Opis:
The codling moth, Cydia pomonella (L.), is the key pest of apple production worldwide. In Morocco, there is a sustainable presence of codling moth causing considerable damage in apple orchards despite frequent applications of broad spectrum insecticides. For 12 years, sexual trapping and chemical control were performed and the development of the codling moth population was analysed in an orchard which was in the region of Azrou. The efficacy of some insecticides (azinphos-methyl, chlorpyriphos-ethyl, diflubenzuron, thiacloprid, methoxyfenozide, spinosad, and deltamethrin) was also evaluated on neonate larvae and compared with a laboratory sensitive strain. This procedure was done to assess an eventual resistance in Moroccan populations. The action threshold was usually exceeded, leading to an intensive chemical control, with an average frequency of 9 to 13 days. The chemical control was done according to the action persistence time of the insecticides and the trap captures. However, those two parameters are compromised in Moroccan conditions because of the high summer temperatures which disrupt the action of insecticides and exacerbate populations. The pheromone traps may become ineffective and useless. Neonate larvae were resistant to five insecticides out of seven. Such results suggest the presence of a cross resistance in local strains. Overall, the insect resistance, the functioning of the sexual traps, and some insecticides properties (persistence action, pre-harvest interval) are the key factors that could explain the failure to control these moths under Moroccan conditions.

Effective control of Leptinotarsa decemlineata remains an urgent problem for agriculture worldwide. Minimization of the use of non-selective neonicotinoid insecticides, such as thiomethoxam, is an actual vector of development of potato cultivation. In this rapid communication, we show the prospect of the topical use of short unmodified antisense fragment of L. decemlineata CYP6B gene as a DNA insecticide. Investigated parameters, namely, number of larvae per plant, aboveground biomass, yield and number of potatoes produced per plant indicate the possibility of this post-genomic approach as a safe and effective method of L. decemlineata control.

Demographic toxicology is recommended for toxicity determination of the long term effects of a pesticide since it gives a more accurate and efficient measure of the effect of a pesticide. Thus, in the current study the sublethal effects of pirimicarb (carbamate insecticide) two concentrations of LC30 and LC10 were used against third instar larvae of Hippodamia variegata (Goeze) in order to determine the effects of the pesticide on demographic parameters of the predator under laboratory conditions. Results showed that pirimicarb did not affect individual life parameters such as development time of larva, pupa, adult longevity, female and male longevity, adult preoviposition period (APOP), and total preoviposition period (TPOP). However, population parameters such as intrinsic rate of increase (r), net reproductive rate (R0), mean generation time (T), and finite rate of increase (λ) was affected by sublethal treatment. For example, intrinsic rate of increase (r) was 0.18 day–1 in the controls but it was 0.13 and 0.14 day–1 in the treated insects with LC10 and LC30 concentrations, respectively. Also, there were significant differences between mean generation time (T) of the treatments and the controls i.e. mean generation time of the controls was 29.03 days while mean generation time in the two treatments of LC10 and LC30 was 33.93 and 31.66 days, respectively. The finite rate of increase was also significantly affected by sublethal effects of the pesticide. The results showed that pirimicarb, even at low concentrations, has potential to adversely affect the predatory ladybird, therefore care should be taken when this insecticide is used in the Integrated Pest Management (IPM) program.

In 1992-1994 studies were carried on the noxiousness, biological effectiveness and effects of chemical control of cereal leaf beetles (Qulema spp.) on the spring wheat. The larvae most frequently injured flag leaves, damaging them to 74.8%. The reduction of flag leaves surface area ranged 20.8-38.0%. The use of insecticides resulted in the increase of the grain yield by 12.5% on an average.
W latach 1992-1994 przeprowadzono badania nad szkodliwością larw skrzypionek dla roślin pszenicy jarej, biologiczną skutecznością zastosowanych insektycydów oraz efektami produkcyjnymi wykonanych zabiegów. Stwierdzono dużą szkodliwość skrzypionek dla pszenicy jarej. Larwy skrzypionek uszkodziły liście flagowe pszenicy jarej od 56,6% do 92,0%, średnio 74,8%. Redukcja powierzchni liści flagowych wynosiła od 20,8% do 38,0%. Otrzymane wyniki badań wskazują na zróżnicowanie podatności badanych odmian pszenicy jarej na uszkodzenia przez skrzypionki. Uzyskano bardzo dobrą skuteczność zwalczania larw skrzypionek w pszenicy jarej. Najlepszymi insektycydami w ich zwalczaniu były: Karate 025 EC, Fastac 10 EC, Bulldock 025 EC, Alpha Combi 26,25 EC. Stosowanie insektycydów spowodowało przyrost plonu ziarna od 0,4% do 24,8%, a liczba ziaren z kłosa uległa zwiększeniu średnio o 21,1%. Znaczący wpływ zastosowanych insektycydów zaznaczył się w stosunku do masy ziaren z kłosa, w porównaniu do kontroli wzrosła ona od 1,4% do 38,3%. W stosunku do kontroli nastąpił również wzrost masy 1000 ziaren. Wahał się on od 4,0% do 28,4%.

Control failure of pests and selectivity of insecticides to beneficial arthropods are key data for the implementation of Integrated Pest Management (IPM) programs. Therefore, the aim of this study was to assess the control failure likelihood of Plutella xylostella and the physiological selectivity active ingredients to parasitoid Oomyzus sokolowskii (Hymenoptera: Eulophidae) and to predators Polybia scutellaris (Hymenoptera: Vespidae) and Lasiochilus sp. (Hemiptera: Anthocoridae). In bioassays, P. xylostella larvae and O. sokolowskii, P. scutellaris and Lasiochilus sp. adults were used. Concentration-mortality curves of six insecticides for P. xylostella were established. These curves were used to estimate the mortality of P. xylostella at the recommended concentration, in order to check a control failure of insecticides to this pest. Furthermore, the lethal concentration for 90% of populations (LC90) and the half of LC90 were used in bioassays with the natural enemies to determine the selectivity of these insects to insecticides. All tested insecticides showed control failure to P. xylostella, indicated by high LC90 and low estimated mortalities (less than 80%). The cartap insecticide was selective in half of LC90 to Lasiochilus sp. and moderately selective in LC90 and the half of LC90, to Lasiochilus sp. and P. scutellaris, respectively. Deltamethrin was moderately selective in the half of LC90 to predator Lasiochilus sp. Cartap, carbaryl, and deltamethrin reduced the mortality of Lasiochilus sp. in the half LC90. The results also showed that the insecticides methamidophos, carbaryl, parathion methyl and permethrin were not selective to any of the tested natural enemies. The role of insecticides in IPM systems of Brassica crops is discussed based on their control failures to P. xylostella and selectivity to their natural enemies.

Efforts have been made during the past three decades to develop insecticides with selective properties that act specifically on biochemical sites present in a particular insect group, but whose properties differ from those present in mammals. This approach has led to the discovery of compounds that affect the hormonal regulation of molting and developmental processes in insects such as ecdysone agonists, juvenile hormone (JH) mimics and chitin synthesis inhibitors. The search for potent acylureas has led to the development of novaluron (Rimon) developed by Makhteshim Chemical Works. The LC-50 value of novaluron on 3rd-instar Spodoptera littoralis fed on treated leaves is approximately 0.1 mg a.i./liter. This value resembles that of chlorfluazuron and is tenfold lower than that of teflubenzuron. Novaluron affects nymphs of Bemisia tabaci more than chlorfluazuron and teflubenzuron. Artificial rain, at a rate of 40 mm/h applied 5 and 24 h after treatment in a cotton field had no appreciable effect on the potency of novaluron on both S. littoralis larvae and B. tabaci nymphs. Hence, novaluron can be used in tropical areas and during rainy seasons. In general, benzoylphenyl ureas had no direct effect on parasitoids and phytoseiids and are considered mildly affect other natural enemies. Novaluron has no cross-resistance with conventional insecticides, the JH mimics pyriproxyfen and neonicotinoids. As such, it is considered an important compound in pest management programs.

The objectives of this work were to describe the residual behaviour of chlorpyrifos (insecticide) and chlorothalonil (fungicide), applied for the protection of carrot and parsley plantations. The field tests were carried out at farms located near Rzeszów (South-Eastern Poland) on processing carrot and parsley destined for baby food production. The results obtained indicated that disappearance rate of chlorothalonil on carrot leaves was significantly faster and its half-life time was about 7 times shorter than that of chlorpyrifos. In weather conditions of 2006, chlorpyrifos residues in rape carrot and parsley roots were reduced to levels lower than MRL and even the rigorous value of 0.01 mg/kg.

Cry4Aa 678 amino acids fragment (60 kDa) was cloned into Escherichia coli. After induction with IPTG the 60 kDa Cry4Aa fragment was purified by Ni chromatography, separated by SDS PAGE and identified by mass spectrometry (MS/MS). The 60 kDa Cry4Aa fragment exhibited high toxicity towards Ae. aegypti larvae. The earlier results [1] show that Pichia pastoris yeast cells expressing tmfA (synthetic gene coding for the Trypsin Modulating Oostatic Factor of Ae. aegypti) together with E. coli cells expressing Bti toxin genes (cry4Aa, cry11Aa, cyt1Aa and p20) are synergistic. Therefore, P. pastoris, which synthesizes high amounts of heterologous proteins was genetically engineered to produce TMOF and Cry4Aa. Codon-optimized synthetic genes, cry4Aa-tmfA, gst-cry4Aa-tmfA, tmfA and gfptmfA that were expressed by P. pastoris and fed to Ae. aegypti larvae caused 90% mortality. GST (glutathione-S-transferase) enhanced the activity of Cry4Aa-TMOF and protected it from heat denaturation and GFP (Green Fluorescent Protein)- TMOF allowed us to follow yeast cells consumption by individual larva using fluorescent microscopy. This report shows for the first time that 60 kDa Cry4Aa and TMOF expressed together in P. pastoris are highly toxic to Ae. aegypti larvae.

Neuropeptides are regulators of critical life processes in insects, but are subject to rapid degradation by peptidases in the hemolymph (blood), tissues and gut. This limitation can be overcome via replacement of peptidase susceptible portions of the insect neuropeptides with non-natural residues or moieties to create analogs with enhanced biostability. Two neuropeptide families, the insect kinins and insectatachykinins, stimulate gut motility and Malpighian tubule fluid secretion in certain insects but unmodified members demonstrate little or no effect when fed to pea aphids (Acyrthosiphon pisum) in an artificial diet. Nonetheless, biostable analogs developed via the strategic introduction of either bulky Aib residues and/or β-amino acids demonstrate potent antifeedant and aphicidal effects when administered orally; whereas other biostable analogs are inactive. Although the precise mechanism of action has not been delineated, the activity may be associated with disruption of the physiological processes that these neuropeptides regulate in insects. The most active of the biostable insect kinin and insectatachykinin analogs show LC50 values of 0.063 nmole/μl (LT50 = 1.68 days) and 0.0085 nmole/μl (LT50 = 1.1 days), respectively; matching or exceeding the potency of some commercially available aphicides. The biostable analogs represent important leads in the development of alternative, environmentally sound aphid control agents.
