Reliability of the ship propulsion system depends, inter alia, on the quality of lubricate oils. Research indicates that exploitive features of oil are reduced with it runtime in the ship engine circuit. At the present the basic parameters representing exploitive features of oil are: water content, total acid number and total base number or viscosity vs. temperature, anti-wear performance, further: content of additives and their concentrations, reaction products, and other contaminating substances, also magnetically separated large ferromagnetic particles, the size, the shape, the composition and concentration of the abnormal wear particles. Additional properties of oil are described based on atomic emission spectroscopy, granulometry of separated from the lubricant sample by magnets and gravity, permeability changes, as well as rheometric and tribometric characteristics. Complexity of composition of lubricate oil raises the idea to use changes of relative content of any compounds for characterization of oil exploitive features. Fluorescence method is possible to use for oil properties characterisation due to the presence mono and polycyclic compounds in oil that induces fluorescence phenomenon. Therefore identification the fluorescence spectra when exploitation time of oil increases could be novel indicator to monitor of lubricate oil exploitive quality. In this study we present changes of oil fluorescence expressed by synchronous spectra, taking into account working time of exemplary lubricate oil in the ship engine.

Opis:
The escalating maritime transport as well as offshore crude oil exploitation and pipeline transportation leads to the increase of feasibility of the discharge of petroleum substances to the sea environment events. Therefore, it is necessary to rapidly detect of oil pollution to minimize the extent of contamination in the seawater. The aim of this study is to discuss the assumptions of the optical methods using for detection of oil pollution in the seawater. We consider the different approaches of oil-in-water detection using spectral active and passive techniques with emphasis on fluorescence techniques. The intensive development of scientific apparatus and methods in light spectroscopy leads to build various airborne or underwater specialized devices (UV-, IR-scanners, fluorosensors), which allows to detect oil pollution in relatively large areas of the sea. The possibility of measurements of oil pollution in the seawater is particularly important for monitoring, especially in very difficult weather conditions and highly waved sea surface. Such possibilities should be given by devices for in-situ measurements based on time-resolved fluorescence. Such measurements allow obtaining the significant information about presence of oil in really short time. As we show in this study, the wide-range development of fluorescence techniques opens the diagnostic opportunity not only for detection of the oil-in-water content but also for identification the type of oil directly in seawater or even determination of the source of oil pollution.

The variable UV-fluorescence of minerals is well known, and this is attributed to some defects in crystallographic structure or some admixtures, e.g. REE elements. So far, UV-fluorescence has not been applied to describe the nature of meteorites. The plates of forty-five meteorites were examined in three simultaneous emission spectra under an epi-fluorescence microscope. Meteorites were characterized quantitatively according to twelve colours of fluorescence, arbitrarily identified. A numerical method of non-metric multidimensional scaling was used to put colour characteristics as well as the meteorites into an ordination space. Each colour characteristic carries its own information independent of the others. NWA 4039, Bilanga, Gujba and NWA 5437 are extremes in the ordination space. Sahara 560 and NWA 791 expressed very poor and very rich fluorescence, respectively. UV-fluorescence can be a good screening method to quickly distinguish space minerals with crystallographic defects and/or those which are contaminated by REE elements. In subsequent research, other more advanced methods will have to be used.

Opis:
General transcription factor TFIIB is one of the basal constituents of the preinitiation complex of eukaryotic RNA polymerase II, acting as a bridge between the preinitiation complex and the polymerase, and binding promoter DNA in an asymmetric manner, thereby defining the direction of the transcription. Methods of fluorescence spectroscopy together with circular dichroism spectroscopy were used to observe conformational changes in the structure of recombinant human TFIIB after binding to specific DNA sequence. To facilitate the exploration of the structural changes, several site-directed mutations have been introduced altering the fluorescence properties of the protein. Our observations showed that binding of specific DNA sequences changed the protein structure and dynamics, and TFIIB may exist in two conformational states, which can be described by a different microenvironment of W52. Fluorescence studies using both intrinsic and exogenous fluorophores showed that these changes significantly depended on the recognition sequence and concerned various regions of the protein, including those interacting with other transcription factors and RNA polymerase II. DNA binding can cause rearrangements in regions of proteins interacting with the polymerase in a manner dependent on the recognized sequences, and therefore, influence the gene expression.

A simple and coumarin-based fluorescence probe has been designed and synthesized with silyl group as recognition group of fluoride ions (F−) in this study. The results showed that the fluorescence intensity of the probe displayed prominent enhancement with addition of F− at 445 nm with incubation of 1 min. There was an excellent linear relationship between fluorescence intensity and fluoride concentration from 0 to 30 μM (0~0.57 ppm), which offered the important condition for the quantitative analysis. In addition, the highly selective response to fluorion, the low detection limit with 28 nM (0.532 ppb), low toxicity and bioimaging afforded an advantage for practical application and detecting fluoride in biological systerms.

Ciliates have a long history of being central in evolutionary and ecological studies on eukaryotic microorganisms. Although thousands of species have been discovered, their total diversity still remains unknown. Here, we will discuss two unsolved problems that hinder the further exploration of ciliate diversity at the species level, and potential solutions to these problems are proposed. First, ciliate morphospecies are difficult to identify because the different silver stains are not scalable (they do not represent high-throughput methods) and basic supplies are lacking (e.g., protargol); a solution may be the development of fluorescent staining techniques. Second, ciliate phylogenetic species are difficult to identify because of extensive paralogy in nuclear-protein-coding genes; a solution may be to concentrate on sequencing mitochondrial genomes. These two approaches could be integrated into a high-throughput fluorescent-single-cell sorting and mitochondrial genomes sequencing process that would enable the observation and better understanding of ciliate species on a massive scale.

Confocal microscopy is commonly used in mineralogy for structural analysis and to investigate the chemical composition of minerals. In our work we performed imaging of structural defects in meteorites i.e. imaging of microstructures, inclusions, fractures and cracks. Information obtained in this way enables better understanding of minerals' origin and evolution. The aim of our work was not concentrated on the deep description of physical processes related to confocal microscope (CSLM) imaging. This paper is limited to the presentation of a few CSLM functions and possibilities, which have practical application in the interpretation of fluorescence microscope imaging of minerals. Confocal microscopy enables identification and preview of localization of selected minerals and therefore can be used to image of the distribution of chemical compounds in minerals.

This study examined the antiproliferative effects of β-escin (E) in cancer cells. The study showed that E inhibited cancer cells growth in a dose-dependent manner. The flow cytometric analysis revealed an escin-induced increase in the sub-G1 DNA content, which is considered to be a marker of apoptosis. Apoptosis was also confirmed by annexin V staining and DNA fragmentation assay. These effects were associated with increased generation of reactive oxygen species (ROS), caspase-3 activation and decreased mitochondrial membrane potential (MMP). Moreover, escin decreased mitochondrial protein content and mitochondrial fluorescence intensity as well as caused depletion of glutathione (GSH). However, activity of glutathione peroxidase (GPx) and glutathione reductase (GR) was not significantly changed in escin-treated cells. In conclusion, our results demonstrated that E has apoptotic effects in human cancer cells through the mechanisms involving mitochondrial perturbation. Although the exact mechanism needs to be investigated further, it can be concluded that E may be a useful candidate agent for cancer treatment.

W pracy przedstawiono podstawy stroboskopowej metody pomiarów czasów życia fluorescencji, aparaturę, opracowaną metodykę pomiarów i analizę procesu dekonwolucji sygnałów. Pokazano, że krzywe zaniku fluorescencji z wykorzystaniem źródeł światła typu LED UV, (280 i 340 nm) mogą znaleźć zastosowanie do charakterystyki aminokwasów, białek, bakterii - w tym symulantów i interferentów bojowych środków biologicznych. Podstawowymi zaletami metody są: jej szybkość, możliwość wykorzystania tanich źródeł wzbudzenia, wysoka czułość, prostota obsługi, modułowa budowa umożliwiająca szybką wymianę źródeł wzbudzenia. Badania przeprowadzone na standardowych wzorcach - NATA, NADH wykazały zgodność czasów życia fluorescencji z danymi literaturowymi.
Stroboscopic method for determination of fluorescence lifetimes, using 280 nm and 340 nm UV LED sources, may be used for characterization of biological substances - including simulants and interferents of Biological Warfare Agents (BWA). Main advantages of this method are: quickness of measurement, cheap UV sources, high sensitivity, simplicity of operation, modular construction allowing quick changing of sources. Principle of stroboscopic method, developed method of measurements and analysis of deconvolution process are presented in this work. Results of measurement of NATA and NADH standards correspond to data published by other authors.

This paper comprises the synthesis and characterization of 3-(4-(dimethylamino)phenyl)-1-phenyl-(2E)-propen-1-one (DMAPPP) and its application as a new laser medium. The absorption and fluorescence spectra of DMAPPP under different solvents and concentrations have been investigated. The amplified spontaneous emission performance of DMAPPP under various concentrations, organic solvents and pump pulse energies of Nd:YAG laser (355 nm) was also studied. The amplified spontaneous emission spectra of DMAPPP in solution were compared with a conventional laser dye of coumarin 503, under the same identical conditions. The gain and the fluorescence quantum yield of DMAPPP were determined. The most important features are: (1) DMAPPP has an excellent photochemical stability, (2) the amplified spontaneous emission from the DMAPPP was tuned in the wavelength region between 515 and 548 nm. This could be the first detailed paper on laser properties of DMAPPP.

Rozwój technologii wyświetlania obrazu doprowadził do opracowania ekranów typu OLED, których użycie jest coraz powszechne. Współcześnie dąży się do poprawy kwantowej wydajności (EQE) OLED. Organiczne diody elektroluminescencyjne zawsze cierpiały z powodu ograniczonej wydajności kwantowej, co jest wywołane zdolnością wykorzystania zaledwie 25% stanów wzbudzonych. TADF, czyli aktywowana termicznie opóźniona fluorescencja, umożliwia użycie do 100% stanów wzbudzonych powstających w diodzie, a zatem zmniejszeniu ulega udział bezemisyjnej relaksacji cząsteczek. Zjawisko to występuje dla cząsteczek o małej całce nakładania HOMO i LUMO, co skutkuje małą przerwą energetyczną singlet-tryplet.
The development of display technology led to invention of OLED screens whose use is becoming more common nowadays. Today one of the goals is to improve OLED external quantum efficiency (EQE). Organic light-emitting diodes have always suffered from limited quantum efficiency due to harvesting of only 25% of excited states. TADF, which is thermally activated delayed fluorescence, allows to harvest up to 100% of excited states formed in the OLED, thus to decrease a contribution of the non-radiative relaxation. This phenomenon occurs for molecules possessing a small integral overlap of HOMO and LUMO, which results in a small singlet-triplet energy gap.

Artificial neural networks have been shown to be able to approximate any continuous nonlinear functions and have been used to build data based empirical models for nonlinear processes. This work studies primarily the performance of neural networks as a tool for predicting the emission spectra of fluorescent materials from their absorbance, and further, tends to the determination of the optimal topology of the neural network for this purpose. In order to do this, spectral data were initially analyzed by a principal component analysis technique. The first four principal components were used as input nodes of neural networks with various training algorithms – namely cascade- and feed-forward algorithms – and also, various numbers of hidden layers and nodes. The obtained results indicate that the RMS error in a testing data set decreased with increasing the number of neurons and the minimal network architecture for a data prediction problem consists of two hidden layers, respectively with 9 and 1 nodes for both neural networks. Additionally, a better performance was obtained with the cascade-forward neural network, especially in a small number of nodes. The obtained results indicate that the neural networks can be used to provide a relationship between the absorbance as an input and the emission as a target.

Photosensitizing dyes are used in fluorescence diagnostics and photodynamic therapy (PDT). These usually hematoporphyrin derivatives (HpD) accumulate preferentially within neoplastic tissues. HpD is a mixture of ether and ester linked porphyrins. Its partially purified form is known under the commercial name of photofrin II (PII). PII emission spectra were studied in a hydrophilic (PBS) and a lipophilic (PC liposomes) environment. Red shift was observed in their emission maxima from 615 nm in buffer solution to 635 nm in lipid. Identical red shift was obtained when the intracellular fluorescence of two cancer cell lines, MCF 7 and Jurkat, incubated with PII was investigated. Thus, intramembrane localization of PII may be suggested. As determined from the total fluorescence intensity, the uptake of PII was only slightly higher for Jurkat than for MCF 7 cells. Nevertheless the kinetics of the uptake was found to be different for both cell lines.

The phenomenon of fluorescence in immunosensors is described in this paper. Both structure and characteristics of biosensors and immunosensors are presented. Types of immunosensors and the response of bioreceptor layers to the reaction with analytes as well as measurements of electrochemical, piezoelectric and optical parameters in immunosensors are also presented. In addition, detection techniques used in studies of optical immunosensors based on light-matter interactions (absorbance, reflectance, dispersion, emission) such as: UV/VIS spectroscopy, reflectometric interference spectroscopy (RIfs), surface plasmon resonance (SPR), optical waveguide light-mode spectroscopy (OWLS), fluorescence spectroscopy. The phenomenon of fluorescence in immunosensors and standard configurations of immunoreactions between an antigen and an antibody (direct, competitive, sandwich, displacement) is described. Fluorescence parameters taken into account in analyses and fluorescence detection techniques used in research of immunosensors are presented. Examples of immunosensor applications are given.

Opis:
The purpose of this study is to analyse the elements and PM10 concentrations in air samples gathered in the winter of 2017/2018 in two small towns, namely Skala and Wadowice. The chemical elements were identified for each sample using the energy dispersive X-ray method. The spectrometer was equipped, among others, with an Mo-X-ray tube which was the source of the photons and the Si(Li) detector. The following chemical elements: Cl, K, Fe, Ca, Zn, Pb, Br, Ti, Cu, Mn, V, Co, Rb, Ni, Sr, and Cr were identifi ed in the samples. In addition, As and Se were identifi ed in Wadowice. First, the results were compared with each other and then with the results for the nearest city. It was observed that the PM10 concentrations were significantly higher than the UE limit value for PM10, which equals 50 gm−3 per 24 h. Moreover, the high concentrations of, among others, K, Pb, Cl or Zn, are likely to be linked with fossil fuels combustion and biomass burning. The levels of element concentrations in Wadowice and Skala resemble the levels observed several years earlier in Krakow.

Opis:
Almost all kinds of fuel applied in combustion engines come from refining industry, in which crude oil serves as basic raw material. However, there are also searched other sources of hydrocarbons which can be used directly or as additives to conventional fuels. The most popular in this regard are vegetable oils such as rapeseed oil, from which are produced esters used as fuel for diesel engines. On the other hand, as additives to gasoline can be used alcohols derived from the fermentation of agricultural products. Another way to obtain liquid hydrocarbons is pyrolysis of scrap tires. In this article, such pyrolytic product is analysed in terms of its fluorescence properties compared to the fluorescent properties of substances derived from the distillation of the pyrolysis product in three following temperatures: 160, 204 and 350 ºC. All the tested substances show relatively strong fluorescence in ultraviolet range. Fluorometric spectra were performed applying the spectrofluorometer Hitachi F-7000 FL with 1x1 cm quartz cuvette. Studied substances were previously diluted in n-hexane to obtain concentration 4 ppm. Amount of fluorescent peaks and their positions depends on kind of distillate. Ipso facto fluorescence method seems be suitable much more – than chemical analyse – for quick identification of substances constituting a components of distillate obtained from pyrolytic product.

Opis:
The fluorescence intensities for various vibronic fine structures in the 1-pyrenecarboxaldehyde (PyCHO) fluorescence show strong environment dependence. In aqueous solvent, the distribution of dye is highly depended on its concentration labels and varies from excimer to monomeric state. UVVis Spectroscopic analysis could not able to detect dye below 10-7M concentration, whereas a New monomeric peak at 342 nm was observed after heating and cooling treatment at above the concentration. At 10-8M concentration, only monomeric distribution of 1-PyCHO reveals a strong temperature (20-50 ?C) induced reversible perturbation of the vibronic band intensities. This suggests the operation of some specific solute-solvent dipole-dipole interaction mechanism strongly influence by heating.

Opisano zastosowania biosensorów w medycynie.
This paper describes the basics of protein biosensors with particular emphasis on their fluorescent properties. Biosensors are classified on receptor layers with particular emphasis on protein biosensors. There is brought closer the base of protein sensor design, taking into account three strategies commonly use in protein engineering in order to improve sensivity, selectivity and stability of protein biosensor: rational design, directed evolution, de novo design. The main construction problems encountered in the design of biosensor and ways of they solving have been described. The recent research on fluorescent protein biosensors receptor layer is presented. In Table 1 there is given the list of proteins and appropriate fluorescent methods: FRET (Föster Resonance energy Transfer), change in fluorescence emission spectra, for Zn2+, Glc ,Gal ,Fruc, Arab, DMAPIP-b, Ca2+, H2O2, Ca2+, β-CD,CB6, CB7, CFP detection. There is described the use of fluorescent protein based biosensors for clinical application. Biosensors as modern and widely used tools in environmental, medical, food industry have been presented and the future trends in scientific research have been given in the paper.

W artykule przedstawiono przegląd analizatorów DNA wykorzystujących technikę real-time PCR i lab-chipy. Zaprezentowano opis opracowanej metodologii i instrumentarium do detekcji fluorescencji z wykorzystaniem miniaturowych komponentów optoelektronicznych i "inteligentnego" oprogramowania analizującego. Przedstawiono dwa przykłady miniaturowych analizatorów real-time PCR/DNA opracowanych w ramach projektów europejskich i krajowych wykorzystujących lab-chipy i nowatorską metodę detekcji fluorymetrycznej.
The paper presents a novel miniaturized optical instrumentation for fluorescence excitation and detection for miniaturized real-time PCR analyzers using lab-on-a-chip (LOC) devices. Application of miniaturized semiconductor laser to fluorescence excitation, CCD-minicamera as fluorescence photodetector and specialized software for optical signal conditioning led to development of low-cost and highly sensitive optical instrumentation. To carry out full characterization of the optical instrumentation, miniaturized thermocycler co-working with LOC was built. The optical instrumentation was tested with three LOC made of different materials and technologies, giving proper detection of fluorescence signals during real-time PCR. Finally, two miniaturized devices for real-time PCR detection and identification of DNA and utilizing described here novel optical instrumentation were briefly described. The scheme and technical realization of the novel instrumentation in laboratory version is shown in Fig. 4. The detection limit of DNA was about 0,1 ng/ml (Fig. 5). It was also found that the optical instrumentation co-works with different constructions of lab-on-a-chips for DNA real-time PCR amplification (Fig. 6). The developed optical instrumentation was successfully applied to two miniaturized devices for DNA detection and identification by real-time PCR. Technical realizations of the devices and views of the lab-on-a-chips are shown in Figs. 7 and 9. In both cases, proper detection of fluorescence signals generated during real-time PCR of Campylobacter j. DNA (Fig. 8) or complementary DNA (Fig. 10) were observed. It confirmed high sensitivity of the developed optical instrumentation.

The in vitro formation of transcription complexes with Escherichia coli RNA polymerase was monitored using fluorescence anisotropy measurements of labeled fragments of DNA. The multicomponent system consisted of holo or core RNA polymerase (RNAP) and lac or gal promoter fragments of DNA (in different configurations), in the presence or absence of CRP activator protein (wt or mutants) with its ligand, cAMP. Values of the apparent binding constants characterizing the system were obtained, as a result of all processes taking place in the system. The interaction of the promoters with core RNAP in the absence of CRP protein was characterized by apparent binding constants of 0.67 and 1.9 × 106 M-1 for lac166 and gal178, respectively, and could be regarded as nonspecific. The presence of wt CRP enhanced the strength of the interaction of core RNAP with the promoter, and even in the case of gal promoter it made this interaction specific (apparent binding constant 2.93 × 107 M-1). Holo RNAP bound the promoters significantly more strongly than core RNAP did (apparent binding constants 1.46 and 40.14 × 106 M-1 for lac166 and gal178, respectively), and the presence of CRP also enhanced the strength of these interactions. The mutation in activator region 1 of CRP did not cause any significant disturbances in the holo RNAP-lac promoter interaction, but mutation in activator region 2 of the activator protein substantially weakened the RNAP-gal promoter interaction.

Picocyanobacterium Synechococcus sp. is very important but still poorly understood component of marine and freshwater ecosystems. In this study, the effect of single and multiple addition of cell-free filtrate obtained from Synechococcus sp. on selected cyanobacteria Synechocystis sp., Geitlerinema amphibium, Nodularia spumigena and Nostoc sp. was investigated. The species present in this work are groups of aquatic phototrophs known to co-occur in the Baltic Sea. The study showed that the picocyanobacterial cell-free filtrate inhibit the growth and changed the cell morphology of filamentous cyanobacteria G. amphibium, N. spumigena and Nostoc sp. It was shown that the addition of cell-free filtrate caused a decline of pigmentation and cell lysis of G. amphibium, N. spumigena and Nostoc sp. compared to the control culture. In addition, it was observed that the filtrate obtained from Synechococcus sp. did not affect the Synechocystis sp. It was found that the filtrate obtained from picocyanobacterium had the strongest effect on growth of G. amphibium, therefore for this cyanobacteria performed additional experiments to showed whether the filtrate affected also photosynthetic pigments, chlorophyll fluorescence and photosynthesis. The study proved that the picocyanobacterial allelopathic compounds reduce the efficiency of photosynthesis, which results in the inhibition of growth of target organisms. This way of interaction may explain the formation of almost monospecific cyanobacterial blooms in many aquatic ecosystems, including in the Baltic Sea.

In this work, we have designed and synthesized the fluorescent probe 1, which was capable to selectively detect fluoride anion (F). More importantly, the probe 1 possessed near-infrared excitation and emission wavelengths (excitation at 650 nm and emission at 695), and the probe solution had changed dramatically from yellow to cyan with the addition of F. In addition, the fluorescence intensity exhibited perfectly positive correlation with concentration of F concentration from 0 to 40 μM (R2 = 0.9972), which offered the important condition for quantitative analysis. The probe 1 owned detection limit of 46 nM. Therefore, this near-infrared probe can be of great benefit for detecting F in practical application.

A new achievement of mobile phone application for fluorometry to identify cocaine concentration using samples made of tested person sweat implementing special designed software has been presented in this paper. Accessibility is one of most dominant features of this method since the main important part is a mobile phone which can be found and used vastly. Reliability of data collected using this technique is strong enough to compete with past time consuming and expensive devices and equipment. The results enable to have a close and precise observation about the amount of cocaine concentration, the same as reference device outputs.

While studying the influence of ionizing radiation or certain chemical agents on cells, it is crucial to not only determine cytotoxicity, but also to follow cell death mechanisms. There are different methods to screen processes of cell death and still very important question remains unanswered about differences in results that could be caused by various experimental steps in procedures. Based on literature review two protocols of cell death determination were compared. First protocol regarded collecting cells floating in medium before trypsinization and following centrifugation of them. In the second protocol floating cells were discarded and attached ones were stained and fixed. In all experiments three different untreated cell lines (A172, DU145 as cancer cell lines and in comparison, fibroblasts (FB CCL 110), as a non- cancerous cell line) were used to test applied protocols. Cells were cultured and death processes were examined at different time points up to 120 h. Compared protocols showed statistically significant differences, especially in terms of necrosis, which was higher when included floating cells from culture medium and then centrifuging them. Therefore, presented results show importance of choosing a valid experimental procedure in case of evaluating cells viability and types of cell death pathways quantitatively.

Lifetime distribution analysis were performed to study the influence of Leu configuration in position 5 on changes of the peptide chain of cyclic analogues of enkephalins containing a fluorescence donor and acceptor in different solvents. The configuration change of Leu5 in all the analogues of enkephalins studied which contain donor-acceptor pairs has no apparent influence on Trp lifetime distributions. In contrast, there is a significant solvent effect on the shape of lifetime distribution.

Fluorescence in situ hybridization (FISH) experiments with specific probes for chromosome 29 and 25 were carried out on a Brown Swiss bull, previously diagnosed as a carrier of 1;29 centric fusion. The hybridization of the chromosome 29-specific probe (BMC 4216 - already located on 29q13), produced signals on two small acrocentrics, but not on the translocated chromosome. The signals appeared on the translocated chromosome and on a single chromosome 25 after hybridization of the chromosome-specific probe (BMC 3224 - previously located on 25q24). According to the actual nomenclature, the analysed aberration is a robertsonian translocation involving chromosomes 1 and 25.

Considering that correctly functioning of diesel ship engine depends on the composition of applied fuel, it is necessary to easy characterise fuels in terms of quality. Therefore taking into account above mentioned, appropriate analyses have been undertaken and the intention of this article is to inform that characterisation of fuel used in marine transport can be described by the specific parameter based on fluorescence excitation-emission spectroscopy. Therefore, for this study fuel typically used in ship engine – Diesel fuel – is considered. For analysis, Diesel fuel dissolved in n-hexane was used and several oil samples for various oil concentration were prepared. Spectrofluorometer Aqualog Horiba was used to record the excitation-emission spectra (EEMs). EEM spectra determined for each of oil concentration were described by two specific peaks for this kind of oil. Based on registered EEMs, the specific indicator - the wavelength-independent fluorescence maximum – coded in two specific wavelengths – excitation and emission, respectively, described as Exmax/Emmax was determined for considered oil concentration. Obtained results indicate the independence from oil concentration only for one detected peak described by Exmax/Emmax = 240/332. Taking into account obtained results, we can conclude that the wavelength-independent fluorescence maximum detected for excitation wavelength at 240 nm corresponds to the emission wavelength at 332 nm due to the independence from the oil concentration, could be a good indicator to characterise this kind of oil – Diesel fuel. We can conclude that the changes in the composition of fuel could be detected in EEM spectra by the change of detected peaks position.

The performance of 1-(9-ethylcarbazol-3-yl)-4,4,4-trifluorobutane-1,3-dione (1) as a fluorescent probe for the monitoring of cationic photopolymerization processes by Fluorescence Probe Technique (FPT) has been evaluated in comparison with the response of 7-diethylamino-4-methylcoumarin (Coumarin 1) (2). Triethylene glycol divinyl ether and diphenyliodonium hexafluorophosphate were used as an example monomer and a cationic photoinitiator respectively. It has been found that the probe 1 withstands the cationic polymerization conditions and provides correct probe response. 1-(9-ethylcarbazol-3-yl)-4,4,4-trifluorobutane-1,3-dione shifts its fluorescence spectrum with progress of cationic photopolymerization of the monomer, which enables the monitoring of the polymerization progress using the fluorescence intensity ratio measured at two different wavelengths as the progress indicator. By comparing the behavior of 1 and 2, it has been documented that the fluorescence spectrum of probe 1 shows a spectacular hypsochromic shift (Δλ = 33 nm) upon the monomer polymerization, while the shift of 2 is three times smaller (Δλ = 11 nm). Moreover, the sensitivity of probe 1 is more than 2.5-times higher than that of any other probes suitable for monitoring cationic polymerization processes, reported previously. Therefore, application of the carbazole derivative (1) as a new probe for the monitoring of the crosslinking process of coatings cured by cationic photopolymerization has been proposed.

Porous carbons obtained from poly(ethylene terephtalate) contained in a mixture with either MgCO3 or Mg(OH)2 were examined as adsorbents for removal of humic acid from water. Adsorption of the model contaminants is discussed in relation to the textural parameters of the obtained carbon materials. Pore structure parameters of the carbonaceous materials were strongly influenced by preparation conditions including temperature and relative amounts of the inorganics used during preparations as template. Porous carbons prepared revealed a potential to purify water from the model contaminant of high molecular weight. The results presented confirmed a key role of mesoporosity in the adsorption of humic acid. Fluorescence spectroscopy was confirmed to be an useful method to evaluate concentration of humic acid in water.

Dansylated analogues of the potent and selective μ opioid peptide agonist [Dmt1]DALDA (H-Dmt-D-Arg-Phe-Lys-NH2; Dmt = 2',6'-dimethyltyrosine) were prepared either by substitution of Nβ-dansyl-α,β-diaminopropionic acid or Nε-dansyllysine for Lys4, or by attachment of a dansyl group to the C-terminal carboxamide function via a linker. All three analogues displayed high μ agonist potency in vitro and the C-terminally dansylated one retained significant μ receptor selectivity. The three analogues showed interesting differences in their fluorescence emission maxima and quantum yields, indicating that the dansyl group in two of them was engaged in intramolecular hydrophobic interactions. These dansylated [Dmt1]DALDA analogues represent valuable tools for binding studies, cellular uptake and intracellular distribution studies, and tissue distribution studies.

This paper presents the results of the evaluation of feed quality conducted on the basis of the content of a tracer coated with a fluorescent substance. The study used the example of compound feed mixture for laying hens. Feed ingredients: maize and kardi primed wet using fluorescent solutions. The substances, such as Tinopal, Uranine and Rhodamine B were excited to shining by ultraviolet radiation. Before mixing process started, two tracers coated with different fluorescent substances were introduced into the mixture. The acquired images of the samples were analysed using the Patan computer program and were the basis of the information about the percentage share of the specified key component. The tests were used to verify the premise that there is a possibility of simultaneous determination of several components in a mixture based on fluorescent method. The results confirmed this assumption.
W pracy przedstawiono wyniki oceny jakości paszy w oparciu o zawartość składnika kluczowego pokrytego substancją fluorescencyjną. Badania przeprowadzono na przykładzie wieloskładnikowej mieszanki paszowej dla kur niosek. Składniki paszy: kukurydze i kardi zaprawiano na mokro rozworami fluorescencyjnymi. Zastosowane substancje: tinopal, uranina i rodamina B wzbudzano do świecenie promieniowaniem ultrafioletowym. Pozyskane obrazy próbek poddawano analizie w programie komputerowym Patan, na podstawie którego uzyskiwano informacje o procentowej zawartości wskazanego składnika kluczowego. Do mieszanki przed rozpoczęciem procesu mieszania wprowadzano dwa trasery pokryte różnymi substancjami fluorescencyjnymi. Testy miały posłużyć do weryfikacji założenia, że istnieje możliwość jednoczesnego oznaczenia zawartości kilku składników w jednej mieszance w oparciu o metodę fluorescencyjną. Uzyskane wyniki potwierdziły założenie.

The effects of crude oils entering marine environment have been investigated since early 1960s when oil pollution became a subject of global environmental concern. Extraction and transportation of crude oils have been contributing to water pollution especially in closed water basins, such as the Baltic Sea. It was estimated that yearly oil input to the Baltic Sea amounts to 21-70 thousands of tons, which is twice higher than in the North Sea and three times higher than in the North Atlantic. Growing pollution loads into the Baltic Sea created a demand for multifarious studies on the environmental effects of oil products. In this study, we focus on bio-optical and ecological aspects of the presence of crude oil in seawater. Dispersed oil droplets occur in seawater as the result of contaminated river inflows, bilge water discharges and as the consequence of mechanical and chemical dispersion of oil spills. Their optical properties depend on oil type, concentration and size distribution. We present further results obtained from the developed fluorescence-based method for determination of the crude oil concentration in natural Baltic seawater and for evaluation of the oil droplets size by applying vacuum filtering. The results have been measured for the samples collected in Southern Baltic Sea during several ship cruises in 2012. We discuss the application of vacuum filtering in the fluorescence analyses in the context of laboratory, in situ and remote detection of dispersed oil.

Despite of numerous actions undertook by the global and local authorities towards protection of marine environment, oil pollution loads into the Baltic Sea still tend to increase and need to be continuously estimated in order to apply the legal regulations. There is a demand for multifarious studies on the environmental effects of oil products. Dispersed oil droplets occur in seawater as the result of contaminated river inflows, bilge water discharges and as the consequence of mechanical and chemical dispersion of oil spills. Their optical properties depend on oil type, concentration and size distribution. Oil content influences many environmental factors, like water quality and bio-optical parameters (e.g. water-leaving radiance, inherent optical properties, seawater fluorescence). We present a unique study of the collected database of crude oil fluorescence spectra for better understanding the correlations between oil optical properties and its concentration, as well as for evaluation of the oil droplets size by the application of vacuum filtering using three filters of different permeability. Fluorescence spectra have been registered for oil-in-water emulsion samples prepared in the laboratory by mechanical dispersion. We discuss the optical properties of crude oil and the relationships between them in the context of potential remote detection of dispersed oil in seawater.

The paper discusses the capabilities of the laser diagnostic system for dispersed multiphase flows measurements, which was recently commissioned at the Institute of Heat Engineering (ITC) at Warsaw University of Technology. The system presented in the paper is multifunctional and is capable to use different measurements techniques such as shadowgraphy, Mie scattering (scattering on the gas-liquid interface) and LIF (laser induced fluorescence). It provides possibility of switching from one technique to another depending on the characteristics of the observed combustion/injection mixing process. The operational features are discussed in-detail. The study presents the required and possible system setups. Special attention was focused on the system components, which are needed for relevant measurement techniques. Moreover, the limitations of the measurement techniques are discussed and the example measurements are presented. The study shows that using different techniques extends the measurement capabilities of the system with minimum investment costs. This results from the fact that certain system components for different techniques are the same. Moreover, one can conclude that the system design allows certain measurement methods to be applied simultaneously, which results in synergy by making possible drawing more advanced conclusions on observed phenomena

A new edition of the standard specification for liquefied hydrocarbon gas EN 589:2018 was introduced in 2018, which changes some parameters for this fuel, including the acceptable sulfur content. Also, this specification constitutes a new standard for determining volatile sulfur content in LPG, i.e. EN 17178: 2019. Studies were carried out to determine the precision of the method according to the above European standard. Repeatability was determined based on tests of 11 LPG samples with different sulfur content and next it was compared with the values given in the applicable standards: EN 17178: 2019 and ASTM D 6667-14 used until now. Each sample was analyzed 7 times. The repeatability values calculated according to EN 17178: 2019 are much lower than the repeatability according to ASTM D 6667-14 and the repeatability determined in this paper for samples with a concentration range above 4 mg/kg. The repeatability values determined in this work are lower than the values calculated according to EN 17178: 2019 and ASTM D 6667-14 for samples with concentrations below 4 mg/kg. The obtained results confirm the legitimacy of reservations of many European laboratories that perform tests of sulfur content in LPG and participants of the work of the CEN/TC 19/WG 23 N 231 working group, with regard of the correctness of determining the precision of the method pursuant to EN 17178: 2019. It was decided consequently to repeat interlaboratory studies that will be aimed at the determination of a new precision, which will soon be carried out. In addition, the intermediate precision, limit of detection (LD) and limit of quantitation (LQ) of the method were determined. The method was verified by examining 139 samples of commercial LPG fuel. The tested samples were eval- uated for compliance with the specifications from 2012 and 2018. It was found that by testing samples according to EN 17178:2019, the criterion for assessing compliance with the requirements of the specification on LPG is tightened compared to testing according to the ASTM D 6667-14 standard.

This paper presents a method of optical fluorescence analysis for the evaluation of homogeneity of multicomponent grain mixtures. This method is based on the evaluation of the content of fluorescent marker. Maize with two degrees of fineness d1 = 1.25 mm and d2 = 2.00 mm was used as a tracer. Maize was covered with Rhodamine B, which emits red light under the influence of ultraviolet radiation. The tracer was introduced into the mixture before the mixing process began. Nine multicomponent grain mixtures were used. The proportion of fluorescent maize was evaluated on the basis of computer image analysis. Additionally, the fraction of the tracer was evaluated using a control method (validation of the accuracy of the proposed method). The results indicate that the degree of the tracer’s fineness influences the results obtained. The use of fluorescent maize with particle size d2 = 2.00 mm allowed to obtain results which differed less from the control method. The average size of the difference in results ranged from 0.20-0.38 for the 2.00 mm tracer and 0.38-1.34 for the 1.25 mm tracer.

Plants of soybean (Glycine max L.) cv. Augusta grew in the soil with addition of Ni in the concentration of 80 (C-80) or 120 mg∙kg-1 dry mass (C-120) or without Ni (C-0), and they were subjected to UV-B (300 nm – 315 nm) dose of 1.8 kJ m-2 d-1 for 28 days. The addition of nickel into soil in the concentration of 120 mg kg-1 (C-120) caused a decrease of the plant height by 35% compared to the control (C-0), and in the presence of UV-B (U-120) – by 43%. The shoot fresh and dry mass in the C-80 and C-120 plants were lower by 33% and 52% than in the C-0. In the presence of Ni, the intensity of net photosynthesis decreased by 55%. UV-B caused an increase of flavonoid content by 25% compared to the control (C-0), and Ni induced a reduction in the content of these compounds from 20% to 40%.

The development of petroleum extraction and transport technology does not ensure complete isolation of these substances from the natural environment. This problem is exacerbated by the location of mining equipment on the sea shelf and the fact that numerous submarine pipelines, tankers and handling terminals can also emit oil pollution. Therefore, the possibility of detecting oil dispersed in the water is particularly important. This paper reports the efforts to identify methods of characterization of the water containing the crude oil emulsion in a very low concentration (a few to several tens of ppm). Due to this, the effect of emulsion concentration on the possibility of its objective characterization using synchronous fluorescence spectra was studied. The similarity of spectra at various oil concentrations was analysed. It has been shown that the stabilization of the shape of synchronous fluorescence spectra occurs at relatively low oil concentrations.

Ciągły rozwój metod wizualizacji śladów wymusił tworzenie nowych, specjalistycznych urządzeń do ich rejestracji. W ostatnich latach na rynku kryminalistycznym pojawił się system TrasoScan do rejestracji i obróbki śladów kryminalistycznych: traseologicznych, daktyloskopijnych lub dokumentów. Obecnie najczęściej wykorzystywany jest w badaniach traseologicznych. Niemniej jednak z dużym powodzeniem może być zastosowany do rejestracji śladów linii papilarnych na foliach daktyloskopijnych, odlewach silikonowych, zabezpieczonych wraz z podłożem, bądź śladów ujawnionych metodami chemicznymi. W niniejszym artykule przedstawiono szerokie spektrum możliwości TrasoScanu w zakresie rejestracji różnego rodzaju śladów daktyloskopijnych.
The continuous development of methods of trace visualization entailed the development of new, specialized trace registration devices. In recent years, the TrasoScan System for registration and processing of forensic traces: traseological traces, fingerprints or documents has appeared on the forensic market. Currently, this device is most often applied to traseological examinations. Notwithstanding the above, it can be successfully used to register fingerprints on dactyloscopic films, silicone casts, those secured together with a surface, or traces revealed by chemical methods. This article presents the broad spectrum of TrasoScan’s capabilities for registering fingerprints of various types.

The depletion of natural resources such as freshwater and cropland makes it necessary to find a new solution for sustainable food production. Aquaponic systems seem to be a great alternative to traditional agriculture, however, there are still many unknowns that need to be explored. It is already known how fish stocking affects water quality in aquaponic systems, but not how it affects the plants’ growth, and especially on chlorophyll fluorescence. In this study, we examined how the density of 0, 2, 4, 8, and 16 stocking fish in five aquaria affects lettuce growth. The first tank was only a hydroponic system with plants but without fish (control). In the remaining four aquaria - 2, 4, 8 and 12 specimens of common carp fry with an average weight of 20 grams (average 8.5-33.2 g) were placed in the aquaponic growing system. Physicochemical analysis of water was conducted to determine the levels of pH, electrical conductivity (EC), N-NO3, N-NO2, N-NH4, P-PO4, O2 and physiological parameters of plants (nitrogen balance index - NBI, chlorophyll content index - CCI, quantum yield - QY, flavonoid content - Flv) were analysed. The results showed that fish stocking density has different effects on plant physiological parameters, but in most cases, was insignificant. It seems that the greater number of fishes and higher density indirectly causes growth inhibition (lower photosynthetic efficiency) due to the increase of N-NO3 and a decrease of O2 in the water.

Human peripheral blood cells stimulated by phytohemagglutinin (which serve as a model of cancerous cells) and resting cells were incubated in dimethyl sulfoxide solutions of various phthalocyanines. In order to diminish the influence of atmospheric oxygen the cells were embedded in a polymer (polyvinyl alcohol) film. Fluorescence spectra of the samples were measured over two regions of excitation wavelengths: at 405 nm (predominant absorption of the cell material) and in the regions of strong absorption of phthalocyanines (at about 605 nm and 337 nm). The intrinsic emission of cell material became changed as a result both of cells' stimulation and of incubation of cells in dye solution. In most cases the stimulated cells when stained by dye exhibited higher long wavelength fluorescence intensity than resting cells. This suggests higher efficiency of dye incorporation into cancerous cells than into healthy cells. The absorption spectra of samples were also measured. The spectra of various phthalocyanines in incubation solvent, in polymer and in the cells embedded in polymer, were compared. The comparison of properties of the cells stimulated for different time periods enabled to establish the conditions of stimulation creating a population of cells incorporating a large number of sensitizing molecules.

Kinetics of the smooth muscle calponin-F-actin interaction was studied by stopped- flow measurements of light scattering and fluorescence intensity of pyrene-labelled F-actin. The intensity and character of the changes in light scattering, and thus the mode of calponin binding to actin filaments leading to changes in their shape and bundling, depend on the molar ratio of the two proteins. Parallel measurements of pyrene-fluorescence quenching upon calponin binding revealed that intrinsic conformational changes in actin filaments are delayed relative to the binding process and are not markedly influenced by the mode of calponin binding. Bundling of actin filaments by calponin was not correlated with fluorescence changes and thus with alterations in the structure of actin filaments.

Molecular dynamics simulations were carried out on tyrosine and phenylalanine and their derivatives with various terminal groups to determine the populations of side-chain rotamers. The obtained populations were compared with those calculated from fluorescence-decay lifetime distributions and NMR studies. It was found that theoretically calculated populations do not match the experimental ones, which suggests that the static rotamer model is inadequate to explain the dynamics of tyrosine and phenylalanine side chain in fluorescence and NMR experiments.

Opis:
The sedimentary organic matter (SOM) assemblages and sedimentology of the Menilite Beds from the Dukla, Grybów and Vrancea units in the Slovakian and Romanian Outer Carpathians are described. Qualitative and quantitative analyses of the SOM help ascertain depositional conditions, while the thermal maturity of the organic matter studied is estimated utilizing the Spore Colour Index and UV light excitation techniques. The sedimentary organic particles were grouped into ten SOM categories: marine palynomorphs (dinoflagellate cysts), sporomorphs (saccate and non-saccate, pollen and spores), freshwater algae (Botryococcus sp., and other freshwater microplankton), phytoclasts (cuticles, translucent wood, opaque wood), resin and amorphous organic matter (AOM). All samples are dominated by AOM. The presence of Botryococcus sp., Pediastrum sp., Pterospermella sp. and Campenia sp., in some samples points to deposition under hyposaline conditions. It is interpreted that the freshwater influx induced water column stratification in the basin, leading to the development of dysoxic to anoxic bottom-water conditions that enhanced the preservation of AOM. Kerogen analysis in UV light and evaluation using the Spore Colour Index demonstrated different thermal maturation patterns from the Slovakian (post-mature) and Romanian (immature) sections. Integrated palynofacies analysis (notably, the presence of freshwater algae) and sedimentological observations (e.g., hummocky cross-stratification) lead to the conclusion that the deposition of the Menilite Beds in the Vrancea Unit (Romania) was relatively proximal to the shoreline, above storm wave base, whereas the Slovakian units (Dukla and Grybów) were deposited in a more distal setting.

The natural seawater contains both dissolved and suspended organic substances originating from natural sources and human activities – like the marine transport fleet among other. To specify the type and quantity of vitally valid as well as dangerous for properly functioning marine ecosystems substances complicated and sophisticated chemical instrumentation and methodologies must be used. Only a small number of seawater components it is possible easily to determine their concentrations – for example, the salt content is determined directly in the bulk of water through simultaneous measurement of electrolytic conductivity and temperature of water. It is worth to search similarly quick method for oil substances directly in the seawater. Taking into account seawater organic pollutants originating from natural sources inter alia crude oils, the presence of refinery petroleum substances due to their fluorescence in ultra-violet light can be determined based on fluorescence spectroscopy. The aim of this paper is to search fluorescence features of oil substances dissolved in natural seawater based on excitation-emission spectroscopy. In the paper fresh and oily contaminated seawater taken from the coastal area of the Baltic Sea were used. As an oil pollution ‘Petro-baltic’ oil were applied. The natural seawater, at first was laboratory exposed to low extremely amount of oil and then it was examined by fluorescence under UV light. The seawater from vicinity of Gdynia (Poland) were tested as fresh and after artificially contaminated by different amount of oil (from 0.5 to 500 ppm). Spectrofluorometer Hitachi F-7000 FL was applied to measure excitation-emission spectra (EEMs). Low amounts of oil (up to several ppm) cause increasing of main peak in excitation-emission spectra (EEMs). Addition of larger amounts of oil results in appearance new peaks, which originate from fluorescence of soluble fractions of oil artificially added to examined water. These specific features of oil describe the spectroscopic signatures of oil, which is the basis to development operational method of the source of oil pollution identification.

The question of identifying the type of oil in exploitation of engine room is associated with both the quality of ship consumables (fuels and lubricants) and the composition and origin of oil in settling tanks or in dumping water. Related issue is the possibility of determining the origin of oil, which is detected in the marine environment. The key question is how oils vary in their ability to fluorescence, understood as the shapes of fluorescence spectra obtained for different wavelengths of light exciting fluorescence. In this paper we present spectra of fluorescence for six chosen oils. Those oils were previously dispersed (emulsified) in the seawater, then extracted into hexane. Fluorescence spectra were obtained using fluorescence spectrometer Perkin Elmer LS55, for excitation wavelengths in the range from 240 nm to 500 nm, and emission wavelengths from 300 nm to 790 nm. In this paper there is shown that in general both total intensity of fluorescence and shapes of spectra of fluorescence depend on the excitation wavelength and are differ for various kinds of oil. In order to visually show the differences in fluorescence abilities of various oils, the results of measurement were placed on a chart of fluorescence intensities in the function of both variables: the excitation wavelength and the emission wavelength (so called the total fluorescence spectra). Analyses of results of the described studies confirm that the different oils fluoresce differently and identification of type of oil is possible by fluorescence spectrophotometry.

The significant part of petroleum substances is transferred to the marine environment after technical activities (inevitable exploitive spills, illegal discharges). However a certain part of petroleum in seawater coming from the natural sources. Therefore, to develop the safety of natural marine environment it is necessary to track not only the anthropogenic pollutants but also the natural sources of petroleum. To distinguish both source of oil the fluorescence properties of oil can be used as the tools to oil identification. The aim of the study is to describe the optical properties of crude oil after it contact with aquatic environment based on fluorescence spectroscopy. For the study, crude oil ‘Petrobaltic’ type potentially found in marine environment due to coming from natural source was applied. To dissolve oil, two different solvent – hexane and Baltic seawater form the Gdynia coast were used. To measure the excitation-emission spectra (EEMs) of oil spectrofluorometer Hitachi F-7000 FL was applied. In the paper, we discuss the changes of oil EEMs in two cases for oil dissolved in n-hexane and for oil dissolved in seawater for various oil concentration. Obtained EEMs for oil dissolved in seawater indicate that EEMs spectrum contain the main peak – describing by the wavelength – independent fluorescence maximum, which was detected for oil dissolved in n-hexane (typical for this kind of oil). However, the intensity of oil fluorescence decreases after oil contact with seawater. This approach could allow expanding the description of complex mixtures of oils as a possible tool proposed for identification the type and origin of oil found in marine environment.

The utilization of X-ray fluorescence technique for the determination of trace element concentrations in environmental and biological samples is presented. The analytical methods used include energy dispersive X-ray fluorescence with polarizing secondary targets, total reflection X-ray fluorescence, direct in-situ X-ray fluorescence, and micro-beam X-ray fluorescence spectrometry. These methods were applied to analysis of different samples including soil, water, plant material and airborne particulate matter collected on polycarbonate filters. The performance and achieved detection limits of elements for different techniques, established by measuring appropriate reference standards, are presented. Also described is the utilization of micro-beam X-ray fluorescence technique for studying element distribution in heterogeneous samples and investigating the 2D- and 3D-morphology of minute samples by means of computerized X-ray absorption and X-ray fluorescence tomography. The different X-ray techniques have their unique advantages. The micro-beam X-ray fluorescence set-up has an advantage of producing very well collimated primary X-ray beam (about 15 mm in diameter), in front of which the analyzed sample can be precisely positioned, providing local information about the sample composition. The energy dispersive X-ray fluorescence with secondary targets offers rapid analysis of broad range of elements (Na–U) combined with a simple sample preparation method. Total reflection X-ray fluorescence technique, characterized by the lowest detection limits of elements, has its leading edge in analysis of liquid samples, and dealing with particle size effects in air particulates collected on filter papers. In-situ X-ray fluorescence spectrometers are truly portable and enable on the spot, in field analysis. It is shown that the combination of several XRF methods

Wykrycie kokainy zażytej przez zawodowego kierowcę jest tak samo ważne jak przeprowadzenie testu na spożycie alkoholu. Skutki zażycia jednej i drugiej używki mogą być tragiczne. Dlatego też, istotne jest wykrycie kokainy lub jej metabolitów, za pomocą prostych w obsłudze i tanich testów, nie tylko w chwili upojenia tym narkotykiem. Pozwoli to na stwierdzenie, że kierowca w czasie pracy (jazdy samochodem) był pod wpływem kokainy. Brak jest w chwili obecnej tego typu testów. W artykule przedstawiono wyniki prac prowadzonych w ramach 7. PR UE LABONFOIL nad tanim testem na obecność kokainy lub jej metabolitów w pocie kierowcy. Test ten bazuje na jednorazowym pasku immunochromatograficznym współpracującym z ręcznym czytnikiem optycznym. Zastosowanie nowatorskiego układu detekcji optycznej umożliwia wykrycie metabolitu kokainy na poziomie do tej pory charakterystycznym dla stacjonarnych urządzeń laboratoryjnych, otwierając tym samym drogę w kierunku nowej generacji przenośnych narkometrów.
Detection of cocaine taken by a professional driver is from safety reasons as dangerous as consumption of alcohol. Therefore, it is crucial to detect cocaine or its metabolite by the use of simple and cheap test not only while the driver is abused. The paper presents a first approach toward portable instrumentation for cheap and rapid quantitative detection of cocaine in a sweat sample developed under EU 7. FP LABONFOIL project. The test consists of a skin patch and a hand-held optical reader (Fig. 1). The reader is connected to a personal computer which collects data and calculates cocaine concentration in the sweat sample (Fig. 2). Details of the immunochromatographic strip and hand-held reader are shown in Figs. 3 and 4. The view of the hand-held reader connected to an ultra-mobile personal computer is shown in Fig. 5. The reader sends images of the flourishing control and test lines to the computer which analyes and calculates fluorescence intensity of the lines (Figs. 6, 7). During experiments immunochromatographic tests with cocaine metabolite concentration corresponding to cocaine concentration from 0 ng/ml to 20 ng/ml were used. The detection areas for different concentrations are presented in Table 1. The fluorescence intensity as a function of the cocaine metabolite concentration is shown in Fig. 8. The detection limit of cocaine was about 2 ng/ml,m which is at least 10-times better than the limit recommended by EC (30 ng/ml).
