Reakcje roślin na fizyczne czynniki środowiska są bardzo złożone. Można je obserwować na różnych poziomach, począwszy od zmian w intensywności podstawowych procesów biochemicznych, takich jak: oddychanie, fotosynteza czy replikacja DNA, aż po zmiany morfologiczne organów lub całych organizmów. Badania reakcji roślin na stresy mają dwa podstawowe cele; pierwszy to lepsze, bardziej dogłębne poznanie otaczającego nas świata, drugi praktyczny - ukierunkowany na zastosowanie zdobytej wiedzy w rolnictwie. Badania aplikacyjne prowadzone są z wysoką intensywnością, ponieważ prawie wszystkie środowiska wykorzystywane w produkcji roślinnej są zmienne i oferują warunki odmienne od wymaganych przez rośliny do optymalnego wzrostu i plonowania. Tolerancja na stres jest pojęciem bardzo ogólnym. Można ją zdefiniować jako zdolność rośliny do utrzymywania procesów życiowych na jak najmniej zmienionym poziomie, w warunkach środowiska znacznie odbiegających od optymalnych. Z punktu widzenia genetyki tolerancja na stres jest cechą o charakterze ciągłym, dziedziczoną wielogenowo. Do najistotniejszych czynników stresowych zaliczyć należy niską temperaturę z podziałem na niskie temperatury powyżej zera i mróz, wysoką temperaturę, suszę, zasolenie podłoża i światło. Ostatni z wymienionych czynników stresowych nie był omawiany ze względu na niedostatek danych opisujących genetyczne uwarunkowanie odporności na ten czynnik stresowy. W pracy zasygnalizowano również znaczenie substancji osmotycznie czynnych w ochronie organizmów przed stresami abiotycznymi.
Plant reactions to physical factors of the environment are very complex. They can be observed and studied at various levels, starting from the changes in the intensity of very basic biochemical processes such as photosynthesis or DNA replication up to alterations in morphology of some organs or the whole organism. Studies of plant reaction to stress have two main aims. The first one is better understanding of the surrounding world. The second one is utilitarian. This approach is focused on application of the gathered knowledge in agriculture. As almost all environments used for plant production are very far from being optimal in regard to plant productivity, significant emphasis is placed on the studies of genes involved in reaction to physical stresses. The main recipients of this research are plant breeders struggling to select new varieties more suited to harsh and unstable conditions. Stress tolerance is a very general notion. In the simplest way it can be described as plant ability to cope with stress. In terms of genetic, stress tolerance has a complex poligenic character. The main types of abiotic stresses plants must cope with are: temperature (high or low with distinct division into low temperatures above zero and frost), drought, light and salinity. In real life very often crops are subjected to two or even more stressors at the same time, e.g. high temperatures are very often accompanied by drought and soil salinity. Situation is further complicated by the fact that many genes are triggered by more than one stressor. Due to scarcity of information, problems of light stress tolerance genetics have not been discussed. Importance of osmoprotecting solutes was underlined in a separate chapter.

Plants have evolved complex of protective mechanisms that allow them to grow and develop in heavy metal-contaminated environments. They show different degrees of tolerance to heavy metals, depending on the species, population or even developmental stage of the plant. This is a review of literature data on the anatomical, physiological and biochemical mechanisms responsible for the formation of "the heavy metal tolerance syndrome " in plants. Numerous factors have been involved in development of heavy metal tolerance. The main ones encompass processes occurring on the root surface of the plants that lead to restriction of heavy metal absorption, mycorrhizae, transport of heavy metals through the apoplastic pathway, transport from the epidermis to the vascular cylinder, transport from the root to the shoot, and transport across the plasma membrane to the cytoplasm. Plants are able to detoxify heavy metal ions inside the cell with phytochelatins, metallothioneins or organic acids. Other factors involved in, and contributing to heavy metal tolerance include excretion of heavy metals from the cytoplasm to the apoplast and from plant organs, specific mineral-handling pathways and water regimes of plants, as well as the ability to synthesize heavy metal-resistant enzymes. Knowledge of the fundamentals of heavy metal tolerance will aid in the effective use of plants in phytoremediation.

In the experiment run at aquatic cultures there was examined sensitivity of 3 plant species (maize, sunflower, lettuce) to aluminium (0 - control, 20, 100, 200 μmol・dm⁻³) at nitrogen nutrition in reduced and oxygenated form. Tolerance of sunflower, lettuce and maize in particular increases when oxygenated form makes a nitrogen source. Reduced form of nitrogen causes Al³⁺ phytotoxicity increase. Plants tolerant to aluminium and fed reduced nitrogen form demonstrate higher carbohydrates content, especially in roots.
W doświadczeniu przeprowadzonym w kulturach wodnych badano wrażliwość 3 gatunków roślin (kukurydza, słonecznik, sałata) na stężenie glinu (0 - kontrola, 20, 100, 200 μmol・dm⁻³) w warunkach żywienia azotem w formie zredukowanej oraz utlenionej. Tolerancja roślin słonecznika, sałaty, a szczególnie kukurydzy wzrasta, kiedy źródłem azotu jest forma utleniona. Azot w formie zredukowanej powoduje wzrost fitotoksyczności glinu. Rośliny tolerancyjne na glin i żywione zredukowaną formą azotu charakteryzują się wyższą zawartością węglowodanów, szczególnie w korzeniach.

Plants are sessile organisms hence environmental factors such as excessive light and high air temperature lead to significant reductions of their productivity and quality of gained yield. In fact, scientific and agriculture hubs make lots of efforts to improve crop tolerance to elevated temperature, selecting more tolerant varieties. We analyzed less expensive and highly efficient method to improve resistance of well-known cultivars of crop plant by reversible modification of xanthophyll cycle. It functions as a safety valve to adjust energy transfer and protects fragile structures of photosynthetic machinery from excessive light, especially accompanied by heat or water stress. Efficiency of modified xanthophyll cycle activity was measured after pre-treatment with four, chemically different regulators, with or without light illumination. Analyses were carried out on barley (Hordeum vulgare L.) cv. Zenek treated with ascorbic acid (AsA), dithiothreitol (DTT), putrescine (Put) and calcium ions (Ca2+). To measure the scale of thermal energy dissipation we traced energy transfer absorbed by PSII with PAM chlorophyll fluorescence technique. Results showed clear correlation between AsA (activator of violaxanthin de-epoxidase) treatment and stimulation of the Φ(NPQ) at increased temperature. DTT (inhibitor of violaxanthin de-epoxidase) decreased the cycle activity at 45 °C at the same time increasing its value at 35°C, caused by interaction with other enzymes. Action of Put (hydrogen ions buffer) concerned mainly a non regulated Φ(NO) energy quenching. We noticed that application of Ca(NO3)2 (Ca2+ source for enzyme activity) reduced the Φ(NPQ) at 45 °C and stimulated it at 25 °C. Obtained results confirmed postulated possibility of creating new type of plant protection products (PPPs) able to precisely manage natural mechanisms of heat resistance.

Zanieczyszczenia gleby metalami ciężkimi mają toksyczne działanie na rośliny, zwierzęta oraz człowieka. Metalofity, rośliny odporne na metale ciężkie, kolonizujące tereny metalonośne, są wykorzystywane do fitoremediacji, czyli oczyszczania gleb z metali ciężkich.
Wykorzystanie roślinnych kultur komórkowych do badań nad toksycznością metali i tolerancją komórek na ich działanie jest stosunkowo nową techniką. W pracy zostały przedstawione możliwości wykorzystania roślinnych kultur zawiesinowych w badaniach nad wpływem metali ciężkich na metabolizm komórek oraz metody oszacowania ich toksycznego wpływu. Zaprezentowane zostały techniki otrzymywania kultur zawiesinowych, oceny żywotności komórek, akumulacji metali ciężkich w komórkach. W ocenie toksyczności metali stosuje się także badania nad programowaną śmiercią komórki (PCD), co pozwala oszacować reakcję komórek na ich wysokie stężenia. Zostały przedyskutowane mechanizmy tolerancji komórek na metale ciężkie. Kultury zawiesinowe są dobrym modelem do badań tolerancji na metale, ponieważ pozwalają zbadać ich wpływ na pojedyncze komórki w jednolitych, stałych warunkach.

Soil pollutants exert toxic effects on plants, animals and humans. Metallophytes, plants tolerant to heavy metals colonizing polluted areas, are being used to phytoremediation - cleaning up soil contaminated with heavy metals.
The use of plant cells in vitro cultures to study heavy metal toxicity and tolerance is a relatively new approach in research of metal toxicity. In this paper the usefulness of plant suspension cultures to study the impact of heavy metals on cells is presented alongside with the methods of obtaining suspension cultures, evaluation of cell viability, metal accumulation and detection of programmed cell death (PCD). The mechanisms by which cells of plant species tolerant to heavy metals develop resistance to metal toxicity are discussed. Cell suspension cultures appear to be a good model to study tolerance to heavy metals because they allow to estimate metal impact to a single cell in stable uniform conditions.

Six-week old sunflower seedlings, cv. Ogrodowy, were treated with 0, 15, 45 and 60 mg Pb dm-3, and then the content of lead and selected physiological and biochemical parameters were measured. Photosynthesis efficiency, water relations (intensity of transpiration, relative water content (RWC)) and gene-encoding metallothionein were measured three times after 24, 48 and 72 hours of exposure to Pb. The content of glutathione and lead was analysed after 72 hours’ exposure to Pb. Most of the lead uptake was accumulated in the roots, then in the stems and leaves, but when re-calculated per plant dry weight, the uptake of the metal did not depend on the lead dose applied. The highest 60 mg Pb dm-3 treatment was accompanied by a significant decrease in dry weight content. Moreover, most of the lead taken up in these plants was transported to the stems and leaves (23.6% of total lead uptake). The lead doses used in this study did not affect the intensity of photosynthesis, but a decrease in transpiration and relative water content was observed. The glutathione level in the plants varied depending on the organ examined and the Pb concentration in the treatment. The expression of the metallothionein gene HaMT1 was observed in the stems only. These results indicate that the sunflower cultivar Ogrodowy is a promising plant for phytoremediation of lead-polluted soils.

Opis:
Valerian (Valeriana officinalis L.) is a medicinal plant, but its cultivation is restricted by weed competition. Therefore, three rates (0.75X, 1X, and 1.25X, where X is equal to the recommended dose of haloxyfop-R (methyl ester), sethoxydim, oxadiargyl, bentazon, oxadiazon, and oxyfluorfen) were applied at the 3–4 leaf stages to valerian plants. This application was done to select the herbicide type and rate for post-controlling broadleaf and grasses weeds in this species. Herbicide injury, Soil-Plant Analyses Development (SPAD) reading, number of leaves per plant, stem diameter, and fresh and dry weights were determined 10, 20, and 30 days after herbicide application. Oxyfluorfen application caused the most herbicide injury followed by bentazon. Injury increased as the rate and the days after application increased. Oxadiazon only caused significant damage 30 days after application under all three rates. Other treatments showed no marked injuries under any rate or date after application, as compared with the control. Effects on other measured traits depended on the trait, herbicide, and herbicide rate. The highest SPAD, leaf number, shoot diameter, fresh weight and dry weight, was recorded under application of 30 mg a.i. ∙ kg–1 soil oxadiargyl and 90 mg a.i. ∙ kg–1 soil oxadiazon, 81 mg a.i. ∙ kg–1 soil haloxyfop-R, 37.5 mg a.i. ∙ kg–1 soil oxadiargyl, 22.5 mg a.i. ∙ kg–1 soil oxadiargyl, 81 mg a.i. ∙ kg–1 soil haloxyfop-R, and 81 mg a.i. ∙ kg–1 soil haloxyfop-R, respectively. To sum up, the results showed that sethoxydim, oxadiargyl, and haloxyfop-R produced no significant symptoms of phytotoxicity or reduction of measured traits. This means that oxadiargyl, haloxyfop-R, and sethoxydim may be used safely for weed control of valerian at the rates used in this experiment under similar conditions.

Chilling stress is of major limiting factors influencing the growth and development of warm-season crops like cucumber. In this research, the possibility of chilling tolerance of cucumber seedlings was investigated through employing the drought and low-temperature pretreatments. The factorial experiment consisted of two factors including cucumber cultivars (i.e. ‘Super Dominos’ and ‘Super Star’) and hardening treatments (control, low temperatures at 10°C, and 15°C and drought simulated by 10% and 20% PEG) based on completely randomized design (CRD) in 3 replications. After applying treatments and providing them 48 h opportunity to be recovered, the seedlings were subjected to 3°C for a six-day period and 6 h for each day. All hardening treatments improved seedlings’ growth, chlorophyll content, total phenol (TP) and antioxidant enzyme activities, while reducing chilling injury index and malondialdehyde (MDA) content. Comparing to temperature hardening, the drought pretreatment showed to have a better effect on inducing the chilling tolerance into cultivars. Overall, the results of this experiment showed that employing drought and low-temperature pretreatments enabled cucumber seedlings to mitigate the harmful effects of chilling.

The aim of the study was to induce somaclonal variability in the callus culture of L. hirsutum f. typicum and L. chilense and to characterize them in respect to tolerance to salinity. Callus was initiated on the cotyledon fragments grown on the medium supplemented with NAA and BAP. The tolerance of callus to salt was tested on MS media containing NaCl in concentration 25, 50, 75, 100, 200 mM. Callus tolerant to 100 mM NaCl was next regenerated by ten weeks on the media with different doses of growth regulators. After this time genetic differences between selected fragments and control (MS medium containing no growth regulators) were determining using ISSR-PCR method. The results show that NaCl concentration significantly affects the regeneration of callus in L. hirsutum f. typicum and L. chilense. The dose 200 mM NaCl of the medium results, in both species, in callus dying. Comparing the genetic similarity of examined callus samples with the control ones in both species, it may be stated that the differences in their response to NaCl and applied growth regulators were generally in the range 2–30%.
Celem pracy była próba indukowania zmienności somaklonalnej w kulturach kalusowych L. hirsutum f. typicum i L. chilense oraz charakterystyka ich tolerancji na zasolenie. Kalus inicjowano na fragmentach liścieni wyłożonych na pożywkę uzupełnioną NAA i BAP. Tolerancję na zasolenie badano, wykładając fragmenty kalusa na pożywkę uzupełnioną 25, 50, 75, 100, 200 mM NaCl. Kalus uznany za tolerancyjny był regenerowany na pożywkach uzupełnionych różnymi kombinacjami roślinnych regulatorów wzrostu. Następnie metodą ISSR-PCR określono różnice genetyczne pomiędzy wybranymi fragmentami kalusa a kontrolą. Na podstawie otrzymanych wyników badań stwierdzono, że stężenie NaCl w sposób istotny wpływa na regenerację w przypadku obu badanych gatunków. Stężenie 200 mM NaCl powoduje zamieranie tkanki kalusowej. Obserwowana u obu gatunków pomidora na poziomie kalusa tolerancja na zasolenie NaCl jest prawdopodobnie w większości przypadków zmiennością dziedziczną.

It is a common characteristic of all life forms that some of the elements present in the environment are accumulated and others are rejected. The rates of accumulation are necessarily governed by physiological requirements rather than toxicity. Because metals like Ca, Mg, Fe, Cu, Zn, Mn, Mo, Ni are essential for normal running a vast number of metabolic processes, plants evoke special systems of metal uptake and transport during evolution. On the other hand, the same metals present in an excess in plant cells may become seriously toxic to the cells. Moreover, the low specificity of the metal uptake and transport systems makes them to function also as an entrance for nonessential and toxic substances (Cd, Pb). The existence of a complex systems for metal uptake and transport processes, able to respond to continuously changing environmental conditions, is necessary for maintaining metal homeostasis in plants. It is highly probable that these complex systems of metal uptake and transport can function as systems of heavy metal tolerance in plants, as well. According to the recent reports, the main processes involved in maintaining heavy metal homeostasis in plants are: metal ions mobilization and uptake from the soil, their short distance transport in roots, complexation by different ligands in cytosol, compartmentation in root cells, xylem loading and long distance transport in xylem, distribution in shoots, xylem unloading, ions trafficking in apoplastic and symplastic passage of leave cells, their chelating in cytosol and compartmentation in leave cells and structures. Although some of these processes are well recognized, a number of them remain still enigmatic. It is strongly believed that further investigation of these processes should help to explain differences in metal tolerance between plant varieties as well as the phenomenon of heavy metal hyperaccumulation in plants.

Endophytes are an important constituent of sustainable agriculture because of their ability to produce a large number of agriculturally important metabolites. A salt-tolerant fluorescent green pigment-producing endophytic bacterium was isolated on 2.5% NaCl-supplemented nutrient agar from the leaf samples of Zanthoxylum alatum Roxb. The isolate Z1B4 was identified as Pseudomonas fluorescens based on morphological features, fatty acid methyl ester analysis, biochemical tests, and 16S rRNA gene sequencing. P. fluorescens Z1B4 showed positive results for tricalcium phosphate solubilization; 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity; and production of auxins, siderophores, hydrogen cyanide, and ammonia. P. fluorescens Z1B4 also showed strong antagonistic activity against Curvularia lunata (MTCC 283), Fusarium verticillioides (MTCC 3322), and Alternaria alternata (MTCC 1362) and exhibited stress tolerance to a wide range of temperature and pH and concentrations of NaCl and calcium salts. Under natural conditions, following inoculation with the isolate Z1B4, a significant increase in the growth of pea and maize test plants in pots was observed compared to that of uninoculated control plants. The rifampicin-resistant mutant Z1B4Rif was recovered from the roots, shoots, and leaves of the test plants, indicating that the isolated endophytic bacterium can grow well within different plant tissues. The present study indicated that the endophytic bacterium P. fluorescens Z1B4 can be used as a bacterial inoculant in stressed environments for sustainable agriculture.

Invasive alien species are non-native plant species that displace native species and pose adverse effects to environment, ecosystem, economy, and human health by diminishing the growth of native plants and by exhibiting higher stress tolerance. In our present study, four invasive alien species, namely Lantana camara, Parthenium hysterophorous, Ricinus communis, and Ageratum conyzoides, were studied from different locations. Plants growing under natural environmental conditions were sampled at random in the vicinity of Jalandhar. To gain insights into the biochemical basis of invasiveness of these plants, the samples were subjected to chemical fingerprinting by using UV-Vis, fluorescent, and Fourier transform-infrared (FT-IR) techniques under natural abiotic stress conditions (moderate and hot conditions). Indices of oxidative stress, such as malondialdehyde (MDA), were also studied. MDA levels were enhanced under hot conditions. Elevated peaks (major and minor) were observed in UV fingerprinting during adverse abiotic conditions. Fluorescent spectroscopy also validated the enhanced levels of secondary metabolites. FT-IR spectroscopy confirmed the presence of alkaloids and phenolics during stress conditions. Peaks were identified as rutin, vanillic acid, ascorbic acid, and glutathione reduced. The obtained results showed that under stressful conditions, the studied plants may produce an increased level of metabolites that might play a role in minimizing the oxidative stress faced by these plants. It was concluded that the studied plants, namely P. hysterophorus, L. camara, R. communis, and A. conyzoides, have the potential to cope with abiotic stress such as high temperature, which could be the reason for their invasiveness and vast adaptability.

Tolerance of several strains of the P-, S-, and F- intersterility groups of Heterobasidion annosum to low pH and high concentration of aluminium ions was studied on malt extract agar. There were marked intraspecific, but not intergroup, variations in tolerance to both factors. Some strains were relatively tolerant or sensitive to low pH and aluminium, while the others were tolerant to low pH but sensitive to Al. There was no relationship between tolerance of the strains to low pH and their tolerance to aluminium stress.

Biological control of plant diseases is strongly emerging as an effective alternative to the use of chemical pesticides and fungicides. Stress tolerance is an important attribute in the selection of bacteria for the development of microbial inoculants. Fourteen salt-tolerant bacteria showing different morphological features isolated from the rhizosphere of maize were evaluated for different plant growth-promoting activities. All isolates showed auxin production ranging from 5 to 24 μg ⋅ ml–1 after 48 h incubation in tryptophan supplemented media. Phosphate solubilization ranged from 15 to 419 μg ⋅ ml–1. 1-aminocycloproprane- 1-carboxylate (ACC) deaminase activity was shown by 6 isolates, ammonia production by 9 isolates, siderophore production by 8 isolates while HCN production by 4 isolates. Four bacterial isolates with all plant growth-promoting properties also showed strong antagonistic activities against Fusarium oxysporum, F. verticillioides, Curvularia lunata and Alternaria alternata and abiotic stress tolerance against salinity, temperature, pH and calcium salts. Two selected bacterial isolates significantly enhanced the growth of pea and maize test plants under greenhouse conditions. The bacterial isolate M1B2, which showed the highest growth promotion of test plants, was identified as Bacillus sp. based on phenotypic and 16S rDNA gene sequencing. The results indicated that Bacillus sp. M1B2 is a potential candidate for the development of microbial inoculants in stressful environments.

Salinization of soils or waters is one of the world’s most serious environmental problems in agriculture. It is necessary to determine the environmental factors under which medicinal and aromatic plants give higher yields and better quality. The problem of salinity is characterized by an excess of inorganic salts and is common in the arid and semi-arid lands, where it has been naturally formed under the prevailing climatic conditions and due to higher rates of evapotranspiration and lack of leaching water. Although more frequent in arid lands, salt-affected soils are also present in areas where salinity is caused by poor quality of irrigation water. Saline soil induces physiological and metabolic disturbances in plants, affecting development, growth, yield, and quality of plants. Plants affects adversely as a result of salinity, seed germination, survival percentage, morphological characteristics, development and yield and its components. In general, salt stress decreases the photosynthesis and respiration rate of plants. Total carbohydrate, fatty acid and protein content were adversely affected due to salinity effect, but increased the level of amino acids, particularly proline. The content of some secondary plant products is significantly higher in plants grown under salt stress than in those cultivated in normal conditions. The salinity tolerance depends on the interaction between salinity and other environmental factors.
Zasolenie gleb oraz wód jest jednym z najpoważniejszych problemów z zakresu ochrony roślin występujących w rolnictwie. Dlatego niezbędne jest określenie czynników środowiskowych wpływających na jakość, a także wysokość plonowania roślin leczniczych i aromatycznych. Zasolenie spowodowane obecnością nadmiaru nieorganicznych soli jest charakterystyczne dla suchych oraz półsuchych terenów powstałych w wyniku działania lokalnych warunków klimatycznych, braku ewapotranspiracji, jak również przenikania wody. Większy stopień zasolenia terenów suchych spotykany jest na obszarach, na których jakość wody jest niższa. Zasolenie gleby wpływa na fizjologię roślin, wywołując zaburzenia w metabolizmie, rozwoju, wzroście, plonowaniu oraz jakości. Zasolenie wpływa także negatywnie na zdolność kiełkowania nasion, wskaźnik przetrwania roślin, ich morfologię, wydajność oraz jakość plonu. Stres wywołany zasoleniem obniża zdolność rośliny do procesów fotosyntezy oraz procesów ich oddychania. Wynikiem tego jest obniżony poziom całkowitej zawartości węglowodanów, kwasów tłuszczowych oraz białek, a także wzrostu zawartości aminokwasów, w szczególności proliny. W hodowli roślin w warunkach stresu wywołanego zasoleniem podłoża zaobserwowano wzrost poziomu produkcji niektórych metabolitów wtórnych w porównaniu z uprawą w normalnych warunkach. Zdolność roślin do tolerancji zasolenia wynika z interakcji między czynnikami środowiskowymi a warunkami zasolenia.

Globally, more than 5.2 billion hectares of farming fields are damaged through erosion, salinity, and soil deterioration. Salt stress-tolerant bacteria have plant growth-promoting (PGP) characteristics that can be used to overcome environmental stresses. Isolation and screening of salt-tolerant endophytes from Bougainvillea glabra were achieved through surface sterilization of leaves, followed by their cultivation on 0.5% NaCl-supplemented media. The performance of isolates for indole acetic acid (IAA) production, phosphate solubilization, ACC deaminase activity, ammonia production, siderophore production, and stress tolerance was determined. In the present study, 5 morphologically distinct salt-tolerant endophytic bacteria were cultured. Out of 5 isolates, 60% salt-tolerant endophytes showed phosphate solubilization, 100% IAA production, 40% ACC-deaminase activity, 100% siderophore production, 40% ammonia production and 60% HCN production. Dendrogram generated based on stress tolerance showed three clusters: clusters 1 and 2 containing two isolates and cluster 3 containing 1 isolate. Based on the highest PGP activities, BoGl17 and BoGl21 isolates were chosen and further tested for wheat and corn growth promotion. The isolates increased wheat shoot and root dry weight by 40% and 80%, and by 69% and 126% for BoGl17 and BoGl21, respectively. The bacterial isolates identified as Pseudomonas reidholzensis BoGl17 had 95.59% identity with Pseudomonas reidholzensis strain ID3, while the isolate Bacillus aerius BoGl21 had 90.96% identity with Bacillus aerius strain 24K based on phenotypic features and 16S rRNA gene sequencing results. Both isolates significantly improved plant growth as compared to uninoculated plants.

Due to the absence of a suitable method and standardized procedures, the root systems of plants have been evaluated to a much lesser extent than aboveground organs. The aim of this article is (i) to provide a detailed description and thus standardization of an upgraded procedure of electrical capacitance measurement for evaluating the size of the root system of plants in situ, which allows for a reassessment to be made during the growing season and subsequent harvest of seeds for the planting of selected progenies, (ii) to demonstrate, through a standardized methodological protocol, the applicability of root electrical capacitance measurement as a field phenotyping method for the selection of superior root systems to improve crop abiotic stress tolerance and resource efficiency, (iii) to suggest a standardized methodological protocol for the application of electrical capacitance measurements in breeding nurseries, and (iv) to discuss the methodological aspects, development and limitations of this method. A methodological overview of the use of electrical capacitance to measure plant root systems, which emerged from working groups directed by the author of this unique method, is presented along with a standardized protocol. An overview of the application of electrical capacitance measurements of roots in breeding is shown along with some examples of successful applications.

In the field study period from 2006 to 2010, the incidence and severity of sharp eyespot caused by Rhizoctonia were recorded on 36 cultivars of triticale at the milk ripe growth stage. Four localities in north-central Poland were included: Chrząstowo, Dębina, Kończewice and Minikowo. The susceptibility of the seedlings of 30 cultivars of triticale to R. cerealis (AG-D subgroup I) and R. solani (AG-5) was studied in the laboratory. There was much variation in incidence and severity of sharp eyespot between years and locations. The disease was most intense at Chrząstowo. At this location, the mean percentage of diseased stems on 28 cultivars was 2.6-35.7 (-55.0), and the mean disease index was 0.7-15.6 (-24.5), with the lowest and highest values in 2008 and 2009, respectively. At Minikowo, the disease was least intense. At this location, the mean percentage of diseased stems on 23 cultivars was 1.0-4.6 (-18.0), and the mean disease index was 0.3-1.4 (-6.3), with the lowest and highest values in 2006 and 2007, respectively. The cultivars with least intense disease were Tulus and Atletico (Chrząstowo), Grenado and Zorro (Dębina), Krakowiak and Tornado (Kończewice), and Woltario and Constans (Minikowo). The cultivars with most intense disease were Alekto (Chrząstowo), Baltiko (Dębina), Pawo (Kończewice) and Borwo (Minikowo). Mostly R. cerealis was isolated from the diseased stems; R. solani was isolated only sporadically. There was a wide variation in the susceptibility of triticale cultivars to Rhizoctonia. Most triticale seedlings inoculated with R. cerealis produced symptoms typical of sharp eyespot. Seedlings inoculated with R. solani formed extended lesions with no defined borders. Most symptoms developed on coleoptiles, with less symptoms on the leaves and the least on the roots. There was much variation in susceptibility of triticale cultivars to both Rhizoctonia species. Cultivars were grouped into six categories according to the intensity of seedling infection. Categories 1, 2 and 3, representing low, moderate and high susceptibility to R. cerealis, included 17, 10 and 3 cultivars, respectively. Categories 4, 5 and 6, representing low, moderate and high susceptibility to R. solani, included 3, 12 and 15 cultivars, respectively. Cultivars Baltiko and Zorro had low, and cv. Cultivo had high susceptibility to both Rhizoctonia species. No cultivar was resistant to Rhizoctonia. There was a positive correlation between infection by R. cerealis and R. solani. Infection of coleoptiles by R. cerealis or R. solani was significantly correlated with infection of leaves. No correlation between intensity of sharp eyespot on triticale plants in the field and on seedlings in controlled conditions was found.

Opis:
Salt stress is one of the most important factors limiting the growth and yield of plants around the world. However, silicon can reduce the harmful effects of salt stress on plants. For this purpose, an experiment was conducted in a factorial arrangement on randomized complete block design with three replications in a research greenhouse on the Satureja hortensis medicinal plant. Experimental treatments consisted of two salinity levels (control and 100 mM) and potassium silicate (Si) at three levels (0, 1, and 2 mM). The results showed that salinity reduced shoot dry weight, photosynthetic pigments and potassium content of shoot. However, sodium, proline, MDA, and H2O2 contents in shoot increased. The highest shoot dry weight, photosynthetic pigment content, proline, RWC, and the lowest content of MDA and H2O2 of the shoot were observed with Si application under salt stress and non-salt stress conditions. The highest yield of essential oil was also observed with Si application under salt stress and non-salt stress conditions. Therefore, the use of silicon in salt stress condition not only minimizes the harmful effects of salt stress by increasing the K+/Na+ ratio and improving the morphological and physiological traits of the Satureja hortensis medicinal plant but also improves the essential oil yield of this medicinal plant in salt stress and non-salt stress conditions.

The purpose of the present study was to determine whether water deficit affects cold hardiness of rhododendron ‘Catawbiense Boursault’, ‘Lee’s Dark Purple’, ‘Prinz Karneval’ and ‘Old Port’ shrubs. Plants were grown in unheated greenhouse and for 14 weeks from June to mid-September were subjected to six irrigation treatments. In the end of September shrubs were left in an unheated greenhouse or planted into the ground and at the beginning of each month from December to March freezing tolerance tests were performed. The results showed that in all rhododendron cultivars the highest cold hardiness was noted in January and February, lower in March but the lowest in December. Application of four-week water deficit period during summer especially between the first and the second vegetative growth may improve the frost resistance of Rhododendron shrubs.
Badania mrozoodporności liści krzewów różanecznika były prowadzone w dwóch sezonach badawczych (od grudnia 2013 roku do marca 2014 roku oraz od grudnia 2014 roku do marca 2015 roku) w Instytucie Ogrodnictwa, w Skierniewicach. Celem przeprowadzonych badań było określenie następczego wpływu regulowanego deficytu nawadniania (RDI) na mrozoodporność krzewów różanecznika odmian ‘Catawbiense Boursault’, ‘Lee’s Dark Purple’, ‘Prinz Karneval’ oraz ‘Old Port’. W poprzednich sezonach wegetacyjnych rośliny były uprawiane w nieogrzewanej szklarni. Podczas 14 tygodni, od czerwca do połowy września, krzewy były nawadniane zgodnie z sześcioma wariantami nawodnieniowymi (T1–T6) wyznaczonymi na podstawie wartości ewapotranspiracji (ETp). Trzy z nich obejmowały jednakowe, stałe nawadnianie roślin: 1 ETp (rośliny kontrolne, dobrze nawodnione – T1), 0,75 ETp (średni deficyt wody – T2), 0,5 ETp (duży deficyt wody – T3). Pozostałe trzy warianty obejmowały zróżnicowane nawadnianie w czasie trzech faz wzrostu roślin: 1–0,5–1 ETp – silny deficyt wody w fazie II (T4), 1–0,25–1 ETp – bardzo silny deficyt wody w fazie II (T5) i 0,5–1–0,5 ETp – duży deficyt wody w fazach I oraz III, i dobrze nawodnione rośliny w fazie II (T6). Fazy nawadniania I i III (każda trwająca po 5 tygodni) odpowiadały wegetatywnej fazie rozwojowej krzewów. Z kolei faza II nawadniania, trwająca 4 tygodnie, odpowiadała fazie inicjacji kwitnienia oraz tworzenia pąków kwiatostanowych. W końcu września odmiany ‘Lee’s Dark Purple’ i ‘Prinz Karneval’ posadzono do gruntu, a ‘Catawbiense Boursault’ i ‘Old Port’ pozostawały w zimie w szklarni. W obydwu sezonach badawczych na początku każdego miesiąca, od grudnia do marca, były przeprowadzane badania mrozoodporności liści, w których określono uszkodzenia mrozowe na podstawie wycieku elektrolitu z uszkodzonych komórek liścia. W pierwszym sezonie badawczym dla odmian ‘Catawbiense Boursault’ i ‘Old Port’ zastosowano wartości temperatury –10°C, –20°C, –25°C, –30°C, a w drugim doświadczeniu odpowiednio dla odmian ‘Lee’s Dark Purple’ i ‘Prinz Karneval’ liście poddano temperaturze –20°C, –25°C, –30°C. Wyniki badań wykazały dla wszystkich czterech odmian różaneczników, że największa mrozoodporność była notowana w styczniu i lutym, średnia w marcu, zaś najmniejsza w grudniu. Wielkość stresu wodnego istotnie wpływała na mrozoodporność krzewów u wszystkich czterech badanych odmian różaneczników. W wyniku zastosowania traktowań nawodnieniowych T2 i T3 rośliny cechowały się gorszą mrozoodpornością, z kolei użycie traktowań nawodnieniowych T4, T5 oraz T6 poprawiało mrozoodporność krzewów podczas zimy w porównaniu do kontroli.

The estimation of the protein content and amino acid composition under the influence of metal nanoparticles (Mn, Cu, Zn, Ag) for seven species of aquatic macrophytes: Limnobium laevigatum (Humb. & Bonpl.ExWilld.), Pistia stratiotes L., Salvinia natans L., Elodea canadensis Michx., Najas guadelupensis (Spreng.) Magnus, Vallisneria spiralis L. and Riccia fluitans L. was conducted. The plants were exposed during 7 days on the experimental solutions of metal nanoparticles at the rate of 1 g of plant per 100 ml of the mixture of stock colloidal solutions of metal nanoparticles (Mn – 0.75 mg/l, Cu – 0.37 mg/l, Zn – 0.44 mg/l, Ag+, Ag2O – 0.75 mg/l) diluted 200 times. In the five investigated species, reduction of the protein content was observed. However, this indicator remained stable only in P. stratiotes (52 mg/ml) and, conversely, increased in V. spiralis (46 mg/ml to 51 mg/ml). The content of the studied amino acids in N. guadelupensis decreased by 46% (from 112.05 mmol/g to 60.15 mmol/g), in R. fluitans – by 44% (from 104.06 mmol/g to 58.25 mmol/g), in S. natans – by 23% (from 90.08 mmol/g to 69.59 mmol/g), in E. canadensis – by 10% (from 143.92 mmol/g to 129.4 mmol/g), and in P. stratiotes as well as in L. laevigatum – by 8% (from 210.65 mmol /g to 193.77 mmol/g and with 155.0 mmol/g to 142.60 mmol/g), but in V. spiralis, on the contrary, increased by 7% (from 91.31 mmol/g to 97.59 mmol/g). Changes in the composition and content of amino acids for each species of aquatic plant were analyzed. It was suggested that the studied plants, which belong to different families, have different defense mechanisms, according to which the amino acid composition of plants varies.

Opis:
Carrot production of valuable carotenes, carbohydrate and protein are hindered by elevated salinity levels in many parts of the world. To assess this problem, germination and growth of two carrot cultivars (Daucus carota cvs Jordan and Napoli) were studied in vivo and in vitro under different salt stress concentrations (0, 75, and 150 mM NaCl). Seeds were directly or gradually exposed to these salt concentrations. With elevated salinity levels, significant reductions in growth parameters (dry shoot weight, fresh shoot weight, shoot length, root length, and root number) were observed. Also, significant difference in germination percentage was observed at 150 mM NaCl in both cultivars when compared with control treatment (90% germination percentage in Napoli and 71% in Jordan cultivar). Growth rate, tolerant index, and relative water content (RWC) declined as salinity increased. The 150 mM NaCl salinity treatment significantly reduced the shoot chlorophyll and protein content, but increased carbohydrate content. Lesser impairment by the gradual exposure of seedling to salinity provides an opportunity to study the acquirement of salt tolerance.

A factorial experiment was performed to evaluate whether wild and primitive Aegilops and Triticum species may be used in wheat breeding as donors of an improved water use efficiency (WUE) and/or tolerance to nutrient shortage. Seventeen lines representative for Aegilops and Triticum species of different origin, ploidy level and genomic structure were compared with three local cultivars of hexaploid wheat T. aestivum. The genotypes were grown till maturity in experimental pots (9 dm3) under high and reduced NPK nutrition. There was a broad genetic variation in the response to nutrient shortage and efficiency of water use in the vegetative and grain mass formation. The variation was dependent upon species, ploidy level and genome present. Results suggest that a search for enhanced tolerance and novel variation in WUE among wild or primitive wheats may be essential for wheat breeders. The tetraploids T. carthlicum, T. dicoccoid.es and T. timopheevii and the hexaploid T. sphaerococcum were found to be the most promising potential sources of stress tolerance. However, only the primitive T. sphaerococcum appeared to be a valuable donor of improved WUE. Despite a high operative heritability of WUE, testing till plant maturity over diverse levels of soil nutrient status would rather be necessary for a precise discrimination of the most efficient genotypes as indicated by the genotype-stage and genotype-nutrition interactions.
Przeprowadzono doświadczenie wazonowe w celu określenia czy dzikie i prymitywne gatunki Aegilops i Triticum mogą być wykorzystane w hodowli pszenicy jako donory efektywniejszego wykorzystania wody (WUE) i/lub tolerancji na niedobór składników pokarmowych. Siedemnaście linii reprezentujących gatunki Aegilops i Triticum o różnym pochodzeniu, ploidalności i strukturze genomowej porównano z trzema krajowymi odmianami pszenicy heksaploidalnej T. aestivum. Genotypy rosły do stadium dojrzałości w wazonach doświadczalnych (9 dm3) przy wysokim i obniżonym nawożeniu NPK. Obserwowano szeroką zmienność genetyczną w reakcji na obniżone nawożenie i efektywności wykorzystania wody w tworzeniu masy wegetatywnej i plonu ziarna. Zmienność ta była zależna od gatunku, poziomu ploidalności i obecności danego genomu. Wyniki sugerują, że poszukiwanie zwiększonej tolerancji i nowej zmienności w WUE wśród dzikich i prymitywnych pszenic dla celów hodowlanych jest uzasadnione. Tetraploidalne T. carthlicum, T. dicoccoides i T. timopheevii oraz hexaploidna Т. sphaerococcum były potencjalnie najlepszymi źródłami tolerancji stresu. Jednak tylko prymitywna T. sphaerococcum okazała się cennym źródłem zwiększonej WUE. Obserwowane interakcje typu genotyp-stadium i genotyp-nawożenie wskazują, że pomimo wysokiej odziedziczalności WUE, dla precyzyjnego doboru najefektywniejszych genotypów raczej niezbędne będzie testowanie roślin uprawianych aż do stadium dojrzałości przy odmiennych poziomach żyzności gleby.

Plant glutathione peroxidases are an important class of enzymes which play key roles in the stress adaptability of plants both in context of biotic and abiotic stress pathways. They have been over the years much studied in animals since the catalytic residues are comprised of selenocysteine a variant amino acid which is ribosomally encoded with the help of an RNA structural element known as SECIS. Various workers over the years have shown that plant glutathione peroxidases play active roles in ROS sequestration, lipid hydroperoxidation as well as regulate glutathione levels. However, each plant has various patterns of glutathione peroxidase expression and action and in some plants certain isoforms have not been detected at all. This work focuses on the prediction and identification of single nucleotide polymorphisms (SNPs) and INDELs in the coding regions of plant glutathione peroxidases, with the help of a Bayesian based algorithm subsequently validated. A large number of informative sites were detected 279 of which had variant frequency of ≥ 50 %. This data should be beneficial for future studies involving genetic manipulation and population based breeding experiments.

As the impact of global climate change increases, the interaction of biotic and abiotic stresses increasingly threatens current agricultural practices. The most effective solution to the problem of climate change and a decrease in the amount of atmospheric precipitation is planting extremely drought-resistant and high-yielding crops. Sorghum can grow in harsh conditions such as salinity, drought and limited nutrients, also it is an important part of the diet in many countries. Sorghum can be introduced in many zones of Kazakhstan. Plant height and yield of green plant biomass of 16 sorghum samples in arid conditions were determined based on a set of agrobiological characteristics for field screening. The height of the studied samples of grain sorghum was 0.47 ±0.03 m, and the height of sweet sorghum was much longer, reaching up to 2.88 ±0.12 m. Also, there was a strong difference in green biomass in cultivated areas under different soil and climatic conditions, the green biomass of sweet sorghum was 3.0 Mg∙ha-1, and in grain sorghum, it reached up to 57.4 Mg∙ha-1. Based on the data of the field assessment for various soil and climatic conditions, the following samples were identified for introduction into production: samples of sweet sorghum for irrigated and rainfed lands of the Almaty Region and in the conditions of non-irrigation agriculture of the Aktobe Region - a promising line ICSV 93046. For non-irrigation agriculture of the Akmola Region, genotypes of sweet and grain sorghum are ‘Chaika’, ‘Kinelskoe 4’ and ‘Volzhskoe 44’.

Opis:
In order to evaluate morphological and physiological traits related to drought tolerance and to determine the best criteria for screening and identification of drought-tolerant genotypes, we grew two tolerant genotypes (MCC392, MCC877) and two sensitive genotypes (MCC68, MCC448) of chickpea under drought stress (25% field capacity) and control (100% field capacity) conditions and assessed the effect of drought stress on growth, water relations, photosynthesis, chlorophyll fluorescence and chlorophyll content in the seedling, early flowering and podding stages. Drought stress significantly decreased shoot dry weight, CO2 assimilation rate (A), transpiration rate (E), and PSII photochemical efficiency (Fv/Fm) in all genotypes. In the seedling and podding stages, PSII photochemical efficiency was higher in tolerant genotypes than in sensitive genotypes under drought stress. Water use efficiency (WUE) and CO2 assimilation rate were also higher in tolerant than in sensitive genotypes in all investigated stages under drought stress. Our results indicated that water use efficiency, A and Fv/Fm can be useful markers in studies of tolerance to drought stress and in screening adapted cultivars of chickpea under drought stress.

Opis:
Tolerance to a new herbicide, pyributycarb, was evaluated both at the plant and cellular levels. Several highly or moderately tolerant strains chosen at the plant level, showed a parallel relation of to tolerance at the cellular level. However, on the whole, correlation between total tolerance indices and survival rates of calli was not significant in 18 out of the 80 studied strains. As a result of somaclonal selection for two herbicides, lines NB-200 and NM-100 were regenerated from the tolerant calli screened with benthiocarb at 200 ppm, and molinate at 100 ppm, respectively. In the R₂ generation, both the lines displaned a stable tolerance both at the plant and cellular levels. Thus the highly tolerant mutant lines were developed from a moderately tolerant line, N-61, via in vitro selection. To achieve a short-cut method in the interspecific genetic exchange, a series of techniques related to cell fusion were established in rice and related species. Two kinds of somatic hybrids between the cultivar Kitaake and tetraploid Oryza species, O. punctata and O. officinalis, were successfully produced. Among the somatic hybrid plants, a wide range of chromosomal variation was observed. Aneuploid plants with a chromosome number around 2n = 72 (hexaploid), which are expected from a symmetric fusion between diploid and tetraploid strains, were obtained showing mixoploidy within a plant. Most of the somatic hybrids were characterized by intermediate features of plant-type showing high sterility, shattering of spikelets and reduced plant height. As an exception, a diploid plant, which was identified by RFLP analysis using the rDNA gene probe, closely resembled Kitaake and produced viable seeds. A tetraploid hybrid plant was also promising for the introduction of economically important characters through the reduction of chromosome numbers by doubled haploids. Gametoclonal variation and gamma radiation was applied to Kitaake. The mutation frequency was prominently increased by gamma ray treatment, especially at high doses of 200 Gy or 300 Gy. In the M₃R₂ or M₄R₃ generations, most of the variants showed unfavourable characters. Most of the mutant characters were governed by single or double recessive genes. Several mutants such as short culm and early flowering time might be used for rice breeding.
