Established taxa within the Enterobacteriaceae wereisolated from cloacal swabs of Strix aluco chicks in nest boxes located at five research sites. ChromID ESBL medium (bioMerieux) was used to select a pool of Enterobacteriaceae strains producing extendedspectrum beta-lactamases. Drug sensitivity of the chosen strains was determined from the full pool of Enterobacteriaceae to 6 chemotherapeutics of different mechanisms of action. The study evaluated the sensitivity of ESBL-synthesizing isolates to substances belonging to penicillins, cephalosporins, cephamycins, clavams, carbapenems and monobactams. Analysis of the results indicated a potential role of Strix aluco in the dissemination of epidemiologically-relevant Enterobacteriaceae, and, importantly, pose health risks to forest service workers, nature protection service and ornithologists. The results can also serve as the basis for further environmental studies.

In the study we tested drug sensitivity to 3 carbapenems (doripenem, imipenem and meropenem) of Gram-negative clinical isolates from Southern Poland. Material and methods. 89 strains were examined: 42 from Pseudomonas genus, 16 Acinetobacter baumannii strains and 31 Enterobacteriaceae strains. Etests were used according to the producers instructions, MIC values were interpreted using EUCAST criteria. Results. Highest in vitro activity against Pseudomonas spp. was shown for doripenem, then meropenem and the lowest for imipenem (MIC values were definitely lower for doripenem; differences were statistically significant); A. baumannii strains showed similar sensitivity to doripenem, meropenem and imipenem (differences non-significant); all Enterobacteriaceae strains showed sensitivity to the tested antimicrobials. Conclusions. As a conclusion-doripenem, which has high in vitro activity (almost the same as imipenem and meropenem) as well as beneficial pharmacologic properties, may be an alternative solution in the treatment of multiresistant Gram-negative bacteria, especially in patients in severe status who require restrictive antibiotic regimens.

Introduction and objective. Infections due to multidrug-resistant (MDR) Enterobacteriaceae are an ongoing global threat in their management. The aim of the study was to investigate the antiimicrobial resistance (AMR) and virulence gene profiles of MDR Gram-negative isolates in Sokoto, north-west Nigeria. Materials and method. A total of 578 clinical samples were collected from patients. Suspected Gram-negative bacteria were isolated from these clinical samples: vaginal swab, pus, stool, blood, wound swab and urine, using Gram-staining and conventional biochemical reactions. These isolates were further identified with an identification kit (Microgen-GN-A), and tested against a panel of 11 antibiotics. A single polymerase chain reaction (PCR) assay targeting 13 virulence gene related to adhesion (fimH, papC, and sfaS), iron chelation (iutA, and fyuA), toxins (astA, stx1, stx2, and eaeA), biofilm (bssS), and serum resistance (traT, iss, and kapsMTII) encoding genes were evaluated. Results. A total of 276 Gram-negative isolates were identified using the Gram stain and biochemical reactions. These organisms were further confirmed with identification kit. Of the 276 isolates, 36 organisms of interest (23 Escherichia coli, 4 Klebsiella pneumoniae and 9 Proteus mirabilis) were identified. Other Gram-negative isolates accounted for the remaining 86.9%. The majority of the isolates were resistant to cefixime (100%) and partially resistant to amikacin (19.4%).The virulence genes bssS (58.3%), fimH (44.4%), and iutA (44.4%) were the most prevalent, whereas kapsMTII (5.6%) and stx2 (2.8%) were least detected, while astA was not detected in any of the isolates. Conclusion. The study elucidated the prevalence of antibiotic resistance and virulence genes in Gram-negative bacteria from clinical isolates in Sokoto, north-western Nigeria. The majority of the isolates were MDR, thereby posing a public health risk.

Foodborne microorganisms harbor and adheres itself to the food material and surrounding surfaces for a long time and influence the food quality and consumers health. Among these microbes the presence of Enteric indicator bacteria in food premises confers the indication of entero-pathogens, i.e. E. coli, Salmonella, Shigella and Campylobacters that could cause severe systemic infections in consumers. In this study, the hygienic status of confectionery and supplementary food processing facility was evaluated. A total of 10497 examinations were performed on 3499 swab samples collected from food premises and handlers for the analysis of Enteric indicator bacteria. From swabs, 1277 (12.2%) isolates were identified in which Enterobacteriaceae were found with higher frequency 604 (47.3%) followed by Coliforms 293 (30.8%) and Escherichia coli 280 (21.9%) respectively. The mean count (CFU/cm2) was found maximum for plain surfaces (floors, walls and door), while the lowest was for equipment and machinery. Overall isolates percent prevalence was determined where Enterobacteriaceae were 47%, Coliforms 31% and Escherichia coli 22%. Majority of the floor surfaces were highly contaminated, where washing and sanitation practices were observed to be inappropriate. Worker hygiene status was lacking essential food safety and hygiene standards. In general, the Enteric bacteria were found with higher ratio, that could affect the food quality and quantity both to a greater extent with some influences on consumers health.

This study was conducted to determine the occurrence, antibiotic susceptibility pattern, and plasmid profile of multidrug-resistant (MDR) Enterobacteriaceae isolated from soymilk hawked in the Ibadan Polytechnic Community, Nigeria. Bacterial isolation and identification were performed using a culture-dependent method and biochemical characterization, respectively, while antibiotic susceptibility was tested using the disk diffusion method. Plasmid analysis and curing were performed using standard procedures. The results revealed the following occurrence rate of bacterial species in soymilk samples collected from all axes (South, North, East, West, and Central) of Ibadan Polytechnic: Citrobacter spp., 64%; Klebsiella spp., 7%; Enterobacter spp., Escherichia coli, and Serratia spp., 6% each; Proteus spp., 5%; and Salmonella spp. and Shigella spp., 3% each. The highest heterotrophic bacterial count of 9.3 × 103 CFU/ml was recorded at Ibadan Polytechnic North and South, while the least count (3.2 × 107 CFU/ml) was recorded at Ibadan Polytechnic Central. The highest enteric bacterial count of 1.3 × 103 CFU/ml was recorded in soymilk samples from Ibadan Polytechnic Central, while the least count of 1.7 × 107 CFU/ml was recorded in soymilk samples from Ibadan Polytechnic North. A total of 26% of the isolates showed resistance to cefpodoxime, while 88% of the isolates were susceptible to trimethoprim-sulfamethoxazole. Four of the MDR isolates possessed plasmid bands ranging from 6 to 1 with molecular weights from 2.7 to 17.2 kbp. Enterobacteriaceae such as isolates O14 (Klebsiella spp.) and B10 (E. coli) retained their resistance to antibiotics even after removal of plasmids, while isolates S13 (Citrobacter spp.) and O4 (Shigella spp.) were susceptible to some antibiotics after curing. Conclusively, soymilk sold in the sampled areas was highly contaminated with Enterobacteriaceae, thereby indicating poor hygiene standards of soymilk production.

The aim of this study was to investigate the bacteriological quality of 5 kinds of vegetables (lettuce, dill, radish, beetroot, carrot) and their rhizosphere soil, originating from conventional farms located in the Lublin Province of Eastern Poland. A total number of 35 samples of fresh vegetables (FV) taken immediately from soil, 35 samples of soil from rhizosphere of these vegetables (SR) and 35 samples of vegetables sold at retail in the markets in Lublin (VR) were examined. The samples were analysed for the content of: aerobic mesophilic bacteria (AMB) grown at 30 °C and 37 °C, Gram-negative bacteria of Enterobacteriaceae family, faecal coliform (FC) bacteria, Salmonella spp., and Clostridium perfringens. Median AMB values determined at 30 °C for FV, SR and VR were 5.27, 5.00, and 5.00 log10 CFU g-1, respectively, being significantly greater compared to those recorded at 37 °C. The exceeding of the threshold value of 6.0 log10 CFU g-1 proposed by Gelosa (1998) was noted only in 5 FV samples grown at 30 °C (14.3%), and in 3 FV samples grown at 37 °C (8.6%). The threshold value was never exceeded in SR and VR samples. Median concentrations of Enterobacteriaceae determined for FV, SR and VR were 4.03, 3.87, and 3.04 log10 CFU g-1, respectively. Eleven species of Enterobacteriaceae were identified in the FV, SR and VR samples. The percent of samples containing Escherichia coli was greatest for VR (22.9%), smaller for FV (17.1%) and smallest for SR (5.7%). The median concentrations of the faecal coliform bacteria (FC), determined by culture at 44oC, were low, amounting to 1.000 log10 CFU g-1 for FV and SR and 0.00 for VR. All examined vegetable and soil samples tested negative for the presence of Salmonella. The median concentrations of Clostridium perfringens were low, amounting to 0.00 log10 CFU g-1 for all categories of samples. This bacterium was relatively common in soil samples with the prevalence of 40.0%, but very rare in vegetable samples (occurring in 5.7% of FV and in none of VR samples). In conclusion, the results of the present study generally indicate that the microbiological quality of Polish vegetables grown on conventional farms is satisfactory and safe for consumers.

Our previous studies showed that glycerol fermentation by Hafnia alvei AD27 strain was accompanied by formation of high quantities of lactate. The ultimate aim of this work was the elimination of excessive lactate production in the 1,3-propanediol producer cultures. Group II intron-mediated deletion of ldh (lactate dehydrogenase) gene in an environmental isolate of H. alvei AD27 strain was conducted. The effect of the Δldh genotype in H. alvei AD27 strain varied depending on the culture medium applied. Under lower initial glycerol concentration (20 gL-1), lactate and 1,3-propanediol production was fully abolished, and the main carbon flux was directed to ethanol synthesis. On the other hand, at higher initial glycerol concentrations (40 gL-1), 1,3-propanediol and lactate production was recovered in the recombinant strain. The final titers of 1,3-propanediol and ethanol were similar for the recombinant and the WT strains, while the Δldh genotype displayed significantly decreased lactate titer. The by-products profile was altered upon ldh gene deletion, while glycerol utilization and biomass accumulation remained unaltered. As indicated by flow-cytometry analyses, the internal pH was not different for the WT and the recombinant Δldh strains over the culture duration, however, the WT strain was characterized by higher redox potential.

Opisano nowe zagrożenia mikrobiologiczne w żywności. W 2011 roku w Niemczech wyizolowano werotoksyczny szczep Escherichia coli O104:H4 pochodzący z partii kiełków kozieradki, sprowadzonej z Egiptu. Szczep ten charakteryzował się unikalnymi cechami: obecnością tylko werotoksyny vtx2 (wariant vtx2a), obecnością genów aatA, aggR, aap, aggA, aggC oraz opornością na większość antybiotyków. Innym, niespotykanym do niedawna w Polsce patogenem jest Yersinia enterocolitica o serotypie O8 biotyp 1B. Ten bio-serotyp należy do najbardziej chorobotwórczych dla ludzi. Najczęściej zatrucia pokarmowe związane z Yersinia związane są ze spożyciem wieprzowiny. Omówiono również problem występowania neurotoksyn botulinowych w żywności. Nowym zagrożeniem żywności mogą być bakterie wytwarzające beta-laktamazy o rozszerzonym spektrum substratowym (Extended-Spectrum Beta-Lactamases - ESBL). Zjawisko to odnosi się do wszystkich gatunków pałeczek z rodziny Enterobacteriaceae. Wśród izolowanych z żywności drobnoustrojów coraz większą rolę w zwiększaniu się oporności na antybiotyki przypisuje się metycylinoopornym szczepom Staphylococcus aureus (Methicillin-Resistant Staphylococcus aureus- MRSA). Występowanie zatruć pokarmowych u ludzi może być związane również z obecnością wirusów w żywności i wodzie. Nośnikiem wirusów mogą być urządzenia i linie technologiczne, opakowania, a także człowiek. Najwięcej zachorowań po spożyciu żywności wywołane było norowirusami, rotawirusami, wirusem hepatitis typu A i wirusem hepatitis typu E.
This paper describes the new microbiological hazards in food. For protecting human health, nowadays food safety authorities face with many challenges, that years ago were largely unheard. In 2011 verocytotoxigenic Escherichia coli O104:H4 has been isolated in Germany. Strain came from fenugreek sprouts originated from Egypt. It was characterized by unique features such as presence of enteroaggregative Escherichia coli genes (aatA, aggR, aap, aggA, aggC) and resistance to most antibiotics. In Poland only three cases of disease caused by O104:H4 strain have been reported. Another emergence pathogen in Poland is Yersinia enterocolitica O8, biotype 1B. It is the most pathogenic bioserotype recently isolated in the USA only. Food-borne is commonly associated with raw or undercooked pork. The source of Yersinia spp. may be also milk and water. The presence of botulinum neurotoxins in food is not new, but still an important issue because of their high toxicity to human. Botulinum neurotoxins are high-molecular thermolabile proteins produced by Clostridium botulinum and some strains of Clostridium butyricum and Clostridium baratii. Based on their antigenic properties, botulin neurotoxins are divided into seven types A-G, however only types A, B, E and F are toxic to humans and some animals. Increasing risk associated with food results from antimicrobial resistance eg. extended spectrum beta-lactamases (ESBLs) producing bacteria, particularly Enterobacteriaceae. Until recently strains ESBL+ were isolated in hospitals, however during last years they have been isolated from healthy humans, animals and food of animal origin. Increasingly common microbiological hazard in food is methicillin-resistant Staphylococcus aureus (MRSA). Although prevalence of this pathogen in food is not high, the thread comes from difficulties of treating of infections caused by MRSA. The occurrence of food-borne in humans may also be associated with presence of viruses in food and water. The carrier of viruses may be equipment in food plant, production line, packaging and man. Most food-borne are caused by noroviruses, rotaviruses, hepatitis A virus and hepatitis E virus. An increased number of food-borne viral outbreaks are recorded in several countries. Reasons for this include the improved diagnostic methods that have enhanced detection of some virus groups, and the increased marketing of fresh and frozen foods that has led to a worldwide availability of high risk food. Viruses may contaminate food either through contamination at source, principally through sewage pollution of the environment, or in association with food processing through inadequate hygiene practices of operatives or systems. Consequently, many different food products such as vegetables, shellfish and a great variety of ready-to-eat foods have been implicated in food-borne viral infections.

Opis:
Sialic acids are important constituents of animal tissue glycoconjugates and are also present in the antigens of some bacterial strains. Capsular polysaccharides with sialic acid (NeuAc) have been extensively studied with regard to sensitivity to the bactericidal action of serum, whereas little is known in this regard about lipopolysaccharides (LPS) which contain NeuAc. Strains of Salmonella 048, able to infect animals and containing the same structures of LPS with NeuAc, were examined for their susceptibility to the bactericidal action of normal bovine serum (NBS). The strains showed varied sensitivity to the bactericidal action of NBS, which indicates that the expression of LPS containing NeuAc residues is not critical for the strains' resistance to the serum's activity. In this study the mechanisms of complement activation responsible for killing serum-sensitive Salmonella 048 rods by NBS were also established. Three such mechanisms were distinguished: activation of the classi- cal/lectin pathways, important (decisive) in the bactericidal mechanism of complement activation, parallel activation of the classical/lectin and alternative pathways, and independent activation of the classical and lectin or the alternative pathway.

Celem badań przeprowadzonych w latach 2009–2011 była ocena wpływu nawożenia łąki trwałej nawozami naturalnymi (obornikiem i gnojówką) i nawozami mineralnymi (wprowadzonymi w trzech różnych dawkach) na liczebność wybranych mikroorganizmów epifitycznych bytujących na runi łąkowej. Próbki roślin do badań pobierano trzykrotnie w trakcie sezonu wegetacyjnego. W ramach badań eksperymentalnych oceniano: ogólną liczebność bakterii tlenowych, bakterii z rodzaju Enterobacteriaceae oraz drożdży i grzybów pleśniowych. W wyniku przeprowadzonych badań nie wykazano różnic w liczebności badanych grup drobnoustrojów bytujących na roślinności łąkowej nawożonej różnymi nawozami i w różnych dawkach. Stwierdzono jednak, że liczebność poszczególnych grup mikroorganizmów zależała przede wszystkim od terminu pobierania próbek. Największe liczebności wszystkich badanych mikroorganizmów stwierdzono w drugim roku badań (2010 r.). Liczba bakterii tlenowych rosła statystycznie istotnie wraz z okresem wzrostu roślin (kolejnym pokosem) i była największa w trzecim terminie zbioru. W próbkach świeżej masy roślin z trzeciego pokosu w drugim roku badań stwierdzano największą liczbę bakterii z rodzaju Enterobacteriaceae (6,61 jtk∙g–1) oraz bakterii tlenowych (8,65 jtk∙g–1). W próbkach z tego samego roku badań, ale z drugiego pokosu, odnotowano największą liczebność grzybów pleśniowych (5,88 jtk∙g–1) i drożdży (3,44 jtk∙g–1).
The study The aim of the research carried out in 2009–2011 was to assess the effect of fertilizing meadow with natural fertilizers (manure and liquid manure) and mineral fertilizers on the number of selected epiphytic microorganisms living on the meadow sward. Samples of plants for testing were collected three times during the growing season. The total number of aerobic bacteria, enterobacteria, yeast and mold fungi were assessed as a part of the study. As a result of the conducted experimental research, there were no differences in the number of examined groups of microorganisms living on the meadow plants fertilized with various fertilizers and doses. However, it was found that the number of individual groups of microorganisms depended mainly on the date of sampling. The largest number of all studied microorganisms occurred in the second year of the research (2010). In the samples of plants from the third cut in the second year of the study, the highest number of bacteria of the family Enterobacteriaceae (6.61 cfu∙g–1) and aerobic bacteria (8.65 cfu∙g–1) were found. In the samples from the same research year, but from the second cut, the highest numbers of mold fungi (5.88 cfu∙g–1) and yeast (3.44 cfu∙g–1) were recorded.
