Opis:
Incomplete oxygen reduction gives rise to reactive oxygen species (ROS). For a long time they have been considered unwelcome companions of aerobic metabolism. Organisms using oxygen developed several systems of ROS scavenging with enzymatic and non enzymatic antioxidants, which allow them control the cellular level of oxygen derived from free radicals. It is well established nowadays that ROS are not necessarily negative byproducts, but they also play an important role in cellular mechanisms. They are involved in many regular cellular processes in all aerobic organisms. When the antioxidant system is overcome and the balance between ROS production and scavenging is disrupted, oxidative stress occurs. It has been reported that oxidative stress may be linked to some human diseases and is also involved in biotic and abiotic stress response in plants.

Opis:
The objective of this study was to assess the dynamics of oxidative damage to cellular macromolecules such as proteins, lipids and DNA under conditions of oxidative stress triggering early stages of estrogen-dependent carcinogenesis. A rodent model of carcinogenesis was used. Syrian hamsters were sacrificed after 1, 3, 5 h and one month from the initial implantation of estradiol. Matching control groups were used. Kidneys as target organs for estradiol-mediated oxidative stress were excised and homogenized for biochemical assays. Subcellular fractions were isolated. Carbonyl groups (as a marker of protein oxidation) and lipid hydroxyperoxides were assessed. DNA was isolated and 8-oxodGuo was assessed. Electron paramagnetic resonance spectroscopy was used to confirm the results for lipid peroxidation. Exposition to estradiol in the rodent model leads to damage of macromolecules of the cell, including proteins and DNA, but not lipids. Proteins appear to be the primary target of the damage but are closely followed by DNA. It has previously been speculated that protein peroxides can increase DNA modifications. This time sequence was observed in our study. Nevertheless, the direct relation between protein and DNA damage still remains unsolved.

Background: Ovarian cancer is the fifth most common form of cancer in the world and is often asymptomatic in its early stages. Development of ovarian cancer-specific biomarkers for the early detection of disease could improve the current dismal survival rate. Evaluation of serum carbohydrate antigen 125 (CA125), alkaline phosphatase (ALP) and oxidative stress in ovarian carcinoma patients may improve the prognosis of the disease through earlier detection. Aim of the study: The aim of this study was to find the relative risk of ovarian cancer in patients screened for CA125, ALP, Nitric oxide (NO) and Malondialdehyde (MDA) as a marker for lipid peroxidation. Material and methods: 451 subjects with ovarian cancer were screened for serum CA125 levels using a chemiluminescence analyser, out of which 164 showed values above 21 U/ml. 80 subjects with higher values were further analysed for MDA and NO using spectrophotometry and AL P by fully automated chemistry analyser. Results: The selected 80 subjects with CA125 values above 74 U/ml had increased ALP, NO and MDA, also showing positive correlation amongst these parameters. Conclusions: Benefits of CA125 screening vary with age group according to blood CA125 levels. Enzyme ALP levels are elevated with higher values of CA125. MDA and NO indicate oxidative stress and increase as the ovarian marker values increase. Positive correlation amongst the parameters indicates a significant increase in oxidative stress in ovarian cancer. For women with various CA125 levels in different age groups, screening and treatment depends upon individual decision and clinical examination.

Opis:
Molecular oxygen (O2), constituting the basis of life on Earth, is classified as a substance with oxidizing properties. Reacting with organic compounds, it leads to their oxidation and at the same time participates in reduction processes. In aerobic organisms, over 90% of oxygen undergoes a total four-electron reduction to produce water molecules (O2 + 4 H+ + 4e- → 2 H2O). The remaining 10% of oxygen, however, is not fully reduced, which results in the production of molecules referred to as reactive oxygen species (ROS). In high concentrations ROS can interact with cellular components (DNA, proteins and lipids), leading to the oxidation of these macromolecules. The resulting oxidation products interfere with the proper functioning of the body by influencing gene expression, intercellular signaling and apoptosis. These changes have been observed in numerous pathological conditions, such as neurodegenerative, cardiovascular, metabolic, autoimmune diseases, and cancer. However, in the context of evolution, living organisms developed specialized repair mechanisms to prevent cellular accumulation of the products of DNA, protein and lipid oxidation, including enzymatic mechanisms (e.g. nucleases, proteases, phospholipases) or removal of damaged DNA, proteins and lipids by apoptosis or autophagy. This article briefly discusses the mechanisms of oxidative modification of cell components and the main repair systems responsible for the removal of lesions in cells by oxidative damage.

The purpose of this study was to examine the effects of oxidative stress caused by hydroperoxide (H2O2) in the presence of iron ions (Fe2+) on mitochondria of the amoeba Acanthamoeba castellanii. We used isolated mitochondria of A. castellanii and exposed them to four levels of H2O2 concentration: 0.5, 5, 15, and 25 mM. We measured basic energetics of mitochondria: oxygen consumption in phosphorylation state (state 3) and resting state (state 4), respiratory coefficient rates (RC), ADP/O ratios, membrane potential (ΔΨm), ability to accumulate Ca2+ , and cytochrome c release. Our results show that the increasing concentrations of H2O2 stimulates respiration in states 3 and 4. The highest concentration of H2O2 caused a 3-fold increase in respiration in state 3 compared to the control. Respiratory coefficients and ADP/O ratios decreased with increasing stress conditions. Membrane potential significantly collapsed with increasing hydroperoxide concentration. The ability to accumulate Ca2+ also decreased with the increasing stress treatment. The lowest stress treatment (0.5 mM H2O2) significantly decreased oxygen consumption in state 3 and 4, RC, and membrane potential. The ADP/O ratio decreased significantly under 5 mM H2O2 treatment, while Ca2+ accumulation rate decreased significantly at 15 mM H2O2. We also observed cytochrome c release under increasing stress conditions. However, this release was not linear. These results indicate that as low as 0.5 mM H2O2 with Fe2+ damage the basic energetics of mitochondria of the unicellular eukaryotic organism Acanthamoeba castellanii.

Background: Primary hyperparathyroidism (PHPT) is one of the most common endocrine disorders and defined as excessive secretion of parathormone. PHPT is a risk factor of several cardiovascular diseases, which could be caused by alterations in oxidant-antioxidant balance. Materials and methods: Blood serum collected from 52 consecutive patients with PHPT treated surgically constituted our study material, whereas 36 healthy volunteers were our control group. Oxidative stress was evaluated in both patients and control subjects by assessment of malondialdehyde (MDA) and lipid hydroperoxides (LHP). Antioxidants were evaluated by the measurement of superoxide dismutase (SOD), ceruloplasmin (CER), catalase (CAT), sulfhydryl (SH) groups, glutathione (GSH), glutathione peroxidase (GSH-Px), glutathione transferase activity (GST) and glutathione reductase (GR). Moreover, total antioxidant capacity (TAC) and total oxidative status (TOS) were measured and oxidative stress index (OSI) was calculated. Results: OSI was increased in patients with PHPT when compared to normal controls, whereas TAC was lower in PHPT. The levels of CER, MnSOD, GR, SH groups and MDA were significantly decreased in PHPT. The levels of serum LHP, catalase and SOD were significantly higher in patients with PHPT than in healthy patients. The erythrocyte CAT activity and GST were significantly increased in patients after parathyroidectomy. The erythrocyte GR and GPx were up-regulated postoperatively, whereas SOD activity decreased. Conclusions: In PHPT there are several alterations in the balance between the production of reactive oxygen species and antioxidant defense system.

Etanol ma istotny wpływ na proces wytwarzania reaktywnych form tlenu, a tym samym na generowanie stresu oksydacyjnego. Dokonano charakterystyki RFT oraz opisano mechanizm ich powstawania. Oksydacyjnym uszkodzeniom ulegają białka, DNA i lipidy. Istotna rolę odgrywają reaktywne formy tlenu w procesach chorobotwórczych u osób przewlekle spożywających alkohol. Wśród chorób związanych z oddziaływaniem RFT i nadużywaniem alkoholu wyróżnia się: choroby wątroby, miażdżycę, nadciśnienie tętnicze, rozwój nowotworów, spadek odporności, zaburzenia funkcji seksualnych, a także przyspieszenie procesu starzenia się skóry. Organizm ochraniają antyoksydanty wewnątrzkomórkowe i zewnątrzkomórkowe.
Ethanol significantly influences formation of reactive oxygen species (ROS) in cells, and thus generation therein of oxidative stress. At present, ROS and mechanisms of their formation and action are well characterized. Under the stress condition, proteins, DNA and lipids undergo oxidative damages. In persons consuming alcohol protractedly, ROS play an important role in etiology of pathogenic processes, in particular such as liver diseases, arteriosclerosis, arterial hypertension, development of tumors, decrease in immunity, disorder of sexual functions, and acceleration of skin aging. Intracellular and outside antioxidants help to protect organism from the oxidative stress.

Oxygen is a biogenic element which determines the course of basic biochemical processes to ensure the survival of the cell. Unfortunately, in particular circumstances, the highly reactive oxygen species (ROS) may cause a toxic effect on the body. Small amounts of ROS are essential for the proper functioning of cells, however, the excessive production of them, which transcend the effectiveness of antioxidant systems – defined as oxidative stress – promotes structural and functional disorders of cells and tissues. Nowadays, the significant participation of oxidative stress is increasingly more frequently indicated in the course and/or etiology of chronic inflammatory diseases, which include atopic diseases such as bronchial asthma, atopic dermatitis, or allergic rhinitis.
Tlen jako pierwiastek biogenny warunkuje przebieg podstawowych procesów biochemicznych gwarantujących przeżycie komórki. Niestety, w szczególnych warunkach wysoce reaktywne formy tlenu (RFT) mogą wywierać toksyczny efekt na organizm. W niewielkich ilościach RFT są niezbędne do prawidłowego funkcjonowania komórki, jednak ich nasilone wytwarzanie, przekraczające skuteczność działania systemów antyoksydacyjnych, określane mianem stresu oksydacyjnego, sprzyja zaburzeniom struktury i funkcji komórek oraz tkanek. Współcześnie coraz częściej wskazuje się na istotny udział stresu oksydacyjnego w przebiegu i/lub etiologii przewlekłych chorób zapalnych, do których zalicza się m.in. choroby atopowe, takie jak astma oskrzelowa, atopowe zapalenie skóry czy alergiczny nieżyt nosa.

Introduction and aim. Oxidative stress is one of the complications that accompany defects in thyroid hormone levels. This study was designed to evaluate oxidative stress markers in patients with hypothyroidism. Material and methods. This case control study was comprised of forty-two hypothyroid patients aged 36–46 years and forty age matched (35–43 years) healthy control participants randomly selected from the Endocrine Clinic of Al-Yarmook Hospital in Iraq. Blood levels of thyroid hormones malondialdehyde, glutathione, and paraoxonase-1 were assessed. Body mass index was calculated for each patient and control participant. Results. Regarding the data of oxidative stress markers in hypothyroid patients compared to controls, a significant elevation was reported in blood levels of malondialdehyde and a significant reduction was found in blood levels of glutathione (p=0.031). On the other hand, the blood levels of paraoxonase-1 were significantly different in hypothyroid patients compared with the control. Conclusion. Elevated blood levels of malondialdehyde and declined blood levels of glutathione in hypothyroid patients are a signal of oxidative stress and consequently increase the risk of cardiovascular complications.

Isoprostanes are a recently discovered group of prostaglandin isomers. Results of previous studies suggest that they can be used as oxidative stress markers, because in a number of cardiovascular, pulmonary and neurological diseases their levels in biological samples considerably increase. It has been found that people suffering from diabetes, obesity, homozygous familial hypercholesterolemia, moderate hypercholesterolemia, and smokers have higher levels of isoprostanes in urine. The same refers to patients with asthma, Alzheimer disease and Down syndrome. This paper reviews the results of relevant studies.

Oxidative stress represents a situation where there is an imbalance between the reactive oxygen species (ROS) and the availability and the activity of antioxidants. This balance is disturbed by increased generation of free radicals or decreased antioxidant activity. It is very important to develop methods and find appropriate biomarkers that may be used to assess oxidative stress in vivo. It is significant because appropriate measurement of such stress is necessary in identifying its role in lifestyle-related diseases. Previously used markers of oxidative stress, such as thiobarbituric acid reactive substances (TBARS) or malondialdehyde (MDA), are progressively being supplemented by new ones, such as isoprostanes (IsoPs) and their metabolites or allantoin. This paper is focusing on the presentation of new ones, promising markers of oxidative stress (IsoPs, their metabolites and allantoin), taking into account the advantage of those markers over markers used previously. Stres oksydacyjny jest stanem braku równowagi między działaniem reaktywnych form tlenu (RFT) a działaniem antyoksydantów. Równowaga ta może być zakłócona w wyniku zwiększonego działania wolnych rodników lub spadku aktywności antyoksydacyjnej. Zaburzenia te mogą występować zarówno na poziomie komórkowym, jak i całego organizmu. Ponieważ stres oksydacyjny może być podłożem wielu zespołów chorobowych, niezwykle istotne jest znalezienie odpowiednich markerów, które mogą być wykorzystane do oceny jego poziomu in vivo. Stosowane od wielu lat markery – ocenę stężenia aldehyd dimalonowy (MDA) i substancji reagujących z kwasem tiobarbiturowym (thiobarbituric acid reactive substances – TBARS) – stopniowo uzupełnia się nowymi, takimi jak alantoina czy izoprostany (IzoP) wraz z ich metabolitami (IzoP-M). W niniejszej pracy skupiono się na zaprezentowaniu nowych, obiecujących markerów stresu oksydacyjnego (alantoina, IzoP, IzoP-M), ukazując korzyści wynikające z ich stosowania i prognozując dalsze kierunki badań nad ich zastosowaniem. High-intensity exercise increases reactive oxygen species formation, which in excess may cause oxidative stress. We assessed nutritional status and exercise-induced oxidative stress in 20 adolescent male soccer players (age: 15-17). Participants were divided into two teams for a 60-minute friendly match and evaluated immediately before (Pre-match), 30 minutes after (Post-match I) and 24 hours after (Post-match II) the game. All players recorded a 3-day dietary intake. Biochemical tests were performed for lipid profile, muscle damage (creatinine and creatinine kinase [CK]) and oxidative stress (thiobarbituric acid-reactive substances [TBARS], protein carbonyls [PC], reduced glutathione [GSH], and vitamins E, C, and A). CK and creatinine were significantly elevated at Post-match I (p < 0.01), returning to baseline at Post-match II. Vitamins E, C and A were significantly elevated at Post-match I (p < 0.01), but only vitamins E and A remained high at Post-match II. TBARS showed no significant changes. GSH showed a significant decrease (p < 0.01) and PC showed a slight but significant increase (p < 0.01) at Post-match II. The recruitment of non-enzymatic antioxidants prevented lipid peroxidation, but dietary and especially endogenous defence responses were insufficient to prevent protein oxidation. Proper nutrition is essential to improve the activity of the antioxidant defence system, preventing exercise-induced oxidative stress.

Stres oksydacyjny definiowany jest jako zachwianie równowagi pomiędzy powstawaniem (nadprodukcją) wolnych rodników, a zdolnościami indywidualnych mechanizmów obronnych. Produkcja wolnych rodników jest stałym elementem metabolizmu tlenowego komórki. Jest powszechnie wiadomo, że stres oksydacyjny wywołuje wiele stanów patologicznych i chorób. Stres oksydacyjny jest przyczyną uszkodzeń wielu ważnych makromolekuł, do których zaliczamy: DNA, białka i lipidy. Wzrost poziomu wolnych rodników wpływa też na transformację nowotworową komórki. Procesowi formowania i rozwojowi nowotworu towarzyszy zachwianie równowagi redox w komórce, nadprodukcja reaktywnych form tlenu i azotu. Obecnie rak piersi jest najczęstszym nowotworem złośliwym u kobiet w Polsce i na świecie. Biologia tego nowotworu i jego kliniczny obraz jest bardzo heterogenny. Stres oksydacyjny i zmiany oksydacyjno-nitracyjne są często obserwowane u chorych z rakiem piersi, ale nadal mechanizmy relacji między stresem oksydacyjnym czy nitracyjnym a rakiem piersi są niejasne. Stres oksydacyjny i nitracyjny obserwowany u pacjentów chorych na raka piersi może również indukować zmiany strukturalne i zmiany funkcji różnych elementów układu hemostazy. Organizm człowieka wyposażony jest w kilka sposobów ochrony przed atakiem oksydacyjnym. Mechanizmy obrony obejmują różne enzymy antyoksydacyjne, do których zaliczamy: peroksydazę glutationową, katalazę, dysmutazę ponadtlenkową i inne substancje, takie jak niskocząsteczkowe tiole: glutation, cysteinę, witaminę E, C i A, które eliminują wolne rodniki. Jest to bardzo istotne szczególnie dla chorych z rakiem piersi podczas chemioterapii wielolekowej, czy przed- lub pooperacyjnej radioterapii.
Oxidative stress may be defined as an imbalance between reactive oxygen species and the individual antioxidant defense system. Reactive oxygen species formation is a constant element of a cell's oxygen metabolism. It is now known that oxidative stress is involved in most of pathological states and diseases. Oxidative stress causes damage to important macromolecules, such as DNA, proteins, and lipids. Growing evidence indicates the participation of free radicals in the cancerous transformation of cells. The process of cancer formation and development is associated with loss of redox balance in the cell and with overproduction of reactive oxygen and nitrogen species. Actually breast cancer is the most common malignant neoplasm in women both in Poland and worldwide. It is biologically and clinically very heterogenous disease. Oxidative stress and oxidative-nitrative changes in the cells are frequently observed in breast cancer patients, but until now mechanisms involved in the relationship between oxidative stress and breast cancer are still unclear. Oxidative stress species may also induce changes to the structure and function in hemostatic elements. Human organisms have developed several ways to protect themselves from oxidant attacks. The defense mechanisms include a variety of antioxidant enzymes like glutathione peroxidase, catalase and superoxide dismutase and other substances such as low-molecular-weight thiols, such as glutathione, cysteine and vitamins E, C and A, which provide to eliminate free radicals are very important for the development of breast cancer and during multichemotherapy with different anti-cancer drugs or pre- and postoperative radiation therapy.

Biofertilizers are the special formulation of specific beneficial microorganisms that promote the growth of plant crops by converting the unavailable form of nutrients into available form. Here, the effect of heavy metal stress on antioxidant enzymes were studied in Clitoria ternatea L. leaves. Clitoria ternatea L. plant was grown for 30 days and the heavy metal mercuric chloride was sprayed after 10 days from the date of planting. Effect of mercuric chloride was observed in treated plants. The selected plant Clitoria ternatea L. was grown under mercuric chloride treatment in a specified concentration 1 μg/10 ml. The control plant maintained without the treatment of mercuric chloride. Antioxidant effect of mercuric chloride was measured under controlled and treated conditions. The selected plant Clitoria ternatea L. was grown under mercuric chloride in treatment. Further it increases H2O2 content and the antioxidant enzyme activities such as superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) were observed in mercuric chloride treated plants when compared with control. Here mercuric chloride was accumulated more in matured leaves. The results are discussed with the literature.

Opis:
Specificity of targeting of the oxidative stress towards lipid and protein fractions in a model of estrogen-induced Syrian hamster nephrocarcinogenesis was evaluated. The amount of proteins modified by oxidative stress was significantly elevated as early as one month after the initial implantation of estradiol to the experimental versus the control group, while the stress did not affect lipids. Subcellular localization of the oxidative stress target was determined by the analysis of protein oxidation in subcellular fractions of kidney cells. The endoplasmic reticulum membranes were the fraction most affected by the oxidative stress.

The aim of the study was to analyze the influence of selenium (Se) on the course of oxidative stress in trained athletes, on the basis of own former published studies and the reference of literature. In a number of references it was shown that the application of Se led to the diminishing of oxidative stress during the physical exercises in athletes. The application of Se to athletes − led to the increase of peroxidase glutathione the main selen − dependent enzyme. The fact of increased concentration of malondialdehyde (MDA) in blood serum, the main metabolite of lipid peroxidation can indirectly evidence about the intensity of physical endurance. This increase is significantly smaller in individuals taking exogenous antioxidative diet supplement containing Se. Moreover there is the evidence that oxidative muscles damage during physical effort could be diminished by compounds with antioxidative properties. Taking into consideration, the obtained results by many of authors empower to the conclusion that application of Se diminishes the degree of peroxidation of lipids in trained athletes.

Homeostasis of an organism and its proper functioning are determined by the oxidative-antioxidant balance. Its disorder is most often associated with overproduction of reactive oxygen species that cause oxidative stress. This, in turn, lies at the heart of the development and course of many diseases, especially those that are classified as civilization diseases. Environmental pollution, chronic stress, low levels of physical activity, and improper nutrition are among the determinants of the production of an increased amount of free radicals. Their destructive action consists in the oxidation of cell components, which in turn leads to damage and disturbance of the physiological functions of the cell. Free radicals change the structure and modify the functions of proteins. In addition, they destroy DNA structures. The present review article outlines the problem of the participation of oxidative stress in the pathogenesis of selected civilization diseases. The concept and role of free radicals and reactive oxygen species in the human body are presented. Next, their participation in the development and course of individual diseases is discussed.
Homeostaza organizmu i jej odpowiednie funkcjonowanie determinowana jest przez równowagę oksydacyjno-antyoksydacyjną. Jej zaburzenie związane jest najczęściej z nadprodukcją reaktywnych form tlenu, które powodują stres oksydacyjny. Ten natomiast leży u podstaw rozwoju i przebiegu wielu chorób, a zwłaszcza tych, które zaliczane są do schorzeń cywilizacyjnych. Zanieczyszczenie środowiska, przewlekły stres, niski poziom aktywności fizycznej oraz nieprawidłowe odżywianie są jednymi z determinantów wytwarzania zwiększonej ilości wolnych rodników. Ich destrukcyjne działanie polega na utlenianiu składników komórki, co w konsekwencji prowadzi do uszkodzeń i zaburzeń jej funkcji fizjologicznych. Wolne rodniki zmieniają strukturę oraz modyfikują funkcje białek. Ponadto niszczą struktury DNA. Prezentowany artykuł poglądowy stanowi zarys problematyki udziału stresu oksydacyjnego w patogenezie wybranych chorób cywilizacyjnych. Scharakteryzowano pojęcie oraz rolę wolnych rodników i reaktywnych form tlenu w organizmie człowieka. Następnie omówiono ich udział w rozwoju i przebiegu poszczególnych schorzeń.

Purpose. In adult liver transplant recipients, coronary artery disease and congestive heart failure are significant cause of morbidity and mortality. This may be attributed to the long-term immunosuppressive treatment, mostly with calcineurin inhibitors and steroids, which in long-term may be associated with hyperlipidemia, oxidative stress and cardiovascular complications. Since such data for children is sparse, the aim of this study was to assess the lipid and oxidative stress markers after pediatric liver transplantation (LTx). Method. We performed prospective analysis of 74 children, at the median age of 7.9 (2.8-11.6) years, 3.2 (1.2-4.3) years after LTx. We assessed the BMI Z-scores, cholesterol fractions (LDLc, HDLc, VLDLc), triglicerides, apolipoproteins (ApoAI, ApoB, ApoE), LCAT, insulin resistance by HOMA-IR and markers of oxidative stress and atherosclerosis: glutathione (GSH), glutathione peroxidase (GPx), asymmetrical dimethyl arginine (ADMA) and oxidized low-density lipoprotein (oxyLDL). At baseline, the results were compared with a healthy age-and-sex matched control group. After 3.1±0.3 year follow-up we repeated all investigations and compared them with the baseline results. RESULTS. At the baseline, we investigated 74 patients 3.2 (1.2-4.3) years after LTx, at the median age of 7.9 (2.8-11.6) years. The prevalence of overweight or obesity (BMI >85th percentile) was 23% and was more common in girls (24% vs 20%). Fourteen patients had TCH >200 mg%, 9 patients had LDLc >130 mg% and TG were at normal levels in all patients. Compared to the controls, there were no significant differences in lipid profiles but we found decreased GSH (p<0.001) and GPx (p<0.001) which play role as an antioxidant defense. OS markers were higher in the study group: ADMA (p<0.001), and oxyLDL (p<0.0001). Insulin resistance by HOMA-IR was increased in the study group (p=0.0002) but fasting glucose remained within normal ranges in all patients. After 3.1-year follow-up, the BMI >95th and >85Th percentile was present in 8% and 14% respectively. ADMA and oxyLDL decreased, whilst GSH and GPx increased when compared to the baseline. There was also significant decrease in apoB and Lp(a). Conclusion. Children after LTx had normal lipid profiles when compared to controls, however there is a tendency for hypercholesterolemia and obesity, which may play a role in cardiovascular complications in the future. Some markers of oxidative stress were increased after LTx, however further investigations are required to establish its clinical significance.

Introduction and aim. Thyroid hormone abnormalities have been associated with oxidative changes in human beings. The aim of the study was to evaluate the oxidative stress marker and antioxidants status in individuals with thyroid hormone dysfunction in Ekiti State. Material and methods. A total of eighty samples were recruited in this study comprising forty subjects with thyroid hormones dysfunction and forty apparently healthy controls. Malondialdehyde (MDA), reduced glutathione (GSH) and catalase were determined spectrophotometerically. Results. MDA was non-significantly higher (p>0.05) in subjects (4.33±0.84 nmol/mL) compared with control (4.12±0.63 nmol/mL), catalase was non-significantly higher (p>0.05) in subjects (199.36±20.21 µm/mL) compared with control (181.55±16.61 µm/mL), while GSH was significantly lower (p<0.05) in subjects (79.31±10.12 µmol/mL) compared with control (127.21±7.29 µmol/mL). Conclusion. It can be concluded that the increase in the reactive oxygen species accompanied with impairment of the antioxidant system occurs in patients with thyroid hormone dysfunction. Hypothyroidism and hyperthyroidism induces disequilibrium of the oxidative/anti-oxidative balance that can lead to subsequent development of inflammation and associated diseases.

Increased oxygen concentration (hyperoxia) induces oxidative damage of tissues and organs. Oxygen toxicity in hyperoxia is controlled by factors such as sex, age, tissue, strain and hormones. In most species females show lower incidence of some age-related pathologies linked with oxidative stress, which has been attributed to a beneficial effect of ovarian hormones. In this study we found that hyperoxia induced hepatic oxidative damage exclusively in male CBA/H mice, followed by their decreased survival. Histopathological examination revealed that the observed differences in survival were not the consequence of acute lung injury induced by hyperoxia. Next, we observed that an increased Sirt1 protein level in hyperoxia-exposed female CBA/H mice correlated with their lower PPAR-γ and higher eNOS and Sod2 protein levels. In males, higher PPAR-γ and lower Sod2 protein levels were associated with unchanged Sirt1 expression. Although these results are of a correlative nature only, they clearly show that females show better survival, increased resistance to hyperoxia and have generally more efficient defense systems, which suggests that their headstart in resistance to hyperoxia could be a consequence of the beneficial effect of ovarian hormones.

Reactive oxygen species are constantly generated in aerobic processes in the organism. At the beginning their appearance in tissues were considered to be dangerous, leading to numerous diseases, mutagenesis, degenerative processes and aging. With time their physiological role was discovered as factors regulating numerous internal processes. Oxidative stress can appear as a consequence of influence of various external and internal factors acting especially as stressors. The factors of diving environment are usually stressors. Physical work performance leads also to enhancement of reactive oxygen generation. As a response there is synthesized several stress proteins, which are involved in homeostasis maintenance, cell survival promotion and in cytoprotection. If this protective role is insufficient there appear a cell destruction and cell death due to apoptosis or necrosis. These two processes are close related. The appearance of oxidative stress and apoptosis does not always result in cell death. Less intensive they act in remodeling of the cell that has an adaptative character. This process is under supervision of stress proteins.
Reaktywne formy tlenu są stale wytwarzane w organizmie w procesach przemian o charakterze tlenowym. Początkowo ich obecność w tkankach była traktowana jako niekorzystne zjawisko, łączone z różnymi schorzeniami, mutagenezą, procesami degeneracyjnymi i biologicznym starzeniem się organizmu. Z czasem poznano jednak ich fizjologiczne znaczenie w zakresie regulacji różnych procesów wewnątrzustrojowych. Stres oksydacyjny może powstać w wyniku wpływu różnych czynników zewnątrz- i wewnątrzpochodnych, zwłaszcza mających charakter stresora. Czynniki występujące w nurkowaniu ten charakter zazwyczaj mają. Obok nich, wykonywanie pracy fizycznej prowadzi także do zwiększenia ilości powstających reaktywnych form tlenu. W odpowiedzi na stres organizm wytwarza szereg białek stresowych, które regulują homeostazę wewnątrzkomórkową, ułatwiają przeżywanie komórki i odgrywają zasadniczą rolę w ochronie komórki. W razie niedostatecznej ochrony powstaję uszkodzenie komórki i jej śmierć poprzez apoptozę lub nekrozę. Oba procesy są ściśle ze sobą związane. Zmiany wywołane stresem oksydacyjnym i pojawienie się apoptozy nie zawsze prowadzą do śmierci komórki. W mniej intensywnym wymiarze służą przebudowie komórki, mającej znaczenie adaptacyjne. Proces ten jest pod nadzorem białek stresowych.

Glaucoma has long been considered exclusively a disease of the eye. Today it is known that glaucoma is an occular manifestation of systemic diseases. The pathomechanisms of glaucoma seem to be similar to those observed in other neurodegenerative disorders such as Alzheimer’s disease where nerve cell death, oxidative damage and inflammatory response are observed. The one known and proved glaucoma risk factor – elevated intraocular pressure (IOP), is neither necessary nor sufficient for the development and progression of glaucoma. What is more, IOP normalization does not always prevent disease progression. The data on oxidative stress in glaucomatous disturbances seem not only to conflict other observations but rather complement the mechanical, vascular, genetic and immunologic theories of glaucoma pathogenesis. The aim of the work is a summary of the latest research on the role of oxidative stress in glaucoma.
Do niedawna jaskra była uważana za schorzenie wyłącznie okulistyczne. Dzisiaj wiadomo, że jest ona oczną manifestacją ogólnoustrojowych zaburzeń. Patomechanizmy występujące w jaskrze wydają się podobne do obserwowanych w pozostałych chorobach neurodegeneracyjnych, takich jak np. choroba Alzheimera, gdzie obserwuje się śmierć komórek nerwowych, uszkodzenia związane z szokiem tlenowym, azotowym i odpowiedzią zapalną. Jedyny znany i potwierdzony czynnik ryzyka jaskry – podwyższone ciśnienie wewnątrzgałkowe (CWG) nie jest ani wystarczający, ani konieczny do rozwoju i progresji jaskry. Normalizacja CWG często nie zapobiega postępowi choroby. Obserwacje związku stresu oksydacyjnego z jaskra zdają się nie tylko nie zaprzeczać innym teoriom rozwoju jaskry (mechanicznym, naczyniopochodnym, genetycznym i immunologicznym), ale je uzupełniać. Celem pracy jest zestawienie najnowszych wyników badań nad rolą stresu oksydacyjnego w patogenezie jaskry.

In the recent years there has been considerable interest in mountain sports, including mountaineering, owing to the general availability of climbing clothing and equipment as well trainings and professional literature. This raised a new question for the environmental and mountain medicine: Is mountaineering harmful to health? Potential hazards include the conditions existing in the alpine environment, i.e. lower atmospheric pressure leading to the development of hypobaric hypoxia, extreme physical effort, increased UV radiation, lack of access to fresh food, and mental stress. A reasonable measure of harmfulness of these factors is to determine the increase in the level of oxidative stress. Alpine environment can stimulate the antioxidant enzyme system but under specific circumstances it may exceed its capabilities with simultaneous consumption of low-molecular antioxidants resulting in increased generation of reactive oxygen species (ROS). This situation is referred to as oxidative stress. Rapid and uncontrolled proliferation of reactive oxygen species leads to a number of adverse changes, resulting in the above-average damage to the lipid structures of cell membranes (peroxidation), proteins (denaturation), and nucleic acids. Such situation within the human body cannot take place without resultant systemic consequences. This explains the malaise of people returning from high altitude and a marked decrease in their physical fitness. In addition, a theory is put forward that the increase in the level of oxidative stress is one of the factors responsible for the onset of acute mountain sickness (AMS). However, such statement requires further investigation because the currently available literature is inconclusive. This article presents the causes and effects of development of oxidative stress in the high mountains.
Znaczne zainteresowanie w ostatnich latach sportami górskimi, w tym alpinizmem, przez popularyzacje tych sportów, ogólną dostępność szkoleń i fachowej literatury postawiła nowe pytania przed medycyną środowiskową i górską – czy alpinizm jest szkodliwy dla zdrowia. Z uwagi na warunki jakie panują w środowisku wysokogórskim tj. obniżone ciśnienie atmosferyczne prowadzące do rozwoju hipoksji hipobarycznej, ekstremalnie ciężki wysiłek fizyczny, wzmożone promieniowanie UV, brak dostępu do świeżej żywności oraz stres psychiczny. Zasadną miarą szkodliwości tych warunków jest określenie wzrostu poziomu stresu oksydacyjnego. Środowisko wysokogórskie może oddziaływać bodźcowo na enzymatyczny układ antyoksydacyjny lecz w szczególnych sytuacjach może przewyższać jego możliwości przy jednoczesnym zużyciu antyoksydantów niskocząsteczkowych czego konsekwencją jest wzmożona generacja reaktywnych form tlenu (RFT). Ta sytuacja jest określana mianem stresu oksydacyjnego. Gwałtowne i niekontrolowane namnażanie reaktywnych form tlenu prowadzi do wielu niekorzystnych przemian prowadzących do ponad przeciętnej ilości uszkodzeń lipidowych struktur błony komórkowej (peroksydacji), białek (denaturacja) oraz kwasów nukleinowych. Sytuacja taka w obrębie organizmu ludzkiego nie może wystąpić bez następczych konsekwencji ogólno ustrojowych. Tym właśnie jest tłumaczone złe samopoczucie po powrocie z wysokości oraz wyraźne obniżenie wydolności fizycznej. Dodatkowo jest wysuwana teoria jako by to wzrost poziomu stresu oksydacyjnego był jednym z czynników odpowiedzialnych za rozwój ostrej choroby wysokogórskiej (ang. acute moutain sickness – AMS). Stwierdzenie to jednak wymaga dalszych badań ponieważ aktualnie dostępne piśmiennictwo na ten temat nie jest jednoznaczne, a artykuł ten dotyczy przyczyn rozwoju stresu oksydacyjnego w górach wysokich oraz jego skutków.

Acrylamide (ACR) is a foodborne toxic agent, formed in food when processed at high temperature. This study aimed at evaluating the biochemical changes induced by ACR and the effect of Quercetin as a treatment against ACR induced cardiotoxicity in rats. Wistar rats of either sex (n=6) were divided into four groups as follows: normal control, an Acrylamide control group, Quercetin groups (25 and 50 mg/kg). Diagnostics characteristics were assessed daily, at the end of the study (4 weeks) evaluate hemodynamic parameters, the blood sample was collect for estimation of biochemical and rats were decapitated excised hearts, cleaned and weighed. Heart homogenate was used to determine antioxidants and oxidative levels, and histopathological evaluations were carried out to determine changes induced by Acrylamide. As compared control groups, ACR treated rats show altered significantly (P < 0.05) general characteristic and also elevated myocardial damage markers, altered hemodynamic, oxidative stress level, increased expression of inflammatory cytokines and induced histopathological changes. Treatment with Quercetin at 25 mg/kg and 50 mg/kg recouped the above changes significantly (P < 0.05), 50 mg/kg being more prominent. The present study has concluded that Quercetin protects against Acrylamide-induced cardiotoxicity.

Background: The etiology of autism spectrum disorders is not well known but oxidative stress has been suggested to play a pathological role. Purpose: The present study aimed to investigate the effects of supervised physical exercise on oxidative stress in children with autism. Methods: Thirty boys with autism (7 to 9 years old) voluntarily participated in this study. They were randomly divided into experimental and control groups. The experimental group performed a physical curriculum known as sports, play and active recreation for kids (SPARK) for 12 weeks (3 sessions of 45 minutes per week), while the control group was exempted to participate in the program. As the clinical parameters, the level of brain-derived neurotrophic factor (BDNF) and malondialdehyde (MDA) were measured before and after the intervention. Results: The results indicated that BDNF levels significantly increased (85.7 %) and MDA concentration significantly decreased (29.2%) in the experimental group (p < 0.05). Conclusions: In conclusion, the supervised physical exercise may result in less oxidative stress in children with autism.

Algae have the ability to detoxify the environment by binding heavy metals. They are also used as natural biosensors. In this study, the effect of barium chloride on saltwater algae was examined. Direct influence on the development of the test organism was visualized by means of infrared spectroscopy. The direct effect of barium chloride describes the amount of polyphenols and chlorophyll content in the culture. Work indicates the toxicity scale of a relatively small amount of toxic factor. The results were compared with the effects for a non-barium chloride culture.

In this study, we examined whether and to what extent oxidative stress is induced in seedlings of two winter triti- cale (Triticosecale wittm.) varieties (susceptible Tornado and resistant witon) in response to infestation by the cereal grain aphid (sitobion avenae l.) and bird-cherry-oat aphid (rhopalosiphum padi l.). we compared the level of hydrogen peroxide (H 2O2) and lipid peroxidation products as well as markers of protein damage (protein- bound thiol and carbonyl groups). The studied parameters were measured at 6, 24, 48 and 96 h post-initial aphid infestation compared to the non-infested control seedlings. Our studies indicated that the cereal aphid feeding evoked oxidative stress in the triticale seedlings. Cereal aphid feeding increased the H2O2 level in triticale tissues, with maximum levels observed at 24 and 48 h post-infestation. Triticale infestation with aphids also increased lipid peroxidation products in triticale seedlings, with the maximal levels at 48 or 96 h post-infestation. Further, there was a reduction in protein thiol content and an increase in protein carbonyl content in the triticale seedlings after infestation with female aphids. Stronger triticale macromolecule damages were evoked by the oligophagous aphid r. padi. There was a more substantial protein thiol content reduction in the resistant witon cultivar and higher accumulation of protein-bound carbonyls in the tissues of the susceptible Tornado cultivar. The changes were proportional to the aphid population and the time of aphid attack. These findings indicate that the defensive strategies against cereal aphid (s. avenae and r. padi) infestation were stimulated in triticale Tornado and witon seedlings. Our results explain some aspects and broaden the current knowledge of regulatory mechanisms in plant-aphid interactions.

The aim of this pilot study was to determine the baseline state of oxidative stress indices in patients with Parkinson's disease (PD). Peripheral blood samples of 15 PD subjects were analyzed and compared with ten age matched healthy controls. Patients with PARK2 mutations were also compared with PD patients without mutations. There was significant increase in malondialdehyde content and superoxide-dismutase (SOD) activity in peripheral blood parameters in PD patients (p < 0.05) in comparison to controls. These findings suggest an important role of oxidative stress in Parkinson's disease evolution and progress. No changes were observed in glutathione peroxidase and nitric oxide levels. We found significant correlation between SOD activity and lipid peroxidation when the biochemical data was further analyzed. In addition, significant increase in the levels of SOD among the PD patients with PARK2 mutations was observed, which can be ascribed to chronic oxidative stress induced by PARK2 mutations.

The role of nitric oxide (NO) in paraquat (PQ)-induced neurotoxicity is still not fully understood. In this study we used NG-nitro-L-arginine methyl ester (L-NAME), a non-selective nitric oxide synthase (NOS) inhibitor, in order to examine the effects of NO, reactive oxygen species (ROS) generation and lipid peroxidation (LPO) development during PQ-mediated neurotoxicity. Oxidative stress development in the striatum of Wistar rats intrastriatally (i.s.) poisoned with PQ (and in some cases pre-treated with L-NAME) was investigated by measuring superoxide anion (O2 •ˉ), malondialdehyde (MDA) and nitrate (NO3ˉ), 30 min, 24 hours and 7 days after treatment. L-NAME pretreatment provided the possibility to distinguish the role of ROS from reactive nitrogen species (RNS) in oxidative stress development induced by PQ. Our results confi rm the involvement of NO in PQ-mediated neurotoxicity and reduced LPO by L-NAME pretreatment implying that the latter has a protective role.

The purpose of this study was establishing the basic energetic parameters of amoeba Acanthamoeba castellanii mitochondria respiring with malate and their response to oxidative stress caused by hydrogen peroxide in the presence of Fe2+ ions. It appeared that, contrary to a previous report (Trocha LK, Stobienia O (2007) Acta Biochim Polon 54: 797), H2O2-treated mitochondria of A. castellanii did not display any substantial impairment. No marked changes in cytochrome pathway activity were found, as in the presence of an inhibitor of alternative oxidase no effects were observed on the rates of uncoupled and phosphorylating respiration and on coupling parameters. Only in the absence of the alternative oxidase inhibitor, non-phosphorylating respiration progressively decreased with increasing concentration of H2O2, while the coupling parameters (respiratory control ratio and ADP/O ratio) slightly improved, which may indicate some inactivation of the alternative oxidase. Moreover, our results show no change in membrane potential, Ca2+ uptake and accumulation ability, mitochondrial outer membrane integrity and cytochrome c release for 0.5-25 mM H2O2-treated versus control (H2O2-untreated) mitochondria. These results indicate that short (5 min) incubation of A. castellanii mitochondria with H2O2 in the presence of Fe2+ does not damage their basic energetics.

Candida species are associated with an increasing number of life-threatening infections (candidiases), mainly due to the high resistance of these yeast-like fungi to antifungal drugs and oxidative stress. Recently, thiamine (vitamin B1) was found to alleviate stress responses in Saccharomyces cerevisiae; however, thiamine influence on defense systems in pathogenic fungi has never been investigated. The current work was aimed to elucidate the role of thiamine in stress reactions of C. albicans, C. glabrata, C. tropicalis and C. dubliniensis, subjected to hydrogen peroxide treatment. As compared to S. cerevisiae, Candida strains exposed to oxidative stress showed: (i) a much higher dependence on exogenous thiamine; (ii) an increased demand for thiamine diphosphate (TDP) and TDP-dependent enzyme, transketolase; (iii) no changes in gene expression of selected stress markers - superoxide dismutase and catalase - depending on thiamine availability in medium; (iv) a similar decrease of reactive oxygen species (ROS) generation in the presence of thiamine. Moreover, the addition of therapeutic doses of thiamine to yeast culture medium revealed differences in its accumulation between various Candida species. The current findings implicate that the protective action of thiamine observed in S. cerevisiae differs significantly form that in pathogenic Candida strains, both in terms of the cofactor functions of TDP and the effects on fungal defense systems.

Background The photocopying occupation contributes to various health issues in workers. Elevated levels of oxidative stress have been found to be associated with respiratory problems in occupational workers. The intention of the current research is to evaluate the association between the pulmonary function parameters, a biomarker of oxidative stress, and the photocopying occupation in Lahore, Pakistan. Material and Methods The study was performed on a group of photocopying operators (N = 100) and a control group (N = 100). A structured questionnaire was developed to gather the demographic attributes. Lung function testing was done with the spirometer. The blood lipid peroxides level, as thiobarbituric acid reactive substances (TBARS), was assessed spectrophotometrically. Results A statistically significant reduction was found in the pulmonary function parameters as compared to the control group. In the study group, the prevalence of respiratory symptoms, including cough (31%), sneezing (18%), wheezing (9%) and breathing issues (19%), was high as compared to the control group. A significantly higher plasma TBARS concentration was observed in the photocopying workers. A negative correlation prevailed between FVC (−0.081), FVC% predicted (−0.038) and FEV1 (−0.043), and the duration of exposure. Conclusions The study showed that the occupational exposure could have a significant effect on the normal pulmonary function and the oxidative potential of well-being. The profound emissions in the vicinity of photocopying centers may be responsible for the variations in the pulmonary function parameters and oxidative stress. Med Pr. 2019;70(4):403–10

Cukrzyca ciążowa (gestational diabetes mellitus – GDM) jest jedną z najczęstszych chorób prowadzących do powikłań w trakcie ciąży. Zauważono, że GDM towarzyszy nasilenie stresu oksydacyjnego. Celem niniejszej pracy jest podsumowanie stanu wiedzy na temat wpływu stresu oksydacyjnego na występowanie i przebieg GDM oraz jej odległe skutki. Przegląd ma także na celu sprawdzenie potencjalnych implikacji klinicznych związanych z GDM. Przeszukano bazy danych takie jak Pubmed, Scopus i Google Scholar. Artykuły wyszukiwano m.in. z użyciem fraz „gestational diabetes mellitus” i „oxidative stress”. Wykazano istotnie wyższy po-ziom stresu oksydacyjnego u kobiet z GDM, zarówno we krwi, jak i w łożysku. Stres oksydacyjny może być ściśle związany z patogenezą GDM. Ze względu na istotną rolę stresu oksydacyjnego w patofizjologii GDM wiele badań skupia się na poszukiwaniu substancji, które mogą zmniejszyć stężenie prooksydantów i zwiększyć udział antyoksydantów, tym samym poprawiając potencjał oksydacyjny. Stres oksydacyjny odgrywa kluczową rolę w patogenezie i patofizjologii GDM. Ponieważ jest to choroba, która może prowadzić do wielu powikłań zarówno u matki, jak i u płodu, jej wczesne wykrycie i skuteczne leczenie wymaga szczególnej uwagi. Ocena możliwości zastosowania parametrów stresu oksydacyjnego jako biomarkerów do wczesnego wykrywania GDM wymaga dalszych szczegółowych badań.
Gestational diabetes mellitus (GDM) is one of the most common diseases that leads to complications during pregnancy. It has been noticed that GDM is accompanied by an increase in oxidative stress. The aim of this study is to summarize the state of knowledge on the influence of oxidative stress on the occurrence and course of gestational diabetes as well as its long-term effects. This review also aims to examine the potential clinical implications of GDM. Databases such as Pubmed, Scopus, and Google Scholar were searched. Articles were searched for using the phrases “gestational diabetes mellitus” and “oxidative stress”. A significantly higher level of oxidative stress was found in women with GDM, both in the blood and in the placenta. Oxidative stress may be closely related to the pathogenesis of GDM. Due to the important role of oxidative stress in the pathophysiology of GDM, many studies have focused on finding substances that can reduce the concentration of prooxidants and increase the proportion of antioxidants, thereby improving the oxidative potential. Oxidative stress plays a key role in the pathogenesis and pathophysiology of GDM. As it is a disease that can lead to a number of complications for both the mother and the fetus, its early detection and effective treatment require special attention. Further detailed research is required to evaluate the possibility of using the parameters of oxidative stress as biomarkers for the early detection of GDM.

Fluoride is regarded as one of the strongest oxidants, which causes oxidative changes in cells of living organisms. It may both increase the content of reactive oxygen species and inhibit the activity of antioxidative enzyme. In recent years, many researchers successfully used the properties of clay minerals in the sorption of fluoride ion from water. This raises the question of the possibility of limiting the effect of fluorine on the negative changes in plants by adding bentonite to soil. A two-year pot experiment was carried out in the Greenhouse of West Pomeranian University of Technology in Szczecin, on loamy sand and sandy loam. Each sample of soil was mixed with three different concentrations of bentonite – 1, 5, 10% of dry weight (DW) of the soil and then treated with 30 mmol of F- per 1 kg of dry weight of the soil in a form of NaF solution. A control series was prepared for each soil, to which no additives were added. The medium prepared in such way was transferred to plastic pots (3 kg each) and seeded with 16 pea seeds of Pisum sativum. In three phases of pea development (4 leaves unfolded, flowering and development of fruit), fresh leaf samples were collected and the concentrations of ascorbic acid, reduced glutathione, total flavonoids and total polyphenols were measured. Sodium fluoride introduced to the soil changed the level of antioxidant parameters in the plant, which may suggest that fluoride is involved in the formation of reactive oxygen species, resulting in oxidative stress. Bentonite in a dosage of 10% reduced the toxic effects of fluoride on the oxidative balance and morphological changes in the plant, which was observed especially for loamy sand, naturally poor in clay minerals.

Objectives: Octabromodiphenyl ether (OctaBDE) was used as a flame retardant applied mostly in the manufacture of plastics utilized in the electrical and electronic industries. Owing to its long half-life and being regarded as an environmental pollutant, OctaBDE, like other polybrominated diphenyl ethers, has been classified as a persistent organic pollutant (POP). This study was carried out to assess the effects of oxidative stress (redox homeostasis) induced in rats by OctaBDE. Material and Methods: Female Wistar rats exposed intragastrically to OctaBDE at single (25, 200 or 2000 mg/kg b.w.), or repeated (0.4, 2, 8, 40 or 200 mg/kg/day) doses during 7–28 days were used in the experiment. Selected oxidative stress parameters were determined in the liver and blood serum. Results: Administration (single or repeated) of OctaBDE to rats resulted in the impaired redox homeostasis, as evidenced by the increased levels of reduced (GSH) and oxidized (GSSG) glutathione in the liver, the reduced total antioxidant status (TAS) in serum and the increased concentration of malondialdehyde (MDA) in the liver. After multiple doses of OctaBDE, elevated activity of glutathione transferase (GST) in the liver was also noted. Conclusions: After repeated administration of OctaBDE at the lowest dose (0.4 mg/kg/day), changes were observed in the parameters (MDA, TAS, GSSG) indicative of oxidative stress.

In the retinoic acid-differentiated neuroblastoma SH-SY5Y cells, IL-1 induced binding activity of NFκB and up-regulated the expression and activity of MnSOD. The IL-1-elicited effects were partly reversed by IL-4 and IL-6. It is proposed that IL-4 and IL-6 may participate in the regulation of the imbalanced oxidant status induced by IL-1 in differentiated neuroblastoma cells. In the SH-SY5Y cell line, TNFα neither activated NFκ nor induced MnSOD expression and activity, but was capable of modulating the IL-1 effects. Pyrrolidine dithiocarbamate (PDTC), an inhibitor of NFκB activation, down-regulated the expression and activity of MnSOD, which may suggest that the regulation of MnSOD by IL-1 in retinoic acid-differentiated neuroblastoma cells was mediated by the nuclear factor κB.

Opis:
Characterization of free radical-induced cell injury processes of placenta cells is of vital importance for clinical medicine for the maintenance of intrauterine fetal life. The present study has analyzed cell injury processes in cells of the choriocarcinoma cell line JAR treated with menadione, an anticancer drug, and Hg2O2 in comparison to osteosarcoma 143B cells using electron microscopic and flow cytometric techniques. Flow cytometry on JAR cells exposed to 100 μM menadione and double-stained with Annexin V and propidium iodide (PI) detected apoptotic cells reaching the maximum after 4 h of incubation with a rapid decrease thereafter. Viable cells became decreased to 46% of the control after 2 h of incubation, reaching 5% after 4 h. Cells stainable with both Annexin V and PI began to increase distinctly after 2 h of incubation, reaching 55% after 4 h. Electron microscopy showed that cells stainable with both dyes specified above had condensed nuclei and swollen cytoplasm, suggesting that they were undergoing a switch of the cell death mode from apoptosis to necrosis. On the other hand, 90% of 143B cells remained intact after 4 h of menadione treatment although the intracellular levels of superoxide were always higher than those of JAR cells treated with the drug. In contrast, JAR cells were more resistant than 143B cells to H2O2-induced cytotoxicity. These results may suggest that cytotoxicity of menadione cannot be explained simply by oxygen free radicals generated from the drug. The resistance of JAR cells to oxygen free radical-induced cytotoxicity may be advantageous for intrauterine fetal life.

Opis:
Oxidative stress (OS) plays a pivotal role in the development of many diseases. Therefore, many efforts have been undertaken to minimize the consequences of OS or improve antioxidant defence systems. One solution expected to improve redox homeostasis is the ketogenic diet (KD). KD is a low-carbohydrate, high-fat diet leading to a ketosis state. The shift in metabolism occurring in ketosis can affect the reactive oxygen species (ROS)-producing pathways and influence the expression of enzymes involved in redox homeostasis. Therefore, this review summarizes and discusses existing knowledge about KD and ROS homeostasis. The available tools for evaluating OS status are presented, listing their potential and drawbacks. An important aspect is the summary of the current knowledge about the effect of KD on OS conducted in vitro, in vivo, and in clinical trials. Finally, the review addresses future studies needed to understand the connection between KD and OS.

Purpose. Topotecan, a semisynthetic water-soluble derivative of camptothecin exerts its cytotoxic effect by inhibiting topoisomerase I and causes double-strand DNA breaks which inhibit DNA function and ultimately lead to cell death. In previous studies it was shown that camptothecin causes ROS formation. The aim of this study was to investigate if Topotecan like camptotecin causes oxidative stress in MCF-7 human breast cancer cell line. Determining the oxidant effect of Topotecan may elucidate a possible alternative mechanism for its cytotoxicity. Experimental design. MCF-7 cells were cultured and exposed to Topotecan for 24 h at 37°C. The viability of the cells (% of control) was measured using the colorimetric 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Lipid peroxidation (TBARS), protein oxidation (carbonyl content), sulfhydryl, glutathione (GSH) levels, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities were determined in MCF-7 cells with and without Topotecan incubation. Results. We found the IC50 concentration of Topotecan as 0.218 µM in MCF-7 cells. This concentration of Topotecan was used in the incubations of the cells. Our data indicated increased oxidative status, as revealed by increased lipid peroxidation and protein oxidation, and decreased GSH and sulfhydryl levels in MCF-7 cells exposed to Topotecan compared to control cells. In contrast, there was a slight increase in SOD and a significant increase in GPx and catalase activity in MCF-7 cells incubated with Topotecan compared to the control. Conclusions. These results support our hypothesis that Topotecan increases oxidative stress in MCF-7 cells.

Effects of nonylphenol-induced oxidative stress in ovary of cichlid fish, Etroplus maculatus (Bloch, 1795)

The present study was designed to evaluate the effects of nonylphenol in the pro-oxidant/ antioxidant system in ovary of the cichlid fish Etroplus maculatus. Fishes were exposed at two sublethal concentrations (one-fifth and one-tenth of LC50) of nonylphenol for 24, 72 and 96 h maintaining control groups. The oxidative stress indices as the activities of antioxidant enzymes, superoxide dismutase, catalase, glutathione reductase along with the levels of hydrogen peroxide generation and lipid peroxidation were monitored in concentration- and time-dependent manner. Activity of superoxide dismutase significantly (P<0.05) increased at both concentrations in timedependent manner. Meanwhile the activities of catalase and glutathione reductase significantly (P<0.05) decreased after 72 and 96 h of nonylphenol treatment. The levels of hydrogen peroxide generation and lipid peroxidation increased in all treatment groups when compared to controls. The present results demonstrated that the induction of oxidative stress in ovary of fish by the generation of lipid peroxidation could be due to the exposure of environmental contaminant, nonylphenol. Therefore, the observed oxidative stress in ovary can be indicated as a mechanism of toxicity in the fish exposed to nonylphenol.

The aim of the present study was to determine the effects of Luteolin (LUT) on semen quality, oxidative stress, apoptosis, acrosomal integrity, mitochondrial membrane potential and dead sperm ratio in rabbits. Ejaculates from six New Zealand rabbits were collected, evaluated and pooled. The pooling was divided into five groups as control (no additive) LUT 25 μM, LUT 50 μM, LUT 100 μM and LUT 200 μM and LUT added. It was then filled into a falcon tube with Tris-based extender at a final concentration of approximately 35 x 106 spermatozoa. Diluated rabbit semen samples were drawn into frozen and thawed. Frozen semen straws were thawed at 37°C in 30 seconds. According to our findings, no statistical difference was found between all doses of luteolin and the control group in the CASA (computer assisted sperm analysis) analysis performed at 4°C. However, total motility, progressive motility and rapid sperm percentage were found to be higher in the frozen and thawed rabbit semen at a dose of LUT 50 μM compared to the other groups (p<0.05). While amplitude of lateral head displacement (ALH) and beat cross-frequency (BCF) values were found at the lowest dose of LUT 200 μM, a statistically significant difference was observed between the other groups. When the flow cytometry results were examined, no statistical difference was found between the rate of dead sperm, acrosomal integrity, mitochondrial membrane potential and apoptosis rate. Morever, the H2O2 percentage was found to be lower in all experimental groups compared to the control group (p<0.001). In conclusion, the addition of LUT in long-term storage of rabbit semen provided a protective effect for spermatozoa with its antioxidative properties against damage caused by cryopreservation.

The effect of some species of the Fabaceae family (the pea Pisum sativum L., broad bean Vicia faba L. and vetch V. sativa L.) on biochemical markers of oxidative stress within tissues of the pea aphid, Acyrthosiphon pisum Harris (Homoptera, Aphididae), has been studied. The highest concentration of superoxide anion radical (O2-), hydrogen peroxide (H2O2) and lipid peroxidation products thiobarbituric acid reactive substances (TBARS) was recorded for morphs that fed on the broad bean. The opposite tendency was observed for the level of total thiols, that were the highest for insects reared on the pea. Among the studied aphid morphs, the highest concentration of superoxide anion and hydrogen peroxide was noted for the wingless females whereas TBARS content was the highest within the winged migrant tissues. Different results were obtained for the total thiols, where apterae as well as migrants reared on the tested plants, had comparable content of these compounds. Our experiments indicate that oxidative stress may play an important role in interactions between the pea aphid and their host plants.

The aim of this study was to compare the effect of flunixin meglumine (FM) and meloxicam (M) on postoperative and oxidative stress in ovariohysterectomized bitches. Twenty four bitches were divided into three groups (n=8 in each) and treated during premedication as follows: FM (2.2 mg/kg, iv, Fluvil, Vilsan, Turkey), M (0.2 mg/kg, sc, Maxicam, Sanovel, Turkey) or 0.9% saline (1 ml, iv, IE, Turkey) - control (C) group. The concentrations of serum cortisol, nitric oxide (NO), malondialdehyde (MDA), antioxidant potential (AOP) and glutation (GSH) were measured in blood samples collected during incision (0 h), closure of incision line (0.5 h) and 1, 2.5, 12 and 24 hours after incision. It was observed that cortisol level was higher at 0.5, 1 and 2.5 h in group C (p < 0.05), 0.5 h in group FM (p < 0.001), and 1 and 2.5 h in group M (p < 0.01), as compared to that determine at 0 h. Group C showed higher cortisol level during 0.5 h (p < 0.05) than that found in the other groups. Group FM displayed lower levels during 1 h (p < 0.01) and 2.5 h (p < 0.05) as compared to those observed in other groups. Concentrations of MDA, AOP and GSH between all the groups did not show any significant differences. MDA level was higher at 0.5 and 1 h in groupM (p < 0.05) than that found in group C and it was the lowest at 2.5 h in group C (p < 0.05). AOP was higher at 2.5 h in group FM and M (p < 0.05) than that observed in group C, and at 12 and 24 h in group M than that found in group C and FM. GSH did not show any significant differences between the groups. NO level in group FM after 12 h was higher (p < 0.05) than that at 0.5, 1 and 24 h. Moreover, NO level was lower at 0.5 (p < 0.01), 1 (p < 0.05) and 24 h (p < 0.05) in group FM than that observed in group C and M. In conclusion, flunixin meglumine decreases cortisol and NO levels more efficiently than meloxicam. Therefore, it is suggested that postoperative stress following ovariohysterectomy may be prevented by flunixin meglumine in bitches.

Thioredoxin (Trx) is a multifunctional protein with a redox-active disulfide/dithiol in the active site. Thioredoxin, with its redox-regulating and reactive oxygen species (ROS) scavenging activities, plays several important biologic roles both in intracellular and extracellular compartments. The purpose of this report was to quantify the relative expression of Trx in rat hippocampus following an oxidative stress-involving treatment such as kainic acid (KA) using real-time PCR and the 2-ΔΔCT method. The relative changes in expression of Trx mRNA in KA-treated and control animals were significantly different as 2.02 ± 0.77 and 1.0 ± 0.26, respectively ( P <0.05). Minimum and maximum n-fold changes in Trx expression in KA-treated and control animals were determined as (1.4-5.2) and (0.8-1.3), respectively. Thus, real-time PCR and the 2-ΔΔCT method for data analysis from real-time PCR were found to be an accurate and sensitive method for quantifying Trx mRNA levels.

In the last few years on old yew trees growing in the parks and gardens extensive disease symptoms were observed on leaves, indicating affection with pathogenic microorganisms. The causal factor was the presence of a fungal pathogen, Pestalotiopsis funerea. Physiological responses involving the oxidative stress, i. e. superoxide anion and hydrogen peroxide levels, and the superoxide dismutase were analyzed as an element of a plant defense mechanism. Diseased leaves from plants growing under sunny and shaded conditions, from May to July, were investigated. The increased generation of superoxide anion-radicals and hydrogen peroxide were observed in the leaves with disease symptoms simultaneously with the activation of superoxide dismutase, which may indicate the induction of host defense response to the P. funerea.

Different maize (Zea mays L.) varieties have been used for thousands of years as a healthy food source in Mesoamerica including pigmented maize. Maize ingestion could contribute to the reduction in the rate of non-communicable diseases and, in turn, to its function as an adjuvant in their management. These diseases are mainly associated with oxidative stress, which is characterized by a redox cell imbalance produced due to pro-oxidant molecules accumulation, inducing irreversible damages. Although the endogenous antioxidant defense system is efficient, exogenous antioxidants are necessary to help to prevent this damage. Bioactive compounds, like anthocyanins, contained in dietary plants exert a major activity against oxidative stress. Could the maize anthocyanins play a curative, preventive or complementary role in the treatment of chronic diseases? Here, we describe the occurrence of anthocyanins from pigmented maize and their chemical structures. Furthermore, the biosynthesis, bioavailability, and stability are also summarized. Finally, many in vitro and in vivo studies of maize anthocyanins are discussed that demonstrated their nutraceutical potential, antioxidant capacity, and other biological effects. Given the importance of the biological properties of maize anthocyanins, it is necessary to understand the current knowledge and propose further research or clinical studies which allows us to better elucidate the biological mechanism of maize anthocyanins derivatives of several varieties and processes of cooking and combination with other ingredients to enhance their nutritional and health benefits.

The present investigation was conducted to appraise the physiochemical adjustments in contrasting maize cultivars, namely, PakAfgoi (tolerant) and EV-5098 (sensitive) subjected to heat shock. Seven-day-old seedlings were exposed to heat shock for different time intervals (1, 3, 6, 24, 48 and 72 h) and data for various physiochemical attributes determined to appraise time course changes in maize. After 72 h of heat shock, the plants were grown under normal conditions for 5 d and data for different growth attributes and photosynthetic pigments recorded. Exposure to heat shock reduced growth and photosynthetic pigments in maize cultivars. The plants exposed to heat shock for up to 3 h recovered growth and photosynthetic pigments when stress was relieved. A time course rise in the relative membrane permeability, hydrogen peroxide (H2O2) and malondialdehyde contents was recorded particularly in the EV-5098 indicating that heat shock-induced oxidative stress. Activities of different enzymatic antioxidants greatly altered due to heat shock. For instance, an increase in superoxide dismutase activity was recorded in both maize cultivars. The activity of ascorbate peroxidase was greater in Pak-Afgoi. However, the peroxidase and catalase activities were higher in plants of EV-5098. Heat shock caused a significant rise in the proline and decline in the total free amino acids. Overall, the performance of Pak-Afgoi was better in terms of having lesser oxidative damage and greater cellular levels of proline. The results suggested that oxidative stress indicators (relative membrane permeability, H2O2 and malondialdehyde) and proline can be used as markers for heat shock tolerant plants.

Objectives A water pipe (hookah) is a tobacco smoking tool which is thought to be more harmless than a cigarette, and there are no adequate studies about its hazards to health. Water-pipe smoking is threatening health of the youth in the world today. The objective of this study has been to investigate the carbon monoxide (CO) levels in breath, examine the changes in pulmonary function tests (PFT) and to assess the change of the oxidative stress parameters in blood after smoking a water pipe. Material and Methods This study is a cross-sectional analytical study that has included 50 volunteers who smoke a water pipe and the control group of 50 volunteers who smoke neither a cigarette nor a water pipe. Carbon monoxide levels were measured in the breath and pulmonary function tests (PFTs) were performed before and after smoking a water pipe. Blood samples were taken from either the volunteer control group or water-pipe smokers group after smoking a water pipe for the purpose of evaluation of the parameters of oxidative stress. Results Carbon monoxide values were measured to be 8.08±7.4 ppm and 28.08±16.5 ppm before and after smoking a water pipe, respectively. This increment was found statistically significant. There were also significant reductions in PFTs after smoking a water pipe. Total oxidative status (TOS), total antioxidant status (TAS) and oxidative stress index (OSI) were found prominently higher after smoking a water pipe for the group of water-pipe smokers than for the control group. Conclusions This study has shown that water-pipe smoking leads to deterioration in pulmonary function and increases oxidative stress. To the best of our knowledge this study is the only one that has shown the effect of water-pipe smoking on oxidative stress. More studies must be planned to show the side effects of water-pipe habit and protective policies should be planned especially for young people in Europe. Celem pracy było zbadanie czy palmitynian retinylu redukuje stres oksydacyjny wywołany wysiłkiem fizycznym. Badania przeprowadzono na rosnących szczurach samcach rasy Wistar, które przez 10 dni biegały na bieżni z prędkością przesuwu 20 m/min. Szczurom podawano palmitynian retinylu w ilości 7,5, 15 lub 60 µg dziennie na zwierzą. Po tym okresie w osoczu oznaczano poziom retinolu, nadtlenki lipidowe i potencjał anty oksydacyjny oraz estry retinyłu w wątrobie. Stwierdzono, że palmitynian retinylu redukował stres oksydacyjny wywołany wysiłkiem fizycznym, a efektywność tego procesu nie zależała od dawki podanego palmitynianu.
The effect of retinyl palmitate dose on oxidative stress reduction induced by physical activity was the aim of study. The experiment was carried out on Wistar strain males rats, which were trained by running on trademill with speed 20 m/min. during 10 days. The rats were administered with 7,5, 15 or 60 µg of retinyl palmitate daily to each one. The level of retinol, lipid peroxides, antioxidant potential in serum and retinyl esters in liver were measured. It was shown that retinyl palmitate reduced oxidative stress, independently to its daily dose.

Objectives: Paints are complex mixtures of solvents and metals that can induce health damages in workers exposed to them. The aim of the present work was to evaluate possible oxidative and genotoxic effects in workers exposed to paints. Material and Methods: Peripheral blood and buccal cell samples were collected from 33 workers exposed to paints and 29 non-exposed workers (controls) during an ordinary working week (Monday morning and Friday evening). Oxidative markers were assessed using thiobarbituric acid assay, carbonylated proteins, superoxide dismutase and catalase activities. Hippuric acid and delta-aminolevulinic acid were determined as biomarkers of toluene and lead exposure, respectively. Genotoxicity was measured through comet assay and micronucleus (MN) frequencies. Results: The exposed group showed higher hippuric acid and delta-aminolevulinic acid levels (Friday samples) and lower superoxide dismutase activity (Monday samples) in relation to control group. DNA damage index (comet assay) was higher in the exposed group, both in Monday and Friday samples, compared to the control group. No differences were observed in frequency of micronuclei (MN) between the groups, either in lymphocytes or buccal cells. However, the exposed group presented an increase (Monday samples) in nuclear buds frequency in lymphocytes — a marker of gene amplification — as well as an increase in condensed chromatin in the buccal cells (Monday and Friday samples), suggesting induction of apoptosis. Furthermore, a decrease in the nuclear division index (Friday samples) was observed in the exposed group, indicating that paint exposure induces cytostatic effects in lymphocytes. Conclusion: The results suggest that individuals exposed to paints have increased levels of DNA damage.

Objectives Perfluorooctane sulfonate (PFOS) and zinc oxide nanoparticles (ZnO-NPs) are frequently detected in the environment but few studies have assessed the joint toxicity of them. Oxidative stress and apoptosis to zebrafish (Danio rerio) embryos induced by the PFOS and ZnO-NPs were investigated in this study. Material and Methods The embryos were exposed to the PFOS (0, 0.4, 0.8 and 1.6 mg/l), ZnO-NPs (12.5, 25, 50 mg/l) and PFOS plus ZnO-NPs (0.4+12.5, 0.8+25 and 1.6+50 mg/l) mixture solutions until 96 h post-fertilization. Activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (Gpx) and malondialdehyde (MDA) content were measured in zebrafish embryos after exposure lasting for 96 h. At the same time, the genes expression related to reactive oxygen species (ROS) generation, oxidative damage and apoptosis were also measured. Results A significant induction of the ROS accompanied by the increase in the activity of the Gpx and MDA contents were found in co-treatment groups. Furthermore, several apoptosis pathway related genes such as Bax, p53, caspase-3 and caspase-9 were significantly up-regulated in the PFOS plus ZnO-NPs exposure groups, while anti-apoptotic gene Bcl-2 was significantly down-regulated in the PFOS plus ZnO-NPs exposure groups. In addition, some oxidative stress-related genes such as Cat, GSH peroxidase 1 (Gpx1a) and superoxide dismutase 1 (Sod1) were also significantly down-regulated after the PFOS plus ZnO-NPs co-treatments. Conclusions The results demonstrated that the PFOS plus ZnO-NPs co-exposure could cause more serious oxidative stress and apoptosis than the PFOS and ZnO-NPs exposure alone at the exposure concentrations above. The synergistic effects of the PFOS and ZnO-NPs may be the important mechanisms of their toxicity to zebrafish embryos. Int J Occup Med Environ Health 2017;30(2):213–229

Opis:
Inflammation is a non-specific immune response to infection, irritation or other injury, the key features being redness, warmth, swelling and pain. A number of mediators are released which alter the resistance of mucosa to injury induced by noxious substances. Oxidative stress is a unifying mechanism of injury in many types of disease processes, including gastrointestinal diseases. It has been defined as an imbalance in the activity of pro and antioxidants. Pro-oxidants favour free radical formation while antioxidants inhibit or retard the same. A number of markers of oxidative stress have been identified. This review provides an overview of various mediators of inflammation and oxidative stress, and diverse approaches for prevention and treatment of gastrointestinal inflammation.

The appearance of a disease is caused by many factors, which for the most part have their source in the external environment and in the attitudes and health habits of a person. Rational prevention, which seems to be underrated, can affect behavior and improve health. One of its elements is certainly a diet rich in antioxidants. They are compounds that inhibit the excessive production of free radicals or convert them into inactive derivatives. Thanks to this, they prevent the formation of free radical damage. They may be of endo- or exogenous origin. From the point of view of their mechanism of action, they can also be divided into enzymatic and non-enzymatic ones. In combination, they form the so-called antioxidant protection system of every organism. The present review article outlines the problem of prevention of oxidative stress, which is a risk factor responsible for the development of many diseases, especially civilization diseases. The structure of the antioxidant protection system of the human body has been discussed. The article also presents the main antioxidants that can be delivered with food, as well as their role in the prevention of increased amounts of free radicals and reactive oxygen species.
Pojawienie się choroby spowodowane jest wieloma czynnikami, które w większości mają swoje źródło w środowisku zewnętrznym oraz w postawach i nawykach zdrowotnych człowieka. Racjonalna profilaktyka, która wydaje się wciąż niedoceniana może odpowiednio wpływać na zachowanie i poprawę zdrowia. Jednym z jej elementów jest z pewnością dieta obfitująca w przeciwutleniacze. Są to związki, które hamują nadmierne wytwarzanie wolnych rodników bądź przemieniają je w nieaktywne pochodne. Dzięki temu przeciwdziałają powstawaniu wolnorodnikowych uszkodzeń. Mogą on mieć pochodzenie endo- i egzogenne. Z punktu widzenia ich mechanizmu działania można podzielić je również na enzymatyczne i nieenzymatyczne. W połączeniu tworzą one tak zwany system ochrony antyoksydacyjnej każdego organizmu. Prezentowany artykuł poglądowy stanowi zarys problematyki profilaktyki stresu oksydacyjnego, który jest czynnikiem ryzyka zachorowania i rozwoju wielu chorób, a zwłaszcza schorzeń cywilizacyjnych. Przybliżono budowę systemu ochrony antyoksydacyjnej organizmu człowieka. Przedstawiono główne przeciwutleniacze, które można dostarczać wraz z pożywieniem, a także ich rolę w prewencji zwiększonej ilości wolnych rodników i reaktywnych form tlenu.

In both forms of muscular dystrophy, the severe Duchenne’s muscular dystrophy (DMD) with lifespan shortened to about 20 years and the milder Becker dystrophy (BDM) with normal lifespan, the gene defect is located at chromosome locus Xp21. The location is the same in the experimental model of DMD in the mdx mice. As the result of the gene defect a protein called dystrophin is either not synthesized, or is produced in traces. Although the structure of this protein is rather well established there are still many controversies about the dystrophin function. The most accepted suggestion supposes that it stabilizes sarcolemma in the course of the contraction-relaxation cycle. Solving the problem of dystrophin function is a prerequisite for introduction of an effective therapy. Among the different factors which might be responsible for the appearance and progress of dystrophic changes in muscles there is an excessive action of oxidative stress. In this review data indicating the influence of oxidative stress on the severity of the pathologic processes in dystrophy are discussed. Several pieces of data indicating the action of oxidative damage to different macromolecules in DMD/BDM are presented. Special attention is devoted to the degree of oxidative damage to muscle proteins, the activity of neuronal nitric oxide synthase (nNOS) and their involvement in defining the severity of the dystrophic processes. It is indicated that the severity of the morbid process is related to the degree of oxidative damage to muscle proteins and the decrease of the nNOS activity in muscles. Estimation of the degree of the destructive action of oxidative stress in muscular dystrophy may be a useful marker facilitating introduction of an effective antioxidant therapy and regulation of nNOS activity.

Objective. Little is known about lead toxic effects on lung tissue. Therefore, the present study was undertaken to estimate the values of selected parameters related to oxidative stress in the lung of rats exposed to lead acetate. Material and methods. Twenty male Wistar rats were randomly divided into two groups: control (n=10) and lead-treated (n=10). The lead-treated group was fed with regular rat chow and distilled water supplemented with lead acetate (1200 ppm) for 7 days. In lung homogenates, the level of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD, CuZn-SOD, Mn-SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR), and glutathione-S-transferase (GST) were determined. Results. The activity of catalase was significantly higher in the lung of lead-treated rats than in controls by 98%. Similarly, the activities of GPx and GR were higher by 57% and 45%, respectively. The level of MDA was also higher in lead-exposed rats than in the control group by 52%. The remaining parameters did not differ between the studied groups. Conclusions. Lead exposure stimulates antioxidant defense mechanisms in rat lung; however, these mechanisms are insufficient to prevent lead-induced oxidative stress development
Wstęp. Toksyczny wpływ ołowiu na tkankę płucną jest słabo poznany. W związku z tym, prezentowany eksperyment miał na celu ocenę zmian w wybranych parametrach związanych ze stresem oksydacyjnym w tkance płucnej u szczurów narażonych na octan ołowiu. Materiał i metody. Dwadzieścia szczurów szczepu Wistar, podzielono na dwie grupy: kontrolną (n=10) i badaną z podawanym ołowiem (n=10). Szczury z grupy badanej były regularnie karmione granulatem dla szczurów i pojone wodą destylowaną z octanem ołowiu (1200 ppm) przez 7 dni. W homogenatach z tanki płucnej oznaczany był poziom malonylodialdehydu (MDA) i aktywność dysmutazy ponadtlenkowej (SOD, CuZn-SOD, Mn-SOD), katalazy (CAT), peroksydazy glutationowej (GPx), reduktazy glutationowej (GR), oraz transferazy glutationowej (GST). Wyniki. Aktywność katalazy była o 98% wyższa w grupie badanej aniżeli w grupie kontrolnej. Podobnie, aktywności GPx i GR były wyższe odpowiednio o 57% i 45%. Poziom MDA był również wyższy w grupie badanej, o 52% w porównaniu do grupy kontrolnej. Pozostałe parametry nie różniły się pomiędzy grupami. Wnioski. Ekspozycja na ołów stymuluje mechanizmy obrony antyoksydacyjnej, jednakże mechanizmy te są niewystarczające by zapobiec stresowi oksydacyjnemu wywołanemu działaniem ołowiu.

Inula viscosa is a perennial herbaceous plant native to the Mediterranean Basin, which is used topically for the treatment of various diseases in folk medicine. This study aimed to evaluate the in vivo intestinal anti-inflammatory activity of the ethanolic extract of I. viscosa (EEIV) and to test its effect on a colorectal cancer cell line. EEIV was administered to rats orally and daily at 100 and 200 mg/kg body weight for 7 days, and then colitis was induced by intrarectal instillation of 2 ml of 4% (v/v) acetic acid (AA) solution. At the end of the experiment, clinical examinations of the rats were conducted by evaluating macroscopic and histological signs of colonic tissues and measuring erythrocyte sedimentation rate (ESR) and the levels of C-reactive protein, fibrinogen, myeloperoxidase (MPO), malondialdehyde (MDA) and nitric oxide (NO). Using MTS assay, the antiproliferative effect of EEIV against human colon carcinoma HT29 cells and cytotoxicity on nondifferentiated Caco-2 cell line was evaluated. EEIV significantly decreased the ESR and fibrinogen levels as compared to control colitic rats (P < 0.001). It also significantly decreased the NO, MDA, and MPO levels in the colon tissue compared with the untreated colitic group (P < 0.001). These results were confirmed by macroscopic and histological examination, which showed significant protection against AA-induced ulcerative colitis. Furthermore, EEIV at a concentration of 369.88 μg/ml did not show cytotoxicity on confluent Caco-2 cells, with significant inhibition of colorectal cancer cell (HT29) growth (EC50 = 62.39 μg/ml). These results demonstrate that EEIV plays a potential role as pharmacological tool in the management of inflammatory bowel disease and prevention of colorectal cancer.

Opis:
Quercetin, a member of the flavonoid family is a major antioxidant acquired in humans by food consumption, while Cadmium fluoride (CdF2) is one of the naturally occurring chemicals having adverse effects. The protective effect of quercetin on time dependent oxidative damage induced in mice liver by CdF2 was studied in the following groups of mice consisting of six mice each: (i) control group; (ii) mice treated with single i.p injection of 2 mg/kg bw CdF2 for 24 h; (iii) mice treated with single i.p injection of 2 mg/kg bw CdF2 for 48 h; (iv) mice treated with single i.p injection of quercetin (100 mg/kg bw); (v) mice treated with i.p injection of 100 mg/kg bw of quercetin followed by i.p injection of CdF2 (2 mg/kg bw) for 24 h; and (vi) mice treated with i.p injection of 100mg/kg bw of quercetin followed by CdF2 (2 mg/kg bw) for 48 h. Administration of quercetin two hours before CdF2 significantly reduced the biochemical alterations in reduced glutathione, ascorbic acid, lipid peroxidation, super oxide dismutase, catalase and total protein (p<0.05). Histopathology also showed the protective effect of quercetin. The livers treated with CdF2 were atrophic, markedly nodular, inflamed and necrotic. However, this effect was reduced to a minimum in the mice pre-treated for two hours with quercetin.

Introduction and objective. Data on the possible role of peritoneal fluid free radical-mediated oxidative damage in the pathogenesis of endometriosis still remains inconsistent. The aim of the study was to determine iron metabolism markers and their influence on oxidative stress parameters in the peritoneal fluid of women with endometriosis. Materials and method. 110 women with endometriosis and 119 patients with benign ovarian cysts were included in the study. All visible peritoneal fluid was aspirated during laparoscopy from the anterior and posterior cul-de-sacs. under direct vision to avoid blood contamination. Haemoglobin, iron, total oxidative status, and total antioxidant status were measured using standard colourimetric kits. Results. Haemoglobin, iron levels, as well as total oxidative status values were significantly higher, whereas total antioxidant status values were significantly lower in the peritoneal fluid of patients with endometriosis, in comparison to the reference groups. No differences were observed in peritoneal fluid concentrations of all parameters measured in relation to the phase of the menstrual cycle. Conclusions. Peritoneal fluid of women with endometriosis is characterized by disrupted iron metabolism. This is most likely related to an increased number of erythrocytes in the peritoneal cavity of endometriotic women, which leads to a higher concentration of haemoglobin in this environment. Impaired iron homeostasis may have a significant influence on the pathophysiology of peritoneal endometriosis by the direct impact of haemoglobin derivatives and/or formation of the pro-inflammatory and pro-oxidative environment. Peritoneal cavity oxidative stress occurs predominantly in women in advanced stages of the disease.

Lithium is one of the most widely used medications for the treatment of bipolar disorder (BD). It also has some side effects on thyroid functions. We aimed to investigate the role of oxidative stress, trace elements, and hemorheological parameters on the pathophysiology of thyroid dysfunctions developed by lithium treatment in patients with BD. Patients with BD were divided into three groups: patients that non-lithium-treated, lithium-treated patients for 4-6 weeks, and lithium-treated patients for 40-68 weeks. Blood samples for analysis were taken before and after the treatment period. After analysis, patients were divided into six groups: non-treatment BD group (Group 1); short-term lithium-treatment group that did not develop thyroid dysfunctions (Group 2); short-term lithium-treatment group that developed hyperthyroidism (Group 3); longterm lithium treatment group that developed hypothyroidism (Group 4), long-term lithium-treatment group that developed hyperthyroidism (Group 5), and long-term lithium-treatment group that did not develop thyroid dysfunctions (Group 6). Plasma and whole blood viscosity levels were significantly increased in Groups 4 and 6 compared to Groups 1, 2, and 3. Hemoglobin levels were lower in Group 4 than in Groups 1, 2, and 5. Fibrinogen values were higher in Groups 4 and 5 than Group 1. Plasma and erythrocyte malondialdehyde levels were higher in Group 4 than In Groups 1, 2, 3, and 5. Also, they were increased in Group 6 in comparison with Groups 2 and 3. Erythrocyte glutathione levels were lower in Groups 4 and 6 than Groups 1, 2, 3 and 5. Plasma protein carbonyls levels were higher in Group 4 than in Group 1, or in Group 5 than in Groups 1, 2, and 3, as well as in Group 6 than Groups 1, and 2. Serum zinc levels were higher in Groups 2, 3 and 6 than in Group1. Serum copper levels increased in Groups 2, 4 and 6 in comparison with Group1. The results of this study indicate that oxidative stress increased with treatment time in lithium-induced thyroid dysfunctions. Also, whole blood viscosity, plasma viscosity, fibrinogen, zinc, and copper levels were affected by lithium treatment and treatment duration induced thyroid dysfunctions.

An extensively used fluoroquinolone antibiotic ciprofloxacin shows a broad-spectrum antibacterial activity against both gram-positive and gram-negative strains. It works mainly by the inhibition of DNA gyrase and topoisomerase IV which results in impaired DNA replication leading towards microbial cell death. Eryptosis is an alternative term used for suicidal erythrocyte death. In current study, eryptotic effect of ciprofloxacin was investigated by exposing erythrocytes for 48 hours to the different concentrations (45-90µM) of ciprofloxacin. The experimental work related to the investigation of eryptosis was done by cell size measurement and confirmation of calcium role in membrane blebbing. As a possible mechanism of eryptosis, oxidative stress induced by ciprofloxacin was determined by catalase, glutathion peroxidase and superoxide dismutase activities measurement. Similarly, necrotic effect of ciprofloxacin was also illustrated by hemolysis measurement. Results of our study revealed that the therapeutical doses of ciprofloxacin may induce oxidative stress by significant decrease in superoxide dismutase, catalase and glutathione peroxidase activities as well as induce eryptosis, featured by erythrocytes membrane blebbing and hemolysis.

Objectives: To study any possible correlation between blood lead levels and some oxidative stress parameters of selected groups of lead-exposed automobile occupational Jordanian workers. Material and Methods: Blood lead levels were determined for a total of 90 male automobile workers aged within the range of 25–45 years old along with the group of 20 agematched healthy males control. To get an idea about the antioxidant status of controls and lead-exposed workers, and to estimate the oxidative stress caused by exposure to lead, we measured the hydrogen peroxide (H2O2) concentration, superoxide dismutase (SOD) activity, lipid peroxidation as malondialdehyde (MDA) and total antioxidant capacity of controls and workers. Results: The study showed that lead levels in the case of workers were approximately 4–5 times as high as in controls 14.5–21 μg×dl–1 vs. 4.3 μg×dl–1, respectively. There was a significant decrease (16–25%) in the level of plasma reduced glutathione and 21–33% decrease in total antioxidant capacity (TAOC) in all worker groups, as compared to controls. The results showed that the concentration of malondialdehyde (MDA) in plasma was higher (120–333%) in the case of workers than controls being the highest in automobile electronics and the lowest – in mechanics. Furthermore, there was 149–221% increase in hydrogen peroxide (H2O2) concentration, and 26–38% increase in SOD activity in the case of workers compared to the control group. Conclusions: There is a strong evidence for the associations between occupational lead exposure and various markers of oxidative stress in Jordanian automobile occupational workers. Thus, there is an urgent need to raise awareness and to initiate suitable protection guidelines for workers. Int J Occup Med Environ Health 2018;31(4):517–525

Opis:
Flavonoids exhibit prooxidant cytotoxicity in mammalian cells due to the formation of free radicals and oxidation products possessing quinone or quinomethide structure. However, it is unclear how the cytotoxicity of flavonoids depends on the ease of their single-electron oxidation in aqueous medium, i.e., the redox potential of the phenoxyl radical/phenol couple. We verified the previously calculated redox potentials for several flavonoids according to their rates of reduction of cytochrome c and ferricyanide, and proposed experimentally-based values of redox potentials for myricetin, fisetin, morin, kaempferol, galangin, and naringenin. We found that the cytotoxicity of flavonoids (n=10) in bovine leukemia virus-transformed lamb kidney fibroblasts (line FLK) and murine hepatoma (line MH-22a) increases with a decrease in their redox potential of the phenoxyl radical/phenol couple and an increase in their lipophilicity. Their cytotoxicity was decreased by antioxidants and inhibitors of cytochromes P-450, α-naphthoflavone and isoniazide, and increased by an inhibitor of catechol-O-methyltransferase, 3,5-dinitrocatechol. It shows that although the prooxidant action of flavonoids may be the main factor in their cytotoxicity, the hydroxylation and oxidative demethylation by cytochromes P-450 and O-methylation by catechol-O-methyltransferase can significantly modulate the cytotoxicity of the parent compounds.

In order to characterize the possible mechanism(s) of cytotoxicity of a neuroleptic agent 6,7-dinitrodihydroquinoxaline-2,3-dione (DNQX) we examined the redox properties of DNQX, and its mononitro- (NQX) and denitro- (QX) derivatives. The irreversible electrochemical reduction of the nitro groups of DNQX was characterized by the reduction peak potentials (Ep,7) of -0.43 V and -0.72 V vs. Ag/AgCl at pH 7.0, whereas NQX was reduced at Ep,7 = -0.67 V. The reactivities of DNQX and NQX towards the single-electron transferring enzymes NADPH:cytochrome P-450 reductase and NADPH:adrenodoxin reductase/adrenodoxin complex were similar to those of model nitrobenzenes with the single-electron reduction potential (E17) values of -0.29 V - -0.42 V. DNQX and NQX also acted as substrates for two-electron transferring mammalian NAD(P)H:quinone oxidoreductase (DT-diaphorase). The cytotoxicity of DNQX in bovine leukemia virus-transformed lamb kidney fibroblasts (line FLK) was prevented by antioxidants and an inhibitor of NQO1, dicoumarol, and was enhanced by the prooxidant alkylating agent 1,3-bis(2-chloromethyl)-1-nitrosourea. A comparison with model nitrobenzene compounds shows that the cytotoxicity of DNQX and NQX reasonably agrees with the ease of their electrochemical reduction, and/or their reactivities towards the used enzymatic single-electron reducing systems. Thus, our data imply that the cytotoxicity of DNQX in FLK cells is exerted mainly through oxidative stress.
