It is not always clear that some equations affected by complicated factors can, actually, be interpreted as a ratio of two polynomials of first degree and so that they can be, in general, represented by rectangular hyperbolas. In this paper we present an easy procedure to rearrange those equations into Michaelis-Menten-type equations and so to make the aspects of these rectangular hyperbolas more clear, particularly for researchers familiar with general biochemistry. As an example, the method is applied to transform the classical rate equation of the Cleland×s Ordered Uni Bi enzyme mechanism.

A combined analysis of enzyme inhibition and activation is presented, based on a rapid equilibrium model assumption in which one molecule of enzyme binds one molecule of substrate (S) and/or one molecule of a modifier X. The modifier acts as activator (essential or non-essential), as inhibitor (total or partial), or has no effect on the reaction rate (v), depending on the values of the equilibrium constants, the rate constants of the limiting velocity steps, and the concentration of substrate ([S]). Different possibilities have been analyzed from an equation written to emphasize that v = Ł([X]) is, in general and at a fixed [S], a hyperbolic function. Formulas for Su (the value of [S], different from zero, at which v is unaffected by the modifier) and vsu (v at that particular [S]) were deduced. In Lineweaver-Burk plots, the straight lines related to different [X] generally cross in a point (P) with coordinates (Su, vsu). In certain cases, point P is located in the first quadrant which implies that X acts as activator, as inhibitor, or has no effect, depending on [S]. Furthermore, we discuss: (1) the apparent Vmax and Km displayed by the enzyme in different situations; (2) the degree of effect (inhibition or activation) observed at different concentrations of substrate and modifier; (3) the concept of Ke, a parameter that depends on the concentration of substrate and helps to evaluate the effect of the modifier: it equals the value of [X] at which the increase or decrease in the reaction rate is half of that achieved at saturating [X]. Equations were deduced for the general case and for particular situations, and used to obtain computer-drawn graphs that are presented and discussed. Formulas for apparent Vmax, Km and Ke have been written in a way making it evident that these parameters can be expressed as pondered means.

Introduction. The standard porcine-derived pancreatic enzyme replacement therapy (PERT) is a lifesaving treatment for patients with diseases causing exocrine pancreatic insufficiency (EPI). An attempt to replace PERT with microbial enzymes were undertaken. The aim was to highlight whether the mode of application, mixed with food or applied directly to the stomach, of pancreatic-like enzymes of microbial origin (PLEM) can affect their activity along the gastrointestinal tract. Materials and method. The activity of amylase, lipase and proteinase in the stomach, duodenum and ileum were tested in EPI pigs (n=6) after supplementation of PLEM, either orally – before and during feed consumption – or via the stomach – before and during feed consumption. Healthy pigs not treated with PLEM (n=3) served as controls. Activity of the enzymes measured in the chyme were obtained together with the digesta pH. Activity of the enzymatic residues in the stool samples was also checked. Results. The highest pancreatic enzyme activities were found in the duodenum of the healthy pigs (amylase 162,68 kU/ mL, lipase 507,34 kU/mL and protolitic (trypsin) activity 357,60 kU/mL). Nevertheless, the microbial enzymes remained also active along the entire length of the GIT – including stomach in EPI pigs, regardless of their route of administration. However, activity level was significantly lower. Discussion. Results indicate that the activity pattern of PLEM in the small intestine mimics the activity of the natural endogenous pancreatic enzymes in healthy pigs. The most physiological features of PLEM were observed when enzymes were offered orally. The magnitude of PLEM activity in the stomach of EPI pigs was essential and significantly higher than that measured in healthy pigs, thus being somewhat not physiological, and for health reasons of the patients should be further explored. Interestingly, specific trypsin-like activity was measured in all parts of the GIT after PLEM application. However, proteolytic activity of the experimental proteaze in in vitro studies did not exhibit trypsin-like activity.

The objective of the study described the importance of L-asparaginase and its importance in the field of medicine. Different types of enzymes are produced based on the adaptation to the environment where the living organisms live to tune the metabolic pathways according to their adapted changes. The enzymes present in various organs are produced by many cell types in multicellular organisms. Except ribosomes all other known enzymes are proteinaceous in nature. L-asparaginase is a potential therapeutic agent for acute lymphoblastic leukemia (ALL) and chronic myelogenous leukemia which is approved by FDA & WHO. L-asparaginase catalyzes the deamination of L-asparagine to L-aspartic acid & ammonia. Unlike normal cells, malignant cells require large amount of L-asparagine for protein synthesis and cell division. From this background the present review is an effort to gather the information on the mechanism, sources, molecular details and application of L-asparaginase enzyme.

Opis:
Taking into account the progressive degradation of soils it is important to assess their quality. Soil quality depends on a large number of physical, chemical, biological and biochemical properties. In the publications available, there are presented three approaches regarding the use of soil properties to estimate soil quality: (1) the use of individual properties, (2) the use of simple indexes and (3) the use of complex indexes derived from combinations of different properties. The purpose of this study was to investigate the possibility to use enzymes as indicators of forest soil quality. Experimental plots (43) were located in central Poland. The study was carried out in a number of diverse fresh forest sites. To assess the quality of forest soils dehydrogenase and urease activity and the degree of base saturation were used. One of the final conclusions point out that enzymatic activity indicates current site condition as well as the changes that occur in soil better than soil physical and chemical properties. In other words, in comparison to soil enzymatic activity, soil physico-chemical properties constitute a less sensitive indicator of soil changes.

Using histochemical and cytophotometric methods, enzymes associated with anaerobie glycolysis: glucose-6-phosphatase, hexokinase, and aldolase in the developing sporocysts of Fasciola hepatica were studied. Highest activity of these enzymes was found in the germ balls in the sporocysts, at all phases of their development, which is related to intensive proliferation and differentiation of the embryos developing inside the germ balls.

The paper reports results of analysis of protein and eight enzymatic systems of three varieties of Helianthus annuus L. (Wielkopolski, Coril, and Frankasol) performed by three electrophoretic methods. Electrophoretic analyses allowed finding protein and isoenzymes markers for each of variety. The two interline hybrids Coril and Frankasol shared more features in common than the population variety Wielkopolski with each of the former. Mean values of the polymorphism index for the protein and the eight enzymes indicate that the variety Wielkopolski is more homogeneous than Coril and Frankasol.
W pracy przedstawiono wyniki analiz białek i ośmiu układów enzymatycznych, rozdzielanych trzema metodami elektroforezy, u trzech odmian Helianthus annuus L. (Wielkopolski, Coril i Frankasol). Elektroforetyczne analizy umożliwiły znalezienie białkowych i izoenzymatycznych markerów dla trzech odmian. U dwóch odmian międzyliniowych mieszańców Coril i Frankasol wykryto więcej cech wspólnych niQ u wymienionych odmian i odmiany Wielkopolski. Średnie wartości indeksu polimorfizmu, obliczone dla białek i ośmiu układów enzymatycznych, pokazały, że odmiana Wielkopolski była bardziej wyrównana niż odmiany Coril i Frankasol.

Celem badań było określenie aktywności ureazy na wybranych glebach Polski: bielicowych i brunatnych wytworzonych z piasków i glin, lessów, czarnych ziem i mad rzecznych. Badaniom poddano próbki z poziomu 0-10 cm, które pobrano z Banku Gleb Instytutu Agrofizyki PAN. Aktywność ureazy zawierała się w przedziale 0,3256 - 1,7896 pM N-NH4 kg-lh-1. Najniższą aktywność ureazową zanotowano w glebach bielicowych i wynosiła ona 0,33 pM N-NH4 kg-lh-1. W glebach brunatnych wytworzonych z piasków i lessów aktywność ureazy wahała się w granicach 0,55 - 1,00 pM N-NH4 kg-lh-1. Najwyższą aktywność stwierdzono w glebach: czarnych ziemiach, madach rzecznych, glebach murszowych i murszowatych 1,43 - 1,79 pM N-NH4 kg-lh-1. W oparciu o analizę statystyczną otrzymanych wyników można stwierdzić, że aktywność ureazowa pozostaje w ścisłym związku z zawartością materii organicznej.
The aim of the investigation was to measure activity of urease in different polish soils such as podzolic and brown developed from sands and loams, brown developed from loess, as well as in black earths and aluvials soils. The measurements were performed on samples taken from 0-10 cm depths. The urease activity was in the range of 0,3256 - 1,7896 pM N-NH4+ kg''h‘'. The lowest activity was noted in the podzolic soil and was on the level of 0,3256 pM N-NH4+ kg’'h''. In the brown and lessive soils the urease activity was in the rangę 0,55-1,00 pM N-NH4+ kg''h_l. The highest urease activity were found in black earths, alluvials soils, Eutric Histosol Soils 1,43-1,79 pM N-NH/kg1 h’’. The results obtained allow to conclude that the urease activity is tightly connected with the organie matter content of the soil.

Chloroplast protease AtDeg2 (an ATP-independent serine endopeptidase) is cytosolically synthesized as a precursor, which is imported into the chloroplast stroma and deprived of its transit peptide. Then the mature protein undergoes routing to its functional location at the stromal side of thylakoid membrane. In its linear structure AtDeg2 molecule contains the protease domain with catalytic triad (HDS) and two PDZ domains (PDZ1 and PDZ2). In vivo AtDeg2 most probably exists as a supposedly inactive haxamer, which may change its oligomeric stage to form active 12-mer, or 24-mer. AtDeg2 has recently been demonstrated to exhibit dual protease/chaperone function. This review is focused on the current awareness with regard to AtDeg2 structure and functional significance.
