Mycobacterium tuberculosis (Mtb), the etiological agent of tuberculosis (TB), is the leading bacterial cause of infectious disease mortality. The current WHOapproved treatment for TB involves a three- or four-drug regimen comprising isoniazid, rifampin, pyrazinamide, and/or ethambutol for a minimum of 6 months. While these first-line agents remain useful in treating susceptible Mycobacterium tuberculosis strains, the emergence of multidrug resistant tuberculosis demands the development of new drugs [1]. Iron acquisition is an essential process for M. tuberculosis as well as almost all other microorganisms. However, this essential micronutrient is highly sequestered in a mammalian host. In response to iron starvation, Mtb produces small-molecule iron chelators, a pair of related peptidic siderophores known as mycobactin and carboxymycobactins that vary by the appended lipid residue termed siderophores [4, 5, 7, 8]. Because mycobactins are critical for growth and virulence of M. tuberculosis, they have emerged as attractive targets for the development of anti-TB agents [4]. Biosynthesis of mycobactin is initiated by the aryl acid adenylation enzyme MbtA which activates salicylic acid forming an acyladenylate intermediate (Sal- AMP). MbtA is also responsible for loading the acyladenylate intermediate onto the thiolation domain of MbtB-SH – the enzyme taking part in the next step of biosynthesis process [10]. Given the documented importance of many siderophores for virulence and lack of human aryl acid adenylation enzymes homologues, several analogues possessing stable linkers as bioisosteres of the labile acyl phosphate function have been synthesized as the potent enzyme inhibitors [13]. The initial lead compound 5’-O-[N- (salicyl)sulfamoyl]adenosine (Sal-AMS) has emerged as a promising inhibitor of MbtA and was shown to possess promising whole-cell activity toward M. tuberculosis.

Two analytical methods, atomic force microscopy and quartz crystal microbalance, were applied to the study of the reaction kinetics occurring between concanavalin A and carboxypeptidase Y, presenting the specific lectin-carbohydrate recognition. The dissociation rate constants for concanavalin A-carboxypeptidase Y complex obtained using both atomic force microscopy and quartz crystal microbalance were of the same order of magnitude: k$\text{}_{diss}$=0.170± 0.060 s$\text{}^{-1}$ and k$\text{}_{diss}$=0.095±0.002 s$\text{}^{-1}$, respectively. In addition, each method alone aided in determining other parameters characterizing the studied interaction. Quartz crystal microbalance permitted us to estimate the association rate (k$\text{}_{ass}$=(5.6 ±0.1)×10$\text{}^{4}$ M$\text{}^{-1}$ s$\text{}^{-1}$) and the equilibrium (K$\text{}_{a}$=(0.59×0.01)×10$\text{}^{6}$ M$\text{}^{-1}$) constants for the binding process occurring between concanavalin A and mannose residues of carboxypeptidase Y under given experimental conditions. Atomic force microscopy in force spectroscopy mode enabled the determination of the energy barrier position of r=2.29±0.04 Å characterizing the dissociation of concanavalin A- carboxypeptidase Y molecular complex. The presented results show that both atomic force microscopy and quartz crystal microbalance can be used to determine quantitative parameters characterizing the specific molecular interaction. Both methods can be easily combined for complementary and/or alternative studies of a chosen molecular interaction. By preparing the samples in the same manner the direct comparison between the data obtained via atomic force microscopy and quartz crystal microbalance can be made.

Natural nucleotide antibiotics such as Agrocin 84, Dinoguellin, Microcin C and Phosmidosine have a N-acylphosphoramidate linkage at the 5’-hydroxyl of the adenosine derivatives (Fig. 1, 2) [1–3]. They exhibit interesting antifungal, antiemetics and anticancer properties. To synthesize these products, the construction of the N-acylphosphoramidate linkages seems to be a key step. Many groups have described the preparation of such a type of analogues but none of those methods was general. Grandas has for the first time reported the synthesis, of N-acylphosphoramidate peptide-oligonucleotide hybrids via condensation of N-phosphitylated carboxyamides with alcohols in the presence of 1H-tetrazole [9]. Based on this strategy Sekine synthesized aminoacyl adenylate (aa-AMP) analogues which could be useful in the studies on the recognition mechanism of the aminoacylation of tRNA and other biochemical reactions [10]. Since aa-AMPs are extremely unstable under aqueous conditions more stable analogues were required. Aminoacyl-adenylate analogues having an N-acylphosphoramidate linkage (aa-AMPN) could behave as potent, selective asparagine synthetase (AS) inhibitors because of its structural similarity to β-aspartyl-AMP (β AspAMP) which is natural product of AS [17]. Among natural N-acylphosphormiadates, Phosmidosine which connects a nucleoside analogue, 8-oxoadenosine, with an L-proline residue is unique because of its significant antitumor activities and property of stopping cell growth at the G1 phase in the cell cycle (Fig. 2) [2, 13]. The main difficulty during the synthesis of this compound is an extreme instability under weak basic conditions which excludes the use of labile protecting group of basic properties [14]. Stability studies have shown that under basic conditions phosphoryl group of Phosmidosine underwent rapid N–N migration (Scheme 9) [16]. Many modifications have been introduced to improve Phosmidosine properties [16]. Analogues such as demethylated species (Phosmidosine B) have proven to be stable under both basic and acid conditions and are also potential candidates for antitumor drugs [14].

Skład komonomerów jak również mikrostruktura łańcucha polimerowego są ważnymi czynnikami determinującymi sposób degradacji polimeru oraz profil uwalniania leku. Celem badań było określenie wpływu mikrostruktury łańcucha na uwalnianie paklitakselu. Dwa rodzaje PLATMC (70:30) zostały użyte do przygotowania matryc z 3% i 5% zawartością leku oraz matryc bez leku. Materiały zostały zsyntezowane w różnych temperaturach i dlatego różniły się pod względem właściwości fizykochemicznych. Wszystkie matryce badano za pomocą spektroskopii NMR, GPC, DSC, SEM zarówno przed jak i w czasie procesu degradacji. Ilość uwolnionego paklitakselu szacowano przy pomocy HPLC. Otrzymane wyniki wskazują, iż podczas inkubacji w mikrostrukturze łańcucha kopolimerowego nie zaszły znaczące zmiany. Wolniejszy profil uwalniania odnotowano w przypadku PLATMC o wyższym współczynniku randomizacji, zsyntezowanego w wyższej temperaturze. Kopolimer ten wydaje się być atrakcyjnym materiałem do tworzenia systemów kontrolowanego uwalniania paklitakselu.
Comonomers composition as well as the chain microstructure are very important factors determining time of polymer degradation and drug release profile. In this study it was aimed to determine the influence of chain microstructure on paclitaxel release. Two types of PLATMC (70:30) were used to prepare matrices with 3% and 5% of drug and also drug free matrices. They were synthesised at different temperature and thus differed in physicochemical properties. All of the matrices were investigated before and during hydrolytic degradation process by means of NMR spectroscopy, GPC, DSC, SEM. The amount of liberated paclitaxel was assessed with the use of HPLC. The obtained results indicated no significant changes in chain microstructure during incubation period of time. Slower release profile was noticed for PLATMC synthesised at higher temperature and possessed higher randomisation ratio. This type of copolymer seems to be very attractive candidate for controlled paclitaxel delivery systems.

Depth distribution of implanted species and microstructure of oxygen-containing Czochralski grown silicon (Cz-Si) implanted with light ions (such as $H^{+}$) are strongly influenced by hydrostatic pressure applied during the post-implantation treatment. Composition and structure of Si:H prepared by implantation of Cz-Si with $H_{2}^{+}$; fluence D = 1.7 × $10^{17} cm^{-2}$, energy E = 50 keV (projected range of $H_{2}^{+}$, $R_{p}(H)$ = 275 nm), processed at up to 923 K under Ar pressure up to 1.2 GPa for up to 10 h, were investigated by elastic recoil detection Rutherford backscattering methods and the depths distributions of implanted hydrogen and also carbon, oxygen and silicon in the near surface were determined for all samples. The defect structure of Si:H was also investigated by synchrotron diffraction topography at HASYLAB (Germany). High sensitivity to strain associated with small inclusions and dislocation loops was provided by monochromatic (λ = 0.1115 nm) beam topography. High resolution X-ray diffraction was also used.

Description of the modeling system of differential GNSS mode for the operation ITS is considered. Differential method of navigation allows to precise determination of the coordinates and is not very complicated for technical realization. The coordinates of the consumer are calculated with high accuracy because the base station and the user are located in a short distance from each other. Determination of the origin location is on the same satellite and method of local correction is implemented.

This article focuses on the development of architecture and technical solutions for implementation intelligent transport systems (ITS). The presented system will perform the following functions: monitoring the movement of an object collecting and displaying information about the state of the road, warning of the approaching object to obstacles, routing, as well as control of movement and speed of the output parameters of the recommendations of the movement, the development of intelligent decisions about the choice of optimal routes, intellectual analysis of data (IAD) of the style and movement of the driver (the influence of alcohol), IAD on the technical parameters of the vehicle. Thus, it is advisable for ITS to be divided into several subsystems, which are related but are responsible for one or more functional tasks. Consequently, the need for monitoring the movement subsystem, the database on the state of the object database on the location of an object, display subsystem, the subsystem of control of the road, the engine safety (prevention of obstacles), the subsystem control parameters of movement, routing subsystem.
Статья посвящена разработке архитектуры и технических решений для реализации интеллектуальных транспортних систем. Представляемая система будет выполнять следующее функции: мониторинг движения объекта, сбор и отображение информации о состоянии дороги, предупреждение о приближении объекта к препятствиям, маршрутизация, а так же контроль скорости движение и вывод рекомендаций о параметрах движения, разработка интеллектуальных решений о выборе оптимальных маршрутов, интеллектуальном анализе данных (ИАД) о стиле и движении водителя (влиянию алкоголя), ИАД о технических параметрах транспортного средства. Таким образом, целесообразно ITS разделить на несколько подсистем, которые будут взаимосвязаны, но отвечают за одну или несколько функциональных задач. Для реализации этих функций, необходима подсистема мониторинга движения, база данных о состоянии объекта, база данных о местоположении объекта, подсистема отображения информации, подсистема контроля дороги, подсистема безопасности движения (предупреждения о препятствиях), подсистема контроля параметров движения, подсистема маршрутизации.

The magnetocaloric effect may be assessed indirectly by expressing it as the change in magnetic entropy in varying magnetic field, H, as the function of temperature, T. Magnetization, M=f(T,H), may be experimentally acquired from a series of isothermal measurements with variable field, or from a series of constant field measurements with variable temperature. The accuracy of magnetic entropy calculation depends on the number of series in these experiments. The aim of this work is to determine how little data is sufficient to obtain accurate results of magnetic entropy change calculations, on the basis of real, magnetocaloric materials. Pure gadolinium and a Ni-Mn-Cu-Ga Heusler alloy were studied. For both materials, the magnetic entropy change and relative cooling power were calculated from both experiments, with the decreasing number of experimental data. For both materials, the constant field experiment with only 6 field values provided only a 5% error of calculations, as compared to the experiment with 100 field values. The Arrott plots were also drawn for constant field mode with 6 field values, easily indicating the order of transition. Comparison of the calculation results suggests that the constant field mode magnetization measurement may be more accurate and faster than isothermal mode.

It is shown in the present paper that gradually devitrified Co-based non-magnetostrictive metallic glass is an excellent model material to verify Louis Néel's theory of the Rayleigh rule. In the course of the calculations, Néel showed that the parameter p=bH$\text{}_{c}$a (where H$\text{}_{c}$ is the coercivity, a and b are the coefficients of a quadratic polynomial expressing the Rayleigh rule) is expected to range between 0.6 (hard magnets) and 1.6 (soft). However, the experimental values of this parameter, reported in the literature for a number of mono- and poly-crystalline magnets, are much greater than those expected from the theory presented by Néel (in some cases even by two orders of magnitude). The measurements, performed for a series of Co-based metallic glass samples annealed at gradually increasing temperature to produce nanocrystalline structures with differentiated density and size of the crystallites, have shown that the calculated values of the parameter p fall within the range expected from Néel theory.

In this work hyperfine fields of two-phase nanocrystalline Fe72Cu1.5Nb4Si13.5B9 alloys were studied in order to verify the existence of surface effects. To obtain a series of nanocrystalline samples with small grains of different sizes, a special non- -isothermal annealing procedure of an initially amorphous ribbon was applied. In the case of samples with a significant amount of crystallites, a high field (about 27.5 T) component of continuous part of the hyperfine field distribution was found that could be attributed to boundary regions between the grains and rest of the sample. The existence of the surface effects was confirmed in the magnetostrictive experiment.

We identified a species level of the fungal cultures isolated from selected crop seeds using traditional method and BIO-PCR. The use of BIO-PCR did not correspond completely to the morphological analyses. Both methods showed increased infection with F. poae in winter wheat seed sample originated from north Poland. Fungal culture No 40 (isolated from faba bean and identified with traditional method as mixed culture with F. culmorum and F. graminearum) did not produce expected product after PCR reaction with species specific primers OPT18F₄₇₀ OPT18R₄₇₀ However, the use of additional primers Fc01F, Fc01R allowed for reliable identification of F. culmorum in the culture.
W pracy określono przynależność gatunkową grzybów z rodzaju Fusarium wcześniej wyizolowanych z nasion wybranych gatunków roślin uprawnych metodą tradycyjną i BIO-PCR. Zastosowanie metody BIO-PCR było podyktowane faktem, że nie wszystkie kultury zostały wstępnie poprawnie sklasyfikowane. Z przeprowadzonych analiz identyfikacji metodą tradycyjną i molekularną wykazano stosunkowo wysokie porażenie ziarna pszenicy przez F. poae. W badaniach wykazano, że kultura nr 40 (wyizolowana z nasion bobiku i oznaczona metodą tradycyjną jako mieszanka F. culmorum i F. graminearum) nie dawała wyniku pozytywnego w reakcji PCR z użyciem primerów OPT18F₄₇₀ OPT18R₄₇₀ specyficznych dla F. culmorum. Natomiast zastosowanie dodatkowej pary primerów Fc01F, Fc01R pozwoliło na wiarygodne oznaczenie F. culmorum.

Opis:
Remarkable thermomechanical and electrical properties of silicon carbide (SiC) make this material very attractive for high-temperature, high-power, and high-frequency applications. Because of very low values of diffusion coefficient of most impurities in SiC, ion implantation is the best method to selectively introduce dopants over well-defined depths in SiC. Aluminium is commonly used for creating p-type regions in SiC. However, post-implantation radiation damage, which strongly deteriorates required electric properties of the implanted layers, is difficult to anneal even at high temperatures because of remaining residual damage. Therefore implantation at elevated target temperatures (hot implantation) is nowadays an accepted method to decrease the level of the residual radiation damage by avoiding ion beam-induced amorphization. The main objective of this study is to compare the results of the Rutherford backscattering spectroscopy with channeling and micro-Raman spectroscopy investigations of room temperature and 500°C $Al^{+}$ ion implantation-induced damage in 6H-SiC and its removal by high temperature (up to 1600°C) thermal annealing.
