Opis:
W pracy przedstawiono nową metodę pomiaru Całkowitego Potencjału Antyoksydacyjnego (CPA) z wykorzystaniem wysokosprawnej chromatografii cieczowej (HPLC) w układzie faz odwróconych z detekcją fluorescencyjną. Jako pułapkę spinową, sensor zastosowano kwas tereftalowy. CPA odniesione jest do występujących naturalnie, najsilniejszych (najbardziej reaktywnych) rodników hydroksylowych, co jest olbrzymią zaletą proponowanej metody. Rodniki te generowane są w reakcji analogicznej do Fentona (Fe(II) z nadtlenkiem wodoru w pH obojętnym).
Antioxidants are crucial group of compounds in food chemistry, cosmetology and medicine. Therefore, many methods of their analysis are already described in the literature. Constitution of real samples is usually unknown and many antioxidants can interact each other. Hence, more useful information frequently provides the summaric value - Total Antioxidant Potential (TAP) which reflects concentration of all antioxidants in the sample multiplied by their antioxidant power. Although many assays are presented in the literature, the methods based on true reactive oxygen species provide the most realistic measure of antioxidant activity and are still needed. The proposed assay is related to the hydroxyl radicals, generated in the Fenton reaction. The product of its reaction with the sensor, terephthalic acid, was analysed using reversed phase high performance liquid chromatography with the fluorescence detection. Decrease of the surface area of its peak, caused by competition between the sample and sensor, is the TAP measure. The elaborated assay was practically tested on the samples of alcoholic drinks, teas as well as herbal infusions.

Opis:
Encapsulation of sinigrin in liposomes with the proliposomal method was performed in order to evaluate the effect of this process on in vitro simulated digestion and antioxidant potential of sinigrin. The recovery of sinigrin after simulated gastric and duodenal digestion of its free and liposomal forms was determined with HPLC-UV using human digestive juices. The antioxidant potential of sinigrin and sinigrin-loaded liposomes was determined with the Rancimat test as their ability to prolong oxidative stability of edible oil . The efficiency of 62% was obtained by encapsulating sinigrin in liposomes. The values of mean diameter, polydispersity index and zeta potential showed satisfactory size uniformity and physical stability of the liposomes containing sinigrin. Liposomes were shown to inhibit the digestion of sinigrin in both human gastric and intestinal juices, clearly enabling its prolonged release. Moreover, sinigrin in the liposomal form significantly prolonged the induction time of edible oil oxidation compared to its free form. The results obtained are encouraging from the point of view of a possible incorporation of the sinigrin-loaded liposomes in real functional food systems or their use as nutraceuticals.

Introduction: Antioxidants, isolated from different plant parts, are widely used due to their ability to prevent the development of so-called oxidative stress. Sea buckthorn (Hippophae rhamnoides L.) is one of the plants with expected antioxidant properties. Objective: The aim of the study was to evaluate the antioxidant activity of ethanolic, methanolic and acetonic extracts of H. rhamnoides leaves, ripe and unripe fruits obtained by ultrasound-assisted extraction. Methods: To estimate the antioxidant potential of the extracts the DPPH, FRAP, ABTS and Folin-Ciocalteu methods were applied. Moreover, the influence of the extrahent, as well as extraction time, on this activity was evaluated. Results: Sea buckthorn leaf extracts showed higher activity, contrary to the fruit extracts. Moreover, higher activity of ripe fruit extracts compared to unripe material extracts was found. To obtain the highest content of antioxidants in the extracts, ultrasound-assisted extraction for 60 min with methanol should be applied. Conclusions: The presented in vitro results could lead to the conclusion that H. rhamnoides seems to be a valuable source of antioxidants to be applied in various branches of industry.

Oxidative stress occurs as a result of an imbalance between production of reactive oxygen species and the regulatory mechanism produced by human cells. The stress contributes to the pathogenesis of inflammatory diseases. Synthetic antioxidants like butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and ascorbic acid, are used in curtailing the menace of the stress. Their potential depends on the type of phytochemicals in the oil. It is on this basis this study aimed at isolating, characterizing and investigating antioxidant potential of rind essential oil of C. reticulata. To achieve this aim, 500g of fresh rinds of C. reticulata were pulverized and hydro-distilled for three hours. GC and GC-MS analysis of the oil revealed the abundance of hydrocarbon monoterpenoids (96%). The most abundant compounds in the oil were D-limonene (82.4%), γ-Terpinene (9.4%) and β-Myrcene (2.2%). The oil exhibited antioxidant activity, with IC50 value (10.73 μl/ml), which was lower to the activity of ascorbic acid (49.14 μl/ml). This shows that the oil is more active than ascorbic acid. Hence, the oil could be explored for the treatment of oxidative stress.

The aim of this study was to determine the value of the antioxidant potential of several domestic herbal plant species derived from conventional and organic crops, which, due to the content of valuable biologically active compounds with anti-inflammatory properties, can be used in the production of cosmetic preparations. The antioxidant potential of methanol extracts of the tested plants was determined using the FRAP method with ascorbic acid as a standard. The content of antioxidants in the studied plants ranged from 1.121 to 13.228 mg/g for samples from conventional crops and from 1.559 to 19.327 mg/g for samples from organic farming. Greater biological value of extracts obtained from plants grown in an organic system than in a conventional systemhas been indisputably demonstrated. The obtained research results may constitute an incentive for producers of cosmetics to choose native organic plants for the production of their products.

Opis:
Phytochemicals and antioxidant potentials of chloroform extract of leaves of Cyathea latebrosa have been investigated. The metabolites detected are Carbohydrates, starch, glycosides, saponins, alkaloids, tannins, flavonoids, sterols, terpenes, and polyuronides. Quantitative Phytochemical analysis gave alkaloid (0.08×102 ±0.02), saponins (0.16×102 ±0.02), tannin (0.11×102 ±0.01), flavonoid (0.16×102 ±0.01), and phenol (0.18×102 ±0.01). FT-IR (KBr) cm-1 analysis showed -NH-(3433.41 weak), -OH- (3851.01 sharp), -C-F (1408.8 strong) –SO-(1101.39) and Ar (1646.3 weak), GC/MS characterization revealed the extract contained [2-Aziridinylethyl]amine (70.7%), Benzeneethamine, 3-fluoro-β,5-dihydroxy-N-methyl (14.0%), N-Methyltaurine (10.7%) and (R)-(-)-2-Amino-1-propanol (3.15%). The presence of these compounds exposed the leaves as a potential remedy for alleviated cold, cancer, malaria and to regulate immune functions. At 400 µg/ml concentration the extract and ascorbic acid produced 54.34% and 96.02% antioxidant activity respectively. The FRAP values were significantly (p < 0.05) low than the value of ascorbic acid (2µ M). The plant extracts showed low antioxidant potential even on higher concentrations when compared to reference standard ascorbic acid.

Consuming a diet high in fruits and vegetables can lower the risk of developing numerous chronic diseases, including cancer and cardiovascular disease, due to the presence of multiple antioxidants. Horticultural produce consumed in Cyprus include a large variety of fruits and vegetables, most of which are common components of a Mediterranean-style diet due to the island’s localization. The aim of the present study was to evaluate the antioxidant composition and activity of the edible portion of ten fruits and ten vegetables commonly consumed in Cyprus. Total phenolics, total anthocyanins, ascorbic acid and carotenoids contents were determined, while the in vitro antioxidant activity was evaluated by three assays. Antioxidant activity showed great variation, with the highest values found in green olives, capers and red chili peppers and in correlation with total phenolic content. Ascorbic acid was detected in large quantities in parsley, coriander, red guavas and red chili peppers, while red chili peppers, capers and coriander had high levels of carotenoids. Furthermore, sweet cherries, red grapes, and red apples contained significant amounts of anthocyanins. Statistical analysis revealed that phenolic compounds are the most potent antioxidants in fruit and vegetables, while total anthocyanins had a weak contribution to their antioxidant activity. The present study could be a guide for Cypriot as well as Mediterranean diet health-conscious consumers to select specific fruits and vegetables as dietary components. To the best of our knowledge, this is the first study thoroughly describing the antioxidant composition and activity of a wide array of important Cypriot horticultural products.

The aim of this work was to determine the total phenolic content (TPC) and the antioxidant activity of methanol extracts of Hericium erinaceus, Hericium americanum and Hericium coralloides, including free radical scavenging method (DPPH), ferric reducing antioxidant power (FRAP) and radical cation scavenging method (ABTS). Hericium spp exhibited moderate to high antioxidant activity. The highest TPC (3.27 ±0.01 mg GAE g–1) and antioxidant activity values (17.0 ±0.68 mmol TE g–1 in FRAP; EC 50 = 4.12 ±0.12 mg mL –1 in DPPH •; EC 50= 2.83 ± 0.10 mg mL–1in ABTS•+ ) were found for methanol extracts of H. coralloides. The TPC and antioxidant activity of H. erinaceus isolates varied from strain to strain. H. americanum possessed considerably lower total phenolic content (2.31 ±0.01 mg GAE g–1) and antioxidant activity (10.5 ±0.59 mmol TE g–1 in FRAP; EC 50 = 7.82 ±0.09 mg mL –1 in DPPH•; EC 50 = 6.36 ± 0.12 mg mL–1 in ABTS •+) than H. coralloides and H. erinaceus. A high correlation was determined between TPC and ABTS •+( r2= 0.855), DPPH•(r2= 0.969) and FRAP (r2 = 0.942). According to results obtained in the present study, Hericium spp., especially H. coralloides and some of H. erinaceus isolates, might be promising natural source of antioxidants for food and pharmaceutical industry.

The growing public awareness of the dangers regarding chemicals used in traditional agriculture has led to consumers seeking valuable and contaminant-free products. Ecological agriculture has become synonymous with high health value and product safety. The aim of this study was to evaluate the antioxidant activity and the total polyphenolic content of infusions of herbal tea bags and loose teas from traditional crops, as well as infusions of loose teas from ecological crops. Raw material comprised dried flowers of Matricaria chamomilla and Tilia cordata, as well as dried leaves of Urtica dioica, Melissa officinalis and Mentha piperita. Herbal infusions were prepared using three brewing times: 5, 10 and 20 min. The analysis of antioxidant potential was performed using in vitro methods such as DPPH, ABTS and FRAP. The polyphenolic content was determined using the Folin-Ciocalteu method. The antioxidant activity of the studied tea infusion depended on the method by which the plants were cultivated and the brewing time. The ecological agriculture conditions seem not to stimulate the synthesis of antioxidants. However, the possibility to obtain other beneficial properties of the studied plants is an indication to carry out ecological cultivation.

Purpose: The present study evaluates antioxidant activity, as well as polyphenol and flavonoid contents in common sprouts, available on the Polish market. The aim of this study was to extend our already published food databases. Materials and methods: Ten seed species from four plant families were analysed. Total polyphenol content of sprout extracts was determined using the Folin-Ciocalteau method. Total flavonoid content was assessed by the aluminium chloride colorimetric method. Total antioxidant status was measured using FRAP and ABTS methods. Results: The FRAP antioxidant potential was 0.60- 2.53 mmol TE (trolox equivalents)/100 g FM (fresh mass), and arranged in descending order it was: white mustard>cress>radish>broccoli>chickpea> sunflower>mung bean>wheat>green lentil>alfalfa), while the ABTS potential was 3.92-16.19 mmol TE/100 g FM (according to decreasing value: white mustard>green lentil>chickpea>sunflower>mung bean>cress> alfalfa>wheat> broccoli> radish). The polyphenol content was 160-774 mg GAE (gallic acid equivalents)/100 g FM, and flavonoid content 15-53 mg QE (quercetin equivalents)/100 g FM. Conclusion: Our results suggest that sprouts in comparison to other foods, despite small weight can be powerful sources of antioxidants. Special attention in human nutrition should be paid to white mustard sprouts as they are excellent source of polyphenol and flavonoid and are characterized by tremendous antioxidant activity.
