The proteins level and activities of acid and alkaline proteases in whole body extracts of drone prepupae of Apis mellifera naturaly infested with Varroa destructor were studied. The infested and a non-infested group did not differ significantly in their total protein content. However, some differences in protein profiles were found. A lack of three protein fractions of moderate and lower molecular weight in infested prepupae was noted. Moreover, some differences in the quantity of protein in most of the fractions were observed. The activity of acid proteases from infested prepupae was lower (p < 0.05) compared with the activity of these proteases from the non-infested one group. The infested drone had higher activity of alkaline proteases than non-infested but this difference was not statisticaly significant.

Opis:
Celem przeprowadzonych badań w doświadczeniu modelowym na glebie płowej wytworzonej z piasku gliniastego mocnego było poznanie wpływu dawki 500 kg S ha-1 r-1 na aktywność proteolityczną, urolityczną, nasilenie amonifikacji i nitryfikacji. Przed wprowadzeniem dawki kwaśnego opadu glebę wzbogacono gnojowicą (60 m3 ha-1 r-1). Przeprowadzone badania wykazały, że najwyższą aktywnością proteolityczną charakteryzowała się gleba wzbogacona gnojowicą z równoczesnym zasiarczeniem. Zasiarczenie gleby oraz wzbogacenie gnojowicą również wpływało stymulująco na aktywność proteolityczną. Na aktywność ureazową gleby największy istotny wpływ miała gnojowica oraz, gnojowica z równoczesnym zasiarczeniem. Na nasilenie procesu amonifikacji hamujący wpływ miało zastosowane zasiarczenie i gnojowica. Natomiast na proces nitryfikacji istotnie stymulujące wpływały zarówno nawożenie w postaci gnojowicy jak i zastosowana dawka kwaśnego opadu.
The experiment was set on lessive soil developed from heavy loamy sand.The aim of study was to evaluate the effect of 500 kg S ha-1 year -1 dose on proteolytical and urolytical activities, as well as intensification of amonification and nitrification processes. Soil was amended with liquid manure (60 m3 ha-1year-1)before acidic rainfall dose application.Studies revealed that soil amended with the liquid manure along with sulfating was characterized with the highest proteolytical activity. Soil sulfating and liquid manure amendment also stimulated the proteolytical activity.Soil sulfating and liquid manure amendment also stimulated the proteolytical activity. Liquid manure as well as liquid manure along with sulfating had the highest impact on urease activity. Sulfating and liquid manure applied inhibited the intensification of amonification process. However, nitrification process was stimulated by both liquid manure fertilization and acidic rainfall dose applied.

The larvae of Anisakis simplex had the largest influence upon decreasing the activity of porcine pepsin. The activity of that enzyme in tests, where the larvae were present during the entire period of incubation, was lower than in the controls. No similar trends were observed in case of the solutions with bovine and porcine trypsin. The activity of those enzymes in the solutions containing the larvae was higher than in the controls. Only the activity of porcine trypsin after 10 h of incubation was slightly lower in the experimental sample than in the control, however, during the later hours the dynamics of the activity decrease of that enzyme in the controls was higher than in the experimental samples. The recorded activity of papain in the samples containing the larvae was higher than that in the controls during the entire time of the experiment.

Opis:
Introduction: The main protease (Mpro) and the papain-like protease (PLpro) are essential for the replication of SARS-CoV-2. Both proteases can be targets for drugs acting against SARS-CoV-2. Objective: This paper aims to investigate the in silico activity of nine xanthophylls as inhibitors of Mpro and PLpro. Methods: The structures of Mpro (PDB-ID: 6LU7) and PLpro (PDB-ID: 6W9C) were obtained from RCSB Protein Data Bank and developed with BIOVIA Discovery Studio. Active sites of proteins were performed using CASTp. For docking the PyRx was used. Pharmacokinetic parameters of ADMET were evaluated using SwissADME and pkCSM. Results: β-cryptoxanthin exhibited the highest binding energy: –7.4 kcal/mol in the active site of Mpro. In PLpro active site, the highest binding energy had canthaxanthin of –9.4 kcal/mol, astaxanthin –9.3 kcal/mol, flavoxanthin –9.2 kcal/mol and violaxanthin –9.2 kcal/mol. ADMET studies presented lower toxicity of xanthophylls in comparison to ritonavir and ivermectin. Conclusion: Our findings suggest that xanthophylls can be used as potential inhibitors against SARS-CoV-2 main protease and papain-like protease.

Bacteria immobilized in carrageenan are widely used in industry to facilitate bacterial handling and storage. Carrageenan is derived from seaweed and its nature is influenced by the condition of the origin of the sea where seaweed grows, one of the Indonesia sea territories that has seaweed that contains caraganen with good properties is Maluku. This study was conducted to determine the effect of storage time of bacteria immobilized in Maluku sea’s carrageenan on proteolytic activity, the bacteria used were Bacillus megaterium. Bacterial immobilization of carrageenan was made at concentrations of 1%, 1.5%, and 2%, storage in cold conditions for up to 9 months. Protease activity was tested using Kunitz method by adding casein as a substrate. The optimal concentration of carrageenan for immobilization of Bacillus megaterium was obtained at a concentration of 1.5%. Protease isolated from immobilized Bacillus megaterium showed increased activity value from storage for 4 months (0.0489 Ug-1) to 7 months (0.1372 Ug-1), and decreased activity after being stored for 9 months (0.0501 Ug-1).

The enzymatic activity (chitinolytic, proteolytic, pectolytic andcellulolytic) of twenty strains of Streptomyces isolated from soil and rhizosphere of Scots pine was analyzed. Most strains produced chitinases, catalyzing the degradation of chitin, the main component of fungal cell walls including fungi pathogenic for plants. This activity was about 4 times higher in the presence of Fusariumoxysporum than Rhizoctonia solani mycelium or chitin flakes. The number of the proteolytic strains was also significant. In general, rhizosphere and soil organisms synthesizedcomparable amounts of these enzymes. Over half of the analyzed Streptomyces strains produced pectolytic enzymes (polygalacturonase, pectin lyase and pectate lyase). This property was more common among rhizosphere than among soil strains. The Streptomyces strains also showedcellulolytic activity (endocellulases, exocellulases) – enzymes decomposing basic components of cell walls of plant and lower fungi (cellulose). The cellulolytic activity was differentiated and depended on the Streptomyces strain. Conclusion of our studies is that Streptomyces are the microorganisms more chitinolytic andproteolytic than pectolytic and cellulolytic.

The study presented in this paper was conducted under the conditions of a field experiment. Microbiological analyses were made at various stages of winter wheat plants developmentie heading, milk ripeness and full ripeness. The objective of the study was to acquire knowledge on the effect of cultivation of various lines of winter wheat on the numbers of bacteria and fungi with proteolytic capabilities, on protease and urease activity, and on the rate of the processes of ammonification and nitrification. The results of conducted study demonstrated that the number of proteolytic bacteria and fungi, as well as the activity of protease and urease, and the intensity of ammonification and nitrification processes in soil depended on both the development stage and cultivated line of winter wheat.

Chloroplast protease AtDeg2 (an ATP-independent serine endopeptidase) is cytosolically synthesized as a precursor, which is imported into the chloroplast stroma and deprived of its transit peptide. Then the mature protein undergoes routing to its functional location at the stromal side of thylakoid membrane. In its linear structure AtDeg2 molecule contains the protease domain with catalytic triad (HDS) and two PDZ domains (PDZ1 and PDZ2). In vivo AtDeg2 most probably exists as a supposedly inactive haxamer, which may change its oligomeric stage to form active 12-mer, or 24-mer. AtDeg2 has recently been demonstrated to exhibit dual protease/chaperone function. This review is focused on the current awareness with regard to AtDeg2 structure and functional significance.

Badania realizowano w glebie bielicowej wytworzonej z piasku luźnego, która cechuje się niewielką żyznością oraz w czarnej ziemi wytworzonej z piasku słabo gliniastego, która posiada dużą ilość części spławialnych, wysoki poziom węgla i azotu organicznego oraz znaczną pojemność sorpcyjną. Gleby te występują w sektorze rolniczym zlewni dwu jezior - Piaseczno i Głębokie (Pojezierze Łęczyńsko-Włodawskie). Próby do analiz biochemicznych pobierano 7, różnych stanowisk czterokrotnie w następujących terminach: kwiecień, maj, lipiec i wrzesień. Aktywność proteazy w glebach badano metodą Ladda i Butlera. Natomiast nasilenie procesu amonifikacji określano na podstawie przyrostu zawartości N-NH4+ (w próbkach glebowych inkubowanych z 0,1 % asparaginy) oznaczonego metodą Nesslera. Przeprowadzone badania wykazały, że aktywność proteolityczna zależała od właściwości gleby. W glebie bielicowej kształtowała się na ogół na niższym poziomie w porównaniu z czarną ziemią i była zróżnicowana w zależności od odległości stanowiska od jeziora. Potencjalna zdolność amonifikacyjna gleb nie była wyraźnie powiązana z ich właściwościami. W glebie bielicowej utrzymywała się w całym sektorze na zbliżonym poziomie. Natomiast w przypadku czarnej ziemi stwierdzono zależność badanego procesu od punktu pobrania gleby. Najwyższą wartość tej aktywności odnotowano w glebie zlokalizowanej na brzegu jeziora.
The investigations were carried out in podzolic soil of poor fertility developed from loose sand and black soil formed from slightly loamy sand with great part of fine fraction, high level of carbon and organic nitrogen and considerable sorp tivity. The soils arc located in the agricultural sector of the two lakes basin - Piaseczno and Głębokie (the Łęczyńsko-Włodawskie Lake District),The samples for biochemical analyses were gathered from different test sites for four times in the following terms: April, May, July and September, The protease activity in soils were examined after Ladd and Butler's method. Intensification of ammonification process was determined on die basis of N-NH4+ content growth (in soil samples incubated with 0,1 % asparagine) determined by Nesslef s method. The examinations proved the proteolytic activity depended on soil properties. In podzolic soil it demonstrated a lower level as compared to black soil and was differentiated in relation to test site distance off a lake. Potential ammonification activity of soils was not clearly connected with their characteristics. In podzolic .soil it maintained a similar level all over the agricultural sector, while in black soil there was stated dependence of a process examined on a sampling point. The highest value of the activity was recorded in the soil located on the lake bank.
