To develop a sensitive, specific, and rapid approach for the detection Getah virus (GETV), a set of primers targeting the conserved region of the E1 gene was created. The TaqMan-based real-time PCR method for GETV detection was developed by optimizing the reaction conditions. The method demonstrated excellent specificity, and amplification did not occur with the causative agents of all prevalent swine viral infections (CSFV, PRRSV, PRV, PEDV, PTV, and JEV), except GETV. Additionally, upon assessing the sensitivity of the method, the minimum detection limit for GETV was found to be 5.94 copies/μL, which is 10 times higher than that of the traditional PCR approach. Further, the intra- and inter-assay variation coefficients were less than 1%, demonstrating good repeatability. Moreover, GETV was found in 10 of the 20 field serum samples using real-time PCR but only in three of the samples using traditional PCR. Consequently, the first GETV TaqMan-based real-time PCR approach based on the E1 gene was developed for GETV pathogenic diagnoses, and this exhibited high specificity, sensitivity, and repeatability. This assay is practical for the pathogenic diagnosis and epidemiology of GETV.

Chlorocresol nanoemulsion disinfectant (CND) is an environmental disinfectant prepared with nanoemulsion as its drug carrier. This study aimed to investigate the bactericidal effect of CND on Staphylococcus aureus ( S. aureus) and its effect on bacterial ultrastructure. The neutralizing effect of CND against S. aureus was first screened by suspension quantitative evaluation experiment procedure of neutralizer. Disinfection performance was evaluated by the determination of Minimal Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC), quantitative bactericidal experiment, and comparative experiment of disinfection performance between 0.1% CND and 0.1% chlorocresol aqueous solution. Meanwhile, the effect of CND on the ultrastructure of S. aureus was investigated with scanning electron microscope (SEM) and transmission electron microscope (TEM) to preliminarily explore the bactericidal mechanism. The results showed that 3% Tween-80 in PBS could be screened as the neutralizer of CND against S. aureus. MIC and MBC were 100 μg/mL and 200 μg/mL, respectively. The bactericidal rates were all 100% when 0.06% and 0.08% disinfectant acted for 15 and 5 min, respectively. Furthermore, compared with 0.1% chlorocresol aqueous solution, the bactericidal effect of 0.1% CND was significantly enhanced (p<0.01). After treatment with CND for 10 min, SEM observation showed that the morphology of S. aureus cells were changed and the integrity destroyed. TEM observation showed that the cell shape changed, and the structures of the cell wall, cell membrane and cytoplasm were damaged in varying degrees. CND showed the strong bactericidal effect on S. aureus and could cause ultrastructure alterations of S. aureus.

Classical swine fever (CSF) and porcine reproductive and respiratory syndrome (PRRS) are responsible for major economic losses and represent a threat to the swine industry worldwide. Routine surveillance serology for CSF and PRRS viruses is critical to maintaining the health status of sow farms in Hunan Province, which is one of the top pig production provinces in China. The aim of our study was to investigate the serological statistics of CSF virus (CSFV) and PRRS virus (PRRSV) in Hunan Province. The cohort serum samples were collected from vaccinated and unvaccinated pigs. Our findings showed that the average rates of CSFV and PRRSV antibody seropositivity were 82.2% (95% CI: 80.1-84.3) and 84.8% (95% CI: 82.5-87.1), respectively, in the immunized group and that these rates were higher than those in the unvaccinated group (58.6% for CSFV and 47.8% for PRRSV). Additionally, the level of CSFV antibody in piglet serum declined gradually with age, whereas PRRSV-specific antibody level increased initially (1 to 2 weeks old) and then declined with age (2 to 4 weeks old). In summary, we investigated the difference in CSFV/PRRSV antibody levels among piglets at various weeks old (1 to 4 weeks) to further establish the duration of maternal immunity in piglets. In addition, routine monitoring of CSFV/PRRSV antibodies in immunized pigs was carried out to evaluate the efficacy of vaccination.

In view of the current situation that it is difficult to obtain the actual cutting force received by the conical pick of Continuous-Miner and to deal with the characteristics of the cutting force. By using contact dynamics, rock mechanics, and finite element methods, based on the display dynamics analysis program ANSYS/LS-DYNA and the Holmquist–Johnson–Cook model, the contact mechanical characteristics between conical pick and rock are analyzed. The resultant force was obtained by obtaining the force curve of the conical pick under X, Y and Z directions, analysis of cutting gear cutting speed of 2.5 m/s, cutting depth of 5, 10, 15, 20 and 25 mm, cutting angle of 30°, 35°, 40°, 45°, 50° parameters of the size of cutting force and cutting ratio energy consumption, thus, the best cutting angle of the conical pick is obtained. The conclusions are as follows: when the cutting depth and cutting speed are constant, and the cutting angle is 45°, the mean cutting force received by the conical pick is the least, the specific energy consumption is the least, and the cutting efficiency is the highest. The best cutting angle of the conical pick should be 45°; When the cutting angle and cutting speed are constant, the energy needed by the cutter to cut the rock increases with the increase of cutting depth. The research results provide a theoretical basis for improving cutting efficiency and cutting life, and for choosing cutting angle and cutting thickness.

Climate change is causing changes in tree species performance and distribution, impacting breeding programme effectiveness. Our aim was to analyse the effects of provenance and climatic factors on the annual ring density of Masson pine (Pinus massoniana Lamb.) at different experimental sites and potential breeding strategies that may be developed in response to future climate change. The study trees represented provenances originating from the western, east-central, northern, and southern regions of P. massoniana distribution in China. The wood density differed significantly among provenances. A multisite variance analysis test showed that the type B correlation coefficients for ring density at the two sites stud- ied were less than 0.8, indicating an interaction effect of genotype by environment (G×E) on tree ring density. Climatic factors directly affected the wood density properties. At Chun’an (CA), the maximum latewood density (MXD) and minimum earlywood density (MND) were positively correlated with absolute maximum temperatures in August and May of the current growing season, respectively. At Taizi Mountain (TZS), MXD was significantly positively correlated with absolute maximum temperature in September of the current year and significantly negatively correlated with precipitation in June. MND was significantly positively correlated with absolute maximum temperature in May of the current year and significantly negatively correlated with precipitation in April. The climatic effects on P. massoniana wood density differed among seed-source origins. This study showed that ring density characteristics differed significantly among provenances, and provenance selection could promote wood density. MXD and MND exhibited significant genotype-by-environment interaction effects, and significant correlations were found between ring density and temperature and precipitation conditions. These findings suggest that climatic factors and site condi- tions in addition to genetics could be strong drivers of wood density variation, and/or that wood density is a highly plastic trait.

The negative landform of sinkholes provides belowground level refugia for a high diversity of forest species compared to the forests on the surface of surrounding karst plateaus in southeastern China. Bryophyte diversity in sinkhole forests is also likely to be high. In this study, bryophytes of an underground forest sinkhole (UFS), and two forests (Forest Karst Mountain 1; FKM1) and (Forest Karst Mountain 2; FKM2) on the surface of the karst plateau were compared to understand the role of the sinkhole forest in the conservation of bryophyte diversity and the relationships between bryophyte diversity, environmental factors, and soil nutrients. Significantly more bryophyte taxa were recorded from the sinkhole forest (71 taxa, 36 genera, 23 families) than those in the forest on the plateau surface, which was the closest to the sinkhole (FKM1; 29 bryophyte taxa, 16 genera, 12 families), and even fewer bryophytes were found in the forest more distant to the sinkhole (FKM2; 22 taxa, 17 genera, eight families). Twenty-four liverwort taxa were collected from the sinkhole forest, two from the closest surface forest (FKM1) and none from the furthest surface forest (FKM2). Ninety-three percent of the bryophytes in karst mountain sinkhole were not found on surface forest. The diversity index trend was as follows: UFS > FKM1 > FKM2, and the evenness index trend was in the opposite direction as follows: UFS < FKM1 < FKM2. The beta diversity showed that the Jaccard index among the three forests was 0–0.25, reflecting a huge difference between the sinkhole forest and the two surface forests. Canonical correspondence analysis showed that light was the most important factor affecting the distribution of bryophytes in the sinkhole forest, while temperature and humidity were key factors for the distribution of bryophytes in the two surface forests. Further, soil parameters, namely, soil organic carbon, total nitrogen, total phosphorus, total potassium, alkali- hydrolyzed nitrogen, and available phosphorus changed gradually from UFS to FKM1 and to FKM2. The sinkhole forest, located in a landform with the unique negative topography enclosed by cliffs, and with a complex microenvironment, provides a natural refuge for bryophyte species in areas where forests have been negatively impacted by karst rocky desertification.

In this study, cubic and cylindrical cement mortar specimens were first subjected to high temperatures, then the cubic and cylindrical specimens were taken out and conducted with uniaxial compressive test and splitting tensile test, respectively. The effect of the length to side ratio on the uniaxial compressive properties and the effect of thickness-to-diameter ratio on the splitting tensile properties of cement mortar specimens after high temperature were studied. Test results show that: (1) With temperature increasing from 25 ºC (room temperature) to 400 ºC, the compressive strength and elastic modulus of cubic specimens with three kinds of side lengths decrease; the decreasing rates of compressive strength and elastic modulus of cubic specimen with side length of 70.7 mm is higher than those of cubic specimens with side length of 100 mm and 150 mm, and the strain at the peak stress of cubic specimens with three kinds of side lengths increase. (2) After the same temperature, the tensile strength of cylindrical specimen decreases with the thickness-to-diameter ratio increasing from 0.5 to 1.0. The decreasing rate of tensile strength of cylindrical specimen with thickness-to-diameter ratio is highest when the temperature is 25 ºC (room temperature), followed by that after the temperature of 200 ºC, and that after the temperature of 400 ºC is the lowest.

Opis:
Cypress (Cupressus funebris Endl.) is an important tree species in the subtropical regions of China; it is also a major tree species for afforestation and forest land restoration under low-fertility soil conditions. Cypress is considered a calcicolous tree, and its growth and development can be promoted significantly by exchangeable calcium (Ca2+) in the soil. However, most of the subtropical regions have low-fertility acidic soils, in which Ca2+ gradually becomes a limiting element for Cypress growth. In this study, different concentrations of Ca2+ fertilizer were added under fertile soil (3 g kg−1 NPK fertilizer added) and low-fertility soil (0 g kg−1 NPK fertilizer added) conditions. Cypress clones responded differently to Ca2+ addition in different soil conditions. The seedling height and dry matter quality of Cypress in fertile soil were significantly greater than those in low-fertility soil, but plant height and dry biomass did not differ significantly among Ca2+ treatments. The accumulation efficiencies of nitrogen (N), phosphorous (P) and Ca all differed significantly among the Ca2+ treatments. In low-fertility soil, the addition of 3 g kg-1 Ca2+ significantly promoted development of roots 0.5–2 mm in diameter, and both the C1 and C2 clones achieved their highest N, P and Ca accumulation efficiencies. When the Ca2+ concentration increased to 6 g kg−1, the seedling height, dry matter quality and root development were lower than those of the 3 g kg−1 Ca2+ treatment. In the fertile soil, the addition of Ca2+ significantly inhibited development of roots 0.5–1 mm in diameter. The highest N accumulation efficiency was achieved under the 0 g kg−1 Ca2+ treatment, and the highest Ca accumulation efficiency was achieved under the 6 g kg−1 Ca2+ treatment. Seedling height, root dry weight, roots 0–1.5 mm in diameter and Ca accumulation showed a significant interaction effect between NPK fertilizer and Ca2+. Therefore, Ca accumulation was more efficient in low-fertility soils. Under low-fertility soil conditions, the addition of CaSO4 can promote the root development of seedlings and advance and prolong the fast growth period of seedling height. Cypress clones can be used as an important tree species for afforestation under low-fertility soil conditions, especially under calcareous soil conditions.

The influence of the variation in water content, sampling location and loading speed on the shearing and across-compression mechanical characteristics of dwarf-denseearly major cotton variety stalks was analysed. The second-order response models of the factors vs. the maximum shear force and critical across-compression failure force were built using the response surface methodology. Besides, the sampling location and water content have a significant influence over the maximum shear force and critical across-compression failure force, especially the sampling location. Both the maximum shear force and critical across-compression failure force reach a theoretical maximum when the water content is 10%, the sampling location is the lower part and the loading speed is 40 mm min-1. When the sampling location is the lower part, optimization is carried out assuming the theoretical minimum of the maximum shear force and critical across-compression failure force as the optimization target. In this case, the loading speed is 20 mm min-1, while the water content is between 30 and 50%. This indicates that the parameter range requires a low power consumption during the crushing of film residue mixtures. The test data can provide some theoretical references for the technical research and equipment development for the crushing of film.
