Oligochitosans obtained through degradation of macromolecules of chitosan with a high degree of deacetylation turned out to be biologically active, contributing to an increase of nitric oxide levels in Ehrlich ascites tumor (EAT) cells through inducing expression of the isoform of inducible nitric oxide synthase (iNOS) gene. An increase of NO levels in EAT cells in the presence of the investigated oligochitosans might contribute to nitrosylation of L-cysteine – an allosteric effector of the M2 isoenzyme of pyruvate kinase (PK), which switches the PK kinase activity, responsible for ATP synthesis, to the histone kinase activity that may participate in histone H1 phosphorylation. Lack of the histone activity of the PK M2 isoenzyme may contribute to decreased histone H1 phosphorylation and thus inhibit EAT cells proliferation.

We investigated the cytotoxic effect of nitric oxide (NO) on primary culture of human hematological malignant cells. Sodium nitroprusside (SNP), an NO donor, had cytotoxic effects on the cells of some patients with malignant lymphoma (ML), acute myelocytic leukemia (AML) or chronic myelomonocytic leukemia (CMMoL), but not with multiple myeloma. Cultured cells from the ML patient remained sensitive to SNP after the cells became resistant to anti-cancer drugs. In contrast, the cells from the patients with AML and CMMoL became resistant to SNP while anti-cancer drugs remained effective. In samples of the cells of the patients with ML and AML, the number of CD3 positive lymphoma cell was decreased by SNP and the number of CD33 negative cells and normal B lymphocytes (CD19 positive cells) were increased. In the cells of the patient with ML, apoptosis was induced by SNP. SNP had no effect on lymphocytes of healthy volunteers. These results suggest that SNP had an anti-tumor effect on human hematological malignant cells.

COVID-19-associated ARDS (cARDS) results from multiple pathogenetic mechanisms involving both parenchyma and circulation of the lungs. Despite authors described two pneumonia phenotypes according to lung elastance values, reported as Low and High phenotype, the evidence suggests that patients with cARDS have similar respiratory mechanics to patients with ARDS from other causes. Consequently, the proper management of patients with cARDS is the same as ARDS,consisting of protective mechanical ventilation strategy and prone position ventilation.However, the hypothesis that impaired alveolar perfusion could have a role in cARDS is interesting. Nitric oxide (NO) is a biatomic gaseous moleculeables to induce smooth muscle relaxation and vasodilation. So, inhaled nitric oxide (iNO) acts as a selective vasodilator because it only dilates vessels in areas participating in gas exchange, preserving hypoxic pulmonary vasoconstriction reflex and reducing ventilation/perfusion mismatch. With this narrative review we summarised the role of iNO for cARDS treatment to improve gas exchange. Furthermore, we briefly described its activity as an antiviral agent. In conclusion, although iNO administration can represent a valid theoretical therapeutic choice for the treatment of severe unresponsive hypoxemia in COVID-19 patients, data presented in the literature are scares and inconclusive, and several clinical trials are still ongoing. Further studies are needed to elucidate the pathogenetic mechanism of respiratory failure related to SARS-CoV-2 infection and inhaled NO role in patients with severe disease.

Introduction. The fraction of exhaled nitric oxide (FeNO) is used as a non-invasive biomarker that reflects inflammation in the airways. It is so versatile that it used to control asthma severity as well as to monitor response to treatment. However, the exact cut-off point of the nitric oxide level which allows one to make a precise diagnosis of asthma is unclear. Aim. To examine the possibility of using advanced statistical methods such as receiver operating characteristic for the analysis of FeNO concentrations for improving the diagnosis of asthma. Materials and methods. Receiver operating characteristic (ROC) was used for analyzing results to determine levels of nitric oxide which may be a prognostic indicator of asthma. The studied group consisted of 111 children including 69 asthmatic patients, and 42 age- and sex-matched healthy subjects. Measurement of exhaled nitric oxide was conducted in all subjects included in this study. Results. FeNO level was higher in asthmatic patients. The analysis of results showed that the cut-off point for the FeNO concentration is 11.5 ppb. Sensitivity and specificity with the FeNO level allowed us to determine a value of the diagnostic variable of FeNO concentration of 14.0 ppb. Investigation of the mechanisms of phenotypic realization of allelic polymorphism of the eNOS gene has shown that the level of eNOS mRNA and activity of this enzyme in platelets depends from genotype. We identified a T-786→C polymorphism in the promoter region, a variable number of tandem repeats (4a/4b) in intron 4 and the G894→T polymorphism in exon 7 of the eNOS gene in isolated human platelets. We measured eNOS mRNA in isolated platelets by reverse transcription-PCR and eNOS enzyme activity by fluorimetric detection system FCANOS-1 using diaminofluorescein diacetate (DAF-2A). It was shown that the level of eNOS mRNA is the lowest for the -786C/C promoter genotype. In exon 7 homozygotes (894T/T) the level of RNA is lower than in normal homozygotes (894G/G), but higher than in heterozygotes (894G/T). The eNOS activity in platelets is lower in carriers of the 786C/C promoter genotype than in normal homozygotes (2.1 × P=0.03), and lower comparing to heterozygotes (2.9 × P>0.05). The eNOS activity accompanying the 894T/T variant of exon 7 is also lower than in normal homozygotes (P>0.05). Regarding the polymorphism in intron 4 - the enzyme's activity is lower in carriers of the 4a/4a genotype comparing to normal homozygotes (1.7 × P>0.05) and lower than in heterozygotes (1.9 × P>0.05). These results allow one to conclude that the T-786→C polymorphism of the eNOS gene promoter most significantly affects the gene expression and eNOS activity.

The paper presents the results of research conducted on cells isolated from the epidermal layer of the skin taken from the ear of domestic swine (Sus scrofa). It was checked whether the increase in the concentration of carbon monoxide (CO) in cell culture, cause changes in their viability, and will affect the changes in the production of nitric oxide. For evaluation of the viability of cells, the MTT assay was used. The concentration of a nitric oxide metabolites (NO3/NO2), in the cell supernatants was measured using test-Measure iT High-Sensitivity Nitrite Assay Kit.

Introduction. Kidney damage during pregnancy constitutes a diagnostic and therapeutic challenge. However, it is not entirely known whether a kidney condition recognised before the pregnancy releases an organism’s response to pregnancy, or whether pregnancy itself worsens kidney function. Objective. The aim of the study was immunohistochemical evaluation of cells of kidneys of pregnant rats under the influence of nitric oxide (NO), with measurement of the immunoexpression of cellular stress markers (p-53, HSP 70). The dose of administered L-arginine (NO substrate) was approximated to that applied in obstetrics in gestosis prevention and treatment in pregnant women. Materials and method. 60 female rats used in experiment were divided into 6 groups: 3 experimental and 3 control. The females from experimental groups were administered L-arginine (40g/kg, per os) every other day starting from the seventh day or pregnancy. The animals were decapitated on the 10th, 20th day of pregnancy, and 10 days after the delivery. Kidneys taken from decapitated rats were evaluated using the immunohistochemical three step method. HSP 70 and p-53 proteins were detected. Conclusions. L-arginine increased the expression of p-53 protein – on the 10th day of pregnancy, which increased at the end of pregnancy; however, 10 days after delivery the level dropped below that observable during physiological pregnancy. Hormonal changes in physiological pregnancy cause an increase in expression of the p-53 (cell stress marker) in the epithelial cells of renal tubules, mainly at the end of pregnancy (20th day). 10 days after the delivery, this expression decreases. The expression of HSP-70 protein increases already on the 10th day of pregnancy and maintains a similar level throughout the pregnancy, but is reduced after the puerperium.

The proper functioning of the perinatal sucking reflex in calves is essential for the prevention of milk leakage into the rumen. The complex process behind its regulation is mediated at the gut level via multiple excitatory and inhibitory neurotransmitters, of which acetylcholine and nitric oxide are of fundamental importance. The aim of our study was to depict age-related alterations in the cholinergic and nitrergic innervation of the esophageal groove (EG) using immunohistochemistry and Real-Time PCR methods. We found out that the highest number of cholinergic nerve cells was present in the second trimester fetuses. From this developmental stage onward, their amount was gradually decreasing and reached the lowest value in 4-year-old cows. The same developmental pattern was observed for nitrergic nerve structures with the highest percentage of nitrergic neurons in the third trimester fetuses. Our observations prove that both neuronal populations are crucial for a proper closure of EG in calves. Therefore, their contribution to a general neuronal activity in the ENS diminishes with age as the high motility of a gastric groove is not necessarily required in older cattle.

Introduction and aim. The aim of this work is to study the parameters of the nitric oxide (NO) system in the blood of guinea pigs under the conditions of the experimental periodontitis (EP) and immobilization stress (IS) formation and to evaluate the effectiveness of thiocetam use. Material and methods. Experimental studies were performed on 50 guinea pigs (males, body weight 0.18–0.21 kg) which were divided into five groups (10 in each): the first group were intact animals as control; the second experimental group were animals with experimental periodontitis under conditions of immobilization stress (3rd day), the third group included guinea pigs with EP and IS on the 5th day of the combined model process, group IV – animals with EP and IS 15th day (without administration of thiocetam) and group V – animals on the 15th day of experiment with EP and IS after use of thiocetam. Results. As a result of this research, changes in the activity of the NO system in the blood were observed, namely an increase in the level of stable metabolites and an increase in the activity of total NO-synthase, which is accompanied by a compensatory inhibition of the L-arginine activity, and these indicators were most pronounced in the late stages of EP and IS formation. Conclusion. The use of thiocetam showed a corrective effect on the changed variables of NO metabolism in the peripheral blood of guinea pigs under the conditions of the EP and IS development.

This study was undertaken to determine whether nitric oxide (NO) can affect platelet responses through the inhibition of energy production. It was found that NO donors: S-nitroso-N-acetylpenicyllamine, SNAP, (5-50 μM) and sodium nitroprusside, SNP, (5-100 μM) inhibited collagen- and ADP-induced aggregation of porcine platelets. The corresponding IC50 values for SNAP and SNP varied from 5 to 30 μM and from 9 to 75 μM, respectively. Collagen- and thrombin-induced platelet secretion was inhibited by SNAP (IC50 = 50 μM) and by SNP (IC50 = 100 μM). SNAP (20-100 μM), SNP (10-200 μM) and collagen (20 μg/ml) stimulated glycolysis in intact platelets. The degree of glycolysis stimulation exerted by NO donors was similar to that produced by respiratory chain inhibitors (cyanide and antimycin A) or uncouplers (2,4-dinitrophenol). Neither the NO donors nor the respiratory chain blockers affected glycolysis in platelet homogenate. SNAP (20-100 μM) and SNP (50-200 μM) inhibited oxygen consumption by platelets. The effect of SNP and SNAP on glycolysis and respiration was not reduced by 1H-[1,2,4]oxadiazolo-[4,3-a]quinoxalin-1-one, a selective inhibitor of NO-stimulated guanylate cyclase. SNAP (5-100 μM) and SNP (10-300 μM) inhibited the activity of platelet cytochrome oxidase and had no effect on NADH:ubiquinone oxidoreductase and succinate dehydrogenase. Blocking of the mitochondrial energy production by antimycin A slightly affected collagen-evoked aggregation and strongly inhibited platelet secretion. The results indicate that: 1) in porcine platelets NO is able to diminish mitochondrial energy production through the inhibition of cytochrome oxidase, 2) the inhibitory effect of NO on platelet secretion (but not aggregation) can be attributed to the reduction of mitochondrial energy production.

It has been reported that concentrations of neopterin in the urine are changed according to the host immunological conditions. In the present study, we measured urinary concentration of neopterin in patients with malignant hematological disorders and investigated the relationship between urinary neopterin levels and laboratory indices for cellular immunity. Urine neopterin levels were correlated with serum sIL-2R levels in the patients with malignant lymphoma, and inversely correlated with lymphocyte reactivity with ConA in the patients with acute myelocytic leukemia. However, no significant correlation was observed between urine neopterin levels and lymphocyte reactivity with phytohemagglutinin and pokeweed mitogen, CD4/8 ratio, CD56+16+ subset or serum IFN-β levels. In the patients with malignant lymphoma, parallel changes in serum sIL-2R and urine neopterin were observed. The presented results suggest that urine neopterin levels are related to the activation of T cells in malignant lymphoma.

The results of experimental investigations on the removal of NOx from gases applying ozone as the oxidizing agent and the absorption of higher nitrogen oxides in the sodium hydroxide solutions are presented. The experiment was conducted using a pilot plant installation with the air flow rate 200 m3/h, being a prototype of a boiler flue gas duct and a FGD scrubber. It was shown that in the range of [NOref] = 50 ÷ 250 ppm the mechanism of NO ozonation depends on the molar ratio X = O3/NOref: for X ≤ 1.0 oxidation of NO to NO2 predominates and NO2 is poorly absorbed, for X >> 1.0 NO2 undergoes further oxidation to N2O5, which is efficiently absorbed in the scrubber. The stoichiometric molar ratio of complete conversion of NO into N2O5 is X = 1.5, in these studies to reach the effectiveness η ≥ 90% the molar ratio X was much higher (2.75).

Badania prowadzono na strumieniu powietrza 200 m3/h w instalacji pilotowej będącej prototypem instalacji kotłowej odprowadzenia spalin i skrubera IOS. Wykazano, że w zakresie [NOref] =50÷250 ppm mechanizm ozonizacji NO zależy od stosunku molowego X = O3/NOref: dla X ≤ 1.0 przeważa utlenianie NO do NO2 a NO2 jest słabo absorbowany, dla X >> 1,0 NO2 ulega dalszemu utlenianiu do N2O5, który jest efektywnie absorbowany w skruberze. Stosunek molowy przy całkowitej konwersji NO do N2O5 wynosi X = 1.5, w przeprowadzonych badaniach, aby osiągnąć efektywność η ≥ 90% stosunek X był dużo większy (2,75).

Objectives: For the purpose of evaluation of exhaled NO as an index of airway inflammation, we assessed changes in fractional exhaled NO (FeNO) across a work shift and its relationship with respiratory complaints. Material and Methods: Chronic and work-aggravated respiratory complaints were assessed using a questionnaire in 89 male textile workers. FeNO and spirometry were performed before and after a work shift and all the changes were registered. Results: A significant increase in FeNO after a work shift was observed. Post-shift FeNO was significantly higher among the subjects with chronic respiratory complaints. There was an obvious decrease in FVC, and FEV₁ after a work shift; however, we couldn’t find a significant relationship between changes in respiratory parameters and concentration of inhalable dusts. Conclusions: FeNO increase after a work shift along with pulmonary function decrement and higher post-shift FeNO among subjects with respiratory complaints makes across-shift FeNO a non-invasive test for assessment of airway hyper-responsiveness in textile workers.

Nitric oxide (NO) is synthesised in the vascular endothelium by nitric oxide synthase (NOS3) and is an important factor in the regulation of blood pressure. Impaired synthesis of NO due to mutations in the NOS3 gene is associated with hypertension. To date several allelic variants of the NOS3 gene have been identified and their possible linkage with hypertension investigated. We studied the distribution of genotypes and frequency of alleles of the G11T polymorphism in intron 23 of the NOS3 gene in patients with hypertension and in a control group of healthy individuals. The polymorphism was determined by PCR-RFLP analysis. The distribution of genotypes in the patients with hypertension and in the healthy individuals did not differ significantly from the values predicted from Hardy-Weinberg equilibrium for the general population. No major differences in the distribution of the G11T polymorphism in the patients and healthy individuals were found (P > 0.05).

Nitric monoxide (NO) exerts a great variety of physiological functions. L-Arginine supplies amino groups which are transformed to NO in various NO-synthase-active isoenzyme complexes. NO-synthesis is stimulated under various conditions increasing the tissue of stable NO-metabolites. The major oxidation product found is nitrite. Elevated nitrite levels were reported to exist in a variety of diseases including HIV, reperfusion injury and hypovolemic shock. Denitrifying bacteria such as Paracoccus denitrificans have a membrane bound set of cytochromes (cyt cd1, cyt bc) which were shown to be involved in nitrite reduction activities. Mammalian mitochondria have similar cytochromes which form part of the respiratory chain. Like in bacteria quinols are used as reductants of these types of cytochromes. The observation of one-e- divergence from this redox-couple to external dioxygen made us to study whether this site of the respiratory chain may also recycle nitrite back to its bioactive form NO. Thus, the aim of the present study was therefore to confirm the existence of a reductive pathway which reestablishes the existence of the bioregulator NO from its main metabolite NO2-. Our results show that respiring mitochondria readily reduce added nitrite to NO which was made visible by nitrosylation of deoxyhemoglobin. The adduct gives characteristic triplet-ESR-signals. Using inhibitors of the respiratory chain for chemical sequestration of respiratory segments we were able to identify the site where nitrite is reduced. The results confirm the ubiquinone/cyt bc1 couple as the reductant site where nitrite is recycled. The high affinity of NO to the heme-iron of cytochrome oxidase will result in an impairment of mitochondrial energy-production. "Nitrite tolerance" of angina pectoris patients using NO-donors may be explained in that way.

The paper presents an aphonia of the effects of hyperbaric exposures on endothelial capability to generate nitric oxide synthesis, expressed as a measurement of the concentration of nitrates/nitrites in the blood serum of volunteers subjected to a hyperbaric exposure. In addition the method of measurement of total concentration of superoxides was used to assess the impact of antioxidant capacity while being subjected to hyperbaric oxygenation. Results: A significantly higher concentration of nitrates/nitrites was observed following a hyperbaric exposure. Similarly, an increased concentration of total superoxides in the researched groups was observed. Conclusions: The obtained results indicate that staying in a hyperbaric environment is not insignificant for endothelial cells. However, the issue that stays open is the one concerning the time and the pressure conditions in which the exposure remains safe.
Praca przedstawia alalię wpływu ekspozycji hiperbarycznych na zdolność śródbłonka naczyń do syntezy tlenku azotu wyrażoną jako pomiar stężenia azotanów/azotynów w surowicy ochotników poddanych ekspozycji hiperbarycznej. Dodatkowo oceniano za pomocą pomiaru stężenia całkowitego stężenia ponadtlenków jaki wpływ na pojemność antyoksydacyjną ma przebywanie w hiperbarii. Wyniki: Zaobserwowano istotnie wyższe stężenie azotanów/azotynów po ekspozycji hiperbarycznej. Podobnie zaobserwowano podwyższone stężenie całkowitego stężenia nadtlenków w badanej grupie. Wnioski: Uzyskane wyniki pokazują, że przebywanie w środowisku hiperbarycznym nie jest obojętne dla komórek śródbłonka. Otwartym pozostaje jednak problem jaki czas i na jakie warunki ciśnieniowe pozostaje bezpieczny.

Opis:
Celem niniejszego opracowania jest przedstawienie roli argininy w organizmie człowieka. Arginina jest aminokwasem względnie egzogennym, co oznacza, że pomimo tego, iż jest produkowana w ludzkim ciele, to dostarczanie jej wraz z pożywieniem jest równie ważne dla zachowania homeostazy organizmu. Główne źródło argininy w diecie stanowią mięso i owoce morza. Aminokwas ten jest syntezowany w czasie cyklu mocznikowego w wątrobie, a także w nerkach. Bierze udział między innymi w wytwarzaniu tlenku azotu, który jest czynnikiem rozszerzającym naczynia. Liczne prace wskazują również na skuteczność suplementacji argininą w zaburzeniach erekcji. Udział argininy w tej reakcji przyczynił się do zainteresowania wielu badaczy. Dowiedziono, że oprócz powszechnie znanego zastosowania argininy jako leku wspomagającego wątrobę w usuwaniu z organizmu amoniaku, odpowiednia suplementacja tego aminokwasu ma pozytywny wpływ na poprawę wartości ciśnienia tętniczego indukowanego ciążą, nadciśnienia płucnego oraz wyrównanie niewydolności nerek, cukrzycy, jaskry, gojenia się ran i profilaktykę udarów mózgu. Uważa się, że stężenie argininy wpływa na rozwój chorób neurodegeneracyjnych postępujących z wiekiem. Aminokwas ten jest też środkiem chętnie stosowanym przez sportowców w celu poprawienia wydolności organizmu. Jednak przeprowadzone do tej pory badania nie potwierdzają jego pozytywnego wpływu w tym zakresie.

The paper presents results of experimental studies on removal of NOx from flue gas via NO ozonation and wet scrubbing of products of NO oxidation in NaOH solutions. The experiment was conducted in a pilot plant installation supplied with flue gas from a coal-fired boiler at the flow rate 200 m3/h. The initial mole fraction of NOx,ref in flue gas was approx. 220 ppm, the molar ratio X = O3/NOref varied between 0 and 2.5. Ozone (O3 content 1-5% in oxygen) was injected into the flue gas channel before the wet scrubber. The effect of the mole ratio X, the NaOH concentration in the absorbent, the liquid-to-gas ratio (L/G) and the initial NOx concentration on the efficiency of NOx removal was examined. Two domains of the molar ratio X were distinguished in which denitrification was governed by different mechanisms: for X > 1.0 NO2 undergoes further oxidation to higher oxides being efficiently absorbed in the scrubber. At the stoichiometric conditions (X = 1) the effectiveness of NO oxidation was better than 90%. However, the effectiveness of NOx removal reached only 25%. When ozonation was intensified (X >/= 2.25) about 95% of NOx was removed from flue gas. The concentration of sodium hydroxide in the aqueous solution and the liquid-to-gas ratio in the absorber had little effect on the effectiveness of NOx removal for X > 2.

Background: Measurement of fractional exhaled nitric oxide (FeNO) is a useful technique for detection of eosinophilic airway inflammation and assessment of efficiency of corticosteroid treatment in patents with respiratory disease. Generally studies agree that measurement of FeNO is a useful non-invasive biomarker in patients with chronic obstructive pulmonary disease (COPD), however, there are reports that do not confirm such a relationship between FeNO and COPD. Aim of the study: The main objective of this study was to investigate FeNO levels in Polish patients with COPD compared to healthy controls. As a secondary objective, we assessed the influence of smoking on FeNO levels in healthy patients, and patients with COPD. Material and methods: FeNO concentration was measured using an electrochemical analyzer in healthy nonsmokers (n=21), healthy smokers (n=25), and former smokers with COPD (n=30) and smokers with COPD (n=38). General characteristics, hematological variables and serum biochemical parameters were also obtained and analyzed using the Kruskal-Wallis test. Results: FeNO measurement revealed significantly reduced NO levels in healthy smokers compared to healthy non-smokers, former smokers with COPD and smokers with COPD (median [range]: 14 [6–17] vs. 21 [15–29], 25 [15–53], and 19 [11–32] ppb, respectively, p<0.001). Moreover, we found increased FeNO levels in ex-smokers with COPD compared with smokers with COPD (p<0.05). No associations between FeNO and other analyzed parameters were found. Conclusions: Levels of FeNO, measured by with an electrochemical analyzer, were elevated among patients with COPD compared to healthy non-smoking controls. Moreover, our study confirms that smoking results in a reduction in FeNO concentration in both healthy patients and patients with COPD.

Both ADAM17, the secretase responsible for the shedding of ectodomains of numerous membrane proteins including TNF and its receptors, as well as nitric oxide synthesized by inducible nitric oxide synthase play regulatory roles in inflammation and tumor progression. We analyzed the effect of endogenous and exogenous nitric oxide on the expression and activity of ADAM17 in murine endothelial cells and a monocyte/macrophage cell line. We found that endogenous nitric oxide influenced neither ADAM17 mRNA level nor the shedding of two ADAM17 substrates, TNF and TNFR1. Exogenous NO significantly diminished the release of TNF and TNFR1 without affecting the ADAM17 transcript level. Our data seem contrary to a previous report that showed the activation of ADAM17 by nitric oxide (Zhang et al., 2000, J Biol Chem 275: 15839-15844). We discuss potential mechanisms of NO-mediated inhibition of ectodomain shedding and possible reasons of discrepancy between our results and the previous report.

ObjectivesAmbient air pollution is related to adverse respiratory effects. Because of a popular habit of recreational physical activity, the effects of exposure to increased levels of air pollution attract increasing attention. It remains unclear whether the allergy status has an impact on acute responses to air pollution exposure during brief exercise in young adults. The aim of the study was to determine if acute respiratory responses to ambient air pollution during physical exercise differ between young subjects with and without a history of upper respiratory allergy.Material and MethodsOverall, 41 young males with (N = 15, 36.6%) and without allergy (N = 26, 63.4%) performed short moderate-intensive cycle-ergometer sessions in winter air pollution exposure conditions. Associations were analyzed between environmental conditions and acute physiological changes in spirometry, fractioned exhaled nitric oxide (FeNO) levels, blood pressure and pulse oximetry.ResultsNo associations between air pollution concentrations and changes in forced vital capacity, forced expiratory volume in 1 s and the Tiffeneau index were found. In the subjects without allergy, the increased air pollution concentrations recorded during exercise were associated with a post-exercise increase/a smaller decrease in FeNO (SO2: Spearman’s ρ = 0.44, NOx: ρ = 0.51, and particulate matter [PM] levels – PM10: ρ = 0.51, PM2.5: ρ = 0.52). This effect was not observed in the subjects with allergy.ConclusionsUpper-respiratory allergy may be a modifying factor in human response to air pollution during exercise. Exposure to air pollution during brief moderate-intensive exercise did not have any acute negative impact on respiratory and cardiovascular function in young males. However, in the case of FeNO, subclinical post-exercise changes related to air pollution were observed in volunteers without allergy.

A number of epidemiological and experimental data indicate that exposures to low doses of low-LET ionising radiation may trigger the activity of natural anti-tumour immune mechanisms and inhibit tumour growth. In the present study, we assessed the cytotoxic activity and production of nitric oxide, superoxide anions, and tumour necrosis factor-alfa in peritoneal macrophages collected from BALB/c mice exposed to single whole-body irradiations with 0.1, 0.2, or 1.0 Gy X-rays. The results indicate that all the tested parameters were significantly up-regulated in macrophages obtained from mice exposed to 0.1 or 0.2 Gy X-rays but not in those collected from the sham-irradiated and 1.0 Gy-exposed animals.

The novel MAO-B inhibitor PF9601N, its cytochrome P450-dependent metabolite FA72 and l-deprenyl were studied as potential peroxynitrite (ONOO-) scavengers and nitric oxide synthase (NOS) inhibitors. The scavenging activity of these compounds was evaluated by measuring the oxygen consumption through peroxynitrite-mediated oxidation of both linoleic acid and brain homogenate. FA72, PF9601N and l-deprenyl caused a concentration-dependent inhibition of ONOO--induced linoleic acid oxidation with an IC50 value of 60.2 µM, 82.8 µM and 235.8 µM, respectively. FA72 was the most potent also in inhibiting ONOO--induced brain homogenate oxidation with an IC50 value of 99.4 µM, while PF9601N and l-deprenyl resulted weaker inhibitors in the same experimental model, showing an IC50 value of 164.8 and 112.0 µM, respectively. Furthermore, both the novel MAO-B inhibitor as well as its metabolite were able to strongly inhibit rat brain neuronal NOS (IC50 of 183 µM and 192 µM, respectively), while l-deprenyl at the highest concentration used (3 mM), caused only a slight decrease of the enzyme activity. Moreover, inducible NOS was strongly inhibited by FA72 only. All these results suggest that PF9601N could be a promising therapeutic agent in neurodegenerative disorders such as Parkinson's disease.

Opis:
Carbon monoxide (CO) is an odorless, tasteless and colorless gas which is generated by heme oxygenase enzymes (HOs). HOs degrade heme releasing equimolar amounts of CO, iron and biliverdin, which is subsequently reduced to bilirubin. CO shares many properties with nitric oxide (NO), an established cellular messenger. Both CO and NO are involved in neural transmission and modulation of blood vessel function, including their relaxation and inhibition of platelet aggregation. CO, like NO, binds to heme proteins, although CO binds only ferrous (FeII) heme, whereas NO binds both ferrous and ferric (FeIII). CO enhances the activity of guanylate cyclase although it is less potent than NO. In contrast, CO inhibits other heme proteins, such as catalase or cytochrome P450. The effects of CO on gene expression can be thus varied, depending on the cellular microenvironment and the metabolic pathway being influenced. In this review the regulation of gene expression by HO/CO in the cardiovascular system is discussed. Recent data, derived also from our studies, indicate that HO/CO are significant modulators of inflammatory reactions, influencing the underlying processes such as cell proliferation and production of cytokines and growth factors.

The effects of cyclosporin A (CsA), a potent immunosuppressive drug with antiparasitic activity, on the innate immunological response in guinea pig lungs during an early period (6th and 14th days) after T. spiralis infection were studied. CsA treatment of T. spiralis-infected guinea pigs caused a significant attenuation of immunological response in lungs by decreasing lymphocyte infiltration into pulmonary alveolar space, inhibiting alveolar macrophage superoxide anion production and lowering both the production of NO metabolites measured in bronchoalveolar lavage fluid and expression of the iNOS protein in lung homogenates, allowing us to speculate that the T. spiralis-dependent immunological response is dependent on lymphocyte T function. Interestingly, CsA itself had a pro-inflammatory effect, promoting leucocyte accumulation and macrophage superoxide production in guinea pig lungs. This observation may have a relevance to the situation in patients undergoing CsA therapy. Macrophage expression of the iNOS protein, evaluated by immunoblotting was not influenced by treatment of animals with CsA or anti-TGF-antibody, indicating different regulation of the guinea pig and murine enzymes.

Praca przedstawia budowę i zasadę działania laserowego spektrometru do detekcji tlenku azotu przy niskich stężeniach (na poziomie sub-ppm). Opracowany przyrząd oparty jest na wewnątrzimpulsowej spektroskopii, laserze kaskadowym (1902 cm⁻¹) i komórce wielokrotnych przejść. Przedstawiono podstawowe charakterystyki kalibracyjne spektrometru, wpływ stężenia, temperatury i ciśnienia gazu na mierzone wartości transmisji promieniowania laserowego. Dla komórki o długości drogi 36 m, granica wykrywalności NO w powietrzu wynosi 14 ppb.
This work presents construction and principle of operation of the spectrometer for nitric oxide detection at low concentration (at ppb level). Designed instrument is based on intra pulse spectroscopy, quantum cascade laser (1900 cm⁻¹) and multipass cell. Main calibration characteristics of the spectrometer, effects of concentration, temperature and pressure of nitric oxide on transmission of laser radiation are presented. For a cell with a path length of 36 m, the limit of detection is 14 ppb.

Opis:
Nitric oxide (NO) is a potent extracellular and intracellular physiological messenger. However, NO liberated in excessive amounts can be involved in macromolecular and mitochondrial damage in brain aging and in neurodegenerative disorders. The molecular mechanism of its neurotoxic action is not fully understood. Our previous data indicated involvement of NO in the release of arachidonic acid (AA), a substrate for cyclo- and lipoxygenases (COX and LOX, respectively). In this study we investigated biochemical processes leading to cell death evoked by an NO donor, sodium nitroprusside (SNP). We found that SNP decreased viability of pheochromocytoma (PC12) cells in a concentration- and time-dependent manner. SNP at 0.1 mM caused a significant increase of apoptosis-inducing factor (AIF) protein level in mitochondria. Under these conditions 80% of PC12 cells survived. The enhancement of mitochondrial AIF level might protect most of PC12 cells against death. However, NO released from 0.5 mM SNP induced massive cell death but had no effect on protein level and localization of AIF and cytochrome c. Caspase-3 activity and poly(ADP-ribose) polymerase-1 (PARP-1) protein levels were not changed. However, PARP activity significantly decreased in a time-dependent manner. Inhibition of both COX isoforms and of 12/15-LOX significantly lowered the SNP-evoked cell death. We conclude that AIF, cytochrome c and caspase-3 are not responsible for the NO-mediated cell death evoked by SNP. The data demonstrate that NO liberated in excess decreases PARP-1 activity. Our results indicate that COX(s) and LOX(s) are involved in PC12 cell death evoked by NO released from its donor, SNP.

There is growing evidence that endothelial dysfunction, which is often defined as the decreased endothelial-derived nitric oxide (NO) bioavailability, is a crucial factor leading to vascular disease states such as hypertension, diabetes, atherosclerosis, heart failure and cigarette smoking. This is due to the fact that the lack of NO in endothelium-dependent vascular disorders contributes to impaired vascular relaxation, platelet aggregation, increased vascular smooth muscle proliferation, and enhanced leukocyte adhesion to the endothelium. During the last several years, it has become clear that reduction of NO bioavailability in the endothelium-impaired function disorders is associated with an increase in endothelial production of superoxide (O2̇̄). Because O2̇̄ rapidly scavenges NO within the endothelium, a reduction of bioactive NO might occur despite an increased NO generation. Among many enzymatic systems that are capable of producing O2̇̄, NAD(P)H oxidase and uncoupled endothelial NO synthase (eNOS) apparently are the main sources of O2̇̄ in the endothelial cells. It seems that O2̇̄ generated by NAD(P)H oxidase may trigger eNOS uncoupling and contribute to the endothelial balance between NO and O2̇̄. That is maintained at diverse levels.

Opis:
Heme oxygenase-1 (HO-1), an inducible enzyme degrading heme to biliverdin, iron and carbon monoxide, is involved in regulation of inflammation and angiogenesis. Tin protoporphyrin (SnPPIX) and zinc protoporphyrin (ZnPPIX) are commonly used as competitive inhibitors of HO-1. We aimed to compare the effects of SnPPIX and ZnPPIX on the production of vascular endothelial growth factor (VEGF), activity of inducible nitric oxide synthase (iNOS) and cell viability. All experiments were performed on rat vascular smooth muscle cells and murine RAW264.7 macrophages treated with 3-10 μM protoporphyrins. Some cells were additionally stimulated with IL-1β or with lipopolysaccharide. After a 24 h incubation period SnPPIX and ZnPPIX significantly reduced the generation of VEGF in vascular smooth muscle cells and RAW264.7, both in resting and stimulated cells. The inhibitory potentials of both protoporphyrins on VEGF synthesis were very similar. In contrast, analysis of iNOS activity revealed that results obtained with different HO-1 inhibitors are discrepant. Generation of nitric oxide by iNOS was significantly increased by SnPPIX but strongly decreased by ZnPPIX. Similar differences were observed when cell viability was compared. SnPPIX improved the cell survival rate, whereas the same doses of ZnPPIX exerted some cytotoxic effects. In summary, SnPPIX and ZnPPIX can be used as HO-1 inhibitors in some experimental models. However, these compounds produce also HO-independent effects, which can make the interpretation of experiments very uncertain. Thus the involvement of the HO-1 pathway should be always confirmed by more specific methods.

This article presents results of research concerning the possibility of reducing the level of toxic nitric oxides (NOx) emission to the atmosphere. The research has been conducted on DKVR 20-13, PTVM-50 and DE 25-14 gas boilers. The complex character of this issue requires individual consideration regarding each boiler configuration. Each case requires consideration of characteristics and details of all elements constituting the boiler-furnace unit. The main problem was to establish the reference level to which the reduction of nitric oxides occurs. The actual maximum emission of nitric oxides was assumed as this level. It was verified with the maximum allowable emission of nitric oxides for each boiler. Three levels of the potential influence of emission on the atmosphere have been taken into account. This experimental research allowed for proposing an effective method, which led to reducing nitric oxides emission by around 30%.

Purpose: The aim of the study was the assessment of the influence of MLS laser therapy on morphological changes in nailfold videocapillaroscopy (NVC), clinical features, and the serum NO level in patients with primary and secondary Raynaud’s phenomenon (RP). Materials and methods: The analysis was performed on a group of 78 patients with RP and 30 healthy volunteers, who underwent NVC examination. NO concentration was assayed using the Griess method in blood serum before and after 3-weeks of laser biostimulation. MLS wasperformed with a Laser-M6 ASA Company device,for 3 weeks with weekend breaks, using thefollowing parameters: a frequency of 1500 Hz, adose of 25 J/cm2, and a time of 2.5 minutes on onehand. Results: After 3 weeks of MLS laser therapy, thebeneficial clinical effects manifested by a decrease of duration and number of RP attacks and degree of pain score on the visual analogue scale (VAS) in patients with primary and secondary RP. Clinical improvement after MLS laser therapy was reflected in the assessment of microcirculation disorders in NVC examination. Moreover, the tendency of normalization of NO concentration in the serum of patients with primary and secondary RP may suggest a favorable effect of laser biostimulation on the regulation of processes taking part in microcirculation disorders. Conclusions: The results showed that NVC is a useful diagnostic tool in the evaluation of dynamic microvascular involvement in RP patients. MLS laser therapy has a beneficial effect in patients with primary and secondary Raynaud’s phenomenon

Wśród różnorodnych działań w obszarze rolnictwo-środowisko konieczność ochrony powietrza na terenach użytkowanych rolniczo jest dzisiaj równie ważnym zadaniem, jak dbałość o czystość znajdujących się tam zasobów wodnych. Substancjami zanieczyszczającymi powietrze, które w znacznych ilościach powstają w toku szeroko rozumianej produkcji rolniczej, są między innymi gazowe, nieorganiczne związki azotu: amoniak [NH3], tlenki azotu - NOx, w tym tlenek azotu (I), powszechnie określany mianem "podtlenku azotu". Emisja każdego z tych związków powoduje określone zakłócenia w środowisku. Na podstawie literatury oraz wyników badań, wykonanych w IMUZ i kontynuowanych w ITP, w sposób syntetyczny omówiono problematykę szacowania, modelowania oraz ograniczania tych zanieczyszczeń powietrza atmosferycznego w warunkach polskiego rolnictwa. Przedstawiono konkretne wyniki pomiarów ilościowych emisji amoniaku i podtlenku azotu z różnych źródeł rolniczych, wykonane w ramach działalności statutowej i współpracy międzynarodowej w Zakładzie Chemii Gleby i Wody IMUZ w Falentach.
Nowadays, the necessity of air protection in rural regions is equally important as the care of water resources situated there. Air contaminants, which originate in significant amounts from broadly understood agricultural production, include inorganic nitrogen compounds: ammonia [NH3] and nitric oxides - NOx with dinitrogen monoxide commonly known as "nitrous oxide". The emission of any of these compounds causes interferences in the environment. Based on literature data and results of presently carried studies in IMUZ, the estimation, modelling and reducing these atmospheric air pollutants in Polish agriculture were discussed in a synthetic way. Specific results from quantitative measurements of ammonia and nitrous oxide emissions from different agricultural sources were presented and described. The results were obtained within the statutory studies and international cooperation of the Department of Soil and Water Chemistry IMUZ Falenty.

To reduce the emission of nitrogen oxides (NOx) during the co-disposal of sludge in a TTF-type precalciner, an optimized co-disposal process of a TTF-type precalciner has been implemented in a cement plant in Hebei. The model was built using ANSYS FLUENT software. The effects of three single-factor perspectives (sludge input ratio, gas flow rate, and tertiary air temperature on NO concentration) were investigated. The response surface method of Box–Behnken design was used. When the sludge ratio increased from 0 to 25%, the NO concentration at the outlet was 122–297 mg/m³. Meanwhile, it increased from 192 mg/ m³ to 241 mg/ m³ since the airflow increased from 95 m³/s to 122 m³/s. The maximum NO concentration was 192 mg/ m³ when the tertiary air temperature was 1170 K. The inter-action between airflow and sludge ratio was more significant than any other interaction between other conditions (P < 0.05). Finally, the optimum conditions were a sludge ratio of 5%, airflow 109 m³/s, and tertiary air temperature 1280 K. NO concentration was 166.9 mg/m³ under this condition.

Fenton reaction is an important method to remove NO from flue gas. • OH radicals generated in the Fenton reaction can effectively oxidize NO to NO2, which is absorbed and removed by alkali sorbent. To supply guidance for engineering research, numerical simulation of NO oxidation by • OH radical clusters in flue gas has been carried out using Fluent software. The average concentration of NO on the cross sections at various positions of a cylindrical pipe with a circular surface was calculated by simulation. Under various working conditions (temperature, • OH/NO molar ratio, NO concentration in flue gas, and jet velocity), NO oxidation efficiency by • OH radical was simulated and the key factors affecting NO oxidation were analyzed. The results show that temperature and • OH/NO molar ratio are the key factors affecting the oxidation of NO by • OH radicals. The injection velocity has also a significant effect on the oxidation efficiency while the moisture and oxygen content are minor factors influencing the process. The optimum oxidation efficiency of NO is obtained at 473–523 K, the molar ratio of • OH/NO ca. 1.4, the jet velocity 10 m/s, and the flue gas velocity of 3 m/s.

Introduction and aim. Violation in the salivary glands will inevitably cause changes in periodontium, and periodontitis can disrupt the functioning of the salivary glands. The purpose of the work is to evaluate changes in NO-synthase and arginase activities, pro- and antioxidant balance in rat salivary glands and soft periodontal tissues during administration of bacterial lipopolysaccharide (LPS) and water avoidance stress (WAS) modeling. Material and methods. The experiment was performed on 24 rats. The animals were divided into 4 groups: control, WAS, animals injected intraperitoneally with 0.4 μg/kg of bacterial LPS of Salmonella typhi, WAS+LPS. Results. Water avoidance stress led to decrease of inducible NO-synthase (iNOS) activity in salivary glands by 1.63 times, but decreased arginase activity by 1.15 times, superoxide production increased by 1.53 times, catalase activity decreased by 1.2 times, and malonic dialdehyde (MDA) increased by 1.19 times compared to the control. Lipopolysaccharide led to increase of constitutive NO-synthases (cNOS) activity in salivary glands by 1.48 times, but decreased arginase activity by 6.15 times, catalase activity increased by 2.6 times and superoxide dismutase activity decreased by 2.74 times, and MDA increased by 6.84 times compared to the control. Water avoidance stress + LPS in salivary glands led to decrease of cNOS and arginase activity by 1.09 and 1.19 times, respectively, superoxide production increased by 1.88 times, catalase and superoxide dismutase activity decreased by 1.06 times and 1.34 times, respectively, and MDA increased by 2.44 times compared to the control. Water avoidance stress led to increase of iNOS activity in periodontium by 1.44 times and arginase activity decreased by 1.37 times, superoxide production increased 1.32 times, catalase activity and superoxide dismutase activities decreased by 1.27 times and by 1.53 times, respectively, and MDA increased by 1.31 times compared to the control. Lipopolysaccharide led to increase of iNOS activity in the periodontium by 3.88 times, arginase activity decreased by 2.69 times, superoxide production increased 1.64 times, catalase activity increased by 4.32 times, and MDA increased by 4.51 times compared to the control. Water avoidance stress + LPS in periodontium led to increase of iNOS and cNOS activities by 1.95 times and 1.53, respectively, arginase activity decreased by 1.39 times, superoxide production increased 1.66 times, catalase activity increased by 1.11 times, and MDA increased by 1.53 times compared to the control. Conclusion. The combination of LPS and WAS leads to changes in NO production and oxidative stress in salivary glands and the periodontium.

Asthma is a common disease, occurring increasingly among both children and adults. It is defined as a chronic inflammatory disease, characterized by hyperresponsiveness and reversible bronchial obstruction. The diagnosis of asthma in children is currently based mainly on clinical and spirometric evaluation as well as on the assessment of response to anti-inflammatory treatment. Currently there are ongoing discussions on the choice of optimal diagnostic and staging methods. Therefore, the measurement of the levels of exhaled nitric oxide (FeNO) is being seen as a viable option. The results of the measurement are obtained easily and non-invasively. High variability in the levels depending on both environmental factors and patient cooperation is a disadvantage of the test. The aim of this study was to determine the relationship between the levels of exhaled nitric oxide and the severity of asthma based on spirometric outcomes. A total of 141 children aged 5–17 years, including 35 patients diagnosed with asthma, among whom eight were in the stage of exacerbation, were qualified for the study. The control group consisted of 106 children admitted to the hospital for other reasons, with the exception of respiratory diseases. Spirometry and FeNO measurements were performed. No statistically significant differences were found between FeNO levels in patients with asthma or asthma exacerbations and the control group. The highest variation of FeNO levels was observed in the control group, indicating intersubject and factor variability of FeNO levels in exhaled gases. Although the utility of FeNO levels as an indicator of the severity of airway inflammation has been demonstrated in numerous studies, this study questions the usefulness of this parameter as a marker of asthma severity. This is probably due to the large intersubject variations in the concentration of exhaled NO, depending on patient cooperation and clinical status (and thus the accuracy of spirometry) and the fact that FeNO is affected by multiple other factors, such as diet rich in nitrogen compounds, hyperventilation, viral infections, tobacco smoke, airway pH. Only some of these factors can be eliminated, which renders the interpretation of FeNO concentration much more difficult and almost excludes its use in the diagnostics and monitoring of asthma on a large scale.
Astma jest powszechnym, coraz częstszym schorzeniem, występującym zarówno u dzieci, jak i u dorosłych. Definiuje się ją jako przewlekłą chorobę zapalną, cechującą się nadreaktywnością i odwracalną obturacją oskrzeli. Rozpoznanie astmy u dzieci opiera się obecnie przede wszystkim na ocenie klinicznej i spirometrycznej, a także na ocenie odpowiedzi na zastosowane leczenie przeciwzapalne. Nadal trwają dyskusje nad wyborem optymalnych metod diagnostyki i oceny zaawansowania schorzenia – w tym kontekście rozważa się pomiar stężenia tlenku azotu w wydychanym powietrzu (FeNO). Stężenie tlenku azotu, jako regulatora czynności układu oddechowego i związku biorącego udział w patogenezie astmy, koreluje z poziomem innych markerów stanu zapalnego. Wynik pomiaru uzyskuje się w sposób łatwy i nieinwazyjny. Wadą badania jest duża zmienność stężenia w zależności od czynników środowiskowych i współpracy pacjenta. Celem pracy była ocena zależności między stężeniem tlenku azotu w wydychanym powietrzu a stopniem ciężkości astmy określanym na podstawie wyniku badania spirometrycznego. Do badania zakwalifikowano 141 dzieci w wieku 5–17 lat, wśród nich 35 pacjentów z rozpoznaniem astmy, w tym ośmiu w stadium zaostrzenia choroby. Grupę kontrolną tworzyło 106 dzieci przyjętych do Kliniki z innych przyczyn, z wyłączeniem chorób układu oddechowego. Wykonywano badanie spirometryczne i pomiar FeNO. Nie stwierdzono istotnej statystycznie różnicy między FeNO w astmie i jej zaostrzeniu a wynikami pomiarów w grupie kontrolnej. Zróżnicowanie rezultatów okazało się największe w grupie kontrolnej, co wskazuje na osobniczą i czynnikową zmienność zawartości tlenku azotu w gazach wydychanych. Choć liczne badania wykazały użyteczność pomiaru FeNO jako wskaźnika nasilenia procesu zapalnego dróg oddechowych, niniejsze badanie podaje w wątpliwość przydatność omawianego parametru jako markera stopnia zaawansowania astmy. Wynika to zapewne z dużej zmienności osobniczej stężenia NO w powietrzu wydychanym w zależności od stopnia współpracy pacjenta i jego stanu klinicznego (a co za tym idzie stopnia poprawności wykonania badania spirometrycznego) oraz z tego, że na FeNO wpływa wiele czynników: dieta bogata w związki azotu, hiperwentylacja, infekcje wirusowe, dym tytoniowy, pH w drogach oddechowych. Jedynie część tych czynników można wyeliminować, co znacznie utrudnia interpretację wyników pomiarów FeNO i niemal wyklucza ich zastosowanie w diagnostyce czy monitorowaniu na szeroką skalę.

Besides the previously described nitric oxide-detoxification activity we identified new features of class-1 non-symbiotic hemoglobin from Medicago sativa (Mhb1). Under in vitro conditions, using peroxidase in-gel activity assay, the Mhb1 protein was shown to possess also peroxidase-like activity. Due to this activity, in the presence of nitrite and hydrogen peroxide, the protein can mediate autonitration and nitration of other proteins at tyrosine residues, as revealed by tandem mass spectrometry and immune assay approaches. Mhb1 through its multifunctional activities can affect different components of signal transduction cascades operating during plant response to infections. This influence is manifested by Mhb1-mediated selective up-regulation of expression of certain pathogen inducible genes in Pseudomonas syringae infected Arabidopsis thaliana plants which overproduce Mhb1, as revealed by reverse transcription-quantitative real-time PCR analysis. Changes in expression level of these genes can influence such processes as synthesis of secondary metabolites, protein degradation and biosynthesis of ethylene. They can also result in alteration of pathogen-induced defense response of Mhb1 transgenic plants.

Opis:
Celem pracy była histologiczna ocena nerek szczura poddanego działaniu egzogennego tlenku azotu. Szczury użyte w niniejszej pracy zostały podzielone na 2 równe grupy. Zwierzęta z grupy doświadczalnej otrzymywały L-argininę - 40mg/kg masy ciała per os, co drugi dzień przez 2 tygodnie i zostały dekapitowane po 3 tygodniach doświadczenia. Szczury z grupy kontrolnej dostawały 2ml wody destylowanej per os co drugi dzień i zostały dekapitowane po 3 tygodniach doświadczenia. Z wycinków nerek pobranych do analizy robiono preparaty, które barwiono Hematoksyliną i Eozyną. Preparaty oglądano pod mikroskopem świetlnym i opisano. Obliczono również indeks apopotozy, a wyniki badań poddano ocenie statystycznej Badania wykazały, że L-arginina jako prekursor egzogennego tlenku azotu podana samicom szczura w dawce uważanej za bezpieczną dla matki i płodu, wywołuje nasiloną apoptozę komórek nabłonka nerek, obserwowaną pod mikroskopem świetlnym i potwierdzoną istotnym statystycznie wzrostem indeksu apoptozy.
The aim of the study was to prepare histological evaluation of rat’s kidneys exposed to exogenous nitric oxide. Rats used in the research were divided into 2 even groups. The animals in the experimental group received L-Arginine --40mg/kg of the body weight, per os, every other day for 2 weeks and were decapitated after 3 weeks of the experiment. The rats in the control groups received 2ml of distilled water, per os, every other day and were decapitated after 3 weeks following the experiment commencement. The preparations for analysis were made on the basis of kidney segments stained with haematoxylin and eosin. They were then studied with an optic microscope and characterised. Also, an apoptosis index was calculated and the research results were subjected to statistical evaluation. The study results showed that L-arginine served to rat females as a precursor to exogenous nitric oxide, in a dosage thought to be safe for the mother and foetus, induced intensified apoptosis of the renal epithelial cells, as it was observed with the use of an optic microscope and confirmed by a statistically significant increase in the apoptosis index value.
