Opis:
Vacuoles play very important physiological roles in plant cells. Pea root nodules, which exhibit distinct zonation (meristematic zone and central tissue zones), may serve as a good experimental model for the investigations of vacuole development and its importance to cell and tissue functioning. Moreover, the nodule central tissue is composed of both infected and uninfected cells which play different physiological roles and differ in the level of vacuolation. Cytological observations revealed that central vacuoles of the infected cells of the effective nodules expand toward cell walls. Thus only thin layers of the cytoplasm separate each central vacuole from plasma membrane and cell wall. This finding is discussed from the viewpoint of improved exchange of solutes and water between the central vacuole and apoplast of the infected cell. Three-dimensional reconstruction of the vacuoles of infected cells within a fragment of effective nodule central tissue, showed their spatial arrangement. Possible advantages coming from the spatial arrangement of vacuoles within the central tissue are discussed. A comparative study of the central tissue (bacteroidal tissue) and meristem vacuolation of the effective and ineffective pea root nodules is also presented. Morphometric measurements revealed that the effective nodule central tissue was more vacuolated than the ineffective one. It was proved that maturation of the infected cells involves dynamic changes in their vacuolation. Having numerous fixing nitrogen bacteroids, the infected cells of effective central tissue were less vacuolated than uninfected cells. On the other hand, both infected and uninfected cells of the effective central tissue showed a much higher level of vacuolation in nitrogen-fixing zone than cells of the same type in ineffective tissue. These results indicate that vacuolation is an important factor in development and functioning of pea root nodule central tissue.

An experiment aimed to review differences in the pattern of canopy forming, solar radiation penetration and its use efficiency (RUE) within the canopy of two contrasting pea morphotypes (leafy and semi-leafless) was conducted in the field. The differences in absorption of PAR were significant for both morphotypes in two consecutive years of different climatic conditions. The canopy architecture influenced absorption of PAR, penetration through the canopy and can cresult in different yield forming. For both morphotypes a common linear correlation between intercepted PAR and dry matter accumulation was found until the beginning of pod formation, but then this association was not linear. Differences in CGR and ULRc were significantly bigger in leafy pea morphotypes than in semi-leafless one and the higher values for both were observed at the beginning of pod formation.

The present study points to an important role of citric acid in limiting toxicity of aluminium ions for pea plants which was confirmed by the plants’ growth, development and yield. Excess of aluminium influenced plant nutrition with Ca, P, Mg, and K as this toxic element did not only gather in the roots of the studied plants but also in their above-ground parts. Limitation of aluminium toxicity by its complexing with citric acid clearly influenced a increase in the Fe uptake by plants and an decrease in the Mn uptake which was statistically proved.

Opis:
Vegetables are considered to be plants with high water requirements. A greater susceptibility to water deficit occurs in the periods of intensive growth and yield formation. At that time even a slight reduction in water supply can deteriorate yielding, thus those periods are referred to as critical periods in terms of water supply. The aim of this paper has been to analyse the pattern of precipitation deficits for the ground cultivation of cucumber, tomato, beans and peas in the Bydgoszcz region in the thirty-year period 1981-2010. The basic of analysis were results of standard meteorological measurements from the Research Station of the Faculty of Agriculture and Biotechnology ‚Mochełek’, University of Technology and Life Sciences. Values of optimal precipitation for medium soils were calculated according to Klatt. Atmospheric precipitation deficits in subsequent month of selected vegetables cultivation were calculated by diminish between the real and optimal precipitation. The trends of deficiency of precipitation in the examined period were estimated too. Very frequent occurrence of the atmospheric precipitation deficits during the entire cultivation period of selected vegetables, and especially in critical periods was demonstrated. Slight decreasing tendency of precipitation deficits in a vegetation periods was indicated, however in a critical periods in terms of water supply, the trend line remains at a practically constant level. The results obtained indicate the necessity of the irrigation in the cultivation of peas, beans, cucumbers and tomatoes.

Phenolic compounds were extracted from pea (Pisum sativum) seeds using 80% aqueous acetone. Crude extract was applied onto a Sephadex LH-20 column. Fraction I of low molecular phenolic compounds was eluted from the column by ethanol. Fraction II of tannins was obtained using water-acetone (1:1; v/v) as mobile phases. Phenolic compounds present in extract and its fractions showed antioxidant and antiradical properties investigated using a β-carotene-linoleate model system, Total Antioxidant Activity (TAA) method, DPPH radical scavenging activity and reducing power. The results of assays performed were the highest when tannins (fraction II) were tested. For example, TAA of tannin fraction was 2.48 µmol Trolox/mg, whereas the extract and fraction I showed only 0.30 and 0.22 µmol Trolox/mg, respectively. The content of total phenolics in fraction II was found the highest (113 mg/g). The content of tannins in this fraction determined using the vanillin method and expressed as absorbance units at 500 nm per 1 g was 368. The HPLC analysis of pea crude extract showed the presence of such phenolic compounds as vanillic, caffeic, p-coumaric, ferulic and sinapic acids (after basic hydrolysis), quercetin and kaempherol, procyanidin B2 and B3.

Opis:
Increasing the production of high-quality agricultural products with minimal negative impact on environmental components is one of the priority tasks of modern agriculture. Due to the global world trends aimed at ensuring a sustainable and safe economy and reducing the use of chemical protection agents, it is predicted that the share of alternative biological methods in the European agricultural sector will increase to 10%, the area of agricultural land for organic farming will increase by 50%. Under the conditions of intensification of agricultural production, a comprehensive approach to the use of microelements and microbial biological preparations, which enhance growth processes, anti-stress, protective effect and increase the yield of agricultural crops, becomes expedient. The article deals with the effects of biological growth-regulating preparations on winter wheat, sunflower hybrids, and vegetable peas. It was established that when processing winter wheat with the “Grainactive-C” preparation, the greatest increase in grain yield was observed under the conditions of 3-fold treatment of crops in the phase of tillering, flag leaf, and grain filling (3.2 t/ha, or 18%). At the same time, the greatest effect was recorded when treating crops in the tillering phase, where the yield increase was 2.9 c/ha, or +16% compared to the control. It has been studied that the effectiveness of the preparation “Nano-Agro” when applied to sunflower hybrids also depends on the time of its application. The same increase – 0.9 t/ha of seeds (+12% compared to the control) was observed during pre-sowing treatment of seeds and during the vegetation phase (before flowering), but the yield increase when treating sunflower after flowering phase was only 0.32 t/ha (4%). In order to restore disturbed soils and increase the efficiency of agricultural production, the introduction of leguminous crops into the crop rotation becomes important, after harvesting them up to 80 kg/ha of biological nitrogen remains and humus accumulates in the arable layer of the soil up to 10.8 t/ha. Treatment of vegetable peas seeds with Rhizohumin contributed to the accumulation of humus by 3.96 t/ha. Through field studies, the article proves the positive effect of growth-regulating preparations on agricultural crops under the conditions of the Steppe zone of Ukraine.

In view of raising prices of crude oil and due to increasing demands the need for alternative sources of bioenergy is expected to increase sharply in the coming years. Cajanus cajan Stalks is the major agricultural residue, more than 25% primary energy demands met through imports mainly in the form of crude oil and natural gas. Biofuels as a domestic renewable energy source can significantly reduce India’s dependence on foreign oil and fossil fuels. The Plant cell wall is a composite material in which cellulose, hemicelluloses & lignin are closely associated. The major constraint in successful bioconversion process is the lignin. The study was conducted for various pretreatment methods. The best is the steam pretreatment method. This method serves as best and effective pretreatment with the maximum removal of lignin and isolation of more reducing sugars and less pentose’s. The alkaline method specially NaOH treatment is used for extraction of more pentose’s in crude extract. Finally, our approach was to find out the best pretreatment method that result in release of maximum reducing sugars. Steam pretreatment serves to be best followed by saccharification by cellulose. Further, these fermentable sugars can be utilized for bioethanol production. Cajanus cajan stalk is mostly cultivated in Kalaburagi district of Karnataka for that can be used for ethanol production. Screening for isolation of S. cereviceae was carried out by using baker’s yeast and was grown in YPD media. The optimization of culture condition was carried out for growth of yeast showed optimum temperature at 30 ºC and optimum pH at 5. Optimum Cellulase enzyme activity was found to be at temperature 40 ºC and pH 4.5. The ethanol production reaction was optimized by the maximum utilization of sugars by yeast was optimized by providing hexose and with combination of xylose at temperature 30 ºC and pH 5.0 and the inoculum size was optimized at 5% volume of total volume for SSF. From the above results, it is clear that the cellulase enzyme isolated from Aspergillus niger has ability to grow and produce hexoses under conditions of optimum temperature and acidic pH. The reducing sugar and pentose’s both can be used together for ethanol production. The obtained reducing sugar and pentose’s sugar conc. were 1.025 gm/500 ml & 0.5 gm/500 ml. This is only possible by using biological enzymatic pretreatment. The resultant substrate can used for making 2-3% ethanol. Thus fermentable sugars by these methods can be utilized for bioethanol production. The ethanol produced from this agro waste is 2-3% in lab scale. Then go for pilot scale 4-5% and later for industrial scale 7-8% by treat with different pretreatment methods using SSF. This helps in increases the utilization of wastes for economic use and improves the biofuel production resources.

Opis:
Neuropeptides are regulators of critical life processes in insects, but are subject to rapid degradation by peptidases in the hemolymph (blood), tissues and gut. This limitation can be overcome via replacement of peptidase susceptible portions of the insect neuropeptides with non-natural residues or moieties to create analogs with enhanced biostability. Two neuropeptide families, the insect kinins and insectatachykinins, stimulate gut motility and Malpighian tubule fluid secretion in certain insects but unmodified members demonstrate little or no effect when fed to pea aphids (Acyrthosiphon pisum) in an artificial diet. Nonetheless, biostable analogs developed via the strategic introduction of either bulky Aib residues and/or β-amino acids demonstrate potent antifeedant and aphicidal effects when administered orally; whereas other biostable analogs are inactive. Although the precise mechanism of action has not been delineated, the activity may be associated with disruption of the physiological processes that these neuropeptides regulate in insects. The most active of the biostable insect kinin and insectatachykinin analogs show LC50 values of 0.063 nmole/μl (LT50 = 1.68 days) and 0.0085 nmole/μl (LT50 = 1.1 days), respectively; matching or exceeding the potency of some commercially available aphicides. The biostable analogs represent important leads in the development of alternative, environmentally sound aphid control agents.
