Opis:
Ionizing radiation applied on food eliminates harmful microorganisms, prevents sprouting and delays ripening. All methods for detection of irradiated food are based on physical, chemical, biological or microbiological changes caused by the treatment with ionizing radiation. When minerals are exposed to ionizing radiation, they accumulate radiation energy and store it in the crystal lattice, by which some electrons remain trapped in the lattice. When these minerals are exposed to optical stimulation, trapped electrons are released. The phenomenon, called optically stimulated luminescence or photostimulated luminescence, occurs when released electrons recombine with holes from luminescence centers in the lattice, resulting in emission of light with certain wavelengths. In this paper, the results of measurements performed on seven different samples of herbs and spices are presented. In order to make a comparison between luminescence signals from samples treated with different doses, unirradiated samples are treated with Co-60 with doses of 1 kGy, 5 kGy and 10 kGy. In all cases it was shown that the higher the applied dose, the higher the luminescence signal.

Large quantity of sewage sludge originating from wastewater treatment plants is becoming a growing problem from environmental and human health points of view. One of the ways to use sewage sludge is agricultural purpose due to its nutrients and organic matter content, but the condition is that it should be deprived of pathogenic bacteria and parasite egg contamination. Application of ionizing radiation to hygienize sewage sludge can make it appropriate for agricultural use. The process does not require addition of chemicals to sludge; it is environmentally friendly and effective in removal of biological threats. In the past, successful attempts of sewage sludge treatment using ionizing radiation were made. Pilot plants and commercial ones proved that pathogens can be easily removed from sewage sludge by ionizing radiation. The concept of ‘zero energy’ biogas plant is based on the construction of a complex system consisting of biogas plant and electron accelerator in the biofertilizer manufacturing line. Digestate originating from the methane fermentation of sewage sludge is irradiated to remove all pathogens using electron beam from an accelerator powered by electric energy obtained from burning biogas in a cogenerator. The product is a high-quality, biologically safe fertilizer.

Recently, significant attention has been paid to the possibility of thwarting cancer progression by inhibition of neoangiogenesis (formation of new blood vessels) in growing tumors. Although general mechanisms of angiogenesis have been elucidated, virtually nothing is known about the effects of low doses of ionizing radiation on pro-angiogenic properties of endothelial cells. In the present study, we evaluated the effects of a low (0.2 Gy), intermediate (1 Gy), and high (4 Gy) doses of X-rays on a few angiogenesis-related parameters of isolated murine endothelial cells. We show here that 24 to 48 hours after irradiation with 0.2 Gy the cell proliferation was inhibited to a similar extent as after the exposure to 1 Gy. Also, adhesion of the 0.2 Gy-irradiated cells to both gelatin and MatrigelŽ was inhibited 24 hours post-exposure, whereas irradiation with 1 or 4 Gy resulted in the increased adhesion of the cells to these substrata. Similar effects were observed during the "wound" migration assay. Finally, 24 hours after exposure of the cells to 0.2 Gy of X-rays, the surface expression of the â3 integrin subunit was down-regulated, whereas irradiations with 1 and 4 Gy of X-rays resulted in the significantly elevated expression of this subunit. These results indicate that proliferating endothelial cells are sensitive in vitro to relatively low doses of ionizing radiation

he movement toward commercialization of the process and the recent actions of various governmental agencies to accept irradiated foods as wholesome, even when high doses are applied has stimulated increased interest in this treatment. Concern for the fate of vitamins when foods are irradiated is almost entirely centered on maintenance of their biological function as essential nutrients. The aim of this work was to assess the effects of ionizing radiation on vitamin A and ßeta-carotene contents in commercialized food products of animal origin, specifically fresh bovine liver and pork pâté de foie. As present Brazilian legislation has no restriction of dose limits to be applied on foods, the 60Co gamma irradiation doses for these experiments were 3 kGy and 30 kGy. The results show a full retention of vitamin content when the applied dose was 3 kGy and a loss of about 60% when the dose was 30 kGy. When applying high doses, it would be necessary to consider this content reduction and proceed to a vitamin supplement when necessary.

Opis:
In recent years there is a trend in industry to limit the usage of chemical compounds. Natural polymers are new promising materials that possess important properties like biodegrability or lack of toxicity. Radiation processing of natural occurring polymers is an area of current research for development of new applications. The aim of this study was to determine the impact of ionizing radiation modification on bioactivity of a natural polysaccharide. Chitosan with different molecular weights was investigated as a biostimulator – a biologically active substance that stimulates some growth processes in plants. Chitosan in solid state was irradiated with electron beam from an electron beam accelerator Elektronika 10-10 with a dose range from 50 to 300 kGy. The effects of irradiation on the molecular weight of chitosan were investigated by viscosity and GPC measurements. Non-irradiated and irradiated chitosan at concentrations 0.001, 0.01, 0.1 and 1 g/dm3 were used for greenhouse tests of its activity for growth promotion of Salix viminalis L. var. gigantea plant. Uniform rooted cuttings (20 per combination) were selected for the test and cultivated in aerated hydroponics culture containing Hoagland’s nutrient solutions plus respective amounts of chitosan. After six weeks of plant exposure to chitosan, data of selected parameters of plant growth were collected. In most cases, except the highest concentration, both forms of chitosan had stimulatory effect on leaf area, length of roots and of newly developed shoots. Also fresh and dry weights of these organs were greater in chitosan treated plants. The highest concentration of chitosan was stimulatory only for a number of roots and newly developed shoots while for other parameters was inhibitory. In comparable concentrations the stimulatory effect was greater for chitosan irradiated in comparison with the non-irradiated one.

Opis:
With increasing population and human activities, dumpsites experience diverse sources of waste with potential radiation hazards. Ionizing radiations have often been overlooked amongst researchers in third world countries, hence, information in this regard is lacking. In this study, nine dumpsites within Imo State University (IMSU) grounds were assessed for radiation levels using Geiger Muller counter Tube Mullard type ZP 1481 with assisted scalar and stopwatch. After calibration, the instrument was placed one meter above ground level and count rates recorded at 10 minutes interval for each location once in the morning, the afternoon and the evening for seven days. Results reveal higher dose equivalents in some morning periods than in the afternoon and evening. Site ETF2 had highest value of 0.69±0.08 mSv·yr-1, while Extension gate had lowest value of 0.56±0.01 mSv·yr-1. Compared to National Council for Radiation Protection maximum permissible level, these values are quite low. In conclusion, ionizing radiations levels at the studied dumpsites in Imo State University grounds are low and may not pose a threat to persons within the university.

In the present work, twin screw extruded films of PLLA and PCL biodegradable homopolymers and 50:50 (w:w) blend were irradiated with gamma rays from Co-60 and electron beam at doses in the range of 50 to 500 kGy in order to evaluate the effect of irradiations on homopolymers and blend. The FTIR results have shown that this technique was neither sensitive enough to observe the degradation promoted by ionizing radiation of studied homopolymers and blends and, nor on the miscibility of the blends. Wide-angle X-ray diffraction (WAXD) of PCL samples, non-irradiated and irradiated showed two strongest reflections at angles 2theta = 21.4 degrees centigrade and 2theta = 23.7 degrees centigrade that have been attributed in the literature to the (110) and (200) reflections, respectively. As for extruded non-irradiated and irradiated PLLA, it was observed broad diffusion peaks corresponding to an amorphous polymer. PLLA annealed samples showed reflections at angles 2theta =16.4 degrees centigrade and 2theta =18.7 degrees centigrade previously attributed in the literature to distorted 103 (alfa-form) helices. It was possible to observe slight alteration of the crystallite size of all irradiated samples of PCL in the dose range studied.

cellular signalling pathways

Opis:
DNA is the universal primary target for ionizing radiation; however, the cellular response is highly diversified not only by differential DNA repair ability. The monitoring system for the ionizing radiation-inflicted DNA damage consists of 3 apparently independently acting enzymes which are activated by DNA breaks: two protein kinases, Atm (ataxia telangiectasia mutated) and DNA-PK (DNA-dependent protein kinase) and a poly(ADP-ribose) polymerase, PARP-1. These 3 enzymes are the source of alarm signals, which affect to various extents DNA repair, progression through the cell cycle and eventually the pathway to cell death. Their functions probably are partly over-lapping. On the side of DNA repair their role consists in recruiting and/or activating the repair enzymes, as well as preventing illegitimate recombination of the damaged sites. A large part of the nuclear signalling pathway, including the integrating role of Tp53 has been revealed. Two main signalling pathways start at the plasma membrane: the MAPK/ERK (mitogen and extracellular signal regulated protein kinase family) "survival pathway" and the SAPK/JNK (stress-activated protein kinase/c-Jun N-terminal kinase) "cell death pathway". The balance between them is likely to determine the cell’s fate. When DNA break rejoining is impaired, the cell is unconditionally radiation sensitive. The fate of a repair-competent cell is determined by the time factor: the cell cycle arrest should be long enough to ensure the completion of repair. Incomplete repair or misrepair may be tolerated, when generation of the death signal is prevented. So, the character and timing of the signals are, to a large part, responsible for the cellular intrinsic radiation sensitivity and depend on the characteristics of the cellular signalling web.

Several lines of evidence indicate that exposure of heart to ionizing radiation increases the risk of cardiotoxicity manifested by heart dysfunction and cardiovascular diseases. It was initially believed that the heart is an organ relatively resistant to radiation. Currently, however, it is suspected that even low doses of radiation (< 2 Gy) may have a negative impact on the cardiovascular system. Cardiotoxicity of ionizing radiation is associated with metabolic changes observed in cardiac cells injured by radiation. In this study, we used human cardiomyocytes as a model system, and studied their metabolic response to radiation using high-resolution magic angle spinning nuclear magnetic resonance techniques (HR-MAS NMR). Human cardiomyocytes cultured in vitro were exposed to ionizing radiation and their survival was assessed by clonogenic assay. Changes in apoptosis intensity and cell cycle distribution after the irradiation were measured as well. NMR spectra of cardiomyocytes were acquired using Bruker Avance 400 MHz spectrometer at a spinning rate of 3200 Hz. Survival of cardiomyocytes after NMR experiments was assessed by the Trypan blue exclusion assay. Exposure of cardiomyocytes to small doses of ionizing radiation had no effect on cell proliferation potential and intensity of cell death. However, analysis of metabolic profiles revealed changes in lipids, threonine, glycine, glycerophosphocholine, choline, valine, isoleucine, glutamate, reduced glutathione and taurine metabolism. The results of this study showed that ionizing radiation affects metabolic profiles of cardiomyocytes even at low doses, which potentially have no effect on cell viability.

Opis:
Ionizing radiation has extensive applications in agriculture for inducing mutagenic changes in major field crops, potential breeding purposes, varietal development and crop improvement. This study was conducted to test the efficacy of 0.8, 1.6, 2.4 and 3.6 kGy gamma irradiation doses (Cobalt-60) upon growth and yield performance of edible pea (Pisum sativum L.) in pot culture experiment during 2009. Results demonstrated that higher radiation doses (1.6 and 3.2 kGy) significantly influenced the studied attributes of P. sativum. It was observed that 3.2 kGy had detrimental effects on shoot and pod lengths of pea which were reduced by 14.60 and 17.71 % respectively when compared to control. Differential response of the number of seeds pod-1, 1000 grain weight and dry biomass of pea were recorded at the applied doses. Significant reduction in number of seeds (-14.21 %) but increase in 1000 grain weight (+13.93 %) and dry biomass (+11.32 %) of pea were recorded at 1.6 kGy which revealed stimulatory effects on grain weight and dry biomass. Conversely, radiation dose 3.2 kGy was found detrimental to all the studied parameters except number of pods plant-1 and number of seeds pod-1 which were not affected.

The aim of the present study was to evaluate the cellular radiosensitivity and radiation-induced DNA damage and repair in Chinese hamster ovary (CHO) cells. Cell survival after irradiation was assessed using the clonogenic assay. The initial, radio-induced and residual DNA damage in Chinese hamster ovary (CHO) cells exposed to 60Co gamma-rays were determined using the alkaline comet assay. A linear-quadratic (LQ) survival curve was observed in CHO line. Data obtained by comet assay demonstrated a linear dose-response correlation in the range of tested doses (0.3-4 Gy). The process of DNA repair was modeled by exponential equation. In addition, we found a good correlation (R2 = 0.995) between clonogenic cell survival and radio-induced DNA damage.

The purpose of the presented study was to investigate the effect of ionizing radiation on selected physicochemical properties of segmented polyurethane (PU). Polyurethane was characterized by electron paramagnetic resonance (EPR), measurement of dynamic contact angle and investigation of thermal and mechanical properties. The results obtained by these techniques revealed insignificant influence of radiation on structure and features of investigated polymer up to 112kGy. Thus it was confirmed that upon exposition to a sterilization dose of about 25kGy polyurethane does not change chemical and mechanical properties.

Introduction and objective. The tendency towards postponement of maternity implies a greater exposure of female germ cells to damaging environmental effects, including ionizing radiation (IR). Progress in paediatric oncology, based on the use of radiotherapy, also implies the occurrence of gonadal dysfunctions and subsequent female fertility disorders. Therefore, it seems justifiable to systematize the state of knowledge concerning the effect of IR on the female reproductive system. Brief description of the state of knowledge. A considerable part of studies concerning the effect of IR on female germ cells have been conducted on animals. Their extrapolation to humans is hindered because in animal studies high acute exposures are applied, which do not reflect human environmental exposures characterized by chronic low dose exposure. Studies on animals provide a heterogenous image, which hinders the formulation of unequivocal conclusions and indicates that radiosensitivity depends, i.a. on IR dose, stage of development of oocytes, the applied marker of the effects of IR, or on the species. LD50 of human oocytes is estimated to be below 2 Gy. The effect of IR depends, i.a. on the dose fractionation and the age (older women are more radiosensitive). In females, the effective sterilizing dose is: at birth 20.3 Gy, at 10 years 18.4 Gy, at 20 years 16.5 Gy, whereas at 30 years 14.3 Gy, which is associated with the available pool of ovarian follicles. Conclusions. Within the range of low doses received as a result of environmental exposure to IR, there is no evidence for the occurrence of either adverse pregnancy outcomes, nor fertility disorders in females. These effects may be related to the cancer radiotherapy, or exposure to high IR doses during nuclear accidents.

The paper is composed of two parts : a review of a group of experiments among irradiated plants and own search for radiation hormesis in a single experiment. In the first part the Bayesian analysis of the hormetic-like data published so far shows that the NOAEL (no observed adverse effect level) point, above which adverse effects appear, may be located between 30 and 100 Gy. In the second part the influence of low doses of ionizing radiation was tested on the particularly fast growing plant, namely the cress (Lepidium sativum L.). Two experimental scenarios were used : in the first one the cress was irradiated during the growth (maximal dose 2.3 Gy), while in the second scenario dry seeds were irradiated (maximal dose 100 Gy). The experiment indicates that the NOAEL point lies above 100 Gy and statistically insignificant hormetic effect can be seen between 0.1 and 14 Gy. No linear reaction is observed in the full range of doses.

Opis:
A report is presented on the advantages of the rapid interphase chromosome assay (RICA) and the difficulties that may be met while implementing this method for application in biological dosimetry. The RICA test can be applied on unstimulated human lymphocytes; this is an advantage in comparison with the dicentric chromosomes or micronucleus tests. In the former two tests, stimulated lymphocytes are examined and hence, 48 h more are needed to obtain cells traversing the cell cycle. Due to the use of unstimulated nondividing cells, higher numbers of cells are available for RICA analysis than for dicentric chromosomes or micronuclei tests. Moreover, the method can be applied after exposure to ionizing radiation doses in excess of 5 Gy. Such doses cause a signifi cant cell cycle delay or result in the loss of G2 phase and mitotic cells because of apoptosis. Therefore, the traditional biodosimetry based on the evaluation of the incidence of damage to chromosomes is very difficult to carry out. This is due to the lack of an adequate number of mitotic cells for analysis. RICA is free of this disadvantage. An automatic microscope can be used to retrieve cell images; automatic image analysis can also be used.

In this study, factors that limit the utilization of electron beam irradiation for decomposition of pollutants were investigated. Potassium hydrogen phthalate (KHP) was used as the model pollutant compound. The accumulated chemical oxygen demand (COD) removal below 2.4 kGy (applied for concentration at 85, 170, 255, 425 and 850 mg/l) was less than 10 per cent, whereas the accumulated COD removal at 215 kGy (applied for concentration at 85, 170, 255, 425 and 850 mg/l) was 198 per cent. At a lower concentration of KHP, 85 mg/l, the accumulated COD removal (for dose ranging from 0.5 to 215 kGy) was 236per cent. As the concentration of KHP increases to 850 mg/l, almost no COD removal was recorded (for dose ranging from 0.5 to 215 kGy). The results show that the removal efficiency and effectiveness of pollutants were influenced by the solute concentration and irradiation dose. When the concentration of KHP increases to a certain level, water radical species were not sufficient to form KHP radicals and, therefore, reduce the decomposition of KHP molecules. Removal of phthalate as measured by COD was restricted at a very high concentration, 850 mg/l. This indicates that the by-products have scavenged the radicals rapidly. This also suggests that, the limitation of radiolytic products of water to decompose KHP at higher concentration. A large quantity of KHP molecules would also stabilize the KHP radicals and resulting in lower removal of KHP. Similar removal trend was also observed for the actual industrial wastewater.

Inverted liquid sugar is a mixture of sucrose, glucose and fructose, which shows its relevant characteristic on high sweetness power. Ionizing radiation has been applied to different kind of food and ingredients for different reasons, such as pathogens reduction, disinfestations, quarantine purposes, ripening delay among others. Radiation from an electron beam can be utilized as a technique to treat this ingredient because it can process a great volume of material per unit of time. The main goal of this paper was to verify the effect of radiation on the properties of inverted liquid sugar. This ingredient was irradiated in an electron accelerator (Radiation Dynamics) at a dose ranging from 5 to 50 kGy. Sucrose content measurements were reduced by 23% at 30 kGy when compared to control and the reduced sugar content increased around 11%. Density and moisture values were not affected by radiation. The total soluble solids (Brix degrees) rose in function of the absorbed dose.

Exosomes are membrane vesicles of endocytic origin that participate in inter-cellular communication. Environmental and physiological conditions affect composition of secreted exosomes, their abundance and potential influence on recipient cells. Here, we analyzed protein component of exosomes released in vitro from cells exposed to ionizing radiation (2Gy dose) and compared their content with composition of exosomes released from control not irradiated cells. Exosomes secreted from FaDu cells originating from human squamous head and neck cell carcinoma were analyzed using LC-MS/MS approach. We have found that exposure to ionizing radiation resulted in gross changes in exosomal cargo. There were 217 proteins identified in exosomes from control cells and 384 proteins identified in exosomes from irradiated cells, including 148 "common" proteins, 236 proteins detected specifically after irradiation and 69 proteins not detected after irradiation. Among proteins specifically overrepresented in exosomes from irradiated cells were those involved in transcription, translation, protein turnover, cell division and cell signaling. This indicated that exosomal cargo reflected radiation-induced changes in cellular processes like transient suppression of transcription and translation or stress-induced signaling.

Introduction. Computed tomography (CT) is inexpensive radiological examination of high diagnostic effectiveness. However, common use of CT tests has led to increased exposure to ionizing radiation in the population. Aim. To assess the effect of organ dose modulation (ODM) technique on the radiation dose received by patients during head CT scan. Material and methods. A retrospective analysis of 120 tests in two groups of patients who had CT scans without and with ODM. Every group consisted of 60 people (30 women and 30 men in each). The ability to perform tests in two algorithms (without and with ODM) resulted from the fact that after installing the apparatus, tests were carried out using standard technique, and only then the ODM function was launched. Results. We found that during examinations with ODM, patients received a reduced dose of ionizing radiation. The mean DLP value with ODM is 9.4% lower than the mean DLP value without ODM by comparing the tests with and without contrasting agent. The mean DLP value obtained in men using ODM was 11.9% lower than the mean dose without ODM, and in women this difference was 6.6%. Conclusion. The mean DLP value received by men with ODM was by 11.9% lower than the mean dose without ODM, and in women this difference was 6.6%. Patients receive a lower dose both in tests with and without a contrasting agent compared to the tests where ODM was not used. A statistical significance of the obtained results was found.

Objectives Geothermal waters may contain soluble, radioactive radon gas. Spa facilities that use geothermal water may be a source of an increased radiation dose to people who stay there. It has been necessary to assess the exposure to radon among people: workers and visitors of spa centers that use geothermal waters. Material and Methods In 2013, workers of the Nofer Institute of Occupational Medicine measured concentrations of radon over the geothermal water surfaces in 9 selected Polish spa centers which use geothermal water for recreational and medicinal purposes. The measurements were performed by active dosimetry using Lucas scintillation cells. Results According to our research, the doses received by the personnel in Polish geothermal spas are < 0.6 mSv/year. In 1 of the investigated spas, the estimated annual dose to the staff may exceed 3 mSv/year. Conclusions In Polish geothermal spas, neither the workers nor the visitors are at risk of receiving doses that exceed the safe limits.

Research done on a set of simple fl uidic (with the fluid used as the ionized medium being air under atmospheric pressure) alphavoltaic cells – small ionizing reactors or “nuclear batteries”, designed in the Faculty of Power and Aerospace Engineering of Warsaw University of Technology, Poland – has shown the possibility of accumulation of usable amount of electric charge. Two simple methods are proposed to describe the fluidic alphavoltaic cells in terms of their efficiency. The results of these methods are presented and compared with the efficiencies of other contemporary types of solid-body (semiconductor junction-based) alpha- and betavoltaic cells. The comparison showed that despite the far-reaching simplicity in design, the designed fl uidic cells are still more efficient than some of the solid-body devices that use the alpha type of decay.

Opis:
Radon in the environment of early Earth was present in sites, determined by location of deposits of uranium, in very different geological formations. According to the decay of uranium-238, the total production of radon at the beginnings was twice as high as now and was continuously diminishing to the present levels. This nuclide could not play as big a role as do radioactive elements connected with the presence of high concentration of 235U, which was high enough in some places to give rise to formation of natural nuclear fission reactors (e.g. Oklo phenomenon in Africa). The main role of ionizing radiation in prebiotic chemistry and biological evolution was played by low LET (linear energy transfer) radiations, as deep penetrating sources of external energy. High LET radiations are of low penetration and could act only superficially. Radon is an exception, due to its easy transfer in the air. Therefore, it could play a role already in the cases of these early organisms which exhibited the gaseous exchange of chemical compounds with the surrounding atmosphere. The action was destructive, but, on the other hand, was also mutagenic. Nevertheless, the general quantitative effect of radon on the global scale could not be larger than of other radioactive nuclides. Presented considerations are part of the chapter by the present author on the role of nuclear and radiation chemistry in astrobiology, in the monograph published by American Scientific Publishers [1].

Freshly isolated PBMCs in peripheral blood are predominantly quiescent, in G0 phase, and with very low amounts of proteins p53 and p21. Using confocal microscopy we detected dose dependent (0.5-5 Gy) induction of foci containing γH2A.X (1 h after γ-ray exposure), which are formed around radiation-induced double strand breaks of DNA. Apoptosis was detected from 24 h after irradiation by the dose of 4 Gy onwards by Annexin V binding and lamin B cleavage. Seventy two hours after irradiation 70% of CD3+ lymphocytes were A+. Neither increase in p53 nor its phosphorylation on serine-392 after irradiation was detected in these cells. However, massive increase in p21 (cyclin-dependent kinase inhibitor 1A) was detected after irradiation, which can be responsible for late occurrence of apoptosis in these quiescent cells. PHA-stimulation itself (72 h) caused an increase in early apoptosis (A+PI-) in comparison to non-stimulated PBMCs (38% A+ resp. 13.4%). After PHA-stimulation also the amount of γH2A.X, p53, and p21 increased, but no phosphorylation of p53 on serine-392 or -15 was detected. Reaction to γ-radiation was different in PHA-stimulated lymphocytes: the p53 pathway was activated and p53 was phosphorylated on serines-15 and -392 4 h after irradiation by the dose of 4 Gy. Phosphorylation of p53 at serine-15 increased in a dose-dependent manner in the studied dose range 0.2-7.5 Gy. Also the amount of p21 increased after irradiation. Seventy two hours after irradiation of PHA-stimulated CD3+ T lymphocytes by the dose of 4 Gy 65% of cells were A+.

Background: In this work we studied the relationship between the enhanced expression of DR5 receptor and the effect of combination of TRAIL and ionizing radiation on cell cycle arrest and apoptosis induction in human leukemia cell line HL-60. Material and methods: DR5, APO2.7 and cell cycle were analyzed by flow cytometry. Proteins Bid and Mcl-1 were analyzed by Western-blotting. For clonogenic survival, colony assay on methylcellulose was used. Results: Ionizing radiation caused significantly enhanced positivity of DR5 receptors 24 h after irradiation with high doses (6 and 8 Gy). An increase of DR5 receptor positivity after a dose of 2 Gy was not statistically significant and application of TRAIL 48 h after irradiation did not increase the apoptosis induction. However, a decrease of radiation-induced G2 phase arrest and an increase of apoptosis were observed when TRAIL was applied 16 h before irradiation with the dose of 2 Gy. Incubation with 6 µg/l TRAIL for 16 h reduced D0 value from 2.9 Gy to 1.5 Gy. The induction of apoptosis by TRAIL was accompanied by Bid cleavage and a decrease of antiapoptotic Mcl-1 16 h after incubation with TRAIL. Conclusion: TRAIL in concentration of 6 µg/l applied 16 h before irradiation by the dose of 1.5 Gy caused the death of 63% of clonogenic tumor cells, similarly as the dose of 2.9 Gy alone, which is in good correlation with the enhanced apoptosis induction.

Opis:
Equipment being used in medical or industrial institutions is often a source of ionizing radiation with different energies and types, which complicates the detection and assessment of doses. Up until now, for dosimetric measurements of ionizing radiation, Gamma-31 dosimeters have been used in the Central Mining Institute for many years. Now, this system will be expanded by a Panasonic system, for which measurement procedures were developed and comparisons with other dosimeters were held. The method is based on a four-element dosimeters UD-802 Panasonic equipped with CaSO and LiBO detectors additionally sheltered by filters of different surface mass. The use of UD-802 dosimeters, in contrast to Gamma-31 dosimeters, permits measuring radiation doses in a different range of photon energy. Consequently, it is possible to obtain a more accurate analysis of the hazards caused by gamma radiation in underground mines. The publication includes a description of the dosimetry system and presents the results of measurements conducted by means of both types of dosimeters. In order to verify the correctness of the indications of the new dosimetry system a series of measurements were carried out, which allowed examining the behaviour of the dosimeters under different environmental conditions. As a place of exposure, the selected laboratories in the Silesian Centre for Environmental Radiometry were chosen, where the work is connected with (TE)NORM and equipment producing ionizing radiation or containing sources of this type of radiation. Moreover, to observe the dosimeters behaviour in difficult environmental conditions, they were exposed in water treatment plants and an underground potassium salt mine.

Opis:
Objectives Ionizing radiation was known to cause disruption of cytoskeleton. However, the disorganization of the cytoskeleton leads to form microparticles (MP) that carry membrane and cytoplasmic constituents from their parent cells they are released from. Therefore, authors investigated the effect of the occupational exposure to low doses of ionizing radiation on MP levels. Material and Methods The current study was conducted on 38 healthy medical workers occupationally exposed to low doses of ionizing radiation and 29 controls matched by gender, age, and smoking habits. The MP levels measured by flow cytometry were classified as positive or negative phosphatidylserine (PS⁺ or PS⁻), and phenotyped according to their cellular origin. Results Total MP (PS–/PS+) levels, regardless of phenotype, were significantly higher in workers occupationally exposed to ionizing radiation than in healthy individuals (p = 0.0004). Negative phosphatidylserine microparticles were predominant in medical exposed workers and, to a lesser extent, in controls (68% and 57%, respectively). With regard to phenotypic characterization of cellular origin, MP derived from platelets (CD41a+), endothelial (CD146+), leucocytes (CD45+) and erythrocytes (CD235a+) MP were significantly enhanced in exposed workers compared with controls (p < 0.0001). However, no significant difference was found in the proportion of the other blood elements in the peripheral circulation between the 2 groups. Serum levels of C-reactive protein were normal for all individuals. In addition, no association was observed between MP levels and the studied confounding factors. Conclusions The results suggest that elevated circulating MP levels represent an indicator of cellular damage caused by medical exposure to low doses of ionizing radiation. By consequence, the quantification of MP seems to be a useful biomarker for assessing the negative effects of occupational exposure to ionizing radiation. Introduction and objective. The male reproductive system constitutes a set of tissues which are particularly sensitive to external factors. The aim of the study was to analyze the relationship between background radioactivity and the quality of the semen of the men who have reported to the infertility treatment facility in 2000–2016 in the Lublin region of eastern Poland. Materials and method. The radioactivity of the ground-level air layer obtained from the Institute of Meteorology and Water Management in Włodawa in the Lublin region was analysed. 4,250 spermiograms of patients who reported to the treatment facility for the first time due to infertility were analysed and correlated to background radioactivity in the Lublin region in 2000–2016. Results. A long-term decrease in the percentage of morphologically normal spermatozoa of the patients was observed (r=-0.970; p<0.001 in 2000–2009 and r=-0.925; p=0.003 in 2010–2016). Men’s age correlated negatively with sperm motility (r=-0.164; p=0.009) and morphology (r=-0.186; p=0.009). The percentage of spermatozoa with normal morphology was lowered by beta-isotopes of artificial origin in the air in 2000–2009 (r=-0.655; p=0.040) and by the exposure to gamma radiation of the ground-level atmosphere in 2010–2016 (r=-0.676; p=0.048). The percentage of sperm vitality was lowered by gamma radiation in the atmosphere (r=-0.636; p=0.006), but improved by beta isotopes in precipitation (r=0.686; p=0.002) in the whole of the analyzed period. Conclusions. The percentage of morphologically normal spermatozoa in patients who reported to the infertility treatment depends not only on the age of patient, but also on beta-isotopes of artificial origin and gamma radiation in the atmosphere. Beta isotopes in precipitation affect the improvement of sperm vitality.

Rapid retrospective biological dosimetry allows absorbed dose evaluation post exposure to ionizing radiation. One of the main tools of biodosimetry is based on the analysis of the effects resulting from the impact of ionizing radiation on the cell. Various cytogenetic tests give possibility of the accurate dose estimation. To investigate cell response to radiation one performs the analysis of biomarkers approved by International Atomic Energy Agency e.g. the analysis of dicentric chromosomes or micronuclei frequency. Micronucleus test is relatively a faster and therefore more effective method to study changes in the genetic material, induced by various genotoxic agents. This study confirms that micronulei frequency and nuclear division index analysis allows for appropriate absorbed dose estimation when it comes to ionizing radiation. In order to further optimize and facilitate the micronucleus assay and other cytogenetic tests in rapid retrospective biological dosimetry, the research are still ongoing.

The aim of this study was to assess the effect of low-radiation exposure on detachment of the vascular endothelial cells. Circulating endothelial cells (CEC) were measured in the peripheral blood by an immunomagnetic separation (IMS) technique in 63 workers occupationally exposed to low-level ionizing radiation at the Atomic Energy Commission of Syria and in 28 controls. We found a significant difference in CEC numbers in the peripheral blood of the workers exposed to low-level ionizing radiation in comparison with those of the control samples (p = 0.0001) and no significant difference in the proportion of other blood elements in the peripheral circulation. Elevated number of CEC in occupational workers exposed to low doses of ionizing radiation may reflect an early affection of the endothelium. Long-term follow-up of these workers must be conducted to determine the clinical value of CEC as an early indicator of cardiovascular disease induced as a result of chronic occupational exposure to low doses of ionizing radiation.

Ionizing radiation is a factor which severely allows to characterize the human environment and hygienic conditions -health, prevailing in buildings intended for human residence. For organisms play two important factors: - background radiation affecting living organisms - alpha radiation decay products of radon affecting the respiratory system. The presence of radon and its derivatives in the areas of residential buildings s may have come primarily from the soil of raw materials and construction materials. If the radiation standards are not exceeded, there is no fear of the harmful effects of radioactive elements. More than that, it is believed that the total absence of radiation is disadvantageous because it can cause an avalanche microbial growth.

Opis:
ATM kinase (ATM) is essential for activation of cell cycle check points and DNA repair in response to ionizing radiation (IR). In this work we studied the molecular mechanisms regulating DNA repair and cell death in human T-lymphocyte leukemic cells, MOLT-4. Apoptosis was evaluated by flow-cytometric detection of annexin V. Early apoptotic cells were determined as sub-G1 cells and late apoptotic cells were determined as APO2.7-positive ones. Proteins involved in ATM signalling pathway were analysed by Western-blotting. We observed a rapid (0.5 h) phosphorylation of ATM declining after 6 h after irradiation by all the doses studied (1.5, 3.0, and 7.5 Gy). Checkpoint kinase-2 (Chk-2) was also phosphorylated after 0.5 h but its phosphorylated form persisted 4, 2, and 1 h after the doses of 1.5, 3.0, and 7.5 Gy, respectively. The amount of p53 protein and its form phosphorylated on Ser-392 increased 1 h after irradiation (1-10 Gy). The lethal dose of 7.5 Gy caused an immediate induction and phosphorylation of p53 after 0.5 h post-irradiation. At the time of phosphorylation of p53, we found simultaneous phosphorylation of the oncoprotein Mdm2 on Ser-166. Neither ATM nor its downstream targets showed a dose-dependent response after 1 h when irradiated by the doses of 1-10 Gy. MOLT-4 cells were very sensitive to the effect of IR. Even low doses, such as 1.5 Gy, induced apoptosis 16 h after irradiation (evaluated according to the cleavage of nuclear lamin B to a 48-kDa fragment). IR-induced molecular signalling after exposure to all the tested doses was triggered by rapid phosphorylation of ATM and Chk-2. Subsequent induction of p53 protein and its phosphorylation was accompanied by concomitant phosphorylation of its negative regulator, oncoprotein Mdm2, and followed by induction of apoptosis.

When analyzing time series we can often observe that they have periodic oscillations (cyclic, seasonal). One of the methods which can be applied to investigate such a phenomenon and which belongs to the group of periodic components models is the method of harmonic analysis. The method consists in analyzing the oscillations around the medium level a0 and building a model as the sum of so called harmonics. The paper presents the applicability potential of harmonic analysis in the forecasting process of radiation hazard on post-mining grounds (mine waters with raised content of radium Ra226 and Ra-228 drained off to the surface), using for that purpose the measurement results of radium concentration from the area of mine water catchment from the years 2004-2013.
