Abiotic stress responses are regulated critically at the transcriptional level. Clarifying the intricate mechanisms that regulate gene expression in response to abiotic stress is crucial and challenging. For this purpose, the factors that regulate gene expression and their binding sites in DNA should be determined. By using bioinformatics tools, the differentially expressed probe sets were studied. A meta-analysis of transcriptomic responses to several abiotic stresses in barley was performed. Motif enrichments revealed that AP2/ERF (APETALA2/Ethylene-Responsive Factor) has the most frequent binding sites. We found that the bHLH transcription factor family has the highest number of transcription factor members. Moreover, network construction revealed that AP2 has the highest number of connections with other genes, which indicates its critical role in abiotic stress responses. The present research further predicted 49 miRNAs belonging to 23 miRNA families. This study identified the probable conserved and enriched motifs, which might have a role in the regulation of differentially expressed genes under abiotic stresses. In addition to shedding light on gene expression regulation, a toolbox of available promoters for genetic engineering of crop plants under such abiotic stresses was developed.

The article analyses the motif of water in theatrical plays of the Belgian writer Michel de Ghelderode. It is demonstrated that in his dramatic texts the motif/image of water is not merely a background to the plot but carries some important meanings, usually oscillating between threat and salvation. The analyses proposed in the article focus on these two meanings, on the basis of four theatrical plays which most visibly emphasise them, giving water a particular place in the presented world.

The poetry of Khajeh Mohammad Hafiz Shirazi has vastly influenced the poetry of Ralph Waldo Emerson, as many critics have noted but have not demonstrated. Emerson is an American poet whose work reflects the influences of Persian poets, among which that of Hafiz is remarkable. The influence of Hafiz on Emerson includes memorable images, themes and motifs. While one can argue that this influence was indirect, it is obvious from the closeness of certain similarities, from Emerson’s intimate knowledge of Hafiz’s poetry, and from his love for Persian poetry, that the influence was more direct than otherwise. Although Emerson knew German and read Hafiz in German translations yet, he embarked on translating the poems of Hafiz in English in order to master Hafiz’s poetry and to introduce him to American readers. These translations themselves are another proof of the claim of influence of Hafiz on Emerson. The methodology of this article is to set the poems of the two poets over against one another and study them watchfully in order to demonstrate the influence of the precursor poet on the belated poet. Therefore the sources of familiarity of Emerson with Hafiz must not be forgotten and should be brought to the surface.

Opis:
In his article Wallenrod’s Garden and the Mystery of Wajdelota the author analyses the role of the motif of garden in Adam Mickiewicz’s epic poem Konrad Wallenrod. The conventionality of the use of the term ‘garden’ is emphasized by pointing to the fact that there were some changes introduced to the order of the valley near the Neman River, which were conducive to his amorous meetings with his Ladylove. The manner in which the picture of ‘the garden’ is outlined in the poem is confronted by the author with the classical and romantic gardening styles. It is stressed that the poet distances himself from the two schools, which results from, among other things, the emphasis on the beauty of the valley, a garden created by Nature herself. The motif of the garden played an important role in Konrad’s last conversation with Aldona, when the main hero of the poem tried in vain to persuade his wife to escape to Lithuania. He tempted her with a mirage of regained happiness, appealed to their mutual memories, in which this old and still existing garden, a witness of their love, occupied the central place. While analyzing the strategy of insistence used by Wallenrod in this situation, the author of the article referred to the conception of memory proposed by a German researcher, Jan Assmann. In the last part of the essay the character of the relationship between Wallenrod and Halban is considered along with the attempt to unravel the mystery hidden in the words “my son!”, which the old Lithuanian bard, the patron of Konrad’s spy mission, used in their last conversation.

The figure of a singer, Orpheus, symbolizes poetry, dilemmas, the misgivings of the artist performing at the beginning of the XX century. One of the most significant problems is the loss of identity, which in turn leads to emptiness and melancholy. Komornicka’s corpus belongs to the current of existentialism.

Each role and func- tion of the mirror has been highlighted in the article by examples from the selected movies. In the next part I focused on showing the mirror from the perspective of superstitions that are associated with it, and I also made an attempt to isolate and clarify its roles and functions which can be found in the movies. The di- vision I proposed was constructed in such a way that it could be easily expla- ined, in one or two words, what lies beneath them, and so the distribution is as follows: wraith (emphasis on the problem of identity), magical mirror (introducing elements of magic to the movies), the gates of truth (revealing the true face of reality), medium (mediating between the worlds), bad omen (the preaching of someone’s death), prison for the soul.

This article is devoted to the analysis of the motif of water in the works of Marina Tsvetaeva. The main goal is to trace how the concrete and simple lexeme “water” participates in the meaning-generating poetic process, and to prove the specialsignificance of this motif as an important sign of her creative thinking, worldview and creative personality in general. The article uses extensive material (about 630 examples) taken from poetic and prose texts to consider the main meanings of the motif of water and its functional features in the individual author’s viewpoint. Water, without losing itsnatural concreteness, becomes one of the important tools for metaphorizing the processes and phenomena of not only the physical, but also the spiritual world of the poet.

The article is devoted to the research of the motif of deception in the fairy‑tales of the Polish and Ukrainian writer S. Ostashevskyi. The work offers arguments for the topicality of the research as well as for the choice of the analysis methodology. It also emphasizes the significance of studying the author’s creative works with a view to revealing those connections between Ukrainian and Polish culture which indisputably prove their kinship. Four types of the deception motif have been distinguished based on the collections of fairy‑tales Half‑a‑Kopa Fairy‑Tales for the Little World and Half‑a‑Hundred Fairy‑Tales for Jolly People. These types are as follows: “a creature with superpowers deceives people by appearing in front of them in an anthropomorphous, intentionally inferior, everyday image”, “an infernal character takes people in without changing its image but promising material benefits and solutions to people’s life problems in exchange for their own souls or their children’s souls”, “a person (usually, a woman) deceives/tries to deceive an infernal character or some supreme forces in general”, and “a person deludes another person”. The comparison of this material with the corresponding folklore and literary heritage has defined the specificity of using and transforming the deception motif by S. Ostashevskyi: dependence on the expression of the religious idea, comical play such as appears in the 19th‑century burlesque and travesty art, and reflection on the harmonious combination of Ukrainian and Polish folklore traditions.

The paper deals with enigmatic symbols presented in Biblical allusions in the texts of contemporary Russian poets Bakhyt Kenzheev, Evgeny Lesin, Ilya Falikov and Oleg Komkov. Three types of artistic symbols may be distinguished according to the criterion of their interpretative depth: empathic symbols are connected with the most important characteristics of existence, eidetic symbols remain in our memory due to the exceptional brightness of images, while enigmatic symbols constitute riddles to be deciphered. Biblical allusions in modern Russian poetry often belong to the third class of symbols. The allusions in the poems analysed are predominantly tragic in tone and develop the ideas of Ecclesiastes, with an emphasis on the incomprehensibility of the divine plan. The images of retribution to Sodom and flights to Egypt and from it illustrate the eternal circle of human existence. Bakhyt Kenzheev offers a new reading of the story of the destruction of Sodom, in which God’s messenger comes to the ruins of the city to see if there are any survivors after the apocalyptical retribution. A new dialogue with Ecclesiastes includes an image of an endless line of lanterns which may symbolise an after-death meeting of souls. In another poem of this author there is an allusion to the world’s annihilation as a necessary stage of the development of the Universe. Evgeny Lesin calls his country the last Sodom, and a place which he is reluctant to leave even though he is aware of its imminent fate. A new interpretation of the Exodus from Egypt is given by Ilya Falikov. In his take it becomes an eternal journey in a circle with no chance of escape. Oleg Komkov also treats this narrative in a new way: as a never-ending chain of death and resurrection. These allusions emphasise the idea that after creation, there inevitably comes destruction. The emphasis in the analysed texts is on a tragic perception of the world blended with an ironic attitude to it. Interpreting enigmatic symbols requires from the readers a creative participation in the dialogue with poets.

Opis:
The article deals with two fairy-tale motives – “youth” and “old age” in Mikhail Osor-gin’s novel A Quiet Street (Sivtzev Vrazhek, 1928). Owing to the author’s outlook and artistic strategy they do not stand in the opposition to one another, but rather depict parallel vectors of human life, its presentation the context of space (micro and macro space) as well as in the context of a philosophically meaningful historical time correlated with eternity. The author’s aim is supported by the novel’s structure – a freely arranged series of short stories. The article exposes the problem of the writer’s comprehension of human life within different time and spatial plans. The time layer covers daily, annual and eternal / universal time. The spatial dimension of the artistic world takes into account the Infinite Universe, the solar system, the Earth, Russia, Moscow and the corner house in Sivtzev Vrazhek, which is the main center of the novel structure.

The article reinterprets the two poems which Juliusz Słowacki wrote while travelling to the Holy Land. Unlike in previous interpretations, here the author uses the motif of tears to uncover the poems’ emotional value and to situate them in the tradition of romanticism. Other poems from the same period set the interpretative context and anticipate the poet’s spiritual growth. Consequently, the article is yet another attempt to demonstrate the emotive characteristics of Słowacki’s lyric poetry. Moreover, it shows how his works are related to his biographical background.

The calcium-activated neutral proteases, μ- and m-calpain, along with their inhibitor, calpastatin, have been demonstrated to mediate a variety of Ca2+-dependent processes including signal transduction, cell proliferation, cell cycle progression, differentiation, apoptosis, membrane fusion, platelet activation and skeletal muscle protein degradation. The cDNA coding for yak calpastatin was amplified and cloned by RT-PCR to investigate and characterize the nucleotide/amino-acid sequence and to predict structure and function of the calpastatin. The present study suggests that the yak calpastatin gene encodes a protein of 786 amino acids that shares 99 % sequence identity with the amino-acid sequence of cattle calpastatin, and that the yak protein is composed of an N-terminal region (domains L and XL) and four repetitive homologous C-terminal domains (d1–d4), in which several prosite motifs are present including short peptide L54–64 (EVKPKEHTEPK in domain L) and GXXE/ DXTIPPXYR (in subdomain B), where X is a variable amino acid. Our results suggest the existence of other functional sites including potential phosphorylation sites for protein kinase C, cAMP- and cGMP-dependent protein kinase, casein kinase II, as well as N-myristoylation and amidation sites that play an important role in molecular regulation of the calpain/calpastatin system. The regulation of the calpain/calpastatin system is determined by the interaction between dIV and dVI in calpains and subdomains A, B, and C in calpastatin.

The SeqA protein of Escherichia coli is not only the main negative regulator of DNA replication initiation but also a specific transcription factor. It binds to hemimethylated GATC sequences and, with somewhat different specificity, to fully methylated GATC regions. Recently, a microarray analysis was reported, in which transcriptomes of wild-type and ΔseqA strains were compared. Although in the seqA mutant the levels of some transcripts were significantly decreased while certain transcripts were evidently more abundant relative to wild-type bacteria, no correlation between the presence of GATC motifs in promoter sequences and transcription activity was found. However, here we show that when larger DNA fragments, encompassing positions from -250 to +250 relative to the transcription start site, are analyzed, some common features of GATC distribution near the promoters activated by SeqA can be demonstrated. Nevertheless, it seems that the GATC pattern is not the only determinant of SeqA-dependence of promoter activity.

A comparative analysis of traditional western Ukrainian and Lithuanian towels provides rich material for new creative inspirations, as well as diversification of individual and mass-produced interior textiles, and also visualizes historical ties between the two countries. Therefore, the objective of the article is to analyse the weaving techniques that prevail in the creation of towel decor, to determine the artistic and stylistic parameters of Ukrainian and Lithuanian towels and to establish the common and unique features of their decor. The techniques of plain and twill weaving, the latter of which could obtain simple textured patterns on the canvas, were widespread in both countries. Various weaving compositions were obtained by the pick-up techniques of weaving, the overshot, and the damask weaving technique, which was much more widespread in Lithuania than in the western part of Ukraine.

The article concerns the way in which the eye topos functions in Paweł Hertz’s Sedan and Kazimierz Brandys’ The Wooden Horse, works representing Polish literature of retribution written in the 1940s. The role of the process of seeing is interpreted by means of the category of melancholy, and the protagonists in these works are presented as melancholics. Standing in front of a window or in the quiet of European museums, members of the intelligentsia do not so much focus on passive contemplation but rather protest against History. The desire to constantly use the eyes has an existential and political sense. Recognition of the phenomenon of a melancholic gaze in the literature of retribution enriches the traditional interpretation of this trend in literature by lending it a new context and allowing rehabilitation of the intellectuals portrayed in it.

Różycki presents himself as an artist aware of his own work and cultural achievements. He creates an original novel which, at the same time, is strongly rooted in tradition.

Christmas and Baptism as symbols of poetic creativity: Evangelical motifs in the poetry of Russian metarealism and the 21st centuries. The author considers the “Epiphany” and “Christmas” images in the lyrics of Yevgeny Daenin, Ivan Zhdanov, Alexei Parshikov, Sergey Solovyov, correlating them both with the text of the Holy Scripture and with the poetic tradition of “Christmas verses” of the 19th century and two boundary periods (the 19th-20th and the 20th-21st centuries), in which the search for the “painful and unsteady” image of God is manifested with a particular force. Traditionally, in poetry there is an intimization of Christmas and Epiphany: so, these events are not perceived as being external, but as internal, occurring in a person’s soul. This is true of all Russian contemporary poetry. And metarealism is no exception. The article sequentially explores (using the hermeneutic method, semantic, intertextual and content analysis) verbal images and motifs associated with Baptism, Epiphany and Christmas. In the interpretation of the evangelical stories, metarealists proceed from the ideas of the divine Word-Logos, embodied in words. Baptism and Epiphany are associated with the Word’s Appearance to the People, their baptism in the Speech as a River, which at the same time becomes synonymous with Christmas and Nativity, the birth of the “mortal” word and the beginning of its Way of the Cross. Poetry is perceived by metarealists as climbing Golgotha, “the crucifix of the throat”. The poet has three hypostases in their texts: the “wise Thief” who ascends to “paradise” by God’s will, but is capable of performing the sacrament of baptism with the “verb fire”; “Carpenter”, “Joseph”, stepfather of the “God-born baby”-text; builder, governor of Heavenly Jerusalem and leader of the “Heavenly Host” of words. Analyzing the concept of Speech as a “Homing,” the author comes to the conclusion that metarealists are not so much engaged in “God-seeking” as they are developing M. Heidegger’s theory of language as a “house of being” and of existing and being.

Moreover, critical attention is paid to the way Riach’s poems forge a construct wherein a cultural agenda represents the clearest way forward for the accomplishment of Scotland’s nationalist aspirations.

The growing body of literature on intensifiers enables us to circumscribe the scope of the phenomenon more accurately and to identify rigorously its linguistic realizations. Yet, the latter are clearly underrepresented or inconsistently treated both in general dictionaries and in textbooks. That is why the need arises to work up a dictionary devoted to means of intensification so that it can serve as a tool to students of French. Rather than simply listing these means, it should be conceived of as a lexicongrammar enabling users both to vary particular forms and to respect syntactic and combinatory constraints. Additionally, it should account for when registers can be conveniently switched. The present paper aims at giving some hints on how such a disctionary should be prepared.

Opis:
The Decameron 2020 started in Croatia as an online literary event in the time of the first quarantine caused by the Covid-19 pandemic and resulted in an e-book of fifty-two selected short stories. Its initiators and editors, Ana Cerovac, Vesna Kurilić and Antonija Mežnarić, recognized the peculiarity of the experience of living in the end times, as well as the potential for comparison of the e-book with Boccaccio’s classic, and set up an online space where authors from Croatia and the region could ( re )create or transform their experiences, or reflect upon them. This paper focuses on four stories from the collection in which nature is givenspecial significance: Algernonova osveta (Algernon’s Revenge) by Nataša Milić, Zaražena (Infected) bySunčica Mamula, Redukcije(Reductions) byAnaKutleša and 2030 byRadmila Rakas. It investigates the range of feelings and attitudes towardsnature in the time of pandemic and quarantine, as reflected in these four stories.

Opis:
Animal motifs were very popular in medieval bestiaries and love poetry, especially in Provençal and Petrarchan poetry. Afterwards they spread throughout European Renaissance love poetry and they are found in love poems by Šiško Menčetić (1457–1527) and Džore Držić (1461–1501), the very first Croatian Renaissance poets. This paper examines some of the animal motifs present in their poems, in order to establish whether they could provide a better understanding of the luralism of Croatian Renaissance love poetry, which cannot be reduced to Petrarchism alone, and of the extraliterary context to which certain animal motifs are related. Our research shows that e.g. motifs of snakes and birds are linked to various types of love discourse; animal motifs related to the topos of love hunt are also analysed. It is shown that animal motifs are potentially polysemous and that their usage follows conventions of the genre and norms of a particular type of love discourse with which they are associated, while their meaning is more precisely definedin the context of the poem.

The issue connected with sense of life appears in the works of the Apostolic Fathers not only as a main subject, but also not occasionally. They used the motif of two roads to show the sense of behavior on the earth, according to the God’s commandments and his will. They do not condemn the earthly world but deeds, which lead people to death. Even in the face of his close martyrdom, they saw a clear aim: life on the earth is important, because it leads to God. The Apostolic Fathers tell also the truth connected with eternal life and hope to be used from the dead, which will take place after death. The sense of our life is – it leads a man to the resurrection.

Obie kobiety – to wielkie postaci literatury ukraińskiej.
One can identify at least a few points at which the artistic careers and personal lives of Lesya Ukrainka (1871-1913) and Olha Kobylianska (1863-1942) had crossed their paths. Both women are great figures of Ukrainian literature. They both lived and worked in the same time, although 8 years older Olga outlived Lesya by nearly 28 years. They shared the literary space, that is the early Ukrainian modernism, and the everyday environment, both living in the Chernivtsi district. The writers were linked by a special spiritual bond which was reflected in their epistolary oeuvre and their work in general. In two works of theirs in particular, that is in Lesya Ukrainka’s essay Голосні струни and Olha Kobylianska’s novelette Valce melancolique one can trace artistic and typological parallels stemming from the similarities between their personal lives and work. These are: timewise similarity, similarity within the genre, autobiographism of both texts and their intellectualism. In both works a conflict between the reality and Art is present, interpreted as a variant of the conflict between an artist and the society, as well as the feature of broadly understood ‘musicality’. Moreover, in both texts the motifs of longing/ melancholy and silence/ quietness, as well as the then popular motif of beauty/ ugliness are intertwined.

Among terracottas found in Tell Atrib in the Nile Delta by the Polish-Egyptian archaeological mission, there are two incense burners with ‘horns’ and vine scroll decoration. Both objects were made from a double mould. The first one, found in 1989 (TA 89/166), is almost fully preserved; the second one, discovered in 1978 (TA 87/41), is just a small fragment of a similar object. Archaeological context allows to date the artifacts to the Ptolemaic or Roman period. The closest analogies are known from necropoleis in Alexandria, but similar objects are also attested outside Egypt. The soot traces visible on the better preserved specimen (TA 89/166) confirm that it was used for incense burning. The find context suggests that both objects could have been rather intended for domestic cult than for official religious practice or funerary rituals. The form of the burners, and the horn-shaped projections in particular, suggest their connection to the cult of Egyptian gods.

Opis:
This essay examines biblical references, inspirations and motifs in 20th-century Italian prose. It investigates the longstanding significance of the Sacred Scripture as one of the fundaments of Western civilisation and the Scripture’s changing influence on the imagination and sensibilities of modern Italian writers; it also attempts to provide an explanation of this trend. The Bible, a source of inspiration with extensive historical presence and numerous translations, has been met with multiple interpretations extending far beyond its purely theological dimension. It has shaped European cultural heritage in terms of its themes as well as its languages, symbols, concepts and metaphors, which have permeated European works of culture in the centuries past. The Bible has encouraged creators, including authors, to continue to operate within its tradition, albeit outside the strict confines of faith itself. It is a timeless code not just because of its longstanding presence, but also because it cannot be dismissed out of hand if we are to understand not simply the works of great, world-renowned classics, and those shaped by religion in particular, but notably the works of modern writers of both Christian and secular backgrounds.

Remarks on language and poetry in the poems of Mirosław Dzień Miroslaw Dzień is perceived as a metaphysical poet. The subject matter of his poems frequently oscillates around existential problems and touches on various religious issues (including eschatological ones). An ambivalent view of language emerges from Dzien’s poems. The language used there serves to capture different aspects of reality and preserve various phenomena. On the other hand, however, it does not allow the poet to address the most important issues and articulate his innermost thoughts. In this context, it is the poetry that is the higher form of communication, though it also has great limitations. This is because everything that matters most is contained in silence.

Opis:
This article examines the concept of German Realism, also known as poetic realism or bourgeois realism, using the example of the motif of marital betrayal. Theodor Fontane’s Effi Briest is the most widely known literary work that portrays infidelity and the punishment inflicted on the wife who breaks her marriage vows. The novel is often compared to Madame Bovary and Anna Karenina. It depicts not only the young wife’s affair but also its consequences: divorce, losing the rights to her child, social isolation and finally her premature death, caused by her “anguish”. A superficial reading suggests that Fontane was inspired by the literature of European Realism and Naturalism; however, even a cursory comparison of Flaubert’s and Fontane’s works reveals the distinctive features of German Realism. It is this specificity of the mid-19 century German novel that constitutes the main focus of this article. The key aesthetic category characteristic of the German 19th century prose was “humour”, defined by Wolfgang Preisendanz as finding the connection between two seemingly unrelated things. Fontane wrote about the “beautifying veil”, which makes it possible to portray extreme social problems in a poetic way, never directly. The reader must uncover aspects of the literary work which evoke the dilemmas of a world on the verge of modernity.

His-tags are specific sequences containing six to nine subsequent histydyl residues and they are used commercially in immobilized metal affinity chromatography (IMAC) as molecular ‘anchors’ that bind to a metal ion (usually nickel), immobilized by chelation with nitrilotriacetic acid (NTA) bound to a solid support [37, 38]. Consecutive histidines are the common denominator for both His-tags used in molecular biology and for quite remote biological phenomena – more than 2000 histidine- rich proteins (HRPs) are found in microorganisms including 60% and 82% of archaeal and bacterial species, respectively and their roles are not well characterized [73]. The physicochemical properties of histidine make it a versatile amino acid that influences protein conformation and enzymatic activity [15]. Many natural proteins with a His-tag domain are assigned to different functions, for example: most bacterial proteins, containing this motif are probably involved in the homeostasis of nickel ions [68, 76], while others, e.g. newly isolated peptides from the venom of the snake genus Atheris contain poly-histidyl-poly-glycyl sequences (pHpG) can act on the cardiovascular system by inhibiting snake venom metalloproteinases and affect its function by acting on specific receptors [58, 62]. His-rich motifs have been found also e.g. in Zn2+ transporters, prion proteins, His-rich glycoproteins, transcription factors or numerous copper-binding proteins [56, 67, 84]. Binding mode and the thermodynamic properties of the system depends on the specific metal ion and the histidine sequence. Despite the wide application of the His-tag for purification of proteins, little is known about the properties of metalbinding to such tag domain. Recent experimental and theoretical studies have shown that metal ions, e.g. Cu2+ can bind to various sets of imidazoles depending on the number of histidine residues that are located in His-rich sequences. The occurrence of polymorphic binding states and the formation of an α-helical structure induced by metal ion coordination suggest that proteins with a His-tag domain may serve as the dynamic site able to ‘move’ metal ions along the tag sequence [99, 100]. This might explain the frequent occurrence of such sequences in bacterial Ni2+ chaperones, which transfer the metal ion between different proteins.

Opis:
The article explores the textual intricacy and the epistemological uncertainty projected by the 1999 novel of Peter Straub titled Mr. X. Already the title of the novel hints at secrecy, hidden identities, and cryptic messages. Indeed, the novel seems to be conceived as a cryptogram and a kind of literary “Russian doll”. This applies as much to the person of the narrator as to the construction of the narrative, its layers upon layers of secrecy and deception. This cryptic character of the novel is reinforced by the lack of closure and the ambiguity of the ending: “the fog” that shrouds everything as well as “undecipherable signs” that Ned meets along the way serve as a trope that projects the implied reader’s loss of certainty. The sense of being lost in the textual maze is compounded by the abundant instances of doubling and mirroring in the text. This is first observed in the composition of the novel as the dual-level narrative, consisting of the part narrated by Ned, and of the diary of Mr. X, the mysterious figure seen by Ned in his dreams, later revealed to be his father. The two, Ned and Mr. X, in their own peculiar ways act out the same pattern: Ned’s quest to find out the identity of his father is paralleled by his father’s (futile) attempts to confirm his own unearthly origins. However, duality is most powerfully expressed by the motif of doppelganger: Ned, the narrator, in time realizes the existence of his not-entirelyhuman brother, Robert, his “shadow self”, his “dark half”. Finally, even the novel, Mr. X, finds itself reflected in the work of fiction written by Mr. X, a story titled Blue Fire. Blue Fire, introduced at length into the narrative, serves as its specular text, mise en abyme, encapsulating its two central themes, as verbalized by the narrator: “the obsession with the ancestral house” and “the flight from and the pursuit of the Other”.

The main goal in gene therapy and biomedical research is an efficient transcription factors (TFs) delivery system. SNAIL, a zinc finger transcription factor, is strongly involved in tumor, what makes its signaling pathways an interesting research subject. The necessity of tracking activation of intracellular pathways has prompted fluorescent proteins usage as localization markers. Advanced molecular cloning techniques allow to generate fusion proteins from fluorescent markers and transcription factors. Depending on fusion strategy, the protein expression levels and nuclear transport ability are significantly different. The P2A self-cleavage motif through its cleavage ability allows two single proteins to be simultaneously expressed. The aim of this study was to compare two strategies for introducing a pair of genes using expression vector system. We have examined GFP and SNAI1 gene fusions by comprising common nucleotide polylinker (multiple cloning site) or P2A motif in between them, resulting in one fusion or two independent protein expressions respectively. In each case transgene expression levels and translation efficiency as well as nuclear localization of expressed protein have been analyzed. Our data showed that usage of P2A motif provides more effective nuclear transport of SNAIL transcription factor than conventional genes linker. At the same time the fluorescent marker spreads evenly in subcellular space.
