The effect of light conditions on the accumulation of phenolic compounds was investigated in the needles of one-year and two-year old shoots, collected from perennial specimens of common yew (Taxus baccata L.), American arborvitae (Thuja occidentalis L.) and common juniper (Juniperus communis L.), growing in an urban garden. The content of total phenols and flavones was partially related to the light requirement or light tolerance of conifer tree. In Taxus needles, a shade-loving species, higher accumulation of phenolic metabolites was observed under shaded condition (in two-year old shoots) and was linked with the growth intensity in spring. In Thuja and particularly in Juniperus, the level of phenolics was related to the insolation intensity, probably so as a part of the adaptation mechanism.

The field experiment on Stevia rebaudiana (SR), (Family: Asteraceae) was carried out in acid soil zone of Shimoga (Karnataka) in the year 2009 and 2010 to investigate the influence of biofertilizers on protein, moisture and ash content in the dried Stevia leaf and their correlation with swelling property, water absorption capacity and mineral element contents. Results revealed that second harvested sample (August) in first year (2009) was better for all aspects than other harvested samples and the results showed significant increase in protein content (16.22 %), swelling index (SI) (5.10 % w/w), water absorption capacity (WAC) (4.91 ml/g) with the treatment T8 where three bio fertilizers were applied togetherly with simultaneous moisture content varied from 6.61 to 7.18 %. The significant higher correlations were observed (significant at 1 %) between protein content with SI (r = 0.99), WAC (r = 0.99) and SI with WAC (r = 0.98) in 2009 harvested samples. The ash content was higher in third harvested sample (13.54 %) in the year 2009 with simultaneous increased amount of Fe (6.19 mg/kg), Mn (1.24 mg/kg), Cu (0.72 mg/kg) and Zn (1.22 mg/kg) contents due to the residual effect of bio fertilizers. Nitrogen (0.74 mg/kg), P (0.40 mg/kg) and K (0.68 mg/kg) contents were higher during second harvested samples and thereafter decreased. Total phenolic content was also recorded highest (64.52 mg/g) with three bio fertilizers applied togetherly during year 2009 compared to the year 2010 (50.12 mg/g). The results suggest that the application of bio-fertilizers either single or more numbers in a balanced way may be beneficial in improving protein, phenol and mineral contents of stevia plant.

Opis:
The influence of light reduction on the growth and phenolic compounds contents in Scots pine trees and seedlings was studied. Results of the first experiment in field conditions show that shade causes an increase of phenolic compounds and in the second experiment with seedlings under controlled shading conditions the results were opposite. It is suggested that the factor insolation should be paid more attention during the studies on the influence of pollution on the content of phenolic compounds.

Opis:
The effects of combined aminoethoxyvinylglycine (AVG) and modified atmosphere packaging (MAP) treatments on quality attributes of ‘0900 Ziraat’ sweet cherry fruit during the cold storage and shelf life were investigated in this study. Significantly lower weight loss and decay ratios were observed in all treatments throughout the cold storage period as compared to the control. A similar case was also observed referring to the shelf life. MAP treatments were found to be more effective in retarding the weight loss and decay ratio. Higher hue angle values were measured from AVG-treated fruit at harvest. Similarly, hue angle of AVG and MAP-treated fruit were also higher than for the control in all periods of cold storage and on the 7th and 21st day of shelf life. AVG-treated fruit had higher firmness values than the control at harvest. However, higher firmness values were measured from MAP-treated fruit during the cold storage and shelf life. At the end of cold storage, lower SSC and higher titratable acidity values were observed in AVG and MAP-treated fruit than in the control. AVG + MAP treatments yielded significantly higher vitamin C, total phenolics and antioxidant activity values than the control. Contrarily, the control fruit had significantly higher total monomeric anthocyanin than the other treatments. Based on current findings, it was concluded that combined AVG + MAP treatments could be used as a beneficial tool to maintain the quality of sweet cherry fruit throughout the cold storage and shelf life.

Throughout the world, charcoal rot, caused by Macrophomina phaseolina, is one of the most destructive and widespread diseases of crop plants such as soybean. In this study, the biological control capability of 11 Trichoderma spp. isolates against M. phaseolina was investigated using screening tests. Among all the tested Trichoderma spp. isolates, inhibition varied from 20.22 to 58.67% in dual culture tests. Dual culture, volatile and non-volatile tests revealed that two isolates of Trichoderma harzianum (including the isolates T7 and T14) best inhibited the growth of M. phaseolina in vitro. Therefore, these isolates were selected for biocontrol of M. phaseolina in vivo. The results of greenhouse experiments revealed that disease severity in the seed treatment with T. harzianum isolates was significantly lower than that of the soil treatment. In most of the cases, though, soil treatment with T. harzianum resulted in higher plant growth parameters, such as root and shoot weight. The effects of T. harzianum isolates on the activity of peroxidase enzyme and phenolic contents of the soybean root in the presence and absence of M. phaseolina were determined in greenhouse conditions. Our results suggested that a part of the inhibitory effect of T. harzianum isolates on soybean charcoal rot might be related to the indirect influence on M. phaseolina. Plant defense responses were activated as an elicitor in addition to the direct effect on the pathogen growth.

A study was conducted in order to determine the content of phenolic compounds and antioxidative activity of extracts from Flammulina velutipes. Water and ethanolic, methanolic, acetone (70% v/v) extracts were prepared from lyophilized fruiting bodies of mushrooms. Different extraction techniques were used: ultrasonication and stirring by a rotary shaker at ambient temperature and at 50oC, and at boiling point for each solvent. Next, total phenolics by Folin-Ciocalteu method, antioxidant capacity (EC) by ferric reducing antioxidant power assay (FRAP) and scavenging ability on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals were analysed in these extracts. It was observed that the extraction method and the kind of solvent influenced the antioxidant activity and concentration of total phenolics in the extracts. The highest concentration of phenolic compounds (7.58±0.17 mg GAE/g extract) were observed for water extract prepared with use of ultrasonification method (at 50oC for 1h). Results showed that water extracts possessed high equivalent capacity (EC) for all applied extraction methods. The highest EC value (9.2±0.18 mM Fe2+/g extract) was observed in water extract prepared by stirring at ambient temperature for 1 h. This extract was characterized by high level of total phenolics. The acetone extracts, prepared by ultrasonic extraction at 50oC for 1 h, at 10 mg/ml showed the highest scavenging activity (57.53±1.18%), although, the value was lower than that of ascorbic acid at 10 mg/ml (60.50±1.32%).
Praca miała na celu określenie ogólnej zawartości związków polifenolowych i właściwości przeciwutleniających ekstraktów z grzyba Flammulina velutipes. Z liofilizowanych owocników F. velutipes przygotowano ekstrakty wodne, etanolowe (70% v/v), metanolowe (70% v/v) i acetonowe (70% v/v). Zastosowano różne techniki ekstrakcji: wytrząsanie i ekstrakcję przy użyciu łaźni ultradźwiękowej w temperaturze pokojowej i w 50oC oraz ekstrakcję we wrzącym rozpuszczalniku. W otrzymanych ekstraktach zbadano ogólną zawartość związków polifenolowych metodą Folin-Ciocalteu i ich właściwości przeciwutleniające (EC – equivalent capacity) metodą FRAP (ferric reducing antioxidant power assay) oraz zdolność do neutralizacji wolnych rodników z użyciem DPPH (1,1-diphenyl-2-picrylhydrazyl). Zaobserwowano, że takie czynniki jak warunki ekstrakcji czy rodzaj użytego rozpuszczalnika mają wpływ na właściwości przeciwutleniające i ogólną zawartość związków polifenolowych w otrzymanych ekstraktach. Najwyższą ilość związków polifenolowych uzyskano w wodnych ekstraktach otrzymanych z użyciem łaźni ultradźwiękowej w 50oC (7,58±0,17 mg GAE/g ekstraktu). Wyniki pokazują, że wodne ekstrakty, otrzymane wszystkimi zastosowanymi metodami ekstrakcji, posiadają wysokie właściwości przeciwutleniające. Najwyższą wartość EC (9.2±0.18 mM Fe2+/g ekstraktu) zaobserwowano dla wodnych ekstraktów przygotowanych w pokojowej temperaturze z użyciem wytrząsarki. Te ekstrakty charakteryzowały się również wysoką zawartością związków polifenolowych. Ekstrakty acetonowe otrzymane poprzez zastosowanie łaźni ultradźwiękowej w 50oC przy stężeniu 10 mg/ml wykazywały najwyższą zdolność do redukcji wolego rodnika DPPH (57,53±1,18%) ale niższą niż kwas askorbinowy przy takim samym stężeniu (60,50±1,32%).

Total phenolic content, concentration of flavonoids and in vitro antioxidant activity of twenty different extracts from the whole plant and plant parts (leaves, flowers and stems) of Teucrium polium were determined. The total phenolic contents ranged between 14.57 to 157.84 mg of GaA/g of extract. The concentrations of flavonoids varied from 6.48 to 139.87 mg of Ru/g of extract. Antioxidant activity was determined in vitro using DPPH reagent and expressed as concentration of each extract required to inhibit radical by 50% (IC50) values that ranged from 26.30 to 2190.75 μg/ml. The methanolic leaves extract contain the greatest concentration of phenolic compounds (157.84 mg of GaA/g) and showed strong antioxidant activity (IC50 = 26.30 μg/ml). Ginkgo and Green tea extracts were analyzed for comparison, and the results indicated that some extracts of T. polium were equal in activity with Ginkgo or Green tea and some appeared to have greater activity. The obtained results suggest strong antioxidant activity and large contribution of separate analysis for the maximum exploitation of active phenolic compounds from T. polium. Based on this information, plant parts of this plant are natural sources of antioxidant substances of high importance.

The present study investigates phytochemical screening and antibacterial efficacy of various solvent extracts of marine algae such as Sargassum swartzii against some selected human and fish pathogenic bacteria. The brown seaweed Sargassum swartzii was collected from Kudankulam, Tirunelveli district, Tamil Nadu, India. Soxhlet extraction method was used to get the extracts of different solvents like aqueous, ethanol, methanol, acetone and were tested for their qualitative phytochemical screening using common standard tests. Quantitative phytochemical analysis such as total phenolic content and total flavonoid content of S. swartzii and antibacterial activity against human pathogens viz., Escherichia coli, Pseudomonas aeruginosa and Stephylococcus aureus and fish pathogens viz., Aeromonas hydrophila and Vibrio vulnificus bacteria using disc diffusion methods. Preliminary phytochemical screening of sixteen different chemical compounds were carried out. The maximum biochemical compounds were present in the ethanol extract and the minimum compounds were present in aqueous extract. The ethanol extract was significantly higher in phenolic content (15.35 ±2.61 mg of GAE/g) and the methanol extract was significantly higher in flavonoides content (26.92±7.14 mg of QE/g). In human pathogen the highest antibacterial activity was present in Pseudomonas aeruginosa (18.00±0.00mm) and the fish pathogen was significantly higher in Aeromonas hydrophila. The present study showed that the ethanol extract of marine brown seaweed Sargassum swartzii contains bio active constituents with highest antibacterial activity against human and fish pathogen.
