Opis:
The main objective of the study was to evaluate the effect of added 0.3% (w/w) oat β-glucan (OG) in set-type yogurt on its protein digestion using an in vitro gastrointestinal model. During gastric digestion phase, the amount of soluble proteins and peptides increased to 25% and 40% for control yogurt (yogurt without OG) and 0.3% OG yogurt, respectively. Buccal digestion has little effect on the structure of yogurts, while large spherical vesicles were formed for both control yogurt and 0.3% OG yogurt after gastric digestion. The presence of 0.3% OG promoted the hydrolysis of yogurt in the gastric digestion phase and caused higher antioxidant activity. Compared with that of control yogurt, the inhibition of cholesterol solubility of 0.3% OG yogurt showed no differences after buccal digestion but significantly higher after gastrointestinal digestion (21.3% for gastric and 22.7% for intestinal digestion). Overall, this study enhances the understanding of digestion characteristics of 0.3% OG-fortified set-type yogurt and provides a theoretical basis for the development of this kind of dairy products.

Background: Cardiovascular diseases with the accompanied elevated level of total cholesterol have been a major problem in society for the last several decades. They belong to the diseases of civilization which affect people at an increasingly young age. For this reason, our aim was to investigate whether the concentrations of selected steroids are related to elevated total cholesterol in people without diagnosed cardiovascular diseases. Material and methods: The study involved 71 plasma samples. 19 of them were obtained from women and men with elevated cholesterol levels, whereas 52 samples were from healthy volunteers (control group). Liquid chromatography coupled with mass spectrometry (LC-MS) validated method followed by solid-phase extraction procedure were applied to measure the plasma concentrations of the three endogenous glucocorticosteroids (cortisol, corticosterone and cortisone). Results: Statistically significant differences between the concentration of cortisol were noted among healthy women and women with elevated cholesterol. The measured concentrations of cortisol in healthy women and men are comparable, 111.19 ng/mL and 112.22 ng/mL. respectively. However, the concentrations of cortisol in the elevated cholesterol group was significantly lower among women with elevated cholesterol than in healthy women (74.13 ng/mL and 111.19 ng/mL respectively). The concentration of cortisol for men with elevated cholesterol was 38.60 ng/mL. Hence, it is much higher than in women with elevated cholesterol and higher than in the case of healthy men. Distinctive changes can be observed also for corticosterone measured for both women and men. Conclusions: The observed differences on the level of steroids between healthy control group and patients with elevated cholesterol can be considered as worthy of further investigation from both biochemical as well as clinical points of view.

Introduction. Lipid is an essential component of the cell and its organelles membrane. The uniqueness and selectivity of lipids to specific functions and asymmetry of lipid distribution in the organelle’s membrane give the cell ability of being highly qualified and specified. Aim. The paper provides a comprehensive review of membrane lipids in different tissues and organelles of the cell in norm and disease. Material and methods. The paper analyzed the present literature data on membrane lipids behavior in physiology and pathology. Analysis of the literature. The major structural and functional lipids of the cell membrane are phosphatidylcholine > phosphatidylethanolamine. The absence/deficiency or augmentation of a specific type of lipid results in serious defects and usually life-threatening with a permanent disability. The observations discussed here suggest, the lipid peroxidation severity depends on the membrane lipid composition of the cell. Some tissue cells can handle lipoperoxidation and protect themselves from the peroxidation damaging products better, while other cells cannot compensate. Therefore, some organs are highly sensitive to peroxidation and irreversible changes occur rapidly. Conclusion. To sum up, the understanding of lipid’s role in norm and disease is clinically crucial to evaluate a novel therapeutic target to treat many metabolic disorders such as metabolic syndrome and some lysosomal storage disorders via targeting specific new signaling pathways, lipid molecules, and enzymes

ObjectivesThis observational follow-up study investigated the associations of nutrition and body composition with cardiovascular disease (CVD) risk factors, including pro-inflammatory biomarkers, in soldiers during a 6-month deployment.Material and MethodsThirty-five male soldiers were assessed at months 0, 3 and 6, and their parameters, i.e., M±SD, were as follows: age 30.0±8.7 years, height 179±6 cm, and BMI 24.2±2.5 kg/m2. Three-day food diaries were used for monitoring macronutrient intake. Body composition was estimated using bioimpedance. Fasting blood samples for lipids and pro-inflammatory biomarkers were collected, and blood pressure measurements were performed.ResultsCarbohydrate intake increased and protein intake decreased at month 3 (p = 0.034, p < 0.001), while body composition remained stable. Systolic blood pressure increased at month 6, while other CVD risk factors remained within the reference values. Fat mass and body fat percentage were associated positively with total and low density lipoprotein (LDL) cholesterol concentrations at all measurement points. A negative association was found between the change in fiber intake vs. the change in total (r = –0.36, p = 0.033) and LDL cholesterol (r = –0.39, p = 0.019).ConclusionsLower fiber intake and a greater amount of body fat were associated with high total and LDL cholesterol concentrations. Nevertheless, the measured CVD risk factors remained within the reference values, except for the higher systolic blood pressure. A regular screening of body composition and a higher consumption of fiberrich foods may promote cardiometabolic health in soldiers.

Introduction and objective. Hyperlipidemia is considered as a serious communal problem in developed countries, caused by an excess level of cholesterol in blood circulation. It leads to chronic illness and even death in human beings. As the currently available drugs cause unexpected side-effect, the aim of this study is to concentrate on naturally occurring flavonoids which can potentially provide defensive and therapeutic effects in atherosclerosis diseases, and investigate the hypolipidemic effect of rutin on Triton WR-1339 triggered hyperlipidemia in a rat blood sample. Materials and method. Rats were randomly prearranged into five different groups of five rats each. Group-I was the non-disease control and administered normal saline. Group-II was the atherogenic control, administered Triton WR 1339 (200 mg/kg BW). Group-III was standard and received Atorvastatin. The last two groups (IV, V) were tested (I&II) by administering administered Rutin (40 mg/kg, 80 mg/kg) orally. The test material (I&II) and the standard drug were administered for seven days. After the last dose, blood samples were collected and the lipid levels estimated in the blood samples. Results. Rats treated with rutin flavonoid at a dose of 40 mg/kg & 80mg/kg exhibited a reduction in Total Cholesterol, Triglycerides, Low Density Lipoprotein (LDL) and Very Low Density Lipoprotein (VLDL). Rutin also increases the High Density Lipoprotein (HDL), compared with control rats. Rutin treated rats exhibited dose-dependent hypolipidemic activity. The protection percentage of rutin against hyperlipidemia was observed as 41.89%, 55.57% whereas the atorvastatin treated group protection was observed at 60.63%. Conclusions. The results of the study revealed that rutin showed a significant hypolipidemic effectiveness on Triton WR- 1339 induced hyperlipidemia in rats.

The present study aimed to determine the role of job components and individual parameters on the raised blood pressure among male workers of textile industry who were exposed to continuous high noise level. Information of all eligible subjects including demographic and individual characteristics, medical history and job characteristics were obtained by direct interview and referring to the medical records. All blood pressure measurements were done using mercury sphygmomanometer in the morning before work. The 8-hours equivalent A-weighted sound pressure level, the level of blood cholesterol and triglyceride, and noise annoyance was determined for each worker. As the result of weighted regression in path analysis (direct effect), only the work shift did not have a significant effect on blood pressure among the studiem variables. It can be seen that variables including the level of triglyceride, cholesterol, and noise exposure have the most direct effects on blood pressure. The results of total effects showed that variables, including using the hearing protection device, age, work experience and visibility of sound source, did not have a significant effect on blood pressure. The results of this study indicate that occupational noise exposure alone and combined with other job components and individual parameters is associated with raised blond pressure. However, noise exposure was probably a stronger stressor for increased blood pressure.

Long-term exposure to hypercholesterolemia is the cause of atherosclerosis, which in turn causes cardiovascular and cerebrovascular events. In developed countries, including Poland, vascular diseases are the main cause of death. They affect an ever younger part of the population, including the working population. The authors address the problem of epidemiology of cardiovascular diseases, unsatisfactory detection and treatment, economic consequences for the health care system, and the possibilities of using occupational medicine services in the prevention of this health problem. Due to the fact that the early detection of diseases caused by high blood cholesterol levels is relatively low in Poland, obligatory occupational medicine examinations seem to be a key element of the second-line prevention. Therefore, it seems natural to consider the idea of extending the scope of obligatory examinations and introducing tests that allow lipid disorders to be detected at an early stage. This can contribute to a general improvement of the health of the population, and to economic benefits, such as a decrease in the costs of treatment of the disorders that have been detected too late. Broadening the scope of occupational examinations is also important from the perspective of public health and epidemiology of cardiovascular diseases, thus being an element of prevention of civilization diseases. It means improving health and building health awareness, and it should translate into regular health examinations. The performance of these examinations should result not only from the obligation, but also from the patient’s conviction about the importance of early detection of disorders, including lipid disorders, for an effective therapy. Introduction: Common ivy (Hedera helix) is a plant used successfully in the treatment of various ailments. This is possible owing to the unique set of substances contained in it such as large amount of saponins, flavonoids, phenolic acids and phytosterols as well as polyacetylenes and coumarins. All these substances have been used in the production of cosmetic and medicinal preparations. Clinical trials on the efficacy and safety of dry ivy leaf extract have shown its high efficacy, comparable to synthetic medications, and better tolerance of herbal drugs. Objective: Investigations were performed on model ivy leaf (Hedera helix) extracts which were created using a medium of diversified polarity (ԐM). Measurements of viscosity and surface activity on phase boundary were performed. During preformulation studies attention was drawn to the possibility of micellar solubilization of cholesterol and selected structures of nonsteroidal anti-inflammatory drugs (NSAIDs) – ketoprofen. Methods: Viscosity measurements of Extractum Hederae helicis e folium aqueous solutions and in 0.1 mol HCl were performed according to the Polish Standard with Ubbelohde dilution viscometer. The surface tension of aqueous solutions – Δγ25 of Hederae helicis e folium extracts was determined according to the Polish Standard with stalagmometric method . Critical micellar concentration (cmc) was calculated. This enabled to evaluate the dependence ΔG0 =2.303 RT× log cmc of the thermodynamic potential of micelle formation (ΔG0 ). Results: It has been confirmed in the conducted comparative studies that aqueous solutions of Extr. Hederae helicis e folium created with maltodextrin as well as with SiO2 – maltodextrin result in micellar solubilization. The increase of granulometric size of cholesterol particle to Ø 1.60 mm decreased the amount of solubilized cholesterol but solubility preferences of the extracts were maintained. Conclusions: Model extracts produced from Hederae helicis e folium with diversified polarity of the extraction medium (water – ethanol) are characterized by appropriate solubility of the components which results not only from the presence of chlorophyll and its derivatives in the extract but also from the technique used for spray drying of the extract.

Celem niniejszej pracy było określenie zdolności Lactobacillus rhamnosus ATCC 9595, Lactobacillus casei ATCC 334 oraz szczepów bakterii fermentacji mlekowej wyizolowanych z kału zdrowych dzieci do wiązania cholesterolu w środowisku modelowego soku jelitowego i podłoża MRS Bulion. Do oznaczenia pozostałości cholesterolu w próbkach zastosowano diagnostyczny zestaw enzymatyczny Cholesterol RTU. Hodowle prowadzono w 24-dołkowych płytkach titracyjnych. Pomiary absorbancji przy oznaczaniu stop- nia wiązania cholesterolu oraz OD600 wykonano przy użyciu czytnika mikropłytek Spark 10M. Szczepy wykazujące największy stopień wiązania cholesterolu oceniono pod względem zdolności wzrostu oraz wiązania cholesterolu ze środowiska przy zwiększającym się obciążeniu cholesterolem [g/dm3 ]: 1,66, 2,85 i 3,75. L. rhamnosus i L. casei wykazywały zdolność do wiązania cholesterolu z podłoża MRS Bulion odpowiednio w 68,18 i 83,6 %, natomiast w modelowym soku jelitowym osiągały odpowiednio 60 i 50 %. Szczepy wyizolowane z kału dzieci charakteryzowały się dużą zmiennością w stopniu wiązania choleste- rolu ze środowiska, przy czym lepsze rezultaty osiągnięto w przypadku modelowego soku jelitowego (1,81 ÷ 77,99 %) niż podłoża MRS Bulion (0,49 ÷ 33,14 %). Przy zwiększającej się zawartości cholestero- lu w środowisku badane szczepy wykazywały zmniejszony przyrost biomasy, co korelowało ze zmniej- szonym stopniem wiązania tego związku. Największą redukcję cholesterolu z modelowego soku jelitowe- go uzyskano przy jego stężeniu – 1,66 g/dm3 . Stężenie cholesterolu 3,75 g/dm3 skutkowało zahamowaniem przyrostu biomasy, a tym samym najmniejszym ubytkiem tego związku ze środowiska.

Opis:
Cholesterol to cząsteczka zbudowana z 17-węglowej struktury cyklopentanoperhydrofenantrenu i dołączonego do niej 6-węglowego łańcucha bocznego, klasyfikowana jako alkohol steroidowy ze względu na obecność pojedynczej grupy hydroksylowej. Główną funkcją cholesterolu jest zależna od temperatury modulacja płynności błon komórkowych. Duża ilość cholesterolu występuje w nanodomenach błonowych (tratwach lipidowych i kaweolach), które pełnią ważną rolę w procesie endocytozy i przekaźnictwie międzykomórkowym. Ponadto, jest on prekursorem hormonów steroidowych produkowanych przez gonady i korę nadnerczy oraz warunkuje właściwy przebieg rozwoju embrionalnego. Poza pełnieniem istotnej roli fizjologicznej, cholesterol może przyczyniać się do rozwoju wielu stanów patologicznych, wynikających zarówno z nagromadzenia jego cząsteczek w ustroju, jak i zaburzeń ich metabolizmu. Jego udział opisano m.in. w rozwoju schorzeń neurodegeneracyjnych, chorób układu krążenia, chorób nerek oraz chorób nowotworowych. Obecnie prowadzone badania zmierzają do opracowania nowych strategii terapeutycznych, pozwalających na skuteczniejszą i bezpieczniejszą kontrolę poziomu cholesterolu i regulację jego metabolizmu, oraz mających na celu wykorzystanie cholesterolu jako składnika nowych, skuteczniejszych leków.
Cholesterol is a molecule build of 17-carbon cyclopentano-perhydro-phenanthrene structure and 6- carbon side chain, classified as steroid alcohol due to the presence of a single hydroxyl group. The main function of cholesterol is temperature-dependent modulation of cell membrane liquidity. Large amounts of cholesterol are found in membrane nanodomains (lipid rafts and caveolae) that are essential for endocytosis and intercellular signaling. Moreover, cholesterol is a substrate in steroid hormones biosynthesis in gonads and adrenal glands, and determines the proper course of embryonic development. Besides its physiological role, cholesterol may contribute to pathogenesis of different diseases, resulting from its accumulation in the system or from metabolic disorders. The significance of cholesterol has already been described in several neurodegenerative disorders, cardiovascular and renal diseases, and in cancer. Therefore, current research focus on providing some new therapeutic strategies, allowing for cholesterol level control, regulation of its metabolism, or for using cholesterol molecules as effective drug component.

Introduction and objective. There are some clinical situations where a high level of HDL cholesterol (HDL-C) may be unfavourable. In these situations, HDL-C may undergo some changes, and even if its quantity is within the reference range, its quality is no longer the same. Brief description of state of knowledge. Diabetes is the state of elevated oxidative stress. Studies conducted to-date have revealed an increased production of the reactive forms of oxygen as the result of tissue damage in diabetes patients. The expression ‘dysfunctional HDL’ has been coined in the literature to describe high-density lipoproteins that lose their antioxidative and anti-inflammatory properties, that is, HDL-C that loses its basic functions. Recent observational studies have confirmed that the atheroprotective activity of properly functioning HDL-C is frequently impaired in clinical situations associated with oxidative stress. The presented review lays the foundation for a new approach to understanding how the functional properties of HDL help reduce cardiovascular risk. Conclusions. In the light of presented findings it seems that there is a need to seek a better diagnostic marker than HDL-C level. This study presents some possible directions for future research to bring us closer to the full understanding of the HDL particle and its role in patients with ischemic heart disease and type 2 diabetes.
