This research was conducted with the aim to analyze the presence of potential pathogenic bacteria in carp that are sold live in supermarkets in the city of Cimahi and to find out the species of these bacteria. Fish samples were obtained from two supermarkets in Cimahi City, Transmart and Superindo. From each supermarket three fish samples were taken once a week and repeated three times. Bacteria were isolated from several parts of the fish body namely body surface mucus, gills, liver, and kidneys. The results of isolation from each target organ were biochemically tested to determine the species of bacteria. Potential pathogenic bacteria found in carp from this research are Aeromonas hydrophila, Aeromonas sobria, Aeromonas schubertii, Aeromonas media, Aeromonas caviae, Aeromonas ecrenophila, Pseudomonas stutzeri, Pseudomonas pseudoalcaligenes, Pseudomonas aeruginosa, Aeromonas caviae, Aeromonas ecrenophila, Pseudomonas stutzeri, Pseudomonas pseudoalcaligenes, Pseudomonas aeruginosa, Plesiomonas shigelloides, Bacillus megaterium, Bacillus thuringiensis, Neisseria mucosa, Citrobacter freundii, Serratia marcescens, Staphylococcus epidermidis, Micrococcus luteus, Enterobacter aerogenes, and Enterobacter sakazakii.

Feed greatly affects the growth and health of fish, however it also functions to increase the value of the color intensity when specific additional ingredients are added. One of the important ingredients, marigold-meal is one of the natural carotenoid sources. This research aims to determine the optimum concentration of marigold-meal in the artificial feed to increase the intensity color of koi fish, specifically the kohaku strain. The method used was an experimental Completely Randomized Design, consisting of five treatments and four replications. Parameters observed were color intensities analyzed using Kruskal-Wallis analysis, if there were significant differences, Z test would be performed. Research results were the color intensity enhancement of koi fish tails and head until the 40th day showed the highest value in the treatment concentration of 0.0% marigold-meal (positive control) with an average score of 8.00 and 10.00. This value was not different from the treatment concentration of 2.0% marigold-meal and concentration 1.5% marigold-meal. However, this value was not significantly different from concentration of 1.5% marigold-meal. The lowest average score on the tail and head was treatment concentration of 0.0% marigold-meal (negative control). The optimum concentration of the addition of marigold-meal resulting the best color intensity increase was in 1.5% concentration of marigold-meal with the color intensity value of 5.97 in the tail and 6.50 in the head of koi fish Kohaku strain.

This study aims to determine the method of generating water flow inside a round container to increase the survival and growth of common carp fingerlings. The approach used in this research is an experimental method which applies a Completely Randomized Design (CRD) and consists of three and five replications. The treatments used are (A) No Water flow (control), (B) 0.1 m/s of water flow, (C) Water flow utilizing an additional venturi system. The test fish used were 450 majalaya common carp of size 3-5 cm. The containers used in this research are in the form of 15 gallons capacity with a diameter of 27 cm x 36 cm height x bottom diameter of 6 cm containing 19 liters of water. The density of common carp during the study was 30 carp per treatment container. The treatment period is 40 days. The food given is 5% of the bodyweight of fish. Water quality parameters (temperature, pH, dissolved oxygen, ammonia and nitrates) were observed every 10 days for 40 days. Other parameters are survival, daily growth rate, feed conversion ratio, and feed efficiency observed every 10 days for 40 days. The results showed that the use of water flows with the venturi system provides the best results - with a life growth rate of 1.5%, a feed conversion ratio of 3.18, feed efficiency of 31.73% and 75% survival.

Dead fish due to reversal of water mass in floating net cages cultivation, most of which cannot be sold because they are not suitable for consumption or processing into food. One effort that can be done to increase the added value of dead fish is processed into silage. Silage is a very good feed ingredient for fish and poultry because of its low metabolic basal energy. This study aims to determine the effect of increasing the scale of production on the characteristics of fish silage quality. The research method used was experimental with a complete randomized design consisting of 3 treatments namely a) scale of production of 1 kg of fish, b) scale of production of 5 kg of fish, and c) scale of production of 25 kg of fish. The three treatments were repeated 3 times. The parameters observed were protein content, fat, ash content, water content and amino acid composition of silage fluid and rendemen. Data obtained from all observation variables except amino acid compositions were analyzed by F test, if significant continued with Duncan test, each test was carried out at 95% confidence level. The results of the study obtained conclusions that the quality characteristics of fish silage made from dead fish due to reversal of water mass were not affected by the scale of the production process. The silage quality characteristics produced in this study were protein content of 16.02%, fat content of 8.98%, ash content of 2.35%, carbohydrate content of 2.45% and moisture content of 70.2%. Rendemen is 62.72%.

Variability of stress proteins concentration in caged carp exposed to transplantation experiment model dam reservoir was caused only by natural (climatic and biological) conditions. Thus, the reference data of stress proteins concentration range in young carp individuals were obtained. Metallothionein, HSP70 and HSP90 protein concentrations as biomarkers were assayed in the livers, gills and muscles of six-month-old (summer) or nine-month-old (autumn) carp individuals in relation to the site of encaging, season (summer or autumn), the term of sampling (1, 2 or 3 weeks after the transplantation) and tissue. Physicochemical analyses of the condition of water as well as pollution detection were conducted during each stage of the experiment. As the result of this study, the range of the variability of the stress protein concentration in young carp individuals was obtained. According to the analyses of the aquatic conditions of a reservoir with no detectable pollutants, we conclude that the variability in the stress protein concentration levels in the groups that were compared is solely the result of the natural conditions. Future regular monitoring of the reservoir using the transplantation method and young carp individuals will be both possible and reliable. Moreover, the range of variability in the stress protein concentrations that were measured in the young C. carpio individuals acquired from the model dam reservoir in relation to all of the studied factors may be applied in the monitoring of any other similar reservoir.

This study aims to determine the best density of Bacillus sp. which is given through feed to increase the body resistance of carp that is infected by pathogenic bacteria, Aeromonas hydrophila. The research was conducted at the Aquaculture Laboratory and Laboratory of Microbiology and Biotechnology at FPIK UNPAD. This study used an experimental method with a completely randomized design consisting of 4 treatments and 3 replications, namely treatment A (control), treatment B (Bacillus sp. 106 cfu/mL feed), treatment C (Bacillus sp. 108 cfu/mL feed) and treatment D (Bacillus sp. 1010 cfu/mL feed) for two weeks later in Aeromonas hydrophila infection with an intramuscular density of 108 cfu/mL for seven days. Observation parameters were clinical symptoms, fish response to feed, fish response to shock and survival. The results of this study showed that treatment C (Bacillus sp. 108 cfu/mL feed) resulted in 89% survival and reduced clinical symptoms.

The aim of the study was to assess the dynamics of cadmium and nickel accumulation in the carp, Cyprinius carpio L., during the rapid growth period between 3rd and 6th month of life, including seasonal changes. Chemical assays were performed on samples of gills, anterior and mid-posterior part of the alimentary tract, liver, kidneys, skin and muscles. Prior to the assay, 1-g tissue samples were digested with 3 ml of concentrated HNO3 in a CEM MDS 2000 microwave oven. The samples prepared this way were assayed for Cd and Ni. Cadmium level was determined with flameless graphite furnace atomic absorption spectrometry (GF-AAS) in a ZL 4110 Perkin Elmer. Nickel was determined inductively coupling plasma atomic emission spectrometry (ICP-AES) in a JY-24 Jobin Yvon apparatus. The results indicated that in the period of growth between 3rd and 6th month of life, cadmium and nickel concentrations in the examined carps fluctuated. Average cadmium content in examined organs ranged between 0.053 ÷ 0.004 µg g-1 wet weight. Average nickel content in examined organs ranged between 0.326 ÷ 0.023 µg g-1 wet weight. The observed fluctuations are supposed to be a result of intensive growth and accompanying accumulation of metals from water.
Oceniano dynamikę kumulacji kadmu i niklu w różnych narządach i tkankach karpi (Cyprinus carpio L.) w okresie intensywnego ich wzrostu, czyli między 3. a 6. miesiącem życia. Badania przeprowadzono na 240 karpiach hodowanych w wodach pochłodniczych. Z każdej ryby do analiz chemicznych pobierano próbki: nerek, wątroby, przewodu pokarmowego, listków skrzelowych, mięśni grzbietowych i skóry. Próbki narządów o masie 1 g zmineralizowano na mokro, w obecności 3 cm3 HNO3, w piecu mikrofalowym CEM MDS 2000. Kadm oznaczono metodą bezpłomieniowej absorpcyjnej spektrometrii atomowej GF-AAS, nikiel z użyciem emisyjnej spektrometrii atomowej w plazmie indukcyjnie sprzężonej (ICP-AES). Stwierdzono, że u przebadanych karpi między 3. a 6. miesiącem życia wystąpily wahania zawartości Cd i Ni. Średnie zawartości kadmu w badanych narządach wynosiły 0.053 ÷ 0.004 µg g-1 mokrej masy, natomiast średnie zawartości niklu - 0.326 ÷ 0.023 µg g-1 mokrej masy. Zaobserwowane niewielkie zmiany poziomu kadmu i niklu są wynikiem intensywnego wzrostu karpi i towarzyszącej temu kumulacji metali ze środowiska wodnego.

This research aims to determine the best currents speed and utilization of venturi to increasing survival, growth of Cyprinus carpio L. and reducing the infection rate of A. hydrophila bacteria. fingerlings. This research used an experimental method completely randomized design (CRD) with four treatments and four replications. The treatments used were the use of currents and venturi on the maintenance media with currents velocities of 0 m/s (A), 0.06 m/s (B), 0.1 m/s (C), and 0.14 m/s (D). Maintenance of the test fish for 40 days then followed by a challenge test for 14 days. Koi fish fingerlings tested to use A. hydrophila with a density of 108 CF /ml. The parameters observed included white blood cells, red blood cells, clinical symptoms, survival, daily growth rate, feed conversion ratio and water quality. The results showed that the use of currents and venturi with a flow rate of 0.14 m/s was effective in improving the performance of the koi fish's immune system and reducing the infection rate of A. hydrophila bacteria. During the flow administration during the 40 days maintenance period, the white blood cell count increased by 20.51% from a value of 9.19 × 104 cells/mm3 to 11.56 × 104 cells/mm3. Red blood cells increased by 40.65% from 1.60 × 106 cells/mm3 to 2.69 × 106 cells/mm3. Currents treatment with a speed of 0.14 m/s produced the highest survival rate after challenged with a value of 86.7%. Also this treatment was effective in preventing infection of A. hydrophila bacteria, it can be seen from the macroscopic clinical symptoms that appear less than the treatment current. The best results of daily growth rate and feed conversion ratio provided in currents treatment with a speed 0.1 m/s.

This study aims to see the condition of hematocrit levels and differential leukosit after immersion in LAB isolated and after in vivo, using Aeromonas hydrophila bacterium. LAB isolated were obtained from the organs of the common carp intestine. The study used a Completely Randomized Design (RAL) consisting of four treatments and three replications. The treatment used was immersion of test fish for 24 hours in several isolates of LAB with a density of 108, namely A: without LAB isolate (control), B: isolate CcB7, C: isolate CcB8, D: isolate CcB15. Immersion was done three times, with a frequency of seven days. Parameters observed included hematocrit levels, leukocyte differentiation and survival rate. The results showed that immersion in LAB isolate CcB15 will enhance the differential leukocytes and hematocrit levels that serve as indicators of carp body resistance, which is shown by high levels of white and red blood cells.

Cyanobacteria (Cyanophyta, Cyanoprocaryota, Cyanobacteria) (blue-green algae) are procaryotic phototrophic microorganisms playing an important ecological role in the freshwater and marine environment as primary producers. However, as a consequence of water eutrophication observed in many reservoirs in different parts of the world, these microorganisms form massive scums, known as water blooms, releasing cyanotoxins hazardous to fish and other aquatic organisms. Cyanotoxins are cyanobacterial secondary metabolites of various chemical structures harmful to humans, terrestial and aquatic animals such as fish. The most abundant cyanotoxins are microcystins and hepatotoxins inducing toxic changes in fish liver, kidney, gills, digestive tract and immune system. Very little is known on the effects of alkaloid neurotoxic anatoxin-a on fish and their immunity. The aim of this study was to assess the in vitro influence of anatoxin-a on immune cells isolated from the common carp (Cyprinus carpio L.). The leukocyte intracellular level of ATP was reduced only at the highest concentration of anatoxin-a. Apoptotic and necrotic leukocytes were observed at the lower and the highest concentrations of anatoxin-a, respectively. Elevated activity of caspases 3/7 after 2 hours and a concentration-dependent decrease in the proliferative ability of T and B lymphocytes was also observed. The results suggest that anatoxin-a could be a possible immunotoxic agent in the aquatic environment and may increase the susceptibility of fish to infectious and neoplastic diseases. Therefore, constant monitoring of anatoxin-a and its producers in lakes and fish ponds should be performed.

The research was conducted at the Aquaculture Laboratory of the Faculty of Fisheries and Marine Science, Universitas Padjadjaran, Jatinangor. The purpose of this research is to determine the optimum temperature for the maintenance of Common carp strain Majalaya fingerlings based on resistance to temperature stress that is known through their blood profile and survival rate. Common carp strain Majalaya fingerlings originating from Ciparay, West Java. Total body length is 8-12 cm with a minimum weight of 10 g. The method used is experimental with a completely randomized design (CRD) consisting of three treatments and five replications. The fish are kept at 24 ºC (control without heater), 29 ºC and 34 ºC. Each experimental unit contains 10 fishes. The results obtained by the survival rate (SR) of Common carp strain Majalaya which were maintained at a treatment temperature of 24 ºC by 100%, the 29 ºC by 70%, the 34 ºC by only 42% remained on the 21st day. The number of erythrocytes of common carp strain Majalaya fingerlings before the treatment temperature was 2.77 × 106 cells/mm3. The average number of erythrocytes of the 24ºC treatment from 7th, 14th and 21st day were 2.82, 2.82 and 2.81 × 106 cells/mm3; the 29 ºC were 2.93, 2.68 and 2.42 × 106 cells/mm3; the 34 ºC were 3.08, 2.52 and 1.99 × 106 cells/mm3. While the number of leukocytes of common carp strain Majalaya fingerlings before the treatment temperature was 13.61 × 104 cells/mm3. The average number of leukocytes of the 24 ºC treatment from 7th, 14th and 21st day were 13.73, 13.71 and 13.88 × 104 cells/mm3; the 29 ºC were 18.40, 16.46 and 15.76 × 104 cells/mm3; the 34 ºC were 20.78, 18.46 and 16.81 × 104 cells/mm3.

The paper evaluates the effect of culture conditions and culture site on levels of certain microelements (Zn, Cu, Fe) and macroelements (Mg, Ca) in three species of freshwater fish: rainbow trout (Oncorhynchus mykiss Walbaum), common carp (Cyprinus carpio L.), and Siberian sturgeon (Acipenser baeri Brandt). The study involved 90 individuals of freshwater fish aged from 6 to 12 months. Samples of blood, liver, kidney, gills, skin and muscles were collected from each fish and subjected to chemical assay of Mg, Ca, Zn, Cu, Fe with inductively coupled plasma-atomic emission spectrometry in a JY-24 Jobin Yvon apparatus. The study revealed that culture site had statistically significant impact on levels of the examined elements among the three fish species. Mg content in kidney and skin was significantly higher in carp than in sturgeon. Similar regularities were observed for Ca content in skin and Zn content in kidney. Liver and kidney levels of Fe and Cu were significantly lower in carp than in rainbow trout. Of all the three examined fish species, rainbow trout had the highest skin levels of Ca and Mg, and the highest blood level of Fe. The results indicate that culture site and culture conditions exerted significant influence on levels of macro- and microelements in freshwater fish.
Celem pracy była ocena wpływu warunków i miejsca prowadzenia hodowli na zawartość mikroelementów (Zn, Cu, Fe) i makroelementów (Mg, Ca) w organizmach ryb słodkowodnych. Ocenie poddano trzy wybrane gatunki ryb: pstrąga tęczowego (Oncorhynchus mykiss Walbaum), karpia (Cyprinus carpio L.) i jesiotra syberyjskiego (Acipenser baeri Brandt). Badaniu poddano 90 ryb słodkowodnych w okresie od 6. do 12. miesiąca życia. Z każdej ryby do analiz chemicznych pobrano próbki: krwi, wątroby, nerek, skrzeli, skóry i mięśni. Próbki poddano analizie na zawartość Mg, Ca, Zn, Cu, Fe, z użyciem emisyjnej spektrometrii atomowej w plazmie indukcyjnie sprzężonej (ICP-AES), w aparacie Jobin Yvon typ JY-24. Wykazano, że miejsce hodowli ma statystycznie istotny wpływ na zawartości badanych pierwiastków u poszczególnych gatunków ryb słodkowodnych. Zawartość Mg w nerce i skórze karpi była istotnie wyższa niż u jesiotra. Podobne spostrzeżenia dotyczyły poziomu Ca w skórze i Zn w nerce. W przypadku Fe i Cu obserwowano niższą zawartość w wątrobie i nerkach karpi niż w tych samych narządach u pstrąga. Badania wykazały, że poziom Zn i Cu we wszystkich narządach przebadanych gatunków kształtował się na najniższym poziomie. Spośród badanych gatunków u pstrąga stwierdzono najwyższą zawartość Ca i Mg w skórze, a Fe we krwi. Otrzymane wyniki pozwalają stwierdził, że miejsce i warunki prowadzenia hodowli mają istotny wpływ na zawartość badanych makro- i mikroelementów w organizmach ryb słodkowodnych.

The aim of the study was to estimate the effect of culture conditions and culture site on magnesium (Mg) and zinc (Zn) concentrations in freshwater fish. The study encompassed dorsal muscles in five fish species: common carp (Cyprinus carpio L.), rainbow trout (Oncorhynchus mykiss Walbaum) and Siberian sturgeon (Acipenser baeri Brandt), northern pike (Esox lucius L.) and grass carp (Ctenopharyngodon idella Valenciennes). A total of 125 fish comprised 25 individuals of each species, aged from 6, 9, and 12 months. The fish were cultured in privately owned fish breeding ponds (Western Pomerania, Poland). For chemical and biochemical assays, samples of dorsal muscles were taken from each fish. Tissue samples were wet mineralised in concentrated HNO3 in a CEM MDS 2000 microwave oven. Magnesium and zinc concentrations were determined by inductively coupled plasma atomic emission spectrometry (ICP-MS) in a Jobin Yvon type JY-24 apparatus. The pursuit of the research we had an approval of the Polish Local Ethics Committee nr 9/05. The magnesium concentration in the dorsal muscles ranged from 95.3÷347.6 mg kg–1 w.w. The highest Mg concentration was found in rainbow trout (347.6±32.2 mg kg–1 w.w.), and the lowest in grass carp (95.3±11.3 mg kg–1 w.w.). Zinc concentration varied from 6.7÷98.8 mg kg–1 w.w. The highest was found in the muscles of Siberian sturgeon (98.8±0.4 mg kg–1 w.w.), and the lowest in rainbow trout (6.7±0.7 mg kg–1 w.w.). It was found that the breeding place significantly affected the Zn and Mg concentrations in the muscle tissue among individual freshwater fish species. The magnesium and zinc concentrations were also significantly affected by the type of feed.

Ocenie poddano mięśnie grzbietowe pobrane od pięciu wybranych gatunków ryb słodkowodnych: pstrąga tęczowego (Oncorhynchus mykiss Walbaum), karpia (Cyprinus carpio L.), jesiotra syberyjskiego (Acipenser baeri Brandt), szczupaka pospolitego (Esox lucius L.) i amura białego (Ctenopharyngodon idella Valenciennes). Badaniami objęto ogółem 125 ryb słodkowodnych, po 25 osobników z każdego gatunku. Materiał do badań pobrano w 6., 9. i 12. miesiącu życia ryb. Z każdej ryby do analiz chemicznych pobrano próbki mięśni grzbietowych. Pobrany materiał zmineralizowano na mokro w stężonym HNO3 w piecu mikrofalowym CEM MDS 2000. Próbki poddano analizie na zawartość Mg i Zn z użyciem emisyjnej spektrometrii atomowej w plazmie indukcyjnie sprzężonej (ICP-AES) w aparacie Jobin Yvon typu JY-24. Na powyższe badania uzyskano zgodę Lokalnej Komisji Etycznej nr 9/05. Zawartość Mg w mięśniach mieściła się w przedziale 95.3÷347.6 mg kg–1 m.m. Najwyższy poziom Mg odnotowano u pstrąga (347.6±32.2 mg kg–1 m.m.), a najniższy u amura (95.3±11.3 mg kg–1 m.m.). Zawartość Zn mieściła się w przedziale 6.7÷98.8 mg kg–1 m.m. Najwyższy poziom Zn odnotowano w mięśniach jesiotra syberyjskiego (98.8±0.4 mg kg–1 m.m.), a najniższy u pstrąga (6.7±0.7 mg kg–1 m.m.). Wykazano, że miejsce hodowli ma statystycznie istotny wpływ na zawartości magnezu i cynku w tkance mięśniowej poszczególnych gatunków ryb słodkowodnych. Na poziom magnezu i cynku w tkance mięśniowej istotne znaczenie miał również wpływ rodzaj spożywanego pokarmu.
