The genus Phytophthora plays an important role not only in agriculture but also in forest ecosystems. As the number of known Phytophthora species continues to grow, identifying new isolates in this genus has become increasingly challenging even by DNA sequencing. Therefore, the development of proper techniques for detection and identification is crucial for monitoring and control of these pathogens in the forestry sector. In recent years, new molecular methods using innovative approaches have indeed been developed. However, the majority of these methods was designed to detect single Phytophthora species. Techniques that are able to target multiple species would offer advantages, especially for the assessment of Phytophthora diversity in the environment. This paper describes a multiplex assay for the identification of eight Phytophthora isolates, down to the species level, based on a Fluidigm platform employing pyrosequencing. The obtained results showed that for an accurate determination of the species, it is sufficient to know the sequence of two markers, ITS and COX1. The sensitivity of this test is sufficient to identify Phytophthora in a pure culture. Unfortunately, analysis based on a pyrosequencing platform does not provide enough data to simultaneous identify multiple Phytophthora species in samples collected in the field. This problem could be resolved in the future by sequencing using more efficient platforms like Illumina or IonTorrent.

The aim of the research was to determine the impact of climatic conditions on the tree−ring width of Swedish whitebeam Sorbus intermedia (Ehrh.) Pers. growing in the urban conditions in Szczecin (NW Poland). The samples were taken from 26 trees growing along Rodziewiczówny, Ejsmonda and Santocka streets (left bank of the Odra river). On the basis of the tree−ring width measurements and cross−dating, two local chronologies (JS1 – Rodziewiczówny and Ejsmonda streets, and JS2 – Santocka street) were developed. Later on we indexed them (double detrending, least squares method and spline function) and subjected to the analysis of correlation and response functions. Growth−climate dependencies in both Swedish whitebeam communities are different. For the JS1 chronology the highest value of the multiple regression coefficient was obtained for air temperature, while for JS2 chronology for insolation and precipitation. For both sequences dependency on climate is similar in August and December of the previous year of vegetation and in January, February, April, June and July of the current year. The observed differences are the result of the impact of various factors related to human activity: traffic volume, soil pollution (mainly heavy metals and salt), damage to roots, trunks and crowns.

Black Alder Alnus glutinosa (L.) Gaertn. is an important tree commonly growing in Poland. Alders are actinorhizal plants that play an important ecological role in riparian ecosystems through atmospheric nitrogen fixation, filtration and purification of waterlogged soils as well as providing a refuge for terrestrial and aquatic organisms thus helping to stabilize stream banks. Black Alder used to be considered a very pest and disease resistant species but, the situation changed in 2000, when an unprecedented decline of Alders was observed in Poland. In the Białowieża Forest District, this decline has been observed on wet meadow habitats and along rivers or watercourses. Currently, there are several hypotheses explaining Alder dieback, among them climatic changes and Phytophthora infections. In terms of climate, Black Alder requires a high atmospheric humidity during all phases of its reproductive cycle. It tolerates neither long-term summer flooding nor a significant decrease in the groundwater level. In terms of pests, oomycete pathogens of the genus Phytophthora are the most destructive plant pathogens known and many of them are present in forests and nurseries all over Europe. The aim of this study was to evaluate the health of Black Alder along the Narewka River in the Białowieża Forest District. Selected areas were monitored in 2012 and 2018, but no relationship between drought and alder health was found. A preliminary analysis of soil and water samples by real time PCR revealed the presence of two Phytophthora species: P. alni and P. cactorum. Further and more detailed research is required to elucidate the role of these pathogens in Alder dieback.

In forestry management, artificially produced planting material is mainly used for renewal the tree population. Seedlings are cultivated in two systems: in the ground (the bare−root seedlings) and in controlled conditions (container seedlings). The aim of the study was to analyse the microsatellite markers of nuclear and chloroplast DNA, in terms of the number and frequency of rare, private, low frequency and common alleles in the planting material of Scots pine. The rare alleles included alleles occurring with less than 1% in analyzed group of seedlings and low frequency alleles occurred with a frequency of less than 25%. The private alleles were detected only in one group of seedlings. Genetic pools of seedlings from traditional (soil) and container production were compared. Planting material came from nurseries in the Olsztynek (N Poland) and the Oleszyce (S Poland) forest district. With the similar number of observed nDNA and cpDNA alleles in both analyzed locations, a higher number of rare, low frequency and private alleles was found within container seedlings. Most private alleles were a rare allele. Rare and private alleles are supposed to be responsible for adaptation to changing climatic conditions and a stressful environment. It seems reasonable to continue research on the meaning of rare and private alleles under conditions of strong selective pressure.

Phytophthora pathogens cannot be controlled with well−known fungicides, because as oomycetes they do not synthesize chitin and ergosterol. Phytopathogenic microorganisms of Phytophthora genus offers an alternative to pesticides. The aim of the study was to understand the interactions among the host silver birch (Betula pendula), a common forest tree species in Polish lowlands and in lower mountain locations, and its primary pathogen Phytophthora plurivora as well as potential Biological Control Agents Bacillus subtilis and Trichoderma asperellum. The 2−year−old silver birch seedlings were selected for this experiment. Interactions between P. plurivora and B. subtilis, producing peptide antibiotics including polymyxin B and subtiline, stimulated growth of birch seedlings. Comparing to the control a stimulation of both height and root collar diameter of plants was observed when B. subtilis was added into the soil of pots. It is a saprophyte, decomposing organic compounds of plant origin. However, the application of bacterium into the rhizosphere soil stimulated more shoots growth than roots. The application of T. asperellum into the soil stimulated development of roots, and in consequences the above−ground parts of plants. However, in the combination with pathogen, T. asperellum protected the roots only partially. The presence of pathogen and its antagonists increases the biomass of birches compared to control plants. Chlorophyll fluorescence studies proved better parameters like total performance index (PI total) after application of B. subtilis, in contrast, interaction between B. subtilis and P. plurivora negatively affected photosynthesis causing weakening of plants. The higher content of carboxylic acids, observed in the variant with T. asperellum + P. plurivora, indicates the initiation of biochemical defence processes in birch leaves cells.

Introduction. Dobrava-Belgrade virus (DOBV) is one of the emerging pathogens which have been reported during the last decades in Europe and have attracted the attention of researchers. The course of infection among humans may have a varied course – from the completely asymptomatic to the more severe forms, such as haemorrhagic fever with renal syndrome (HFRS). DOBV is hosted and carried by rodents like Apodemus flavicollis or A. agrarius, which occur commonly in Europe. Objective. To-date, orthohantaviruses have been reported in Poland, both in humans and animals, but detailed countryscale studies have not yet been carried out. The aim of the study was molecular characterization of a strain which was found in A. flavicollis in south-eastern Poland. Materials and method. The phylogenetic analysis of the first Dobrava-Belgrade virus found in A. flavicollis in the subcarpathian region of south-eastern Poland, presented in this study, was performed after virus proliferation in cell culture and sequencing of specific PCR products. Results. Based on genetic sequences of fragments of three segments (S, M and L), the isolated virus was assigned to the Dobrava genotype, taking into consideration the most current classification of the DOBV species. Conclusions. The Dobrava-Belgrade virus strain isolated from A. flavicollis in the subcarpathian region of south-eastern Poland, has been molecularly characterized and assigned to Dobrava genotype, thereby the occurrence of that genotype in Poland has been confirmed by molecular techniques.

Opis:
Oak decline phenomenon has been observed in Poland in the 1980s. Especially, Krotoszyn Forest District was affected in the years 1982−1984, when oaks were dying in a mass extend causing economic and ecological problems. Twenty five years later many Phytophthora species were isolated from rhizosphere soil suggesting their important role in fine root damage. This research was focused on application of fertilisers increasing oak resistance to soil−borne pathogens. In total, 60 soil samples were collected c.a. 1 meter from trunks of oaks representing control and treated variant with Actifos® and Kalex® fertilisers. The fine root parameters (smaller than 2 mm) were evaluated with the water scanner and WinRhizo software. The same soil samples were analysed for the presence of macro− (N, P, K, Ca, Mg) and microelements (Al, Cu, Fe, Mn, Na, Zn). The use of ammonium and potassium phosphites in the form of foliar spray had a beneficial effect on the health of oak stands (Quercus robur L.) in the Krotoszyn Forest District. Three parameters: defoliation (ICP Forest), vitality according to Roloff method and synthetic index of damage were applied in order to evaluate the status of oaks health. Two aforementioned crown parameters were obtained from visual assessment from the ground, while the synthetic index of damage was calculated according to the formula developed by Dmyterko and Bruchwald. The parameters of fine roots after treatments, especially their number, length and surface improved. Trees with better quality fine roots had more chance to survive in unfavourable environmental conditions like drought. Phosphite treatments of oak stand positively influenced the chemical properties of soil, decreased acidity, but increased availability of Zn for plants, and probably reduced root damage by toxic aluminium ions. An improvement in the health of fine roots (that were saved by phosphites from damage caused by pathogenic species of Phytophthora genus present in the soil) will have a positive effect on the reconstruction of crown architecture (shoots development from dormant buds).

Opis:
The production of forest tree species in forest nurseries is performed via two main breeding systems: i) the traditional (conventional) way with the seedlings grown in soil, and ii) plants cultivated in the containers. The aim of the study was to assess the level of genetic variability in the populations of the mother stands and the progeny populations of Scots pine cultured with traditional way (in soil) and in containers in two nurseries in Olsztynek (N Poland) and Oleszyce (S Poland) forest districts. Four polymorphic microsatellite markers (SPAG 7.14, SPAC 11.6, SPAC 12.5 and SsrPt_ctg4363) were used to evaluate the genetic variability of the studied populations. The basic hypothesis assumed that higher gene pool characterizes the seedlings grown in the containers comparing to the seedlings grown in the ground. The results confirmed that. Seedlings from containerized breeding had larger gene pool and were more diverse than plants with conventional breeding, both in Olsztynek and Oleszyce. Our study revealed a significant human impact on shaping the pool of forest genetic resources of Polish forests at the early stage of nursery production and showed the need for a broader study on further stages of cultivation of forests.

Umowa o partnerstwie publiczno-prywatnym ma kluczowe znaczenie dla prawidłowej realizacji przedsięwzięcia, jednakże ustawa o partnerstwie publiczno-prywatnym w ograniczonym zakresie normuje zarówno samą umowę o partnerstwie, jak i umowę przenoszącą prawo własności składnika majątkowego jako wkładu własnego. Prawidłowa ocena sytuacji prawnej wymaga zatem wykładni przepisów kilku aktów prawnych i określenia ich wzajemnego stosunku. Artykuł wskazuje jakie regulacje i w jakim zakresie powinny być stosowane w przypadku wnoszenia wkładu w postaci nieruchomości, polegającego na przeniesieniu jego własności.
The public-private partnership agreement is of key importance for the proper realization of the undertaking, however the Public-Private Partnership Act to a limited extent norms both the partnership agreement itself and the contract transferring the ownership of the property as own contribution. A correct assessment of the legal situation therefore requires an interpretation of the provisions of several legal acts and the determination of their mutual relationship. The article indicates what regulations and to what extent should be applied in the case of making a contribution in the form of real estate, consisting in the transfer of its ownership.

During plant propagation changes in genetic stability may occur, not only during the in vitro multiplication but also in the ex vitro conditions in response to the sum of stress factors affecting plants. It is therefore necessary to evaluate planting material in terms of its genetic stability. The aim of the work was to assess by the RAPD method genetic stability of the ex vitro rooted blueberry (Vaccinium corymbosum ‘Bluecrop’ and ‘Duke’) microcuttings. Also an effect of biostimulator Goteo (0.2%) on rhizogenesis was tested as compared to the auxin indole-3-butyric acid IBA (50 mg·l–1) applied as water solution and a commercial rooting powder Rhizopon AA containing 1% IBA. All the rooting stimulators improved the percentage and degree of rooting in blueberry microcuttings. The biostimulator gave results only slightly poorer than the auxin. Both cultivars were genetically stable and no DNA polymorphism was found between the traditionally propagated stock plants growing in a nursery and those rooted in vivo, regardless of the rooting enhancer used.
