The aim of this project was to compare the effect of Rhizobacterium rhizogenes infection, and subsequent hairy root induction, on the antioxidant capacity of in vitro root cultures derived from two tomato cultivars differing in flavonoid biosynthetic potential. Tomatoes and especially processed tomatoes are rich in lycopene ,which is one of the most powerful antioxidants. Antioxidan capasity of normal (wild) roots from Kumato and Cherry seeds and transformed roots from infected cotyledons with agrobacterium was determined by the DPPH metod. The results presented in this paper indicate that the normal roots extracts and extracellular had higher antioxidants capasity than transformed roots but antioxidant capasity was higher in the extracellular than in the roots extract. Standard deviation was higher in extracellular than roots extract. This experiment was valuable because antioxidants protect human body against oxidative stress which can cause many diseases such as diabetes and accelerate the aging process of the organism.

Opis:
Inhibition of jack bean activity by 2,5-dichloro-1,4-benzoquinone (DCBQ) was studied in phosphate buffer, pH 7.0. It was found that DCBQ acted as a strong, time and concentration dependent inactivator of urease. Under the experimental conditions obeyed the terms of pseudo-first-order reaction, urease was totally inactivated. Application of Wilson-Kitz method proved that the urease-DCBQ interaction followed a simple bimolecular process and the presence of intermediate complex was undetectable. The determined second order rate constant of the inactivation was 0.053 (μM min)-1. Thiols such as l-cysteine, glutathione and dithiothreitol (DTT) protected urease from inhibition by DCBQ but DCBQ-modified urease did not regain its activity after DTT application. The thiol protective studies indicated an essential role of urease thiol(s) in the inhibition. The irreversibility of the inactivation showed that the process was a result of a direct modification of urease thiol(s) by DCBQ (DCBQ chlorine(s) substitution). The decomposition of DCBQ in aqueous solution at natural light exposure was monitored by visible spectrophotometry, determination of the total reducing capacity (Folin-Ciocalteu method) and DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging ability. The DCBQ conversion resulted in a decrease of the inhibition power and was well correlated with the increase of the total reducing capacity and DPPH scavenging ability. These findings were attributed to DCBQ transformation by photolysis and the hydrolysis effect was found to be negligible.

In this study, DPPH radical scavenging activity, ABTS radical cation decolorization assay, FRAP reducing power, total phenolic content, total flavonoid content and phenolic compounds of leaves of plants, grown in Afyon/Turkey were investigated. These plants were poppy, daisy, dandelion, manger, chickweed, black chicory and white chicory. Phenolic acids including gallic acid, ferulic acid, chlorogenic acid, coumaric acid, ellagic acid, vanilic acid, caffeic acid, cinnamic acid, 4-hydroxybenzoic acid, 2,5-dihydroxybenzoic acid and flavonoids including catechin, apigenin, naringin, rutin and quercetin amounts in leaves were determined. Among plants leaves, leaves of daisy and manger had statistically highest DPPH value and leaves of poppy and dandelion had statistically highest ABTS value, while leaves of White chicory had statistically highest FRAP value, total phenolic content and total flavonoid content. Leaves of black chicory and white chicory had higher phenolic compounds compared to that of other plants. These results suggest the using of these leaves as sources of natural antioxidants.

We used DPPH scavenging assays to study the antioxidant activity of three native Polish species of blackberry leaves (Rubus kuleszae Ziel., R. fabrimontanus (Sprib.) Sprib. and R. capitulatus Utsch.). All the studied extracts (methanolic, water, methanolic-water) showed high DPPH free radical scavenging activity (IC50 450.0–186.0 μg/ml). The most effective of the studied species was Rubus kuleszae. Total content of phenolic compounds (70.50–136.04 mg GAE/g) and phenolic acids (14.70–38.26 mg CAE/g) was determined spectrophotometrically. Antioxidant activity correlated positively with total content of phenolic compounds and phenolic acids.

Introduction:. It is well documented that many species from Passifloraceae family can provide edible and nutritious fruits while the leaves of cultivated plants are renewable and waste material. This biomass may be further used in various sectors, especially as a bioactive food additive and as source of innovative pharmaceuticals, cosmetics or feed additives. The biomaterials and green chemistry are new sectors bioeconomy according to the high-level horizontal strategies and bio-based industries in Europe. In recent years, attention has been paid to the biological activity and phytochemical profiles of extracts from different species of Passiflora. However, there is little comparative studies using the same procedures and techniques in the same laboratory conditions for study of plant material obtained from the similar greenhouse conditions. Objective: This study was focused on the examination of antioxidative activities of low concentrations of crude extracts from leaves of Passiflora incarnata L., Passiflora caerulea L., and Passiflora alata Curtis. Methods: The activity was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging and ferric reducing antioxidant power (FRAP) methods. Results of study were supported by estimation of chemical composition with secondary metabolites profiling in extracts which were carried out previously for the same extracts from three Passiflora species. One-way ANOVA analysis revealed significant differences in the antioxidant activity of various concentrations of the extracts using the DPPH and ABTS radical models, and FRAP method. Results: Measurement of antioxidant capacity (expressed as trolox equivalent, TE) showed that the most active was extract of P. caerulea > P. alata > P. incarnata. Phytochemical analysis for extracts of P. caerulea and P. incarnata showed greater similarities in metabolites content than P. alata. However, comparative statistical analysis of antioxidant activity showed that despite this phytochemical similarities, extract from P. alata leaves had higher activities than extract from leaves P. incarnata. Antioxidant effect of extract from P. alata can be explain by terpenoids presented in this extract. In this work, there have been discussed activities against Acanthamoeba castellanii strain, antibacterial and antifungal activities against selected clinical microorganisms (Enterococcus faecalis, Escherichia coli, Staphylococcus aureus, and Candida albicans, Microsporum gypseum), and anti-leukemic activities tested in human acute lymphoblastic leukemia cell lines for this extracts, which have been described in previous authors’ publications. Conclusion: Our current and previous studies showed that the same crude extracts from leaves of P. alata, P. caerulea, P. incarnata exerted not only antioxidant potential in vitro but also few interesting properties such as antibacterial, antifungal, amoebostatic, amoebicidal activities, which indicate the possibility of using these extracts in both a healthy diet and natural cosmetics. Leaves of this species may become an interesting source of biomaterials which can exert health-promoting effects.

INTRODUCTION: Fungal lesions are accompanied by inflammation, during which large amounts of free radicals are formed. Antifungal drugs, which have an additional antioxidant effect, could contribute to a faster recovery. An important role in ensuring effective and safe pharmacotherapy of fungal lesions is played by proper drug storage and appro-priate patient actions during treatment. Ultraviolet radiation can generate free radicals in a drug due to photolysis. A drug containing free radicals may cause toxic effects in the body. The aim of the study was to research the antioxidant properties of selected antifungal drugs of azole derivatives. Addi-tionally, the impact of UVC radiation on the tested drugs and their interaction with the DPPH free radical was studied. MATERIAL AND METHODS: The tested azole samples were exposed to 24-hour UVC radiation. The interaction of drug samples with a model DPPH free radical, before and after exposure to UV radiation, was studied. UV-Vis spectroscopy and colourimetry in the CIE Lab colour analysis system were used as the research techniques. RESULTS: Among the initial samples of the tested drugs only ketoconazole interacted with the DPPH free radical, causing its extinction. Exposure to UVC radiation increased the antioxidant properties in ketoconazole and miconazole. The CIE Lab parameters, UV spectra and interaction kinetics of those two drugs with DPPH were different after UVC exposure, which may indicate changes in ketoconazole and miconazole. The highest stability to ultraviolet radiation was shown by fluconazole, whose CIE Lab parameters, UV spectra and interaction with DPPH did not change after exposure of the sample to light radiation. CONCLUSIONS: The research using UV-Vis spectrophotometry and colourimetry enabled the impact of UVC radiation on selected antifungal drugs to be evaluated, including evaluation of their interaction with the model DPPH free radical. It is recommended to protect ketoconazole and miconazole from exposure to light. It is also advisable to protect patients from the sun during pharmacotherapy with these drugs.
WSTĘP: Zmianom grzybiczym towarzyszy stan zapalny, w trakcie którego wytwarzane są duże ilości wolnych rodników. Leki działające przeciwgrzybiczo, wykazujące dodatkowo działanie antyoksydacyjne, mogłyby się przyczynić do szybszego powrotu do zdrowia. Ważną rolę w zapewnieniu skutecznej i bezpiecznej farmakoterapii zmian grzybiczych odgrywają prawidłowe przechowywanie leku oraz odpowiednie postępowanie pacjenta w trakcie leczenia. Promieniowanie ultrafioletowe może generować wolne rodniki w leku w procesie fotolizy. Lek zawierający wolne rodniki może powodować efekty toksyczne w organizmie. Celem pracy było zbadanie właściwości antyoksydacyjnych wybranych leków przeciwgrzybiczych pochodnych azoli. Dodatkowo sprawdzono wpływ promieniowania UVC (ultraviolet C) na badane leki oraz ich oddziaływanie z modelowym wolnym rodnikiem DPPH (2,2-Diphenyl-1-picrylhydrazyl). MATERIAŁ I METODY: Badane próbki azoli poddano 24-godzinnej ekspozycji na promieniowanie UVC. Zbadano oddziaływanie próbek leków z modelowym wolnym rodnikiem DPPH przed i po naświetlaniu ich promieniowaniem ultrafioletowym. Jako techniki badawcze zastosowano spektroskopię UV-Vis (ultraviolet-visible) oraz kolorymetrię w układzie analizy barwy CIE Lab. WYNIKI: Spośród wyjściowych próbek badanych leków tylko ketokonazol oddziaływał z wolnym rodnikiem DPPH, powodując jego wygaszenie. Ekspozycja na promieniowanie UVC powodowała wzrost właściwości antyoksydacyjnych w ketokonazolu oraz w mikonazolu. Parametry CIE Lab, widma UV oraz kinetyka oddziaływania z DPPH obu leków były odmienne po naświetlaniu promieniowaniem UVC, co może wskazywać na zmiany zachodzące w ketokonazolu i mikonazolu. Największą stabilność na promieniowanie ultrafioletowe wykazywał flukonazol, którego parametry CIE Lab, widma UV oraz oddziaływanie z DPPH nie zmieniły się po naświetlaniu próbki. WNIOSKI: Badania z zastosowaniem spektrofotometrii UV-Vis oraz kolorymetrii pozwoliły na ocenę wpływu promieniowania UVC na wybrane leki przeciwgrzybicze, w tym na ocenę ich oddziaływania z modelowym wolnym rodnikiem DPPH. Zaleca się ochronę ketokonazolu i mikonazolu przed światłem. Wskazana jest również ochrona przed słońcem pacjentów w trakcie farmakoterapii tymi lekami.

In this study, the ability of hexane (HSE), chloroform (CSE), ethyl acetate (EASE) and methanolic (MSE) stems extracts from Phragmites australis or EDTA (as standard) to chelate iron using ferrozine method or zinc and copper using the murexide method is carried out in vitro. When the increased volumes of the HSE studied were taken from a stock solution of a fixed concentration 1 mg/ml at 25–175 μl for the iron and zinc chelating assay, 1–7 mg/ml for the copper chelating assay gave a significant (p≤ 0.01) activity. The obtained results showed that the HSE have the highest capacity to chelate ferrous ions below the EDTA (standard chelator) with absorbance arrive to the lesser extent 0.24±0.005, 0.04±0.013 which expresses 86% and 97% (compared to the control) of inhibition, respectively. For the murexide chelation, the results obtained also showed that the HSE and EDTA have a good (p≤ 0.01) chelating dose dependent effect towards zinc and copper ions with increased absorbance 0.45±0.02 and 0.42±0.02 with 54% and 56% of inhibition, respectively, for the zinc chelation and 0.66±0.03, 0.13±0.005 represent 44% and 88% for copper chelation. In contrast to the antioxidant capacity, the extract of hexane, ethyl acetate, chloroform and methanol from leaves, stems and roots of the Phragmites australis plant have a very low scavenger effect to the radical DPPH where the maximum inhibition is approximately 13.79%, obtained with the maximum volume. Finally, the HPLC analysis of effective extract (HSE) confirmed the presence of oxalic, citric, malic, succinic, fumaric, formic, acetic, propionic and butyric or iso butyric acid.

Introduction: Our study is a part of a trend of studies on the antioxidative properties of Chelidonium majus extracts or their fractions suggesting that antioxidant activities may depend on total flavonoid and/or alkaloid contents. Objective: This study focused on the examination of antioxidative activities of full water extract, non-protein fraction and protein fraction of the extract from aerial parts of mature plants and young seedlings. Methods: Total flavonoid and alkaloid contents were evaluated by spectrometric methods. Quantitative determination of chelidonine, coptisine, sanquinarine, berberine was made by HPLCUV. The antioxidative activities were evaluated using (1) 2,2-diphenyl-1-picrylhydrazyl (DPPH), (2) 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging and (3) ferric reducing antioxidant power (FRAP) methods. Results: All concentrations of herb extracts exhibited higher antioxidant capacities than extract from seedlings. Two antioxidant tests (DPPH, FRAP) showed that full water extract from herb had the highest antioxidant activity, while its non-protein fraction and protein fraction showed lower antioxidant activity. It was found that the full water extract from herb contained the highest concentrations of flavonoids and alkaloids when compared with other samples. Conclusion: Our findings suggest that chelidonine and coptisine especially could be responsible for the observed changes in the extract antioxidant activity, because these alkaloids were determined in the highest concentration in full water extract from herb. It cannot be also excluded that the observed variables values between extracts and their fractions from herb or from seedlings may also be the result of interactions between flavonoids and other chemical compounds.
Wstęp: Nasze badania, będąc częścią trendu badawczego skupiającego się na ocenie aktywności antyoksydacyjnej ekstraktów i ich frakcji z Chelidonium majus wskazują, że działanie przeciwutleniające może być zależne od całkowitej zawartości flawonoidów i/lub alkaloidów. Cel: W badaniu skupiono się na ocenie aktywności antyoksydacyjnej ekstraktów z ziela dojrzałych roślin oraz siewek Ch. majus (pełny ekstrakt wodny, jego frakcja bezbiałkowa i frakcja białkowa). Metody: Całkowita zawartość flawonoidów oraz alkaloidów była oznaczana metodami spektrofotometrycznymi. Ilościową zawartość chelidoniny, koptyzyny, sanquinaryny, berberyny określono metodą HPLC-UV. Aktywność antyoksydacyjną oceniano przy użyciu (1) wolnego rodnika DPPH (2,2-difenylo-1-pikrylohydrazyl), (2) ABTS (kwas 2,2’–azynobis-(3-etylobenzotiazolino-6-sulfonowy)) oraz metodą (3) oznaczania zdolności redukowania jonów żelaza (FRAP). Wyniki: Wszystkie stężenia ekstraktów z ziela wykazywały większą pojemność antyoksydacyjną w porównaniu z ekstraktami z siewek. W dwóch testach antyoksydacyjnych (DPPH, FRAP) wykazano, że pełny ekstrakt wodny z ziela wywierał największą aktywność antyoksydacyjną, natomiast frakcje białkowa i bezbiałkowa tego ekstraktu wykazywały niższą aktywność. Pełny ekstrakt wodny z ziela zawierał najwyższe stężenie flawonoidów i alkaloidów w porównaniu z innymi analizowanymi próbkami. Wnioski: Wyniki badań sugerują, że szczególnie chelidonina i koptyzyna mogą być odpowiedzialne za obserwowane zmiany w aktywności antyoksydacyjnej z uwagi na to, że te alkaloidy oznaczono w największym stężeniu w pełnym ekstrakcie wodnym z ziela. Nie można również wykluczyć, że obserwowane różnice w wartościach badanych zmiennych pomiędzy ekstraktami z ziela Ch. majus i otrzymanymi z siewek mogą wynikać z zachodzenia interakcji flawonoidów z innymi związkami chemicznymi.

Opis:
The article presents research on the exploring of extraction process of biologically active substances from the leaves of the Moringa oleifera tree using ethanol. Ethanolic extracts were obtained using three different techniques: maceration with shaking, ultrasound-assisted extraction and extraction in Soxhlet apparatus, in different time variants: 1, 2 and 4-hours. After solvent evaporating and drying, the yields of dry extracts obtained in particular processes were calculated. The antioxidant activity of extracts was analyzed spectrophotometrically using DPPH radical scavenging method, and total phenolic content (TPC) was determined by Folin-Ciocalteu method. By means of gas chromatography with mass selective detector (GC-MS), 11 biologically active compounds present in ethanolic extracts were identified, among which α-tocopherol had the greatest share. Based on the results, the influence of the extraction technique and time on the yield and antioxidant activity of M. oleifera leaf extracts were discussed.

The increase in the costs of storage and disposal of post-production residues has resulted in the search for new directions for their recycling, which is closely related to the necessity of protecting the natural environment and promoting a circular economy. Moreover, the apparent interest shown by the food market in raw materials with high antioxidant activity implies an increasing use of by-products. The objective of the study was to determine the effect of the type and concentration of the solvent on the efficiency of extracting polyphenols from distillery stillage as well as their antioxidant activity by using several solvents: methanol:water (70:30 v/v), methanol:water (100:0 v/v), ethanol:water (70:30 v/v) or ethanol:water (100:0 v/v). The DPPH radical method was used to determine the antioxidant activity of the obtained extracts. The normalised variable (NV) and statistical measure (MS) were determined, based on which the effectiveness of the solvents was evaluated. The highest polyphenolic content and the antioxidant activity were obtained by using ethanol:water (70:30 v/v) as a solvent in the extraction of polyphenolic compounds from distillery stillage.

Spirulina platensis is a microalga that contains a lot of secondary metabolites and is utilized as a dietary supplement. It can inhibit fat peroxidation better compared to the chemical antioxidant. Laboratory-scale closed reactor system was designed for Spirulina cultivation. This research aims to analyze identified secondary metabolite (Phytochemical contents) and the antioxidant activity or value of Inhibition Concentration 50% (IC50) of the fresh lab-scale cultivated Spirulina platensis. The antioxidant content was evaluated using DPPH method (1,1–diphenyl-2-pikrilhidrazil) on various sample concentrations as well as using Vc (Ascorbic Acid) as a positive control. The principle of hydrogen absorption by free radicals from antioxidants was demonstrated with the absorption value using a spectrophotometer at a wavelength of 517 nm. The results show that Spirulina platensis lab-scale cultivation was resulting in 5.0±0.01 g/10.0 L wet basis on the 10th day. The results show that Spirulina platensis has positive compounds of flavonoid, steroids, triterpenoids, phenolic, and saponins. Fresh Spirulina platensis has a value of IC50 647.045 ppm and Vc as a positive control has the IC50 of 2.085 ppm. The potential as a source of natural antioxidants was categorized as a very weak capacity. Therefore this study can be concluded that Spirulina platensis cultivated in Laboratory-scale has a potential to act as antioxidant candidate.

The aim of this study was to elucidate the influence of thermal processing at 22-95°C on the antioxidant ability of Brussels sprouts juice and its thiosulfinate content. The antioxidant power was determined by FRAP, Folin-Ciocalteu and DPPH radical scavenging methods. Thiosulfinate concentration was assayed by Han's procedure. It was shown that the increase of processing temperature decreased of the antioxidant ability determined by Folin-Ciocalteu method. However, FRAP and DPPH scavenging showed increase by 20% of the antioxidant power in the final heating step (95°C). The increasing processing temperature resulted in decreasing thiosulfinate concentration until the total loss. This proved that the thiosulfinate present in Brussels sprouts juice is thermolabile compound (not resistant for high temperature). The total loss of the thiosulfinate in high temperature did not reflect the change in antioxidant power. This fact indicated that there was no impact of the thiosulfinate on antioxidant abilities of Brussels sprouts juice or the impact was insignificant among the abundance of other antioxidants. The results showed a good correlation between FRAP and DPPH assays of antioxidant power determination in contrast to correlation with the data of Folin-Ciocalteu procedure.

In this work, synthesis of three pyrazoline derivatives (6-8) is described. (E)-1,3-(phenylsubstituted)-prop-2-en-1-one (3-5) is prepared by the reaction of substituted benzaldehyde with 4-methylacetophenone, whereas condensation cyclization of the same chalcones (3-5) with phenylhydrazine hydrate in ethanol yielded 6-8. The structures of the title compounds (6-8) were characterized by chemical reactions, elemental analysis, and spectral methods such as IR spectra. The synthesized chalcone and pyrazolines were evaluated for in-vitro antibacterial and antioxidant activities against standard. The zone of inhibition for some of the newly synthesized compounds showed notable antibacterial activity against selected bacterial strains when compared with ampicillin. Significant antioxidant activities were also shown by chalcone and pyrazolines.

Comparative analysis of antioxidant capacity was performed using FRAP and DPPH methods on extracts containing carotenoids acquired from fruits of Sea buckthorn. The examination included nine varieties of Sea buckthorn growing in the comparative cultivation. Conducted analysis allowed to compare the antioxidant capacity with carotenoids content measured with spectrophotometric and HPLC methods. Three of the examined cultivars indicating high antioxidant activity in both, FRAP and DPPH methods, also revealed highest ('Aromatnaya') and high ('Botanicheskaya', 'Arumnyj') total carotenoids content in HPLC analysis.
