Opis:
Acoustical attenuation spectra in the frequency range 12 kHz – 2 GHz and nonequilibrium time domain measurements are briefly reviewed for aqueous solutions of various mono- and disaccharides as well as alkyl glycosides. Several relaxation regimes emerge with relaxation times between 10^-11 s and 10^3 s. In this paper relaxation terms reflecting conformational changes are discussed, particularly mutarotation (10^3 s), chair-chair ring inversion (1 ąs), two modes of pseudorotation (100 ns, 10 ns), disaccharide ring isomerisation (10 ns), and exocyclic side group rotation (1 ns).

Concentrations of total nonstructural carbohydrates (TNC), soluble carbohydrates, starch, sucrose, glucose, fructose, raffinose, galactose, stachyose, mannitol and specific activities of soluble acid (AI) and neutral (NI) invertases, sucrose synthase (SuSy), hexokinase (HK), fructokinase (FK), glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and glucose 6-phosphate dehydrogenase (G6PDH) were analyzed in fine roots of Populus deltoides Bartr. ex Marsh growing at a polluted site (near copper smelters) and a control site (free from heavy pollution). Also chemical properties of the soil from both sites were assessed. In comparison with the control, fine roots from the polluted site contained greater concentrations of TNC, soluble sugars, starch and sucrose but less hexoses, so they had higher values of sucrolysis index (sucrose/hexoses). The activity of AI, NI and SuSy declined insignificantly, while specific activities of HK, FK, GAPDH and G6PDH were significantly inhibited. The results suggest that a long-term heavy metal stress leads to an accumulation of carbohydrates and altering activities of glycolysis and the oxidative pentose phosphate pathway in fine roots.

Results from broadband acoustical spectroscopy for aqueous solutions of mono- and disaccharides with salts added, of various alkyl glycosides, and of α-cyclodextrin with n-octyl-β-D-glucopyranoside added are briefly summarized in view of their relevance in the study of molecular association and complexation processes. Mono- and tridentate complexes of alkali earth ions with saccharides are discussed as well as the monomer exchange between micellar structures and the suspending phase in the alkyl glycoside surfactant solutions. Particular attention is given to the behaviour at solute concentrations close to the critical micelle concentration or aggregate concentration, respectively. Also described is the competition between inclusion complex formation and self-aggregation in solutions containing cyclodextrin and alkyl glycoside surfactant.

Neoplastic transformation is often associated with characteristic changes in the expression of the sialyl Lewisa and sialyl Lewisx antigens, representing typical tumor-associated carbohydrate antigens. High amounts of sialyl Lewisa are present in human adenocarcinomas of the colon, pancreas and stomach. A growing amount of data suggests that this carbohydrate structure is the ligand for E-selectin. Sialylated Lewis structures present on the surface of tumor cells are carried by the carbohydrate chains of glycoproteins and glycolipids. There are several lines of evidence showing that sialyl Lewisa is responsible for the adhesion of human cancer cells to endothelium. E-selectin present on endothelial cells mediates these interactions. Selectins and their carbohydrate ligands can thus play an important role in the selective homing of tumor cells during metastasis. However, the presence of sialyl Lewisa antigen on the surface of tumor cells and their adhesion to E-selectin-expressing cells in in vitro adhesion assay by itself can not be directly related to metastatic properties of all cancer cells.

The aim of this 3-year study was to determine the influence of harvest time on dry matter, total dietary fibre, fractional composition of dietary fibre (acid detergent fibre (ADF) and neutral detergent fibre (NDF)), water-soluble carbohydrates (WSCs) and inulin content in tubers of different cultivars of Jerusalem artichoke (Helianthus tuberosus L.): ‘Rubik’, ‘Albik’ and ‘Sauliai’. The tubers were harvested in spring (March and April) and autumn (October and November). Our study shows that the largest amount of WSC to exist in ‘Rubik’ tubers harvested especially during October. In contrast, the largest amount of total dietary fibres and their fractions (NDF and ADF) accumulated in ‘Albik’ tubers during the spring harvest month (April). The largest amount of inulin was obtained from ‘Sauliai’ tubers harvested in October. Based on our results, it is advisable to use the ‘Sauliai’ tubers from the harvest time during October to obtain the largest amount of inulin.

With the improvement of people’s living standards and rapid economic development, the incidence of diabetes mellitus (DM) is increasing in most parts of the world. DM presents an important potential threat to human health. In the present study, a model of diabetes in female mice was established, and fasting blood glucose was detected at week 4, after which the biochemical profiles were evaluated by histopathological analysis. The success rate of modeling in the normal control (NC) group and the low/ middle/high-dose streptozotocin (STZ) group were 0, 0, 25% and 60%, respectively. In the middle-dose and high-dose STZ groups, the liver index was increased significantly compared with the NC group (p<0.05). The blood biochemical indicators of total cholesterol and low density lipoprotein cholesterol in three STZ injection groups were as follows: alanine aminotransferase and aspartate transaminase in middle- and high-dose STZ groups, high-density lipoprotein cholesterol and serum creatinine in the high-dose STZ group, and blood urea nitrogen in the middle-dose STZ group were significantly increased (p<0.05). The level of total triglycerides was lower, obviously, in the high-dose STZ group than in the NC group (p<0.05). The mice showed marked steatosis, green-dyed fiber tissue coloring in varying degrees, and the contour of the hepatic lobules basically disappeared in STZ injection groups. The results suggest that to establish a diabetes model for female ICR mice, the optimum dose of STZ is 100 mg/kg.

Seasonal changes in the levels of water-soluble carbohydrates, free proline and tolerance to freezing and water deficit were studied in leaves of cherry laurel (Prunus laurocerasus) and ivy (Hedera helix). Frost and water deficit tolerance was estimated based on the measurement of electrical conductivity of aqueous media containing leaf discs that were previously treated with frost (–7°C) or polyethylene glycol solution (PEG 600), respectively. Carbohydrate content in leaves of examined species changed differently throughout the measurement period. In laurel leaves the highest carbohydrate level was found in February, while it was lowest in June. In contrast, in ivy leaves the highest carbohydrate content was recorded during summer (June, July), while the lowest in February. However, a lack of correlation between soluble carbohydrate level and membrane injury index was shown in this study. Free proline content in leaves of investigated species was the highest in early spring, i.e. April. The lowest level of free proline in laurel leaves was found in July, September and October, but in ivy from July to February. A negative correlation between proline content and membrane injury index observed in frost and PEG treated leaves of both species suggest that this amino acid may have resulted on membrane protection.

Introduction: The role of cross-reactive carbohydrate determinants (CCDs) in diagnostics of occupational allergy remains unclarified and its clinical relevance is still questioned. The aim of the study was to assess the frequency of positive response to CCDs in the subjects with suspected occupational allergy and the relationship between other diagnostic test results and final diagnosis. Material and methods: The study group included 201 patients. They underwent clinical examination, skin prick test (SPT) to common and occupational allergens, specific serum immunoglobulin (sIgE) determinations, spirometry and specific inhalation challenge test. Moreover, sIgE to CCDs from bromelain was assessed in all subjects. Results: Occupational respiratory allergy was recognized in 64.3% of CCD-positive and 52.4% of CCD-negative patients. Positive SPT results to common and occupational allergens were found in 64.3% and 35.7% of CCD-positive subjects, respectively. In all subjects with CCDs, the sIgE to grass pollens as well as to occupational allergens were detected. The total IgE level > 100 kU/l was significantly associated with the presence of sIgE to CCDs. Conclusions: sIgE to CCDs were found in 7% of subjects suspected to suffer from occupational respiratory allergy. The presence of CCDs is not significantly associated with occupational respiratory allergy. It is also not more frequent in subjects reporting work-related respiratory symptoms in whom occupational allergy was not confirmed. The elevated total IgE level was related with CCD positivity. In patients with suspected occupational allergy, the presence of sIgE to CCDs in serum did not indicate the irrelevance of positive sIgE to occupational allergens.

Germinability and the content of soluble carbohydrates were analysed in cereal seed (winter rye, cv. Warko; spring wheat, cv. Santa; hexaploid winter triticale, cv. Fidelio and cv. Woltario). Seed moisture content (mc) was equilibrated over silica gel to 0.08 g H2O/g dry mass and stored in a desiccator at 20oC for up to 205 weeks or were equilibrated to mc 0.06, 0.08 or 0.10 g H2O/g dm and subjected to artificial aging at 35oC in air-tight laminated aluminium foil packages for 205 weeks. It was shown that the rate of seed aging depended on the species and seed moisture content. The fastest decrease of germinability upon storage was observed in seed with the highest moisture level. Complete germinability loss for winter rye, winter triticale cv. Fidelio, winter triticale cv. Woltario and spring wheat seed with mc 0.10 g H2O/g dm3 occurred after 81, 81, 101 and 133 weeks, respectively. Fructose, glucose, galactose, myo-inositol, sucrose, galactinol, raffinose, stachyose and verbascose were the main soluble carbohydrates found in the seed. The obtained data on the contents of specific sugars and the composition of soluble sugars fraction in seed of rye, wheat and triticale did not corroborate any profound effect of reducing sugars, sucrose and oligosaccharides on seed longevity.

The differences between the metabolism and the physiology of cancer cells and the cells of the human body are assessed and used in most anticancer treatments. These differences encompass, among others, increased glucose metabolism in the changed cells. The aim of the paper was to discuss the results of studies concerning the relationship between lowcarbohydrate diets and fasting and the course of cancer. An inappropriately composed diet consisting of high amounts of simple sugars supplies cancer cells with nutrients, which may impair the effectiveness of cancer patients treatment. Lowcarbohydrate diets may, therefore, constitute an element of supplementary therapy in cancer treatment. The mechanism of low-carbohydrate diets in combination with standard treatment has not been completely explained, though. In initial studies it was proven that patients who were able to continue low-carbohydrate diets showed improvement in health and reduction of tumor mass or its slower growth. Moreover, it was observed that the inability of cancer cells to adapt in new environmental conditions that occur while fasting may have toxic effect on them. Introduction of fasting may sensitize cancer cells to chemotherapy, decrease concentration of growth factors and lead to repair of normal cells. On the other hand, fasting may also promote autophagy and, as can be concluded from the literature, its mechanism may have twofold activity: as a process impacting the survival or death of cancer cells.
W większości terapii przeciwnowotworowych oceniane i wykorzystywane są różnice między metabolizmem i fizjologią komórek nowotworowych, a komórkami ciała człowieka. Różnice te obejmują między innymi nasilony metabolizm glukozy w zmienionych komórkach. Celem pracy było omówienie wyników badań na temat związku diet niskowęglowodanowych i głodówek z przebiegiem choroby nowotworowej. Nieodpowiednio skomponowana dieta składająca się ze znacznej ilości cukrów prostych dostarcza składniki odżywcze komórkom nowotworowym, co może pogarszać skuteczność leczenia pacjentów onkologicznych. Diety niskowęglowodanowe mogą więc stanowić element terapii uzupełniającej w chorobie nowotworowej. Mechanizm działania diet niskowęglowodanowych w połączeniu ze standardowym leczeniem nie został jednak w pełni wyjaśniony. We wstępnych badaniach wykazano, że pacjenci, którzy byli w stanie kontynuować diety niskowęglowodanowe wykazywali poprawę stanu zdrowia, zmniejszenie masy guza lub jego spowolniony wzrost. Ponadto zaobserwowano, że niezdolność komórek nowotworowych do adaptacji w nowych warunkach środowiska, jakie występują podczas głodzenia, może działać na nie toksycznie. Wprowadzenie postu może uwrażliwić komórki nowotworowe na chemioterapię, zmniejszać stężenie czynników wzrostu i prowadzić do naprawy prawidłowych komórek. Z drugiej strony post może również promować proces autofagii, a jak wynika z piśmiennictwa, jej mechanizm może mieć działanie dwukierunkowe: jako proces wpływający na przeżycie lub śmierć komórek nowotworowych.

From 1994 to 1996 changes in the concentrations of dissolved mono- (MCHO) and total dissolved polysaccharides (TCHO) as well as the activities of carbohydratedegrading enzymes (α- and β-glucosidase, glucosaminidase) were investigated during mixing of water from the River Odra and the open Pomeranian Bight. This study addresses the question of whether their distribution was a result of physical dilution alone or if biological interactions were detectable. Within the salinity gradient, ranging from 1.9 to 7.8 PSU, TCHO declined from 13.2 μmol l−1 near the Świna mouth to 2.8 μmol l−1 after mixing. Concentrations of MCHO decreased from 3.4 μmol l−1 to 1.1 μmol l−1 but its distribution pattern varied more between summer and autumn than that of TCHO. The hydrolysis rate (Hr) by glucosidase and glucosaminidase activities was reduced from 13.9% h−1 to 0.3% h−1 and 9.9% h−1 to 0.2% h−1, respectively, and correlated with the uptake rate of glucose (To) by bacteria. In summer, the To/Hr ratio increased from about 1.2 to 29.4, mainly because of stronger decreases in Hr than in To. It was shown that the relationship between enzymatic release and uptake of carbohydrates influences the concentration of dissolved carbohydrates within the salinity gradient. Most probably, the decrease in carbohydrate-degrading enzymes is the result of reduced substrate stimulation and the lower number of particle-associated bacteria.

Carbohydrates were analyzed in Norway maple embryo axes and cotyledons after imbibition, in the middle of cold stratification, before germination and during radicle protrusion to 8-10 mm and 20-25 mm. Simultaneously desiccation tolerance of seeds was determined by tetrazolium (TTC) test, after desiccation of seeds to 10-20% of water content. The cotyledons were tolerant to desiccation throughout all stratification and germination period. Embryo axes became sensitive to desiccation when hypocotyls-radicle protrusion reached 20-25 mm length. In this period the significant increase of monosaccharides: glucose, fructose and galactose in embryo axes occurred. This was not observed in cotyledons. During the germination period significant decrease of sucrose and raffinose content was noted in embryo axes and cotyledons. Relatively less changes appeared in stachyose content in embryo axes while in cotyledons it decreased evidently. The mass ratio of sucrose to oligosaccharides was higher in cotyledons of germinated seeds. The marked decrease of mass ratio of oligo to monosaccharides was observed in embryo axes in the last period of germination. The role of carbohydrates in loosing tolerance to desiccation in germinated Norway maple seeds is discussed.

Opis:
The treatment of type 2 diabetes should be supported with a proper diet paired with physical activity at every stage of therapy. Carbohydrates are key macronutrients with a direct effect on the level of postprandial glycaemia. For the past several decades, dietary intervention studies investigating and comparing the effects of limiting carbohydrate intake in favour of other macronutrients have been carried out. A low-carbohydrate diet has been and still is suggested by some experts as well as patient organizations as the most effective option that is associated with the smallest risk of side effects. The recommendations, however, have not been supported with evidence from high quality randomized control trials. The aim of this work is to review the published meta-analyses of randomized control trials, which compared the parameters relevant for the treatment of diabetes, including fasting plasma glucose, glycated haemoglobin, the lipid profile and weight loss. Currently, there is insufficient scientific evidence allowing a uniform recommendation to be made regarding the optimum amount of carbohydrates in the daily diet, hence the relevant recommendations should be tailored to each patient. In dietary interventions among type 2 diabetes, lowering the patient’s diet energy content is of primary importance, as well as the quality of ingested macronutrients, especially carbohydrates and fats.
Leczenie cukrzycy typu 2 na każdym etapie powinno być wspomagane odpowiednią dietą połączoną z aktywnością fizyczną. Kluczowym makroskładnikiem diety, mającym bezpośredni wpływ na wzrost glikemii po posiłku, są węglo-wodany. Od kilkudziesięciu lat badane są efekty interwencji dietetycznych polegających na ograniczeniu procentowego udziału węglowodanów w diecie na rzecz innych makroskładników. Dieta niskowęglowodanowa była i nadal jest sugerowana przez niektórych ekspertów oraz środowiska pacjentów jako najskuteczniejsza i niosąca najmniejsze ryzyko skutków ubocznych. Sugestie te nie są jednak poparte dowodami pochodzącymi z wysokiej jakości randomizowanych kontrolowanych badań klinicznych. Praca stanowi przegląd publikowanych w ostatnich latach metaanaliz randomizowanych kontrolowanych badań porównujących parametry mające znaczenie dla leczenia cukrzycy: glikemię na czczo, hemoglobinę glikowaną, lipidogram oraz redukcję masy ciała. Obecnie brak wystarczających dowodów naukowych do zalecania jednej, optymalnej ilości węglowodanów w diecie, wobec czego zalecenia te powinny być indywidualizowane. Większą rolę w podejmowanych interwencjach dietetycznych u chorych na cukrzycę typu 2 mają obniżenie całkowitej wartości energetycznej diety oraz jakość dostarczanych makroskładników, szczególnie węglowodanów i tłuszczów.

Carbohydrates are known as sources of immunological cross-reactivity of allergenic significance. In celery and in cypress pollen, the major allergens Api g 5 and Cup a 1 are recognised by antisera raised against anti-horseradish peroxidase and by patients' IgE which apparently bind carbohydrate epitopes; mass spectrometric analysis of the tryptic peptides and of their N-glycans showed the presence of oligosaccharides carrying both xylose and core α1,3-fucose residues. Core α1,3-fucose residues are also a feature of invertebrates: genetic and biochemical studies on the fruitfly Drosophila melanogaster, the parasitic trematode Schistosoma mansoni and the nematode worm Caenorhabditis elegans indicate that these organisms possess core α1,3-fucosyltransferases. Various experiments have shown that fucosyltransferases from both fly and worm are responsible in vivo and in vitro for the synthesis of N-glycans which cross-react with anti-horseradish peroxidase; thus, we can consider these enzymes as useful tools in generating standard compounds for testing cross-reactive carbohydrate epitopes of allergenic interest.

Opis:
Enzymatic depolymerisation of starch to glucose or maltose is carried out by starch- degrading amylases during a two-stage hydrolysis: liquefaction using bacterial α-amylase followed by saccharification with glucogenic (fungal amylase) or maltogenic (fungal or bacterial) amylases. As a rule, these enzymes are applied separately, following the recommendations concerning their action provided by the enzyme manufacturers. The study presents our attempts to determine the reaction conditions for a simultaneous action of liquefying and saccharifying enzymes on pre-treated potato starch. Hydrolysis was run by Liquozyme Supra, Maltogenase 4000L and San Super 360L enzymes (Novozymes) at different temperatures. During the single-stage method of starch hydrolysate production the most desirable results was obtained for the maltose hydrolysate at 80°C (51.6 DE) and for the glucose hydrolysate at 60°C (96 DE). The analyses indicate that the application of a single-stage hydrolysis of starch to maltose or glucose makes it possible to obtain a degree of starch saccharification comparable with that obtained in the traditional two-stage hydrolysis.
