Opis:
Introduction. Squamous cell carcinoma (SCC) of the larynx accounts for a significant percentage of all head and neck cancers. Aim. In this paper we determine the differences in magnetic resonance relaxation time (MRI) of water in cancerous and healthy larynx tissues. Material and methods. This study is aimed on T2 MRI modalities for monitoring morphology of larynx tissue. Results. Our results showed that T2 MRI relaxation time measured in larynx tissue can be used to assess early cancer condition of larynx tissues. The changes of T2 MRI correspond to tumor growth within normal tissue. Conclusion. The study showed potential of MRI for the non-invasive monitoring of larynx condition.

Opis:
Chronic rhinosinusitis with nasal polyps (CRSwNP) has a significant impact on the well-being and social functions of the patient. The generalized inflammatory process with the formation of nasal polyps and excess eosinophils in the mucosa of the paranasal sinuses is called type 2 inflammation, which is mediated by Th2 lymphocytes – cells of the immune system responsible for chronic inflammatory processes. Today, we also know the key pro-inflammatory mediators against which new drugs have been developed, the so-called biological drugs, are produced in cell lines. In this document, we present currently available biologicals approved for the treatment of patients with T2-related chronic rhinosinusitis.
Przewlekłe zapalenie błony śluzowej nosa i zatok przynosowych z polipami nosa (PZZPzPN) wywiera duży wpływ na samopoczucie i funkcje społeczne chorego. Uogólniony proces zapalny przebiegający z powstawaniem polipów nosa i nadmiarem eozynofilów w obrębie błony śluzowej zatok przynosowych to zapalenie typu 2., w którym pośredniczą limfocyty Th2 – komórki układu immunologicznego, odpowiadające za przewlekłe procesy zapalne. Dziś znamy też kluczowe mediatory prozapalne, przeciwko którym powstały nowe leki, tzw. leki biologiczne, wytwarzane w liniach komórkowych. W dokumencie przedstawiamy dostępne, w obecnej chwili, leczenie biologiczne zatwierdzone do leczenia pacjentów z T2 – zależnym przewlekłym zapaleniem zatok przynosowych.

Introduction. Type 2 Diabetes Mellitus (T2DM) is a group of metabolic disorders resulting from insufficient action of insulin. The etiology of T2DM is multi-factorial that includes genetic factors, obesity and lifestyles. Recent reviews of overall and stratified meta-analyses demonstrated the association between MTHFR polymorphism (C677T) including fat distribution and risk of T2DM. Publications of Indian context regarding fat patterning and MTFHR genetic polymorphism of the North East Indian population are insufficient and scant among the ethnic population of Tripura. Aim. In this backdrop, the present study is the first attempt to understand the relationship of fat patterning, MTHFR gene polymorphism and T2DM among two Tibeto-Burman speaker endogamous ethnic populations (Chakmas-the migrant group and Tripuris – the aboriginal group) of Tripura, North East India. Material and methods. The present study consists of age matched 280 males (Chakmas 147 and the Tripuris 133) from Tripura. Anthropometric and metabolic (Fasting Blood Glucose) variables and to discern obesity, blood glucose level and genotyping of MTHFR was performed following standard techniques. Results. The result revealed significant (p<0.05) association of obesity, TT genotypes and fasting blood glucose among the Chakmas with in comparison to the Tripuris. Conclusion. In this first attempt from North East India on the aspects of association of fat Patterning, Type 2 Diabetes Mellitus and MTHFR gene polymorphism suggests that the Chakmas are more diabetic, and this might be due to the concomitant effects of T alleles and higher central obesity and Percent Body Fat (PBF). More population screening from other under-represented indigenous populations of North East India is needed for prevention of metabolic disorders.

Surveys of oats in the UK and Nordic countries have identified high concentrations of HT-2 and T-2 can occur in unprocessed oats. HT-2 and T-2 are two closely related type A trichothecenes and two of the most toxic fusarium mycotoxins. There is currently no legislation on HT-2 and T-2, however, there is a discussion limit of 500 µg kg-1 in unprocessed oats. A previous survey identified that variety, previous crop, cultivation, fungicide use and some other factor(s) within organic oat production, which was not within the model, were all significant agronomic factors in the determination of HT-2 and T-2 concentrations of UK oats. Possible agronomy within conventional compared to organic agriculture would include the use of inorganic fertilisers and plant growth regulators (PGR). Oats harvested from two series of agronomic field experiments were analysed for the combined concentration of HT-2 and T-2 (HT2+T2) using ELISA. Experiments were repeated for both winter and spring varieties over two years. The first experiments were of a factorial design with three varieties, three nitrogen rates and plus/minus a PGR (chlormequat). The second series had twelve fungicide regimes. The results identified that there were no significant differences in HT-2+T-2 between samples from oat plots that received different rates of inorganic nitrogen, a PGR, or a range of different fungicide regimes. Opis:
Nuclear Magnetic Resonance (NMR) data provide affluence of information about porous space saturation distribution, porosity itself and permeability, thanks to close relationship between pore sizes and transverse relaxations time distribution. Having known values of these parameters, and especially irreducible water saturation of rocks it is possible to determine reservoir properties of rocks much more precisely. The study presents results of laboratory tests carried out with use of NMR method on the Miocene sandstones representing Dzików gas field. The samples featured extensive variability of reservoir properties within profile of individual boreholes. The highest number of rocks having good reservoir and filtration properties, originated from Dzików-12 borehole, and they were characterised by low irreducible water saturation coefficient and high free water volume.

Species within the genus Fusarium produce a diverse range of mycotoxins, many of which have signifi- cant impacts on human health. Of the five generally recognised major mycotoxins, three (fumonisins, de- oxynivalenol (DON) and zearalenone (ZON)) are produced by Fusaria. Apart from DON, other trichothecenes such as T-2 toxin, have received considerable international attention due to their impact on human health. The fumonisins, which occur ubiquitously in maize and its products, have been linked to oesophageal cancer, liver cancer and neural tube defects. DON, a frequent contaminant of maize, wheat and their products, although showing no carcinogenic potential, is immunomodulatory and produces emesis and growth retardation in animals. ZON is a naturally occurring endocrine disrupting chemical. Acute exposure to these mycotoxins has in each case been linked to outbreaks of human disease – gastro-intestinal effects in the case of fumonisins and DON, and precocious pubertal changes in the case of ZON. Concern over their toxicological effects has led to risk assessments by the Joint FAO/WHO Expert Committee on Food Additives (JECFA), which has set maximum tolerable daily intakes (TDI) of 2 µg/ kg body weight (bw) for fumonisins and 0.5 µg/kg bw for ZON. The initial TDI set for DON, namely 1 µg/kg bw has recently been updated by JECFA to include both 3- and 15-acetylDON. Apart from the above mycotoxins, a number of other secondary metabolites (moniliformin, beauvericin and fusaproliferin) are produced by different Fusaria and their effects on human health, either alone or in combination with other mycotoxins, is largely unexplored.

In the present work Fusarium DNA levels of deoxynivalenol (DON)-producing F. graminearum and F. culmorum and T-2/HT-2-producing F. langsethiae and F. sporotrichioides were compared to DON and T-2/HT-2 levels in Finnish (19 oat samples) and Estonian (3 oat, 8 wheat and 9 barley) grain samples of the year 2008. The Finnish oat samples were selected based on high (<870 ppb) DON levels. Some of them also contained high levels of zearalenone (ZEN). The main aim was to find out, which Fusarium species is the main DON producer in oats in Finland. In Estonian grain sam- ples all mycotoxin levels were relatively low and below the EU limits. The highest ZEN levels were found in oat in Satakunta in Finland. This is the first study, in which F. graminearum has been found in Estonian grains. The correlation between F. graminearum DNA and DON levels in the combined Finnish and Estonian oat samples was higly significant (R2 = 0.88). In Finnish samples a significant correlation was also found between F. graminearum DNA and ZEN levels (R2 = 0.42). No correlation was found between F. culmorum DNA and DON levels in the combined Finnish and Estonian oat sam- ples. There was also a significant correlation between the combined T-2 and HT-2 and combined F. langsethiae and F. sporotrichioides levels (R2  = 0.38) in the combined Finnish and Estonian oat samples. According to our results F. graminearum is clearly the main DON producer in Finnish oat.

The ubiquitin-conjugating enzyme E2E 2 (UBE2E2) gene plays an important role in insulin synthesis and secretion under conditions in which stress to the endoplasmic reticulum is increased in β-cells. In this case-control study, we have selected rs7612462 polymorphism within UBE2E2 gene to identify in a Saudi population the type 2 diabetes mellitus (T2DM) subjects. In total, 376 subjects with T2DM and 380 controls were enrolled in this study. We have collected 5 mL of peripheral blood from each participant for biochemical and molecular analyses. PCR-RFLP was used to generate genotypes at rs7612462 in all of the study subjects. We report on the photoresponse of mid-wavelength infrared radiation (MWIR) type-II superlattices (T2SLs) InAs/InAsSb high operating temperature (HOT) photoresistor grown on GaAs substrate. The device consists of a 200 periods of active layer grown on GaSb buffer layer. The photoresistor reached a 50% cut-off wavelength of 5 μm and 6 μm at 200 K and 300 K respectively. The time constant of 30 ns is observed at 200 K under 1 V bias. This is the first observation of the photoresponse in MWIR T2SLs InAs/InAsSb above 200 K..

Oilseeds are a perfect medium for microfungi spread and mycotoxin production. With increasing demand for oil crop produce such research has gained a special relevance since research evidence on this issue is scarce. During 2007-2009, prevalent fungi genera, including Fusarium genus, potential producer of deoxynivalenol (DON), zearalenone (ZEA), T-2 toxin (T-2) etc. were determined in our tests. The ELISA immunoenzymatic method and Veratox Fast kits were used to identify and quantify mycotoxins, while Fusarium fungi species were identified using conventional fungi determination techniques. Higher Fusarium fungi contamination level was found on linseed compared with that on spring or winter rapeseed. The difference was even more obvious in different experimental years, however, having identified Fusarium species, F. avenaceum and F. oxysporum were found to be prevalent in the seed of all crop species tested. In 2009, spring rape samples were found to contain F. dimerum which is a significant human pathogen.The presence of DON was identified in 18.2-100%, ZEA in 40 -100%, and T-2 toxin in 100% of seed samples of all oil crop species tested. From the food safety viewpoint, the concentrations determined did not exceed the levels hazardous for health, laid out in the EU regulations, however, the effect of low toxin concentrations is slow and the negative consequences manifest themselves only after some time and in various forms, which poses a serious health risk for humans and animals.

Introduction This literature review explores the role of physical activity (PA) in managing and preventing type 2 diabetes mellitus (T2DM), synthesizing the latest guidelines for all T2DM patients. Following PRISMA guidelines, it identifies specific areas for further research. Material and Methods Indexing services (PubMed and Scopus) were used to identify relevant studies, emphasizing original research, review articles, and updated institutional guidelines spanning 2017-2023. The following keywords were used: diabetes, physical activity, type 2 diabetes mellitus, PA, and exercise. Proposal and opinion articles, non-English papers (or those lacking full-text translation), studies involving non-human subjects, pediatric populations or with skewed gender distribution were excluded. Results PA improves quality of life, BMI, glycemic control and well-being. Effects vary by activity domain, such as work or leisure time. Recommendations suggest 60 daily minutes of exercise for children, 150 weekly minutes of moderate intensity exercise for adults, and screening for adverse events. A “sit-less” approach is proposed for those unable to maintain regular activity, with glycemic monitoring for those with variability. Conclusions PA is crucial in T2DM management. Conflicting findings regarding glycemic control warrant further investigation to ascertain causes, whether related to bias or other factors.

W celu realizacji krajowego planu monitoringu żywności opracowano, zwalidowano i akredytowano metodę oznaczania toksyn T-2 i HT-2 w produktach zbożowych. Do analizy toksyn zastosowano metodę pojedynczej ekstrakcji i oznaczania techniką HPLC/MS-MS ESI+. Prametry skuteczności metody (odzysk, precyzja i niepewność wyniku) są zgodne z kryteriami podanymi w Rozporządzeniu Komisji (WE) nr 401/2006. Granica wykrywalności metody dla toksyny T-2 wynosi 3 µg/kg, a dla toksyny HT-2 4 µg/kg. Próbki do badań były pobierane przez stacje sanitarno - epidemiologiczne z terenu całego kraju. Oznaczenia wykonano w 107 próbkach przetworów zbożowych, otrzymanych głównie z owsa. Toksyny T-2 i HT-2 stwierdzono w 43 % próbek, średni poziom zanieczyszczenia w próbkach produktów pochodzących z owsa wynosił 22,5 µg/kg (najwyższy- 109 µg/kg w płatkach owsianych), w pozostałych (płatki pszenne i jęczmienne, kasze, mąka) 7,0 µg/kg. Uzyskane dane wskazują, że wartości tolerowanego dziennego pobrania (TDI) dla toksyn T-2 i HT-2 nie są przekroczone.
In framework of the national monitoring program in Poland HPLC method for determination of T-2 and HT-2 toxins in cereal products was developed, validated and accredited. Simply, one-step extraction and HPLC MS/MS ESI+ method was used for determination both toxins. Performance of method (recovery, precision and uncertainty of results) is in line with Commission Regulation No 401/2006. Limit of detection for T-2 and HT-2 toxins is 3 ad 4 |µg/kg, respectively. Samples were taken by sanitary inspection from all region of country. 107 samples cereal products (mainly from oats) were tested. T-2 and HT-2 toxins were detected in 43 % samples, mean level in oats products was 22.5 µg/kg (maximum level 109 µg/kg in oat flakes), in other samples (wheat and barley flakes, grouts, flours) - 7.0 µg/kg. Intake of T-2/HT-2 toxin by the consumer in Poland is much lower than the TDI.

Opis:
The aim of the study was to assess the frequency of occurrence and concentrations of mycotoxins in grass silages depending on meadow location, term of harvest and type of fertilizer used. The research material consisted of 24 grass silage samples collected in 2013 from three farms located in: Falenty, Kodeń and Kąty. The following mycotoxins were evaluated: deoxynivalenol (DON), nivalenol (NIV), T2 and HT2 toxin, zearalenone (ZEA) and roquefortine C (ROQ C). 22 samples were positive for at least one mycotoxin. The most frequently detected toxins were NIV (14 samples contaminated), DON and ZEA (10 samples). The concentration of mycotoxins in silage samples coming from individual farms varied and significant differences between farms were obtained only in the case of T2 toxin and ZEA. Silage made out of sward from the third cut was more likely to be contaminated, than silage made out of sward from the first cut. The type of fertilizer had no significant impact on mycotoxin concentrations except for NIV and HT2 toxin.
Celem pracy była ocena częstotliwości występowania i zawartości mykotoksyn w kiszonce z runi łąkowej w zależności od lokalizacji gospodarstwa, terminu zbioru runi łąkowej oraz rodzaju nawożenia stosowanego do nawożenia łąki. Materiał badawczy stanowiły 24 próbki sianokiszonki pobrane w 2013 r. w trzech gospodarstwach: w Falentach, Kodniu i Kątach. Oceniono zawartość następujących mykotoksyn i toksyn: deoksyniwalenolu (DON), niwalenolu (NIV), toksyn T2 i HT2, zearalenonu (ZEA) i roquefortyny C (ROQ C). 22 próbki były skażone co najmniej jedną mykotoksyną. Najczęściej wykrywanymi toksynami były NIV (14 próbek) DON i ZEA (po 10 próbek). Zawartość poszczególnych mykotoksyn w próbkach kiszonki pochodzących z poszczególnych gospodarstw była bardzo zróżnicowana, ale istotne różnice między gospodarstwami stwierdzono tylko w przypadku dwóch mykotoksyn: toksyny T-2 i ZEA. Kiszonki sporządzone z runi łąkowej zebranej w trzecim pokosie zwykle były bardziej zanieczyszczone niż kiszonki z pierwszego pokosu. Rodzaj nawożenia nie miał istotnego wpływu na poziom skażenia mykotoksynami, z wyjątkiem toksyn NIV i HT2

Opis:
Fusarium langsethiae has been identified as the primary producer of HT-2 and T-2 in European cereals. HT-2 and T-2 are considered as two of the most potent trichothecenes mycotoxins and a public health concern in Europe. There is currently no legislation on HT-2 and T-2, however, there is a discussion limit of 500 µg kg -1  in unprocessed oats, 200 µg kg-1 for oat products and 50 µg kg-1 for infant food. There are limited data regarding F. langsethiae’s pathogenicity and mycotoxin production, but it is evident that its behaviour deviates from traditional trichothecene-producing Fusarium species. This experiment was aimed at assessing the aggressiveness (measured by lesion length) of 20 different F. langsethiae isolates on wheat, barley and oats using an in vitro detached leaf assay. There was a significant (P<0.001) difference between lesion lengths formed by different F. langsethiae isolates used. Isolate Fl/2004/17(a) caused the shortest lesion on all cereals and this was significantly (P<0.001) different from that caused by isolate Fl/0/08/009/1 which caused the longest lesion on all cereals used. A highly significant difference (P<0.001) was also observed between lesions on the different cereals (wheat, barley and oats). Lesions on oats were the longest, followed by barley with wheat showing the shortest lesions.

Przeprowadzono ocenę zawartości wody nieruchomej metodą magnetycznego rezonansu jądrowego. Określenie ilości wody nieruchomej w przestrzeni porowej jest niezbędne do wyznaczenia porowatości dynamicznej, a co za tym idzie - charakterystyki własności zbiornikowych skał. Kluczowym parametrem w tego typu badaniach jest czas graniczny T2G (parametr umożliwiający rozdzielenie wody nieruchomej od wolnej). Parametr T2G wyznaczono dla piaskowców reprezentujących różne baseny sedymentacyjne. Zmienność wyznaczonych wartości T2G (3-15 ms) związana jest ze zróżnicowaniem wykształcenia przestrzeni porowej i składu mineralnego badanych skał. Średnia wartość wyznaczonego czasu granicznego (9.14 ms) znacząco odbiega od wartości średniej (33 ms) podawanej dla piaskowców w literaturze. Przyjęcie wartości T2G = 33 ms może spowodować, że błędy w określeniu porowatości dynamicznej będą sięgać nawet 100%. Widoczne jest więc, że określenie rzeczywistych czasów odcięcia badanych skał jest bardzo istotne dla prawidłowej interpretacji pomiarów NMR.
The irreducible water content has been evaluated using nuclear magnetic resonance (NMR) method. Estimation of the irreducible water content in the pore space is indispensable for the productive porosity evaluation and characteristics of the reservoir properties of rocks. The irreducible water content has been established calculating the cutoff time T2G (parameter which enables the division between irreducible and free water). Different types of sandstones were investigated. Variability of the estimated T2G values (3-15 ms) is connected with the diversity of the pore space structure and mineralogy of the investigated rocks. The average T2G value (9.14 ms) established experimentally differs considerably from the average value presented in the literature (33 ms). Use of an average T2G = 33 ms value in the interpretation of the NMR measurements may result in great, even 100 percent errors in the estimation of the producible porosity. Thus, it is clear that investigations of real T2 values for rocks from different sedimentary basins is necessary.
