Opis:
In the article a new method of determining the so-called markers of inflexion, tense and mode of synthetic forms within the group of weak verbs was presented, and the whole concept is based on the innovative model of a linguistic analysis developed by Jozef Darski (1987; 22004). The model mentioned above is adapted for diachronic analysis, which allows to depict the development of the markers enumerated above from mid- to new-high-German.

Opis:
Rowing is a sport discipline, which requires extreme physical strength and endurance and appropriate aerobic and anaerobic capacity as well. However, when the workout intensity and load is very high, exercise is associated with temporary changes in cellular metabolism and the immune system. The study included one male rower aged 28 years - the highly-skilled and experienced athlete. We determined basic cardiorespiratory fitness measures, complete blood count, and 24 clinical chemistry parameters including relevant biochemical and haematological parameters and matrix metaloproteinases activities. Maximal exercise on the rowing ergometer induced 2-fold increase in absolute counts of all leukocytes subsets. There was observed an increase in C-reactive protein concentrations as well. MMP-9 activity increased 1,3-fold compared to the baseline value. Exhaustive exercise caused significant changes in creatinine and urea serum levels, but the most prominent changes were found in total and direct bilirubin concentrations. Maximal exercise induced also a decrease in the iron and magnesium levels. No changes in ALT, GGT and ALP activity were observed, while increase in CK, AST and LDH activity in post-exercise time and the decrease during the recovery was found. Therefore acute specialized movement on the rowing ergometer is not the cause of muscular damage, but rather indicate efficient adaptation to the physical exercise. Moreover, it seems that maximal exercise induces an inflammatory response characterized by greater count of all subpopulations of leukocytes, elevated levels of CRP and MMP-9 serum activity.

Opis:
The aim of the present study was to determine the effect of one-day wrestling tournament on magnitude and direction of changes in selected immunological and endocrine status indices. The participants of the study were male (n = 12) and female (n = 13) wrestlers. The earlobe blood samples and unstimulated saliva were collected at three time points: in the morning before the competition (1), immediately after the last match (2) and in the next morning (3). Total protein, IgA and cortisol concentration and α-amylase activity in saliva, as well as concentration of cortisol and interleukin-6 in serum were measured. Significantly lower ratio of sIgA/protein and significantly higher levels of salivary and serum cortisol, interleukin-6, and α-amylase activity were observed at the second time point compared with the first and the third in both groups of athletes. Regardless of the examined time point, concentrations of serum and saliva markers were similar in female and male wrestlers. However, a tendency to lower sAA activity on the next morning after the tournament compared to basal activity of this enzyme was seen in both examined groups. In general, the changes of examined markers were short-lived, except for α-amylase activity, showing that tournament-wrestling matches had no sustained negative effects on endocrine and immunological body systems.

Opis:
The analysis of the utility of serum SCC determination, combined with other tumor markers and cytokine assessment in cervical cancer patients, was performed. However, only CA 125 levels were found to relate to the histopathological type of the tumor, and were higher in adenocarcinoma. In addition, in patients with adenocarcinoma, CA 125 presented higher diagnostic sensitivity than CEA, standard tumor marker for cervical cancer. We have also shown that complementary IL-6 and SCC assessment in patients with cervical cancer improve the diagnostic sensitivity, independent of histopathological type. This would be especially important for early diagnostics and monitoring of patients. We have also demonstrated the possible use of bone metabolism markers in patients after oncological treatment with posttreatment osteoporosis, which is a common side effect of cancer treatment. Actually, bone metabolism markers join the array of tumor markers, to monitor osteoporosis treatment. For the monitoring purposes, these markers are more efficient than densitometry methods. Our preliminary results on osteocalcin, PINP and Ctx point at PINP as the most promising marker.
Do najczęściej wykorzystywanych markerów należą: osteokalcyna, PINP i Ctx. Wyniki naszych wstępnych badań sugerują jednak, że największą przydatność ma oznaczanie stężeń PINP.

Opis:
Cadmium is a toxic and carcinogenic heavy metal that nowadays constitutes a serious environmental health problem. The aim of this study is to review the effects of cadmium on selected inflammatory mediators and markers, such as NF-κB and AP-1 transcription factors, IL-6, TNF-α, IL-1β cytokines, IL-8 or MIP-2 chemokine, MPO, iNOS, MMPs and COX-2 enzymes, PGE2 (product of COX-2 enzyme), ICAM-1, VCAM-1 and PECAM-1 adhesion molecules, and CRP. The research strategy identified articles available in Medline, published between 1998 and 2012; we included both in vivo and in vitro studies carried out on humans and rodents. Most of the reviewed research findings suggest that cadmium in micromolar concentrations (especially in the 1-10 μM range) causes up-regulation of the mediators and markers of inflammation, and appears to have pro-inflammatory properties. However, it is worth mentioning that a contradictory or even opposite hypothesis exists, which suggests cadmium to be an anti-inflammatory factor. Further research including detailed histological analyses should solve this discrepancy. Nevertheless, it appears that the main reason for these contradictory findings is the experimental setup: different biological systems analyzed and different doses of cadmium applied.

Aims. The aim of the study was to assess the usefulness of TNF-α and CK-18 as diagnostic markers of alcoholic liver cirrhosis. Additionally, the effects of the stage of liver cirrhosis on concentrations of TNF-α and CK-18, as well as their correlation, were evaluated. Materials and method. Sixty-two patients with alcoholic liver cirrhosis treated in various hospitals were randomly enrolled. The stage of cirrhosis was assessed according to the Child-Turcotte-Pugh scoring system. The control group consisted of 31 healthy people without liver disease. Concentrations of TNF-α and cytokeratin-18 in blood plasma of patients and controls were measured using the sandwich enzyme immunoassay technique, with commercially available quantitative ELISA test kits. Results. The concentration of CK-18 was statistically higher in patients with alcoholic liver cirrhosis, compared to the control group. The concentration of TNF-α was significantly higher in patients with alcoholic liver cirrhosis, compared to the control group. Higher concentrations of TNF-α were found only in patients with stage C and B alcoholic liver cirrhosis, compared to healthy persons. Conclusions. The levels of TNF-α and total CK-18 were higher in patients with alcoholic liver cirrhosis than in healthy individuals. No correlation was found between the level of CK-18 and stage of liver cirrhosis.

Since about 20 years linguists attempt to apply the grammaticalisation concept to the description of pragmatic phenomena. In this context some linguists speak about ‘pragmatisation’ – a kind of language change, which concerns the linguistic codification of certain pragmatic contents. The term ‘pragmatisation’ suggests a new process, which runs in opposite direction to the development of grammatical elements. Irrespective of how we define the relation between both processes, this means always a reinterpretation (mostly extention) of the grammaticalisation concept. The present article focus on the question whether such attempts indeed result in a new insight into development of pragmatic units, or lead to terminological confusion by different terming already well described linguistic mechanisms.

Opis:
Sclerostin, which is a glycoprotein produced by osteocytes, reduces the formation of bones by inhibiting the Wnt signal pathway. Thyroid hormones are related with Wnt signal pathway and it has been reported that increased thyroid hormones in hyperthyroidism fasten epiphysis maturation in childhood, and increase the risk of bone fractures by stimulating the bone loss in adults. The aim of this study was to examine the sclerostin serum levels, the relation between sclerostin and thyroid hormones as well as the biochemical markers of the bone metabolism in patients with hyperthyroidism (including multinodular goiter and Graves' disease), whose treatments have not started yet. No difference was found in the serum sclerostin levels between the hyperthyroidism group (n=24) and the control group (n=24) (p=0.452). The serum osteocalcin levels and 24-hour urinary phosphorus excretion were found to be higher in the hyperthyroid group than in the control group (p<0.001, p=0.009). A positive correlation was determined between the sclerostin and bone alkaline phosphatase levels (p<0.001); a negative correlation between the osteocalcin and thyroid stimulating hormone (TSH) (p<0.05); a positive correlation between the osteocalcin and thyroid hormones (FT3,FT4) (p<0.001); and a positive correlation between the deoxypyridinoline and hydroxyproline (p<0.001). No correlation was determined between sclerostin and TSH,FT3,FT4 (p>0.05). Therefore, we consider that a long-term study that covers the pre-post treatment stages of hyperthyroidism, including both the destruction and construction of the skeleton would be more enlightening. Moreover, the assessment of the synthesis of sclerostin in the bone tissue and in the serum level might show differences.

The natural observation method was used to collect data from Kiswahili speakers in their normal daily conversations, which was then analyzed using Grice’s (1975) implicature theory. It was observed that the pragmatic marker tu conveys various implicatures depending on the context of use. These include expressing inadequacy or insufficiency, signaling dissatisfaction, showing contempt, inevitability and simplicity. Interestingly, the findings of the study also showed that intonation plays a role in determining the implicature invoked by this marker.

Challenges concerning fruit trees and grapevines as long lived woody perennial crops require adapted biotechnological approaches, if solutions are to be found within a reasonable time frame. These challenges are represented by the need for correct identification of genetic resources, with the foreseen use either in conservation or in breeding programmes. Molecular markers provide most accurate information and will be the major solution for questions about plant breeders rights. Providing healthy planting material and rapid detection of newly introduced pathogens by reliable methods involving serological and molecular biological tools will be a future challenge of increases importance, given the fact that plant material travels freely in the entire European Union. But also new breeding goals and transgenic solutions are part of the biotechnological benefits, e.g. resistance against biotic and abiotic stress factors, modified growth habits, modified nutritional properties and altered processing and storage qualities. The successful characterization of transgenic grapevines and stone fruit trees carrying genes of viral origin in different vectors constructed under ecological consideration, will be presented. Beyond technical feasibility, efficiency of resistance, environmental safety and Intellectual Property Rights, also public acceptance needs consideration and has been addressed in a specific project. The molecular determination of internal quality parameters of food can also be addressed by the use of biotechnological tools. Patient independent detection tools for apple allergens have been developed and should allow to compare fruits from different production systems, sites, and genotypes for their content of health threatening compounds.

In the official assessment of wart resistance the modified Glynne-Lemmerzahl method is used. A full cycle of assessment of the wart disease resistance requires 42 - 45 tubers per cultivar/breeding line. Forty two tubers are used in laboratory tests. To complete the laboratory tests, another 10 tubers are inoculated with winter sporangia of the fungus, using ring test. The final results are available after 3 years of investigation. If necessary, the full cycle of resistance tests to S. endobioticum can be performed during one year on 15 tubers in each of the 3 replications (45 tubers in total).Molecular verification of potato genotypes resistance to pathotype 1(D1) is possible with the use of SCAR marker Nl25-1400. Nevertheless, an official phenotypical assessment of advanced breeding lines, as the final verification of their resistance, is required.

Polish Markers (się, siebie)
Conceptual Distiction
Opis:
The article discusses reflexivity as a departure from the canonical event model and examines the distinction between two Polish markers of reflexivity: siebie and się. Both these markers code the direct reflexive situation type but semantically and syntactically they are not always interchangeable. This distinction is explained within Langacker's Cognitive Grammar framework and attributed to the difference in profile, trajector/landmark alignment and construal that these markers impose on a given situation. The article suggests that siebie functions as a nominal reflexive marker, which profiles a thing, and in a reflexive construction stands as the landmark of a relation. Thus siebie imposes the construal of volitionality and contrastiveness, and syntactically, it is obligatory as a prepositional complement and after verbs of perception. Się is a relational reflexive, and as such it profiles a relation, and specifies the identity of trajector and landmark of a relation, which become conflated in the single participant, i.e. the referent of the sentence subject. Consequently, the się-constructions are neutral with respect to intentionality of the profiled action, and conceptually simpler than siebie-constructions, which accounts for their higher frequency of occurrence.

Opis:
The aim of this study was to use a two-marker assay for the detection of breast cancer cells circulating in patients' blood. We have applied a PCR-based methodology to follow up the possibility of the development of metastatic disease in stage I and II patients who had undergone curative surgery. Since the number of circulating cancer cells in peripheral blood is very low, the technique for their detection needs to be not only highly sensitive, but also very specific. The reverse transcriptase-polymerase chain reaction (RT-PCR) technique may improve the sensitivity of breast cancer cell detection up to only a few cells per one million. The principle of the RT-PCR assay is to amplify a messenger RNA characteristic for breast epithelial cells in a blood sample. Since we do not expect such cells to be circulating in peripheral blood of healthy subjects, detection of the characteristic mRNA should indicate the presence of circulating breast cancer cells. We analyzed the usefulness of three mRNA markers: cytokeratin 19 (CK19), mammaglobin (hMAM) and β subunit of human chorionic gonadotropin (β-hCG) for this test. Blood samples (112) were obtained from 55 patients, in stages I and II, with or without metastasis to regional lymph nodes (N0 or N1). We found that a two-marker assay increases the sensitivity of detection of breast cancer cells in comparison with a single-marker one. Combination of two tumor-specific mRNA markers, hMAM/CK19 or β-hCG/CK19, allowed the detection of circulating breast cancer cells in 65% of N1 patients and 38% of N0 patients. By comparison, the combination hMAM/β-hCG allowed the detection of circulating breast cancer cells in the blood of 68% of N1 patients and 46% of N0 patients. Addition of the third marker did not significantly increase the detection sensitivity.

Forty eight inbred lines of winter rye, of various origin and pedigree, were analysed using 19 RAPD (random amplified polymorphic DNA) primers, 8 ISSR (inter-simple sequence repeats) primers and 13 SSR (simple sequence repeats) primer pairs. On the basis of particular marker types, there were created three separate dendrograms and one combined similarity tree, prepared on account of the whole data. Correlation coefficients for individual technique based on genetic similarity matrices were not significant. By comparing the GS data obtained on the basis of singular methods with collective matrix, it was observed that the highest correlation rate was for ISSR method (r=0.68). The utility of each marker technique was compared by using marker index MI. Diversity detecting index (DDI) was suggested in the paper, which may prove helpful in planning and comparing researches on phenetic relationships...

pragmatic markers, spoken Swahili, contextualization theory
Opis:
This study examines the pragmatic functions of the marker sawa in spoken Swahili. The data have been obtained from informal conversations made by Swahili speakers in informal social settings. These settings include “vijiwe vya kahawa” (setting of informal conversations created around people drinking coffee) and "vijiwe vya mamantilie” (setting of informal conversations created around women preparing and selling food on the streets). The analysis of the data, performed within the framework of the contextualization theory (Gumperz 1982), shows that, apart from its basic connotation of agreement, the marker sawa conveys other meanings, depending on these very contexts of communication, and therefore acquires also various pragmatic functions. The pragmatic functions identified in this work include: to show that the speaker agrees with what has been said but on a condition (I agree, but…), as a receipt marker, as a tag-positioned-comprehension check, as an answer to the question showing that the speaker has understood what has been said (Yes, I understand), as a continuer, as a negative releasing marker, and as a gap filler. Interestingly, the study shows also that intonation and other paralinguistic features (like gestures) play a role in determining the pragmatic functions of this marker. The article concludes that the pragmatic markers in spoken Swahili are rich in meanings and are used to show speakers’ attitudes and emotions, therefore manifesting a deep and meaningful interconnection between the language and its contextualized experience.

Opis:
Breast cancer is a very serious disease from both an economical and social point of view, because of its high incidence and mortality, as well as its prevalence. Genetic and epigenetic changes as well as the resultant deregulation of mechanisms that affect cellular processes are reflected in the final stage, like significant variability in phenotype. These changes have multiple factors in clinical, morphological but also molecular terms, and they greatly affect the possibility of accurate diagnosis of breast cancer in its early stages. Since no combination of clinical examination, mammography and ultrasonography together with CT and MRI can achieve 100% sensitivity, clinical practice is supported with diagnostic methods based on the monitoring of tumour markers with the emphasis on molecular markers. This review describes the most common molecular markers already used for earlier clinical diagnostics and also proposes new possible markers from the tumour vascular markers group. Any new oncomarkers to improve diagnostic sensitivity and specificity will be of great benefit, and in clinical practice, they may provide an opportunity for early diagnostic and therapeutic intervention, and subsequently better prognosis for the patient as well.
Niniejsza recenzja opisuje najbardziej typowe markery molekularne wykorzystywane we wczesnej diagnostyce klinicznej, a także proponuje nowe potencjalne markery z grupy markerów nowotworowych TVMs . Wszelkie nowe markery nowotworowe poprawiające czułość i dokładność diagnostyki będą bardzo pomocne, a w praktyce klinicznej mogą pozwolić na wczesną diagnostykę i leczenie, co polepszy także rokowania pacjenta.

Background: The entry and dispersal of modern humans is India of remains unclear and extending with many interesting evidences South India, assumed to be a major corridor for their with many ancient genetic deposits such as Dravidian tribal with Negrito features. As the relationships between the genetic polymorphs and diseases in human being revealed globally, it is worthy to investigate the genomic architecture of population in south India. Objective: To examine what evolutionary forces have most significantly impacted south Indian tribal genetic variation, and to test whether the phenotypic similarities of some south Indian tribal groups to Africans represent a signature of close relationship to Africans or are due to convergence. Methods: Blood samples from 193 unrelated individuals of both sexes are drawn from the Dravidian tribal settlements of Tamil Nadu and Kerala. South India are genotyped for four Aluindel (Alu FXIIIB, Alu ACE, AluTA25 and Alu PLAT) allele profile by PCR genotyping method. Results: All loci are highly polymorphic and average heterozygositiesare substantial (range: 0.37-0.44). Genetic differentiation is high (Gst = 3.7%) in all the study populations.

Opis:
Introduction and objective. Bone sialoprotein (BSP) is present in a non-collagenous fraction of bone matrix, in dentine, cement and calcified cartilage. The characteristic place where sialoprotein is produced makes it highly specific in relation to the bone tissue and the possibility of using it as a marker of bone metabolism. The study aimed to assess the BSP level in the saliva of female patients at the menopausal and postmenopausal period. Materials and method. The study included 71 women aged 45–74 years. In the examined subjects, bone mineral density and bone sialoprotein levels in saliva were determined. Patients also responded to survey questions about place of residence and the frequency of consumption of foods rich in calcium. The obtained results were statistically analyzed. Results. In the study population of women, BSP saliva concentration ranged from 0.40–14.97 ng/ml. The highest BSP values were observed in the control group (average – 5.66 ng/ml, median – 6.73 ng/ml,), the lowest in the osteopenia group (average – 3.09 ng/ml, median – 1.71 ng/ml). This relationship was close to statistical significance (p = 0.056). Place of residence of the surveyed women had neither effect on BSP concentration in saliva nor on the value of the T-score ratio. Similarly, there was no statistically significant relationship between the frequency of consumption of foods rich in calcium and BSP and T-score. Conclusions. It was found that the BSP level can be determined in saliva. Biochemical tests of saliva considering bone metabolism are an issue that should be continued in future, as the availability of material (saliva) for laboratory analyses, carries potential diagnostic opportunities.

The starting hypothesis of the article is that the discursive operators of Spanish have not been enabled by grammaticalization processes identical to those of other grammatical components (such as verbal or nominal morphemes or prepositions and conjunctions), since there has been no reduction in the functional scope nor a total bleaching, so that its link with the original lexical items has not disappeared. The consequence is that markers are not integrated into paradigms as closed as grammatical paradigms and both their obsolescence and their renewal, therefore, follow similar patterns to what happens in the evolution of the lexicon. This hypothesis is tried to verify in the article by means of the examination of some discursive markers that, having been used diachronically in Spanish, have been abandoned ina relatively short time.

Introduction. Inflammatory response in Diabetes Mellitus (DM) begins with chronic sub-clinical inflammations as a result of insulin resistance and activation of both innate and adaptive immune system as the disease progresses to complicated diabetes. Hence, the present study investigated the neutrophil phagocytic, complement function (CH50), and some cytokine profiles among diabetic and non-diabetic patients attending the National Hospital in Abuja, Nigeria. Aim. To evaluate the neutrophil phagocytic, complement function (CH50), and some cytokine profiles among post-operative septic diabetic and post-operative septic non-diabetic patients at the National Hospital in Abuja, Nigeria. Material and methods. Subjects were recruited by convenient sampling technique through interviewer-administered questionnaires. Subsequently, blood samples were collected. Fasting blood sugar (FBS) (mmol/L) was determined using glucose oxidase method. Neutrophil function test (Fmol/phag) was assayed using nitroblue tetrazolium reduction test (NBT). Hemolytic complement function (CH 50) test was conducted using serum harvested from sheep sensitized with human group (ORh D +ve) red blood cells. While serum Interleukin-4, -6, -10 and TNF- α were determined using Enzyme Linked Immunosorbent Assay (ELISA). Results. Mean ± Standard deviation (SD) of FBS concentration of 10.5 ± 1.3 (mmol/|L) among diabetic and 4.7 ± 0.9 (mmol/L) among non-diabetics was recorded. There is a decrease in neutrophil phagocytic function with a mean ± SD of 5.4 ± 2.1 (Fmol/ phag) in diabetics compared to 9.2 ± 2.1 (Fmol/phag) in non-diabetics. Similarly, complement (CH 50) function and C-reactive protein were significantly lower in diabetics when compared to non-diabetics (p<0.001). There was a significant difference in IL-6 concentration between diabetics and non-diabetics groups, but no significant difference was observed in TNF-α, IL-4 and IL-10 concentrations between study groups (p>0.05). TNF-α and IL-6 was significantly higher in diabetics with cardiovascular disorders compared to non-diabetics subjects with cardiovascular disorders (p<0.001). Conclusion. Findings from this study revealed the association of complement, neutrophil phagocytic function, CRP and IL-6 among septic diabetic patients,. In addition TNF-α and IL-6 expression was higher in DM patients with cardiovascular disorders.

Opis:
Keratoconus (KC) is a corneal disease associated with structural abnormalities in the corneal epithelium, Bowman's layer and stroma and altered concentration of tear components. KC corneas show a different pattern of collagen lamellae than their normal counterparts. Also, a reduction of several collagen types in KC epithelium and stroma was observed. Altered expression and/or activity of lysyl oxidase, a critical enzyme of the biogenesis of connective tissue detected in KC corneas, may weaken covalent bonds between collagen and elastin fibrils, what may lead to biomechanical deterioration of the cornea. Increased activity of matrix metalloproteinases observed in KC may induce the degradation of the extracellular matrix causing damage to the cornea. Oxidative and nitrative stress play an important role in KC pathogenesis and KC corneas are characterized by the disturbed lipid peroxidation and nitric oxide pathways. Malfunctioning of these pathways may lead to accumulation of their toxic by-products inducing several detrimental effects, along with apoptosis of the corneal cells, which may result from the loss of β-actin or increased levels of cytokines, including interleukin-1 and -6. Change in the expression of genes associated with wound healing, including the nerve growth factor and the visual system homeobox 1, may contribute to increased susceptibility of KC corneas to injury. Consequently, biochemical changes may play an important role in KC pathophysiology and, therefore, can be considered in prevention, diagnosis, prognosis and in the therapy of this disease as well.
