Opis:
Escherichia coli is Gram-negative optionally anaerobic roads which belongs to Enterobacteriaceae family. Includes in a physiological bacterial flora of human and warm-blooded animals large intestine. Escherichia coli is being met in abiotic elements of the environment so as waters, wastewater, sewage sludge, soil and the food. This bacterium is showing the pathogenicity in named terms for the peoples, triggering diseases mainly: gastrointestinal tract and urinary. Quality and quantitative proposed detections method of the bacteria E. coli contains five/six steps: - appointment dry suspended solid, - preparation averaged, test of sample and resuscitation of bacteria, - making dilutions, - enrichment and differentiation in chromogenic-selective medium, - enumerating the amount of cfu E. coli in 1 g of a dry weight, - optionally, the biochemical identification.
Escherichia coli jest Gram-negatywną, względnie beztlenową pałeczką należącą do rodziny Enterobacteriaceae. Wchodzi w skład fizjologicznej flory bakteryjnej jelita grubego człowieka oraz zwierząt stałocieplnych. Spotyka się ją w elementach abiotycznych środowiska, takich jak wody, ścieki, osady ściekowe, gleba i żywność. Bakteria ta w określonych warunkach wykazuje chorobotwórczość dla człowieka, wywołując głównie schorzenia układów pokarmowego i moczowego. Proponowana metoda jakościowa i ilościowa wykrywania bakterii E. coli zawiera pięć/sześć etapów: - oznaczenie suchej masy osadu, - przygotowanie próbki uśrednionej i badawczej oraz przywrócenie bakteriom aktywności fizjologicznej, - wykonanie rozcieńczeń, - namnażanie i identyfikacja na podłożu chromogennym-selektywnym, - wyliczenie ilości jtk E. coli w 1 g suchej masy osadu, - opcjonalnie identyfikacja biochemiczna.

Introduction: Escherichia coli is a component of human physiological flora. Pathogenic E. coli strains are a significant etiologic factor for numerous infections, mainly the urinary system, digestive system, respiratory system as well as bacteraemia and post-operative infections. Purpose: To compare the genetic similarity of Escherichia coli strains, isolated from biological material collected for routine microbiological diagnostics. Materials and methods: The examination performed on the Escherichia coli strains, isolated from material collected from patients hospitalized in various clinics and delivered for routine laboratory diagnostics. The analysis was conducted using the ADRSSR method.Results: As a result of the analysis of genetic similarities of examined strains using the ADRSSR method, nine clones were distinguished, clones A and B considered being most numerous. Clone A was predominant in samples from internal diseases clinics while cloning B – from neonatological clinics. Conclusions: The results point to a significant role of monitoring of homogeneity of bacteria strains isolated in the range of the health care providers. It is directly connected with the safety of hospitalized patients as well as effectiveness and course of the treatment. The use of the ADSRRS method gives the opportunity of early detection of the moment of colonization in the monitored place

Opis:
Rapidly growing commercial poultry production generates large amounts of waste. Waste that accumulates during the poultry slaughter process often remains unprocessed, becoming a serious threat to people’s health and the natural environment. Poultry production waste constitutes problems odour threat and dangerous sanitary threat. The aim of this study was to determine the population size of Escherichia coli and Clostridium perfringens in poultry waste and in successive stages of waste composting. Research material consisted of raw feathers collected immediately after the slaughter, samples of biological sludge from the centrifuge, mixture of straw, feathers and lime and processed compost. Microbiological analyses were conducted with the use of spread plate count method and the substrate was used in accordance with research standards. The size of population of microorganisms in the samples analyzed corresponded to the waste processing stage and group of microbes. The presence of C. perfringens strains was ascertained in all samples, whereas E. coli strains were identified only in raw feathers and centrifuge sludge, being most numerous in unprocessed material. A reverse trend was observed in case of C. perfringens with the highest population density in centrifuge sludge and least density in raw feathers.
Intensywnie rozwijająca się produkcja drobiarska wiąże się z generowaniem dużej ilości odpadów. Powstające produkty uboczne podczas uboju pozostawione w stanie surowym stają się poważnym zagrożeniem dla środowiska naturalnego i zdrowia ludzi. Mogą stwarzać problemy odorotwórcze oraz istotnie groźne zanieczyszczenie sanitarne. Celem przeprowadzonych badań było określenie liczebności E. coli i C. perfringens w odpadach drobiarskich oraz kolejnych etapach ich kompostowania. Materiał badawczy stanowiły świeże pióra bezpośrednio po uboju, osad biologiczny z wirówki, mieszanina słomy, pierza i wapna oraz kompost przerobiony. Analizy mikrobiologiczne wykonano metodą płytkową, wykorzystując wybiórcze podłoża zgodne z normami. Liczebność mikroorganizmów w badanych próbach zależała od fazy przerobu odpadu i analizowanej grupy drobnoustrojów. Obecność bakterii C. perfringens stwierdzono we wszystkich próbach, natomiast bakterie E. coli tylko w piórach surowych i osadzie z wirówki, przy czym najliczniej występowały w materiale nieprzetworzonym. Odwrotną tendencję zaobserwowano w przypadku C. perfringens, które najliczniej zasiedlały osad z wirówki, natomiast ich mniejszą liczebność stwierdzono w piórach surowych.

Enteropathogenic E. coli (EPEC) is a human pathogen using type III secretion system for delivery of proteins directly into the human host. The system contains a single ATPase, EscN, which is essential for uncoupling of proteins from their complexes with chaperones before the delivery. The structure of EscN ATPase (PDB code: 2obm) was used to screen computationally for small molecule inhibitors blocking its active site. Two lead candidates were examined but only one, Compound 54, was selected for further optimization. After extended QSAR optimization, two derivatives were found to be competitive inhibitors of EscN capable of blocking ATPase activity with a Ki below 50 µM. One candidate, WEN05-03, with a Ki=16±2 µM, was also minimally toxic to mammalian cells as determined by other assays. In the cell infection model of HeLa cells with EPEC, Compound WEN05-03 completely blocked actin cluster formation at 100 µM concentration, when analyzed by confocal microscopy. The second best inhibitor of EscN ATPase activity was WEN04-34 with a Ki=46±2 µM. However, the compound was highly toxic to the BALB/3T3 cell line. In summary, the work identifies a compound blocking bacterial ATPase in its active site without causing cellular toxicity to the host cells. It is the first report showing feasibility of using bacterial virulence system ATPase as a target for safe, non-toxic compounds and offering a proof-of-concept for non-antibiotic alternatives.

Opis:
Water is an essential component necessary for sustaining life. Nevertheless, it may pose a serious threat to human life resulting from possible occurrence of pathogenic and potentially pathogenic microorganisms. Therefore, monitoring and continuous microbial surveillance of the water intended for consumption is extremely important. The requirements referring to qualitative parameters which should be met by water are set out in the Regulation of the Minister of Health of 29 March 2007, as amended on 20 April 2010, and the EU directives. The quality of water being supplied to consumers depends on many factors, among other on the type of water intake (surface water or groundwater), efficiency of water treatment and disinfection, and technical condition of the water supply network. The analysis of the sanitary condition of drinking water has been conducted form water intakes in the city of Gorzow Wielkopolski, ie Centralny Kosynierow Gdynskich intake, Klodawa intake and Siedlice intake, and in neighbouring localities, ie. Maszewo and Marwice. The research material was raw and treated water samples. The population size of Faecal streptococci, coliform bacteria, including E. coli, and total mesophilic and psychrophilic bacteria counts was determined. The analysis of the sanitary condition of drinking water has been conducted. The number of coliform bacteria, including E. coli, and streptococci was determined by the method of membrane filters, whereas total mesophilic and psychrophilic bacteria counts using the plate method under incubation conditions conforming to the standards. The presence of mesophilic and psychrophilic bacteria was observed in all examined water intake points. Differences in their counts over the whole year being significant. In three research points, ie Centralny Kosynierow Gdynskich intake, Maszewo intake and Marwice intake, psychrophilic bacteria count was exceeded (100 CFU ۰ cm–3). In the case of pathogenic bacteria, their occurrence was occasionally observed in the drinking water intake points. The water treatment processes applied and, as the final result, water sanitary assessment according to the existing standards and the provision stipulated allow it to be collected for consumption purposes and do not pose the threat to human health and life.
Woda jest podstawowym składnikiem niezbędnym do utrzymania życia. Jednakże, może ona stworzyć poważne zagrożenie dla zdrowia człowieka wynikające z możliwości wystąpienia mikroorganizmów chorobotwórczych i potencjalnie chorobotwórczych. W związku z tym, niezwykle istotna jest kontrola oraz stały nadzór mikrobiologiczny nad wodą przeznaczoną do spożycia. Wytyczne co do parametrów jakościowych, jakim powinna odpowiadać, zawarte są w Rozporządzeniu Ministra Zdrowia z dnia 29 marca 2007 r. wraz ze zmianą z dnia 20 kwietnia 2010 r. oraz Dyrektywach Unii Europejskiej. Jakość wody dostarczanej konsumentom zależy od wielu czynników m.in.: rodzaju ujęcia (woda powierzchniowa lub podziemna), skuteczności procesów uzdatniania i dezynfekcji oraz stanu technicznego sieci wodociągowej. Przeprowadzono analizę stanu sanitarnego wody pitnej pochodzącej z ujęć wodnych zaopatrujących miasto Gorzów Wielkopolski, tj. ujęcie Centralne Kosynierów Gdyńskich, Kłodawa, Siedlice oraz sąsiadujące miejscowości, tj. Maszewo i Marwice. Materiałem badawczym były próbki wody surowej i uzdatnionej. Określono liczebność paciorkowców kałowych, bakterii z grupy coli, w tym E. coli oraz ogólnej liczebności bakterii mezofilnych i psychrofilnych. Liczebność bakterii z grupy coli, w tym E. coli oraz paciorkowców kałowych oznaczono metodą filtrów membranowych, zaś ogólną liczebność bakterii mezofilnych i psychrofilnych metodą płytkową w warunkach inkubacji zgodnych z normami. We wszystkich badanych punktach stwierdzono obecność bakterii mezofilnych i psychrofilnych. Różnice w ich liczebności na przestrzeni całego roku były istotne. W punktach badawczych, tj. Centralne Kosynierów Gdyńskich, Maszewo oraz Marwice odnotowano przekroczenie norm ilości psychrofilii 100 j.t.k. ۰ cm–3. Obecność bakterii chorobotwórczych w badanych próbkach wody pitnej wykryto sporadycznie. Zastosowane procesy uzdatniania i w efekcie końcowym ocena sanitarna wody według obowiązujących norm i zawartej klauzuli pozwalają na jej pobór do spożycia i nie stanowią zagrożenia dla zdrowia i życia ludzi.

Opis:
The aim of conducted studies was to determine the impact of fluidal ashes obtained from coal combined with fermented sewage sludge and straw and using effective microorganisms, on the overall abundance of bacteria, fungi, actinomycetes and coli bacteria. The experiments were conducted in a pot environment. An increase in the number of bacteria and actinomycetes in the samples containing fluidal ashes and various organic components was observed in comparison with the first year of studies. However, the number of coli bacteria, compared with the first two years of the experiment, was significantly reduced.
Celem przeprowadzonych badań było określenie następczego wpływu popiołów fluidalnych z węgla kamiennego w połączeniu z przefermentowanym osadem ściekowym i słomą, przy użyciu efektywnych mikroorganizmów, na ogólną liczebność bakterii, grzybów, promieniowców oraz bakterii z grupy coli. Badania prowadzono w warunkach wazonowych. Stwierdzono wzrost liczebności bakterii i promieniowców w próbkach zawierających popiół fluidalny oraz różne komponenty organiczne w porównaniu z pierwszym rokiem badań. Natomiast liczebność bakterii z grupy coli w porównaniu z dwoma pierwszymi latami doświadczenia uległa istotnemu zmniejszeniu.

The use of antibiotics to treat bacterial infections is becoming less and less effective year by year due to the increasing resistance of bacteria. The microbial evolutionarily acquired resistance to antibiotics increases the threat to man’s life due to difficulties regarding effective therapies to fight infections. Therefore, apart from treatment, it is necessary to introduce appropriate prophylaxis which limits the multiplication of bacterial colonies on everyday use objects. Due to the antibiotic resistance phenomenon, it is important to find a new material with antibacterial properties for FDM 3D printing in medical applications. The work contains research on a new chemical compound used as an additive to thermoplastics. The rhodamine derivative was synthesized via the 4-diphenylaminobenzaldehyde reaction with 1.3-indendione in a boiling mixture of EtOH/H2SO4. The obtained chemical compound was used as a bacteriostatic modifier of the polycarbonate (PC) properties, as such a modification enables application e.g. for medical device housings or for surfaces frequently touched by people. The modifier and the commercially available polymer were compounded with a high-temperature screw extruder and a filament for FDM 3D printer was created. The modified polymer revealed antibacterial properties relative to Escherichia coli and good thermal stability during the processing.

The objective of this study was to evaluate the efficiency of electrocoagulation in the removal of Escherichia coli from domestic and urban wastewaters and to determine the effects of the main operational parameters on the process. An electrocoagulation reactor with aluminum and iron electrodes was built for this purpose. A factorial design was applied, where amperage, treatment time, and pH were considered as the factors and E. coli percent removal was the response variable. After 20 min of treatment, >97% removal efficiency was achieved. The highest E. coli removal efficiency achieved was 99.9% at a neutral pH of 7, amperage of 3 A, and treatment time of 60 min. However, the removal efficiency of close to 99% was also achieved at natural wastewater pH of 8.5. The statistical analyses showed that the three tested factors significantly affected the E. coli removal percentage (p < 0.05). These results indicate that electrocoagulation has a high disinfection power in a primary reactor in removing water contaminants as well as simultaneously removing pathogenic microorganisms when compared to biological treatment processes. This represents an additional benefit, because it will considerably reduce the use of chlorine during the final disinfection stage.

The phosphate rock flotatio n test was carried out using Escherichia coli E. coli ) as a depressant of dolomite. The results showed that E. coli had a great selective depression on dolomite during flotation. With E. coli as a depressant of dolomite, a useful beneficiation index of phosphorus concentrate with P2O5 grade of more than 30% and MgO content less than 1.2% can be obtained by closed circuit of one stage roughing and one stage cleaning. Furthermore, the depression mechanism of E. coli was studied by ad sorption experiments, infrared spectrum, and zeta potential. This study shows that the adsorption ability of E. coli onto dolomite is stronger than that of collophanite. When the pH is greater than 6, E. coli are chemically adsorbed on the surface of the c ollophanite and dolomite,which also increases the negative charge on the surface of the two minerals. The selective adsorption of E. coli cells to dolomite was best when the pH value is about 7.8.

Type 1 fimbriae are one of the most important factors of Escherichia coli adaptation to different niches in the host. Our study indicated that the genetic marker - fimH gene occurred commonly in commensal E. coli derived from healthy humans but expression of the type 1 fimbriae was not observed. Identification of fim structural subunit genes (fimA-fimH) and recombinase fimE and fimB genes showed that many of the strains were carrying an incomplete set of genes and the genes expression study revealed that in strains with complete set of fim genes, the fimC gene, encoding the chaperone protein, was not expressed.

In Sri Lanka, among 2588 Salmonella positive cases, the highest incidences were recorded from Jaffna peninsula during 2005 to 2013. Therefore, the present study aimed to identify the microbiological and chemical contamination status of groundwater (40 well water) sources in Jaffna during November 2016. The total coliform, E. coli, Salmonella spp. and Shigella spp. along with some physico-chemical parameters of groundwater were studied. The results revealed that entire peninsula was contaminated with total coliform and E. coli bacteria and the parameters recorded were not within the WHO and SLS (Sri Lanka Standards) drinking water quality standards. 38% of sampling locations were positive for Salmonella spp. and among them six sampling locations were being used for drinking purposes. The results of the study correlates with the statistics of typhoid cases recorded in Jaffna. Results of the study also revealed that around 80% of wells were not within the values specified in guidelines of the SLS for drinking water quality on electrical conductivity. Further, 15% of wells recorded greater than 10 mg∙dm–3 nitrate, which is still below the SLS drinking water standards (45 mg∙dm–3). According to the water quality data, PCA analysis showed that Jaffna town, Nallur, Tellippalai and Kopay DS divisions has similar characteristics for water quality.
Spośród 2588 przypadków zatrucia salmonellą zanotowanych na Sri Lance największą liczbę zachorowań stwierdzono na półwyspie Jaffna w latach 2005–2013. Dlatego prezentowane badania miały na celu identyfikację chemicznego i mikrobiologicznego zanieczyszczenia wód gruntowych z 40 studni w listopadzie 2016 r. Analizowano całkowitą liczbę bakterii z grupy coli, Escherichia coli, Salmonella spp., Shigella spp. oraz wybrane parametry fizyczne i chemiczne wód gruntowych. Wyniki ujawniły, że cały półwysep jest skażony bakteriami z grupy coli i bakterią Escherichia coli, a analizowane parametry nie spełniają standardów WHO i standardów Sri Lanki co do jakości wody. W 38% badanych stanowisk stwierdzono obecność Salmonella spp., przy czym sześć z nich wykorzystywano jako źródła wody pitnej. Wyniki badań korelowały z rejestrowanymi przypadkami duru brzusznego na półwyspie. Stwierdzono także, że 80% studni nie spełniało wymogów stawianych wodzie pitnej przez normy Sri Lanki w odniesieniu do przewodnictwa elektrolitycznego. Ponadto stężenie azotanów w 15% studni było większe niż 10 mg∙dm–3, ale nie przekraczało granicznej wartości 45 mg∙dm–3 wyznaczonej przez normy dla wody pitnej. Analiza PCA wykazała, że miasto Jaffna oraz gminy Nallur, Tellippalai i Kopay mają wody o podobnych właściwościach.

The phenomenon of antibiotic resistance has dramatically increased in the last few decades, especially in enterobacterial pathogens. Different strains of Escherichia coli have been reported to produce a variety of structurally different siderophores. In the present study, 32 E. coli strains were collected from different clinical settings in Cairo, Egypt and subjected to the antibiotic susceptibility test by using 19 antibiotics belonging to 7 classes of chemical groups. The results indicated that 31 strains could be considered as extensively drug-resistant and only one strain as pan drug-resistant. Siderophores production by all the tested E. coli strains was determined qualitatively and quantitatively. Two E. coli strains coded 21 and 49 were found to be the most potent siderophores producers, with 79.9 and 46.62%, respectively. Bacterial colonies with cured plasmids derived from strain 49 showed susceptibility to all the tested antibiotics. Furthermore, E. coli DH5α cells transformed with the plasmid isolated from E. coli strain 21 or E. coli strain 49 were found to be susceptible to ansamycins, quinolones, and sulfonamide groups of antibiotics. In contrast, both plasmid-cured and plasmid-transformed strains did not produce siderophores, indicating that the genes responsible for siderophores production were located on plasmids and regulated by genes located on the chromosome. On the basis of the obtained results, it could be concluded that there is a positive correlation between antibiotic resistance, especially to quinolones and sulfonamide groups, and siderophores production by E. coli strains used in this study.

Adhesion of enterotoxigenic (ETEC) E. coli to host intestinal cells is mediated by lectin-like fimbriae that bind to specific glycan moieties on the surfaces of enterocytes. To prevent in vitro binding of E. coli F4 fimbriae (F4 ETEC+) to piglet enterocytes, neoglycans were synthesized by the Maillard reaction conjugating lactose (Lac), galacto-oligosaccharides (GOS) or chitin oligosaccharides (Ochit) to porcine serum albumin (PSA). Neoglycans were characterized by SDS-PAGE, intrinsic tryptophan fluorescence and recognition by plant lectins, as well as by F4 ETEC variants. Electrophoretic patterns suggested the binding to PSA of 63, 13 and 2 molecules of Lac, GOS and Ochit, respectively. All neoglycans displayed quenching of tryptophan fluorescence consistent with the degree of glycation estimated by SDS-PAGE. Plant lectins recognized the neoglycans according to their specificity, whereas antigenic variants of F4 ETEC (ab, ac and ad) recognized PSA-Ochit and PSA-Lac with higher affinity than that for GOS. Neoglycans partially hindered the in vitro binding of F4+ ETEC to piglet enterocytes in a dose-dependent manner. The most effective blocking was observed with PSA-Lac that partially inhibited the adhesion of bacteria to enterocytes in a dose dependent manner, as quantified by flow cytometry. Increased production of the cytokines IL-6 and TNF-α was observed in response to F4+ ETEC infection of enterocytes and production was reduced in the presence of PSA-Ochit and PSA-GOS. These results suggest that neoglycans synthesized by the Maillard reaction could be useful in the prophylaxis of diarrhea in piglets.

The ability to form different types of biofilm enables bacteria to survive in a harsh or toxic environment. Different structures of biofilms are related to different surfaces and environment of bacterial growth. The aim of this study was analysis of the biofilm formation of 115 clinical uropathogenic Escherichia coli strains under different growth conditions: surface for biofilm formation, medium composition and time of incubation. The biofilm formation after 24 h, 48 h, 72 h and 96 h was determined spectrophotometrically (A531) after crystal violet staining and it was correlated with bacterial growth (A600). The live and dead cells in biofilm structures was also observed on the glass surface by an epi-fluorescence microscope. Additionally, the presence of rpoS, sdiA and rscA genes was analyzed. The statistical significance was estimated by paired T-test. The observed biofilms were different for each particular strain. The biofilm formation was the highest in the rich medium (LB) after 24 h and its level hasn't changed in time. When biofilm level was compared to bacterial growth (relative biofilm) - it was higher in a minimal medium in comparison to enriched medium. These results suggest that most of the bacterial cells prefer to live in a biofilm community under the difficult environmental conditions. Moreover, biofilm formation on polyurethane surface did not correlate with biofilm formation on glass. It suggests that mechanisms of biofilm formation can be correlated with other bacterial properties. This phenomenon may explain different types of biofilm formation among one species and even one pathotype - uropathogenic Escherichia coli.

Most industries in developing countries of the world, especially hospitals and other clinical settings, lack wastewater treatment facilities, and as such, untreated wastewater from their operations are discharged into water bodies without any form of treatment. This study aimed at the antibiotic sensitivity profile and ESBL production in E. coli isolated from untreated hospital wastewater before discharge into the environment. Untreated wastewater from two hospitals, a State Government-owned hospital (SGH) and a privately-owned hospital (POH) with no wastewater treatment facilities were sampled for a period of four months. Isolation of E. coli was carried out using the pour plate technique on Eosin Methylene Blue agar, while identification was carried out using conventional methods. Determination of ESBL production was done by means of the Double Disc Synergy Technique and antibiotic sensitivity testing was carried out by employing the disc diffusion method. A total of fifty-eight (58) E. coli were obtained: SGH at 55 and POH at 3. Herein, in 100% of the total count, resistance was indicated for ampicillin and ertapenem, while 14%, 11%, 16% and 57% of the total count were resistant to ceftazidime, cefpodoxime, cefotaxime and amoxicillin-clavulanate, respectively. In addition, 94.8% showed resistance to tetracycline, 19% to ciprofloxacin, 6.9% to gentamycin, 39.7% to chloramphenicol and 55% and 47% to sulfamethoxazole-trimethoprim and nalidixic acid, respectively. Furthermore, 94.8% of all the isolates were multidrug resistant (MDR), while 29.3% were ESBL positive. Wastewater from the two hospitals under study contained ESBL positive and MDR E. coli, suggesting a need to forestall a potential threat to public health by treating the wastewater generated by both hospitals before discharge into the environment.

Introduction. Shopping habits have changed over the past several years and consumers choose shopping centres, where they can spend time and visit restaurants. People usually wash hands before meals, but toilets in shopping centres are visited by hundreds of users every day which= makes their cleanliness questionable. Therefore, the aim of this study was to assess the sanitary condition of electrical hand dryers in toilets of the selected shopping centres in Kraków. Material and methods. Air was sampled at the outlet of electrical hand dryers and surface swabs of hand dryers were taken in toilets of five shopping centres. Air sampling was conducted using the MAS-100 impactor. The number of mesophilic bacteria, fungi, actinomycetes, staphylococci and Escherichia coli was determined. The results of air analyses were compared with the limits provided by the Polish Standards on air quality. Results. The number of microorganisms varied between the studied centres. In terms of mesophilic bacteria and fungi the air was unpolluted. The concentration of actinomycetes however significantly exceeded the limits set by the Polish Standards. Numerous Escherichia coli were found both in the air and in swabs in one of the centres. However the number of E. coli and staphylococci did not exceed the minimum infective dose. Conclusions. The studies showed high concentrations of airborne actinomycetes and staphylococci. Also the presence of E. coli in the air and swabs allows the conclusion that legal regulations for concentrations of airborne microorganisms are necessary to make people aware of this problem. However, it was found that using electrical hand dryers does not have impact on human health.
Wstęp. W przeciągu ostatnich lat zwyczaje konsumentów znacznie się zmieniły i klienci coraz częściej rezygnują z osiedlowych sklepów na rzecz centrów handlowych, w których mogą spędzić czas, a także skorzystać z bogatych ofert restauracji. Wielu ludzi odruchowo myje ręce przed posiłkami, jednak toalety w centrach handlowych odwiedzane są przez setki osób dziennie, a ich czystość może być wątpliwa. Celem badań była ocena stanu sanitarno-higienicznego suszarek do rąk w toaletach wybranych centrów handlowych w Krakowie. Materiały i metody. Pobrano powietrze u wylotu suszarek elektrycznych oraz wymazy powierzchniowe wylotów powietrza w suszarkach w toaletach pięciu centrów handlowych. Próbki powietrza pobrano przy użyciu impaktora MAS-100. Oznaczono liczebność bakterii mezofilnych, grzybów, promieniowców, gronkowców i Escherichia coli. W przypadku badań powietrza wyniki porównano z zaleceniami Polskich Norm dotyczących jakości powietrza atmosferycznego. Wyniki. Liczebność drobnoustrojów w badanych centrach handlowych była zróżnicowana. Pod względem liczby bakterii mezofilnych i grzybów powietrze we wszystkich toaletach było niezanieczyszczone. Stężenie promieniowców znacznie przekraczało wartości dopuszczalne w Polskich Normach. Również liczba gronkowców była bardzo wysoka. W jednym z badanych centrów stwierdzono liczne bakterie Escherichia coli, zarówno w powietrzu, jak i w wymazach. Jednak liczebność zarówno E. coli jak i gronkowców nie przekroczyła granicy minimalnej dawki infekcyjnej. Wnioski. W badaniach stwierdzono wysokie stężenia promieniowców i gronkowców. Obecność E. coli w powietrzu i w suszarkach daje podstawę do stwierdzenia, że normy prawne regulujące stężenie mikroorganizmów w powietrzu są potrzebne, aby podkreślić znaczenie problemu. Stwierdzono jednakże, że korzystanie z elektrycznych suszarek do rąk nie zagraża zdrowiu użytkowników.

The most frequent cause of salmonelloses are food poisonings. However, many studies prove the occurrence and possibility of survival of rods of the genus Salmonella in lakes. The bacteria are often isolated from water sources, which act as a bacterial reservoir. Rods of Salmonella are constantly released to the environment, which results in infections of people and animals. In comparison with other microorganisms, bacteria of the genus Salmonella are characterized by a high survival rate in water environments, particularly in waters of highly eutrophicated lakes. Temperature, the shortage of nutrients, osmotic stress and light has the most negative effect on their survival. Due to its common accessibility, water is an environment which plays an essential role in spread of diseases. The aim of this study was to estimate the effect of selected temperatures on the survival rate of bacilli Salmonella Enteritidis, and the dynamics of quantitative changes of the bacteria in time in bathing waters used for recreation, tested under laboratory conditions. As well as to determine the dynamic of quantitative changes of the bacteria in time under laboratory conditions. The samples for the study were collected from a lake in the Bydgoszcz district. In the conducted experiment it was indicated that Salmonella Enteritidis survived longer in water at 4°C (51 days), as compared with 20°C (39 days). In the present study the elimination rate of bacteria population, according to the analysis of regression, amounted to 0.16 log daily at 4°C, whereas at 20°C it assumed a value of 0.15 log daily. Consequently, it may be concluded that a lower temperature has a more stabilizing effect on rods of the genus Salmonella.

Escherichia coli is a Gram-negative, facultatively anaerobic, rod-shaped, coliform bacterium, which is a primary cause of urinary tract infections. Resistance to antibiotics has become a particular problem in recent decades. Consequently, there is an unmet need for new therapeutic options. It has been observed that essential oils have bactericidal effects. The antimicrobial susceptibility testing for Escherichia coli isolates obtained from urine of patients with urinary tract infections was determined via disk diffusion method according to the European Committee on Antimicrobial Susceptibility Testing (EUCAST, 2015). Essential oil from clove – Syzygium aromaticum (L.) Merill et L.M. (Myrtaceae) was analyzed by GC-FID-MS. Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) were detected by using the micro-dilution broth method. Escherichia coli clinical isolates are characterized by high resistance to ampicillin, amoxicillin with clavulanic acid, norfloxacin, trimethoprim/sulfamethoxazole, tetracycline, tobramycin and ticarcillin. Clove oil possesses strong inhibiting and killing properties against E. coli isolates, among them the ones resistant to recommended antibiotics. The results of this study highlight the need for testing the efficacy of new agents to inactivate bacteria in clinical settings.

The presence of Escherichia coli in the air of facilities involved in management and composting of post-slaughter poultry wastes in selected plants of West Western Pomerania region was studied. Measurements were made on four dates in a variety of weather conditions during the year. The study was conducted at 5 objects that differ in the type of waste and the degree of preparation for composting. These were: chemical treatment and preliminary processing plant, liquid wastes reservoir, platform for preparation of materials for composting, storage of biological sediments, and composting facility. Measurement of bacteria count was carried out in accordance with the applicable procedures on selective chromogenic TBX medium. The assays revealed the presence of E. coli at all test objects, but not always on all measurement dates. It has been shown that the presence of E. coli was from 20 to 3047 CFUm-3 of air, although the largest quantities were most frequently detected in the air of the building for post-slaughter waste pre-treatment in chemical treatment plant.

Rzeka Wilga należy do najbardziej zanieczyszczonych rzek w Krakowie. Zanieczyszczenie tej rzeki wynika głównie z obecności obiektów przemysłowych w jej pobliżu. Analizy miana coli próbek wody pobranych w 16 miejscach (począwszy od źródła rzeki w Raciborsku aż do Podgórza w Krakowie) wzdłuż rzeki przeprowadzone zostały w 2014 roku. Analiza ta jest podstawową metodą służącą do wykazania obecności ścieków w ciekach wodnych i jest oparta na pojawieniu się pęcherzyka powietrza w rurce Durhama, co z kolei jest efektem fermentacji laktozy prowadzonej przez bakterie Escherichia coli. Wodę o największej czystości wykazano w środkowym biegu rzeki, gdzie zaobserwowano szybki nurt oraz brak zabudowań gospodarczych w pobliżu rzeki. Wodę złej jakości stwierdzono nawet przy źródle rzeki, gdzie miano coli wynosiło 0,1. Wilga wykazuje tendencję do samooczyszczania się, co można było zaobserwować na odcinkach: Janowice-Sygneczów-Gołkowice oraz Swoszowice-Łagiewniki. Rzeka ta wykazałaby sporą tendencję do samooczyszczania się w przypadku braku ścieków odprowadzanych w sposób pośredni i bezpośredni. Samooczyszczanie się rzek jest procesem długotrwałym, dlatego przy znacznym dopływie ścieków proces ten jest hamowany i przez to trudny do zaobserwowania.

The present study was investigated to determine the competitive interaction between Gram positive (B. subtilis) and Gram negative (P. aeruginosa and E. coli) bacteria in vitro. Bacteria of interest were isolated from soil sample and were identified through morphological and biochemical tests. Bacterial cultures were grown alone and as co- culture in test tubes containing sterile broth. Bacterial growth and their growth inhibition in co- culture tubes were analyzed till 96 h through spectrophotometric assay and colony forming unit (CFU) assay. Spectrophotometric analysis and CFU/mL assay showed that the effect of B. subtilis on P. aeruginosa and E. coli in co- culture tubes was growth inhibitory. This is accompanied by the reduction in absorbance value and CFU/mL of the co-culture tubes. The bactericidal activity of B. subtilis was measured on P. aeruginosa and E. coli using agar well diffusion method. B. subtilis showed zone of inhibition of 8 mm and 6 mm against P. aeruginosa and E. coli, respectively. The antagonistic activity test of B. subtilis favoured the spectrophotometric and colony forming assay results. These results suggest that B. subtilis develop adaptational pathways by extracellular signaling molecules and antibacterial factors in order to compete with bacterial neighbors.

A trehalose synthase gene from Deinococcus radiodurans (DSMZ 20539) containing 1659 bp reading frame encoding 552 amino acids was amplified using PCR. The gene was finally ligated into pET30Ek/LIC vector and expressed after isopropyl β-d-thiogalactopyranoside induction in Escherichia coli (DE3) Rosetta pLysS. The recombinant trehalose synthase (DraTreS) containing a His6-tag at the C-terminus was purified by metal affinity chromatography and characterized. The expressed enzyme is a homodimer with molecular mass of 126.9 kDa and exhibits the highest activity of 11.35 U/mg at pH 7.6 and at 30°C. DraTreS activity was almost unchanged after 2 h preincubation at 45°C and pH 7.6, and retained about 56% of maximal value after 8 h incubation at 50°C. The DraTreS was strongly inhibited by Cu2+, Hg2+, Zn2+, Al3+ and 10 mM Tris. The Km value of maltose conversion was 290.7 mM.

W środowisku coraz powszechnej stwierdzana jest obecność lekoopornych szczepów Escherichia coli. Alternatywną drogę ich eliminacji mogą stanowić biodegradowalne i nietoksyczne substancje, między innymi olejki eteryczne. Celem pracy była ocena działania olejków eterycznych na środowiskowe lekooporne szczepy Escherichia coli. Materiał badawczy stanowiło 10 wyizolowanych ze środowiska naturalnego lekooopornych szczepów E. coli, w tym 8 glukuronidazo-dodatnich. Wrażliwość szczepów na takie antybiotyki, jak: ampicylina, amoksycylina, cefotaksym, chloramfenikol, cyprofloksacyna, ceftazydym, doksycyklina, gentamycyna, kanamycyna, trimetoprim, tetracyklina, streptomycyna, kwas nalidyksowy oraz na sulfonamidy, oznaczono na podłożu Mueller-Hinton. Metodą dyfuzyjną płytkowo-cylinderkową oceniono wrażliwość szczepów na olejki: tymiankowy, oregano, herbaciany, lemongrasowy, cytrynowy i kminkowy oraz tymol w stężeniach 1,5%; 1,0%; 0,5%; 0,25%. Na podstawie uzyskanych wyników stwierdzono, że testowane szczepy E. coli wykazywały oporność na co najmniej połowę testowanych antybiotyków, a 90% z nich było oporne na ampicylinę i amoksycylinę. Także wrażliwość szczepów na testowane olejki eteryczne była zróżnicowana. Tylko olejek tymiankowy w stężeniach 1 i 1,5% zahamował rozwój 90% badanych szczepów E. coli. Olejki herbaciany, lemongrasowy, cytrynowy, oregano i kminkowy w testowanych stężeniach nie hamowały rozwoju testowanych szczepów Escherichia coli.
In an environment increasingly common finding is the presence of antibiotic-resistant strains of Escherichia coli. An alternative way to eliminate them can provide biodegradable and non-toxic substances, including essential oils. The aim of this study was to evaluate the effect of essential oils on environmental, antibiotic-resistant strains of Escherichia coli. The research material consisted of 10 isolated from the environment, antibiotic-resistant strains of E. coli, including 8 glucuronidase-positive. Using the Mueller-Hinton medium was evaluated the sensitivity to antibiotics such as ampicillin, amoxicillin, cefotaxime, chloramphenicol, ciprofloxacin, ceftazidime, doxycycline, gentamycin, kanamycin, trimethoprim, tetracycline, streptomycin, nalidixic acid and sulfonamides. Using the platelet-cylinders diffusion method was assessed the sensitivity of the strains on essential oils: thyme, oregano, tea tree, lemongrass, lemon, and caraway, at concentrations of 1.5%; 1.0%; 0.5%, 0.25%. Based on the results, it was found that the tested strains of E. coli were resistant to at least half of the used antibiotics and 90% of them showed resistance to ampicillin, and amoxicillin. Also, the sensitivity of the strains to tested essential oils has been diverse. Only thyme oil in a concentration of 1.5% inhibited growth of 90% of the tested strains of E. coli. On the other hand, oregano oil in a concentration of 1.5% inhibited the development of 3 out of 10 tested strains. Oils of tea, lemongrass, lemon and caraway, at the mentioned concentrations have been not bactericidal against strains of Escherichia coli.

Water treatment with electrolysis was performed in a specially made electrolytic cell containing TinO2n-1 ceramic anode and Escherichia coli was used as a model organism for disinfection tests. The results showed that even relatively low energy input (0.5-2.5kWh*m-3, depending on water conductivity) in water samples with chloride ions concentration below 50mg*l-1, TinO2n-1 ceramic electrodes generated active chlorine in the range of 0.4-3.5mg Cl2*l-1, which is the level of chorine used for water disinfection. The results also confirmed that disinfection effect is a result of generation of oxidant species from chlorine rather than effect of electricity per se, or formation of radicals in water. At chloride ion concentration about 7mg*l-1 E. coli is not culturable, not able to divide and not respiring. Results showed that increase of the current above 0.02A at chloride ion concentration of 7mg*l-1 was sufficient to inactivate both culturable and viable but nonculturable (VBNC) E. coli. Notably, the ability of bacteria to divide (DVC positive) was lost more rapidly than their ability to respire. Kinetics of disinfection was studied in water sample which was treated with 0.02A at chloride ion concentration of 7mg*l-1. After about 15 minutes of exposure no culturable or able-to-divide E. coli were detected in the sample. Using the TinO2n-1 electrode in the electrolysis process with the presence of chloride ions, in concentration range which is common in raw waters, one can create a level of active chlorine that kills more than 99% of E. coli within 15 minutes. A practically applicable simple model for prediction of disinfection efficacy with electrolytic cell has been proposed.

Background & Aims: Infrared spectroscopy is an increasingly common method for bacterial strains' testing. For the analysis of bacterial IR spectra, advanced mathematical methods such as artificial neural networks must be used. The combination of these two methods has been used previously to analyze taxonomic affiliation of bacteria. The aim of this study was the classification of Escherichia coli strains in terms of susceptibility/resistance to cephalothin on the basis of their infrared spectra. The infrared spectra of 109 uropathogenic E. coli strains were measured. These data are used for classification of E. coli strains by using designed artificial neural networks. Results: The most efficient artificial neural networks classify the E. coli sensitive/resistant strains with an error of 5%. Conclusions: Bacteria can be classified in terms of their antibiotic susceptibility by using infrared spectroscopy and artificial neural networks.

Members of Enterobacteriaceae are known to produce extended-spectrum beta-lactamases (ESBL) which hydrolyze the beta-lactam group of antibiotics. The existence of ESBL-producing Salmonella enterica (S. enterica) and Escherichia coli (E. coli) harbored by urban avifauna was investigated in this study. Dropping samples (n= 180) were collected from six different bird species in the district Jhang, Punjab province, Pakistan. Isolation and identification of ESBL isolates were made by using cefotaxime- (4 mg/L) supplemented MacConkey agar and double disc synergy test (DDST). Polymerase chain reaction (PCR) was performed for the detection of four different ESBL genes including bla CTX-M, bla TEM, bla SHV and bla OXA. A total of 42.69% isolates were confirmed as ESBL via DDST including 30.64% S. enterica and 49.54% E. coli. The incidence of ESBL S. enterica and ESBL E. coli was found highest in egret (Ardea alba) and pigeon (Columba livia) as 64.28% and 78.95%, respectively. The bla CTX-M gene was detected in 57.89% and 64.81% of isolates of S. enterica and E. coli, respectively. Among other genes in S. enterica and E. coli, bla TEM (21.05%, 20.4%); bla SHV (15.78%, 9.26%), and bla OXA (5.26%, 5.56%) were detected, respectively. All of the tested isolates were found resistant to at least one of the thirteen antimicrobial agents except meropenem. To the best of our knowledge, this is the first study reporting the incidence and genetic diversity of ESBL bacteria associated with urban avifauna in Pakistan. The urban avifauna can serve as a potential subject of bio-surveillance to monitor the emergence of antimicrobial-resistant bacteria.

Recombinational repair is a well conserved DNA repair mechanism present in all living organisms. Repair by homologous recombination is generally accurate as it uses undamaged homologous DNA molecule as a repair template. In Escherichia coli homologous recombination repairs both the double-strand breaks and single-strand gaps in DNA. DNA double-strand breaks (DSB) can be induced upon exposure to exogenous sources such as ionizing radiation or endogenous DNA-damaging agents including reactive oxygen species (ROS) as well as during natural biological processes like conjugation. However, the bulk of double strand breaks are formed during replication fork collapse encountering an unrepaired single strand gap in DNA. Under such circumstances DNA replication on the damaged template can be resumed only if supported by homologous recombination. This functional cooperation of homologous recombination with replication machinery enables successful completion of genome duplication and faithful transmission of genetic material to a daughter cell. In eukaryotes, homologous recombination is also involved in essential biological processes such as preservation of genome integrity, DNA damage checkpoint activation, DNA damage repair, DNA replication, mating type switching, transposition, immune system development and meiosis. When unregulated, recombination can lead to genome instability and carcinogenesis.

The rough mutants of Gram-negative bacteria are widely used to induce protective antisera but the nature of the target epitope for such antibodies is not precisely defined. Endotoxin is one of several antigens present on the surface of bacterial cells, which are able to elicit specific antibodies. We studied the specificity of antibodies produced against a conjugate of E. coli J5 endotoxin core oligosaccharide with tetanus toxoid. The use of chemically defined antigen for immunisation excludes the possibility of production of antibodies against other cell surface antigens. A comparison of this monospecific anti-endotoxin serum with antiserum against E. coli J5 whole cells was performed in order to distinguish the role that endotoxin core oligosaccharide plays in the interaction with humoral host defences from that of other potentially important Gram-negative bacterial surface antigens. The reactivity of both sera with smooth and rough lipopolysaccharides was determined in ELISA, immunoblotting and by flow cytometry. Both antisera reacted with similar specificity with most lipopolysaccharides of identical or related core type. Less distinct reactions with endotoxins of the antibacterial serum in comparison with the anti-conjugate serum were found in all serological tests. LPS of E. coli O100 that showed the strongest reactions with both sera was used to stimulate IL-6, TNFα and nitric oxide production by the J-774A.1 cell line. Both sera were used to inhibit that stimulation and no inhibitory effects of the examined sera in comparison with non-immune serum were observed.

The effectiveness of removing indicator bacteria from domestic sewage using a vertical flow filter filled with natural materials has been analysed. The count of Escherichia coli and coliform bacteria was measured in raw and treated sewage and effectiveness of their removal under variable conditions of hydraulic load and depending on the filter filling (type, size and filling proportions) was assessed. A single-layer sand or zeolite filter was the most effective in reducing bacterial contamination. The sand-filled filter allowed the removal of 99.993–99.997% of Escherichia coli and the one filled with fine zeolite of 99.995%. The principal component analysis was used to comprehensively evaluate the filter performance. Three principal components were identified and they were responsible for 81% of raw data variability. The effectiveness of indicator bacteria removal depended mainly on the filling grain size and did not on filling proportions or its type.

Superoxide dismutase 1 (SOD1) is a metalloenzyme that catalyzes the disproportionation of superoxide into molecular oxygen and hydrogen peroxide. In this study, the human SOD1 (hSOD1) gene was cloned, expressed and purified. The hSOD1 gene was amplified from a pool of Bxpc3 cell cDNAs by PCR and cloned into expression vector pET-28a (+). The recombinant soluble hSOD1 was expressed in E. coli BL21 (DE3) at 37°C and purified using nickel column affinity chromatography. Soluble hSOD1 was produced with a yield of 5.9 μg/mL medium. As metal ions can have a certain influence on protein structure and activity, we researched the influences of different concentrations of Cu2+ and Zn2+ on hSOD1 activity at induction and the time of activity detection. The results implied that Cu2+ and Zn2+ do not enhance SOD1 expression and solubility; they can, however, improve the catalytic activity at induction. Meanwhile, Cu2+ and Zn2+ also enhanced the enzyme activity at the time of detection. Furthermore, most other bivalent cations had the potential to replace Zn2+ and Cu2+, and also improved enzyme activity at the time of detection.

Opis:
Recent studies have suggested that carcinoembryonic antigen (CEA)-promoter sequences are active only in CEA-positive cells, filing in the criteria for tumor specific targeting of suicide genes. However, the present study on gene therapy of colon cancer and cell-specificity of CEA promoter, provide evidence that CEA-positive and CEA-negative cells transfected with E. coli cytosine deaminase (CD) gene under the control of CEA promotor sequence are sensitive to enzyme/pro-drug therapy with 5-fluorocytosine (5-FC). Individual clones derived from the CEA-negative cell lines: melanoma Hs294T and glioblastoma T98G after transfection with CD differed profoundly in their sensitivity to 5-FC. The IC50 values for several clones of the CEA-negative cells were almost the same as for CEA-positive colon cancer cells. Such 5-FC-sensitive clones derived from the population of CEA-negative cells, present even in small number, because of the very effective bystender effect of this enzyme/pro-drug system can cause severe problems during therapy by efficiently killing surrounding normal cells. Safety is the major issue in gene therapy. Our data suggest that the safety of gene-directed enzyme pro-drug therapy (GDEPT) with CEA promoter driven expression of therapeutic genes is not so obvious as it has originally been claimed.

Foodborne microorganisms harbor and adheres itself to the food material and surrounding surfaces for a long time and influence the food quality and consumers health. Among these microbes the presence of Enteric indicator bacteria in food premises confers the indication of entero-pathogens, i.e. E. coli, Salmonella, Shigella and Campylobacters that could cause severe systemic infections in consumers. In this study, the hygienic status of confectionery and supplementary food processing facility was evaluated. A total of 10497 examinations were performed on 3499 swab samples collected from food premises and handlers for the analysis of Enteric indicator bacteria. From swabs, 1277 (12.2%) isolates were identified in which Enterobacteriaceae were found with higher frequency 604 (47.3%) followed by Coliforms 293 (30.8%) and Escherichia coli 280 (21.9%) respectively. The mean count (CFU/cm2) was found maximum for plain surfaces (floors, walls and door), while the lowest was for equipment and machinery. Overall isolates percent prevalence was determined where Enterobacteriaceae were 47%, Coliforms 31% and Escherichia coli 22%. Majority of the floor surfaces were highly contaminated, where washing and sanitation practices were observed to be inappropriate. Worker hygiene status was lacking essential food safety and hygiene standards. In general, the Enteric bacteria were found with higher ratio, that could affect the food quality and quantity both to a greater extent with some influences on consumers health.

The role of plasmids of drug resistance R577 and R785 in the development of susceptibility of Escherichia coli K12 strain W1485 to the bactericidal action of serum was studied. Plasmids R577 and R785 were transfered to cells of strain W1485 by means of conjugation. The susceptibility of cells containing the plasmid to serum was compared to that of cells lacking the plasmid. It was found that plasmids R577 and R758 sensitize bacteria to the action of serum.

This comparative study was carried out to investigate the effect of nanosilver, silver-clay nanohybrid, and silver-milled clay nanohybrid coatings on the antibacterial characteristics of paper. Nanosilver (25 ppm) was used as a singleand double-layer coating. The results demonstrated that treatment with pure nanosilver was more efficient in the reduction of microbial growth. Also, milling of clay enabled better maintenance of silver nanoparticles, and led to a greater decrease in bacterial growth than in the case of the original silver-clay nanohybrid. Evaluation of the sustainability of antibacterial characteristics confirmed that, although pure nanosilver treatment achieved better performance in the first 15 minutes than nanohybrid samples, the performance of the nanohybrids improved with the passing of time. As expected, the treatments decreased the brightness of paper, while the opacity increased significantly; pure nanosilver treatment led to lower brightness than the others, and the opacity was higher in the case of the silver-clay nanohybrid than with the other treatments.

The aim of the study was to optimize conditions for producing Salmonella Enteritidis recombinant heat shock protein 60 (rHsp60). Seven Escherichia coli host strains (Rosetta, Turner, C41, C43, Origami, BL21pLys, Rosetta pLys) were transformed by a recombinant plasmid containing Hsp60 gene from Salmonella Enteritidis, and then cultured and induced by isopropyl-β-D-thiogalactopyranoside (IPTG). The highest S. Enteritidis rHsp60 yield was obtained using E. coli strain C41. Induction of this strain using IPTG allowed the yield 400 μg of S. Enteritidis Hsp60 protein/2L of culture, but by autoinduction the yield exceeded 800 μg/2L.

W pracy przedstawiono wyniki badań wody pochodzącej ze śródmiejskiego jeziora Rusałki, zlokalizowanego na terenie Szczecina. Próbki wody badano przez pięć miesięcy, określając w nich obecność F-specyficznych bakteriofagów RNA oraz bakterii z grupy coli. Na podstawie tych badań wykazano duże zanieczyszczenie badanej wody pod względem sanitarnym.
The paper presents results of preliminary studies of water samples originating from the urban lake called Rusałka situated in Szczecin. Water samples were analysed over five months (November 2008-March 2009) and the presence of F-specific RNA bacteriophages and coliform bacteria were investigated. Results obtained in our study show a high pollution of analysed lake. This was confirmed by the presence and a high number of F-specific RNA bacteriophages that are proposed as an alternative pollution indicator in studies of the sanitary state of water basins.

Escherichia coli producing extended-spectrum beta-lactamase (ESBL) are antimicrobial-resistant Enterobacteriaceae important in the livestock production sector, especially dairy cows because these antimicrobial-resistant bacteria can be transferred to consumers via consumption. If antimicrobials are continually used in dairy farms, this may result in antimicrobial resistance. Therefore, investigation of antimicrobial resistance and finding new alternative methods for inhibiting ESBL-producing E. coli is essential. Hence, the aim of this study was to examine the ability of selected antimicrobials to inhibit E. coli ATCC 25922, control bacteria and ESBL-producing E. coli isolated from dairy farms. We also investigated the capacity of polyphenol extract from 10 varieties of medicinal plants to inhibit ESBL-producing E. coli using a broth microdilution method. It was found that control bacteria were susceptible to all antimicrobial agents, i.e., ampicillin, cefotaxime, ciprofloxacin, chloramphenicol, gentamycin, imipenem, nalidixic acid, tetracycline, and sulfamethoxazole/ trimethoprim. However, ESBL-producing E. coli exhibited both susceptibility and resistance to selected antimicrobials. The polyphenol extracted from Psidium guajava Linn at the lowest concentration was 4.5 mg/mL, which could inhibit control bacteria, but at the same concentration could not inhibit ESBL-producing E. coli. These phenomena indicated that ESBL-producing E. coli had both susceptibility and resistance to antimicrobials. Polyphenol, which could inhibit non-resistant E. coli, could not inhibit ESBL-producing E. coli.

Infections caused by extra-intestinal pathogenic Escherichia coli (ExPEC) are a serious public health problem worldwide. The most troublesome are urinary tract infections, severe neonatal meningitis, serious intraabdominal infections, and more rarely, pneumonia, intravascular-device infections, osteomyelitis, soft-tissue infections or sometimes bacteraemia. These strains are also able cause significant economic losses in animal husbandry. A thorough understanding of ExPEC ecology, reservoirs, chains and dynamics of transmission can greatly contribute to a reduction in the burden of ExPEC-associated disease. The ability of E. coli (including ExPEC) to exist and survive in various ecological niches impedes the precise recognition and indication of transmission routes most important for individual infections cases. Among many identified ExPEC reservoirs, animal companion and animals providing food seem to be important sources of infection for human; however, the real level of risk connected with potential transmission of these bacteria remains unclear. Food is indicated as one of potential ways of transmission. Despite a quite high number of reports, many of the uncertainties are expected to be reliably elucidated. This review presents most important data on the current state of knowledge concerning the potential role of food in ExPEC transmission. The possible consequences of ExPEC infections in human and animals are briefly described.

The Escherichia coli AlkB protein encoded by alkB gene was recently found to repair cytotoxic DNA lesions 1-methyladenine (1-meA) and 3-methylcytosine (3-meC) by using a novel iron-catalysed oxidative demethylation mechanism that protects the cell from the toxic effects of methylating agents. Mutation in alkB results in increased sensitivity to MMS and elevated level of MMS-induced mutations. The aim of this study was to analyse the mutational specificity of alkB117 in a system developed by J.H. Miller involving two sets of E. coli lacZ mutants, CC101-106 allowing the identification of base pair substitutions, and CC107-CC111 indicating frameshift mutations. Of the six possible base substitutions, the presence of alkB117 allele led to an increased level of GC→AT transitions and GC→TA and AT→TA transversions. After MMS treatment the level of GC→AT transitions increased the most, 22-fold. Among frameshift mutations, the most numerous were -2CG, -1G, and -1A deletions and +1G insertion. MMS treatment appreciably increased all of the above types of frameshifts, with additional appearance of the +1A insertion.

Adaptive (starvation-associated) mutations occur in non-dividing cells and allow growth under the selective conditions imposed. We developed a new method for the determination of adaptive mutations in Escherichia coli. The system involves reversion to prototrophy of the argE3OC mutation and was tested on AB1157 strains mutated in the mutT and/or mutY genes. The bacteria that mutated adaptively grow into colonies on minimal medium plates devoid of arginine (starvation conditions) when incubated longer than 4 days. Using the replica plating method we solved the problem of discrimination between growth-dependent and adaptive argE3→Arg+ revertants. Phenotype analysis and susceptibility of the Arg+ revertants to a set of T4 phage mutants create an additional possibility to draw a distinction between these two types of Arg+ revertants.

In the face of shortage of fossil fuel supplies and climate warming triggered by excessive carbon dioxide emission, alternative resources for chemical industry have gained considerable attention. Renewable resources and their derivatives are of particular interest. Glycerol, which constitutes one of the by-products during biodiesel production, is such a substrate. Thus, generated excess glycerol may become an environmental problem, since it cannot be disposed of in the environment. The most promising products obtained from glycerol are polyols, including 1,3-propanediol, an important substrate in the production of synthetic materials, e.g. polyurethanes, unsaturated polyesters, and epoxy resins. Glycerol can be used as a carbon and energy source for microbial growth in industrial microbiology to produce 1,3-propanediol. This paper is a review of metabolic pathways of native producers and E. coli with the acquired ability to produce the diol via genetic manipulations. Culture conditions during 1,3-PDO production and genetic modifications of E. coli used in order to increase efficiency of glycerol bioconversion are also described in this paper.

The gene encoding β-D-galactosidase from Pyrococcus woesei was PCR amplified, cloned, expressed in Escherichia coli under the control of an inducible T7 promoter, purified and characterized. The expression system was developed by the construction of recombinant plasmid, based on the high copy number pUET1 vector, giving four times more efficient expression of P. woesei β-D-galactosidase (20 mg of enzyme from 1 liter of culture) than that obtained from a previously constructed one. The recombinant enzymes were purified in a two-step procedure: double heat-denaturation of E. coli cell proteins and affinity chromatography on p-aminobenzyl 1-thio-β-D-galactopyranoside-agarose. To achieve efficient purification of P. woesei β-D-galactosidase by immobilized metal-ion affinity chromatography (IMAC), a His-tag was placed either at the N- or the C-terminal of the coding sequence. The obtained fusion proteins revealed the same specific activity of approximately 5400 U/mg, which was 10 times lower than the wild-type β-D-galactosidase (51100 U/mg). The activity of P. woesei β-D-galactosidase was enhanced by thiol compounds, Mg2+ ions and D-galactose, and was inhibited by heavy metal ions and D-glucose, while Ca2+ ions had no effect.
