Although vitamin D is included in the group of fat-soluble vitamins, it must be considered as a prohormone. Its active forms, including calcitriol, have pleiotropic effects and play an important role in the regulation of cell proliferation, differentiation and apoptosis, as well as in hormone secretion, and they demonstrate anti-cancer properties. Since calcitriol delivery can be beneficial for the organism, and Syrian golden hamsters represent a unique experimental model, we decided to investigate its toxicity in this species. In this study, we injected calcitriol intraperitoneally at doses 0 (control), 0.180±0.009 µg/kg and 0.717±0.032 µg/kg. Animal behavior was observed for 72 hrs after injection, and afterwards blood, liver and kidneys were collected for post-mortem examination, electron microscopy, and hematology analyses. The highest dose of calcitriol induced a change in animal behavior from calm to aggressive, and the liver surface showed morphological signs of damage. Following injection of calcitriol, ultrastructural changes were also observed in the liver and kidneys, e.g. vacuolization and increased number of mitochondria. There was also a trend for increased serum levels of aspartate aminotransferase (AST), but not of alanine aminotransferase (ALT) or GGTP (gamma-glutamyl transpeptidase). There was no change in Ca, Mg and P levels, as well as in blood morphology between experimental and control groups. These results indicate that calcitriol at 0.717, but not at 0.180 µg/kg, may induce acute damage to the liver and kidneys, without inducing calcemia. We propose that the hepatotoxic effect of calcitriol in hamster constitutes the primary cause of behavioral changes.

Objectives The objective of the study was to assess prenatal toxicity of N-methylaniline (NMA) administered by gavage to pregnant female rats. Material and Methods Pregnant female rats were administered N-methylaniline in corn oil by gavage at daily doses of 0.8 mg/kg of body weight (b.w.), 4 mg/kg b.w., 20 mg/kg b.w. and 100 mg/kg b.w. from implantation (the 5th day post mating) to the day prior to the scheduled caesarean section (the 20th day of pregnancy). General behavior, body weight, food and water consumption, hematological, biochemical analyses and pathomorphological changes of the dams were recorded. Results All the females survived until the end of the study. The test substance was toxic to pregnant females, even at the lowest of the used doses, i.e., 0.8 mg/kg b.w./day. Lower weight gain during pregnancy and significantly higher NMA-dose-dependent absolute weight of the organs were noted in the exposed females. The females from the groups exposed at doses of 20 mg/kg b.w./day and 100 mg/kg b.w./day developed anemia and showed higher concentrations of free thyroxine (FT3) and free triiodothyronine (FT4) thyroid hormones. Total protein concentration exhibited an increase in all the exposed groups of females. In the prenatal toxicity study, administration of N-methylaniline throughout the embryonic and fetal periods produced embryotoxic effects at doses ranging 4–100 mg/kg b.w./day. Conclusions Considering the data obtained in this study, it is reasonable to assume that N-methylaniline administered orally to pregnant rats is toxic for mothers even at a low dose of 0.8 mg/kg b.w./day. However, this dose was not associated with any significant effects to their offspring. This prenatal exposure level may be considered as no-observed-adverse-effect level (NOAEL) for the progeny and a dose of 4 mg/kg b.w./day as the lowest-observed-adverse-effect level (LOAEL) for the progeny.

One of the ways for toxic emissions reduction from internal combustion engines, beside external Exhaust purification systems and alternative fuels, is direct intervention into engine combustion process. The inner catalyst application, in form of ceramic active coating applied inside of combustion chamber, is one of the examples of that kind of solution. The application of active coating inside of a compression-ignition engine (diesel engine) may have an important impact on several stages of combustion process: fuel cracking, fore-flame phase, combustion phase and secondary combustion phase. Investigations of such construction are very rare in the literature. The paper presents the results of the researches which aim was analysis of inner catalyst application impact on toxic emissions. As a catalyst platinum and rhodium was used. As a catalyst support and local thermal barrier a zirconium ceramic coating was used. The catalyst was located on the gasoline engine valves surface. The research was done in Emission Research Laboratory of the Division of Motor Vehicles and Internal Combustion Engines. The analysis was done according to European standard (EN ISO 16017-1: 2006) on gas chromatograph (Yarian 450 GC) equipped in capillary column and flame-ionization detector (FID). The results of the researches proofed effectiveness of that solution on toxic emissions limitation.

Introduction and aim. Poorly soluble isoflavonoid genistein is known as an anti-nematode agent and also it decreases the risk of certain types of cancer. The biological activity of genistein is limited mostly by its low solubility. Therefore many attempts to increase genistein solubility in water were reported. We applied a polyamidoamine dendrimer, modified its surface by glycidylation, and used this macromolecule as a guest for genistein. Material and methods. Polyamidoamine dendrimer 3rd generation was substituted with 64 glycidol residues to obtain a macromolecule host for genistein. The stoichiometry of this host-guest complex was determined. The complex was tested for skin model permeability, toxicity on fibroblast (BJ) and keratinocyte (HaCaT) cell lines in vitro and anthelmintic activity on the Caenorhabditis elegans nematode. Results. The partition coefficient of genistein between octanol and water was determined (KO/W). The 1:1 host-guest complex was isolated and used as drug delivery system for genistein delivery. PAMAM G3 glycidyled dendrimer containing genistein indicated an anthelmintic activity at 50 μM concentration. Conclusion. The solubility of genistein in water increases 640 times in presence of an equimolar concentration of the dendrimer. One molecule of host dendrimer encapsulates 3 molecules of genistein. The encapsulate is an efficient anti-nematode formulation.

The aim of the research was to study the impact of "Oława" smelter on surroundings. For this purpose, water, soil and sediments were collected in the vicinity of smelter and subjected to selected ecotoxicological tests. The authors used Spirodela polyrhiza duckweed and Brachinious calyciflorus rotifer for the assessment of the water from Odra River, and Heterocypris inconqruens ostracod for the sediment studies. Triticum sativum and Avena sativa monocotyledons as well as Sinapis alba and Lepidium sativum dicotyledonous plants were used to perform the soil toxicity tests. The obtained results proved slight sediment toxicity, but no toxicity of water was observed. The inhibition of root growth of tested plants was observed suggesting the impact of toxicity on soil. The studies proved that only the area in the nearest vicinity of the "Oława" smelter is still contaminated with heavy metals and may pose a serious threat to the environment.

The aim of this study was to reveal the potentially genotoxic effect of zearalenone on bovine lymphocytes by comet assay in vitro. The bovine lymphocytes were exposed to various zearalenone concetrations (50; 10; 2; 0.4 and 0.08 ppm). The viability and DNA damage of lymphocytes was monitored after 2 h, 24 h, 48 h and 72 h. After 2 hours of zearalenone exposure, statistically significant DNA damage occurred at all tested concentrations of 0.08 ppm (12.2±1.25; p<0.05), 0.4 ppm (12.7±0.88; p<0.01), 2 ppm (12.0±0.51; p<0.01), 10 ppm (11.2±0.47; p<0.01) and at 50 ppm (14.2±0 61; p<0.001). Significantly greater DNA damage was also found after 24 h, 48 h and 72 h. The obtained results showed that zearalenone may induce DNA damage of the bovine lymphocytes.

Introduction. Over 300 species of parasites can possibly be passed on humans. Most of the parasitic infections are defined based on their pathogenicity; however, some positive effects of a parasite existence within the human body have recently been suggested. Beneficial outcomes of parasite infections might result from the production and release of metabolites, modification of host immune response or products uptake of the host. Objective. The aim of the study was a comprehensive analysis of a wide range of effects of parasites on the human body, including an overview of the toxic and positive effects. State of knowledge. In the light of the latest research presenting the unconventional use of parasites in medicine, the widely understood of their impact on the human body can also be considered in a positive context. Clinical cases from diseases caused by the toxic effects of parasites, as described in recent years, indicate that the problem of parasitic infections still persists. Despite a great deal of knowledge about the toxic effects of parasites on the human organism and, above all, despite the improvement in sanitary conditions, there is a resurgence of parasitic infections, as evidenced, e.g. by the examples presented in this review. Conclusions. The examples of positive effects of parasites presented so far give hope for the future in terms of fighting many diseases for which pharmacological treatment has not yet brought a positive effect. A better understanding of those processes might lead to the development of new methods of unconventional medical treatment.

Ionic liquids (ILs) have gained a significant attention as alternative solvents in many chemical and biological reactions. ILs have evolved as a new type of non-aqueous solvents for biocatalysis, because they can stabilize and activate enzymes. Ionic liquids to be considered as so called “green solvents” should possess only very low or no toxicity to living organism and the environment. Currently, very little is known about the biotoxicity of ionic liquids. Within this paper a relative toxicity of two ionic liquids 1,3 dimethylimidazolium methyl sulfate [MMIM][MeSO4] and 1-hexyl-3-methylimidazolium chloride [HMIM][Cl]) on the growth of Escherichia coli (E.coli) was evaluated. In this study we focused on ionic liquids, which potentially could increase activity of β-galactosidase produced by selected strain of E. coli. The inhibition of cell growth in the presence of various ionic liquids was determined using both, solid and liquid cultures. Liquid culture was incubated in culture medium with different concentrations of ionic liquids (0.1, 1, 5, 10% (v/v)). Determination of the minimal inhibitory concentration (MIC50) of ionic liquids was based on monitoring of E. coli growth by optical density measured at 600 nm using densitometer. In the second method the solid culture plate with addition of various amount of ILs has been used. The number of colonies formed on the solid culture plate was converted to colony forming units (CFUs). The obtained results have confirmed, that prescreening based on cytotoxicity tests is a key element in selection procedure of ILs for any biotechnological processes based on application of microorganisms.

The paper presents results of analysis of Polycyclic Aromatic Hydrocarbons (PAHs) concentration and toxicity emitted from 1,9 TDI self-ignition. Because of unstable parameters of self-ignition engine work (pressure and temperature jumps), PAHs were extracted from two phases: gas phase and solid phase (particle matter - PM). A chromatographic method (Capillary gas Chromatography) of polycyclic aromatic hydrocarbons identification and analysis, because of their low level of concentration in exhaust gases, needed to be supported by sample purification and enrichment stages. Calibration of the chromatograph was made by attested mixture of 16 model samples (according to EPA, USA). Two different methods for toxicity estimation was used in this researches. The authors used relative carcinogenic coefficients (RCC) which was determined by Nisbet and LaGoy for individual polycyclic aromatic hydrocarbons in relation to benzo(a)pirene. Samples consist PAHs was also tested for cytotoxicity in standardized cell-culture system (human cell line A549, mouse fibroblasts line cell L929). Cell growth, cell morphology and cell viability were used as parameters to determine the cytotoxicity of the materials. The measure the lethality effect on cells was determined spectrophotometrically with the use of a mitochondrial enzyme activity assay for mitochondrial succinate dehydrogenase activity by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT assay). Cells were exposed to the biomaterials for 24, 72 and 120 h. The results of in vitro tests are discussible. A lach of correlation between toxicity measurement methods which was used in these researches was observed.

This work is the third part of the articles published on the effects of the shape and position of the combustion chamber in piston compression diesel engines on the toxicity of exhaust gases. In two previous articles presented at the KONES 2015 conference, the impact of position of the combustion chamber in relation to position of the injector, and the influence of the shape of the chamber (diameter, depth, lip on the CO, HC and NOx emissions was analysed. In the current article, the shape of the “central body” in a combustion chamber is analysed. “Central body” is the protrusion located in the central part of a toroidal combustion chamber. Subsequent modifications to the basic combustion chamber consisted of reducing the size of this protrusion. The study involved four versions of combustion chambers. Modifications caused a slight decrease in the compression ratio, which could have an impact on the unambiguousness of the results, as the effect of changes in shape of the “central body”. However, to maintain a constant compression ratio would require a change in diameter or depth of the chamber cavity, which would obscure the obtained results with even greater impact. Emission tests in discussed study were performed according to ECE-R4. During the tests, the completion of the engine and the engine settings were not changed.

This study was carried out to monitor the presence of organochlorine in drainage water in Kafr-El-Sheikh Governorate, Egypt. Furthermore, to evaluate the efficiencies of different remediation techniques (advanced oxidation processes [AOPs] and bioremediation) for removing the most frequently detected compound (lindane) in drainage water. The results showed the presence of several organochlorine pesticides in all sampling sites. Lindane was detected with high frequency relative to other detected organochlorine in drainage water. Nano photo-Fenton like reagent was the most effective treatment for lindane removal in drainage water. Bioremediation of lindane by effective microorganisms (EMs) removed 100% of the lindane initial concentration. There is no remaining toxicity in lindane contaminated-water after remediation on treated rats relative to control with respect to histopathological changes in liver and kidney. Advanced oxidation processes especially with nanomaterials and bioremediation using effective microorganisms can be regarded as safe and effective remediation technologies of lindane in water.

Studies on the currently used organophosphorus insecticides with respect to their environmental levels and effective remediation technologies for their residues in water have been considered as a source of major concern. This study was carried out to monitor the presence of organophosphorus in drinking water plants (Kafr-El-Shiekh, Ebshan, Elhamoul, Mehalt Aboali, Fowa, Balteem and Metobess) in Kafr-El-Shiekh Governorate, Egypt. Furthermore, it was carried out to evaluate the efficiency of different remediation technologies (advanced oxidation processes and bioremediation) for removing chlorpyrifos in drinking water. The results showed the presence of several organophosphorus pesticides in water sampling sites. Chlorpyrifos was detected with high frequency relative to other compounds in drinking water. Nano photo-Fenton like reagent (Fe2O3(nano)/H2O2/UV) was the most effective treatment for chlorpyrifos removal in drinking water followed by ZnO(nano)/H2O2/UV, Fe3+/H2O2/UV and ZnO/H2O2/ UV, respectively. Bioremediation of chlorpyrifos by effective microorganisms (EMs) removed 100% of the chlorpyrifos initial concentration after 23 days of treatment. There is no remaining toxicity in chlorpyrifos contaminated-water after remediation on treated rats with respect to cholinesterase activity and histological changes in kidney and liver relative to control. Advanced oxidation processes especially with nanomaterials and bioremediation with effective microorganisms can be regarded as safe and effective remediation technologies for chlorpyrifos in drinking water.

Assessment of gentamicin effect on oxidoreductive balance and microstructure of trunk kidney in Prussian carp (Carassius gibelio). The aim of this study was to investigate the potential toxic effects of gentamicin on the Prussian carp (Carrasius gibelio) by determining its effect on oxidant-antioxidant balance and by histological image analysis of trunk kidney. The fi shes were injected with single standard therapeutic dose of gentamicin of 5 mg·kg–1. The samples of trunk kidney were collected in 3 days post antibiotic administration. Concentration of reduced glutathione (GSH) and activity of enzymes: superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT) were examined. The concentration of GSH and the activity of studied enzymes did not show statistically signifi cant differences between control and gentamicin-exposed group. The pathological changes were not observed in the trunk kidney structure. Renal tubules as well as renal corpuscles had correct structure. The obtained results indicate that a single injection of gentamycin at a dose of 5 mg·kg–1 does not lead to disturbance of oxidant-antioxidant balance or histopathological changes in the trunk kidney of Prussian carp. Gentamicin administration did not change oxidoreductive balance and trunk kidney microstructure in Prussian carp (Carassius gibelio).

The toxicity and bioaccumulation of cadmium in a marine benthic protist community were examined at different depths within the sediment. For this purpose, sediment-water microcosms with 1000 μgCd dm−3 of the pollutant were used in two assays. The addition of cadmium caused a significant reduction in protist density, number of species and biomass. There was also a decrease in these three parameters with depth. During the treatment the density of protist groups was strongly depth-dependent. The dominant groups of protists at the different depths during the assay were also considered. The most dominant protist group in terms of density were the heterotrophic flagellates, both in the control and in the treatment with cadmium. In the treatments with cadmium, these were followed by ciliates and by dinoflagellates in both assays. In the control, all protist groups were present during the assay, whereas in the treatments with cadmium, autotrophic flagellates, diatoms and sarcodines were found in reduced proportion or not at all. Cadmium bioaccumulation increased towards the end of the assay. At any time during the assay, the proportion of cadmium bioaccumulated was an increasing function of depth.

Many studies have reported on the toxicity and related oxidative stress of mercury. Antioxidants play an important role in counteracting metal-induced neurotoxicity under in vivo conditions. In this study, the effect of diallylsulphide (DAS) was evaluated on mercuric chloride induced activities of catalase, superoxide dismutase, glutathione peroxidase and glutathione content in brains of rats. Pretreatment of rats with DAS in the Hg-treated group also inhibited an increase in lipid peroxidation and elevated acetyl cholinesterase and glutathione content. Activities of antioxidant enzymes were also restored concomitantly when compared to the control rats after DAS administration. DAS also caused a decrease in tumor necrosis factor-α level which was higher in HgCl2-treated group. The results indicate that DAS augments antioxidant defense with anti-inflammatory response against HgCl2-induced neurotoxicity. The increased level of antioxidant enzymes enhances the antioxidant potential of the organ to reduce oxidative stress.

Chromium (Cr) is a micronutrient that occurs in the natural environment in different oxidation states. Natural compounds contain chromium in the +3 oxidation state, whereas chromic(VI) acid derivatives are industrial products with strong toxicity. Hexavalent chromium compounds can adversely affect the respiratory system and the gastrointestinal tract, cause skin changes, and exert carcinogenic, mutagenic, embryotoxic as well as teratogenic effects. Research indicates that both organic and inorganic trivalent chromium compounds improve carbohydrate, lipid, and protein metabolism. These compounds are found in many enzymes, and they influence antioxidant processes, regulation of immune responses, and the secretion of hormones and selected vitamins in the body. In animals, Cr has been found to increase fat-free body mass, improve reproductive parameters, enhance growth, increase feed conversion efficiency, boost immunity, and decrease mortality. It has been suggested that Cr supplements can intensify the effects of insulin, decrease the plasma levels of non-esterified fatty acids (NEFAs), decrease triglyceride concentrations in the liver, and improve glucose tolerance, thus improving the performance and productivity of cattle in the perinatal period. Symptoms of chromium deficiency, including decreased feed intake, lower weight gains, reproductive disorders, and higher lipid levels, are observed in cattle fed diets that are low in this nutrient, and in animals that are exposed to considerable and prolonged stress. In ruminants, chromium supplementation is recommended during thermal stress, in the perinatal period, and during infections.

Spent of water based mud (WBM) were tested in this study. Bioassays were done on spent WBM, brine after filtration and solid phase (SP) after filtration on the press. As test organisms, green algae Kirchneriella obesa and Botrydium granulatum were used, along with the cladoceran Simocephalus vetulus and the plant Lepidium sativum. Electrolytic conductivity was between 6.43 and 240 mS/cm. The liquid phase was toxic for K. obesa in the range of dilutions between 0.21 and 0.019, considered as LC50. The parameter LC50 in the category of dilutions was 0.125 to 0.0078 SPP for S. vetulus, 0.25 to 0.094 SPP for green alga Botrydium granulatum when diluted by brown soil and 0.56 to 0.039 SPP for Lepidium sativum. Maximum quantum efficiency (QY) of plants’ photosystem II (PS II), growing on mixtures of drilling fluids with soil, did not depend on the kind of drilling fluid, whereas minimal fluorescence (F0) did. Rinsing the salt out of drilling fluids in a 1:1 proportion was effective only for bentonite mud in the test with L. sativum.

Environmental pollution particularly heavy metal pollution into aquatic ecosystem has led to multiple damage in almost all life forms. Cadmium is one of the priority pollutant listed by United States Environmental Protection Agency or US EPA that widely known to have an adverse effect to organisms as well as human health. One type of organism that is susceptible to the effects of metal pollution is phytoplankton. Phytoplankton is a group of microalgae that are easy to find and are primary producers in aquatic environments. Phytoplankton plays an important role in aquatic ecosystems because they serve as primary producers. They are representing the water total primary productivity and reinforcing the aquatic life on the higher food chain. Thus, any threats that endangered the population of phytoplankton can lead to trophical cascade or even worse, biodiversity loss. The exposure of cadmium in high concentration to phytoplankton can lead to various impact including cell damage and disruption, biosynthesis inhibition of photosynthetic pigment (e.g. chlorophyll and carotenoid), chlorphyll degradation or known as chlorosis, thylakoid membrane degradation, inhibition of cellular metabolism and cell division. It is important to study the impact of cadmium to phytoplankton in a cellular level to better understand what mechanism lies and to what extent that the cadmium will be transferred to higher trophical organism via bioaccumulation or biomagnification.

An important issue in construction of combustion chamber in compression-ignition engines is proper selection of its shape and size. Both features are dependent on several factors such as fuel injector location and angle, spray atomizer selection (amount of holes, their diameter and angular position), location of inlet valve and air turbulence. By doing research on prototype engine T370, an analysis of influence of combustion chamber size and its modifications going towards lip construction on flue gas toxicity was done. The diameter and depth of combustion chamber was being changed while maintaining the same compression ratio. After that, a modification of tested combustion chambers was made by creating so called “lip”, which aim was to create a swirl of injected fuel in the vertical plane. To visualize the changes in swirl, a numerical analysis of fuel injection into combustion chamber was made. In discussed study, emission tests were performed according to ECE-R49. During the research six combustion chambers with diameters 60, 63 and 66 mm (and their modification) were investigated. Tests were performed for several types of injectors and different injection timing. However, for analysis of combustion chamber size the results are presented for the same injectors but with optimized injection timing.

Histamine was discovered in the year 1910 and subsequently histamine toxicity was reported from several marine foods especially from fish. Detailed research has proved that different species of fishes are known to accumulate large quantity of histamine and the histamine is heat stable. Reason for the histamine toxicity was due to the consumption of fishes with high level of histamine. Ancient systems of medicine such as Ayurveda and Siddha are 5000 years old. Ayurveda and siddha clearly dictates the avoidance of fish by patients who suffer from various diseases such as psoriasis, vitiligo, eczema, urticaria, anemia, duodenal ulcer, irritable bowel syndrome, rheumatoid arthritis etc. In all the above diseases histamine is either the trigger or the cause of the disease, therefore reducing histamine burden is also one of the treatment strategies. Histamine toxicity in fish and sea foods are great concern and the histamine is harmful to many diseases. Ancient siddhars have advised the patients to suffer from various diseases to avoid fish may be due to histamine toxicity. In the light of the present revelation about histamine toxicity in fish and the fish avoidance prescription of Ayurveda and Siddha clearly indicates that the Ayurveda and Siddha scholars were clearly aware of histamine toxicity 5000 years ago. Regimen-related mucosal toxicity is extremely common following cytotoxic chemotherapy and radiotherapy. Mucositis is as an important determinant of the inflammatory response and infectious complications in cancer treated patients. Most assessment scales for mucosal damage are focussed on oral mucositis, since it is easy to evaluate. Measuring gastrointestinal musocal damage objectively remains difficult because it cannot be seen directly or readily detected. One of potential non-invasive biomarkers of gastrointestinal mucosal damage is plasma citrulline level. Citrulline is an amino acid produced by small bowel enterocytes. Low concentration of free circulating citrulline signifies severe intestinal mucosal damage in humans with nonmalignant disorders, such as villous atrophy-associated diseases, short bowel syndrome, Crohn's disease, and is used in follow-up after small bowel transplantation. The plasma citrulline level is a reliable and objective biochemical marker of enterocyte mass and function in humans, and therefore can be used to monitor enterocyte toxicity resulting from chemotherapy and radiotherapy during anticancer therapy in patients with severely disturbed gut integrity.

Coins are foreign objects that are commonly ingested by children and pets due to their shiny appearance and bright colors. The current study investigated whether Turkish coins can lead to zinc (Zn) and copper (Cu) toxicity as a result of exposure to simulated gastric juice (i.e. hydrochloric acid solution simulating the gastric environment). Five groups of coins were exposed to simulated gastric juice (0.15 N, pH:1-2) for a period of 4 (Group 1), 12 (Group 2), 24 (Group 3), 48 (Group 4), 72 (Group 5) and 120 h (Group 5) at body temp. (37°C). Zinc and copper levels were determined in the gastric acid solution by using an inductively coupled plasma optical emission spectrophotometer (ICP-OES). The coins were also evaluated for corrosive damage and weight loss. Group 5 had statistically higher Cu and Zn levels versus the other groups. However, at body temp. (37°C), copper and zinc levels increased steadily in parallel to the duration for which the coin remained in the stomach after ingestion. After 120-hour exposure at 37°C, all coins had various types of damage compared to the baseline, such as color alteration, erosion, and visible surface cavities. The mechanisms relating to local and systemic copper and zinc toxicity caused by coin ingestion is yet to be clarified for both Turkish and international coins. Therefore, it can be foreseen that intervention is required in the first 24 h after ingesting 5 kuruş coins and, unless removed spontaneously in 48 h, such intervention is needed for the other coins.

Introduction and aim. COVID-19 is a viral infectious disease, which was first reported in patients with unusual pneumonia in December 2019. However, as the pandemic progressed, extrapulmonary manifestations including various neurologic complications have been started to be increasingly reported. In this retrospective study, we tried to search the neurological complications seen in our patients with positive rRT-PCR test for COVID-19 and examine the underlying associated risk factors. Material and methods. We have retrospectively analyzed the neuroimaging studies performed in our patients with positive rRT-PCR test for COVID-19 between April, 2020 and August, 2021. Both computed tomography (CT) scans and magnetic resonance imagings (MRI) of brain, head & neck region and the spine were retrospectively evaluated for the presence of any complications in patients with positive rRT-PCR test for COVID-19. Results. There were 147 patients having neuroradiological imaging studies performed for various neurological symptoms. Among these patients we detected 10 acute neurologicalcomplications.The most common was acute ischemic stroke in 5 patients and intracranial hemorrhage in 3 patients, two of which were intraventricular hemorrhage. The other complications included a preasumed cytotoxic lesion of corpus callosum in a 18 year old girl and lumbar spondylodiscitis complicated with psoas abcess in a 47 year-old man. Conclusion. In COVID-19 patients severe neurological complications can occur even as a presenting manifestation. Early cytotoxic endothelial injury can be the underlying cause in these patients and should be further studied in larger series in terms of what the susceptibility factors in these patients.

Introduction: Median lethal dose (MLD) has been a controversial subject among biologists and animal ethicists since its inception in 1927 by Trevan. Toxicologists use MLD (LD50) as the first step to assess the toxicity of a substance. Animal ethicists criticize LD50 tests because animals suffer pain, and LD50 is irreproducible. The disadvantage of classifying chemicals based on LD50, the importance of the ‘characteristics’ proposed by Trevan, and the ideal mortality range for determining the best estimate of LD50 are also discussed. Objective: The aim of this review was to understand Trevan’s concept of LD50 and the method of Litchfield and Wilcoxon (L and W), and Finney’s probit analysis for LD50 determination Materials and method: A literature survey was conducted using Google search and Pubmed. Simulated data set was used for identifying the ideal mortality range for calculating the ‘best estimate’ of LD50. Brief description of the state of knowledge: After Trevan, the extensively used classical methods for LD50 determination are Finney’s probit analysis and the L and W method. Animal ethicists questioned LD50, because of its irreproducibility. Presently used methods for LD50 tests do not provide information on the dose-response, hence assessing the complete spectrum of toxicity is not possible. However, LD50 is used to classify chemicals. Conclusions: 'The 'characteristic' is more specific than the slope or LD50 of a dose-response curve. LD50 does not manifest the exact nature of the toxicity of a substance; hence, classifying chemicals based on LD50s may have little relevance

Introduction: Different parts of Grewia mollis Juss. (Malvaceae) are commonly used in folk medicine to treat several ailments, including diarrhea, ulcers, rickets, cough and fever. Although several studies have proved its therapeutic effectiveness, there are very few toxicological studies on the plant. Objectives: This study was carried out to evaluate the acute and sub-chronic toxicity of the aqueous extract of G. mollis stem bark (GM) in animals. Methods: In the acute study, rats were orally administrated with GM at doses of 150, 300, 600, 1200, 2400, 4800 and 9600 mg/kg to determine the oral medial lethal dose (LD50). In the chronic study, rats received three doses of GM (150, 300 and 600 mg/kg) for 28 days. After the treatments, food intake, body weights, biochemical, hematological and histopathological parameters were analyzed. Results: The LD50 was estimated to be >9600 mg/kg. No significant alterations in the animal’s body weight gain, relative organs weight, serum biochemical analysis, hematological or histopathological analyses of liver, kidneys, lungs, heart and spleen were observed. Conclusions: The results of this study provided evidence that oral administration of GM at dose of 600 mg/kg is relatively safe in rats and may not exert severe toxic effects.
Wstęp: Poszczególne części Grewia mollis Juss. (Malvaceae) powszechnie są stosowane w medycynie ludowej w leczeniu wielu schorzeń, w tym biegunek, krzywicy, kaszlu, gorączki. Chociaż wiele badań udowodniło skuteczność terapeutyczną, niewiele jest badań toksykologicznych tej rośliny. Cele: Badanie to zostało przeprowadzone w celu oceny ostrej i subchronicznej toksyczności wodnego ekstraktu z kory G. mollis (GM) u zwierząt. Metody: W badaniu toksyczności ostrej szczurom podano dożołądkowo GM w dawkach 150, 300, 600, 1200, 2400, 4800 i 9600 mg/kg w celu wyznaczenia średniej dawki śmiertelnej (LD50). W badaniu nad oceną toksyczności subchronicznej szczury otrzymywały GM w trzech dawkach (150, 300 i 600 mg/kg) przez 28 dni. Zwierzęta analizowano oceniając je pod względem: spożycia pokarmu, zmian masy ciała, oznaczania parametrów biochemicznych, hematologicznych i histopatologicznych. Wyniki: Wartość LD50 oznaczono na poziomie >9600 mg/kg. Podawanie wyciągu nie wpłynęło istotnie na: zmiany przyrostu masy ciała zwierząt, średniej masy narządów, wartości parametrów biochemicznych surowicy, parametrów hematologicznych lub obrazu histopatologicznego wątroby, nerek, płuc, serca i śledziony. Wnioski: Wyniki tego badania dostarczyły dowodów, że subchroniczne podawanie dożołądkowe GM w dawce 600 mg/kg jest stosunkowo bezpieczne u szczurów i może nie wywierać poważnych objawów toksycznych.

W celu przeprowadzenia badań zebrano miejski pył drogowy w siedemnastu punktach badawczych w sezonie wiosennym na terenie Dolnego i Górnego Śląska w Polsce. Są to regiony znane z podwyższonego poziomu zanieczyszczeń. Celem badań było określenie stężeń metali ciężkich w ekstraktach wodnych w obu regionach, co ma bezpośrednie przełożenie na zawartość tych metali w wodach powierzchniowych zasilanych spływami pochodzącymi z deszczu, oraz ocena toksyczności wymywanego pyłu miejskiego w odniesieniu do bakterii A. fischeri, co pozwoliło na ocenę wpływu spływów z obszarów miejskich na organizmy żywe. W wyniku badań stwierdzono, że ekstrakty wodne badanych próbek miejskiego pyłu drogowego wykazywały zróżnicowane stężenia metali ciężkich na badanych stanowiskach w obu regionach – na Dolnym i Górnym Śląsku, wskazując na istniejące zanieczyszczenie obu badanych obszarów oraz na ryzyko zanieczyszczenia wód wskutek spływu wód opadowych. Powodem tego zanieczyszczenia jest wzmożony transport miejski, przemysł oraz obecne odpady poprzemysłowe, a także sposób ogrzewania domów (niska emisja pochodząca ze spalania odpadów oraz węgla i drewna). Przeprowadzone badania toksykologiczne w odniesieniu do bakterii A. fischeri wskazują jednoznacznie na toksyczność ekstraktu wodnego pyłu miejskiego pobranego ze stanowisk K1 (Bytom), K2, K3 (tereny poprzemysłowe, Piekary Śląskie) na Górnym Śląsku oraz W1 (Wrocław, ul. Obornicka), W5 (Bielany Wrocławskie) i W6 (Wrocław, Most Grunwaldzki) na Dolnym Śląsku. Punkty K1, K2, K3 odznaczały się także podwyższonymi stężeniami większości badanych metali (Mn, Ni, Zn, As). W przypadku Dolnego Śląska punkty W1, W5 i W6 stężenia te nie były najwyższe w regionie, co sugeruje, że na toksyczność tych próbek mogą wpływać także inne związki obecne w pyle (np. WWA, dioksyny), których nie identyfikowano w tych badaniach. Jednakże, bazując na badaniach toksyczności oraz oznaczonych stężeniach metali, można jednoznacznie stwierdzić, że oba regiony są zanieczyszczone i istnieje wysokie ryzyko wpływu tych zanieczyszczeń na biocenozę wodną, a pewnie też na cały ekosystem.
In the presented study, urban road dust was collected at seventeen research points in the spring season in Lower and Upper Silesia in Poland. These regions are known for their elevated levels of pollution. The aim of the study was to determine the concentration of heavy metals in water extracts in both regions, which has a direct impact on the content of these metals in surface waters derived from rainwater runoff, and to assess the toxicity of urban dust on the basis the toxicity test with bacteria A. fischeri to allow making an evaluation of the impact of runoff for living organisms. It was found that the water extracts of the tested samples of urban road dust had different concentrations of heavy metals at the studied sites in both regions, in Lower and Upper Silesia, indicating the existing pollution of both areas and the risk of water pollution due to rainwater runoff. The reason of this pollution is increased urban transport, industry and the presence of post-industrial waste and house heating systems (low emissions from burning waste, coal and wood). The toxicological studies on bacteria A. fischeri indicate the toxicity of the water extracts of urban dust samples collected from K1 (Bytom), K2, K3 (post-industrial areas, Piekary Śląskie) sites in Upper Silesia and W1 (Wrocław, ul. Obornicka), W5 (Bielany Wrocławskie) and W6 (Wrocław, Most Grunwaldzki) sites in Lower Silesia. Sites K1, K2, K3 were also characterized by increased concentrations of metals (Mn, Ni, Zn, As). In the case of Lower Silesia, sites W1, W5 and W6 were not characterised by the highest concentrations of metals in the region, which suggests that the toxicity of these samples may also be affected by other compounds present in the dust (e.g. PAHs, dioxins) that had not been identified in these studies. However, based on the toxicity studies and the concentrations of metals, both regions are polluted and therefore the high risk of the impact of these pollutants on the aquatic biocenosis and probably on the entire ecosystem does exist.

Etery alkilowe glikolu etylenowego (ethylene glycol alkyl ethers – EGAE) i propylenowego (propylene glycol alkyl ethers – PGAE) są szeroko stosowane w przemyśle i gospodarstwie domowym, głównie jako rozpuszczalniki. Niektóre EGAE wykazują działanie gonadotoksyczne, embriotoksyczne, fetotoksyczne i teratogenne zarówno u ludzi, jak i zwierząt doświadczalnych. Ze względu na szkodliwe działanie tych eterów na rozrodczość i rozwój organizmu EGAE są zastępowane znacznie mniej toksycznymi PGAE. W niniejszej pracy przedstawiono dane na temat mechanizmów toksycznego działania EGAE na komórki rozrodcze, zarodek i płód. Szczególną uwagę zwrócono na metabolizm niektórych EGAEs i ich toksyczność narządową, a także na apoptozę spermatocytów związaną ze zmianami ekspresji genów, które kodują czynniki stresu oksydacyjnego, kinazy białkowe i jądrowe receptory hormonów. Both ethylene and propylene glycol alkyl ethers (EGAEs and PGAEs, respectively) are widely used, mainly as solvents, in industrial and household products. Some EGAEs demonstrate gonadotoxic, embriotoxic, fetotoxic and teratogenic effects in both humans and experimental animals. Due to the noxious impact of these ethers on reproduction and development of organisms EGAEs are replaced for considerably less toxic PGAEs. The data on the mechanisms of testicular, embriotoxic, fetotoxic and teratogenic effects of EGAEs are presented in this paper. Our particular attention was focused on the metabolism of some EGAEs and their organ-specific toxicities, apoptosis of spermatocytes associated with changes in the expression of various genes that code for oxidative stress factors, protein kinases and nuclear hormone receptors. Cadmium and its derivatives are considered the most toxic compounds between heavy metals. Beside cadmium have very few independent minerals it is associated in many cases with zinc. Cadmium interacts with zinc in organisms and is a big dependence on the rate of these elements in various humans, animals and plants. It was demonstrated that cadmium compounds can influences to fall ill with cancer. A lot of scientists studied the damage of DNA due to exposure at cadmium compounds in various conditions. It is very interesting to mention the role of trace elements including cadmium in agrosystems. A real source of cadmium is the use of fertilizers. One important studied on cadmium toxicity is the induced oxidative stress. The paper presents some data obtained by the authors concerning the presence of cadmium in waters, foods as well as in various types of clinical and pharmaceutical compounds.
Kadm i jego związki są uważane za najbardziej toksyczne wśród metali ciężkich. Kadm występuje w bardzo niewielu minerałach, często wraz z cynkiem. Ze względu na interakcje pomiędzy kadmem a cynkiem ich zawartość w organizmach ludzkich, zwierzęcych i roślinnych jest zróżnicowana. Wykazano, że związki kadmu wpływają na zwiększenie zachorowań na raka. Badano wpływ związków kadmu na występowanie uszkodzeń w DNA w różnych warunkach. Interesująca jest rola mikropierwiastków, w tym kadmu, w agrosystemach. Dużym źródłem kadmu są stosowane nawozy. Jednym z ważnych kierunków badań jest wpływ toksyczności kadmu na wywołanie stresu oksydacyjnego. Przedstawiono dane uzyskane przez autorów dotyczące obecności kadmu w wodach, żywności, jak i w różnego rodzaju substancjach klinicznych oraz farmaceutycznych.

Oil based muds (OBM) are highly important in well drilling. Water based muds (WBM) and synthetic based muds are not providing sufficient qualities in fulfilling all of the functions of OBM. OBMs ensure more efficient drilling but also disadvantages such as higher initial cost, more stringent pollution controls and reduced effectiveness of some logging tools. Expenses for mud are reaching 10–15% of total well cost. However, high costs are still low compared to expenses for corrective measures in the case of using mud with poor properties, which could lead to drilling disruption as well as excessive time and cost. OBM and cuttings are saturated with toxic compounds and if discharging to sea poses ecological threats. Perhaps the most hazardous oil component for aquatic organisms are low-boiling aromatics, which consist of benzenes and naphthalenes both soluble in water. Additionally, the higher-boiling aromatic fractions are of high environmental interest due to their persistence in sediments, leading to enzyme induction, cellular dysfunctions, genetic alterations, and chronic effects on organisms. There is a difference in volume of generated waste between using water- and oil-based muds. WBM produces 7,000–13,000 bbl of waste per well. Depending on well depth and diameter 1,400–2,800 bbl are drill cuttings. OBM generates mush less waste as the mud is usually recycled and only drill cuttings with volumes of 2,000–8,000 bbl per well need treatment prior discharge. This paper discusses the hazardous effect of toxic compounds in OBM and evaluates the efficiency of different OBM treatment towards zero discharge.

Abstract: The main problem of modern cancer therapy is their resistance to the drugs used and the toxicity of drugs to healthy cells of the human body. Hope is natural substances that are protective of cells and often support the action of standard anticancer chemotherapy. Among them are selenium compounds. In the present study, we examined the cytotoxic and genotoxic potency of doxorubicin (DOX) metal complexes as new anticancer drug candidates. We also measured the effect of simultaneous administration of seleno –L-methionine as biologically active agent with DOX and its complexes with Mn, Mg, Fe, Co and Ni in prokaryotic model - Escherichia coli RFM443 with plasmid transcriptional fusion of recA promoter and luxCDABE as a reporter gene. The results obtained by us, indicate high biological activity of metal complexes with doxorubicin. The strongest modulatory effect was observed using the DOX complexes with Co and Ni compared to the DOX. The DOX complexes with Mg, Mn and Fe also showed relatively high biological activity. Simultaneous bacteria culture treatment with SeMet decreased the values of the DOX and its metal complexes cyto- and genotoxic parameters. These results encourage further research to assess the value of the doxorubicin metal complexes as new cytostatic drug candidates. While SeMet may be considered as therapeutic option in cancer treatment as modulation agent in anti-cancer therapy.

The paper presents the results of experiments on ecological toxicity assessment performed for 12 soil samples collected at 3 obsolete pesticide “tombs” in Poland, before their final disposal. Bioavailability of the main pollutants: p,p’-DDT, lindane and methoxychlor was assessed for selected samples by consecutive solid phase extraction using a Tenax TA sorbent. Several toxicity bioassays were also carried out, including: reducers (Microtox Soild Phase), producers (Phytotox) and consumers (tests of avoidance, acute toxicity and reproduction with use of the earthworm Eisenia foetida). Data from toxicity tests were discussed against the results of the chemical analysis of a wide range of pesticides determined by GC-ECD and GC-NPD. This part of the study enabled the evaluation of the applicability of the aforementioned bioassays in the assessment of pesticide soil pollution. Results of toxicity tests showed a slight to severe impairment of habitat function for all of the contaminated samples, which was only partially reflected by the analytical data. The most sensitive biotest was earthworm reproduction, followed by Phytotox and earthworm acute toxicity. Earthworm avoidance and Microtox tests were found to be of rather limited usability.

W artykule przedstawiono charakterystykę budowy i działania deoksyniwalenolu będącego wtórnym metabolitem grzybów strzępkowych z rodzaju Fusarium. Najwięcej mikotoksyny produkują komórki grzybów należące do gatunków Fusarium graminearum i Fusarium culmorum. Optymalne podłoże dla rozwoju grzybni z zarodników tych grzybów stanowią ziarna zbóż we wszystkich strefach geograficznych. Deoksyniwalenol (DON) może być produkowany w komórkach grzybni zarówno w okresie przedżniwnym, pożniwnym oraz w czasie przechowywania, transportu i przetwarzania ziarna zbóż. Według wielu autorów opracowań naukowych DON wykazuje silne działanie toksyczne u ludzi i zwierząt.
This paper presents the characteristics of the constitution and operation of deoxynivalenol which is a secondary metabolite of fungi of the genus Fusarium. Fungi of the genus Fusarium graminearum and Fusarium culmorum produce mycotoxins at most. The optimal substrate for mycelial growth of these fungi spores are the grains in all geographical areas. Deoxynivalenol (DON) can be produced in the cells of the mycelium on grains before harvest, after harvest, during storage, transport and processing. According to many authors of scientific papers, DON has a strong toxic effect in humans and animals.

Introduction: Everolimus is a selective mTOR inhibitor which received approval for treatment of advanced renal cell carcinoma (mRCC) after progression on or after treatment with VEGF-targeted therapy. The aim of this study was to evaluate the efficiency and toxicity profile of everolimus in second line therapy of mRCC. The authors also assessed the impact of clinicopathological factors on the effectiveness of everolimus. Methods: The retrospective analysis was conducted on the medical records of 33 mRCC patients who were treated with everolimus in second line therapy after progression on interferon or tyrosine kinase inhibitors (sunitinib or pazopanib) during the years 2010–2016. Results: Median time of treatment with everolimus was 4 months (range from 1 to 58 months). Median progression free survival was 4 months and overall survival (OS) was 11 months. The best response (PR + CR + SD) was reported in 57% of patients. Toxicity in grade 3–4 was reported in 9 (27%) of patients. Clinicopathological factors associated with progression during everolimus therapy were: smoking and alcohol abuse (p = 0.029), higher Furman grade (p = 0.166), tumor necrosis (p = 0.383), fat tissue infiltration (p = 0.040), lymph node (p = 0.193) and adrenal metastases (p = 0.067). Factors which increase the risk of everolimus toxicity were worse performance status (p = 0.333) and more advanced disease at the beginning (lymph nodes metastases, p = 0.05) and higher Furman grade (p = 0.04). Conclusions: Cigarettes use and/or alcohol abuse, adrenal metastases, fat tissue had significantly negative influence on survival. Grade 3–4 toxicity were reported more frequently in patients with worse performance status and more advanced disease at the time of diagnosis.

The paper presents results of comparative tests of exhaust gases toxicity of GTD-350 turboshaft engine powered by Liquefied Petroleum Gas (LPG) and conventional JET-A1 fuel. Structure of GTD-350 engine’s test bed was discussed. Because of explosion danger of LPG vapour the test stand was arranged in the open air. Paper comprises specification of LPG supply system, gas injector’s construction and visualization of LPG injection. The supply system was based on the newly constructed pressurized injectors. Required LPG operating pressure was obtained by pressurizing LPG tank using nitrogen and LPG-flow was controlled using needle valve. A series of photographs presents shape of fuel streams for new injector supplied by JET-A1 and LPG. Photos of flame torches for multi-hole and conical type injectors fuelled by LPG are inserted as well. A comparative study of carbon monoxide, nitrogen oxides, hydrocarbons and carbon dioxide as well as oxygen concentration for GTD-350 equipped with new injectors fuelled by LPG and standard supplied by Jet-A1 was carried out. The study comprised turbocharger speed range between 40 and 80% of NTS. The exhaust gas temperature comparison for above-mentioned configuration is presented as well. All data shown in presented figures are mean of 6 measurements. All completed tests were carried out for standard GTD-350 engine combustion chamber.

Opis:
Thallium is a toxic and quite common element in the environment, but it is poorly examined, despite the known toxic activity of this metal. The ionic radius of the element is similar to the ionic radius of potassium, and at the same valence (K+,Tl+) allows substitution of this element in the nervous system and muscle, which results in its toxic activity. Thallium interferes with many enzymes and also has a strong affinity to the group – SH. The most dangerous symptoms of poisoning come from central nervous system. This element is easily cumulated by many living organisms, which poses a serious threat. In the past, it was used in rodenticides, and despite the risks, in many countries is still used for this purpose due to its low production costs. Contamination of this element of industrial areas is associated with the steel industry and cement production, while residents of these areas are often not aware of the threat and grow in these area vegetables that can absorb thallium, and in particular here talking about the Brassicaceae family. This element in the environment should be constantly monitored. Its isotope such as 201Tl (Thallium-201) has some significance in medicine, as radiopharmaceutical.

Butyrylcholinesterase (BChE) is synthesized in the liver and found in high concentrations in blood plasma, liver, heart, pancreas, vascular endothelium, skin, brain white matter, smooth muscle cells and adipocytes. BChE is a non specific enzyme that hydrolyzes different choline esters (succinylcholine, mivacurium) and many other drugs such as aspirin, cocaine and procaine. The enzyme is also considered as a bioscavenger due to its ability to neutralize the toxic effects of organophosphorus compounds (nervous system fs agents) such as soman. BChE displays several polymorphisms that influence its serum activity; therefore they could determine the individual sensitivity to chemical nerve agents. In this study, we investigated the correlation between BChE variants and the degree of enzyme inhibition and reactivation after soman application on blood samples of 726 individuals. The blood samples of individuals expressing abnormal variants, were more sensitive to soman compared to variants of homozygotes and heterozygotes for U-allele. We found significant differences in the degree of enzyme reactivation between different variants (with and without U-presence).

Arsenic is a metalloid of great environmental concern because of its highly toxic nature and wide abundance. Arsenic contamination in groundwater has been reported in Bangladesh, India, China, Taiwan, Vietnam, USA, Argentina, Chile and Mexico. The clinical symptoms of arsenic toxicity in human body include skin itching to sun rays, burning and watering of the eyes, weight loss, loss of appetite, weakness, fatigue, limited physical activities and working capacities, chronic respiratory problems, moderate to severe anemia etc. The Bengal basin is regarded to be the most acutely arsenic infested geological province in the world. Heavy withdrawal of groundwater for fulfilling the needs of the increasing population in Bengal Basin resulted in increased arsenic level in the groundwater. Mobilization of arsenic in Bengal delta is further interfered by microbial activities and interactions. Different microbial strains have been isolated from Bengal Delta which can tolerate, transform and resist arsenic. The use of arsenic contaminated groundwater for irrigation purpose in crop fields elevates arsenic concentration in surface soil and in the plants grown in these areas. Several plant species have been studied for their ability to accumulate arsenic in the Bengal Delta. Rice is generally grown in submerged flooded condition, where arsenic bioavailability is high in soil. As arsenic species are very much toxic to plants and can execute oxidative stresses, they can also affect the overall production of rice and other vegetables, and can affect the agricultural and economic development of Bengal Basin. Cattle population also consume arsenic infested water in those areas and usually eat edible plants contaminated with arsenic, which, in turn, can further increase the toxicity level in their bodies and also can increase the arsenic bioaccumulation in meat and milk. In the rural areas of the Bengal Delta, arsenic contamination raised a number of social problems which are continuously weakening the structural integrity of rural society. Uses of phytoremediation and microbial remediation technologies, rainwater harvesting, use of arsenic resistant plant varieties for cultivation are some sustainable methods which can be applied for arsenic remediation. Besides, nano-agglomerates of mixed oxides have been synthesized and successfully employed for arsenic removal from aqueous solutions. Intensive investigation on a complete food chain is urgently needed in the arsenic contaminated zones, which should be our priority in future researches.

A higher sensitivity and the cytotoxic effects of garlic have been previously reported on culture cells. No studies, however, have investigated the developmental toxicity of fresh garlic in zebrafish model. In this study freshly made garlic juice, which chemically is a combination of sulphur containing compounds, was evaluated to investigate the developmental toxicity during a 5-day assay using zebrafish embryos, starting from 2 h post fertilization. The results showed that toxicity endpoints such as the hatching rate, survival rate, malformation rate and growth rate had a significant dose-response relationship with increasing concentrations of garlic juice. The average hatching rate and time to hatch of zebrafish exposed to lower concentrations (0.001% and less) were significantly increased around 48-60 h of treatment (P < 0.05), strongly emphasizing the acceleration of hatchability caused by garlic juice. Mortality was also increased with exposure to garlic, exhibiting the maximal toxicity after 12 h of exposure to the concentration of 0.08%. A significant reduction in the heart rate by 35.5% was recorded due to the higher concentrations of juice (above 0.01%) starting form 36-hour exposure (P < 0.05). Growth retardation, pericardial oedema with heart malformations, deformity of yolk, lack of pigmentation and body malformation have been noticed at the concentration of 0.03% starting from 48 h of exposure. No severe malformations in any of the experimental zebrafish were produced at concentrations less than 0.01% of garlic juice. The results indicate that developing zebrafish are sensitive to garlic juice and further studies are required to investigate the molecular basis of the observed effects that garlic constituents exert upon aquatic vertebrates.

Herbal medications are gaining popularity in many countries. Although most can be used without any problem, serious toxicities do occur. Adverse events can be anticipated when herbal medications are used at excessive dose, long-term, for non-approved indications or by patients who are using multiple medications. Adverse events should be anticipated when these herbal medications with identified pharmacological effects or toxic ingredients are used. Healthcare professionals need to discuss or advise patients regarding their use. Physician-obtained medication histories towards specific herbal product use could provide relevant pharmacologic information and uncover cases of adverse events or toxicity from the used herbal products.

The aim of this work was to evaluate the ecotoxicity of the waste water from chosen sewage treatment plants. Microtox M500 and Aliivibrio fischeri luminescent bacteria were used for the assessment of toxicity as well as acute tests using aquatic organisms: Daphnia magna crustacea and Chironomus tentans larvae. The performed ecotoxicological tests showed different ranges of sensitivity of living organisms when it comes to the individual types of industrial sewage. The Aliivibrio fischeri bacteria turned out to be the most sensitive bioindicator. In addition, the inclusion of toxicity tests in a wastewater management system in industrial plants will allow the elimination of toxic sewage disposal into the surface water and soil.

Nikotyna jest bezbarwną, bezwonną i oleistą cieczą otrzymywaną z liści tytoniu przez destylację z parą wodną w środowisku zasadowym i ekstrakcję eterem. Największe zużycie nikotyny jest związane z produkcją wyrobów tytoniowych, a także z produkcją środków, których zażywanie ma na celu odzwyczajenie się od palenia. Nikotyna jest składnikiem niektórych pestycydów. Narażenie zawodowe na nikotynę możliwe jest przy produkcji i suszeniu tytoniu. Zatrucia śmiertelne zdarzały się w latach 20. i 30. XX w. w trakcie opryskiwania roślin preparatami z nikotyną. Obecnie w Polsce tylko 8 osób było narażonych na nikotynę o stężeniu w powietrzu przekraczającym wartość NDS, tj. 0,5 mg/m3 (dane z 2002 r.). Do śmiertelnego zatrucia zawodowego nikotyną dochodzi bardzo rzadko. Objawami ostrego zatrucia małymi dawkami nikotyny są: pobudzenie oddechu, nudności, wymioty, bóle i zawroty głowy, biegunka, częstoskurcz, wzrost ciśnienia krwi oraz pocenie i ślinienie się. Po dużych dawkach nikotyny stwierdzono ponadto pieczenie w jamie ustnej, gardle i żołądku. Później następowało wyczerpanie, drgawki, osłabienie czynności oddechowej, zaburzenie rytmu serca oraz zaburzenia koordynacji ruchowej i śpiączka. Śmierć może wtedy nastąpić w czasie od 5 min do 4 h. Zatrucia przewlekłe nikotyną prowadzą do zaburzeń układu krążenia. Zmiany naczyniowe sprzyjają powstawaniu dusznicy bolesnej oraz zawałom serca, a także powodują: osłabienie pamięci, zwolnienie procesów psychicznych i koordynacji myśli, brak energii oraz ogólne wyczerpanie. Obserwuje się również zaburzenia ze strony przewodu pokarmowego. Nikotyna jest związkiem, który powoduje uzależnienie fizyczne i psychiczne. W dostępnym piśmiennictwie nie znaleziono danych epidemiologicznych dotyczących zawodowego narażenia na nikotynę w postaci czystej. Nikotyna jest substancją o dużej toksyczności ostrej dla zwierząt – po podaniu dożołądkowym wartość DL50 mieści się w granicach 3,34 ÷ 188 mg/kg masy ciała. Informacje na temat toksyczności nikotyny wskazują na jej wielokierunkowe działanie. Narażenie drogą pokarmową szczurów na dawkę 1 mg/kg/dzień nikotynę przez 9 dni nie spowodowało żadnych zmian. Podobnie żadnych skutków nie zanotowano po podawaniu nikotyny szczurom w dawce 1,14 mg/kg/dzień przez 34 tygodnie. Dawka czterokrotnie większa powodowała wzrost aktywności niektórych enzymów w sercu szczurów narażonych przez 34 tygodnie. Podobna dawka podawana przez 9 dni wywoływała zmiany w zapisie EEG. Narażenie szczurów na nikotynę w dawce 3,5 mg/kg/dzień przez 90 dni oraz na nikotynę w dawce 12,5 mg/kg/dzień przez 28 dni (dawka skumulowana wynosiła odpowiednio: 315 lub 350 mg/kg) powodowało u zwierząt zaburzenia w gospodarce lipidowej i węglowodanowej. Z obserwacji zależności efektu toksycznego od wielkości narażenia po podaniu dożołądkowym nikotyny można przyjąć za wartość NOAEL dawkę 1,14 mg/kg/dzień, a za wartość LOAEL dawkę 4,56 mg/kg/dzień. Nikotyna nie wykazuje działania mutagennego, ale jest jednak genotoksyczna (wymiana chromatyd siostrzanych i aberracje chromosomowe) oraz fetotoksyczna. Udowodnione działanie rakotwórcze wykazują nitrozoaminy – związki powstające w wyniku palenia się tytoniu (NNN i NNK). Nikotyna dobrze wchłania się przez drogi oddechowe, przewód pokarmowy i skórę. Największe stężenia nikotyny stwierdzono w mózgu, nerkach, błonie śluzowej żołądka, rdzeniu nadnerczy, błonie śluzowej nosa i śliniankach. Nikotyna wiąże się z białkami osocza w 5 20% i przenika przez łożysko oraz do mleka matek karmiących. W trakcie metabolizmu nikotyna może ulegać: C-oksydacji, demetylacji połączonej z C-oksydacją, N-oksydacji oraz N-metylacji. Jej głównymi metabolitami są: kotynina i nikotyno-1’-N-tlenek. Nikotyna i jej metabolity są szybko wydalane przez nerki. Mechanizm działania nikotyny jest wypadkową aktywacji cholinergicznych receptorów nikotynowych powodujących pobudzenie komórek nerwowych i desensytyzacji powodującej zablokowanie przekaźnictwa sympatycznego. Działania obwodowe wywołane małymi dawkami nikotyny są wynikiem pobudzenia zwojów autonomicznych i obwodowych receptorów czuciowych, głównie w sercu i płucach. Pobudzenie tych receptorów wywołuje częstoskurcz, zwiększenie wyrzutu serca, wzrost ciśnienia tętniczego, zmniejszenie perystaltyki przewodu pokarmowego i pocenie się. Najbardziej rozpowszechnionym wśród ludzi przykładem działania łącznego nikotyny z innymi związkami jest palenie papierosów, w których – oprócz nikotyny – znajdują się setki innych substancji. Jednoczesnemu narażeniu szczurów na nikotynę i etanol towarzyszyło znaczące zmniejszenie ich płodności oraz zaburzenie reakcji immunologicznych u potomstwa. Nikotyna nasila hepatotoksyczne działanie CCl4. Na podstawie danych literaturowych przyjęto dawkę 1,14 mg/kg/dzień (po której nie zaobserwowano żadnych szkodliwych skutków) za wartość NOAEL nikotyny, zaś dawkę 4,56 mg/kg/dzień – za jej wartość LOAEL Po analizie danych literaturowych i wykonanych obliczeniach pozostano przy obowiązującej w Polsce wartości najwyższego dopuszczalnego stężenia (NDS) nikotyny wynoszącej 0,5 mg/m3 z oznaczeniami związku literami „Sk” i „Ft”. W dostępnym piśmiennictwie nie znaleziono informacji uzasadniających wyznaczenie dla nikotyny wartości najwyższego dopuszczalnego stężenia chwilowego (NDSCh).
Nicotine is an oily, colourless and odourless liquid obtained from leaves of tobacco plants. The most widespread use of nicotine is in tobacco as well as in remedies for nicotine abuse. Nicotine is a component of certain pesticides. Occupational exposure to nicotine is possible during its production and the tobacco drying process. To date only 8 people have been exposed in Poland to nicotine concentration in the air exceeding the TWA value which is 0.5 mg/m3 (data from 2002). Deadly occupational nicotine intoxication is very rare. The symptoms of severe nicotine intoxication with its small doses are: increased breath stimulation, nausea, vomitting, headache and vertigo, diarrhea, tachycardia, high blood pressure as well as sweating and excessive saliva production. After the administration of high doses of nicotine the following symptoms occured: burning sensations in the oral cavity, throat and stomach, fatigue, palpitations, weakening of the respiratory functions, disturbances of cardiac rhythm, dizziness, weakness, lack of coordination and coma. Death can then occur within 5 minutes up to 4 hours. Chronic nicotine intoxication leads to disturbances in the circulatory system. Vascular changes may lead to angina pectoris and heart attacks; they also cause: a weakening of memory, a slowdown of physical processes and thought coordination, lack of energy and exhaustion. Disturbances in the digestive system may also occur. Nicotine causes both physical and mental abuse. No epidemiological data was found concerning occupational exposure to nicotine in pure form. Nicotine is a substance of high acute toxicity to animals. After intragastrical administration the LD50 value is between 3.34 ÷ 188 mg/kg of body weight. Information concerning toxicity of nicotine indicates its multidirectional influence. Exposure of rats at oral doses (1 mg/kg/day, 9 days or 1.14 mg/kg/day, 34 weeks) caused no changes. When fourfold higher doses were administered to rats, after 34 weeks they caused an increase in the activity of certain enzymes in the heart, and the EEG changed after 9 days. Exposure to nicotine for 28 and 90 days (the accumulated dose was 350 or 315 mg/kg respectively) caused a disturbances in lipid and carbohydrate metabolism. Nicotine has no mutagenic potential, yet it is genotoxic (sister chromatid exchanges and chromosomal aberrations) as well as fetotoxic. Nitrosoamines (compounds produced due to tobacco smoking) have proved to show carcinogenic potential. Nicotine is well absorbed via respiratory tracts, the alimentary canal and the skin. The highest concentrations were detected in the brain, kidneys, stomach mucosa, adrenal medulla, nasal mucosa and salivary glands. Nicotine binds with plasma proteins in 5 - 20%. It penetrates through placenta and gets to the milk of nursing mothers. During metabolism nicotine can undergo: C-oxidation, demethylation with z C-oxidation, N-oxidation and N-methylation. Nicotine’s core metabolites are: cotinine and nicotine-1’-N-oxide. Nicotine and its metabolites are rapidly discharged by the kidneys. Smoking cigarettes is the most common example of nicotine activity together with many other compounds. In addition to nicotine, they include hundreds of other substances. Rats simultaneously exposured to ethanol and nicotine have shown impaired fertility and disturbance of immunological reactions occured in the offspring. Nicotine increases the hepatotoxic activeness of CCl4. Basing on the literature data 1.14 mg/kg/day has been accepted as a NOAEL value of nicotine (no negative results have been observed) whereas 4.56 mg/kg/day has been taken as its LOAEL value. After an analysis of published data and after conducting necessary calculations the MAC of nicotine in Poland remains unchanged: 0.5 mg/m3 with ‘Sk’ and ‘Ft’ compound symbols.

Zanieczyszczenie środowiska metalami ciężkimi jest jednym z ważniejszych zagrożeń dla zdrowia ludzi. Do organizmu człowieka metale ciężkie są wchłaniane przez skórę, wdychane, spożywane z produktami roślinnymi i zwierzęcymi. Przechodzenie pierwiastków do kolejnych ogniw łańcucha pokarmowego, a w konsekwencji do organizmu człowieka jest ograniczone działaniem barier biologicznych. Należy jednak podkreślić, że przy nadmiernym stężeniu pierwiastków działanie tych barier jest zmniejszone, co wiąże się z ryzykiem negatywnego ich oddziaływania na środowisko, a przede wszystkim na zdrowie człowieka. Metale mogą wywołać natychmiastowe ostre zatrucia lub stany przewlekłe. Zatrucia ostre powodują takie metale, jak: As, Zn, Cd, Cu, i Hg. Zatrucia przewlekłe mogą wywoływać m.in. As, Zn, Cd, Cr, Cu, Hg, Pb, Sn, Co, Ni, Mn, Se, Fe i Ag. Schorzenia przewlekłe występują przez długi czas w formie utajonej. Po pewnym czasie mogą wywołać bardzo niebezpieczne zmiany mutagenne lub uszkodzenia centralnego systemu nerwowego. Do najbardziej toksycznych metali ciężkich należą: ołów, rtęć i kadm. Stwierdzono istotne zależności między stężeniami metali ciężkich w spożywanych pokarmach czy wdychanym powietrzu a umieralnością na nowotwory.
Contamination of the environment with heavy metals is one of the major threats for human health. Heavy metals are absorbed by skin, inhaled and consumed with plant and animal products. Transport of elements to subsequent links of food chain, and in consequence to a human organism, is limited by functioning of biological barriers. It has to be pointed out that at the excessive concentration of these elements the functioning of these barriers is impaired, and thus leading to risk of negative impact on the environment, and primarily on human health. Heavy metals do not undergo biological degradation. Detoxification of heavy metals by living organisms mostly occurs through "hiding" active ions of heavy metals in protein structures such as metallothioneins or storing heavy metals in insoluble forms in intracellular granules for long-time accumulation or excretion with faeces. Heavy metals can cause acute toxicity or persistent conditions. Acute toxicity is caused by As, Zn, Cd, Cu, and Hg whereas persistent conditions are mostly due to As, Zn, Cd, Cr, Cu, Hg, Pb, Sn, Co, Ni, Mn, Se, Fe and Ag. Persistent conditions are in latent form for long periods. After some time they can cause extremely dangerous mutanogenic changes or damages to the central nervous system. The most toxic heavy metals include lead, mercury and cadmium. There is a significant correlation between the concentrations of heavy metals in consumed foods or inhaled air and cancer mortality. The correlations between malicious cancers of the gastro-intestinal tract, lymphatic system, bone marrow, breasts and the intake of cadmium or lead have been proven.

Fosforowe związki organiczne zmniejszające palność (flame retardants – FRs) stosowane są od kilkudziesięciu lat w wielu gałęziach przemysłu, m.in. w produkcji barwników, lakierów, klejów, syntetycznych żywic, polichlorku winylu, płynów hydraulicznych, tworzyw sztucznych i tekstyliów. Ostatnio ich znaczenie wzrasta, co spowodowane jest m.in. znacznym ograniczeniem stosowanych dotąd polibromowanych difenyloeterów (PBDEs) – niebezpiecznych dla środowiska trwałych zanieczyszczeń organicznych. Celem pracy był przegląd dostępnych danych literaturowych dotyczących fosforowych FRs przede wszystkim pod kątem ich działania neurotoksycznego, wpływu na płodność i rozrodczość oraz działania kancerogennego. Analiza dotyczyła najczęściej stosowanych substancji, którymi są: fosforan trietyloheksylu (TEHP), fosforan tributoksyetylu (TBEP), fosforan trifenylu (TPP), fosforan tris(2-chloroetylu) (TCEP), chlorek tetra(hydroksymetylo)fosfoniowy (THPC), fosforan tributylu (TBP), fosforan trikrezolu (TCP), fosforan tris(2-chloro-1-metyloetylu) (TCPP), fosforan tri(1,3-dichloroizopropylu) (TDCP) oraz siarczan tetrakis(hydroksymetylo) fosfoniowy (THPS). Działanie neurotoksyczne wykazano w badaniach na zwierzętach po narażeniu na TBEP, THPC, TBP i TCP. U ludzi działanie neurotoksyczne zaobserwowano tylko po narażeniu na TCP. Z kolei TCEP, THPS, TBP, TCP i TDCP powodowały zaburzenia płodności i/lub w rozwoju płodów zwierząt. Niekorzystny wpływ na rozrodczość u ludzi mogą powodować TPP, TCP i TDCP. Nowotwory u zwierząt laboratoryjnych powodowały podawane w wysokich dawkach THEP, TCEP, TBP i TDCP. Żaden z tych związków nie jest jednak klasyfikowany jako rakotwórczy dla ludzi. Toksyczność środowiskowa fosforowych FRs jest niewielka (z wyjątkiem TPP, TCEP i TBEP). Nie są one związkami trwałymi, w organizmach żywych podlegają przemianom metabolicznym i są z nich szybko wydalane. Mogą być więc alternatywą dla stosowanych do niedawna PBDEs. Med. Organophosphorus flame retardants (flame retardants, FRs) have been used for several decades in many industries, including the production of dyes, varnishes, adhesives, synthetic resins, polyvinyl chloride, hydraulic fluids, plastics and textiles. Their importance in recent times has increased due to i.a., significantly reduced use of polybrominated diphenyl ethers (PBDEs) – persistent organic pollutants, dangerous for the environment. The aim of this study was to review the available literature data concerning phosphorous FRs primarily for neurotoxic, fertility, reproductive and carcinogenic effects. The analysis concerned the following most commonly used substances: tris(2-ethylhexyl)phosphate (TEHP), tris(2-butoxyethyl)phosphate (TBEP), triphenyl phosphate (TPP), tris(2-chloroethyl)phosphate (TCEP), tetrakis(hydroxymethyl)-phosphonium chloride (THPC), tributyl phosphate (TBP), tricresyl phosphate (TCP), tris(2-chloroisopropyl)phosphate (TCPP), tris(1,3-dichloroisopropyl)phosphate (TDCP) and tetrakis(hydroxymethyl) phosphonium sulphate (THPS). In animal studies neurotoxic effects were found after exposure to TBEP, THPC, TBP and TCP, while in humans they were observed only after exposure to TCP. TCEP, THPS, TBP, TCP and TDCP caused disorders in fertility and/or fetal development of animals. Adverse effects on reproduction in humans may be caused by TPP, TCP, and TDCP. In laboratory animals the development of tumors was observed after high doses of TEHP, TCEP, TBP and TDCP. None of these compounds is classified as a human carcinogen. The environmental toxicity of phosphate FRs is low (except for TPP, TCEP and TBEP). They are not stable compounds, in living organisms they are metabolised and quickly excreted. Rational chemical modification of amphotericin B (AMB) led to the synthesis of sterically hindered AMB derivatives. The selected optimal compound, N-methyl- N-D-fructosyl amphotericin B methyl ester (MF-AME) retains the broad spectrum of antifungal activity of the parent antibiotic, and exhibits a two orders of magnitude lower toxicity in vivo and in vitro against mammalian cells. Comparative studies of MF-AME and AMB comprising the determination of the spectroscopic properties of monomeric and self-associated forms of the antibiotics, the investigation of the influence of self-association on toxicity to human red blood cells, and of the antibiotic-sterol interaction were performed. On the basis of the results obtained it can be assumed that the improvement of the selective toxicity of MF-AME could in part be a consequence of the diminished concentration of water soluble oligomers in aqueous medium, and the better ability to differentiate between cholesterol and ergosterol.

Coccidiosis continues to be one of the most serious diseases in poultry breeding, causing major economic losses in this industry. Many trials are conducted to prevent and control this disease. However, only a few medications are approved for use in the prevention and treatment of coccidiosis. Ionophore coccidiostats (e.g. salinomycin and narasin) act by altering the transmembrane movement of monovalent or divalent ions (Na+, K+, Ca2+, Mg2+, Rb+, Cs+), resulting in altered ionic gradients and disturbed physiological processes in coccidia. In broiler and turkey breeding, these agents are used for nearly the whole fattening period. Ionophore coccidiostats have a narrow safety margin. Their toxicity is probably due to a disturbed ion balance or to oxidative damage. Although ionophore coccidiostats are considered to be relatively safe for target animals, there are numerous reports of poisoning cases caused by these medications in birds. This paper summarizes the current state of knowledge on the toxicity of salinomycin and narasin in turkeys. It reviews the data concerning the symptoms, mortality rate and possible causes of poisoning with these agents. Moreover, the paper discusses the legal regulations regarding the use of these drugs in poultry.
Kokcydioza jest nadal uznawana za jedną z najważniejszych chorób w hodowli drobiu, stanowiąc główną przyczynę strat ekonomicznych w przemyśle drobiarskim. Podejmuje się wiele działań na rzecz zapobiegania i kontroli tej choroby. Do profilaktyki i terapii kokcydiozy zatwierdzono tylko kilka leków. Mechanizm działania kokcydiostatyków jonoforowych (np. salinomycyny i narazyny) jest związany z zakłóceniem transbłonowego transportu monowalentnych i dwuwalentnych jonów (Na+, K+, Ca2+, Mg2+, Rb+, Cs+), prowadzącym do zmiany gradientu jonowego i wtórnego zaburzenia procesów fizjologicznych u kokcydiów. U brojlerów i indyków leki te podawane są przez prawie cały okres tuczu. Kokcydiostatyki jonoforowe mają wąski margines bezpieczeństwa. Ich toksyczność jest związana z zaburzeniem równowagi jonowej lub uszkodzeniami oksydacyjnymi. Chociaż kokcydiostatyki jonoforowe są uznawane za stosunkowo bezpieczne dla zwierząt docelowych, w literaturze można znaleźć liczne doniesienia na temat zatruć tymi lekami u ptaków. W artykule przedstawiano bieżący stan wiedzy na temat toksyczności salinomycyny i narazyny u indyków, a także dane dotyczące objawów, upadków i możliwych przyczyn zatruć wywołanych tymi lekami. Omawiano również regulacje prawne w zakresie ich stosowania.

Influence of melittin on viability and integrity of cell membrane on grade IV glioma. The grade IV glioma is one of the malignant human tumours. Today there is no effective treatment for this type of cancer. Alternative methods are sought-after in glioma treatment, and lately melittin has been found to be useful in anticancer therapy. The aim of the study was to investigate the effect of melittin on the viability and the integrity of cell membranes of the grade IV glioma cells. The U87 glioma line cells were treated of melittin in increasing concentrations (5, 10, 15, 20 and 50 µg/mL) and incubated for 24 hours. After incubation, the tests were performed in order to investigate the cell morphology, cell viability, membrane integrity and the way of cell death. The results have shown the devastating effect of melittin on the glioma cells. The melittin causes disintegration of cell membranes and induces cell death by apoptosis and less by necrosis.
Wpływ melityny na żywotność i integralność błon komórkowych glejaka IV stopnia. Glejak IV stopnia jest jednym ze złośliwych nowotworów ludzkich. Do dziś nie wynaleziono skutecznego leku do walki z tym nowotworem. Melityna to jeden ze składników jadu pszczelego, który wykazuje działania antyrakowe. Celem badań było zbadanie wpływu melityny na żywotność i integralność błon komórkowych glejaka IV stopnia linii U87. Komórki glejaka IV stopnia linii U87 były traktowane roztworami melityny we wzrastającym stężeniu (5, 10, 15, 20 and 50 µg/mL) i inkubowane przed 24 godziny. Po inkubacji przeprowadzono badania w celu sprawdzenia morfologii komórek, ich żywotności, integralności błon komórkowych oraz drogę śmierci komórkowej. Wyniki wskazują na niszczący wpływ melityny na komórki glejaka. Melityna powoduje dezintegrację błon komórkowych oraz indukuje śmierć komórkową na drodze apoptozy oraz, w mniejszym stopniu, nekrozy.
