Introduction and objective. It is easier and non-invasive to obtain faecal samples compared with blood samples. Molecular techniques may enable detection of parasites even in tiny amounts of blood-containing faeces. We aimed to compare the sensitivity of detection of three Babesia species and Hepatozoon canis in blood and faecal samples, including samples derived from naturally infected hosts. Materials and method. Three groups were involved: 1) Nine BALB/c mice infected with Babesia microti sampled during acute (n=3), post-acute (n=3) and chronic phases of infection (n=3); 2) Eight dogs with symptoms of babesiosis; 3) Six red foxes infected with B. vulpes, one fox infected with B. canis, four foxes infected with H. canis. Genomic DNA was extracted from blood and faeces by use of commercial kits and amplified with genus-specific primers in one-step or nested PCR reactions. Selected PCR products were sequenced. Results. No positive results for faecal samples were obtained from H. canis-positive foxes in contrast to Babesia spp. infections. Positive results from PCRs were obtained for all BALB/c mice (100%), five dogs (62.5%) and four of seven foxes (57.1%). Successful sequencing was obtained for six selected murine samples (B. microti), four canine samples (B. canis) and for one fox sample (B. vulpes). The success of B. microti detection in murine faecal samples from acute, post-acute and chronic phases was identical (100%). Conclusions. Detectability of Babesia spp. infections was lower in naturally infected dogs and foxes, compared to experimentally infected mice. Detection of DNA in faecal samples can be useful in the detection of Babesia infection in populations from which blood samples are hard to obtain, but due regard must be given to the possibility that prevalence of infection may be severely underestimated.

Opis:
The article deals with the gas development of the geopolymer binder system hardened by heat and provides the comparison with organic binder systems. The GEOPOL® W technology is completely inorganic binder system, based on water. This fact allow that the gas generated during pouring is based on water vapour only. No dangerous emissions, fumes or unpleasant odours are developed. The calculated amount of water vapour generated from GEOPOL® W sand mixture is 1.9 cm3/g. The measured volume of gas for GEOPOL® W is 4.3 cm3/g. The measurement of gas evolution proves that the inorganic binder system GEOPOL® W generates very low volume of gas (water vapour) in comparison with PUR cold box amine and Croning. The amount of gas is several times lower than PUR cold box amine (3.7x) and Croning (4.2x). The experiment results are consistent with the literature sources. The difference between the calculated and the measured gas volume is justified by the reverse moisture absorption from the air after dehydration during storing and preparing the sand samples. Minimal generated volumes of gas/water vapour brings, mainly as was stated no dangerous emissions, also the following advantages: minimal risk of bubble defects creation, the good castings without defects, reduced costs for exhaust air treatment, no condensates on dies, reduced costs for cleaning.

The Dermacentor reticulatus tick is a main vector of Babesia canis in Europe. The risk of canine babesiosis is unpredictable, due to significant differences in the prevalence of B. canis between ticks originating from closely situated regions. This phenomenon may be explained by vertical transmission of the pathogen in a vector population. Thus, molecular techniques were applied to investigate the occurrence of transovarial transmission in D. reticulatus ticks. DNA of B. canis was detected in 20.7% (6/29) of engorged female ticks collected from dogs, in every pool of eggs laid by positive females (100%, 6/6) and in larvae hatched from these eggs. In the pools of eggs collected from two positive females (2/6; 33.3%), no larvae hatched and no embryos were observed inside the eggs. Conclusions. Transovarial transmission of B. canis can be an important mechanism supporting maintenance of the pathogen in the environment without the presence of a reservoir vertebrate host. However, the efficiency of transovarial transmission in the maintenance of B. canis in natural conditions requires further field research.

Introduction and objective. Tick-borne infections constitute an increasing health problem in dogs and may lead to death, especially in young or elderly individuals. Canine babesiosis constitutes a serious health problem in dogs worldwide. The aim of the study was to verify the probability of vertical transmission of Babesia canis between the bitch and the pups. Materials and methods. In Autumn 2011, cases of babesiosis were diagnosed in a litter of 6-week-old puppies of a Central Asian Shepherd dog. Immediately following the first case of infection, blood samples were collected from all the pups in the litter (n=10) and from the female. Detection of Babesia infection was performed by molecular and microscopical techniques. Results. The presence of B. canis DNA was detected using PCR in three pups, presenting at the time or 24–48 hours later with babesiosis symptoms, and in their asymptomatic mother. The isolates derived from the pups and the female – 520 bp 18S rRNA gene fragment – were compared and analyzed. All isolates from the pups and their mother were identical and showed 100% homology with B. canis group B (EU622793), supporting the same source of infection. Additionally, the USG of the peritoneal cavity was performed in the female, presenting evidence for splenomegaly. Conclusions. On the basis of (1) the same timing of three pup cases; (2) the identical B. canis sequences derived from all positive dogs; (3) evident splenomegaly in the asymptomatic female, this provides the first evidence of the vertical transmission of this piroplasm in dogs.

Introduction and objectives. Sporting dogs, including sled dogs, are particularly prone to tick-borne infection either due to training/racing in forest areas or through visits to endemic areas. The aim was to present tick-borne infections in a 6-dog racing team after a race in Estonia. Materials and methods. On the 4th day after return to Poland, the first dog presented with babesiosis symptoms and was diagnosed and treated accordingly. Next morning, the dog showed neurological symptoms and was diagnosed with tickborne encephalitis (TBE). Diagnosis was confirmed by a high level of IgG antibodies (922 IU/ml), detected in serum 3 months later. The second dog presented with babesiosis symptoms on the 7th day after return. Babesia DNA was extracted from blood, amplified and sequenced to answer the question of whether the dogs became infected during the race in Estonia or in Poland. Results and conclusions. Sequencing of a fragment of Babesia 18S rDNA revealed that these two isolates were identical to one another and closely related to the B. canis sequence originally isolated from the dog and Dermacentor reticulatus ticks in Poland. Thus, this is the first confirmed case of B.canis and TBEV co-infection and first confirmed case of TBE in a dog in Poland.

Microsporidia are intracellular parasites that cause opportunistic infections in humans of various immunological status. Only a few case reports exist on microsporidial infection in solid organ transplant recipients worldwide. The presented study demonstrates the first case in Poland of Enterocytozoon bieneusi infection in a liver transplant patient. Parasites were diagnosed in stool samples using both modified trichrome staining and PCR.
