Opis:
Somatic embryogenesis (SE) of conifers for clonal propagation has emerged from its earliest beginnings in 1980 to become an integral component of tree improvement strategies. With its capacity for long-term germplasm preservation and scale-up technologies,it is seen to be the preferred avenue to accelerate selection and operational deployment of value-added genotypes. At present,programs for numerous species from the Pinaceae family are underway worldwide,with activities ranging from selection trials to pilot production for plantation forestry. This paper will provide a review of current efforts in conifer SE including cryopreservation,commercialization and deployment strategies,and transgenics. A discussion of challenges and issues is directed at genetic fidelity, intellectual property and future needs.

Opis:
Somatic embryogenesis is the latest, and potentially the most efficient, method for the vegetative micropropagation of plants. Over the past three decades, numerous laboratory studies have investigated somatic embryogenesis of forest trees, yielding positive results for a number of economically important tree species. The first test trials were run and plantations were planted with interior spruce in the 90s by CellFor Inc. (Canada). However, at the beginning of the XXI century, the program to produce spruce and Douglas fir somatic seedlings was stopped for economic reasons. Thus, currently no operational program is ongoing except on a small scale in New Brunswick. In order to integrate somatic embryogenesis technology into operational reforestation programs, the production costs of forest tree somatic seedlings needs to be reduced, and the awareness of foresters and forest landowners that the material obtained through somatic embryogenesis is valuable needs to be increased. This awareness would enable implementation of this technology on a large scale for production and forest management throughout Europe including Poland. In this review, the importance of somatic embryogenesis in scientific research and in global and European forestry is presented. Our main aims are to provide basic information on the challenges in researching somatic embryogenesis of forest trees and to raise interest in this tree propagation technique in both scientists and foresters.

Opis:
The discovery of conifer somatic embryogenesis (SE) and the subsequent development of SE protocols for a range of genera and species have opened new research opportunities to forest biotechnologists and a means towards mass clonal propagation for the forest industry. This paper provides a general description of the conifer SE process, followed by a review of protocols developed specifically for several conifer species in the Pinaceae family for which production of somatic trees has been demonstrated. Additionally, future research needs, including approaches for developing markers to optimize SE process and the production of high quality embryos, are discussed.

Morphogenetic theories investigate the mechanisms of creation and regulation of definite biological forms in living organisms. The incredible diversity of shapes and sizes is generated through a barely unknown coordination of biochemical processes occurring at molecular levels. Such a crosstalk not only defines the rules of a robust scheme of matter differentiation, but it also has the capacity to adapt with respect to some variations of the environmental conditions. In this work, we propose a continuum model of growth and mass transport for biological materials during morphogenetic processes. Using the theory of configurational forces, we define the thermomechanical bases for understanding how both the mechanical and the biochemical states can orchestrate growth. The model is successfully applied to describe the morphogen-driven growth control in the imaginal wing disc of Drosophila melanogaster.

Embryogenic cultures of plants are exposed to various stress factors both in vitro and during cryostorage. In order to safely include the plant material obtained by somatic embryogenesis in combination with cryopreservation for breeding programs, it is necessary to monitor its genetic stability. The aim of the present study was the assessment of somaclonal variation in plant material obtained from embryogenic cultures of Picea abies (L.) Karst. and P. omorika (Pančić) Purk. maintained in vitro or stored in liquid nitrogen by the pregrowth-dehydration method. The analysis of genetic conformity with using microsatellite markers was performed on cotyledonary somatic embryos (CSE), germinating somatic embryos (GSE) and somatic seedlings (SS), obtained from tissues maintained in vitro or from recovered embryogenic tissues (ETc) and CSE obtained after cryopreservation. The analysis revealed changes in the DNA of somatic embryogenesis-derived plant material of both Picea spp. They were found in plant material from 8 out of 10 tested embryogenic lines of P. abies and in 10 out of 19 embryogenic lines of P. omorika after in vitro culture. Changes were also detected in plant material obtained after cryopreservation. Somaclonal variation was observed in ETc and CSE of P. omorika and at ETv stage of P. abies. However, most of the changes were induced at the stage of somatic embryogenesis initiation. These results confirm the need for monitoring the genetic stability of plants obtained by somatic embryogenesis and after cryopreservation for both spruce species.

We aimed to produce tissue cultures and plant regeneration from endangered Crocus species: C. scepusiensis, C. tommasinianus, C. vittatus (“Verni” series of the genus) and C. banaticus. For initiation of cultures we used a plant growth regulator (PGR) combination used for in vitro culture of saffron and its relatives: 10 mg L-1 α-naphthaleneacetic acid (NAA) and 1 mg L-1 6-benzyladenine (BA). Shoot tips of young seedlings (C. scepusiensis) and corms (for the rest of species) were used as explants. C. scepusiensis explants developed into organogenic calli. On media with decreased NAA and with or without increased BA concentration, calli produced stigma-like structures and/or shoots and whole plants. In the other species, callus initiation medium induced callus formation with abundant somatic embryos. In C. tommasinianus, embryos developed shoots when auxin content of medium was decreased. In C. banaticus, a decrease of auxin with or without an increase in cytokinin content led to shoot or whole plant regeneration, as in C. scepusiensis. In the case of C. vittatus and C. banaticus, initiation and/or maintenance of cultures on indole-3-butyric acid (IBA) and increased sucrose concentration stimulated whole plant regeneration and in vitro cormlet development. C. scepusiensis and the rest of cultures (organogenic vs. embryogenic) differed at the biochemical level: C. scepusiensis cultures had higher (yet still low) enzymatic antioxidant (catalase, peroxidase) activities. With respect to catalase isoenzyme patterns, C. banaticus was different from the rest of cultures, demonstrating its distinct taxonomical position. Besides germplasm preservation use of the present cultures, they have a potential biotechnological value.

The studies were carried out on seed explants (mature zygotic embryos) of four spruce species: Picea omorika (Pancić) Purk., P. pungens ‘Glauca' Beisnn., P. breweriana S. Watson and P. abies (L.) Karst. The explants required darkness during the induction of embryogenic tissue. Temporary exposure of explants to light did not increase their embryogenic capacity. It was observed that the temperature of 25°C was the optimal for embryogenic tissue induction from mature zygotic embryos of Picea omorika. Cold treatment at 4°C during one week improved embryogenic tissue initiation in the explants of P. pungens ‘Glauca'. Seed explants of Picea omorika produced embryogenic tissue on medium lacking glutamine. Only explants of Picea abies and P. omorika showed embryogenic tissue formation. For effective proliferation of embryogenic tissue in Picea abies and P. omorika it was necessary to supply the medium with 2,4-D (4.5-9 μM) and BA (2.25 μM). Two of three analized lines showed that ABA promote somatic embryo maturation, at the ABA concentration 20 μM in the medium.

Celem pracy było określenie przy pomocy pomiarów termowizyjnych tempa oddawania ciepła przez jajo kurze z rozwijającym się zarodkiem. Stwierdzono, że temperatura powierzchni skorupy jaja (EST) wzrasta w kolejnych etapach embriogenezy i może przewyższać temperaturę powietrza w inkubatorze. Niewielki spadek EST w 19. dobie inkubacji jaj umieszczonych w temperaturze pokojowej przez okres 15 minut, wskazuje na wykształcenie przez zarodek na tym etapie rozwoju zdolności ograniczonej termoregulacji.
The aim of this study was to investigate the rate of heat transfer by a hen's egg with a developing embryo with use of infrared measurements. The study was conducted on 600 eggs for hatching chicken Ross 308 broiler line. The eggs were incubated in the setter S576 Petersime ® at the air temperature (T) equal to 37.8°C and relative humidity (RH) in the range 49-50%. Thermographic measurements of the eggshell surface temperature (EST) were taken with use of a thermal imaging camera ThermoVision A20 of a detector resolution 120x160 pixels. For statistical analysis of each egg there were used the EST highest values measured. Subsequent sessions were measured in the following days of incubation: E1, E3, E4, E7, E8, E9, E11, E12, E13, E16 and E19. Trays with eggs were removed from the incubator and placed at T = 22.4°C and RH = 75%, and recording of EST was performed: 1) immediately after removing the eggs from the incubator, 2) after 15 min, 3) after 30 min. It was found that the EST increased during the subsequent stages of embryogenesis. Before the start of the hatching process the temperature value may exceed the air temperature in the incubator by about 2 °C. The increase in the surface temperature of the egg shell is particularly intense from the 10th day of embryogenesis, reflecting the intensification of metabolism of the embryo. The slight decrease in the crust surface temperature of eggs placed at the room temperature for 15 minutes in the 19th day of incubation, compared to earlier stages of embryogenesis, shows that the embryo at this stage of development has the limited ability of thermoregulation.
