Opis:
Somatic embryogenesis (SE) of conifers for clonal propagation has emerged from its earliest beginnings in 1980 to become an integral component of tree improvement strategies. With its capacity for long-term germplasm preservation and scale-up technologies,it is seen to be the preferred avenue to accelerate selection and operational deployment of value-added genotypes. At present,programs for numerous species from the Pinaceae family are underway worldwide,with activities ranging from selection trials to pilot production for plantation forestry. This paper will provide a review of current efforts in conifer SE including cryopreservation,commercialization and deployment strategies,and transgenics. A discussion of challenges and issues is directed at genetic fidelity, intellectual property and future needs.

Opis:
Somatic embryogenesis is the latest, and potentially the most efficient, method for the vegetative micropropagation of plants. Over the past three decades, numerous laboratory studies have investigated somatic embryogenesis of forest trees, yielding positive results for a number of economically important tree species. The first test trials were run and plantations were planted with interior spruce in the 90s by CellFor Inc. (Canada). However, at the beginning of the XXI century, the program to produce spruce and Douglas fir somatic seedlings was stopped for economic reasons. Thus, currently no operational program is ongoing except on a small scale in New Brunswick. In order to integrate somatic embryogenesis technology into operational reforestation programs, the production costs of forest tree somatic seedlings needs to be reduced, and the awareness of foresters and forest landowners that the material obtained through somatic embryogenesis is valuable needs to be increased. This awareness would enable implementation of this technology on a large scale for production and forest management throughout Europe including Poland. In this review, the importance of somatic embryogenesis in scientific research and in global and European forestry is presented. Our main aims are to provide basic information on the challenges in researching somatic embryogenesis of forest trees and to raise interest in this tree propagation technique in both scientists and foresters.

Opis:
The discovery of conifer somatic embryogenesis (SE) and the subsequent development of SE protocols for a range of genera and species have opened new research opportunities to forest biotechnologists and a means towards mass clonal propagation for the forest industry. This paper provides a general description of the conifer SE process, followed by a review of protocols developed specifically for several conifer species in the Pinaceae family for which production of somatic trees has been demonstrated. Additionally, future research needs, including approaches for developing markers to optimize SE process and the production of high quality embryos, are discussed.

Morphogenetic theories investigate the mechanisms of creation and regulation of definite biological forms in living organisms. The incredible diversity of shapes and sizes is generated through a barely unknown coordination of biochemical processes occurring at molecular levels. Such a crosstalk not only defines the rules of a robust scheme of matter differentiation, but it also has the capacity to adapt with respect to some variations of the environmental conditions. In this work, we propose a continuum model of growth and mass transport for biological materials during morphogenetic processes. Using the theory of configurational forces, we define the thermomechanical bases for understanding how both the mechanical and the biochemical states can orchestrate growth. The model is successfully applied to describe the morphogen-driven growth control in the imaginal wing disc of Drosophila melanogaster.

Embryogenic cultures of plants are exposed to various stress factors both in vitro and during cryostorage. In order to safely include the plant material obtained by somatic embryogenesis in combination with cryopreservation for breeding programs, it is necessary to monitor its genetic stability. The aim of the present study was the assessment of somaclonal variation in plant material obtained from embryogenic cultures of Picea abies (L.) Karst. and P. omorika (Pančić) Purk. maintained in vitro or stored in liquid nitrogen by the pregrowth-dehydration method. The analysis of genetic conformity with using microsatellite markers was performed on cotyledonary somatic embryos (CSE), germinating somatic embryos (GSE) and somatic seedlings (SS), obtained from tissues maintained in vitro or from recovered embryogenic tissues (ETc) and CSE obtained after cryopreservation. The analysis revealed changes in the DNA of somatic embryogenesis-derived plant material of both Picea spp. They were found in plant material from 8 out of 10 tested embryogenic lines of P. abies and in 10 out of 19 embryogenic lines of P. omorika after in vitro culture. Changes were also detected in plant material obtained after cryopreservation. Somaclonal variation was observed in ETc and CSE of P. omorika and at ETv stage of P. abies. However, most of the changes were induced at the stage of somatic embryogenesis initiation. These results confirm the need for monitoring the genetic stability of plants obtained by somatic embryogenesis and after cryopreservation for both spruce species.

We aimed to produce tissue cultures and plant regeneration from endangered Crocus species: C. scepusiensis, C. tommasinianus, C. vittatus (“Verni” series of the genus) and C. banaticus. For initiation of cultures we used a plant growth regulator (PGR) combination used for in vitro culture of saffron and its relatives: 10 mg L-1 α-naphthaleneacetic acid (NAA) and 1 mg L-1 6-benzyladenine (BA). Shoot tips of young seedlings (C. scepusiensis) and corms (for the rest of species) were used as explants. C. scepusiensis explants developed into organogenic calli. On media with decreased NAA and with or without increased BA concentration, calli produced stigma-like structures and/or shoots and whole plants. In the other species, callus initiation medium induced callus formation with abundant somatic embryos. In C. tommasinianus, embryos developed shoots when auxin content of medium was decreased. In C. banaticus, a decrease of auxin with or without an increase in cytokinin content led to shoot or whole plant regeneration, as in C. scepusiensis. In the case of C. vittatus and C. banaticus, initiation and/or maintenance of cultures on indole-3-butyric acid (IBA) and increased sucrose concentration stimulated whole plant regeneration and in vitro cormlet development. C. scepusiensis and the rest of cultures (organogenic vs. embryogenic) differed at the biochemical level: C. scepusiensis cultures had higher (yet still low) enzymatic antioxidant (catalase, peroxidase) activities. With respect to catalase isoenzyme patterns, C. banaticus was different from the rest of cultures, demonstrating its distinct taxonomical position. Besides germplasm preservation use of the present cultures, they have a potential biotechnological value.

The studies were carried out on seed explants (mature zygotic embryos) of four spruce species: Picea omorika (Pancić) Purk., P. pungens ‘Glauca' Beisnn., P. breweriana S. Watson and P. abies (L.) Karst. The explants required darkness during the induction of embryogenic tissue. Temporary exposure of explants to light did not increase their embryogenic capacity. It was observed that the temperature of 25°C was the optimal for embryogenic tissue induction from mature zygotic embryos of Picea omorika. Cold treatment at 4°C during one week improved embryogenic tissue initiation in the explants of P. pungens ‘Glauca'. Seed explants of Picea omorika produced embryogenic tissue on medium lacking glutamine. Only explants of Picea abies and P. omorika showed embryogenic tissue formation. For effective proliferation of embryogenic tissue in Picea abies and P. omorika it was necessary to supply the medium with 2,4-D (4.5-9 μM) and BA (2.25 μM). Two of three analized lines showed that ABA promote somatic embryo maturation, at the ABA concentration 20 μM in the medium.

Celem pracy było określenie przy pomocy pomiarów termowizyjnych tempa oddawania ciepła przez jajo kurze z rozwijającym się zarodkiem. Stwierdzono, że temperatura powierzchni skorupy jaja (EST) wzrasta w kolejnych etapach embriogenezy i może przewyższać temperaturę powietrza w inkubatorze. Niewielki spadek EST w 19. dobie inkubacji jaj umieszczonych w temperaturze pokojowej przez okres 15 minut, wskazuje na wykształcenie przez zarodek na tym etapie rozwoju zdolności ograniczonej termoregulacji.
The aim of this study was to investigate the rate of heat transfer by a hen's egg with a developing embryo with use of infrared measurements. The study was conducted on 600 eggs for hatching chicken Ross 308 broiler line. The eggs were incubated in the setter S576 Petersime ® at the air temperature (T) equal to 37.8°C and relative humidity (RH) in the range 49-50%. Thermographic measurements of the eggshell surface temperature (EST) were taken with use of a thermal imaging camera ThermoVision A20 of a detector resolution 120x160 pixels. For statistical analysis of each egg there were used the EST highest values measured. Subsequent sessions were measured in the following days of incubation: E1, E3, E4, E7, E8, E9, E11, E12, E13, E16 and E19. Trays with eggs were removed from the incubator and placed at T = 22.4°C and RH = 75%, and recording of EST was performed: 1) immediately after removing the eggs from the incubator, 2) after 15 min, 3) after 30 min. It was found that the EST increased during the subsequent stages of embryogenesis. Before the start of the hatching process the temperature value may exceed the air temperature in the incubator by about 2 °C. The increase in the surface temperature of the egg shell is particularly intense from the 10th day of embryogenesis, reflecting the intensification of metabolism of the embryo. The slight decrease in the crust surface temperature of eggs placed at the room temperature for 15 minutes in the 19th day of incubation, compared to earlier stages of embryogenesis, shows that the embryo at this stage of development has the limited ability of thermoregulation.

The objective of the presented study was to compare the effect of osmotic stress, induced by various concentrations of polyethylene glycol (PEG) MW 4000, on the development, maturation, and conversion of somatic embryos of Picea abies (model species) and P. omorika (endemic species) into somatic seedlings. Embryogenic tissue (ET) of P. abies (line A and B) and P. omorika were incubated on a maturation medium with 20 μM abscisic acid (ABA), 1 μM indole-3-butyric acid (IBA) and PEG 4000 at 0, 5, and 7.5%. After 5 weeks, the number of somatic embryos produced, and the number of cotyledonary somatic embryos per 1 gram of ET, was assessed. Cotyledonary somatic embryos were selected and transferred to a Margara germination medium for two months. Their growth, as measured by hypocotyl and radicle length, was evaluated after one month, and the somatic embryos were transferred to fresh medium. After the second month of incubation, the conversion frequency of germinated embryos into the seedlings was assessed. In general, ETs of both of the tested spruce species cultured on a maturation medium amended with 5% PEG 4000 exhibited a tendency to produce a higher total number of embryos, as well as a higher number of cotyledonary embryos. The B line of P. abies produced a significantly greater number of cotyledonary embryos, relative to the control (no PEG). PEG only stimulated hypocotyl growth in embryos of both lines (A and B) of P. abies during the germination stage. Somatic embryos of Picea omorika were characterized by poor growth of both hypocotyls and radicles, irrespective of PEG treatment. The addition of PEG 4000 to the maturation medium did not improve the conversion frequency of germinated somatic embryos into somatic seedlings in either spruce species after a month of incubation on Margara medium. The effect of PEG 4000 on somatic embryogenesis efficiency was more evident in the ET of the two P. abies lines, relative to the P. omorika ET line, during both the maturation and the germination stage. Results indicated that the P. omorika ET line was almost completely insensitive to all of the PEG concentrations tested.

Opis:
In vitro tissue culture technique, especially micropropagation, is attracting increasing attention in the production of ornamental plants. This technique will probably dominate the horticultural market in the near future. Light is one of the several factors affecting the success of in vitro plant tissue cultures. It directly affects the widely understood morphogenetic response of the explant, i.e., the ability of the explant to grow or regenerate, produce roots, etc. Lighting conditions provided during the in vitro stage may also greatly affect the plant vigor after transferring to nonsterile conditions. Moreover, the necessity of providing artificial light significantly contributes to the total cost of maintaining tissue cultures (related to energy consumption and the need to cool down the heat generated by lamps). Light quantity (intensity) and quality (spectral composition) are the two main parameters that determine its influence on in vitro cultures. This impact depends on the species and other accompanying factors. The aim of this mini-review is to summarize information on the influence of light on the morphogenetic and biochemical response of explants of some selected ornamental plant species grown under in vitro conditions.

Using doubled haploid technologies inbreeding can significantly reduce the time to obtain homozygous parental lines required for the production of F1-hybrid of vegetable crops. This study aims to investigate the influence of factors on the efficiency of carrot embryogenesis in isolated microspore culture to optimise the elements of protocol for producing doubled haploids. Microspores were isolated from inflorescences of 21 genotypes and incubated in NLN13 medium supplemented with 0.1 mg·dm-3 2,4-dichlorophenoxyacetic acids, 0.1 mg·dm-3 1-naphthyl acetic acids, 130 g·dm-3 sucrose, and 400 mg·dm-3 casein hydrolysate and its modifications. Embryoids and their groups were formed after 2–6 months, in some cases after 12 months of cultivation. Depending on the variant, the embryogenesis efficiency averaged from 0 to 4.9 embryoids or groups of embryoids per Petri dish (10 cm3). Embryoids within the group were formed from different microspores. No significant effects of inflorescence position on the plant (branching order), sucrose, and casein hydrolysate concentration in the medium were observed. Significant advantages (p ≥ 0.05) for some genotypes were shown: 1) microspore suspension density 4·104 cells·cm-3 (5.0 embryoids per Petri dish were formed at a microspore suspension density of 4·104 cells·cm-3, 0.0 embryoids per Petri dish at a density of 8·104 cells·cm-3); 2) cultivating microspores of tetrad and early mononuclear stage (4.9 ±3.1 embryoids per Petri dish were obtained by culturing tetrads and early mononuclear microspores, while 0.6 ±0.7 embryoids per Petri dish were obtained by culturing of later developmental stages); 3) high-temperature treatment duration of five days (4.9 ±2.1 embryoids per Petri dish were obtained after five days of high-temperature treatment, 2.7 ±2.6 embryoids per Petri dish formed after two days of high-temperature treatment; 9.8 ±4.7, 10.1 ±6.1, 0.0 ±0.0 embryoids per Petri dish formed after two, five and eight days of high-temperature treatment respectively); 4) adding colchicine 0.5 mg·dm-3 to the nutrient medium for two days of high-temperature treatment, followed by medium replacement (3.3 ±2.6 embryoids per Petri dish were obtained by using a nutrient medium with colchicine, while 1.7 ±1.5 embryoids per Petri dish were obtained by culturing in the reference variant).

This paper provides results of somatic embryogenesis study in our laboratory. General description of somatic embryogenesis (SE) induction, maturation of somatic embryos and plantlets regeneration of the Abies species, followed by a comparisons of some characteristics of zygotic and somatic embryos, seedlings and emblings (somatic seedlings). Own results are supplemented with some literature data. Also aplication of SE for improving of plantlet regeneration of elite fir trees from Dobroč primeval is described as well as initiation of the SE from seeds of incompatible crossings of firs where zygotic embryos abort usually several weeks after pollination

Opis:
Stale wzrastające zanieczyszczanie środowiska oraz presja obcych gatunków raków, a przede wszystkim wyniszczenie przez dżumę raczą, spowodowało znaczne ograniczenie występowania stanowisk raka szlachetnego (Astacus astacus L.) w Polsce. Lepsze poznanie biologii rozrodu i embriogenezy tego gatunku raka jest konieczne nie tylko do jego ochrony przed wyginięciem, ale również poprawy warunków chowu i hodowli. Rozwój zarodkowy raka szlachetnego w warunkach średniej temperatury 8 ± 2°C trwał 2100 D°. Podział żółtka przebiegał w bardzo wolnym tempie i kończył się dopiero po upływie 352 D°. Pierwsze półkoliste zawiązki zarodka zaobserwowano w 592 D°. Zamykanie się blastoporu nastąpiło w 928 D°. W 1200 D° zarodek przybrał postać naupliusa. Na tym etapie widoczne były przydatki: anteny I i II pary oraz żuwaczki. Około 1500 D° serce podjęło pracę. Na krótko przed wylęgiem (1800 D°) pojawił się w oczach pigment, zaś na powierzchni zarodka zaobserwowano równomierne rozmieszczenie komórek barwnikowych. Po przeanalizowaniu zmian morfometrycznych jaj raka szlachetnego stwierdzono, że objętość w trakcie embriogenezy zwiększała się z 9,26 ± 0,44 mm³ do 13,5 ± 0,44 mm³ na krótko przed wylęgiem. Pomiary wytrzymałości mechanicznej osłonek jajowych ujawniły, że była ona dość znaczna na początku embriogenezy (ponad 150 ± 20,2 g) i zmniejszała się w trakcie rozwoju embrionalnego do wartości 83,5 ± 7,1 g w chwili wylęgu. Współczynnik S/V na początku rozwoju zarodkowego wynosił 2,291 i w miarę trwania embriogenezy zmniejszał się do 2,017.
Constantly increasing pollution of the environment, invasive species, and a great harm caused by the plague (Aphanomyces astaci) contributed to a significant decrease of the noble crayfish (Astacus astacus) in Poland. Expanding the knowledge of the biology, reproduction and embryogenesis of the noble crayfish is necessary not only for its conservation, but also to improve its farming. Our study shows that embryogenesis of the noble crayfish at an average temperature of 8 ± 2°C lasted 2100 D°. The partition of the vitellus showed a rather slow rate and ended after 352 D°. First semicircular germs of the embryo were observed at 592 D°. Blastopore closed off at 928 D°. At 1200 D° embryo assumed the shape of nauplius, and at this stage the antennae of I, II pair and mandibles were visible. At about 1500 D° the heart became active. Shortly prior to hatching (1800 D°) eyes were pigmented, while steady distribution of pigment cell was observed on the surface of the embryo. After the analysis of morpho-mechanical changes in eggs of the noble crayfish, we found that the volume of eggs in the course of embryogenesis increased from 9.26 ± 0.44 mm³ to 13.5 ± 0.44 mm³ just shortly prior to hatching. Mechanical resistance of egg membrane to applied pressure was considerable - 150.0 ± 20.2 g at the beginning of embryogenesis, and dropped at hatching to 83.5 ± 7.1 g. The S/V coefficient at the beginning of embryonic development was 2.291 and decreased during embryogenesis to 2.017.

Opis:
Embryogenesis, microsporogenesis and pollen development in eastern galega (Galega orientalis Lam.) have been analysed. Generally, the embryogenesis in eastern galega is similar to that of goat’s rue (Galega officinalis L.). It is typical for eastern galega to form linear proembryos to the phase of 8 or 9 cells. The proper embryo develops from apical cells of the proembryo, while the remaining cells form the suspensor. The embryo of eastern galega typically forms a long, massive suspensor consisting of several irregular rows of cells on the side of the proper embryo and one large cell on the side of the micropyle. Suspensors were preserved in developing seeds possessing embryos with elongated cotyledons and a massive hypocotyl. In the initial phase of embryogenesis the endosperm is of a nuclear type. The transformation of the nuclear into cellular type of endosperm begins at the heartlike embryo phase and proceeds from the micropylar to chalazal end. Observations on the embryogenesis in eastern galega revealed the presence of degenerating ovules. Causes of degeneration were not analysed.Meiotic division in PMCs leads to the formation of microspore tetrads via simultaneous cytokinesis. Mature pollen grains are two-celled with well-formed sporoderm and three germ pores through which pollen tube can emerge.

The research was conducted on explants of silver fir (Abies alba Mill.) deriving from several forest districts in southern Poland. The study encompassed the influence of the origin of plant material, type of explants, kind of substances used for explants sterilization, PPM and the type of medium on the ability to form embryogenic callus and to develop somatic embryos in silver fir explants. From the plant material collected in three sites, 57 clones were obtained from mature zygotic embryos; this produced an embryogenesis frequency of 6%. Embryogenic callus was obtained with a diameter of 65–70 mm depending on the material origin. The best medium for development of callus inducted on embryos isolated from mature silver fir seeds was the SH medium. Somatic embryos were formed in a globular stadium (24 pieces) on this medium. The 10% solution of NaOCl (used for 15 minutes) turned out to be the most effective substance for seed sterilization.

Opis:
The article presents methodological research and theories of Hans Spemann which allowed to coin the concept of ‘organizer' of developmental processes. The concept of ‘organizer' acquires a fundamental cognitive sense not only in relation to the processes of embryogenesis but also relates to the research conducted in the fields of organic regeneration, and tissue or organ transplants in living organisms. The presentation of Spemann's experimental research and his philosophical generalizations in relation to the aforementioned issue focuses not only on the descriptive aspects of historical heuristic but also shows how complex is the problem which has not yet been resolved.
Artykuł ukazuje metodologiczną ścieżkę badań i rozważań Hansa Spemanna, prowadzącą do ukucia tzw. koncepcji „organizatora” procesów rozwojowych. W świetle opisanych zjawisk biologicznych koncepcja „organizatora” nabiera fundamentalnego wymiaru poznawczego w odniesieniu do procesów embriogenezy, organogene- zy oraz regeneracji organicznej. Ukazanie badań eksperymentalnych Spemanna oraz jego uogólnień filozoficznych w tym zakresie, stanowią swoistą wartość opisową, odnoszącą się do uwypuklenia historycznej heurystyki badawczej. Takie podejście metodologiczne wskazuje także na złożoność problemu dotyczącego integracji procesów rozwojowych, który współcześnie nie jest ostatecznie rozwiązany.

Many chrysanthemum mutants are chimeras built from tissues of a varied genetic composition. Regeneration in vitro of somatic embryos from the whole mutated ligulate florets or only from their small mutated fragments can lead to the separation of chimera components and, as a result, producing a new original cultivar. There was determined the effect of growth regulators (2,4-D; KIN or BAP) and explant type (whole or transversely-cut-into-half ligulate florets) on the efficiency of somatic embryogenesis of Chrysanthemum × grandiflorum (Ramat.) Kitam. ‘Cool Time’. For the induction of the somatic embryogenesis ant the regeneration of somatic embryos the MS medium [Murashige and Skoog 1962] with 18.08 μM 2,4-D as well as with this auxin and 4.44; 8.88; 22.20 μM BAP or 4.65; 9.30; 23.25 μM KIN was used. The best results were obtained when transversely-cut-into-half ligulate florets were inoculated onto the medium with 4.65 μM KIN and 18.08 μM 2,4-D. This results may increase the probability of success in separation of chimera components in chrysanthemum breeding.
Wiele mutantów chryzantem jest chimerami zbudowanymi z tkanek o różnym składzie genetycznym. Regeneracja in vitro zarodków somatycznych z całych objętych mutacją kwiatów języczkowatych lub tylko z ich niewielkich zmutowanych fragmentów może doprowadzić do separacji komponentów składowych chimery i w rezultacie do uzyskania nowej, oryginalnej odmiany. W badaniach określono wpływ regulatorów wzrostu (2,4-D; KIN lub BAP) oraz rodzaju eksplantatu (cały lub przecięty poprzecznie na pół kwiat języczkowaty) na wydajność embriogenezy somatycznej u Chrysanthemum × grandiflorum (Ramat.) Kitam. ‘Cool Time’. Do indukcji embriogenezy somatycznej oraz do regeneracji zarodków somatycznych zastosowano pożywkę MS [Murashige and Skoog 1962] uzupełnioną jedynie 18,08 μM 2,4-D, a także tą auksyną oraz 4,44; 8,88; 22,20 μM BAP lub 4,65; 9,30; 23,25 μM KIN. Najlepsze rezultaty uzyskano, inokulując przecięte poprzecznie na pół kwiaty języczkowate na pożywkę z 4,65 μM KIN i 18,08 μM 2,4-D. Uzyskane wyniki mogą przyczynić się do zwiększenia prawdopodobieństwa separacji komponentów chimer w hodowli chryzantem.

Opis:
Modified versions of the Cry3A gene of Bacillus thuringiensis (Bt) were transferred into Norway spruce (Picea abies). Both the biolistic approach and Agrobacterium tumefaciens mediated procedure were employed for transformation of embryogenic tissue (ET) cultures. The latter method proved to be more efficient yielding 70 transgenic embryogenic tissue lines compared with 18 lines obtained by biolistics. The modified Cry3A genes were driven by a 35S promoter and the nptII screenable selection marker gene was used in all vectors. The transgenic ETs were molecularly characterized and converted into mature somatic embryos. Germinating embryos formed plantlets which were finally planted into perlite and their Cry3A gene transcription activities were demonstrated by RT-PCR.

Opis:
The effects of TDZ and paclobutrazol on the primary regeneration on tulip flower stalk explants of six cultivars and subsequent shoot multiplication were examined. Explants, flower stalk slices, were excised from cooled and subsequently forced bulbs. The explants were incubated for two months in darkness on medium containing NAA and cytokinins, 2iP and BAP, as control, or TDZ (0.5-4 mg l-1) and paclobutrazol (0.05-0.4 mg l-1). Then, the regenerating explants were subcultured on medium with TDZ and NAA applied at low concentrations. Different regeneration capabilities were found depending on cultivar and growth regulators. The percentage of explants forming leaf-like structures ranged, on the control medium, from 80% in 'Blue Parrot' and 'Prominence' to below 30% in 'Apeldoorn' and 'Mirjoran'. TDZ, applied at optimum for each cultivar concentration, greatly increased the regeneration potential up to 70-100%. Paclobutrazol, added to the TDZ-containing medium, significantly enhanced the response of explants, resulting in high numbers of leaf-like structures formed per explant (13.7-22.8). The structures developed gradually into characteristic forms: the growing up cotyledonary leaf, the probable root primordium formed at its base, the growing downwards stolon and the shoot meristem developed finely on its tip. It is suggested that such primary regeneration may have a nature of somatic embryogenesis. Then, the adventitious shoots developed and formed clusters, which were divided into 2-3 smaller ones every two months. The growth regulators, used at initial stage, markedly influenced subsequent shoot multiplication. Thus, the most intensive shoot formation was noted with TDZ at concentrations of 0.5-2 mg l-1 and paclobutrazol of 0.05-0.1 mg l-1.

The potential of immature zygotic embryos to produce embryogenic tissue was tested using culture media differed in inorganic salt composition as well as plant growth regulator content. Explants cultured on DCR as well as MLV media gave the highest initiation percentages with maximum values of 8.8% (MLV) to 10.4% (DCR). Plant growth regulators content (standard or reduced) had no profound effect on embryogenic tissue initiation. Somatic embryo maturation as well as germination was dependent on the cell line and salt composition of the medium. Plantlet regeneration occurred in three cell lines out of five tested. During the maturation process profound changes occurred in the internal organisation of somatic embryos such differentiation of root meristem in precotyledonary somatic embryos and formation of provascular strands in cotyledonary somatic embryos.

We investigated direct and indirect formation of somatic embryogenesis in Brassica oleracea var. botrytis (cauliflower), a very important vegetable crop worldwide. Direct somatic embryogenesis, which is rather rare, was achieved in culture of 2-week-old hypocotyl explants of Brassica oleracea var. botrytis on MS medium supplemented with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5; 1.0; and 1.5 mg/l kinetin. Initial induction of embryogenic callus was achieved on MS supplemented with very low concentrations of 2,4-D (0.05 mg/l and 0.1 mg/l). Indirect somatic embryogenesis from leaf sections was obtained on MS supplemented with 0.05 or 0.1 mg/l 2,4-D. We examined various stages of somatic embryos (globular, heart, torpedo, cotyledonary). More embryos per explant were produced through the indirect pathway (23–25) than through the direct pathway (14–19). The number of embryos produced was high. There is a potential for recurrent, repeated or secondary somatic embryogenesis, possibly an unlimited source for mass propagation and ideal for synthetic seed production in this species. Plant regeneration was achieved on half-strength MS medium without any hormones.

Experiments on somatic embryogenesis in selected spruce, fir and larch species were performed to determine if this method of micropropagation enables production of quality seedlings for forest nurseries. High frequencies of embryogenic callus in spruce (23-31%) and fir (29%) were achieved when mature zygotic embryos were used as explants, while in larches (36%) only when megagametophytes (endosperms with immature embryos) were used. The possibility of somatic embryogenesis initiation also from somatic embryos (resulting in secondary and third generation of embryogenic callus) indicate high efficiency of this method of micropropagation. The best results at all stages of somatic embryogenesis (good proliferation of embryogenic callus, high rate of embryo regeneration and their survival) were obtained for Norway spruce, European larch and some hybrid larches.

Somatic embryogenesis was studied in four spruce species (Picea abies, P. omorika, P. pungens 'Glauca' and P. brewenana) to determine if this method can be used for in vitro propagation of coniferous trees. The highest frequency of initiation of embryogenic tissue was obtained when mature zygotic embryos were used as explants. It ranged then from 10.8% (P. brewenana) to 23.75% (P. omorika and P. pungens 'Glauca'). The frequency of embryogenic tissue initiation was strongly affected by medium composition, i.e. addition of appropriate auxins (2,4-D, NAA, Picloram) and sucrose concentration (10-20 g-1"1). A lower frequency was obtained in Picea omorika (10%) when megagametophytes (endosperms with immature zygotic embryos) were used as explants. No emryogenic tissue was produced from hypocotyls, cotyledons and needles. A satisfactory frequency was achieved with the use of somatic embryos of Picea abies (30%). The proliferation of embryogenic cell lines of spruces was affected by medium type. The experiments resulted in production of somatic plantlets of P. abies and P. omorika. This enables the application of this method of spruce micropropagation for genetic and breeding research or for nursery production.

Opis:
The isolation of rye ß-Amy1 and ß-Amy2 gene promoters from nuclear DNA using the inverse polymerase chain reaction (IPCR) technique and characterization of their sequences are presented. The conservation of ß-amylasc coding sequences allowed for simultaneous IPCR amplification of two different promoters with primers designed on the basis of the single known cDNA sequence. Two ß-amylasc gene promoters display a low sequence similarity (47%). Beside consensus TATA and CCAAT boxes, other sequence motives common to both promoters were found. In addition, the homology of amino acid sequences of plant ß-amylases available in the database is discussed.

This text aims to show that the core of human divinity according to Aristotle is exercising the divine mind for its own sake. Being happy and thus divine is auto-teleological, and must not be reduced to any sort of instrumental value. This reading of Aristotle excludes the theist interpretations of Prime Mover as well as the attempts at identifying the human mind with God, mainly because both these (different) interpretations seem to make auto-teleological bios theoretikos impossible. The first do this by introducing the divine provision which makes people act for God’s sake and not for their own sake. The others reduce the special status of humans by taking away the divine part, in my opinion being the sine qua non condition of the concept of human divinity. All the interpretations of human divinity which I have presented above can be useful nowadays in the ethical, (bio)ethical, social or even political discourse. This shows that the history of philosophy is not only about the past, but also about the future.

Opis:
Fenomen poronienia klinicznego wzbudza coraz większe zainteresowanie badaczy z dyscyplin pozamedycznych. Choć nadal dostrzegalna jest znacząca dysproporcja pomiędzy obszerną literaturą traktującą o poronieniu spontanicznym z perspektywy medycznej w porównaniu do wzrastających liczebnie studiów prowadzonych w zakresie nauk społecznych i filozoficznych, to sukcesywnie postępuje proces zmiany orientacji badawczej wczesnych strat prokreacyjnych. Dominujące dotychczas jednostronne podejście biomedyczne ustępujące miejsca holistycznym ujęciom aksjomedycznym. Niniejsze studium wpisuje się w aksjomedyczny model poronienia klinicznego. Jego celem jest pogłębiona analiza czterech kluczowych kontekstów straty prokreacyjnej występującej na początkowych etapach rozwoju prenatalnego ‒ antropologicznego, relacyjnego (przywiązania rodzicielskiego), psychologicznego i pomocowego. W rezultacie prowadzonych badań dochodzimy do wniosku, iż główną dominantą charakterystyk poronienia klinicznego jako zdarzenia tanatycznego jest płaszczyzna rozstrzygnięć antropologicznych statusu płodu i jego implikacji etycznych.
The controversy over the animation (personalisation) of the human embryo is nowadays becoming a particularly pressing theological issue due to the widespread methods of in vitro fertilisation whose adequate ethical evaluation requires a clear definition of the ontological and anthropological status of the human being in the early stages of embryogenesis. The starting point for the ontological analyses is the empirical case of monozygotic twins. The paper provides a critical assessment of the concept of delayed animation developed in the neo-Thomistic tradition (T. Ślipko). The main models of arguments for simultaneous animation developed by leading Polish bioethics are presented. An own model of argumentation justifying animation at conception is also presented. This model ascertains twinness of the zygote from the conception and refers by analogy to the case of inseparable Siamese twins.
