All over the world, the use of medicinal plants is gaining more acceptability due to the possibility of discovering novel drugs from them and solving the problem of antimicrobial resistance associated with conventional antibiotics. The phytochemical composition and antimicrobial properties of crude extracts of the leaves, stems, and bark of Annona muricata were evaluated on Escherichia coli, Staphylococcus aureus, and Salmonella typimurium, while the antifungal properties were evaluated against Candida albicans and Candida tropicalis. The Agar well method was used for the study. At concentrations of 150 mg/ml and 300 mg/ml, inhibitory effects were observed on E. coli and S. aureus, with a visible zone of inhibition ranging from 15 mm to 21 mm respectively, and with respect to N- hexane, an antimicrobial activity range of 5 mm to 20 mm, for the leaf extract, which shows effective antimicrobial action against E. coli and S. aureus. Hot water extracts were observed to possess more bioactive compounds compared to organic solvent extracts, and exhibit higher ranges of activity against the tested bacterial species. All extracts exhibited low anti-fungal activity in the range of 8 mm to 15 mm. The phytochemical screening of the extracts of different parts of A. muricata revealed the presence of secondary metabolites such as tannins, alkaloids, saponins, flavonoids, steroids, and cardiac glycosides. The antimicrobial activity of the extracts was compared with a standard antibiotic, ketoconazole, and with ampicillin, which served as the controls). The results showed that A. muricata can be used as an anti-bacterial substance, since it shows broad spectrum activity against a range of bacteria responsible for the most common bacterial illnesses. Further research will be necessary to ascertain its full spectrum of efficacy.

Objectives: The study presents data concerning occupational exposures among the staff of 5 hospitals in the Małopolska province in 2008–2012, taking into account the frequency and circumstances of exposure formation, occupational groups of hospital workers, as well as diversification of the reported rates in subsequent years between the hospitals and in each of them. An additional objective of the analysis was to assess the practical usefulness of the reported data for planning and evaluation of the effectiveness of procedures serving to minimize the risk of healthcare workers’ exposure to pathogens transmitted through blood. Material and Methods: Data were derived from occupational exposure registries kept by 5 hospitals of varying sizes and operational profiles from the Małopolska province from the years 2008–2012. Results: Seven hundred and seventy-five cases of exposure were found in a group of 3165 potentially exposed workers in the analyzed period. Most cases were observed in nurses (68%) and these were mainly various types of needlestick injuries (78%). Exposure rates with respect to all workers ranged from 2.6% to 8.3% in individual hospitals, but the differences in their values registered in the hospitals in subsequent years did not bear any statistical significance, in a way similar to the rates calculated separately for each occupational group. Conclusions: There was no upward or downward trend in the number of reported cases of exposure to bloodborne pathogens in the studied period in any of the hospitals. Statistically significant differences in the percentages of exposures were reported between individual hospitals in some years of the analyzed period, which confirms the need for registries in individual units in order to plan and evaluate the effectiveness of preventative measures.

Opis:
Progressive degradation of the environment caused, among others, by the excessive use of chemical pesticides forces us to look for alternative methods of protecting crops against pathogens. Definitely beneficial for the environment, but also, as confirmed by numerous studies, the use of biological control mechanisms can be an effective solution. The bacteria can inhibit the growth of fungi through the production of enzymes that lyse their cell walls, such as chitinases and glucanases, but also by limiting the availability of microelements important to growth, such as iron, by chelating them to siderophores, and finally producing antibiotics and secondary metabolites, like 2,4-DAPG. The present study examined 16 strains of bacteria isolated from Medicago sativa rhizosphere for their suitability for the control of fungal pathogens such as Fusarium culmorum and Phoma medicaginis. Among of bacteria were strains belonging to the family Bacillaceae (genus Bacillus, Lysinibacillus and Paenibacillus), the family Pseudomonadaceae (genus Pseudomonas), the family Xantomonadaceae (genus Stenotrophomonas) of the Enterobacteriaceae family (genus Citrobacter, Leclercia and Raoultella) and of the family Rhizobiaceae (genus Sinorhizobium). In vitro, both on solid and liquid media, all bacterial strains were able to limit the growth of Fusarium culmorum and Phoma medicaginis. The effective inhibitors of Fusarium culmorum Cul-3 were Roultella planticola KK 8a, Pseudomonas corrugata KK 12, and both strains belonging to Stenotrophomonas maltophilia KK 8b and KK 9b. Limiting the development of the second Medicago pathogen, Phoma medicaginis, was the most effective after the use of Pseudomonas corrugata KK 7, Leclercia adecarboxylata KK 6 and Pseudomonas brassicacearum KK 5. By studying the mechanisms that may be potential for bacteria to inhibit the growth of fungi was tested production of lytic enzymes the cell walls, and siderophores. All bacterial strains showed β-1,3-glucanase activity in the range of 2.3 to 72.5 μmol glucose /mg protein/min. Five strains showed chitinolytic activity, with Stenotrophomonas maltophilia KK 8b being the most active. Thirteen of the 16 analyzed strains of rhizobacteria were able to produce iron chelating compounds, siderophores. In addition, the presence of the phlD gene was confirmed in Pseudomonas brassicacearum KK 5 and P. corrugata KK 12, which may indicate the production of 2,4-DAPG. The results obtained indicate that the bacteria isolated from the rhizosphere have the potential for biological control of fungal pathogens, which limit the plant cultivation, using various mechanisms. The use of rhizobacteria as biopesticides is an environmentally friendly alternative to chemical plant protection products.
Postępująca degradacja środowiska spowodowana między innymi nadmiernym wykorzystywaniem chemicznych środków ochrony roślin zmusza nas do poszukiwania alternatywnych metod zabezpieczania upraw przed patogenami. Wśród bakterii były szczepy należące do rodziny Bacillaceae (rodzaju Bacillus, Lysinibacillus i Paenibacillus), z rodziny Pseudomonadaceae (rodzaju Pseudomonas), z rodziny Xantomonadaceae (rodzaju Stenotrophomonas), z rodziny Enterobacteriaceae (rodzaju Citrobacter, Leclercia i Raoultella) oraz z rodziny Rhizobiaceae (rodzaju Sinorhizobium). W warunkach in vitro zarówno na pożywkach stałych jak i płynnych wszystkie analizowane szczepy bakterii były zdolne do ograniczenia wzrostu Fusarium culmorum oraz Phoma medicaginis. Do skutecznych inhibitorów rozwoju Fusarium culmorum Cul 3 należy zaliczyć szczepy Roultella planticola KK 8a, Pseudomonas corrugata KK 12, oraz oba szczepy należące do Stenotrophomonas maltophilia KK 8b i KK 9b.

Objectives. Antimicrobial efficacy and toxicity analysis of 5 different concentrations of silver zeolite (SZ) compound against 5 common oral pathogens Streptococcus mutans, Streptococcus pyogenes, Lactobacillus sp., Staphylococcus aureus, Candida albicans. Material and methods. The antimicrobial efficacy of 5 different concentrations of SZ was tested against 5 common oral pathogens using the agar well diffusion method and the MIC and MBC values were determined using the micro broth dilution method. The toxicity of all 5 different concentrations was evaluated using brine shrimp assay and lethal concentration (Lc50) value was determined. Results. At 10 μg/mL the antimicrobial activity of SZ was almost negligible. The antimicrobial activity was observed in an increasing trend against all the test microorganisms as the concentration increased. At 75 & 100 μg/mL the zone of inhibitions was more than the control. Furthermore, MIC and MBC values of SZ with concentrations 25, 50, 75, and 100 μg/mL were determined and recorded. SZ was equally effective against all the test organisms. The LC25 (lethal concentration25) value was 1.6 μg/mL, whereas the LC50 value was 1.77 μg/mL and the LC75 value was 1.90 μg/mL, calculated from the probit computational method. Conclusion. SZ has the potential to change the ongoing system and bring about a revolution as an antimicrobial drug. However, the dose must be regulated as it can be toxic in higher concentrations. SZ compounds with the correct study of physicochemical properties and toxicity analysis can increase their pharmacological use and market value.

Opis:
A fast development of a wide variety of proteomic techniques supported by mass spectrometry coupled with high performance liquid chromatography has been observed in recent years. It significantly contributes to the progress in research on the cell wall, very important part of the cells of pathogenic fungi. This complicated structure composed of different polysaccharides, proteins, lipids and melanin, plays a key role in interactions with the host during infection. Changes in the set of the surface-exposed proteins under different environmental conditions provide an effective way for pathogens to respond, adapt and survive in the new niches of infection. This work summarizes the current state of knowledge on proteins, studied both qualitatively and quantitatively, and found within the cell wall of fungal pathogens for humans, including Candida albicans, Candida glabrata, Aspergillus fumigatus, Cryptococcus neoformans and other medically important fungi. The described proteomic studies involved the isolation and fractionation of particular sets of proteins of interest with various techniques, often based on differences in their linkages to the polysaccharide scaffold. Furthermore, the proteinaceous contents of extracellular vesicles ("virulence bags") of C. albicans, C. neoformans, Histoplasma capsulatum and Paracoccidioides brasiliensis are compared, because their production can partially explain the problem of non-classical protein secretion by fungi. The role assigned to surface-exposed proteins in pathogenesis of fungal infections is enormously high, thus justifying the need for further investigation of cell wall proteomes.

Sewage sludge is a by-product in the wastewater treatment and is an inherent hazardous issue because of the pathogenic contamination of natural resources. Therefore, in this study, domestic sludge was treated with premontane forest soil, macronutrients, and also pasteurization to reduce the content of volatile solids and pathogens. The best biostabilization treatment using premontane forest soil and pasteurization obtained a volatile solids reduction of 87% according to the environmental regulations, in which a biosolid is stable in a range of 38% of volatile solids reduction. In less than 30 days in a mesophilic range, the coliform count was reduced up to 71% when using forest soil and pasteurization. Thus, a biosolid-class B was obtained using gravimetric means as a platform to promote fast quality control.

Consumption of water contaminated with pathogens and heavy metals is responsible for the demise of millions of people around the globe. To find a low cost, facile and benign route for developing a biosorbent capable of handling the challenges associated with unsafe drinking water, the study report the preparation, activation, adsorption and removal of cadmium and pathogenic organisms from water samples. Modified plantain peel biochar (MPPB) was characterized using Braunauer-Emmett-Teller (BET) surface area and X-Ray diffraction (XRD). Surface area and pore size of the MPPB determined from BET surface area plot are 8.79 cc/g and 16.69 m2/g respectively with XRD crystallite size of 14.56 nm. The maximum adsorption capacity was observed to be 366.300 mmolkg-1. Equilibrium isotherm indicated the coefficient of determination to increase from Jovanovich →Elovic→Temkin→Freundlich→Langmuir model. Surface interaction and mass transfer kinetics shows a mixture of intra-particle diffusion and mass transfer process. The adsorption kinetics and surface interaction models coefficient of determination increased from Bangham pore diffusion →mass transfer model→Liquid film diffusion→pseudo first order→Dunwald-Wagner→Elovic→Pseudo-second order→Weber and Moris model. The adsorption indicated a chemisorption process with rate limitimg step likely to involve electron pair donor-acceptor interactions, hydrogen bonding and dipole dipole interactions. Antimicrobial evaluation of MPPB showed that it is a potential antimicrobial agent against common micro-organisms.

Biological control of plant diseases has received worldwide attention in recent years mainly as a response to use of hazardous chemicals in the environment. Soil actinomycetes particularly Streptomyces spp. enhance soil fertility and have antagonistic activity against wide range of soil-borne plant pathogens. In study of antifungal activity of streptomycetes against plant fungal-pathogens Fusarium oxysporum (Schlecht.), F. culmorum (Wm. G. Sm.) Sacc. and Rhizoctonia solani (Kühn) causing damping-off of pine seedlings, in vitro and in vivo tests were carried out. Two types of in vitro tests in mixed cultures of actinomycetes and fungi were performed. In majority of the cases, an inhibitory effect of actinomycetes on fungi was noticed. Three isolates (SG9, SR2, SR4) were highly active against pathogenic fungi and were chosen for exopolysaccharides extraction and in vivo tests. The sensitivity to exopolysaccharide action was noted in fungi from the genus Fusarium. Fusarium oxysporum showed a higher sensitivity to the analyzed exopolysaccharides than F. culmorum. Biocontrol interactions both in sterile and non-sterile soil were found for the actinomycete Streptomyces cyaneus (strain SG9) isolated from root free soil of the pine. The latest experiment confirmed the strong pathogenicity of the fungus Rhizoctonia solani. Antifungal properties of Streptomyces cyaneus (strain SG9) need further comprehensive studies.
