Opis:
Simple methods for detection and isolation of protein-porphyrin complexes were elaborated in our laboratory. They are based on the separation of protein-porphyrin complexes in native polyacrylamide gel and measurement of their fluorescence, with the use of two detection systems: the commercially available Gel DocTM 2000 system, and a system specially designed for the purpose of these investigations, concerning protein-porphyrin interactions. The fluorescent complexes can be electro-transferred from the gel onto PVDF membrane, eluted and analyzed in order to identify the protein interacting with porphyrins.

Opis:
Low molecular proteins were extracted and isolated from rapeseed. The main protein were characterised with molecular mass of 14 000 determined using capillary electrophoresis-SDS technique. UV spectrum showed that this protein appears as a complex with phenolic acids.

Opis:
Based on the polymorphism of gliadin and glutenin proteins relationships of 45 cultivars and strains of winter wheat were evaluated. The cluster analysis showed a considerable variation of the investigated genotypes. The similarity indices were calculated using the Nei and Li formula. The genetic distances between the cultivars ranged from 1.00 to 0.12. The highest similarity index - SI=1.00- being proof of the identical physicochemical composition of storage proteins, was found for the pair Farmer and Elena. The groups of similar and genetically distant cultivars have been presented in the form of a dendrogram. The possibility of using the results obtained from the cluster analysis in breeding programmes has been discussed.

Opis:
One- and two-dimensional polyacrylamide gel electrophoresis were used to compare the composition of nuclear polypeptides from normal and В-cell chronic lymphocytic leukemia mononuclear cells. Against two electrophoretically-specific nuclear proteins with molecular weight of 38/39 and 44/46 kD a from leukemic cells rabbit sera were obtained. As it was analyzed by Western blot technique the available antisera recognized the 38/39 kDa antigen in 53 of the 56 (94.6%), while the 44/46 kDa in 46 of the 49 (93.9%) of examined В-CLL nuclear fraction preparations, but not in normal ones. The pi values of described leukaemia-specific antigens were determined; p38/39 had pi in the range of pH 6.55 -7.00 and p44/46 - in the range of pH 6.2-6.4.

Opis:
Badano osocze krwi 72 nurków w wieku 21 do 23 lat nurków metodą elektroforezy bibułowej. Wartości frakcji białkowych w badaniu wyniosły: albuminy 61,7%, α1 globuliny 26%, α2 globuliny 6%, β globuliny 9,5% γ globuliny 20,2%. Wartości te mieściły się w normie fizjologicznej, ale największy procent odchyleń obserwowano w grupie I – nurków początkujących. Autorzy wiążą tę obserwację z możliwością działania czynnika emocjonalnego i chwilowymi zaburzeniami regulacji hormonalnej.
The study consisted in an analysis of the blood plasma of 72 divers aged between 21 and 23 years with paper electrophoresis method. The examined protein fractions reached the following values: albumins 61.7%, α1 globulins 26%, α2 globulins 6%, β globulins 9.5% γ globulins 20.2%. The obtained values were within physiological norm, with the most significant deviations observed in group I – beginner divers. The authors link this observation with a possible impact of an emotional factor and momentary disorders in hormonal regulation.

Opis:
Influence of aqueous phase properties and process parameters on kaolinite particle zeta potential was quantified by electrophoresis experiments. The results indicated that pH strongly altered the zeta potential of kaolinite and it decreases at the beginning and then increases in the range of pH = 2–13. The activity of different cations changes the zeta potential and has the following tendency of Al3+ > Ca2+ > Mg2+ > Na+ and the zeta potential increases due to heterocoagulation of different mineral particles in suspension. It was found that the zeta potential of kaolinite particles increases after the suspension was stirred and decreases at the beginning, and then increases with soaking time. The FTIR results showed that the zeta potential takes into account ion adsorption and the change of Si–O, Al–O and Al–OH groups on the surface of the kaolinite particles.

Influenza viruses are the worldwide major causative agents of human and animal acute respiratory infections. Some of the influenza subtypes have caused epidemics and pandemics among humans. The varieties of methods are available for the rapid isolation and identification of influenza viruses in clinical and environmental samples. Since nucleic acids amplification techniques such as RT-PCR have been adapted, fast and sensitive influenza type and subtype determination is possible. However, in some ambiguous cases other, more detailed assay might be desired. The genetic material of influenza virus is highly unstable and constantly mutates. It is known that single nucleotide polymorphisms (SNPs) results in resistance to commercially available anti-viral drugs. The genetic drift of the virus could also result in weakening of immune response to infection. Finally, in a substantial number of patients co-infection with various virus strains or types has been confirmed. Although the detection of co-infection or presence of minor genetic variants within flu-infected patients is not a routine procedure, a rapid and wide spectrum diagnostics of influenza virus infections could reveal an accurate picture of the disease and more importantly, is crucial for choosing the appropriate therapeutics and virus monitoring. Herein we present the evidences that native gel electrophoresis and MSSCP - a method based on multitemperature single strand conformation polymorphism could furnish a useful technique for minor variants, which escape discovery by conventional diagnostic assays.

Opis:
An analytical method for determination of pesticide Glyphosate in water as a combination of cation-exchange chromatography and capillary electrophoresis is presented. Pure water was spiked with Glyphosate at concentrations 0.1, 0.25, 0.5 and 1 mM and percolated through a strong cation-exchange column packed with Dowex 50WX4-400 resin in its H+ form. The extract was further analyzed by capillary electrophoresis in indirect detection mode. The calibration curve for the pesticide in the range 0.1–2.5 mM was linear and with high degree of reproducibility. The obtained recoveries for all the studied concentrations amount 85%. Afterwards, the possibility to determine Glyphosate at the concentration 0.001mM (0.17 µg/ml) was checked by percolation of 100 ml of water sample through a column. The calculated recovery was 97.7%
