Purpose: Enterococcus faecium strains have been reported worldwide as etiologic factors of many nosocomial infections, which are difficult to manage because of the constantly increasing resistance of these microorganisms to antibiotics and the ability to form biofilm. The aim of this study was to analyze the ability to produce a biofilm in E. faecium strains, depending on the patient’s clinical material. Materials and methods: Sixty-six E. faecium strains were investigated. Identification and susceptibility testing were conducted by the VITEK2 system. The ability to form biofilm was assessed by phenotypic methods. The presence of selected virulence genes was established by PCR followed by gel electrophoresis and sequencing. Results: Among the tested E. faecium isolates, 72.7% were biofilm-positive (BIO+) and 27.3% biofilm-negative (BIO-). Strains were collected mostly from rectal swabs (30.4%) and blood (18.3%). BIO+ strains from infections constituted 31.8% (52.4% isolated from blood) and from colonization 40.9% (48.2% from rectal swabs). 91.7% of the Blood Group strains and 68.5% of the Other Group strains produced biofilm. Strains from the Colonization Group produced biofilm in a proportion similar to the Infection Group (about 75%). There were no statistically significant differences in virulence and resistance, except for vancomycin (more resistant BIO+ Other than the BIO+ Blood Group, and more resistant BIO+ Colonization than BIO+ Infection Group) and teicoplanin (more resistant BIO+ Colonization than the BIO+ Infection Group). Conclusion: The majority of E. faecium isolates carries high levels of resistance to many antimicrobials, is well equipped with virulence genes, and possesses the ability to form biofilm.

Histophilus somni is an opportunistic pathogen causing respiratory, genitourinary and generalized infections in cattle. An important virulence factor is its ability to produce a biofilm. The aim of this work was to confirm that H. somni Hsp60 (Gro-EL) is a constituent of the biofilm produced by this bacterium in vitro and to check whether or not the presence of a specific antibody within the culture medium can inhibit biofilm production. Biofilm production by H. somni cultured in vitro was confirmed by crystalline violet staining. The presence of Hsp60 in the biofilm was confirmed by using specific antibodies produced in a mouse and goat hyperimmunized with H. somni recombinant Hsp60 (rHsp60). Large complexes of biofilm stained with Hsp60 antibodies were microscopically detected. This indicates that the Hsp60 protein is a common constituent of the biofilm produced by H. somni in vitro. In a second experiment, mouse serum containing anti- H. somni rHsp60 antibodies was added to an H. somni culture. It was found that the presence of anti-rHsp60 antibodies in the culture medium inhibited biofilm production in vitro. Only small biofilm particles were seen in the presence of the specific antibody, whereas in control cultures (without specific antiserum) large biofilm complexes were produced. The results indicate that antibodies specific to Hsp60 may be useful for preventing H. somni biofilm formation in vitro. If this also occurs in vivo, it may be helpful for eradicating H. somni infection in cattle through the elimination of carriers. Further in vivo studies are needed to confirm this idea.

Opis:
Mastitis is one of the most crucial diseases of dairy animals. Especially subclinical mastitis (SCM) has negative impacts on of dairy economy in term of reducing milk quality and quantity also premature culling and cost of therapy. Staphylococci are important etiological agents in SCM. The aim of the study was to investigate the biofilm production and antibiotic resistance profiles of Staphylococcus spp. other than S. aureus isolated from milks of Anatolian water buffalo with subclinical mastitis. Twenty-two coagulase negative staphylococci (CNS) identified phenotypically were also identified with PCR as Staphylococcus spp. other than S. aureus. Biofilm productions were investigated both by Congo Red Agar Method and PCR. The antibiotic resistance profiles of the isolates were determined by Disc Diffusion Method and they were antibiotyped. Only three (13.6%) isolates were biofilm positive both phenotypically and genotypically. All isolates except for two were resistant against at least two antibiotics. Multidrug-resistance among the isolates was low (13.6%). Antibiotyping results showed that the similarities among the strains were between 30-100%. Genotyping of the strains revealed that a genetic heterogeneity was found among CNS isolates and their similarities were between 43% and 93%. In conclusion, CNS isolates identified as subclinical mastitis agents in buffaloes showed a high antibiotic resistance profile especially against oxacillin and vancomycin. Further studies should be conducted to investigate new mechanisms and/or genes responsible for antibiotic resistance in buffaloes.

Sonication is one of the new and innovative approaches that is being increasingly used in food industry to control fermentation processes and to eradicate spoiling. Recently, this approach has seen new industrial applications such as enhancing microbial productivity. The present study aimed to assess the effects of ultrasound conditioning on the metabolism and extracellular matrix production of Streptococcus thermophilus. Bacterial suspensions were treated in ultrasonic bath (35 kHz, 240/60 W peak/nominal power, 1.8 l capacity) for different time periods (5, 10, 15, 20, 30, 45, and 65 min), and the growth improvement, adhesion ability, biofilm formation, and exopolysaccharide production of the bacterial strain were measured. The bacterial strain exhibited resistance to the treatment, and the conditioning improved the growth, adhesion, membrane permeability, biofilm formation, and exopolysaccharide production ability. An optimal treatment was obtained for 30 minutes of conditioning. An excellent yield of desirable exopolysaccharides (1788 mg glucose equivalent/l) was achieved. Ultrasound conditioning may be used as a potential approach to enhance certain biotechnological properties of industrial microorganisms.

The molecular-typing strategy, ERIC-PCR was used in an attempt to determine the genomic relationship of 28 P. aeruginosa strains isolated from faeces of healthy bovine, bovine mastitis and from faeces of hospital patients as well as from environment. ERIC-PCR fingerprinting revealed large molecular differentiation within this group of isolates. Twenty two out of 28 strains tested generated unique patterns of DNA bands and only three genotypes consisted of two isolates each were identified. We also tested the P. aeruginosa isolates for their ability to form a biofilm on abiotic surfaces including polyvinylchloride and polystyrene. Different biofilm-forming abilities were demonstrated among strains; however, most of them (64.3%) showed moderate-biofilm forming ability. The strains with increased swimming and twitching motility displayed elevated biofilm formation. However, a negative correlation was found between slime and initial biofilm production. On the basis of the results obtained, we suggest that there are no major differences in phenotypic properties between P. aeruginosa strains isolated from different sources.

Rozwój mikroorganizmów tworzących biofilm na powierzchniach użytkowych w zakładach produkcji i przetwarzania żywności często prowadzi do jej psucia się oraz stanowi poważne zagrożenie dla zdrowia publicznego. Szczególnie niebezpieczne dla zdrowia człowieka są biofilmy wytwarzane przez chorobotwórcze bakterie, takie jak Campylobacter jejuni, patogenne szczepy Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella enterica, Staphylococcus aureus czy drożdże z rodzaju Candida. Celem artykułu jest przybliżenie czytelnikom wiedzy na temat mechanizmów powstawania biofilmów, zagrożeń wynikających z rozprzestrzeniania się biofilmotwórczych patogenów, a także metod zapobiegania i usuwania biofilmu w zakładach produkcji i przetwórstwa żywności.
The development of biofilm-forming microorganisms on usable surfaces in food production and processing facilities often results in food spoilage and poses a serious threat to public health. Particularly dangerous to human health are biofilms formed by pathogenic bacteria such as Campylobacter jejuni, pathogenic strains of Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella enterica, Staphylococcus aureus and yeasts of the genus Candida. The purpose of this article is to provide readers with an overview of the mechanisms of biofilm formation, the risks associated with the spread of biofilm-forming pathogens, and methods of biofilm prevention and removal in food production and processing plants.

Bacterial biofilm is a complex structure of microorganisms with variable qualitative composition depending on the raw materials used, parameters of the production process and localization in hardly accessible places for antimicrobial agents. At the same time, the life of bacteria in the biofilm structure increases the resistance of pathogens and the probability of their survival in adverse conditions. However, the effectiveness of biofilm removal is still not satisfactory, despite the constant improvement of the cleaning and disinfection procedures of industrial surfaces. The methods of biofilm eradication used so far can be divided into three groups (physical, biological, chemical). Numerous in vitro studies indicate that alternatives to commonly used disinfectants may be natural substances such as essential oils that have bactericidal and bacteriostatic activity. The present review will focus on describing biofilm formation and performance. In addition, the paper describes an overview of the methods used to prevent and eradicate biofilms.

The first purpose of the study was to evaluate the influence of incubation parameters for efficiency of bacterial adhesion on surface of three solid, mineral porous carriers. Second aim of the study was the use of the immobilized Lactobacillus casei cells, in the production of lactic acid and selection, best carriers for continuous production using column bioreactor. The results showed that bacterial adhesion was different depending on the incubation conditions. In all tested samples, the highest efficiency of bacterial adhesion achieved after incubation at 30 °C without shaking which was related to bacterial viability. The results showed that, the highest efficiency of lactic acid production by free cells has been obtained at 30 °C after 48h – 110% (w/w) (lactic acid yield from consumed glucose). This results have been confirmed by further tests which have shown that in stationary culture, the highest production of lactic acid was reached by immobilised bacteria cells on pumice and kermiste after 48 hours at 30 °C.

This study aimed to assess the prevalence of enterococci (including vancomycin-resistant, VRE) strains in meat plants and evaluate their biofilm-forming potential. In two Polish meat plants, 75 samples of raw pork meat, swabs from work surfaces (floors, tables, machine parts and tools) and employees’ hands were collected. The analyses indicated that enterococci were present in more than 72% of the tested samples. In addition, VRE isolates were found in more than 25% of the tested samples (especially in cutting and processing rooms). VRE strains of Enterococcus faecium and E. faecalis were resistant to penicillin, ampicillin, erythromycin, and rifampicin. Moreover, 77% of E. faecium and 43% of E. faecalis isolates showed biofilm-forming ability. The observed high biofilm-forming potential among the analyzed VRE strains indicates that these agents may play an essential role in spreading drug resistance in the food chain through contaminated surfaces, meat, and workers’ hands.

Mulching with plastic materials that cover the soil creates a physical barrier to soil water evaporation, preserves good soil structure, controls weeds and protects plants from soil contamination. The removal and disposal of non-recyclable and non-degradable plastic wastes after harvest is difficult and expensive. Using alternative biodegradable polymers as covers has been of increasing concern in recent years. This paper provides a presentation of results concerning the physical and chemical properties of soil under biodegradable nonwoven covers. Biodegradable PLA (polylactide) and Bionolle (an aliphatic polyester of butylene glycol and succinic and adipic acid) films covering the soil on tomato and cucumber fields were evaluated to estimate the changes in several physical soil properties including bulk density, water field capacity, wet soil aggregate stability and chemical soil properties including soil acidity, EC, organic matter and soil mineral status. Favourable changes in the physical structure of soil can be achieved by mulching the soil surface of biodegradable polymers. The obtained results show that using covers with PLA and Bionolle biofilm significantly increased the amount of large aggregates and decreased the percentage of the smallest size aggregates in soils; however, the observed effects were strongly affected by weather conditions. Under wet conditions, mulching increased the soil bulk density and decreased soil water capacity. The results of soil chemical analyses demonstrated the low impact of treatments on macro- and microelement concentration measured after tomato and cucumber harvesting. Soils under PLA and Bionolle covers had smaller low ion concentrations in relation to bare soils.
Mulcze z tworzyw sztucznych tworzą fizyczną barierę dla parowania wody, pozwalają zachować dobrą strukturę gleby, kontrować zachwaszczenie oraz chronią rośliny przed zanieczyszczeniami glebowymi. Usuwanie i unieszkodliwianie po zbiorach odpadów z tworzyw sztucznych nienadających się do recyklingu i niepodlegających degradacji jest trudne i kosztowne. W ostatnich latach stosowanie alternatywnych włóknin biodegradowalnych jako mulczy lub okryć w uprawie roślin cieszy się coraz większym zainteresowaniem. Praca jest prezentacją wyników dotyczących właściwości fizycznych i chemicznych gleb mulczowanych biodegradowalnymi włókninami. W badaniach oceniano zmiany niektórych fizycznych właściwości gleby, takich jak: gęstość objętościowa, pojemność wodna i wodoodporność agregatów glebowych po uprawie pomidora i ogórka. Do ściółkowania gleby wykorzystano włókninę PLA (polilaktyd) i Bionolle (poliester butylenu, alifatycznego glikolu, kwasu bursztynowego i adypinowego). Chemiczne właściwości gleby oceniano, oznaczając w nich odczyn, stężenie soli, zawartość węgla organicznego, dostępne dla roślin składniki pokarmowe oraz zawartość metali ciężkich. Okrywanie powierzchni gleby włókninami z polimerów biodegradowalnych wpłynęło korzystnie na strukturę gleby. Biowłókniny PLA i Bionolle zwiększały ilość makroagregatów wodoodpornych, a zmniejszały odsetek najmniejszych agregatów w glebie, jednak obserwowane zmiany były silnie modyfikowane poprzez warunki atmosferyczne w poszczególnych latach prowadzenia badań. W bardzo mokrym roku okrywanie gleby zwiększało jej gęstość objętościową oraz zmniejszało pojemność wodną. Mulczowanie gleby z użyciem biodegradowalnych włóknin nie wpływało znacząco na zawartość składników pokarmowych i metali ciężkich w glebie. Obserwowano nieznaczny spadek EC w glebach pod ściółkami w porównaniu do gleby nieokrywanej.
