancreatic enzyme activities were found in the duodenum of the healthy pigs (amylase 162,68 kU/ mL, lipase 507,34 kU/mL and protolitic (trypsin) activity 357,60 kU/mL). Nevertheless, the microbial enzymes remained also active along the entire length of the GIT – including stomach in EPI pigs, regardless of their route of administration. However, activity level was significantly lower. Discussion. Results indicate that the activity pattern of PLEM in the small intestine mimics the activity of the natural endogenous pancreatic enzymes in healthy pigs. The most physiological features of PLEM were observed when enzymes were offered orally. The magnitude of PLEM activity in the stomach of EPI pigs was essential and significantly higher than that measured in healthy pigs, thus being somewhat not physiological, and for health reasons of the patients should be further explored. Interestingly, specific trypsin-like activity was measured in all parts of the GIT after PLEM application. However, proteolytic activity of the experimental proteaze in in vitro studies did not exhibit trypsin-like activity.

The objective of the study described the importance of L-asparaginase and its importance in the field of medicine. Different types of enzymes are produced based on the adaptation to the environment where the living organisms live to tune the metabolic pathways according to their adapted changes. The enzymes present in various organs are produced by many cell types in multicellular organisms. Except ribosomes all other known enzymes are proteinaceous in nature. L-asparaginase is a potential therapeutic agent for acute lymphoblastic leukemia (ALL) and chronic myelogenous leukemia which is approved by FDA & WHO. L-asparaginase catalyzes the deamination of L-asparagine to L-aspartic acid & ammonia. Unlike normal cells, malignant cells require large amount of L-asparagine for protein synthesis and cell division. From this background the present review is an effort to gather the information on the mechanism, sources, molecular details and application of L-asparaginase enzyme.

Opis:
Taking into account the progressive degradation of soils it is important to assess their quality. Soil quality depends on a large number of physical, chemical, biological and biochemical properties. In the publications available, there are presented three approaches regarding the use of soil properties to estimate soil quality: (1) the use of individual properties, (2) the use of simple indexes and (3) the use of complex indexes derived from combinations of different properties. The purpose of this study was to investigate the possibility to use enzymes as indicators of forest soil quality. Experimental plots (43) were located in central Poland. The study was carried out in a number of diverse fresh forest sites. To assess the quality of forest soils dehydrogenase and urease activity and the degree of base saturation were used. One of the final conclusions point out that enzymatic activity indicates current site condition as well as the changes that occur in soil better than soil physical and chemical properties. In other words, in comparison to soil enzymatic activity, soil physico-chemical properties constitute a less sensitive indicator of soil changes.

Using histochemical and cytophotometric methods, enzymes associated with anaerobie glycolysis: glucose-6-phosphatase, hexokinase, and aldolase in the developing sporocysts of Fasciola hepatica were studied. Highest activity of these enzymes was found in the germ balls in the sporocysts, at all phases of their development, which is related to intensive proliferation and differentiation of the embryos developing inside the germ balls.

The paper reports results of analysis of protein and eight enzymatic systems of three varieties of Helianthus annuus L. (Wielkopolski, Coril, and Frankasol) performed by three electrophoretic methods. Electrophoretic analyses allowed finding protein and isoenzymes markers for each of variety. The two interline hybrids Coril and Frankasol shared more features in common than the population variety Wielkopolski with each of the former. Mean values of the polymorphism index for the protein and the eight enzymes indicate that the variety Wielkopolski is more homogeneous than Coril and Frankasol.
W pracy przedstawiono wyniki analiz białek i ośmiu układów enzymatycznych, rozdzielanych trzema metodami elektroforezy, u trzech odmian Helianthus annuus L. (Wielkopolski, Coril i Frankasol). Elektroforetyczne analizy umożliwiły znalezienie białkowych i izoenzymatycznych markerów dla trzech odmian. U dwóch odmian międzyliniowych mieszańców Coril i Frankasol wykryto więcej cech wspólnych niQ u wymienionych odmian i odmiany Wielkopolski. Średnie wartości indeksu polimorfizmu, obliczone dla białek i ośmiu układów enzymatycznych, pokazały, że odmiana Wielkopolski była bardziej wyrównana niż odmiany Coril i Frankasol.

Celem badań było określenie aktywności ureazy na wybranych glebach Polski: bielicowych i brunatnych wytworzonych z piasków i glin, lessów, czarnych ziem i mad rzecznych. Badaniom poddano próbki z poziomu 0-10 cm, które pobrano z Banku Gleb Instytutu Agrofizyki PAN. Aktywność ureazy zawierała się w przedziale 0,3256 - 1,7896 pM N-NH4 kg-lh-1. Najniższą aktywność ureazową zanotowano w glebach bielicowych i wynosiła ona 0,33 pM N-NH4 kg-lh-1. W glebach brunatnych wytworzonych z piasków i lessów aktywność ureazy wahała się w granicach 0,55 - 1,00 pM N-NH4 kg-lh-1. Najwyższą aktywność stwierdzono w glebach: czarnych ziemiach, madach rzecznych, glebach murszowych i murszowatych 1,43 - 1,79 pM N-NH4 kg-lh-1. W oparciu o analizę statystyczną otrzymanych wyników można stwierdzić, że aktywność ureazowa pozostaje w ścisłym związku z zawartością materii organicznej.
The aim of the investigation was to measure activity of urease in different polish soils such as podzolic and brown developed from sands and loams, brown developed from loess, as well as in black earths and aluvials soils. The measurements were performed on samples taken from 0-10 cm depths. The urease activity was in the range of 0,3256 - 1,7896 pM N-NH4+ kg''h‘'. The lowest activity was noted in the podzolic soil and was on the level of 0,3256 pM N-NH4+ kg’'h''. In the brown and lessive soils the urease activity was in the rangę 0,55-1,00 pM N-NH4+ kg''h_l. The highest urease activity were found in black earths, alluvials soils, Eutric Histosol Soils 1,43-1,79 pM N-NH/kg1 h’’. The results obtained allow to conclude that the urease activity is tightly connected with the organie matter content of the soil.

Chloroplast protease AtDeg2 (an ATP-independent serine endopeptidase) is cytosolically synthesized as a precursor, which is imported into the chloroplast stroma and deprived of its transit peptide. Then the mature protein undergoes routing to its functional location at the stromal side of thylakoid membrane. In its linear structure AtDeg2 molecule contains the protease domain with catalytic triad (HDS) and two PDZ domains (PDZ1 and PDZ2). In vivo AtDeg2 most probably exists as a supposedly inactive haxamer, which may change its oligomeric stage to form active 12-mer, or 24-mer. AtDeg2 has recently been demonstrated to exhibit dual protease/chaperone function. This review is focused on the current awareness with regard to AtDeg2 structure and functional significance.

In this study, polyvinyl alcohol (PVA) hydrogels designed for osteochondral regeneration were mineralized with calcium phosphate (CaP) and magnesium phosphate (MgP) by incorporation of the enzyme alkaline phosphatase (ALP) and incubation in solutions of 0.1 M calcium or magnesium glycerophosphate (CaGP, MgGP). Hydrogels incubated in water served as controls. Dry mass percentage, i.e. the percentage consisting of polymer and mineral and not water, was markedly higher for samples incubated in CaGP and MgGP and decreased in the order: CaGP>MgGP>water. CaP and MgP formation was confirmed by FTIR. SEM revealed spherical deposits characteristic for apatite in samples incubated in CaGP and plate-like crystals of the magnesium phosphate bobierrite in samples incubated in MgGP, while samples incubated in water were devoid of deposits. Rheometric testing showed that storage modulus of hydrogels incubated in CaGP and MgGP was markedly higher and decreased in the order: CaGP>MgGP>water. The results prove the principle of enzymatic mineralization of PVA with CaP and MgP to improve their suitability for bone regeneration applications.

The objective of this study was to investigate the effect of exogenous selenium (Se) supply (0, 2, 4, 8, 16 μM) on the growth, lipid peroxidation and antioxidative enzyme activity of 100 mM NaCl-stressed melon (Cucumis melo L.) seedlings. Salt stress significantly reduced the growth attributes including stem length, stem diameter, dry weight and increased antioxidative enzyme activity [superoxide dismutase (SOD), peroxidase (POD), catalase (CAT)]. Moreover, the plant exhibited a significant increase in electrolyte leakage and malondialdehyde (MDA) content under NaCl stress. Se supplementation not only improved the growth parameters but also successfully ameliorated the adverse effect caused by salt stress in melon seedlings. However, the mitigation of NaCl-stressed seedlings was different depending on the Se concentration. At lower concentrations (2–8 μM), Se improved growth and acted as antioxidant by inhibiting lipid peroxidation and increasing in SOD and POD enzymes activity under salt stress. At higher concentrations (16 μM), Se exerted diminished beneficial effects on growth. Whereas CAT activity was enhanced. The result indicated that Se supplementation had a positive physiological effect on the growth and development of salt-stressed melon seedlings.

Using histochemical and cytophotometric methods, enzymes associated with anaerobie glycolysis: glucose-6-phosphatase, hexokinase, and aldolase in the developing sporocysts of Fasciola hepatica were studied. Highest activity of these enzymes was found in the germ balls in the sporocysts, at all phases of their development, which is related to intensive proliferation and differentiation of the embryos developing inside the germ balls.

Boron is a mineral used for various purposes in glass, ceramics, automotive and paint industries. Recently, boron has been confirmed to be an essential element for plants, animals and humans, although the knowledge of its biological effects is rather scanty. Boron is a dynamic trace element, and inorganic borates are transformed into boric acids and absorbed from mucosal surfaces, even when they are in low levels of physiological pH. It has been determined that boron affects many enzymes, bone development, mineralization, Ca, P, Mg and energy metabolism. Boron mineral compounds can be effective in optimizing the performance of an organism, treatment of bone structure disorders, reduction of cholesterol and triglyceride levels. Beside the effects it produces specifically on fat and lipid metabolisms, boron can influence the activity of vitamin D and affect some disorders connected with its deficiency. Although several studies on effects of boron on some mechanisms have been conducted over the last ten years, the available information remains insufficient.

Opis:
A general dependence of the enzyme catalytic rate on its mass was revealed when a statistical analysis of 17065 records from the EMP database was performed. The estimated activation energy of the catalytic process decreases asymptotically with the enzyme molecular mass increase. The proposed theoretical model postulates the existence of an intermediate complex of the enzyme and the departing product. It allows for the explanation of the discovered mass-energy relationship, as an effect of the global enzyme-product interactions during complex dissociation. Fitted parameters of the model seem to be in agreement with those widely accepted for the van der Waals energy of molecular interactions. Their values also agree with the picture of the hydrogen bonding in the catalytic process and suggest that surface walk can be the favorable way of the product departure.

Trehalase (α, α-trehalose-1-C-glucohydrolase, EC 3.2.1.28) is a glycoside hydrolase enzyme that catalyzes the conversion of trehalose to glucose. Trehalose hydrolysis by trehalase enzyme is an important physiological process for various organisms, such as fungal spore germination, insect flight, and the resumption of growth in resting cells. So by studying biochemical properties of trehalase we can control the spore germination of harmful fungus, Apergillus niger. Till now trehalase has been reported from many other organisms including plants and animals, Trehalase has been purified and characterized from various organisms such as Saccharomyces cerevisiae, Lentinula edodes, Acidobacterium capsulatum, and Medicago sativa, as well as root nodules of Phaseolus vulgaris, soybean and from the from the seeds of Triticum aestivum. In this study, trehalase was purified from Aspergillus niger grown on cellulosic medium. And this purified trehalase was tested for various requirements it may need for enzymatic activity such as inorganic phosphate and Magnesium ions (Mg2+).

Hydroxyapatite is used as a matrix for immobilization of protease from Aspergillus oryzae by a process of adsorption. The matrix obtained has the surface area of 26 m2/g and particles in the shape of flakes of diameters no greater than 650 nm. The efficiency of the proposed method was confirmed by the Fourier transform infrared spectroscopy, elemental analysis and by analysis of parameters of the pore structure of matrix and products after immobilization. On the basis of the Bradford method it was found that the greatest amount of enzyme (132 mg/g) was immobilized from a solution of initial enzyme concentration of 7 mg/cm3 after 24 h of the process.

Change in hydrolytic enzyme efficiency over time. The purpose of this study was to determine the action of hydrolytic enzymes (by Dyadic Cellulase CP CONC, and the Dyadic Xylanase 2 XP CONC) over time. Chromatographic analysis of holocellulose samples subjected to enzymatic hydrolysis was performed. The following hydrolysis parameters were used: time 48h, temperature 45 ⁰C, acetate buffer pH 5.4, commercial enzymes Dyadic. Holocellulose extracted by the sodium chlorite method from white poplar wood (Populus alba L.) was used. The final yield of enzymatic hydrolysis was determined. The results of hydrolysis performed at intervals were compared. The results obtained show that the hydrolysis yield of holocellulose after five months decreased by 40 p.p. for glucose yield and by 25 p.p. for xylose yield. The yield for glucose after two and a half years decreases by 68 p.p. and 62 p.p. for xylose compared to the initial yield.
Zmiana wydajności enzymów hydrolitycznych w czasie Celem pracy było określenie działania enzymów hydrolitycznych w czasie. Przeprowadzono analizę chromatograficzną próbek holocelulozy poddanych hydrolizie enzymatycznej. Zastosowano następujące parametry hydrolizy: czas 48h, temperatura 45 ⁰C, bufor octanowy pH 5,4, enzymy komercyjne Dyadic. Zastosowano holocelulozę pozyskaną metodą chlorynu sodowego z drewna topoli bialej (Populs alba L.). Określono końcową wydajność hydrolizy enzymatycznej. Porównano wyniki hydrolizy przeprowadzonej w odstępach czasu. Uzyskane wyniki wskazują, że wydajność hydrolizy holocelulozy po pięciu miesiącach zmniejszyła się o 40 p.p. dla wydajności glukozy i o 25 p.p. dla wydajności ksylozy. Wydajność glukozy po dwóch i pół roku obniża się o 68 p.p., a ksylozy o 62 p.p. w stosunku do wydajności początkowej.

The effects of externally applied sodium nitroprusside on superoxide dismutase (SOD), glutathione peroxidase (GPx), ascorbate peroxidase (APX), and catalase (CAT) antioxidant enzyme activities, nitric oxide (NO) levels have been investigated in salt stress resistant and sensitive sunflower plants. NaCl treatments and SNP treatments simultaneously with salt application (NaCl+SNP) were performed. SOD, GPx, APX and CAT antioxidant enzyme activities and NO levels showed differences in leaf tissues treated with 100 µM SNP, different concentrations of NaCl, and NaCl+SNP. SOD, GPx and APX enzyme activities were generally increased in sensitive variety, but decreased in tolerant variety. However, while general increase in CAT enzyme activity was determined in tolerant type, a reduction was established in sensitive type. An increase was determined in both types in NO levels. It is evident from these results that administration of NO donor SNP can cope with reactive oxygen species in both varieties. This study indicates that negative effects of salt stress on different sun flower varieties can be recovered by nitric oxide.

The aim of the study was to determine how adenine affected biological proprieties of soil. The performance of this precursor cytokinine was tested in a pot trial. The question posed was whether it was possible to improve efficacy of adenine by enlarging populations of bacteria from Azotobacter species in soil. The experiment was carried out on proper brown soil, formed from dust clay sand with pHKCl 6,9. Pots were filled with 3.2 kg of soil. The investigations were performed in two series: with and without addition Azotobacterin to soil. Tow rates of nitrogen fertilisation: 0 and 50 mg N·kg-1 of soil were applied in test. Adenine was applied in the following quantities: 0; 5; 10 and 15 mg·kg-1 of soil. Radish, 6 plants per pot, was the test plant. It was confirmed that the adenine had a significant effect on growth and development of radish. It positively affected microbiological and the biochemical proprieties of soil. The counts of total oligotrophic bacteria, oligotrophic sporulation bacteria, total copiotrophic bacteria, copiotrophic sporulation bacteria, ammonifying bacteria, immobilizing bacteria, celulolytic bacteria, Azotobacter sp., Artrobacter sp. and Pseudomonas sp. were increased, and the number of fungi diminished. Adenine also stimulated activities of dehydrogenase, urease and alkaline phosphatase, although it depressed the activity of acid phosphatase. The inoculation with bacteria from Azotobacter species applied to soil failed to improve efficacy of adenine. Nevertheless, it increased counts of these bacteria, which had a beneficial influence on the development of oligotrophic bacteria, immobilizing bacteria, celulolytic bacteria and actinomyces, while negatively affecting fungi, ammonifying bacteria and Arthrobacter.
Celem badań było określenie wpływu adeniny na biologiczne właściwości gleby. Działanie tego prekursora cytokinin testowano w doświadczeniu wazonowym. Sprawdzano możliwość wzmocnienia efektywności adeniny przez zwiększenie w glebie puli bakterii z rodzaju Azotobacter. Badania wykonano w próbkach gleby brunatnej właściwej, wytworzonej z piasku gliniastego pylastego o pHKCl 6,9. W wazonach umieszczono po 3,2 kg gleby. Badania obejmowały dwie serie: bez i z dodatkiem do gleby szczepionki Azotobakteryny. W doświadczeniu zastosowano zróżnicowane nawożenie azotem: 0 i 50 mg N·kg-1 gleby. Adeninę stosowano w następującej ilości: 0; 5; 10 i 15 mg·kg-1 gleby. Rośliną doświadczalną była rzodkiewka - 6 roślin w wazonie. Stwierdzono, że adenina istotnie wpływała na wzrost i rozwój rzodkiewki. Korzystnie oddziaływała na mikrobiologiczne i biochemiczne właściwości gleby. Zwiększała liczebność bakterii: oligotroficznych ogółem, oligotroficznych przetrwalnikujących, kopiotroficznych ogółem, kopiotroficznych przetrwalnikujących, amonifikacyjnych, immobilizujących azot, celulolitycznych, Azotobacter sp., Arthrobacter sp., Pseudomonas sp. oraz promieniowców, a zmniejszała liczbę grzybów. Stymulowała także aktywność dehydrogenaz, ureazy i fosfatazy alkalicznej, natomiast hamowała aktywność fosfatazy kwaśnej. Zastosowana szczepionka złożona z bakterii z rodzaju Azotobacter nie poprawiała efektywności adeniny. Zwiększała jednak liczbę tych bakterii w glebie, co wpłynęło korzystnie na rozwój bakterii oligotroficznych, immobilizujących azot, celulolitycznych oraz promieniowców, natomiast negatywnie - na grzyby oraz bakterie amonifikacyjne i bakterie z rodzaju Arthrobacter.

Dehydrogenases are exclusively intracellular enzymes, which play an important role in the initial stages of oxidation of soil organic matter. One of the most frequently used methods to estimate dehydrogenase activity in soil is based on the use of triphenyltetrazolium chloride as an artificial electron acceptor. The purpose of this study was to compare the activity of dehydrogenases of forest soils with varied physicochemical properties using different triphenyltetrazolium chloride assays. The determination was carried out using the original procedure by Casida et al., a modification of the procedure which involves the use of Ca(OH)2 instead of CaCO3, the Thalmann method, and the assay by Casida et al. without addition of buffer or any salt. Soil dehydrogenase activity depended on the assay used. Dehydrogenase determined by the Casida et al. method without addition of buffer or any salt correlated with the pH values of soils. The autoclaved strongly acidic samples of control soils showed high concentrations of triphenylformazan, probably due to chemical reduction of triphenyltetrazolium chloride. There is, therefore, a need for a sterilization method other than autoclaving, ie a process that results in significant changes in soil properties, thus helping to increase the chemical reduction of triphenyltetrazolium chloride.

Butyrylcholinesterase (BChE) is synthesized in the liver and found in high concentrations in blood plasma, liver, heart, pancreas, vascular endothelium, skin, brain white matter, smooth muscle cells and adipocytes. BChE is a non specific enzyme that hydrolyzes different choline esters (succinylcholine, mivacurium) and many other drugs such as aspirin, cocaine and procaine. The enzyme is also considered as a bioscavenger due to its ability to neutralize the toxic effects of organophosphorus compounds (nervous system fs agents) such as soman. BChE displays several polymorphisms that influence its serum activity; therefore they could determine the individual sensitivity to chemical nerve agents. In this study, we investigated the correlation between BChE variants and the degree of enzyme inhibition and reactivation after soman application on blood samples of 726 individuals. The blood samples of individuals expressing abnormal variants, were more sensitive to soman compared to variants of homozygotes and heterozygotes for U-allele. We found significant differences in the degree of enzyme reactivation between different variants (with and without U-presence).

Papain activity was studied in water-organic solvent mixtures using the fluorogenic substrate Dabcyl-Lys-Phe-Gly-Gly-Ala-Ala-Edans. The increase of organic solvent (MeOH, EtOH, iPrOH, TFE, MeCN, (MeO)2Et and DMF) concentration in the mixture caused a substantial decrease the initial rate of papain-catalyzed hydrolysis. Moreover, the number of papain active sites decreased with the increase of DMF and MeOH concentration.

In the presented study the ribavirin-TP - an established inhibitor of the NTPase activity of the superfamily NTPase/helicases II - was investigated as an inhibitor of the unwinding activity of the hepatitis C virus (HCV) NTPase/helicase. The kinetics of the reaction revealed that ribavirin-TP reduces the turnover number of the helicase reaction by a mechanism that does not correspond to that of the inhibition of the NTPase activity. Our results suggest that derivatives of ribavirin-TP with enhanced stability towards hydrolytic attack may be effective inhibitors of the enzyme.

Applying pesticides to crops is one of the causes of water pollution by surface runoff, and chlorpyrifos, trifluralin and chlorothalonil are used respectively as insecticide, herbicide and fungicide for crop plants widely. To explore effects of three pesticides on aquatic organisms, superoxide dismutase (SOD) and glutathione S-transferase (GST) activities were determined after 24 h and 48 h exposure of D. magna with ages of 6–24 h to several low concentrations of chlorpyrifos (0.36, 0.72, 1.43, 2.86, 5.72 μg∙L−1), trifluralin (0.17, 0.33, 0.66, 1.33, 2.65 mg∙L−1) and chlorothalonil (0.09, 0.18, 0.36, 0.72, 1.43 mg∙L−1) respectively. Main reproductive parameters including first pregnancy time, first brood time, the number of first brood and total fecundity after 21 d exposures at the same concentrations of pesticides as described above were also measured. The results showed that the activities of GST increased in lower concentrations and decreased in higher concentrations after 24 h exposure to three pesticides, respectively. The activities of SOD showed the same changes after 48 h exposure. With the time prolonged, the activities of GST decreased while the activities of SOD increased. After 21 d exposure, the first pregnancy time and first brood time were delayed, while the number of the first brood and total fecundity per female decreased with increasing concentrations. These results corroborated that GST activity was more sensitive to those pesticides than SOD activity, and there was a significant relationship between total fecundity and pesticides-dose(r>0.94, n=6), GST activity after 48 h exposure and total fecundity after 21 d exposure (r>0.92, n=6).

Prowadzenie procesów biotransformacji przy optymalnym sterowaniu temperaturą jest bardziej korzystne aniżeli w przypadku zastosowania warunków izotermicznych. Aby to oszacować przeanalizowano proces z enzymem natywnym ulegającym zarówno dezaktywacji niezależnej od stężenia substratu jak i równoległej. Stosując klasyczną metodę rachunku wariacyjnego uzyskano równanie analityczne pozwalające odpowiedzieć na pytanie, ile enzymu można oszczędzić w procesie prowadzonym przy optymalnym sterowaniu temperaturą w porównaniu z jego przebiegiem w warunkach izotermicznych. Rozważono przy tym wpływ ilorazu energii aktywacji na uzyskane korzyści. Wykazano, że niskie końcowe aktywności enzymu, dla względnie wysokich wartości ilorazu energii aktywacji, powodują znaczące oszczędności enzymu, dwukrotne lub większe, gdy zamiast warunków izotermicznych zastosuje się optymalne sterowanie temperaturą.
It is known that the running of biotransformations at optimal temperature control is more profitable than in a case of isothermal conditions application. In order to quantify it, biotransformation process with native enzyme deactivation both parallel and independent of substrate concentration, running in a perfect batch reactor was analyzed. Making use of variational calculus method the analytical expression was derived enabling one to answer the question how much of enzyme can be saved if instead of commonly applied isothermal conditions the optimal temperature control is used. An effect of the activation energy quotient on the magnitude of such benefits was considered. It was proved that the lower catalyst activity for relatively high values of activation energy quotient caused significant enzyme savings - two or even more time greater than in a case of optimal temperature control application compared to isothermal process.

The aim of the study was the electrophoretical identification of molecular forms of selected antioxidant enzymes in dog semen. Ejaculates to be studied were chosen from five dogs, aged from two to eight years. Polyacrylamide gel electrophoresis was carried out under non-denaturing conditions and then gels were stained for the activity of the following enzymes: superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Sperm homogenates and all fractions (pre-spermatic, spermatic and post-spermatic) of dog ejaculate demonstrated one protein band with SOD activity characterized by low electrophoretic mobility. Based on the confirmed sensitivity to H₂O₂, it can be assumed that the detected SOD is an enzyme containing ions of Zn²⁺ and Cu²⁺ (Cu,Zn SOD). In sperm homogenates one protein band with GPx activity was characterized by high electrophoretic mobility, whereas in the spermatic and post-spermatic fractions of dog ejaculate three protein bands with different (low, medium and high) electrophoretic mobility were identified. CAT molecular forms were not found in either sperm homogenates or in the analyzed fractions of ejaculate.

The protozoan Neospora caninum (Apicomplexa: Sarcocystidae), respectively infects canids and ruminants as the most definitive and intermediate hosts. A diagnosis of neosporosis is generally made on the basis of clinical signs together with the detection of high levels of antibodies in serum. The present study compares the serum electrolyte profile (Ca, K, Mg, Na, P) and serum enzyme level (ALP, AST, CPK) of non-infected dogs with those of stray dogs naturally infected with N. caninum. The indirect fluorescent antibody test (IFAT) revealed that 17 of the 137 analysed serum samples (12.41%) of the stray dogs were seropositive (Sp) to N. caninum. Serum levels of theelectrolytes and the enzymes were evaluated in the 17 Sp and 28 seronegative (Sn, 20.44%) dogs using common enzyme kits, spectrophotometry and flame photometry techniques. The average serum level of ALP, AST, and CPK were found to be significantly higher in the Sp stray dogs. Measurements of the average serum levels of Ca, K, Mg, and P were higher in Sp than Sn stray dogs, with the average K level being significantly higher in seropositive stray dogs. It was concluded that evaluation of serum enzyme and electrolyte levels may be used to screen N. caninum infection in stray dogs.

Maxilact L2000 (β-galactosidase from Kluyveromyces lactis) was aggregated using ammonium sulphate and the resultant aggregates, on cross-linking with glutaraldehyde, produced an insoluble and catalytically active enzyme. These cross-linked enzyme aggregates (CLEAs) Maxilact were obtained in the optimal conditions. The activity of a biocatalyst, immobilized with the obtained preparation, was 5.89 U/mg. The operating stability of the immobilized Maxilact was determined for 10 successive batches. Residual activity was 8.24 U/mg after 10 cycles of batch operation. The above studies show that moderately stable preparation of catalase can be economically prepared by the cross-linked enzyme aggregates. The preparation and characterization of CLEAs Maxilact is reported for the first time.

Presence, uptake and production of serotonin and its effect on the production of other hormones were studied using immunocytochemical flow cytometric method. In a serotonin (10–12 M) containing medium up to 15 min. serotonin level does not elevate in the cells, but after 30 min. there is a significant elevation which remains till 4 h. In cells starved in salt solution the elevation is higher which calls attention to the effect of (starvation) stress. Using four enzyme blockers tryptophane hydroxylase inhibitor PCPA decreased (in serotonincontaining medium) and MAO B blocker deprenyl increased serotonin content, while serotonin reuptake inhibitor fluoxetine and MAO-A blocker clorgyline were ineffective. Extremely low concentrations of serotonin (10–15 M in case of histamine and 10–18 M in case of ACTH and T3) in the milieu was sufficient for increasing hormone (ACTH, T3, histamine) levels inside the cells. In conclusion; serotonin can be taken up by the cells and can be produced by induction, as Tetrahymena has enzymes for building it up and decomposing it. For synthesizing serotonin; basic molecules from outside are not needed. Serotonin in a minute amount can induce production of different hormones.

Optimization of product in enzyme kinetics is successful by the showers of mathematical analysis with control measures. Enzymes are an important functional aspects of all biochemical processes, as they catalyze numerous reaction taking place within living organisms. With this view, optimization and quantification of product is stressed upon and in such a context, optimal control approaches have been applied in our study. In this article, we have formulated a mathematical model of enzymatic system Dynamics with control measures with a view to optimize the product as well as process conditions. Here, Pontryagin Minimum Principle is used for determination of optimal control with the help of Hamiltonian. We discuss the relevant numerical solutions for the concentration of substrate, enzyme, complex and product with respect to a specified time interval by varying control factors.

Glucose is a very important raw material for the pharmaceutical and beverage industries. Currently industrial production of glucose involves enzyme hydrolysis of potato starch. Potato tuber is carbohydrate rich crop, but less exploited for glucose production. In this project we attempt to produce glucose from potato tuber the process involve extracting extract followed by acid hydrolysis. We also produce a sulphonated salicylic acid (SSA). The SSA was explored as a solid acid for the potato starch hydrolysis. The SSA was converted into polymeric frame work using Bakelite reaction. The rate of glucose formation using HCl was compared to the polymeric SSA.

Alterations in the formation and metabolism of bradykinin (Bk) are hypothesized to play a role in the pathophysiology of hypertension, atherosclerosis and vascular complications of diabetes. However, despite its prominent role in cardiovascular regulation, studies on bradykinin have been limited by various difficulties in accurate measurements of this peptide in biological samples. In this study, using the LC-ESI-MS method we estimated the conversion of exogenous Bk to its main metabolites - Bk-(1-5) and Bk-(1-7) - in endothelial cell culture and in fragments of aorta of normotensive (WKY) and hypertensive rats (SHR). The effects of angiotensin converting enzyme (ACE) and neutral endopeptidase (NEP) inhibitors were more pronounced in SHR: perindoprilat inhibited Bk-(1-5) formation by 49 % and 76 % in WKY and SHR rats, respectively, and tiorphan tended to decrease formation of Bk-(1-5) in both groups of animals. The degradation of bradykinin and generation of both metabolites were significantly higher in the aorta of SHR rats than in WKY controls. Our results show that even in relatively early hypertension (in 4-month old SHR rats) inactivation of Bk by aorta wall is enhanced.

Skonfrontowano wyniki badań dotyczące produkcji enzymów proteolitycznych przez Bacillus licheniformis PCM 1847 z dostępną w literaturze wiedzą i wynikami badań uzyskanymi dla rodzaju Bacillus. Przeprowadzone eksperymenty stanowią etap przygotowawczy do hodowli prowadzonej w bioreaktorze membranowym. W wielu przypadkach zagęszczenie biomasy dzięki zastosowaniu selektywnej membrany przynosi wymierne skutki. Zgodnie z naszą wiedzą, nieznane są przypadki zastosowania bioreaktora membranowego w ciągłej syntezie hydrolaz.
Results received for Bacillus licheniformis PCM 1847 were compared with the data presented in literature. The carried out experiments are the first step to prepare the culture in a membrane bioreactor. Under a high biomass concentration thanks to membrane application a lot of processes run at the high efficiency. To the best of our knowledge the membrane bioreactor has never been applied in the continuous hydro-lases production.

Opis:
The major purpose of the present study was to determine the chemical composition of dried Gomphrena celosioides Mart., including crude protein, lipid, total sugar, reducing sugar, ash, crude fiber, moisture, crude pectin, and total saponins in particular. This plant contains a large percentage of total saponins, which are widely used in food technology and pharmaceutical industries. In our study, the conditions for the extraction of total saponins from the plant were optimized, with special focus on cellulase concentration, pH, hydrolyzation time, and temperature. The highest amount of total saponins content (TSC) was 1.550±0.016% under optimal extraction conditions (cellulase concentration: 0.7% (v/w), hydrolyzation time: 4 h, hydrolyzation temperature: 60EC, and pH value: 5). Accordingly, the total saponins from G. celosioides Mart. were easily extracted.

Oxidative metabolism is essential for the gamete and the embryo energy production and is unavoidably associated with generation of reactive oxygen species (ROS). Enzymatic antioxidant defenses are present in the mammalian oocytes, embryos and follicular fluid (FF). An addition of porcine FF to maturation media have positive effects on the IVM and IVF results. The aim of this study was to study the CAT, SOD and GSH-Px activity, as well as Cu, Mn, and Zn concentration in porcine FF collected from the left and the right ovary. The ovaries were collected from 77 gilts at age 8 months. All the analyzed samples of FF revealed active enzymes (24.2·10–3 U · l–1, 2.65·10–3 U · l–1, and 525 U · l–1 for CAT, GSH-Px, and SOD, respectively) and contained Zn, Cu and Mn (13.8·103mol · l–1, 33.3·103 mol · l–1, and 133·10–9 mol · l–1, respectively). In the pFF collected from the left ovary, SOD and GSH-Px activity was higher compared to pFF from the right ovary. On the contrary, the concentration of Cu and Mn was significantly lower in the left ovary pFF. The concentration of Cu ions was negatively correlated with SOD activity. The CAT activity in pFF form left or right ovary did not show any differences.
Metabolizm oksydacyjny jest niezbędny do produkcji energii na potrzeby gamet i zarodków, co związane jest z wytwarzaniem reaktywnych form tlenu. Antyoksydacyjny mechanizm ochrony enzymatycznej znajduje się w oocytach i zarodkach ssaków, a także w płynie pęcherzykowym. Dodatek świńskiego płynu pęcherzykowego do odżywek hodowlanych korzystnie wpływa na wyniki zapłodnienia i dojrzewania in vitro. Celem badań było określenie aktywności katalazy (CAT) dysmutazy ponadtlenkowej (SOD) i persoksydazy glutationowej (GSH-Px) oraz koncentracji Cu,Mn i Zn wpłynie pęcherzykowym świń pobranym z prawego i lewego jajnika. Ogółem pobrano jajniki od 77 loch w wieku 8 miesięcy.Wewszystkich analizowanych próbkach badane enzymy wykazywały aktywność (24,2; 2,65 10–3 U · l–1 i 525 U · l–1 odpowiednio w przypadku CAT, GSH-Px i SOD) i zawierały jomy Zn, Cu i Mn (odpowiednio 13,8 · 103 mol · l–1; 33,3 · 103 mol · l–1 i 133 · 10–9 mol · l–1). W płynie pęcherzykowym pobranym z lewego jajnika aktywność SOD i GSH-Px była wyższa niż w płynie pęcherzykowym prawego jajnika. Natomiast koncentracja Cu i Mn była istotnie niższa w płynie z lewego jajnika. Koncentracja jonów Cu była skorelowana ujemnie z aktywnością SOD. Aktywność CAT w płynie pęcherzykowym z prawego lub lewego jajnika nie wykazała różnic.

We report the synthesis of isotopomers of L-phenylalanine and L-tyrosine selectively labelled with hydrogen isotopes in the 2-position of the side chain. The deuterium or tritium label was introduced using reductive amination activity of enzyme L-phenylalanine dehydrogenase (EC 1.4.1.20). This way p-phenylpyruvic acid was converted into [2-2H]-, [2-3H]-, and doubly labelled [2-2H/3H]-isotopomers of L-phenylalanine, using deuteriated, tritiated, and mixed (DTO) incubation media, respectively. Similarly, p-hydroxyphenylpyruvic acid was converted into [2-2H]-, [2-3H]-, and [2-2H/3H]-L-tyrosine. Deuterium labelled isotopomers of L-phenylalanine and --tyrosine can be used as markers in the investigation of abnormal metabolism of these amino acids observed in patients with inborn genetic diseases such as phenylketonuria and tyrosinemia.

Inflammatory bowel disease (IBD) are a heterogeneous group of disorders in the course dominated by chronic, recurrent gastrointestinal inflammation. It is believed that the activation of IBD occurs in patients with a genetic predisposition to their development. Chronic inflammation develops as a result of an excessive reaction of the immune system principally under the influence of environmental risk factors. Among them, it has been shown that the mechanism of oxidative stress is associated with the pathophysiology of inflammatory bowel disease, responsible for the commencement and progress of these diseases. The aim of the study was the relationship between single nucleotide polymorphisms (SNPs) of individual antioxidant enzymes, and the prevalence of inflammatory bowel disease that may be associated with increased levels of oxidative stress. Material and methods. A total of 111 IBD patients, including 65 patients with ulcerative colitis (UC) and 46 with Crohn’s disease (CD) and 125 healthy controls recruited from the Polish population, were genotyped for CAT -262C / T (rs1001179), SOD + 35A / C (rs2234694), GPx Pro 197 Leu polymorphisms. Genotyping of CAT, SOD, GPx gene polymorphism was performed by a RFLP-PCR. Results. The performed analysis of genetic polymorphisms of antioxidant enzymes showed that polymorphic variant of the CAT -262 C / T may have protective effects in patients with ulcerative colitis in the range of genotype C / T; OR = 0.49 (0.25-0.99), p = 0.044. Trend protective, but statistically unrelated, it was also observed for genotype T / T and T allele of the same polymorphism and genotypes and alleles + 35A / C SOD1 in UC as well as polymorphic variants CAT -262 C / T, Pro197Leu of GPx1, + 35A / C SOD1 in CD. The results were compared with a control group of potentially healthy individuals without such diseases. Conclusions. It has been shown that the polymorphism of antioxidant enzymes CAT gene -262 C / T may have protective effects in patients who are carriers of a genotype C / T at the UC. The potential protective effect without statistical relationships were also observed for other genotypes and alleles studied polymorphic variants of antioxidant enzymes in CD and CAT- 262C / T and + 35 A / C SOD1 in UC. Conducted our audit should be extended to more group of patients in order to assess whether or not to confirm the observed during analysis, the protective effect of CAT-262 C / T in ulcerative colitis and other trends observed for other polymorphic variants tested genes.

W ziarnie zbóż występują w znacznych ilościach białkowe substancje o charakterze inhibitorów enzymów hydrolitycznych. Pomimo, iż występowanie tych substancji w ziarnie zbóż stwierdzono już w latach trzydziestych, to do tej pory nie udało się dokładnie ich poznać. W pracy tej scharakteryzowano inhibitory enzymów amylolitycznych występujące w ziarnie pszenicy, żyta i pszenżyta oraz przedstawiono współczesne poglądy na temat ich znaczenia.
Cereal seeds contain a lot of protein substances which are inhibitors of hydrolytic enzymes. Although, they were discovered in the early 1930's till today remain a number of question to be answer. In this paper protein nature alpha-amylase inhibitors found in grain of wheat, rye and triticale were discussed on the grounds of the present-day knowledge.

Restricting food intake to a level below that consumed voluntarily (85%, 70% and 50% of the ad libitum energy intake for 3 or 30 days) and re-feeding ad libitum for 48 h results in an increase of malic enzyme (ME) gene expression in rat white adipose tissue. The increase of ME gene expression was much more pronounced in rats maintained on restricted diet for 30 days than for 3 days. The changes in ME gene expression resembled the changes in the content of SREBP-1 in white adipose tissue. A similar increase of serum insulin concentration was observed in all groups at different degrees of caloric restriction and refed ad libitum for 48 h. Caloric restriction and refeeding caused on increase of ME activity also in brown adipose tissue (BAT) and liver. However, in liver a significant increase of ME activity was found only in rats maintained on the restricted diet for 30 days. No significant changes after caloric restriction and refeeding were found in heart, skeletal muscle, kidney cortex, and brain. These data indicate that the increase of ME gene expression after caloric restriction/refeeding occurs only in lipogenic tissues. Thus, one can conclude that caloric restriction/refeeding increases the enzymatic capacity for fatty acid biosynthesis.

Doświadczenia przeprowadzono w warunkach laboratoryjnych na próbkach gleby o składzie granulometrycznym gliny lekkiej pylastej, do których dodano wodne roztwory siarczanu(VI) kadmu(II) w ilościach 0,05; 0,50 i 5,00 mmol·kg⁻¹ oraz bentonit w ilości 1, 5 i 10%. W tak przygotowanych próbkach gleb dokonano pomiaru aktywności dehydrogenaz, ureazy, fosfatazy kwaśnej i zasadowej. Wyniki przeprowadzonych badań wskazują, że zanieczyszczenie gleby siarczanem(VI) kadmu(II) spowodowało inhibicję aktywności wszystkich badanych enzymów glebowych. Stopień inhibicji uzależniony był od stężenia metalu w glebie. Zastosowanie bentonitu do gleby zanieczyszczonej solą kadmu w I stężeniu (0,05 mmol·kg⁻¹) spowodowało zmniejszenie stopnia inhibicji aktywności dehydrogenaz (przy każdej stosowanej dawce bentonitu) oraz zmniejszenie stopnia inhibicji ureazy (przy dawce 5 i 10% bentonitu). W przypadku zastosowania materiału sortującego do gleby zawierającej sól kadmu w stężeniu 5,00 mmol·kg⁻¹ zaobserwowano zmniejszenie toksycznego oddziaływania kadmu na aktywność dehydrogenaz, fosfatazy kwaśnej i zasadowej (przy wszystkich dawkach bentonitu) i na aktywność ureazy (przy dawce 5% bentonitu).
Experiments were carried out under laboratory conditions. Samples of light silty boulder loam soil were treated with water solution of cadmium(II) sulphate(IV). The doses used were 0.05; 0.50 i 5.00 mmol·kg⁻¹. In all soil samples the activities of dehydrogenase, urease, acid- and alkaline phosphatase were measured. Then, for each cadmium dose, three next experimental combinations were realized - with 1, 5 and 10% bentonite addition. The results showed, that soil pollution with cadmium, results in inhibited activity of all tested enzymes, and the intensity of this inhibition depends on metal concentration in soil. After addition of bentonite, to soil polluted with I cadmium concentration (0.05 mmol·kg⁻¹), the inhibition of dehydrogenase activity was reduced (for all used bentonite doses). Similar effect was stated for urease activity, however only for 5 and 10% bentonite content in soil. Application of bentonite to soil containing the highest cadmium dose (5 mmol·kg⁻¹) results in decreased toxic effect of this metal on dehydrogenase, acid and alkaline phos phatases activities (all used bentonite doses) as well as on the urease activity (only at bentonite dose 5%).

2,3,5-triphenyltetrazolium chloride (TTC) is a dye largely used for determination of microbiological activity in soil samples by dehydrogenase assay. TTC is colourless in the oxidised form and red when reduced, due to formation of the product 1,3,5-triphenyl formazan (TPF). In this study, different doses of TTC ranging from 0.2 to 3.0% were added to six different soil types with the aim to verify the effect of chemical TTC reduction and to optimise the substrate dose in order to prevent a toxic effect on microorganisms. Regarding the technical simplicity and cost-effectiveness, the TTC assay is commonly recommended as an efficient method for determination of soil biological activity, provided that an appropriate concentration is applied, depending on the soil type. The lowest TTC levels of 0.2 and 0.5% are recommended for Albic Luvisol and Eutric Fluvisol, respectively, and the highest (2%) for Brunic Arenosol and Haplic Luvisol soils. It has also been demonstrated that DHA has biological origin in more than 90%, whilst chemical TTC reduction accounts for less than 10%, and therefore does not interfere with the correctness of the results obtained.
2,3,5-chlorek trifenylotetrazoliowy (TTC) to powszechnie używany barwnik w oznaczeniach mikrobiologicznej aktywności prób glebowych za pomocą testu na aktywność dehydrogenazową. W utlenionej formie TTC jest bezbarwny zaś w zredukowanej przyjmuje barwę czerwoną z powodu wytworzenia się produktu, jakim jest 1,3,5-trifenyloformazan (TPF). W bieżącej pracy różne dawki TTC w stężeniach od 0,2 do 3% dodawano do sześciu różnych typów gleb celem zweryfikowania efektu jego chemicznej redukcji oraz zoptymalizowania dawki substratu tak by nie działała toksycznie na mikroorganizmy. Z uwagi na prostotę i stosunkowo niski koszt oznaczenia, test z użyciem TTC jest powszechnie rekomendowany do oznaczeń aktywności biologicznej gleb, pod warunkiem że będzie stosowana jego właściwa dawka, zależna od typu gleby. Wykazano, że najniższe stężenie TTC 0,2 oraz 0,5% jest właściwe odpowiednio w odniesieniu do gleby płowej spiaszczonej oraz mady rzecznej, podczas gdy wyższa jego dawka (2%) powinna być stosowana w przypadku gleb płowych właściwych (zerodowanych) i gleb płowych typowych. Wykazano ponadto, że aktywność dehydrogenaz w ponad 90% jest biologicznego pochodzenia, zaś chemiczna redukcja TTC stanowi mniej niż 10%, stąd nie wpływa na prawidłowość uzyskiwanych wyników.

The importance of ADA (adenosine deaminase) in the immune system and the role of its interaction with an ADA-binding cell membrane protein dipeptidyl peptidase IV (DPPIV), identical to the activated immune cell antigen, CD26, has attracted the interest of researchers for many years. To investigate the specific properties in the structure - function relationship of the ADA/DPPIV-CD26 complex, its soluble form, identical to large ADA (LADA), was isolated from human blood serum, human pleural fluid and bovine kidney cortex. The kinetic constants (Km and Vmax) of LADA and of small ADA (SADA), purified from bovine lung and spleen, were compared using adenosine (Ado) and 2'-deoxyadenosine (2'-dAdo) as substrates. The Michaelis constant, Km, evidences a higher affinity of both substrates (in particular of more toxic 2'-dAdo) for LADA and proves the modulation of toxic nucleoside neutralization in the extracellular medium due to complex formation between ADA and DPPIV-CD26. The values of Vmax are significantly higher for SADA, but the efficiency, Vmax /Km, in LADA-catalyzed 2'-dAdo deamination is higher than that in Ado deamination. The interaction of all enzyme preparations with derivatives of adenosine and erythro-9-(2-hydroxy-3-nonyl)adenine (EHNA) was studied. 1-DeazaEHNA and 3-deazaEHNA demonstrate stronger inhibiting activity towards LADA, the DPPIV-CD26-bound form of ADA. The observed differences between the properties of the two ADA isoforms may be considered as a consequence of SADA binding with DPPIV-CD26. Both SADA and LADA indicated a similar pH-profile of adenosine deamination reaction with the optimum at pHs 6.5 - 7.5, while the pH-profile of dipeptidyl peptidase activity of the ADA/DPPIV-CD26 complex appeared in a more alkaline region.

The aim of the study was to determine the activity of dehydrogenases and urease in forest peat soils of different fertility. There were selected 23 experimental plots localised in central and northern Poland. The research was conducted on forest fens, transition bogs and raised bogs. The biggest differences in soil physical and chemical properties were detected between fen and raised bog soils while raised bog soils and transition bog soils differed the least. Statistically significant differences between particular subtypes of peat soils were observed for soil pH-H2O, pH-KCl, C/N ratio as well as the content of organic carbon, nitrogen, calcium and potassium. The highest average dehydrogenase activity in the soil surface level was observed in fen soils, and the lowest – in raised bogs soils. The results obtained on urease activity were similar. Differences in urease activity in the studied soil types were shown. Dehydrogenase activity did not reveal statistically significant diversity. The activity of urease was negatively correlated with the content of carbon, C/N ratio, hydrolytic acidity and moisture. Also the increase in enzymatic activity accompanied by the increase in pH has been observed.

The Bacillus licheniformis ydaP gene encodes for a pyruvate oxidase that catalyses the oxidative decarboxylation of pyruvate to acetate and CO2. The YdaP form of this enzyme was purified about 48.6-folds to homogeneity in three steps. The enzyme was recovered in a soluble form and demonstrated significant activity on pyruvate using 2, 6-dichlorophenolindophenol (DCPIP) as an artificial electron acceptor. HPLC analysis of the YdaP-enzyme catalysed conversion of pyruvate showed acetate as the sole product, confirming the putative identity of pyruvate oxidase. Analysis of the substrate specificity showed that the YdaP enzyme demonstrated preference for short chain oxo acids; however, it was activated by 1% Triton X-100. The YdaP substrate-binding pocket from the YdaP protein differed substantially from the equivalent site in all of the so far characterized pyruvate oxidases, suggesting that the B. licheniformis YdaP might accept different substrates. This could allow more accessibility of large substrates into the active site of this enzyme. The thermostability and pH activity of the YdaP enzyme were determined, with optimums at 50ºC and pH 5.8, respectively. The amino acid residues forming the catalytic cavity were identified as Gln460 to Ala480.

The added value of nilem fish skin needs to be increased. The purpose of this study is to determine the concentration of acetic acid solution combined with the pepsin enzyme in the extraction of collagen from nilem fish skin that is necessary to obtain the highest yield/renderment. The study employed an experimental research method that used a completely randomized factorial design. The first treatment is the concentration of acetic acid solution. This consists of three levels, namely 0.5M, 0.7M and 0.9M. The second treatment is the concentration of the enzyme pepsin. This in turn consists of three levels, namely 0.5 %, 1.0 % and 1.5% (weight / weight). The parameters observed were collagen renderment. The results showed that the combination treatment concentration of 0.7 M solution of acetic acid by the pepsin enzyme at 1.0%, in the extraction of collagen from fish skin, produce the highest yield compared to other combinations. The renderment yield is 6.18%.

Papain activity in a buffer containing Me2SO was studied using fluorogenic substrates. It was found that the number of active sites of papain decreases with increasing Me2SO concentration whereas the incubation time, in a buffer containing 3% Me2SO does not affect the number of active sites. However, an increase of papain incubation time in the buffer with 3% Me2SO decreased the initial rate of hydrolysis of Z-Phe-Arg-Amc as well as Dabcyl-Lys-Phe-Gly-Gly-Ala-Ala-Edans. Moreover, an increase of Me2SO concentration in working buffer decreased the initial rate of papain-catalysed hydrolysis of both substrates. A rapid decrease of the initial rate (by up to 30%) was observed between 1 and 2% Me2SO. Application of the Michaelis-Menten equation revealed that at the higher Me2SO concentrations the apparent values of kcat/Km decreased as a result of Km increase and kcat decrease. Opis:
To enhance the inhibitory potential of 1-β-D-ribofuranosyl-1,2,4-triazole-3-carboxamide (ribavirin) vs hepatitis C virus (HCV) NTPase/helicase, ribavirin-5'-triphosphate (ribavirin-TP) was synthesized and investigated. Ribavirin-TP was prepared with the use of modified Yoshikawa-Ludwig-Mishra-Broom procedure (cf. Mishra & Broom, 1991, J. Chem. Soc., Chem. Commun, 1276-1277) involving phosphorylation of unprotected nucleoside. Kinetic analysis revealed enhanced inhibitory potential of ribavirin-TP (IC50=40 μM) as compared to ribavirin (IC50 > 500 μM). Analysis of the inhibition type by means of graphical methods showed a competitive type of inhibition with respect to ATP. In view of the relatively low specificity towards nucleoside-5'-triphosphates (NTP) of the viral NTPase/helicases, it could not be ruled out that the investigated enzyme hydrolyzed the ribavirin-TP to less potent products. Investigations on non- hydrolysable analogs of ribavirin-TP or ribavirin-5'-diphosphate (ribavirin-DP) are currently under way.

The ability of probiotic strain Lactobacillus plantarum LS/07 to modify the activity of intestinal bacterial enzymes - β-glucuronidase (β-GLUCUR), β-galactosidase (β-GAL), and β-glucosidase (β-GLU) in prevention of chronic diseases - cancer, atherosclerosis and dysbiosis was investigated. The male Sprague-Dawley rats were randomly divided into 12 experimental groups: controls groups - C (control), AT (atherosclerotic), CC (carcinogenic), dysbiotic groups - each group in combination with antibiotics (ATB), probiotics groups - in combinatioan with probiotic (PRO) alone, and each group with combination of antibiotic and probiotic (ATB+PRO). In the control group the β-glucuronidase activity did not change throughout the experiment. High fat diet in atherosclerotic group significantly increased the activity of β-glucuronidase (P<0.001) and β-glucosidase (P<0.01). Azoxymethane application in carcinogenic group significantly increased β-glucuronidase (P<0.01), but reduced β-glucosidase (P<0.01) activity. Daily application of probiotics alone and in combination with antibiotic increased β-galactosidase, of β-glucosidase, and decreased β-glucuronidase activity. In control antibiotic group we observed significant increase in β-glucuronidase (P<0.05) and decreased β-glucosidase (P<0.01) activity which can be caused by the change of microflora in favor of coliform bacteria. These findings indicate the positive effects of probiotic Lactobacillus plantarum LS/07 and suggest its use in disease prevention in human medicine and some animal species.

Polygonum orientale with beautiful red flowers can be found as one dominant species in the vicinity of most water bodies and wetlands in China. However, its phytoremediation potential has not been sufficiently explored because little is known about its resistance to inorganic or organic pollutants. We investigated P. orientale response to low and moderate levels of phenol stress (≤ 80 mg L-1). Endpoints included phenol tolerance of P. orientale and the removal of the pollutant, antioxidant enzyme activities, damage to the cell membrane, osmotic regulators and photosynthetic pigments. In plant leaves, phenol stress significantly increased the activities of peroxidase (POD) and catalase (CAT), as well as the contents of proline, soluble sugars and carotenoids, whereas superoxide dismutase (SOD), H2O2 and electrolyte leakage (EL) levels remained unaltered. On the other hand, there were significant decreases of soluble protein and chlorophyll contents. We demonstrated that, in combination with phenol tolerance and its removal, P. orientale has efficient protection mechanisms against phenol-induced oxidative damage (≤ 80 mg L-1). We propose that P. orientale could be used as an alternative and interesting material in the phytoremediation of phenol.

Zastosowanie hydrożeli jako skafoldów dla inżynierii tkanki kostnej umożliwia integrację substancji bioaktywnej takiej jak enzym fosfataza alkaliczna (ALP), który sprzyja mineralizacji. W niniejszej pracy, hydrożele z gumy gellan (GG) zawierające ALP poddano mineralizacji z wytworzeniem fosforanów wapnia (CaP) i magnezu (MgP) przez inkubację w płynach mineralizujących zawierających glicerofosforan wapniowy (CaGP) i magnezowy (MgGP). Porównano pięć różnych stosunków stężeń CaGP:MgGP, mianowicie 0.1:0, 0.075:0.025, 0.05:0.05, 0.025:0.075 and 0:0.1 (mol dm-3). Tworzenie CaP i MgP potwierdzono za pomocą FTIR, spektroskopii Ramana, SEM, EDS, IC- P-OES, XRD, TGA oraz poprzez wyznaczanie zmian suchej masy. Stwierdzono, że wapń ulega integracji w fazie mineralnej w znacznie większym stopniu niż magnez. Wzrost ilości magnezu spowodował zmiany w morfologii otrzymanej fazy mineralnej. Badania mechaniczne oraz reometryczne wykazały, że moduł Younga oraz składowa rzeczywista modułu reometrycznego hydrożeli ulegających mineralizacji przy stosunków stężeń CaGP:MgGP 0:0.1 były przynajmniej trzy razy większe w porównaniu do wszystkich innych próbek. Obecność MgP w próbkach zmineralizowanych sprzyjała adhezji oraz żywotności komórek osteoblastycznych MC3T3-E1 i nie wpłynęła negatywnie na ich cytozgodność. Wyniki dowodzą, że można indukować mineralizację hydrożeli z gumy gellan za pomocą CaGP oraz MgGP i tym samym wpływać na skład mineralny oraz właściwości mechaniczne kompozytów hydrożelowych-ceramicznych przez zmodyfikowanie stosunków stężeń CaGP:MgGP.
The use of hydrogels as bone tissue engineering (TE) scaffolds enables incorporation of bioactive substances such as the mineralization-promoting enzyme alkaline phosphatase (ALP). In this study, gellan gum (GG) hydrogels containing ALP were mineralized with calcium phosphate (CaP) and magnesium phosphate (MgP) by incubation in mineralization solutions of calcium and magnesium glycerophosphate (CaGP, MgGP). Five different CaGP:MgGP concentration ratios were compared, namely 0.1:0, 0.075:0.025, 0.05:0.05, 0.025:0.075 and 0:0.1 (all values mol dm-3). CaP and MgP formation was confirmed by FTIR, Raman, SEM, EDS, ICP-OES, XRD, TGA and monitoring dry mass percentage changes. Ca was incorporated into mineral to a greater extent than Mg. Incorporation of increasing amounts of Mg into mineral formed led to changes in mineral deposit morphology. Mechanical and rheometric testing revealed that Young's modulus and storage modulus of hydrogels mineralized at a CaGP:MgGP concentration ratios of 0:0.1 were at least three times higher than those of all other samples. The presence of MgP in mineralized samples promoted attachment and vitality of osteoblastic MC3T3-E1 cells and did not negatively impact cytocompatibility. The results prove the principle of enzymatic mineralization with CaP and MgP and adjusting the composition and mechanical properties of hydrogel-ceramic composites by altering the CaGP:MgGP concentration ratio.

Deacetylaza chitynowa może być użytecznym narzędziem w produkcji chitozanu o niskich stopniach acetylacji oraz wysokich masach cząsteczkowych. W prezentowanej pracy przedstawiono wyniki modelowania enzymatycznej deacetylacji chitozanu prowadzonej w reaktorze okresowym. Pokazano, wpływ uwzględnienia inhibicji produktem oraz dezaktywacji termicznej na model oraz porównano zaproponowane modele z danymi doświadczalnymi.
Chitin deacetylase is an enzyme that can be useful for enzymatic deacetylation of chitin. Kinetic models with and without product inhibition and thermal deactivation of chitin deacetylase were tested and compared with the experimental data obtained in a batch reactor. It was shown that the system is more sensitive to enzyme deactivation than to product inhibition.

The article discusses two series of the biodegradation tests of organic blockers used in drilling mud technology. Degradation process of the selected cellulose blockers in terms of quality was assessed by the colorimetric method. In all test solutions these tests revealed the presence of free glucose which is the final product of degradation of modified cellulose from which blockers were made. The beginning of the degradation process was noted already after 48 hours of the impact of enzymatic preparations on samples of blockers. The progress of the degradation process after 30 and 40 days of incubation was assessed by gravimetric method by draining sludge and marking blocker dry matter after enzymatic impact, comparing the sludge matter in samples with the specified enzyme to control samples without the addition of the enzyme. The research described in the work may be useful in the oil industry as they contain data (from laboratory-scale tests) on potential treatments with the use of enzymatic preparations. Detailed data on the degree of degradation of individual enzymatic preparations is included in the tables.

A change in the dinucleotide reactant from NAD+ to the more oxidizing APAD+ in the malic enzyme reaction results in a change in the mechanism of oxidative decarboxylation of malate from stepwise to concerted [7]. In order to determine whether this is a phenomenon general to metal ion dependent â-hydroxyacid oxidative decarboxylases, tartrate dehydrogenase, which catalyzes a reaction diastereotopic to malic enzyme, was studied using the technique of multiple isotope effects. A primary deuterium isotope effect of 1.41 on V/Kmalate was measured, as well as a primary 13C-isotope effect of 1.0096. A decrease in the measured value of the 13C-isotope effect to 1.0078 is consistent with a stepwise mechanism, as observed for malic enzyme. The 13C-isotope effect with thioNAD+ also decreases from a value of 1.0053 using D-malate to 1.0009 using D-malate-2-D, consistent with stepwise oxidative decarboxylation with this alternative dinucleotide substrate. Naringinaza z Penicillium decumbens została unieruchomiona na aktywowanych nośnikach polisacharydowych o właściwościach magnetycznych. W badaniach wykorzystano gumę guar, karobową, tragakantową, ksantanową oraz karagenianin. Nośniki otrzymano poprzez pokrycie mikrocząsteczek magnetytu (Fe3O4) polisacharydem, a następnie aktywując je aldehydem glutarowym lub polietylenoiminą. Celem niniejszej pracy była immobilizacja naringinazy z P. decumbens na magnetycznych nośnikach polisacharydowych oraz charakterystyka otrzymanego biokatalizatora. Najwyższą aktywność enzymu uzyskano poprzez immobilizację naringinazy na nośniku otrzymanym na bazie gumy karobowej aktywowanej polietylenoiminą. Immobilizacja naringinazy na tak otrzymanym nośniku wpływała na zmniejszenie optymalnej wartości pH oraz optymalnej temperatury aktywności badanego enzymu. Immobilizacja powodowała zwiększenie wartości stałej Michaelisa oraz zmniejszenie maksymalnej szybkości reakcji w porównaniu z enzymem wolnym.
Naringinase from Penicillium decumbens was immobilized on activated polysaccharide supported with magnetic properties. Magnetic microparticles were prepared by coating a magnetite (Fe3O4) with a polysaccharide followed by activating with glutaraldehyde or polyethyleneimine. Guar gums, locust bean, tragacanth, xanthan and carrageenan were used in the research. The main goals of this work were the immobilization of naringinase on magnetic polysaccharide carriers and the characterization of such immobilized enzyme. The naringinase immobilized on a carrier obtained on the basis of carob gum activated with polyethyleneimine had the highest activity. The immobilization of the naringinase on the carrier thus obtained influenced the reduction of the optimal pH value and the optimal temperature. Immobilization increased the Michaelis constant value and decreased the maximum reaction rate, as compared to the free enzyme.

Wciąż wzrastające zanieczyszczenie środowiska związane z częstym przedostawaniem się do środowiska toksycznych substancji jest jednym z największych problemów ekologicznych współczesnego świata. Wzrasta więc zainteresowanie nowymi, ekologicznymi technologiami remediacji, niewymagającymi dużych nakładów finansowych, pozwalającymi na całkowite lub częściowe oczyszczenie środowiska. Technologie bioremediacji, wykorzystujące potencjał mikroorganizmów, mogą być z powodzeniem wykorzystywane do usuwania ze środowiska zanieczyszczeń alifatycznymi bądź aromatycznymi związkami pochodzenia naftowego. Jednakże, w przypadku substancji o charakterze ksenobiotyków efektywność mikrobiologicznego rozkładu może ulec ograniczeniu. Wśród czynników biologicznych enzymy posiadają wysoki potencjał do efektywnego przekształcania i detoksykacji zanieczyszczeń i potencjalnie mogą one zostać wykorzystane do oczyszczania zanieczyszczonego środowiska. Celem pracy było przedstawienie, na podstawie literatury, niektórych grup enzymów zdolnych do przekształcania ksenobiotyków w nieszkodliwe związki. Znaczna uwaga została poświęcona enzymom pochodzącym z grzybów białej zgnilizny, charakteryzujących się wysokim potencjałem do efektywnego rozkładu ksenobiotyków. W artykule zestawiono zarówno zalety, jak i wady stosowania enzymów w bioremediacji zanieczyszczonego środowiska, a także perspektywy aplikacji in situ bioremediacji z wykorzystaniem enzymów.
Environmental pollution is growing more and more due to the frequently deliberate release of hazardous, toxic substances into the environment and it has become one of the biggest ecological problems of the world. Therefore, a growing interest is being devoted to develop new, cost-effective and eco-friendly remediation technology capable of partial or total recovery of polluted environment. Bioremediation that uses naturally existing catabolic potential of microorganisms can be efficiently used to clean up certain pollutants such as aliphatic or aromatic hydrocarbons. However, for chemicals exhibiting high xenobiotic character, like polyaromatic hydrocarbons, chlorophenols, dioxines, PCBs, etc., microorganisms can turn out to be ineffective. Among biological agents, enzymes have a great potential to effectively transform and detoxify pollutants and are potentially suitable to restore polluted environments. Moreover, the use of enzymatic proteins may represent a good alternative for overcoming most disadvantages related to the use of microorganisms. They are active in the presence of microbial predators and antagonists, can be used under extreme conditions limiting microbial activity and are effective at low pollutant concentrations. This work will examine the possibility of using enzyme preparations as an element of bioremediation technology. The main terms which must be fulfilled while using this type of technology will be presented. This review will also examine some class of enzymes, mainly oxidoreductases and hydrolases, that are capable of transforming xenobiotics effectively into innocuous products. Particular attention will be devoted to enzymes from white-rot fungi, such as Mn-peroxidase, lignin proxidase and laccase, which have a great potential towards xenobiotic compounds transformation. Also the use of lipase in biodegradation of phtalanes and as an agent for monitoring of bioremediation progress will be discussed. The main advantages as well as disadvantages that are present in the application of enzymes in the bioremediation of polluted environments will be examined in details. The future perspective for the in situ application of enzymatic bioremediation of polluted with xenobiotics environments will be discussed.

The natural resistance against the majority of potential pathogens that exist in most plant species is known as non-host resistance. Several reports suggest the role of antioxidant enzymes in non-host resistance. We assayed the expression or activity of four scavenging enzymes during non-host pathogen-plant interaction (Xanthomonas hortorum pv. pelargonii/mung bean) and host pathogen-plant interaction (Xanthomonas axonopodis pv. phaseoli/mung bean). The expression of superoxide dismutase (SOD) and ascorbate peroxidase (APX) and the enzyme activity of catalase (CAT) and peroxidase (POX) were investigated. The activities of CAT and POX were higher during non-host pathogen invasion vs. host pathogen attack. The expression of SOD and APX were also different between compatible and incompatible interactions. The expression of SOD and APX were higher in the incompatible compared to the compatible interaction. Additionally, induction of the antioxidant enzymes in response to non-host pathogen was earlier than induction in response to host pathogen. Such information is important for plant breeders, and useful when looking for alternative control strategies as well.

Heavy metals are a serious source of soil pollution, especially in the vicinity of metal smelters. His study examines the effect of selected heavy metals on the activity of soil enzymes (acid phosphatase, dehydrogenase, β-glucosidase, urease and protease) in topsoil (0–30 cm) in the vicinity of the zinc smelter “Miasteczko Slaskie”. Soil enzyme activity in the polluted site was compared with a non-polluted site in the outskirts of the Pazurek nature reserve (near Olkusz). Soil samples were collected in coniferous communities. The activity of soil enzymes were compared at distances of 0.5 km, 1 km and 1.5 km from the emitter. In the vicinity of the emitter (all distances) Cd, Pb and Zn levels in topsoil exceeded acceptable limits. Also, Pb content at a non-polluted site (Pazurek nature reserve) was higher than permissible levels. Zn (HNO 3 extracted) were the highest in the topsoil samples collected at a distance of 1.5 km from the Miasteczko Slaskie plant, Cd (HNO 3 and CaCl 2 extracted) and Pb (CaCl 2 extracted) contents were highest at the distance 1 km from the emitter. However, the pollution index in the superficial soil layer (0–10 cm) was similar at all three distances from the emmiter. The Zn, Cd and Pb bioavailable concentrations (CaCl 2 extracted) ranged: 15–220, 0.1–7.2, 2.8–55.7 mg kg –1 respectively. The greatest decreases were found for acid phosphatase, dehydrogenase and protease activity at the distance of 0.5 km from the emitter. Results show that soil enzymatic activity should accompany basic chemical analysis to track disorders and adverse effects in polluted soil.
Zanieczyszczenie gleby metalami ciężkim zwłaszcza w pobliżu hut metali to poważny problem. Oceniano wpływ metali ciężkich na aktywność enzymatyczną gleby. Celem badań była ocena aktywności enzymów glebowych: fosfatazy kwaśnej, dehydrogenaz, β-glukozydazy, ureazy i proteazy w wierzchniej warstwie gleby w najbliższym sąsiedztwie huty cynku Miasteczko Śląskie. Aktywność enzymatyczną gleby na stanowiskach zanieczyszczonych porównywano z aktywnością na stanowisku niezanieczyszczonym, próbki gleby pobierano również w pobliżu rezerwatu przyrody Pazurek koło Olkusza ze zbiorowisk borowych. Aktywność enzymatyczna w glebie najbliższego sąsiedztwa emitora była porównywana w odległości 0,5, 1, 1,5 km od emitora. Wykazane zawartości Cd, Pb i Zn przekraczały dopuszczalne normy przyjęte dla gleby. Zawartość Pb w próbkach gleby pobieranych w pobliżu rezerwatu Pazurek była wyższa niż dopuszczalne wartości dla gleby. Zawartość Zn (ekstrakcja HNO3) była najwyższa w odległości 1,5 km od emitora (0–10 cm), z kolei zawartości Cd (ekstrakcja HNO 3 i CaCl 2 ) i Pb (ekstrakcja CaCl 2 ) były najwyższe 1 km od emitora. Wskaźnik zanieczyszczenia wykazany dla wierzchniej warstwy gleby (0–10 cm) był na porównywalnym poziomie w glebie w sąsiedztwie emitora. Koncentracje biodostępnych form metali (ekstrakcja CaCl2) mieściły się w przedziałach odpowiednio: 15–220, 0,1–7,2, 2,8–55,7 mg kg –1 . Najniższe aktywności fosfatazy, dehydrogenazy i proteazy wykazano w próbkach gleby pobieranej w odległości 0.5 km. Badania aktywności enzymatycznej gleby obok analiz chemicznych powinny być prowadzone w celu śledzenia zmian w glebach zanieczyszczonych.

Celem pracy było określenie wpływu sytemu uprawy roli na aktywność enzymatyczną gleb. Badania, uwzględniające dwa systemy uprawy roli: klasyczny (płużny) i uproszczony (bezorkowy), przeprowadzono na glebach zróżnicowanych typologicznie, rodzajowo i gatunkowo: płowych i rdzawych. W próbkach glebowych oznaczono aktywność 5 enzymów: dehydrogenaz, fosfatazy kwaśnej, fosfatazy alkalicznej, ureazy i proteazy. Zastosowane testy enzymatyczne okazały się dobrymi wskaźnikami różnicującymi badane obiekty glebowe w zależności od systemu uprawy roli. Stosowanie uprawy uproszczonej stymulowało istotnie aktywność analizowanych enzymów, niezależnie od typu gleby. Efekt ten zaznaczył się najwyraźniej w powierzchniowej warstwie gleb (0-10 cm). Szczególnie szeroki zakres aktywności uzyskano dla dehydrogenaz, co wskazuje na przydatność tej grupy enzymów do oceny zmian w środowisku glebowym pod wpływem systemu uprawy roli. Obserwowanej stymulacji aktywności badanych enzymów towarzyszyły korzystne zmiany stanu chemicznego gleb.
The aim of this study was to determine the effect of the soil tillage system on soil enzymatic activity. The performed investigations, employing two soil tillage systems: classical (ploughing) and simplified (no-tillage), were carried out on lessives soils (Luvisols) and rusty soils (Arenosols) differing typologically, with regard to their kind and species. The activity of the following five enzymes was determined in soil samples: dehydrogenases, acid phosphatase, alkaline phosphatase, urease and protease. The applied enzymatic tests turned out to be good indicators differentiating the examined soil objects depending on the employed tillage system. The employment of the simplified tillage system stimulated significantly the activity of the analysed enzymes irrespective of the soil type. This effect was particularly apparent in the top layer (0 - 10 cm) of the soil. An exceptionally wide range of activity was obtained for dehydrogenases indicating the usefulness of this group of enzymes for the evaluation of changes in the soil environment under the influence of the soil tillage system. The observed activity stimulation of the examined enzymes was accompanied by advantageous changes in soil chemical conditions.

Farnesyl diphosphate synthase (FPPS) is a key enzyme in isoprenoid biosynthesis which supplies sesquiterpene precursors for several classes of essential metabolites including sterols, dolichols, ubiquinones and carotenoids as well as substrates for farnesylation and geranylgeranylation of proteins. It catalyzes the sequential head-to-tail condensation of two molecules of isopentenyl diphosphate with dimethylallyl diphosphate. The enzyme is a homodimer of subunits, typically having two aspartate-rich motifs with two sets of substrate binding sites for an allylic diphosphate and isopentenyl diphosphate per homodimer. The synthase amino-acid residues at the 4th and 5th positions before the first aspartate rich motif mainly determine product specificity. Hypothetically, type I (eukaryotic) and type II (eubacterial) FPPSs evolved from archeal geranylgeranyl diphosphate synthase by substitutions in the chain length determination region. FPPS belongs to enzymes encoded by gene families. In plants this offers the possibility of differential regulation in response to environmental changes or to herbivore or pathogen attack.

This paper presents a kinetic analysis of the whole reaction course, i.e. of both the transient phase and the steady state, of open multicyclic enzyme cascade systems. Equations for fractional modifications are obtained which are valid for the whole reaction course. The steady state expressions for the fractional modifications were derived from the latter equations since they are not restricted to the condition of rapid equilibrium. Finally, the validity of our results is discussed and tested by numerical integration. Apart from the intrinsic value of knowing the kinetic behaviour of any of the species involved in any open multicyclic enzyme cascade, the kinetic analysis presented here can be the basis of future contributions concerning open multicyclic enzyme cascades which require the knowledge of their time course equations (e.g. evaluation of the time needed to reach the steady state, suggestion of kinetic data analysis, etc.), analogous to those already carried out for open bicyclic cascades.

The sensitivity of soil enzymes to soil contamination with zinc was analyzed. A laboratory experiment was performed on sandy loam at pH 7.0, sampled from arable land at a depth of 0 to 20 cm. Soil samples were passed through a sieve with 2 mm mesh size and contaminated with the following zinc doses: 0, 300, 600, 1200 and 2400 mg Zn2+ kg-1 soil. Zinc was applied in the form of aqueous solution of ZnCl2. Soil was mixed thoroughly with zinc, and its moisture content was brought to 50% capillary water capacity. The samples were incubated at 25°C. Beakers with soil samples were weighed once a week to replenish evaporated water. The activity of soil enzymes: dehydrogenases, urease, acid phosphatase, alkaline phosphatase, catalase, arylsulfatase and b-glucosidase, was determined after 15, 30, 60 and 120 days of the experiment. The results were used to calculate soil resistance (RS), ED20 and ED50 values. The results of the study indicate that soil enzymes are characterized by varied sensitivity to excessive zinc concentrations, and that the RS index is a reliable measure of enzymatic responses to zinc pollution. The analyzed enzymes were classified in the following decreasing order in terms of their resistance to zinc: b-glucosidase> acid phosphatase > urease >arylsulfatase = alkaline phosphatase> catalase > dehydrogenases. Zinc continued to exert a negative effect on soil enzymes throughout the experiment (120 days). ED20 values for the analyzed enzymes in mg Zn2+ kg-1 DM soil were determined at: 103 for dehydrogenases, 184 for alkaline phosphatase, 233 for urease, 247 for arylsulfatase, 416 for acid phosphatase, 419 for catalase and 1373 for b-glucosidase.
Celem badań było określenie wrażliwości enzymów na zanieczyszczenie gleby cynkiem. Doświadczenie przeprowadzono w warunkach laboratoryjnych na glinie piaszczystej o pH 7,0, pobranej z użytku rolnego z warstwy od 0 do 20 cm. Przed rozpoczęciem badań glebę przesiano przez sito o oczkach 2 mm i zanieczyszczono następującymi dawkami cynku: 0, 300, 600, 1200, 2400 mg Zn2+ kg-1 gleby. Cynk stosowano w postaci wodnego roztworu ZnCl2. Następnie po dokładnym wymieszaniu gleby jej wilgotność doprowadzono do 50% kapilarnej pojemności wodnej i poddano inkubacji w cieplarce w temp. 25°C. Jeden raz w tygodniu zlewki przeważano w celu uzupełnienia ewentualnych ubytków wody. Po 15, 30, 60 i 120 dniach inkubacji część zlewek likwidowano i oznaczono aktywność enzymów glebowych: dehydrogenaz, ureazy, fosfatazy kwaśnej, fosfatazy alkalicznej, katalazy, arylosulfatazy i b-glukozydazy. Na podstawie wyników obliczono indeks oporności enzymów (RS) oraz wskaźniki ED20 i ED50. Stwierdzono, że enzymy glebowe mają zróżnicowaną oporność na nadmiar cynku w glebie, a wskaźnik oporności RS jest dobrą miarą ich reakcji na zanieczyszczenie tym metalem. Pod względem zmniejszającej się oporności można je uszeregować następująco: b-glukozydaza> fosfataza kwaśna > ureaza >arylosulfataza = fosfataza kwaśna > katalaza > dehydrogenazy. Negatywne działanie cynku na enzymy glebowe utrzymywało się przez cały okres badań (120 dni).Wartość ED20 dla poszczególnych enzymów w mg Zn2+ kg-1s.m. gleby wynosiła: dehydrogenazy – 103, fosfataza alkaliczna – 184, ureaza – 233, arylosulfataza – 247, fosfataza kwaśna – 416, katalaza – 419, b-glukozydaza – 1373.

This research aimed to investigate the potential screening of wood degrading fungi to degrade the textile dye. Nine fungi were selected based on their ability to remove the turquoise blue textile dye on liquid potatoes dextrose médium. The fungi which have a high color removal efficiency were identified and analyzed qualitatively in terms of the manganese peroxidase and lignin peroxidase enzymes and their enzyme activity was determined. The selected fungi were investigated for their ability to degrade turquoise blue under variations in pH, dye concentration, and contact time. The results showed that three of the nine strains of wood-degrading local fungi, identified as Trametes hirsuta, Microporus xanthopus, and Ganoderma applanatum, were able to degrade textile dye. The color removal efficiency of dye treatment was monitored under the initial pH, dye concentration, and contact time variation. The study analyzed that the optimal color removal efficiency of the turquoise blue textile dye with a concentration of 30 mg/L in an aqueous solution was achieved at 78.50 and 85.84% at pH 6 for Microporus xanthopus and Ganoderma applanatum, while the color removal efficiency was 82.17% at pH 5 for Trametes hirsuta with seven days of incubation time.

Although pear is one of the most preferred fruits globally due to its high nutrient content, its juice products have not received equal consumer acceptability due to their undesirable haziness and turbidity. However, enzymatic treatment of juice has been noted to improve the overall acceptability of juices better than traditional decantation, filtration, and heating methods, which are either expensive or detrimental to the product quality. In this study, pear juice was effectively clarified using partially purified polygalacturonase from an entomopathogenic fungal endophyte Beauveria bassiana SAN01. The optimization of the juice clarification process led to 137% improvement in clarification compared to control, under optimal conditions of 39.37 U/mL enzyme load, 36.87°C temperature and 2.75 h treatment time. Results also showed that the polygalacturonase treatment resulted in a significant decrease in viscosity and an increase in the pseudoplasticity of the pear juice as the rheological data of the juice was observed to fit adequately into the power law model (R2>0.9). Furthermore, the enzyme-assisted juice clarification was found to reduce browning index (-14.34%) and turbidity (-19.72%) while increasing the reducing sugar content (9.55%). Unlike some conventional juice treatments, the polygalacturonase treatment preserved the antioxidant potential and the total phenolic contents of the pear juice as no significant changes were observed after the treatment. This study thus demonstrates the efficacy of a fungal polygalacturonase in improving pear juice quality as well as the potential applicability of B. bassiana enzymes in the food industry.

In the present study, rice husk ash, which is a renewable and abundant material, was utilized as a carrier for lipase immobilization for the first time. Poly (ε-caprolactone) synthesis was successfully achieved by the new enzymatic catalyst: Candida antarctica lipase B immobilized onto surface-modified rice husk ashes by covalent binding. It was aimed to obtain optimum polymerization conditions at which highest molecular weight was reached and characterize the polymer produced. Moreover, thermal stability and effectiveness of the new biocatalyst in non-aqueous media were also shown with successful polymerization reactions. In addition, by using the new enzyme preparation, ε-caprolactone was able to be polymerized even at 30°C, which was promising for an energy saving process. Consequently, this work provides a new alternative route for poly (ε-caprolactone) synthesis.

Gold nanoparticles are emerging as promising agents for various areas of material science as well as nanotechnology, electronics and medicine. The interest in this material is provided due to its unique optical, electronic and molecular-recognition properties. This paper presents results of preparation, characterization and biofunctionalization of gold nanoparticles. Nanoparticles have been conjugated with the cholesterol oxidase enzyme in order to prepare the active element for biosensors. Cholesterol oxidase is one of the most important analytical enzyme, used for cholesterol assay in clinical diagnostics, and there is still a necessity in improvement of existing analytical techniques, including bio-nanotechnological approaches based on modern nanosystems. The prepared bio-nanosystem was characterized by the enzyme activity test. Obtained results showed a stable binding of the enzyme with nanoparticles and preserved the bioactivity approves which gives possibility to use the prepared bio-nanosystems for analytical purposes. The Surface Plazmon Resonance and the Surface Enhanced Raman Scattering as specific for nanocurriers effects were observed what enable us to research the oscillation spectra of enzyme. These spectra were compared with ones obtained by IR-spectroscopy. The vibrational lines are attributed to chemical functional groups existing in enzyme, for example, amino acids, amide groups as well as flavin – adenine dinucleotide cholesterol oxidase prosthetic group. By this way the identified spectral lines can be used as a distinguishing mark of enzyme.
Nanocząstki złota zdają się być niezwykle obiecującym materiałem, który z powodzeniem może być wykorzystywany w różnych dziedzin inżynierii materiałowej, nanotechnologii, elektroniki czy medycyny. Zainteresowanie tym materiałem wynika z posiadanych przez niego unikatowych właściwości optycznych, elektronicznych i identyfikacji na poziomie molekularnym. W artykule przedstawiono wyniki prowadzonych badań dotyczących przygotowania, charakterystyki i biofunkcjonalizacji nanocząstek złota. Celem wytwarzania i łączenia nanocząstek złota z enzymem oksydazą cholesterolową było opracowanie aktywnego biologicznie elementu biosensora. Oksydaza cholesterolowa jest jednym z najważniejszych enzymów analitycznych, stosowanych w diagnostyce klinicznej do oznaczenia stężenia cholesterolu. Istnieje potrzeba doskonalenia istniejących metod analitycznych, między innymi poprzez wykorzystanie nowoczesnych nanomateriałów. Przeprowadzono badania aktywności enzymu immobilizowanego, jak również dokonano charakterystyki danego bionanosystemu za pomocą wybranych metod optycznych. Na podstawie uzyskanych wyników można stwierdzić, że wiązanie enzymu z nanocząstkami jest stabilne, a aktywność biologiczna została zachowana, co pozwala na przygotowanie bio-nanosystemu do celów analitycznych. Zaobserwowano również wystąpienie dwóch efektów optycznych charakterystycznych w przypadku zastosowania odpowiednich nanonośników: Powierzchniowego Rezonansu Plazmonów oraz Powierzchniowo Wzmocnionego Rozproszenia Ramanowskiego. Uzyskane widma były porównywane z widmami w podczerwieni. Zaobserwowane linie przypisano do charakterystycznych grup funkcyjnych, aminokwasów, wiązań peptydowych, jak również dinukleotydu flawinoadeninowego czyli grupy prostetycznej oksydazy cholesterolowej. Zidentyfikowane linie mogą być wykorzystywane jako marker analityczny dla danego enzymu.

The use of sewage sludge in agriculture is known to be environmentally friendly as it is a practical way of recycling municipal waste. A short-term experiment was carried out to study the changes in soil chemical and microbiological properties influenced through the addition of sewage sludge along with saline irrigation under a pearl millet-wheat crop rotation. There were three irrigation (canal water (0.35 dS m-1), 8 and 10 dS m-1 electrical conductivity saline water) and five fertilizer treatments (control-F1, sewage sludge (5 t ha-1)-F2, sewage sludge (5 t ha-1) +50% recommended dose of fertilizer (RDF)-F3, sewage sludge (5 t ha-1)+75% RDF-F4 and RDF-F5). The results showed that soil organic carbon, and available nitrogen, and phosphorus were reduced significantly under saline conditions but there was an increase in available potassium with the increasing salinity levels of the irrigation water. A significant reduction in soil microbial biomass carbon and enzyme activities was caused by 8 and 10 dS m-1 as compared to 0.35 dS m-1 at different growth stages of crops. Treatment F4 attained the highest soil microbial activity at each crop growth stage by a significant margin among all of the fertilizer treatments, which is associated with a substantial build-up of organic carbon and available NPK in the soil. Soil microbial activities followed a particular trend: at 35 and 75 days after sowing>harvest>sowing in pearl millet and wheat, respectively. Sewage sludge dumping is a major problem in India, and the present study suggests that by applying sewage sludge, soil biological health is improved; therefore its use in agriculture is recommended to farmers.

Leaf scald, caused by the necrotrophic fungus Monographella albescens, is one of the main threats to rice (Oryza sativa L.) around the world. This disease decreases yields in rice by up to 30% because of dead leaf tissue, damaged seeds, and sterile flowers. Currently, there is limited knowledge about the molecular mechanisms involved in rice plant resistance against this pathogen. For this purpose, six commercial cultivars of rice were primarily screened for M. albescens infection and development. Dasht and Salari were found to be the most resistant and susceptible to M. albescens infection, respectively. The plants were kept in a greenhouse at 29 ± 2°C during the day and 26 ± 2°C at night with a relative air humidity of 85 ± 5%. Forty-five days after sowing, the plants with three biological replications were inoculated by transferring a PDA disc (0.3 cm2) containing M. albescens mycelia to the middle third of the 7th, 8th, and 9th completely open leaves. The leaves were collected 24, 48, 72, 96 and 120 hai. Leaf samples were also collected from the non-inoculated plants (0 h) to serve as controls. Real-time quantitative PCR (RT-qPCR) showed rapid induction and significant accumulation of jasmonic acid (JA) and ethylene (ET) responsive genes such as lipoxygenase (LOX), allene oxide synthase 2 (Aos2), jasmonic acid carboxyl methyltransferase 1 (JMT1) and ACC synthase 1 (ACS1) in the resistant Dasht cultivar after infection with M. albescens. Furthermore, the transcripts of salicylic acid (SA) responsive phenyl alanine ammonia lyase 1 (PAL1) and nonexpressor of pathogenesis-related genes 1 (NPR1) genes were induced in the incompatible interaction. The activities of the defense enzymes superoxide dismutase (SOD), peroxidase (POX) and glutathione reductase (GR) increased strongly in Dasht in response to M. albescens infection. In addition, there was an increase in the H2 O2 levels in the leaves of the Dasht cultivar during the infectious period of M. albescens associated with the enhancement of catalase (CAT) activity as well as higher levels of malondialdehyde (MDA). This is the first study on the interaction between rice and M. albescens at the molecular level. It can contribute to understanding how rice responds to pathogen infection, as well as assist with future research plans of molecular breeding regarding the tolerance to leaf scald disease.

Background. Resistant starch (RS) is part of potato starch that is not digested in the gastrointestinal tract. RS4 is a chemically modified starch (for example by oxidation and esterification) and physically (by heating). Objective. The study was aimed at determining the effect of resistant starch on lipid metabolism and activity of hepatic enzymes in Wistar strain rats fed high-fat diets containing 15% of lard or 15% of soybean oil. Material and methods. Four types of diets were administered to the animals (4 groups of males, n=32): control diet (K1) containing 15% of soybean oil; control diet (K2) containing 15% of lard as well as two groups receiving the same diets with 10% addition of resistant starch RS4 (K1S and K2S). Results. The mean concentration of total cholesterol was lower in the group of animals fed a diet with vegetable oil (39.9 mg/dl) as compared to that reported in the group of rats fed the lard-supplemented diet (55.2 mg/dl). Compared to the control groups in both groups of animals receiving the diet supplemented with resistant starch RS4 the total cholesterol concentration in serum decreased by ca. 25% (differences were statistically significant). In groups of rats receiving oil- or lard-containing diets with the addition of the resistant starch preparation the concentration of triglycerides in serum decreased by ca. 47% and 10%, respectively. Conclusions. A beneficial effect of the resistant starch RS4 added to Wistar rats diets on the lipid metabolism has been shown. The concentrations of total cholesterol and triglycerides in the serum were lower and concentration of HDL-cholesterol was higher in the rats fed with the diets containing the addition of the RS4 preparation as compared to the control groups. Based on the activity of hepatic enzymes the degree of liver damage was lower in groups of rats fed with diets containing resistant starch RS4 as compared to the control groups.
Wprowadzenie. Skrobia oporna (resistant starch, RS) jest to część skrobi ziemniaczanej, która w przewodzie pokarmowym nie ulega strawieniu. Skrobia oporna RS4 to skrobia modyfikowana chemicznie (np. przez oksydację lub estryfikację) oraz fizycznie (przez działanie termiczne). Cel. Celem badań była ocena wpływu skrobi opornej RS4 na gospodarkę lipidową oraz aktywność enzymów wątrobowych szczurów rasy Wistar karmionych dietami wysokotłuszczowymi zawierającymi 15% smalcu lub 15% oleju sojowego. Materiał i metody. Zwierzęta (4 grupy samców n=32) karmiono 4 rodzajami diet: kontrolną (K1) zawierającą 15% oleju sojowego i kontrolną (K2) zawierającą 15% smalcu oraz odpowiednio 2 grupy takich samych diet z 10% dodatkiem skrobi opornej RS4 (K1S i K2S). Wyniki. Średnie stężenie cholesterolu całkowitego w grupie zwierząt karmionych paszą z tłuszczem roślinnym było niższe (39,9 mg/dl) w porównaniu do odnotowanego w grupie szczurów karmionych dietą ze smalcem (55,2 mg/dl). W porównaniu do grup kontrolnych, w obu grupach zwierząt karmionych paszą zawierającą olej lub smalec z dodatkiem skrobi RS4 stężenie cholesterolu całkowitego w surowicy krwi obniżyło się o około 25% (różnice były istotne statystycznie). W grupach szczurów karmionych paszą z dodatkiem preparatu skrobi opornej RS4 stężenie triglicerydów w surowicy krwi obniżyło się odpowiednio o około 47% i 10%. Wnioski. Wykazano korzystny wpływ skrobi opornej RS4 dodawanej do diet szczurów Wistar na ich gospodarkę lipidową. Stwierdzono zmniejszenie stężenia cholesterolu całkowitego, TGC oraz zwiększenie stężenia cholesterolu HDL w surowicy krwi zwierząt otrzymujących diety z dodatkiem skrobi opornej RS4 w porównaniu do grup kontrolnych. Na podstawie aktywności enzymów wątrobowych stwierdzono niższy stopień uszkodzenia wątroby w grupach szczurów karmionych dietami z dodatkiem skrobi opornej RS4 w porównaniu do grup kontrolnych.

Opis:
Po mutagenizacji promieniowaniem UV i NTG szczepu Trichoderma reesei M-7 i selekcji na podłożu z dodatkiem żelatyny wyizolowano około 960 niskoproteazowych mutantów. Dziesięć z nich zostało przetestowanych podczas hodowli okresowych w zakresie produkcji celulaz, proteaz i beta-galaktozydazy w obecności serwatki i laktozy jako źródła węgla. Aktywności proteolityczne oznaczone po hodowlach 9 mutantów w obecności laktozy były niższe od szczepu wyjściowego. Zaobserwowano większy indukcyjny wpływ serwatki niż laktozy, na sekrecję proteaz przez wszystkie badane szczepy. Czysta laktoza okazała się bardziej efektywnym induktorem produkcji beta-galaktozydazy niż zawarta w serwatce. Plony biomasy większości mutantów po hodowlach na serwatce były tylko nieznacznie wyższe od oznaczonych po hodowlach na czystej laktozie. Było to prawdopodobnie związane z litycznym mechanizmem kompensującym proteaz, przejawiających wyższe aktywności w filtratach pochodzących z hodowli w obecności serwatki. Praca jest wstępem do dalszych badań nad zależnościami pomiędzy proteazami i innymi enzymami w filtratach pohodowlanych Trichoderma reesei.
It has been isolated 960 low-protease mutants after mutagenisation using UV radiation and NTG and selection on medium with addition of gellatine. Ten of them have been tested during batch cultivation for the production of cellulases, proteases and beta-galactosidase in the presence of whey and lactose as a source of carbon. Proteolytic activities estimated after cultivation of 9 mutants in the presence of lactose were lower then parental strain. It has been observed better inductive effect of whey then lactose on the secretion of proteases by all mutants tested. Pure lactose appeared to be more effective inducer of the production of beta-galactosidase then contains in whey. Biomass concentration of most mutants after cultivation on whey was only slightly higher then estimated after cultivation on pure lactose. It was probably connected with lytic compensatory mechanism of proteases exhibited higher activities in culture filtrates from cultivation in the presence of whey. This work is an introduction for future studies on correlation between proteases and other enzymes in culture filtrates of Trichoderma reesei.

Opis:
The study was carried out on two highbush blueberry cultivars: Bluecrop and Darrow in 2001 and 2002 seasons. Analyses for phytochemical contents in berries were made during commercial maturity of fruit. Concentration of anthocyanins and low molecular weight thiol compounds was significantly higher in ´Darrow´ berries in comparison to Bluecrop cv., irrespective of the harvest year. Tested cultivars did not differ in the ratio of the reduced form of glutathione in its total quantity. As opposed to this, the higher proportion of the reduced form of ascorbate in its total concentration was noted for ´Bluecrop´, but it was statistically proven only in the first year of research. A very low activity of antioxidative enzymes was noted, however on the average, considerably higher activity of glutathione reductase showed ´Darrow´, whereas ´Bluecrop´ had higher catalase activity. The ascorbate peroxidase activity was not detected in the extracts of ´Bluecrop´s’ berries. Berries of ´Darrow´, which exhibited, in general, the higher antioxidants content in the first year of the study, were harvested at two different dates in 2002: in the middle of July and at the end of August. The harvest date had a significant effect on the level of some phytochemicals. The fruit harvested in August had lower content of cysteine, glutathione, phenolics, flavonols and anthocyanins than the ones harvested in July. Ascorbate content was similar at both harvest dates. Growing season also had the influence on antioxidant properties, especially on the content of ascorbic acid.
Badania przeprowadzono w latach 2001–2002. Ocenie poddano dwie odmiany borówki wysokiej: Bluecrop i Darrow. Analizy chemiczne dotyczące zawartości fitozwiązków w owocach wykonano w fazie dojrzałości zbiorczej. Stężenie antocyjanów oraz niskocząsteczkowych związków tiolowych było istotnie wyższe w owocach odmiany Darrow w porównaniu z odmianą Bluecrop, niezależnie od roku badań. Udział zredukowanego glutationu w jego całkowitej zawartości kształtował się na zbliżonym poziomie u obu ocenianych odmian. Przeciwnie, istotnie wyższą zawartością zredukowanego askorbinianu charakteryzowała się odmiana Bluecrop, ale tylko w pierwszym roku badań. Aktywność enzymów oksydacyjnych w dojrzałych owocach borówek była bardzo niska. Jakkolwiek przeciętnie, wyższą aktywnością reduktazy glutationowej charakteryzowała się odmiana Darrow, natomiast ekstrakt z owoców odmiany Bluecrop wykazywał wyższą aktywność katalazy. Ekstrakty z owoców odmiany Bluecrop nie wykazywały aktywności peroksydazy askorbinianowej. Owoce odmiany Darrow, które charakteryzowały się na ogół wyższym stężeniem antyoksydantów w pierwszym roku badań, w roku kolejnym poddano analizie w dwóch terminach: w połowie lipca i końcu sierpnia. Termin zbioru wpłynął istotnie na poziom fitozwiózków. W owocach zebranych w sierpniu stwierdzono niższą zawartość cysteiny, glutationu, związków fenolowych ogółem jak i flawonoli oraz antocyjanów w porównaniu do zbioru wcześniejszego. Poziom askorbinianu był zbliżony, niezależnie od terminu zbioru. Stwierdzono wyraźny wpływ sezonu wegetacyjnego na poziom analizowanych antyoksydantów, szczególnie dotyczyło to stężenia askorbinianu.

The purpose of this study was to evaluate in detail both the in vivo and in vitro efficacy of the enzyme agents, ZYMOX® Plus Otic (ZYMOX-P), in the treatment of canine otitis externa (OE). Eight dogs with a diagnosis of non-seasonal severe chronic OE were recruited for the study. ZYMOX-P was administered for 2-4 weeks. The Otitis Index Score (OTIS3) and bacteria or yeast colony growth were measured. Also, minimum biofilm (BF) formation inhibition concentration (MBIC) and BF bactericidal concentration (BBC) were measured in vitro. OTIS3 showed a statistically significant reduction after treatment (88.2%, p<0.001; pre-treatment = 11.0 ± 0.9; post-treatment = 1.3 ± 0.4, mean ± SEM). The individual OTIS scores, erythema, edema, erosions/ ulcerations, exudate and pruritus showed significant reduction (85.7%, 95.7%, 83.3%, 80.0%, and 89.3%, respectively). Microscopic examination revealed the presence of BF exopolysaccharide in all 8 ear samples when stained with alcian blue. Seven of the 8 dogs (87.5%) showed a reduction in colony growth. ZYMOX-P was effective at 34-fold and 16-fold dilutions on MBIC and BBC, respectively. These findings indicate that ZYMOX-P has efficacy against BF-related infection and is beneficial when used for the management of canine OE.

Arsenic is a prevalent and pervasive environmental contaminant with varied amounts in drinking water. Arsenic exposure causes cancer, cardiovascular, liver, nerve, and ophthalmic diseases. The current study aimed to find the best conditions for eliminating arsenic from simulated wastewater and their effect on biomarkers of hepatic in mice. Adsorption tests including pH, contact duration, Al-kheriat dosage, and arsenic concentrations were evaluated. Seventy-two healthy albino mice (male) were accidentally allocated into nine groups (n = 8), the first group was considered as healthy control, the second group (AL-Kheriat), and other groups received AL-Kheriat and arsenic 25, 50, 75, 100, 125, 150 and 175 mg/kg, respectively. Next 10 days, the following were examined: LD50 level, ALP (alkaline phosphatase), ALT (alanine aminotransferase), and AST (aspartate aminotransferase), besides the histological condition of the liver. The results showed that the best time for arsenic removal was 4 hours, pH 8, Alkheriat dose 1 gram, and 50 ppm of pollutants. The level of alkaline phosphatase ALP, alanine transaminase ALT, and aspartate transaminase AST was increased to 150.96 (U/L), 143.1(U/L), and 32.8(U/L), respectively, in Al-Khriet and arsenic exposed population than the healthy control group, When the appropriate dose of Al-Khriet and arsenic mixture is used, it can aid in the selection of a safe way of disposing of the adsorbed residue. Additionally, it can serve as a low-cost rodent pesticide, increasing the commercial viability of this removal strategy.

This research aims to investigate the effect of adding potassium diformate to commercial feed on the increase of absolute growth rate and the activity of protease enzyme. The research was conducted from July to October 2019 in the Aquaculture Laboratory Faculty of Fisheries and Marine Sciences, Padjadjaran University. The method applied in this research was an experimental method using a Completely Randomized Design (CRD), which consists of four treatments and four replications. The treatments were: (A) without addition of Potassium diformate (control), (B) addition of Potassium diformate by 0.3%, (C) addition of Potassium diformate by 0.5%, and (D) addition Potassium diformate by 0.8%. The test fish were 300 giant gouramis with 4-6 cm in length. The containers used in this research were 16 rearing aquaria with a size of 40 30 40 cm3. The density of studied giant gourami fingerlings was 10 fish per aquarium. The rearing period was 40 days. The feeding rate was 3% from biomass. Water quality parameters (temperature, pH, and dissolved oxygen), the absolute growth rate, feed conversation ratio and survival rate were observed every 10 days. The protease enzyme activities were observed at the end of the research. Data on the absolute growth rate, feeding conversion ratio, the characteristics of protease enzyme and survival rate were analyzed using the Analysis of Variance (ANNOVA) continued with Duncan’s Multiple Range Test at the level of 95%, while the water quality was analyzed descriptively. The results show that the addition of potassium diformate by 0.3% gave the best result with the absolute growth rate of 1.50%, feed conversion ratio of 2.70, protease enzyme activity by 634.2 μ/mL and survival rate of 100%.

Taking as the starting point a recently suggested reaction scheme for zymogen activation involving intra- and intermolecular routes and the enzyme-zymogen complex, we carry out a complete analysis of the relative contribution of both routes in the process. This analysis suggests the definition of new dimensionless parameters allowing the elaboration, from the values of the rate constants and initial conditions, of the time course of the contribution of the two routes. The procedure mentioned above related to a concrete reaction scheme is extrapolated to any other model of autocatalytic zymogen activation involving intra- and intermolecular routes. Finally, we discuss the contribution of both of the activating routes in pepsinogen activation into pepsin using the values of the kinetic parameters given in the literature.

5´-Nucleotidase specific towards dCMP and AMP was isolated from avian breast muscle and characterized. It was found to be similar to a type-I form (cN-I) identified earlier as the AMP-selective 5´-nucleotidase responsible for adenosine formation during ATP breakdown in transfected COS-7 cells. Expression pattern of the cN-I gene in pigeon tissues indicated breast muscle as a rich source of the transcript. We purified the enzyme from this source using two-step chromatography and obtained an active homogenous preparation, free of ecto-5´-nucleotidase activity. The tissue content of the activity was calculated at 0.09 U/g wet weight. The specific activity of the enzyme preparation was 4.33 U/mg protein and it preferred dCMP and AMP to dAMP and IMP as a substrate. Its kinetic properties were very similar to those of the enzyme purified earlier from heart tissue. It was strongly activated by ADP. Inhibition by inorganic phosphate was more pronounced than in heart-isolated cN-I. Despite this difference, a similar physiological function is suggested for cN-I in both types of muscle.

Heavy metal contamination of soil occurs when heavy metals are introduced to soil through human activities, leading to the gradual deterioration of the ecology and environment. Microorganism activity reflects the intensity of various biochemical reactions in soil, and changes in it reflect the level of heavy metal pollution affecting the soil. The effects were studied of heavy metal Cd on the microbial activity of soil at different concentrations by investigating the respiratory intensity, urease activity, and catalase activity in forest soil and garden soil. The results showed that the respiratory intensity, urease and catalase activities in the garden soil were all higher than in the forest soil. Cd has obvious inhibitory effects on microbial activities. The three parameters exhibited a downward trend with increasing concentrations of Cd. Catalase activity increased when the mass concentration of Cd reached 1.0 mg/kg, indicating that low concentrations of Cd can promote the activity of some microorganisms. Respiratory intensity and urease activity also increased when the concentration reached 10.0 mg/kg, showing that respiratory intensity and urease activity have strong response mechanisms to adverse conditions. The effective state of Cd in soil, as well as inhibition of microbial activity, decreased with incubation time.

Allozyme structure of the tick population Ixodes persulcatus coded by nine enzyme gene loci has been studied using two subsamples of ticks from Irkutsk suburban areas. Nine loci out of fourteen were polymorphic in ticks of Baikal highway population, whereas five loci were polymorphic in tics sampled from Goloustnoye highway. High level of intra-population heterogeneity of ticks has been revealed. No distinction between two populations from different nidi of infection has been recorded. It was probably due to the high rate of gene migration between the studied populations and to their high genetic similarity.

The chemical composition of eight Triticale varieties cultivated in Poland was investigated. The contents of the basic chemical components as rule ranged between those in the parental species, and there were only slight differences between the various Triticale varieties in this respect. Protein content averaged at around 12.2% dry maller Triticale varieties contained more P, K, Ca, Mn and Zn than the parental species, and similar quantities of Mg, Na, Fe and Cu. Despite some differences, none of the varieties stood out as regards the content of the determined mineral components. Samples were obtained from three plant breeding farms, and the region of cullivation had only a minor effect on the content of the determined components, only some of the differences were statistically significant.
Zbadano skład chemiczny 8 polskich odmian pszenżyta wyhodowanych w 3 stacjach hodowli Roślin: Choryń, Dańków i Laski i porównano je z macierzystymi gatunkami pszenicy i żyta. Stwierdzono, że wskaźniki oceny towaroznawczej ziarna badanych odmian pszenżyta były mało zróżnicowane. W porównaniu z pszenicą i żytem pszenżyto charakteryzowało się niższą masą gatunkową i wyższą masą 1000 ziarn. Zawartość podstawowych składników chemicznych w ziarnie pszenżyta była z reguły pośrednia pomiędzy zawartością łych składników w gatunkach rodzicielskich i mało zróżnicowana pomiędzy odmianami. Zawartość białka wynosiła średnio ok. 12,2%. Badane próbki pszenżyta zawierały więcej P, K, Ca, Mn i Zn w porównaniu z gatunkami macierzystymi, a zawartość Mg, Na, Fe i Cu był a zbliżona do zawartości tych składników w ziarnie pszenicy i żyta. Stwierdzono wprawdzie różnice w zawartości niektórych składników mineralnych między odmianami, ale żadna z badanych odmian pszenżyta nie wyróżniała się pod tym względem. Rejon uprawy miał minimalny wpływ na zawartość składników mineralnych w ziarnie i tylko niektóre różnice znalazły potwierdzenie w ocenie statystycznej.

The recombinant catalase isolated from a psychrotolerant microorganism belonging to Serratia genus exhibits a high activity in a wide range of pH. Due to a great catalytic potential in operational conditions, it can be used in various industrial applications whereby it acts as a hydrogen peroxide scavenger. To reduce the cost of biocatalyst the enzyme encapsulation into a hydrogel structure was proposed. The obtained results showed a high activity of encapsulated catalase in acidic conditions (pH in range 4.4 - 6.6) and at low temperatures (6-15°C). Moreover, immobilized catalase exhibited a high stability in natural media, especially in milk. Its activity during peroxide decomposition in milk, the possibility of re-using, as well as the fixed bed reactor performance confirmed wide application possibilities. High values of enzyme and substrate concentrations led to the beads burst due to rapid oxygen diffusion from the capsules, thus they are limited.
