Opis:
Creating innovative methods of treatment and regeneration of damaged tissues or organs is a key challenge of the twenty-first century. The aim of this study was to determine the possibility of producing and characterising the properties of self-organising chitosan hydrogels prepared with the use of chitosan lactate/chloride and disodium hydrogen phosphate dodecahydrate as a cross-linking agent. The structure and supramolecular architecture of the biomaterials were evaluated by Fourier-transform infrared spectroscopy and polarised optical microscopy. Biological studies assessed cytotoxicity by contact with a human colon adenocarcinoma cell line. The colourimetric resazurin assay showed that the obtained chitosan hydrogels are non-cytotoxic materials. Thus, self-organising biomaterials hold great promise for application in tissue engineering.

Opis:
The aim of the research was to employ radiation to produce flexible carboxymethyl chitosan (CMCS) based hydrogels of uniform structure to characterise their swelling properties and cytocompatibility for potential applications as hydrogel wound dressings. CMCS in aqueous solution was irradiated with an electron beam in the presence of a poly(ethylene glycol) diacrylate (PEGDA) macromonomer as a crosslinker, at 12 different compositions, i.e. 3–20% CMCS, 3 and 5% PEGDA. The obtained hydrogels were subjected to sol–gel analysis. The amount of insoluble fraction (up to 100%) rose with an increase in the PEGDA/polysaccharide ratio. Moreover, the equilibrium degree of swelling, ca. 15 to 200 g of water per g of gel, which was higher for lower content of crosslinker, decreased with the delivered dose, which was associated with an increase in crosslinking density. The in vitro XTT cell viability assay (murine fibroblasts, L929 cell line) showed no significant cytotoxicity of CMCS gels.

Opis:
Solutions of chitosan (CS) and carboxymethyl chitosan (CMCS) were subjected to irradiation by electron beam in presence of a crosslinking agent (poly(ethylene glycol) diacrylate - PEGDA) in order to produce carboxymethyl chitosan- and chitosan-based hydrogels. PEGDA macromonomer itself undergoes simultaneous polymerization and crosslinking either in neutral water or in acidic medium. Acidic solutions of chitosan of 0.5, 1 and 2% can be effectively crosslinked with PEGDA to form a gel. Although CMCS undergoes radiation-initiated crosslinking only at high concentration in water (over 10%), the presence of PEGDA in solution facilitated hydrogel formation even at lower concentration of CMCS. Formation of CS and CMCS hydrogels required irradiation doses lower than those needed for sterilization, i.e. 25 kGy, in some cases even as low as 200 Gy. Sol-gel analysis revealed relatively high gel fraction of obtained hydrogels, up to 80%, and good swelling ability. Both parameters can be easily controlled by composition of the initial solution and irradiation dose. Possible mechanisms of crosslinking reactions were proposed, involving addition of the polysaccharide macroradicals to a terminal double bond of PEGDA.

Opis:
Damage to the nervous system, in particular spinal cord injuries, is a burden for the patient and is usually the cause of irreversible disability. The progress observed in the last decade in the fields of biology, biomaterial engineering and neurosurgery has created new treatment solutions while preventing further neurodegenerative processes. The most important research is focused on the implementation of polymer structures in clinical practice, especially chitosan hydrogels, which are the scaffolds for regenerating axons. This article presents a new generation of biomaterials that have the ability to gel in response to temperature changes; they are intended for injectable scaffolds for nerve cell cultures. Two types of hydrogels were prepared based on chitosan lactate and chitosan chloride using uridine 5’-monophosphate disodium salt. The structure of the systems was observed under a scanning electron microscope and examined using Fourier transform infrared spectroscopy. In addition, thermal properties were tested using differential scanning calorimetry.

Opis:
Thermosensitive chitosan hydrogels containing sodium beta-glycerophosphate (ß-GP), whose gelation is induced by increasing temperature to body temperature, were functionalized by incorporation of Alkaline Phosphatase (ALP), an enzyme involved in mineralization of bone. ALP incorporation led to acceleration of gelation upon increase of temperature for four different chitosan preparations of differing molecular weight, as demonstrated by rheometric time sweeps at 37°C. Hydrogels containing ALP were subsequently incubated in calcium glycerophosphate (Ca-GP) solution to induce their mineralization with calcium phosphate (CaP) in order to improve their suitability as materials for bone replacement. Incorporated ALP retained its bioactivity and induced formation of CaP mineral, as confirmed by SEM, FTIR, Raman spectroscopy, XRD, ICP-OES, and increases in dry mass percentage, which rose with increasing ALP concentration and incubation time in Ca-GP solution. The results demonstrate that ALP accelerates formation of thermosensitive chitosan/ß-GP hydrogels and induces their mineralization with CaP, which paves the way for applications as injectable bone replacement materials.

Opis:
Chitosan is a natural cationic polymer that dissolves in an acidic environment and forms gels. Its properties depend on the degree of deacetylation and molecular weight. It is a bioactive compound with antibacterial and film-forming properties that allow to increase the regenerative capacity of the skin. Moreover, it is biodegradable, biocompatible, non-toxic, and stable. In this research, chitosan was combined with mandelic and lactobionic acids which are characterized by biological activity and low toxicity. This combination not only has a positive effect on the chitosan solubility, but it also allows to obtain new biomaterials whose positive features of the base ingredients are enhanced by their synergistic effect. The obtained hydrogels were assessed regarding the interaction of chitosan and hydroxy acid molecules, and the stability of the resulting structures was examined. The research was performed by using rheological methods and IR spectroscopy. Chitosan hydrogels made with mandelic acid are characterized by higher viscosity values, as compared to hydrogels containing lactobionic acid. The samples of the obtained hydrogels stored for 7 days showed no signs of degradation and their viscosity values were constantly increasing, which proves the ongoing process of creating new bonds between hydroxy acid molecules and chitosan chains. After this time, the hydrogels with mandelic acid revealed higher viscosity values in comparison to hydrogels made with lactobionic acid. Based on the obtained IR spectra, the shifts of the characteristic chitosan bands as a result of interaction with the tested hydroxy acids were analyzed.

Opis:
In this paper, the mechanical properties of hybrid chitosan hydrogels were investigated using the texture profile analysis test. Scaffolds obtained from a polysaccharide of various molecular weights were studied, which, with the addition of glycerophosphate salt, formed a three-dimensional structure in vivo. The obtained systems were also enriched with collagen and calcium carbonate to improve the mechanical properties. The determined texture parameter values indicate that the mechanical properties of the hybrid hydrogel depend on the molecular weight of the polymer, the type of solvent and, the pH-neutralizing substance, as well as the type and concentration of the filler. Moreover, in some cases the TPA test was the only way to evaluate the mechanical properties of the obtained hydrogels due to the inability to determine the Young's modulus. Consequently, the texture analysis test is a valuable tool for selecting solutions depending on the intended application of the scaffolds.

Opis:
Thermosensitive injectable chitosan hydrogels can be formed by neutralization of acidic chitosan solutions with sodium betaglycerophosphate (Na-β-GP) coupled with increasing temperature to body temperature. Such hydrogels have been considered for applications in bone regeneration. In this study, chitosan hydrogels were enriched with glycerol and the enzyme alkaline phosphatase (ALP) with a view to improving their suitability as materials for bone tissue engineering. Mineral formation was confirmed by infrared spectroscopy (FTIR) and increases in the mass fraction of the hydrogel not consisting of water. Incorporation of ALP in hydrogels followed by incubation in a solution containing calcium ions and glycerophosphate, a substrate for ALP, led to formation of calcium phosphate within the hydrogel. MG-63 osteoblast-like cells were cultivated in eluates from hydrogels containing ALP and without ALP at different dilutions and directly on the hydrogel samples. Hydrogels containing ALP exhibited superior cytocompatibility to ALP-free hydrogels. These results pave the way for the use of glycerol- and ALP-enriched hydrogels in bone regeneration.

Rheological properties of chitosan hydrogels forming polymeric network for active substances are presented in the paper. Bovine serum albumin and vancomycin were used as active substances. A change in solution tem-perature acted as an inhibitor of sol-gel phase transition. The determined rheological properties and temperature curves of solutions showed properties similar to those of initial chitosan solution. In case of vancomycin the gelation process initiated by a temperature change led to the formation of irreversible structure.

Opis:
Biodegradable materials for drug delivery and bone tissue engineering are currently intensively developing and improving, but there are still a lot of problems to solve related with bioactivity, biocompatibility, release profile etc. Osteosarcoma is an aggressive malignant neoplasm arising from primitive transformed cells of mesenchymal origin that exhibit osteoblastic differentiation and produce malignant osteoid. It is the most common histological form of primary bone cancer [1,2]. Treatment is most destinations and made up for: intensive multidrug short induction chemotherapy, amputation or tumor resection within the limits of normal tissue and in the last phase again chemotherapy. This kind of neoplasm is most recently detected in young male till 25 age, therefore improving methods of treatment is so important. Chitosan is a natural-based polymer obtained by alkaline deacetylation of chitin received from powdered shrimp shells was purchased from Acros Organics. Its main advantages are non-toxic, non-immunogenic, non-carcinogenic degradation products, biocompatible, bioactive and biodegradable. These properties cause chitosan a very good candidate for novel hydrogel drug delivery systems. Chitosan easily forms hydrogel particles and entraps biomolecules through a number of mechanisms, including chemical crosslinking, ionic crosslinking, and ionic complexation [3,4]. A possible alternative of chitosan by the chemical modification also has been useful for the association of bioactive molecules to polymer and controlling the drug release profile. There are few methods of modification, or example.: copolimerization, grafting, chemical and ionic crosslinking, polyelectrolyte complexes, etc [5]. Previous studies demonstrated that chitosan could promote the proliferation and osteogenesis but only with moderate swelling ratio of composite, too high ability of entrapment water solutions are not recommended. There are a lot of advantages in chitosan properties that can be used in research work to obtain the material of the best expected properties. Laponite® (LA) is a plate-like synthetic clay hectoritetype belongs to a family of phyllosolicates type 2:1 [6]. Its structure represent empirical formula: Na0.7+[Si8Mg5.5Li0.3 O20(OH)4]0.7-. The plates size is about 25 nm x 0,92 nm. LA has a large surface area, anionic surface charges and exchangeable Na+ cations in hydrated interlayers. Presence of sodium cations causes better adsorption properties for cationic drug molecules. Moreover, the exfoliated LA particles may act as multifunctional crosslinkers in forming the nanocomposite hydrogels, and the polymer chains were anchored to the particles and entangled to form a network [7]. Used synthetic clay has got the same type structure and but better sorption properties to montmorillonite but it has got serious advantage - as a synthetic compound shows low heavy metal content. The initial results indicate that the incorporation of clay improved the swelling behavior in contrast to the pure chitosan beads. There also had been revealed significant disproportion of viscosity received hydrogels according to different type of LA or different concentration. Increasing content causes telling rise of viscosity, especially reported in higher content of used crosslinker. The aim of research is to develop a bioactive system biopolymer/layered silicate intelligent nanocomposite based on chitosan and synthetic clay by a cross-linking reaction using sodium tripolyphosphate as the gel factor. The resultant composite were characterized by Fourier transform infrared spectroscopy, scanning electronic microscope and X-ray diffraction analysis. The bioactivity in physiological pH solution (SBF pH=7.40) [8], drug encapsulation efficiency and controlled release behaviour were also investigated by using the model drug to reveal the effects of introduced LA.

Opis:
This paper discusses the influence of the direction of applied deformation on the ability to gelation of thermosensitive chitosan hydrogels. The application of the shear rate equal in value to the classically performed oscillatory measurements leads to significantly different shapes of experimental curves. It was found that the type of mechanically applied deformation has a significant impact on the gelation ability of colloidal chitosan solutions and conditions of sol-gel phase transition. Simple shear leads to a phase transition at a lower temperature or in a shorter time compared to oscillatory tests. Moreover, based on the final values of dynamic viscosity in rotational measurements, it was found that stronger crosslinking of the polymer structure was observed.

Opis:
Polysaccharides, such as chitosan (CS), are widely used in many biomedical applications. However, they require crosslinking agents to achieve chemical stability and appropriate mechanical properties. In this work, chitosan-based hydrogels were crosslinked using vanillin and/or sodium tripolyphosphate, as chemical and physical crosslinking agents, respectively. Microstructural (digital microscope, SEM), structural (FTIR-ATR), mechanical (static compression test), and in vitro biological (chemical stability and swelling ratio in PBS, cytotoxicity) properties of the obtained materials were evaluated to assess materials potential as biomedical scaffolds. The optimal ratio of vanillin to chitosan (DD = 89%) to crosslink the polymer was found to be 1.2:1. Moreover, the double crosslinking with vanillin caused a two-time increase in the compression strength of the samples and led to the slower biodegradability. Cytotoxicity studies showed that the cells prefer double vanillin crosslinked hydrogels over those treated with TPP. Further studies, such as bioactivity are required to determine the specific functionality of the hydrogels and the specific tissue which may be treated with the tested materials. The optimal material was chosen to the next step of the study, which may be obtaining composite hydrogels with hydroxyapatite and/or graphene oxide to tailor or improve properties towards specific tissue regeneration.

Opis:
In order to achieve hydrogel and drug release profiles, a comprehensive knowledge of the types, properties and syntheses of hydrogel polymer networks are needed. For this reason, a natural biopolymer hydrogel based on chitosan was described. Chitosan has many advantages, which meet the requirements necessary for the preparation of medical materials; for example, wound dressings. This article focused on the biomedical use of a chitosan hydrogel: chitosan–poly(vinyl alcohol) (PVA). The method of preparation of hydrogels containing a drug as an active wound dressing was described. To obtain a hydrogel dressing to be applied in patients with burns or difficult curative wounds, gentamicin (an aminoglycoside antibiotic) was used as a medicament. The effect of the PVA concentration in hydrogels on the release rate of the antibiotic was examined. For this, the crosslinking agent of the hydrogel, glutaraldehyde, was used. The release process of gentamicin was described by using an equation of first order kinetics.

The damage to the central nervous system is one of the most difficult cases of trauma to treat. Over the last few years, increasing attention has been focused on the development of strategies based on biomaterials for regeneration and repair of the spinal cord injury. In particular, materials in the form of hydrogels based on chitosan are being actively investigated due to their intrinsic properties that are favorable in spinal cord tissue regeneration. The purpose of this study was to develop a thermo-gelling chitosan solution that will be prepared with the use of acids that naturally occur in the human nervous tissue. For this purpose, two types of chitosan gels were prepared based on chitosan glutamate and chitosan lactate. In order to reduce toxic action of the system obtained gels were conditioned in distilled water with pH 5.00. The changes in the structures of systems obtained were determined with the use of FTIR method. Biocompatibility was primarily evaluated through cytotoxicity testing by MTT assay with respect to mouse fibroblast cells.

Opis:
This review considers articles on the formation of hydrogels based on chitosan as well as succinylated and quaternized chitosan derivatives. They are synthesized using low toxicity reagents, under ordinary conditions (low production costs). Chitosan derivatives are soluble in an extended range of pH values and characterized by mucoadhesiveness, bioavailability and biodegradability, which extends the potential of their medical applications. One of the most important properties of chitosan and its derivatives is the ability to form hydrogels. Depending on the nature of the bonds that occur during formation, hydrogels are divided into chemically or physically crosslinked, or a mixture of the two. Chemically crosslinked gels have covalent bonds, while physically crosslinked gels are formed by noncovalent interactions, for example, ionic. Mixed hydrogels have both types of crosslinking.

The aim of this paper was to conduct preliminary instrumental tests to determine the possibility of injection applications for thermosensitive chitosan systems, including injection needles and application conditions. Among the many biomedical and pharmaceutical applications of chitosan, the minimally invasive thermosensitive scaffolds that form in vivo are an interesting solution. Despite many studies on colloidal chitosan systems undergoing sol-gel phase transition, almost no studies have examined their injectability. It has been stated that the use of acetic acid as a solvent reduces the forces needed for injection. Moreover, the key impact of injection temperature was determined. Storing the medium at room temperature before the injectability test led to a decrease in the value of forces needed for injection. The obtained results are discussed based on the change of the rheological properties of the chitosan hydrogels.

The effect of chitosan concentration on the properties of dermatological preparations prepared with 2% methylcellulose and 10% polyvinylpyrrolidone was studied. As the levels of chitosan, propylene glycol-1,2 and glycerol increased, the hardness, consistency and cohesiveness of the hydrophilic gels increased, while the dissipation capacity decreased. Gels based on methylcellulose and chitosan, compared to formulations containing polyvinylpyrrolidone, have a higher rate of loss of volatile compounds, higher pH values for 1,2-propylene glycol formulation and higher texture parameters. The tested hydrogels have good rheological properties, allowing extrusion from the tube and spreading on the skin.

Ionically crosslinked chitosan membranes were prepared and their swelling behaviour was analysed. Dynamic swelling of noncrosslinked (Ch) as well as singly (Ch/TPP, Ch/NaAlg) and doubly (Ch/NaAlg/TPP) crosslinked chitosan membranes was performed in buffered aqueous solutions of various pH to determine the mechanism of water diffusion into these hydrogels. It was stated out that mechanism of water diffusion into studied chitosan hydrogel membranes depends both on pH of swelling solution as well as on the type of membrane (type of crosslinker). The water transport mechanism was determined to be: i) Fickian or ‘Less Fickian’ diffusion, ii) non‑Fickian diffusion (anomalous diffusion) and iii) Case II or Super Case II diffusion.

Currently, increasing attention is being paid to issues related to environmental protection, waste management, as well as to the development of polymers with useful properties. The research presented here involved preparation of hydrogels based on Beetosan® - a chitosan derived from the multi-stage processing of dead bees. Moreover, hydrogels were additionally modified with natural substances - i.e. bee pollen and extract of Salvia officinalis (sage) that are well known for the presence of many compounds with beneficial properties from a medical point of view. Materials have been first obtained by photopolymerization. Then, their surface morphology, wettability and cytotoxicity to selected cell lines have been determined. It can be stated that such combination of Beetosan® hydrogel matrix and the mentioned additives resulted in a preparation of polymers characterized by negative impact on cancer cells. Impact of hydrogels with sage is slightly more intense due to the presence of substances such as ursalic or rosmaric acid that are characterized to have anticancer activity. Such negative impact has not been observed in case of studies using fibroblasts. Furthermore, addition of natural substances into hydrogels resulted in a more homogeneous surface and in the decrease of wettability angle of the tested polymers. It can be concluded that the use of natural-derived reagents and synthesis of polymers using these reagents (as a result of environmentally friendly photopolymerization) yields materials with interesting properties for medical purposes, with particular emphasis on antitumor activity, and without significant negative impact on fibroblasts.

Rheological measurements were performed to determine the viscoelastic properties and phase transition temperatures of these solutions. The study was conducted in the temperature range of 5–60°C with the use of classical techniques of rotational rheometry in the cone-plate measurement system. A significant influence of a collagen addition to chitosan chloride solutions on the viscoelastic properties of the systems was observed. The addition of collagen in all the cases increased the sol–gel phase transition temperature in comparison with the chitosan chloride solution containing β-glycerophosphate.

Zsyntezowano szereg materiałów różniących się zawartością fotoinicjatora oraz przeprowadzono badanie zdolności pęcznienia i inkubację. Wykonano pomiar kątów zwilżania i analizę spektroskopową FT-IR.
The paper presents a synthesis of the hydrogel materials by photopolymerization. A series of materials differing in the amount of photoinitiator was prepared. In these studies the ability of swelling of attained materials was determined. What is more, incubation studies were also carried out. Measurements of contact angles was conducted and a chemical structure of obtained materials was defined using FT-IR spectroscopy. On the basis of the research the impact on the amount of the photo-initiator on the structure of a polymeric material have been defined. Synthesized materials were characterized by a different degree of the cross-linking, and therefore they have different properties. Hence the conclusion that possibility of the structure modifying of the material creates more opportunities for its use.
