FURNITURE BEETLES - FORMALIN FUMIGATION
WOOD CONSERVATION
Opis:
This work gives the attention to a problem of furniture beetles fumingation with formalin. Steams of formaldehyde after four full days of fumingation in temperature of 21°C degrees gave 100% of mortality amongst larvas of furniture beetle, increasing the temperature by 30°C degrees shortened the time of fumingation up to twenty four hours. Formalin is the 40% solution of formaldehyde in water. The main problem is the increase of humidity in gas chamber amounting to 54% at the beginning and going up to relative humidity of 100% within 5 hours. The influence of the steams on gilts and polichromes was also tested. After two weeks of fumingation with formaldehyde there were found dark stains on the gilts. Also on one of polchromed samples there was found resin spil in the spot where the knot was.

Opis:
The potential carcinogenicity of formaldehyde (FA) has prompted increasing preventive measures in anatomic pathology (AP) laboratories and new strategies aimed at innovating airborne FA monitoring systems. This review provides an updated overview of the most recent improvements in preventive measures, safe practices, and exposure monitoring tools in the FA usage and handling. A computer-based search of scientific and non-scientific sources was performed on PubMed, Web of Science, Google and Google Patents databases, querying the main topics of real-time, in-continuous FA monitoring instruments for sale, and commercially available tools for improving preventive measures in formalin management. In order to simplify the sampling process and to choose a better analytic solution to FA assessment, the main characteristics of each FA monitoring instrument were described. The novel technical tools recently introduced on the global market, aimed at reducing FA emissions in AP laboratories, were summarized. This review is directed at anatomic pathologists to draw their attention to the rapidly growing field of safe formalin practices. A repeated exposure assessment is recommended to evaluate technical changes in air monitoring programs to keep FA emissions low, in compliance with the limit value; thus, evolved monitoring devices are needed.

Introduction: This study was conducted to correlate the personal formaldehyde (FA) exposure levels of instructors and students with the indoor FA concentrations in gross anatomy laboratory at King Saud University. The personal FA levels of instructors and students are higher than the indoor FA concentration in the gross anatomy laboratory. Materials and Methods: The gross anatomy laboratory at college of medicine, King Saud University Riyadh, was observed for indoor FA concentration and the personal exposure levels of instructors and the medical students during the 4th, 10th and 14th weeks of the dissection sessions. All air samples were collected by the diffusive sampling device and analyzed by using high performance liquid chromatography (HPLC). Results: The personal exposure level of FA was higher than the indoor concentration, and the personal exposure levels of instructors were higher than that of the students. The concentration of FA was also higher in the center of the room than the corners and near the doors. Conclusion: Both the indoor FA concentrations and personal FA exposure levels are higher near the dissecting table than at locations away from it during the gross anatomy laboratory sessions. Thus, the instructors and students are exposed to the higher concentration of FA than the general population.

Opis:
SNP-minisequencing has become common in forensic genetics, especially for analysing degraded or low copy number DNA (LCN DNA). The aim of this study was to examine the usefulness of five SNP (single nucleotide polymorphism) markers for analyzing degraded and LCN DNA recovered from archival samples. DNA extractions of eight formalin-fixed paraffin-embedded (FFPE) tissues were performed and DNA fragments were amplified in one multiplex PCR (polymerase chain reaction). SNPs were identified in a minisequencing reaction and a gel electrophoresis in ABI Prism® 377 Sequencer. The research confirmed the usefulness of SNP-minisequencing for analysing FFPE tissues.

Opis:
To evaluate the effect of formalin disinfection on the oxidative stress biomarkers in the cardiac and hepatic tissue, rainbow trout (Oncorhynchus mykiss Walbaum) were assigned into test and control groups. The test group was disinfected by formalin in final concentration 200 mL per m3. Fish were bathed for 20 min, three times, every 3 days. Two days after the last bathing fish were sampled. 2-Thiobarbituric acid reactive substances (TBARS) and carbonyl derivatives of protein oxidative destruction, as well as antioxidant defense biomarkers (superoxide dismutase, catalase, glutathione reductase, glutathione peroxidase activity, total antioxidant capacity) were determined. The formaldehyde- exposed animals showed decrease of aldehydic and ketonic derivatives of oxidatively modified proteins and increased superoxide dismutase activity in the hepatic tissue compared to untreated group. In cardiac tissue, TBARS level, aldehydic and ketonic derivatives of oxidatively modified proteins were increased in formalin-exposed trout and down-regulated antioxidant status versus control group. It could be concluded that the disinfection of rainbow trout by formalin not contributed to the hepatic injury and may not be toxic. Toxic effect to cardiac tissue was more considerable.
Określono poziom biomarkerów stresu oksydacyjnego (produktów reagujących z kwasem 2-tiobarbiturowym (TBARS) i karbonylowych pochodnych oksydacyjnie zmodyfikowanych białek) oraz obrony antyoksydacyjnej (aktywności dysmutazy ponadtlenkowej, katalazy, reduktazy glutationowej, peroksydazy glutationowej, całkowitej zdolności antyoksydacyjnej).

Opis:
Novel aryl/alkyl azole derivative compounds C1 and C2 were screened for in vivo anti-nociceptive and anti-inflammatory activities by using the hot plate test and formalin-induced hind paw edema test, respectively. Ability of these compounds to inhibit cyclooxygenase-1/2 (COX-1/2) and 5/15-lipoxygenase (5/15-LOX) were evaluated in vitro by using a colorimetric method. C1 (30 and 50 mg/kg, i.p.) and C2 (3 and 30 mg/kg, i.p.) increased the latency to withdrawal in the hot plate test, indicating an anti-nociceptive activity. C1(30 and 50 mg/kg) and C2 (3,30 and 50 mg) were able to decrease formalin-induced edema, indicating their anti-inflammatory properties. C1 and C2 exhibited inhibitory effects on the activity of COX(1-2) and 15-LOX. However, neither C1 nor C2 showed an inhibitory effect on 5-LOX. This study demonstrates that C1 and C2 have anti-nociceptive and anti-inflammatory activities, that is partially mediated by inhibition of COX and LOX enzymes. Our results suggest that C1 and C2, novel aryl/alkyl azole compounds, could serve as lead compounds to develop novel therapeutic options for the treatment of pain and inflammation.

Opis:
Deep freezing and storing in formalin are some of the common techniques of human tissue preservation. However, the preservation modes affect the biomechanical properties of the tissues. In this work, the effects of the above-stated preservation techniques are compared with that of fresh cadaveric samples. Methods: FATC samples from male cadavers of age between 60 and 70 years were tested under tensile loading at a strain rate of 0.8 s–1. Fourteen FATC samples from soft embalmed cadavers were preserved for 3 weeks by two methods: (a) 10% formalin and (b) deep freezing at –20 C followed by thawing. Seven FATC samples from fresh cadavers were experimented as control samples. The results were evaluated by a two-stage statistical process of Kruskal–Wallis H test and Mann–Whitney U-test. Results: It was observed that the failure force of fresh cadavers was the highest while that of preserved samples were approximately half the value. Failure elongation of frozen samples exceeded fresh samples while formalin samples failed at lesser elongations. Higher incidence of tibial insertion point or mid-section failures were observed in fresh samples while the higher incidence of ruptures at femoral insertion point was observed in the two preservation methods. Conclusion: Tensile properties of fresh tissues vary significantly from that of formalin preserved or frozen preserved samples.
