Opis:
Scanning electron microscopy (SEM) is a perfect technique for micro-/nano-object imaging [1] and movement measurement [2, 3] both in high and environmental vacuum conditions and at various temperatures ranging from elevated to low temperatures. In our view, the magnetic field expanding from the pole-piece makes it possible to characterize the behaviour of electromagnetic micro- and nano-electromechanical systems (MEMS/NEMS) in which the deflection of the movable part is controlled by the electromagnetic force. What must be determined, however, is the magnetic field expanding from the e-beam column, which is a function of many factors, like working distance (WD), magnification and position of the device in relation to the e-beam column. There are only a few experimental methods for determination of the magnetic field in a scanning electron microscope. In this paper we present a method of the magnetic field determination under the scanning electron column by application of a silicon cantilever magnetometer. The micro-cantilever magnetometer is a silicon micro-fabricated MEMS electromagnetic device integrating a current loop of lithographically defined dimensions. Its stiffness can be calibrated with a precision of 5% by the method described by Majstrzyk et al. [4]. The deflection of the magnetometer cantilever is measured with a scanning electron microscope and thus, through knowing the bias current, it is possible to determine the magnetic field generated by the e-beam column in a defined position and at a defined magnification.

Opis:
Introduction: Knowledge about the physiology of a healthy middle ear is essential for understanding the activity and mechanics of the ear as well as the basics of ossiculoplasty. Trauma of the epithelial lining of the tympanic cavity as well as the ossicular chain may be the result of chronic inflammation and surgery. Depending on the observed changes of the middle ear lining, there are several types of distinguished chronic inflammatory changes: simple, with cholesteatoma, with the formation of inflammatory granulation tissue, in course of specific diseases. Purpose: The aim of the article is presentation of the microstructure and vasculature of the ossicular chain in the Scanning Electron Microscope. Particular attention is drawn to the anatomical aspects of the structure and connections of auditory ossicles as vital elements for reconstruction of the conduction system of the middle ear. Material and method: The analysis covered auditory ossicles standardly removed in accordance with the methodology of the investigated surgical procedures. The preparations were evaluated in a scanning electron microscope. Results: The exposure of bone surface promotes deep erosion. The advanced process of destruction of bone surface in the case of chronic otitis media correlates with a significant degree of damage to both the lining covering the auditory ossicles and that surrounding articular surfaces. Conclusions: (1) The ossicles in the image of the Scanning Electron Microscope are covered with lining. It passes from the surface of the ossicles to the vascular bundles, forming vascular sheaths; (2) Damage to lining continuity on the surface of the auditory ossicles promotes the rapid destruction of bone tissue in the inflammatory process; (3) The dimensions of the individual ossicles are respectively: malleus – 8.36 +/- 0.01, incus – 8.14 +/- 0.0, stapes – 3.23 +/- 0.01 mm. Behavior of the anatomical length of ossicular chain during tympanoplasty appears to be essential to maintaining adequate vibration amplitude of the conductive system of the middle ear.

Opis:
The process of vitrification of the cumulus-oocyte complex (COCs) often results in cold shock. When warming, heat shock occurs which can disrupt the balance of intracellular calcium (Ca2+) intensity. Drastic changes in temperature cause Reactive Oxygen Species (ROS), affecting changes on Ca2+ in COCs. The role of calcium is needed for oocyte activation in the fertilization process. The purpose of this study was to measure the expression of Ca2+ and the intensity of Ca2+ in COCs after vitrification. The study was divided into 2 groups, the control group (C) of fresh COCs, and the treatment group (T) of COCs after vitrification. After vitrification for 24 hours, then thawing, the expression of Ca2+ was examined using the Immunocytochemistry (ICC) method and the intensity of calcium (Ca2+) with a Confocal Laser Scanning Microscope (CLSM). The research data obtained were analyzed statistically by T-Test. The results showed that the expression of Ca2+ in the control group (12.00±0.00) was different from the treatment group (0.35±0.79). The intensity of Ca2+ in the control group (1059.43±489.59) was different from the treatment group (568.21±84.31). The conclusion of this study is that cryopreservation affects calcium in COCs; there were differences in the expression and the intensity of Ca2+ between fresh COCs and COCs after vitrification. Ca2+ intensity of COCs after vitrification was concentrated in the nucleus, while in fresh COCs it was concentrated in the cytoplasm.

Introduction: Chronic purulent cholesteatoma of the middle ear, as well as to a lesser extent chronic granulomatous otitis media, lead to destruction of bone structures within the middle ear space. The above process is controlled by the OPG/RANKL/RANK system. Aim: The aim of the study is a comparative analysis of surgical material obtained from patients with diagnosed chronić cholesteatoma and chronic otitis media with inflammatory granulation in the assessment of the Scanning Electron Microscope (SEM). Material and methods: An analysis of 140 patients operated on due to chronic otitis media was performed. Forty patients who had been diagnosed with chronic cholesteatoma of the middle ear and chronic granulomatous otitis media were selected for a detailed analysis in the SEM. The final study under SEM included 20 patients. Results: The regular structure of cholesteatoma depicted in the SEM concerned 5 patients. In the remaining 7 patients, the system was irregular and even chaotic. The lack of regularity can also be observed in the case of granulation tissue, which in the SEM image presented itself as an irregular tissue mass without detectable regularities. Conclusions: (1) The regular pattern of the cholesteatoma matrix cells observed in some patients with chronic cholesteatoma of the middle ear reduces the molecular permeability of inflammatory cytokines, concurrently limiting the destructive activity on bone structures; (2) the presence of inflammatory granulation tissue in the middle ear is accompanied by an influx of leukocytes: neutrophils and lymphocytes, which are the source of pro-inflammatory cytokines, the growth of which activates the processes leading to the damage of bone tissue and the development of inflammation; (3) no specimen of acquired cholesteatoma revealed the presence of commensal organisms from Demodex species on the surface of the exfoliated human epithelium.

Opis:
At present, Al-Si-Cu based alloys (with a typical representative AlSi9Cu3 alloy) represent more than half of the castings used in various industries (automotive, aerospace and electrical engineering). These are most often sub-eutectic (exceptionally eutectic) alloys with a content of 6 to 13 wt. % Si and 1 to 5 wt. % Cu. The aim of the paper is to point out the importance of the evaluation of input raw materials that determines the overall properties of the casting and the costs invested in its production. A negative impact on performance can be expected when using an alloy made up of a high proportion of recycled material, despite its economic benefits. Experimental alloys were evaluated based on the results of crystallization process and a combination of scanning electron microscopy (SEM), energy dispersive X-ray analysis (EDX), and deep etching. The effect of remelting and increasing the remelted returnable material in the batch was manifested especially in the crystallization of iron-rich phases. The negative effect of remelting on the structural components was manifested after the fourth remelting. Gradual increase of remelted returnable material in the batch causes harmful changes in the crystallization process.

The acceleration of the dynamic microobjects (MO) movement, the motion of which is accelerated uniformly or non-uniformly is an important task for television measuring systems. Most existing measuring systems usually have a small range of acceleration with a short period of growth. Such period of growth of the acceleration is characteristic for many dynamic MO. Better instruments to determine the non-uniform velocity of the various objects are based on various laser devices. These instruments are contactless and high-speed enough. In the paper the basics of the television scanning optical microscope (TSOM) are presented. In the Chapter 2 the schematic diagram of the TSOM and its main functionalities are pointed out. In the Chapter 3 an adaptation of the TSOM for movement acceleration of microobjects is discussed. Modified structural and functional diagram of the instrument is presented.

Opis:
An experimental-theoretical investigation have been presented to exploring phenomena of mirror effect. The Polyethylene Terephthalate (PET) material is chosen to be a case study throughout this work. Where the scanning electron microscope (SEM) is used for executing the experiments. Attention has been focused on determination of the number of electrons that accumulated at PET surface, since it is regarded to be the most important factor for producing mirror effect images. Thus two different procedures are produced to achieve such a task namely Magnification Factor Method and Disappearing Method. Results obtained from these two approaches have clearly shown that mirror effects can accurately be used as an excellent tool to determine dielectric constant of insulator.

Opis:
The microstructural evaluation of complex cementitious materials has been made possible by the microscopic imaging tools such as Scanning Electron Microscope (SEM) and X-Ray Microanalysis. Particularly, the application of concrete SEM imaging and digital image analysis have become common in the analysis and mapping of concrete technology. In this study, six samples of two-dimensional (2D) SEM images were spatially resampled to produce Geo-referenced SEM sample images. Subsequently, they were analyzed and the intensity histogram plot was produced to facilitate visual interpretation. The consecutive digital image analysis performed was the enhancement and noise removal process using two filtering methods i.e. median and adaptive box filter. The filtered resampled images, then undergone the unsupervised K-Means classification process to collectively separate each individual pixel corresponds to the spectral data. By spatial segmentation of K-Means algorithms, the cluster groups generated were carefully reviewed before proceeding to the final analysis. From the resulting data, the mapping of the spatial distribution of k-cluster and the quantification of micro-cracks (voids) were performed. The results of the SEM images (1st - 4th sample) showed a higher percentage of k-cluster data indicating a good correlation with the major elemental composition of EDX analysis, namely Oxide (O), Silicon (Si) and Carbon (C). Meanwhile, the subjective visual assessment of the image (5th and 6th sample) has confirmed the micro-crack developments on the concrete SEM images upon which the crack density was 3.02 % and 1.30 %, respectively.

The performed studies focused on the surface structure of floral nectaries of four species from the following genus: C. coccinea L , C. crus-galli L, C. curvisepala Lindm and C. prunifolia (Poiret) Pers. The observations of the epidermis area were made in a scanning electron microscope (SEM). A nectary appears to be shaped like a slightly curved disk situated between the pistil style and the basal part of the stamens filaments. The nectary area of the studied species differed substantially as regards the cuticle sculpture and stomata number. The nectary secretion in Crataegus flowers proceeds through the stomata located below a level of the other epidermis cells, in the deep indents of the secretory tissue. The highest stomata number in 1 mm² nectary epidermis was recorded in C. crus-galli, C. coccínea, C. prunifolia and finally, C. curvisepala. Analyzing the nectary cuticular structure in respect of its increasing complexity (absence or presence of stripes), the investigated taxons can be ordered as following: Crataegus curvisepala, C. coccinea, C. crus-galli and C. prunifolia.

In this paper, an attempt was made to use Prosopis juliflora fibres (PJFs) as a reinforcing agent for phenol formaldehyde (PF) composites. Mechanical properties of the composites were studied for various fibre aspect ratios (FAR) and fibre loadings (FL). A scanning electron microscope (SEM) was used to study the fractured surface of the composites. The peak range of mechanical properties was identified for composites with a FAR of 136 and fibre loading of 23.53 wt%. This study shows that the optimum FAR and fibre loading for PJFs were found to be 136 and 23.53 wt% in order to achieve good reinforcement with better mechanical properties in the PF resin matrix. Experimental results were observed to be in very good agreement with the theoretical.
