Opis:
Scanning probe microscopy (SPM) since its invention in the 80’s became very popular in examination of many different sample parameters, both in university and industry. This was the effect of bringing this technology closer to the operator. Although the ease of use opened a possibility for measurements without high labour requirement, a quantitative analysis is still a limitation in Scanning Probe Microscopes available on the market. Based on experience of Nano-metrology Group, SPM still can be considered as a tool for quantitative examination of thermal, electrical and mechanical surface parameters. In this work we present an ARMScope platform as a versatile SPM controller that is proved to be useful in a variety of applications: from atomic-resolution STM (Scanning Tunnelling Microscopy) to Multi-resonance KPFM (Kelvin Probe force microscopy) to commercial SEMs (Scanning electron microscopes).

Nowadays nanostructures are more and more often designed as carriers for drug delivery, especially to improve the drug pharmacokinetics and pharmaco-dynamics. Numerous kinds of nanostructures are considered a good prospect for medical applications thanks to their small size, acceptable biocompatibility and toxicity. Due to the fact that nanotechnology is a new field of science, every nano-scale product must be thoroughly examined regarding its toxicity to the human body. This study provides new insights into effects of exposing endothelial cells to the selected nanostructures. Dendrimers of the fourth generation (PAMAMs), multi-walled carbon nanotubes (MWCNTs) and silver nanoparticles (SNPs) were used to evaluate nanostructures influence on endothelial cells in vitro. The nanostructures were evaluated via transmission electron microscopy and dynamic light scattering technique. The cells previously exposed to the nanostructures were observed and analyzed via the atomic force microscopy and scanning electron microscopy to obtain a quantitative evaluation of the cells morphology. The presence of multi-walled carbon nanotubes and silver nanoparticles on the cells surface was confirmed by the scanning electron microscopy. Our results confirm that the surface association and/or uptake of nanostructures by the cells resulting from physicochemical and biological processes, affect the cells morphology. Morphological changes can be induced by the membrane proteins interaction with nanomaterials, which trigger a sequence of intracel-lular biological processes.

There are no papers examining the labellar micromorphology in Malaxidinae in detail. The aim of this paper is to present the results of labellar micromorphology of our studies on Malaxidinae. The micromorphology of nine species was examined and described (calli, nectaries, OP structures, concavities). These species are divided into six groups of which the lip morphology probably represents different forms of attracting and rewarding pollinators. Group one consists of Disticholiparis gregaria and Platystyliparis aurita with characteristic globular nectary and pad-like callus. The second group with Liparis nervosa has two conelike convexities. The third one is represented by Liparis sutupensis with spread lip, the basal callus, and the epichile often eroding with age. The fourth group (Stichorkis cespitosum) has a massive hypochil with basal callus and delicate epichil. The fifth group (Microstylis ophioglossoides, Dienia ophrydis) is characterized by a lip with developed lateral lobes, three deep concavities in the middle part, an erect callus, and many raphide cells.Crepidium cf. rheedii and Crepidium dryadum from the sixth group have a three-lobed lip with entire or toothed mid-lobe, basal callus, cavity surrounded by a rim and OP structures with raphide cells. We also emphasized the different way and place of nectar secretion in the subtribe and gave some suggestions about pollination strategies.

W artykule przedstawiono charakterystyki cienkich warstw SnO2 na czystym szkle. Warstwy SnO2 nanoszone były metodą magnetronowego sputteringu (nanoszenie) na podłoże szklane w różnych temperaturach. Wykorzystano do tego urządzenie Magnetron Line 440. Do badania składu i morfologii warstwy użyto mikroskopii skaningowej i atomowej (STM/AFM). Na podstawie badań stwierdzono, że powłoki wykazują dobre połączenie z materiałem podłoża, charakteryzują się zróżnicowaną chropowatością i są jednorodnie chemiczne.
Atomic force microscopy is one of the most popular method used in surface imaging. This method allows to measure the surface topography and determine the dimensions of the structures in the subatomic resolution [1]. Due to its properties, it can be applied to the measurement of conductors and semiconductor surfaces prepared in various processes. The experiment is fo-cused on SnO2 and ITO thin layers which can be used as transparent electrodes [2]. The authors are trying to illustrate the correlation between process parameters - creation of semiconductor in magnetron sputtering by different process conditions (temperature and cooling process, gas pressure and composition), surface of the sample and its other electrooptical parameters.

Skaningowa mikroskopia elektronowa wymaga aby próbka była przewodząca, lub pokryta warstwą przewodzącego materiału. Pomimo tego że złoto jest metalem najczęściej używanym do pokrywania próbek, jest także bardzo drogie, więc autorzy postanowili sprawdzić czy inne metale lub stopy metali będą nadawać się do pokrywania próbek. Próbki były pokrywane w procesie sputteringu magnetronowego przy pomocy urządzenia LINE 440 (Alliance Concept), a obserwacje przeprowadzone na mikroskopie Hitachi TM3000. Próbki były pokryte miedzią, stopem miedzi i galu, cyną i tlenkiem cyny. Obrazy otrzymane w czasie eksperymentu były porównywalne z próbkami pokrytymi złotem.

The cracking of materials and fracture surface is of great practical and academic importance. Over the last few years the development of the fractography of crystalline alloys resulted in a useful tool for the prediction or failure analysis. Many attempts have been made to observe cracks using optical microscopy, X-ray topography and transmission electron microscopy (TEM). Of these techniques, the resolution of optical microscopy and X-ray topography is too poor. By contrast, the resolution of TEM is high enough for detailed information to be obtained. However, in order to apply TEM observations, a thin foil specimen must be prepared, and it is usually extremely difficult to prepare such a specimen from a pre-selected region containing a crack. In the present work, deformation mechanisms fracture surfaces of Ni-based metallic glass samples have been studied by specially designed experiments. In order to study the deformation mechanisms and fracture the Ni-based metallic glasses have been investigated in the tensile test. The structure and fracture surfaces after the decohesion process in tensile tests were observed using transmission electron microscope (TEM) and scanning electron microscope (SEM), respectively. The studies of structure were performed on thin foils. Moreover the investigated tape was subjected to a banding test. Then, the tape was straightened and the thin foil from the area of maximum strain was prepared. This thin foil sample was deformed before the TEM investigation to obtain local tears.

Mg60 Zn35 Ca5 amorphous powder alloys were synthesized by mechanical alloying (MA) technique. The results of the influence of high-energy ball-milling time on amorphization of the Mg60 Zn35 Ca5 elemental blend (intended for biomedical application) were presented in the study. The amorphization process was investigated by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM). Initial elemental powders were mechanically alloyed in a Spex 8000 high-energy ball mill at different milling times (from 3 to 24 h). Observation of the powder morphology after various stages of milling leads to the conclusion that with the increase of the milling time the size of the powder particles as well as the degree of aggregation change. The partially amorphous powders were obtained in the Mg60 Zn35 Ca5 alloy after milling for 13-18h. The results indicate that this technique is a powerful process for preparing Mg60 Zn35 Ca5 alloys with amorphous and nanocrystalline structure.

This paper presents the results of investigations into the structure, microstructure and magnetic properties of Fe61Co10Y8W1B20 amorphous alloy. The alloy samples were in two physical forms: (1) plates of approximate thickness 0.5 mm (so-called bulk amorphous alloys) and (2) a ribbon of approximate thickness 35 μm (so-called classic amorphous alloy). The investigations comprised: X-ray diffractometry, Mössbauer spectrometry, transmission electron microscopy, and selected magnetic measurements; all of the investigations were carried out on samples in the as-quenched state. Analysis of the obtained SEM and TEM images, X-ray diffraction patterns, Mössbauer spectrometry results and measurements of the magnetisation in a high magnetic field facilitated collectively the detailed description of the structure of the investigated alloy, which was found to depend on the quenching speed.

The knowledge whether and how chemical species react with tissues is important because of protection against harmful factors, diagnose of dermatological diseases, validation of dermatological procedures as well as effectiveness of topical therapies. In presented work the effects of chemical agents on plates of human fingernails were studied using Atomic Force Microscopy and Scanning Electron Microscopy. Apart from that, mapping of the elastic properties of the nails was also carried out. To obtain reliable measures of spatial evolution of the surface variations, recorded images were analyzed in terms of scaling invariance brought by fractal geometry, instead of common though not unique statistical measures.

A wide range of abrasive tools is used in modern removal machining techniques. Their condition has significant influence on the quality of the shaped surfaces of the produced elements and the functions they perform. Abrasive tools diagnostics is in this case an essential element of the production process, connected with assessment of the abrasive tool surface condition. Such an assessment can be carried out with microscopic methods The work presents selected microscopic techniques on the cutting edge of technology such as: digital microscopy, confocal laser scanning microscopy and scanning electron microscopy. Characteristics of each of the methods and the applied devices were briefly presented. The experimental part presents exemplary results of measurements and analyses carried out using the described methods. The works were carried out on, among others, grinding wheels with ceramic bond used in internal cylindrical grinding processes. The assessed tools were characterized by a number of features such as impregnated active surface, modified bond microstructure or smearing the active surface with chips of the machined material. The obtained results confirmed the possibility of application of the above-mentioned microscopic techniques in abrasive tools diagnostics.

Cytoszkielet to sieć białkowych polimerów oraz związanych z nimi setek białek motorycznych, regulatorowych i łączących cytoszkielet z innymi strukturami komórkowymi. Rozwój wiedzy o cytoszkielecie jest nierozerwalnie zwiększany z postępem technik mikroskopowych używanych do jego obserwacji. Początki tych badań to niespecyficzne, nieskomplikowane barwienia utrwalonego materiału biologicznego, które później rozwinęły się w nowoczesną mikroskopię strukturalną, pozwalającą na precyzyjne znakowanie określonych białek tworzących cytoszkielet, badanie ich stanu fizjologicznego czy też oddziaływań cytoszkieletu z luźno związanymi białkami błony czy cytoplazmy. Obecnie możliwe jest nie tylko obrazowanie struktury i funkcji cytoszkieletu ze znacznie lepszą rozdzielczością przestrzenną, ale także prowadzenie tych obserwacji na żywym materiale biologicznym. Z drugiej strony, stabilność cytoszkieletu umożliwia poszukiwanie nowych metod jego obrazowania, co niewątpliwie należy do kół napędowych postępu, jaki dokonał się i wciąż dokonuje się w dziedzinie mikroskopii.
Cytoskeleton is basically a network of protein polymers, but it also contains thousands of motor, regulatory and scaffolding proteins that interact with this network. Discoveries related to the cytoskeleton were strictly connected to the development of microscopy techniques used to observe the cytoskeletal structures. At first, the imaging involved only unspecific, very simple staining of fixed material. Then, the methods evolved into advanced structural microscopy, which enabled accurate detection of specific cytoskeletal proteins, their physiological status, and interactions with loosely bound membrane and cytoplasmic proteins. Today, it is possible not only to visualize the structure and function of the cytoskeleton with better spatial resolution but also to perform the imaging in vivo on live biological specimens. On the other hand, one should also notice that observations of the stable, well defined cytoskeletal structures from their very discovery have continuously stimulated the progress in the microscopy field.

Good quality in composite castings can be confirmed primarily by their user. The producer is obliged to create a good product that meets the user’s needs that should undergo quality control. Omission of technological procedures and improper quality control may result in various defects like porosity. This paper presents the characteristics of the casting defect, namely porosity, with a particular focus on porosity in suspension and saturated metal composites. This defect is different, specific to composite castings, making its identification very difficult. The aim of the study is to detect and describe porosity in composites with the use of microscopic and submicroscopic examinations. The assessment of the porosity of the microstructure of composite castings allowed for the formulation of the following conclusions: in addition to the porosity that occurs in castings of classic materials (cast steel, cast iron, and non-ferrous metal alloys), metal composites also distinguish between primary and secondary agglomerates, leaving the reinforcement space not filled, occluded bubbles, and separated gas bubbles.

Results of positron annihilation lifetime spectroscopy (PALS) and microscopic studies on simple microorganisms, brewing yeasts, are presented. Lifetime of ortho-positronium (o-Ps) were found to change from 2.4 to 2.9 ns (longer-lived component) for lyophilized and aqueous yeasts, respectively. Also hygroscopicity of yeasts in time was examined, allowing to check how water – the main component of the cell – affects PALS parameters, thus lifetime of o-Ps were found to change from 1.2 to 1.4 ns (shorter-lived component) for the dried yeasts. The time sufficient to hydrate the cells was found below 10 hours. In the presence of liquid water, an indication of reorganization of yeast in the molecular scale was observed. Microscopic images of the lyophilized, dried, and wet yeasts with best possible resolution were obtained using inverted microscopy (IM) and environmental scanning electron microscopy (ESEM) methods. As a result, visible changes to the surface of the cell me mbrane were observed in ESEM images.

The aim of this work was to investigate the microstructure and mechanical properties of the two-component melt-spun (TCMS) alloy produced from Ni40Fe40B20 and Ni70Cu10P20 melts. The Ni40Fe40B20, Ni70Cu10P20, Ni55 Fe20 Cu5 P10 B10 alloys were arc-melted. Then the alloys were melt-spun in the two different ways i.e.: by casting from a single-chamber crucible and from the two-chamber crucible. All of the above mentioned alloys were processed in the first way and the Ni40Fe40B20 and Ni70Cu10P20 were simultaneously cast on the copper roller from the two-chamber crucible. The microstructure of the alloy was studied using transmission electron microscopy (TEM), scanning electron microscopy (SEM) with energy dispersive spectrometry (EDS) and light microscopy. The mechanical properties were investigated using tensile testing and nanoindentation. The two-component melt-spun (TCMS) amorphous Ni55 Fe20 Cu5 P10 B10 alloy present hardness, tensile strength and Young modulus on the significantly higher level than for a single phase amorphous Ni55 Fe20 Cu5 P10 B10 alloy and slightly below the corresponding values for the Ni40Fe40B20.

Mucopolysaccharidoses (MPS) are severe inherited metabolic disorders caused by storage of glycosaminoglycans (GAGs). The level of accumulated GAGs is an important parameter in assessment of the severity of the disease and the efficacy of treatment; unfortunately, biochemical methods are often unreliable and only semi-quantitative. Therefore, finding other methods for estimation of GAG levels and/or assessment of the efficacy of applied therapy is very important. Although monitoring of GAG levels during therapy is crucial, in this work it is proposed that analysis of the ultrastructure of MPS cells by electron microscopic techniques can be considered as an alternative and reliable method for assessment of lysosomal storage. The number of complex lysosomal structures was found to be significantly higher in MPS cells relative to controls, while it decreased significantly in response to either enzyme replacement therapy or substrate reduction therapy. Thus, this parameter, easily assessed by electron microscopy, appears to correspond to the physiological state of MPS cells.

In this paper we summarize the results of our research concerning the diagnostics of micro- and nanostructure with scanning probe microscopy (SPM). We describe the experiments performed with one of the scanning probe microscopy techniques enabling also insulating surfaces to be investigated, i.e., atomic force microscopy (AFM). We present the results of topography measurements using both contact and non-contact AFM modes, investigations of the friction forces that appear between the microtip and the surface, and experiments connected with the thermal behaviour of integrated circuits, carried out with the local resolution of 20 nm.

Opis:
Confocal microscopy is commonly used in mineralogy for structural analysis and to investigate the chemical composition of minerals. In our work we performed imaging of structural defects in meteorites i.e. imaging of microstructures, inclusions, fractures and cracks. Information obtained in this way enables better understanding of minerals' origin and evolution. The aim of our work was not concentrated on the deep description of physical processes related to confocal microscope (CSLM) imaging. This paper is limited to the presentation of a few CSLM functions and possibilities, which have practical application in the interpretation of fluorescence microscope imaging of minerals. Confocal microscopy enables identification and preview of localization of selected minerals and therefore can be used to image of the distribution of chemical compounds in minerals.

Porphyrin photosensitizers tend to localize in mitochondria. The depolarization of mitochondrial membrane is one of the early stages of apoptosis and Laser Scanning Fluorescence Microscopy allows to determine changes in transmembrane mitochondrial potential under influence of PDT depending on the kind of photosensitizer (PP(Arg)2, Hp(Arg)2), the energy dose (5, 10, 30 and 50 J/cm2) and time periods (24 and 48 hours after irradiation) in the LNCaP (lymphonodal metastasis of prostate carcinoma, the androgen dependent cell line). Cyototoxicity induced by PP(Arg)2- and Hp(Arg)2-based PDT depending on energy dose and time after irradiation in prostate carcinoma is determined with MTT. Generally, it was shown that lower energy doses induce greater changes in transmembrane mitochondrial potential. Hp(Arg)2-based PDT was more effective causing greater mitochondrial membrane depolarization and cell viability decrease in comparison to PP(Arg)2-mediated PDT (in the case of maximal nontoxic photosensitizer doses used).

Objectives. The Afghans, living in poor socioeconomic conditions, are estimated to be a community with a high rate of intestinal parasitic infections. The aim of the study was to estimate the prevalence and species of intestinal parasites among children’s population in eastern Afghanistan and to present the methods of optimizing the techniques for identification of pathogens in light microscopy. The research was carried out as a part of humanitarian project Capacity building of health care system in Ghazni Province. Materials and method. The study involved 500 children aged 7–18 attending the Share Kona and the Khuija Ali High Schools in Ghazni, eastern Afghanistan in the period November 2013-April 2014. Three stool samples were collected from each patient at 2-day intervals, the samples were fixed in 10% formalin, transported to the Military Institute of Medicine in Poland, where they were pooled and examined using five different diagnostic methods in light microscopy (direct smear in Lugol’s solution, Fülleborne’s flotation, decantation in distilled water, Kato-Miura thick smear, and DiaSys/PARASYS sedimentation system). Results. Pathogenic intestinal parasites were detected in 217 patients (43.4%), with the most common Ascaris lumbricoides (35.3%), Giardia intestinalis (31.1%), and Hymenolepis nana (15.7%). The use of direct smear method allowed for the detection of intestinal parasites in 161 individuals. The application of four following testing methods has improved the detection rates of infected patients by 11.2%. Conclusions. The variety of detected intestinal pathogens in examined children’s population has required the use of combination of multiple diagnostic methods in light microscopy, and finally improved the detection rates of intestinal parasites and helped eliminate infections with nematodes, cestodes, trematodes, and protozoa using appropriate treatment in the study population.

In this work we present recent results on nanoscale imaging in the extreme ultraviolet and soft X-ray spectral ranges, describing three novel imaging systems dedicated for high spatial resolution imaging of nanoscale objects with the extreme ultraviolet and soft X-ray radiations. The extreme ultraviolet and soft X-ray full field microscopes operate at 13.8 nm and 2.88 nm wavelengths and are capable of imaging of nanostructures with a sub-50 nm spatial resolution. A soft X-ray contact microscope operates in the "water-window" spectral range from 2.3 to 4.4 nm wavelength, to obtain images of an internal structure of the investigated object in a thin surface layer of soft X-ray light sensitive photoresist. The development of such compact imaging systems may, in the near future, be important from the point of view of new research related to biological, material science, and nanotechnology applications. Such preliminary applications are also shown in the studies of biological samples, including carcinoma cells, diatoms, and neurons.

In view of the rapid growth of interest in AFM technique in surface property investigation and local surface modification we describe here an AFM microscope with optical tip oscillation detection. The modular shear-force/tunneling microscope for surface topography measurement and nanoanodisation is described. The measurement instrument presented here is based on the fiber Fabry-Perot interferometer for the measurement of the conductive microtip oscillation that is used as nano e-beam for local surface anodisation. An advantage of this system is that quantitative measurements of tip vibration amplitude are easily performed.

Opis:
In the paper authors present a special measurement system for observing phase objects. The diffraction phas microscopy makes it possible to measure the dimensions of a tested object with a nanometre resolution. To meet this requirement, it is proposed to apply a spatial transform. The proposed setup can be based either on a two lenses system (called 4f) or a Wollaston prism. Both solutions with all construction aspects are described in the paper. To make a full analysis of the object shape the authors developed an accurate image processing algorithm, also presented in the paper.

In this paper we present the investigation aimed at the photoresist roughness change determination as a reliable estimator of the exposition rate in the processing verification in semiconductor industry. By employing atomic force microscopy as the 3D high resolution surface imaging tool, we tested twelve popular photoresists in terms of the morphological properties changes, while the following radiation doses were applied. Basing on high precision, and repetitive sample positioning, it was possible to perform the tests with high degree of confidence and observe the roughness change dynamics. Various profiles of roughness changes were observed, showing the need for individual study of each material. Moreover, it was possible to select the photoresists which due to poor homogeneity and small roughness changes are not suitable to such a verification. According to our knowledge, no such study was performed so far.

Thin film samples (d ≈40 nm) of tetrahedral amorphous carbon (ta-C), deposited by filtered cathodic vacuum arc, were implanted with Ga⁺ at ion energy E =20 keV and ion fluences D=3×10¹⁴-3×10¹⁵ cm¯² and N⁺ with the same energy and ion fluence D=3×10¹⁴ cm¯². The Ga⁺ ion beam induced surface structural modification of the implanted material, displayed by formation of new phase at non-equilibrium condition, which could be accompanied by considerable changes in the optical properties of the ta-C films. The N⁺ implantation also results in modification of the surface structure. The induced structural modification of the implanted material results in a considerable change of its topography and optical properties. Nanoscale topography and structural properties characterisation of the Ga⁺ and N⁺ implanted films were performed using atomic spectroscopy analysis. The observed considerable surface structural properties modification in the case of the higher fluence Ga⁺ implanted samples results from the relatively high concentration of introduced Ga⁺ atoms, which is of the order of those for the host element.

Plasmodiophorid sporosori (aggregations of resting spores) reach their most complex form in Spongospora subterranea f. sp. subterranea, the biotrophic plant pathogen which causes the economically important disease powdery scab of potato (Solanum tuberosum). Resting spores are the perennation life cycle stage of plasmodiophorids, allowing them to survive for long periods and infect subsequent host generations. Light microscopy was used to measure resting spores and sporosori of Sp. subterranea, and enumerate resting spores in individual sporosori. Mean resting spore diameters differed for two sporosorus collections, being 4.0 μm (from New Zealand) and 4.3 μm (from Switzerland). Counts of resting spores in 4 μm thick serial sections of sporosori from one collection gave a mean of 667 (range 155 to 1,526) resting spores per sporosorus. Number of resting spores per sporosorus was closely related to sporosorus volume, and could be accurately estimated using the formula; number of resting spores = 0.0081 × sporosorus volume (assuming sporosori to be spheroids). Using this formula, mean numbers of resting spores in sporosori from 37 Sp. subterranea collections from field-grown potato tubers from 13 countries were determined to range from 199 to 713. Differences in numbers of resting spores between the collections were statistically significant (P < 0.05), and independent of country or host cultivar of origin, indicating that enumeration should be carried out for individual sporosorus collections to accurately quantify inoculum. Morphology, using scanning electron microscopy, also showed that between 2 and 51% (average 20%) of resting spores released zoospores after exposure to roots of host plants. The formula for resting spore enumeration validated in this study can be used to standardise Sp. subterranea resting spore inoculum for plant pathology studies, and possibly to assist determination of soil inoculum potential for disease risk evaluations.

Opis:
A fractured nutcracker was examined for determining the root cause/s for premature fracture/failure. This is one of the common tools used typically for cracking hard nuts. In this study, metallurgical failure analysis techniques namely, visual inspection, optical microscopy, SEM, and hardness tests were used in investigating the broken product. From the metallurgical analysis, it was determined that the combined effect of low carbon equivalent and presence of inclusions contributed to the sudden fracture of the nut cracking tool.

Opis:
The study involved the use of a bacterial strain isolated from environmental samples which produce the biopolymer in the form of pellets in the submerged culture. This material (bacterial exopolysaccharide) is produced by bacteria of the Komogateibacter xylinus which are prevalent in the environment. The aim of this study was to characterize bacterial exopolysaccharides and commercial dextran-based “microcarriers” in terms of their roughness and cell culture effects, including the morphology and viability of the human hybridoma vascular endothelial cell line EA.hy926. The pellets were characterized using scanning electron microscopy (SEM) and atomic for¬ce microscopy (AFM). The resulting structures were used for cell culture of adherent cells (anchorage¬-dependent cells). At the same time, the cultures with commercial, dextran-based “microcarriers” were carried out for comparative purposes. After com¬pletion of the cell culture (24 hours of culture), the cellulose and commercial “carriers” were analyzed using SEM and AFM. Finally, the obtained cell dens¬ities (fluorescence labelling) and their morphological characteristics (SEM) were compared. The obtained results strongly support the applicability of bacterial exopolysaccharide (EPS) in tissue engineering to build innovative 3D scaffolds for cell culture, the more so that it is technologically possible to produce EPS as spatially complex structure

Presented work is focused on the use of correlation methods for numerical analysis of magnetic stray field over the surface of materials. Obtained results extend our previous findings about application of the autocorrelation function and the fractal analysis for characterization of magnetic surfaces. Several domain images are recorded at various tip-sample gaps (i.e. the lift heights), and then their average widths were extrapolated down to the zero distance in order to estimate the width seen right on the surface. Apart from that, fractal parameters were derived from autocorrelation function, which turned out to be sensitive to the lift height, and might constitute universal measure (the critical lift height), above which the MFM signal became dominated by thermal noises and non-magnetic residual interactions.
Niniejsza praca dotyczy zastosowania metod korelacyjnych do numerycznej analizy obrazów rozkładu pola magnetycznego emitowanego z obszarów spontanicznego namagnesowania. W pracy przedstawiono kontynuację badań nad zastosowaniem funkcji autokorelacji oraz metod analizy fraktalnej w badaniach struktury domenowej oraz charakterystyki emitowanego z nich pola magnetycznego. Wyniki badań wykazują zależność mierzonej metodami mikroskopii sił magnetycznych (MFM) szerokości domen od wysokości skanowania nad badaną powierzchnią, sugerując przeprowadzanie serii pomiarów na różnych wysokościach (h), ich aproksymację z następną ekstrapolacją do powierzchni (h=0). Przeprowadzona analiza fraktalna, wskazuje na możliwość jej aplikacji do charakterystyki zmian sygnału magnetycznego rejestrowanego przez MFM. Pozwala ona również na wyznaczenie wysokości krytycznej (hkr), na której sygnałem dominującym stają się nakładające się na sygnał magnetyczny pozostałości niemagnetycznych interakcji między igłą sondy i badaną powierzchnią, stanowiące rejestrowany przez MFM szum.

Opis:
In this study, the addition of natural fibers to biodegradable PCL, PLLA blend and the effect of ionizing radiation on the surface of composites were investigated by force modulation microscopy (FMM), atomic force microscopy (AFM) and scanning electron microscopy (SEM). Hot pressed sheets were prepared using pellets of twin extruded PCL:PLLA 20:80 (w:w) blend containing 5% or 10% weight concentration of coconut fiber. In this study coconut fibers non treated chemically and acetylated ones were used. Irradiation was performed using an electron beam (EB) accelerator and an absorbed dose of 100 kGy. FMM images acquisition was obtained using a silicon cantilever, intermittent contact mode (tapping mode). Also, AFM images were obtained using tapping mode but J scanner. By FMM, it was possible to observe regions with different elasticity indicating fiber presence under the surface of the composite. Furthermore, it appears that the spherical structures size decreased on composites. This probably was induced by ionizing radiation.

The genus Calciarcus is revisited here with the purpose of providing, based on additional sampling from both polar regions, an update on species diversity and morphology that can serve as a reference for future work. The geographic realm of the genus is significantly widened and a case is built based on consistency in appearance in favour of adding Calciarcus spp. to the well-defined community of bipolar weakly calcified coccolithophorid genera. Despite the multitude of specimens available for analysis and the fact that the specimens examined distribute themselves within three clusters based on morphological features of coccolith superstructures, it has not been possible at this stage to define a robust framework for differentiation among species of Calciarcus. Circumstantial evidence exist linking species of Wigwamma with species of Calciarcus in haploid-diploid life cycles.

The aim of this paper is to analyse high-resolution optical images of the surface layers of titanium oxide and copper oxide. The materials were produced using the PREVAC Modular Platform for layer deposition located at the University of Rzeszów. Images with a magnification of 50x were obtained using an optical microscope. On the basis of the results obtained, the quality of the layers was evaluated based on the image analysis. The quantitative method of analysis was applied, as a result of which the conditions of the manufacturing process for which the obtained layers had the highest homogeneity were indicated. The minimum range of grey shade counting was from 65 to 150 and the maximum range from 135 to 220. The standard deviation was from 8 to 40%.

In order to measure the Seebeck coefficient of nanometer-scale thermoelectric materials, we propose a new technique in which the thermoelectric-motive force (TEMF) is evaluated by Kelvin-probe force microscopy (KFM). In this study, we measured the Seebeck coefficient of an n-type Si wafer. The surface-potential difference between the high- and low-temperature regions on the Si wafer increases with increasing temperature difference. This indicates that the TEMF can be measured by KFM. The Seebeck coefficient evaluated from the surface-potential difference is 0.71š0.08 mV/K, which is close to that obtained by the conventional method.

New NWA XXX ureilite found in 2006 was investigated by optical microscopy in reflection and in transmission. Olivines, pigeonite, metal veins and dots, and carbon phases have been revealed and their features and distribution studied. Twinning of pigeonite, reduction rims, and poikilitic inclusions of olivine in large pigeonite crystals have been identified and characterized. NWA XXX meteorite belongs to olivine-pigeonite ureilites.

Benzanthrone derivates are now widely used in many industrial and scientific applications as dyes for polymers and textiles. In biochemical, biomedical and diagnostics investigations benzanthrone dyes are used as a lipophilic fluorescent probe since many benzanthrone derivates demonstrate bright fluorescence and they have ability to intercalate between membrane lipids. The aim of research presented here was to assess the luminescence ability of benzanthrone derivatives using microscopic visualization of biological objects. Accordingly, specimens of freshwater trematodes: Diplostomum spathaceum, Diplodiscus subclavatus and Prosotocus confusus, were stained by novel benzanthrone dyes using different fixatives. The samples were examined under a confocal laser scanning microscope. All of the dyes tested demonstrated good results for digestive and reproductive system visualization. Based on obtained results we conclude that benzanthrone dyes could be used for internal and external structure confocal laser scanning microscopic imaging of trematode specimens.

Structural changes in rapeseed following hydrothermal treatment are reported . SEM and TEM micrographs revealed significant changes in the seed microstructure. It was found that the intensity of these changes depends on the kind of treatment.
W prezentowanej pracy przedstawiono wyniki badań zmian mikrostruktury nasion rzepaku poddanych wybranym procesom mającym na celu inaktywację enzymu tioglukozydazy. Za pomocą skaningowego i transmisyjnego mikroskopu elektronowego wykazano zróżnicowany wpływ tych procesów na podstawowe składniki chemiczne nasion rzepaku - białko i tłuszcz. W kontrolnych nasionach rzepaku odmiany Skrzeszowicki zaobserwowano wy stępowanie dwóch typów ciał białkowych. Różniły się one głównie wielkością i rodzajem powierzchni (fot. 1-3). Wysunięto przypuszczenie, że ciała te mogą mieć znaczenie w lokalizacji tzw. ,,bomby olejku gorczycznego". Wykazano, że parowanie nasion w warstwie fluidalnej oraz działanie mikrofal powodują praktycznie identyczne zmiany mikrostruktury. Przejawiają się one w silnej denaturacji ciał białkowych, gromadzeniu się masy białkowe j w centralnej części komórki, a tłuszczu w pobliżu ścian komórkowych (fot. 4-7). Zastosowanie promieniowania γ spowodowało jedynie nieznaczną denaturację ciał białkowych, a także miało minimalny wpływ na zanik membran wokół kropel tłuszczu (fot. 8 i 9).

INTRODUCTION: Adenoids are nasopharyngeal lymphoid tissue with a relevant role in host defence against infection of upper respiratory tract. Nevertheless, adenoids are also a reservoir of microorganisms that can cause infections of upper respiratory tract and otitis particularly in children. OBJECTIVE: Evaluate and compare the association between biofilm assembly on adenoids and the incidence of recurrent infections in a paediatric population submitted to adenoidectomy by either infectious or non-infectious indication. METHODS: Scanning electron microscopy was used to assess biofilms on adenoid surface; biofilm assembly in vitro was monitored by crystal violet assay; antibiotic susceptibility was assessed following EUCAST guidelines; Hinfluenzae capsular typing was performed by PCR. RESULTS: Biofilms were present in 27.4% of adenoid samples and no statistical difference was found between infectious and non-infectious groups. In vitro, the most clinically relevant bacteria, H.influenzae, S.aureus, S.pyogenes, S.pneumoniae and M.catarrhalis, were mostly moderate biofilm assemblers (71.7%). 55.3% of these bacteria were intermediate/resistant to at least one of the tested antibiotics. No association was found between the ability to assemble biofilms in vitro and the presence of biofilms on adenoids nor antibiotic resistance. All H.influenzae were characterized as non-typeable. CONCLUSION: The presence of biofilms on adenoid surface was independent from clinical sample background. Bacterial ability to assemble biofilms in vitro cannot be used to predict biofilm assembly in vivo. The lack of correlation between biofilm formation and infectious respiratory diseases found contributes to question the relevance of biofilms on the pathogenesis of infectious diseases.

Metal-intermetallic layered (MIL) composites attract considerable attention due to their remarkable structural and ballistic performance. This study aimed to develop a Ti/Al-based multilayered MIL material by adding ceramic powders, since they can improve the composite’s impact resistance. To this end, an experiment was conducted which a stack of alternating Ti and al sheets bonded by hot pressing; Ti/Al multilayers containing additional layers of Al2O3 and SiC powders were also produced. The samples obtained were examined using electron microscopy techniques. The clads’ mechanical properties were investigated using a Charpy hammer. In the reaction zone, only one intermetallic phase occurred: the Al3Ti phase. The model with an additional Al2O3 layer showed the highest impact energy. none of the Ti/Al clads broke during the Charpy impact test, a result proving their high ductility.

Pure aluminium (6N) and commercially pure aluminium (99.7) was deformed by KOBO method. Microstructure and texture of both materials after deformation was analyzed by means of scanning and transmission electron microscopy. Advanced methods of crystallographic orientations measurements like Electron Backscatter Diffraction - EBSD (SEM) and microdiffraction (TEM) was used. Grain size distribution and misorientation between grains in cross and longitudinal sections of the samples were analyzed. Differences in size and homogeneity of the grains were observed in both materials. Pure aluminium was characterized by larger grain size in both sections of extruded material. Whereas commercially pure aluminium reveals smaller grain size and more homogeneous and stable microstructure.

The article presents the possibilities of using acoustic microscopy to evaluate defects in resistance spot welding joints. For this purpose, the welded joints were made from two grades of aluminium plates EN AW5754 H24 and EN AW6005 T606, which were then subjected to non-destructive testing using acoustic microscopy and conventional destructive testing using traditional light microscopy techniques. Additionally, the study examined the influence of the typical contaminants found in industrial conditions on the quality of the joint.

To study the occurrence of Cryptosporidium infection in laboratory rats (Rattus norvegicus) raised for experimental usage, 134 faecal samples were obtained from two rearing houses in Ibadan and examined for the presence of Cryptosporidium oocyst using the modified acid fast staining technique. Cryptosporidium species in 2 samples positive for microscopy were further characterized by a nested polymerase chain reaction (PCR) amplifying the 18S rRNA gene. Two of 134 samples were positive for the Cryptosporidium oocysts. Sequencing of the small-subunit rRNA amplicons identified the species in the two PCR positive samples as Cryptosporidium andersoni and Cryptosporidium rat genotype. These findings showed that laboratory rat is a potential reservoir for diverse Cryptosporidium species and suggests that laboratory rats should be screened for Cryptosporidium infection prior to experiments, especially where pathogen free animals are not available. This the first report to identify Cryptosporidium species infecting laboratory rats in Nigeria.

The paper presents results on whether and how the magnetic domain structure in maraging steel undergoes any change due to the aging process. It continues the works on application of correlation methods and fractal analysis into studies of magnetic properties of various steel alloys using Scanning Probe Microscopy. It is aimed at verifying the usefulness of the structure function for description of spatial changes in shape and orientation of magnetic domains, and the magnitude of magnetic stray field affected by the heat treatment. Obtained results suggest that despite vanishing magnetic tip-surface interaction, and hence vanishing magnetic stray field, magnetic domains remain nearly perfectly isotropic although randomly oriented. Similar to isotropic real surfaces, the fractal dimension of the magnetic domains turns out to be independent of the structural changes induced by the aging process, whereas the topothesy is found to vary within order of magnitude along with changes in the magnetic stray field. After all, obtained results lead to conclusion that the aging process leaves its fingerprints in the structure of the steel alloys, which can be studied using numerical analysis of images of magnetic interactions between vibrating tip and the stray field.

Opis:
It was found out that participation of lactobacilli, streptococci and yeasts in kefir grailns' microflora amounted to 66, 16 and 18% respectively. Scanning and transmission microscopy revealed that lactic acid bacteria formed microcolonies mainly inside the grains and yeasts at the surface layers.
W pracy określono proporcje pomiędzy różnymi formami morfologicznymi drobnoustrojów występujących w ziarnach kefirowych oraz badano strukturę i budowę ziarn w mikroskopie elektronowym. Przy liczeniu bakterii jako jednostkę traktowano zarówno komórki pojedyncze, jak też dwoinki i łańcuszki, przy liczeniu drożdży - poszczególne komórki. Liczenie komórek w preparatach barwionych sporządzonych z ziarn kefirowych rozpuszczonych w O, 1 N NaOH wykazało, że liczbowo pałeczki stanowiły średnio 66% mikroflory (wahania 62-69%), zierniaki 16%(wahania 11-22%), a drożdże 18% (wahania 16-20%). Obserwacje przeprowadzone w elektronowym mikroskopie skaningowym pozwoliły stwierdzić występowanie na powierzchni ziarn dużych skupień komórek drożdży i pałeczek,. co sugeruje, że rozwijają się one w mikrokoloniach tworzących masę ziarna. Ziarniaki występowały w obrębie tych skupień jako dwoinki i krótkie łańcuszki (rys. 2). W barwionych octanem uranylu ultracienkich skrawkach z ziarn kefirowych obserwowanych w elektronowym mikroskopie transmisyjnym zaobserwowano wewnątrz ziarn głównie komórki bakterii. Drożdże występowały nielicznie, co wskazuje na fakt silniejszego ich rozwoju w powierzchniowych częściach ziarn. Komórki drobnoustrojów zatopione były w bezpostaciowej substancji (rys. 3 i 4).

This geochemical study aims to resolve genesis of precipitation spots on the walls on the Nydeggbrücke in Bern, Switzerland. The bridge is composed of Jurassic limestone and dolomites and coated on both sides with Miocene flysch sandstone. As a result of infiltration of aqueous solutions derived directly from the road embankment into the sandstone, sulfate encrustation on the walls of sandstone has been formed.The study of these precipitates using optical and electron microscopy clearly shows dominant sulfate phases are gypsum (calcium sulfate), mirabilite (sodium hydrated sulfate) and polyhalite (potassium, calcium and magnesium sulfate). Impurities of Ni, Cu, Zn and Pb was encountered by ICP-MS analysis. Identified polymetallic mineralization is associated with the infrastructure of the bridge and the accumulation of pollution from vehicular traffic. This is also confirmed by sulfur and oxygen stable isotope analyses of sulfates.
