The aim of this study was to use a two-marker assay for the detection of breast cancer cells circulating in patients' blood. We have applied a PCR-based methodology to follow up the possibility of the development of metastatic disease in stage I and II patients who had undergone curative surgery. Since the number of circulating cancer cells in peripheral blood is very low, the technique for their detection needs to be not only highly sensitive, but also very specific. The reverse transcriptase-polymerase chain reaction (RT-PCR) technique may improve the sensitivity of breast cancer cell detection up to only a few cells per one million. The principle of the RT-PCR assay is to amplify a messenger RNA characteristic for breast epithelial cells in a blood sample. Since we do not expect such cells to be circulating in peripheral blood of healthy subjects, detection of the characteristic mRNA should indicate the presence of circulating breast cancer cells. We analyzed the usefulness of three mRNA markers: cytokeratin 19 (CK19), mammaglobin (hMAM) and β subunit of human chorionic gonadotropin (β-hCG) for this test. Blood samples (112) were obtained from 55 patients, in stages I and II, with or without metastasis to regional lymph nodes (N0 or N1). We found that a two-marker assay increases the sensitivity of detection of breast cancer cells in comparison with a single-marker one. Combination of two tumor-specific mRNA markers, hMAM/CK19 or β-hCG/CK19, allowed the detection of circulating breast cancer cells in 65% of N1 patients and 38% of N0 patients. By comparison, the combination hMAM/β-hCG allowed the detection of circulating breast cancer cells in the blood of 68% of N1 patients and 46% of N0 patients. Addition of the third marker did not significantly increase the detection sensitivity.

The results of the current study demonstrate that exercise interferes with the hormonal balance system. The size and direction of these changes depend on the volume and intensity of a physical effort. Biochemical monitoring can support the process of training and gathered data can assist in countering the effects of overtraining. The article presents the latest reports on the character of the changes in cortisol influenced by various types of physical effort, the impact of the training units, time of their execution and the influence of the supply of carbohydrates on the level of this hormone in the body. This work also presents the advantages of saliva analysis, as a reliable material to measure the level of cortisol in athletes.

Forty eight inbred lines of winter rye, of various origin and pedigree, were analysed using 19 RAPD (random amplified polymorphic DNA) primers, 8 ISSR (inter-simple sequence repeats) primers and 13 SSR (simple sequence repeats) primer pairs. On the basis of particular marker types, there were created three separate dendrograms and one combined similarity tree, prepared on account of the whole data. Correlation coefficients for individual technique based on genetic similarity matrices were not significant. By comparing the GS data obtained on the basis of singular methods with collective matrix, it was observed that the highest correlation rate was for ISSR method (r=0.68). The utility of each marker technique was compared by using marker index MI. This study examines the pragmatic functions of the marker sawa in spoken Swahili. The data have been obtained from informal conversations made by Swahili speakers in informal social settings. These settings include “vijiwe vya kahawa” (setting of informal conversations created around people drinking coffee) and "vijiwe vya mamantilie” (setting of informal conversations created around women preparing and selling food on the streets). The analysis of the data, performed within the framework of the contextualization theory (Gumperz 1982), shows that, apart from its basic connotation of agreement, the marker sawa conveys other meanings, depending on these very contexts of communication, and therefore acquires also various pragmatic functions. The pragmatic functions identified in this work include: to show that the speaker agrees with what has been said but on a condition (I agree, but…), as a receipt marker, as a tag-positioned-comprehension check, as an answer to the question showing that the speaker has understood what has been said (Yes, I understand), as a continuer, as a negative releasing marker, and as a gap filler. Interestingly, the study shows also that intonation and other paralinguistic features (like gestures) play a role in determining the pragmatic functions of this marker. The article concludes that the pragmatic markers in spoken Swahili are rich in meanings and are used to show speakers’ attitudes and emotions, therefore manifesting a deep and meaningful interconnection between the language and its contextualized experience.

Breast cancer is a very serious disease from both an economical and social point of view, because of its high incidence and mortality, as well as its prevalence. Genetic and epigenetic changes as well as the resultant deregulation of mechanisms that affect cellular processes are reflected in the final stage, like significant variability in phenotype. These changes have multiple factors in clinical, morphological but also molecular terms, and they greatly affect the possibility of accurate diagnosis of breast cancer in its early stages. Since no combination of clinical examination, mammography and ultrasonography together with CT and MRI can achieve 100% sensitivity, clinical practice is supported with diagnostic methods based on the monitoring of tumour markers with the emphasis on molecular markers. This review describes the most common molecular markers already used for earlier clinical diagnostics and also proposes new possible markers from the tumour vascular markers group. Any new oncomarkers to improve diagnostic sensitivity and specificity will be of great benefit, and in clinical practice, they may provide an opportunity for early diagnostic and therapeutic intervention, and subsequently better prognosis for the patient as well.
Rak piersi jest bardzo poważną chorobą, zarówno z punktu widzenia ekonomicznego jak i społecznego, ze względu na dużą zachorowalność i umieralność, a także na zakres jego występowania. Zmiany genetyczne i epigenetyczne, jak również wynikająca z nich deregulacja mechanizmów, które wpływają na procesy komórkowe są odzwierciedlone w ostatnim stadium choroby, jak znaczna zmienność fenotypu. Zmiany te mają wiele przyczyn z punktu widzenia klinicznego, morfologicznego, ale również molekularnego oraz znacznie wpływają na możliwość dokładnego rozpoznania raka piersi w jego wczesnych fazach. Ponieważ żadna kombinacja badań klinicznych, mammografii i USG wraz z tomografią komputerową i obrazowaniem magnetyczno-rezonansowym nie może osiągnąć 100% czułości, praktyka kliniczna jest wspierana metodami diagnostycznymi opartymi na monitoringu markerów nowotworowych, ze szczególnym uwzględnieniem markerów molekularnych. Niniejsza recenzja opisuje najbardziej typowe markery molekularne wykorzystywane we wczesnej diagnostyce klinicznej, a także proponuje nowe potencjalne markery z grupy markerów nowotworowych TVMs . Wszelkie nowe markery nowotworowe poprawiające czułość i dokładność diagnostyki będą bardzo pomocne, a w praktyce klinicznej mogą pozwolić na wczesną diagnostykę i leczenie, co polepszy także rokowania pacjenta.

Background: The entry and dispersal of modern humans is India of remains unclear and extending with many interesting evidences South India, assumed to be a major corridor for their with many ancient genetic deposits such as Dravidian tribal with Negrito features. As the relationships between the genetic polymorphs and diseases in human being revealed globally, it is worthy to investigate the genomic architecture of population in south India. Objective: To examine what evolutionary forces have most significantly impacted south Indian tribal genetic variation, and to test whether the phenotypic similarities of some south Indian tribal groups to Africans represent a signature of close relationship to Africans or are due to convergence. Methods: Blood samples from 193 unrelated individuals of both sexes are drawn from the Dravidian tribal settlements of Tamil Nadu and Kerala. South India are genotyped for four Aluindel (Alu FXIIIB, Alu ACE, AluTA25 and Alu PLAT) allele profile by PCR genotyping method. Results: All loci are highly polymorphic and average heterozygositiesare substantial (range: 0.37-0.44). Genetic differentiation is high (Gst = 3.7%) in all the study populations.

Introduction and objective. Bone sialoprotein (BSP) is present in a non-collagenous fraction of bone matrix, in dentine, cement and calcified cartilage. The characteristic place where sialoprotein is produced makes it highly specific in relation to the bone tissue and the possibility of using it as a marker of bone metabolism. The study aimed to assess the BSP level in the saliva of female patients at the menopausal and postmenopausal period. Materials and method. The study included 71 women aged 45–74 years. In the examined subjects, bone mineral density and bone sialoprotein levels in saliva were determined. Patients also responded to survey questions about place of residence and the frequency of consumption of foods rich in calcium. The obtained results were statistically analyzed. Results. In the study population of women, BSP saliva concentration ranged from 0.40–14.97 ng/ml. The highest BSP values were observed in the control group (average – 5.66 ng/ml, median – 6.73 ng/ml,), the lowest in the osteopenia group (average – 3.09 ng/ml, median – 1.71 ng/ml). This relationship was close to statistical significance (p = 0.056). Place of residence of the surveyed women had neither effect on BSP concentration in saliva nor on the value of the T-score ratio. Similarly, there was no statistically significant relationship between the frequency of consumption of foods rich in calcium and BSP and T-score. Conclusions. It was found that the BSP level can be determined in saliva. Biochemical tests of saliva considering bone metabolism are an issue that should be continued in future, as the availability of material (saliva) for laboratory analyses, carries potential diagnostic opportunities.

The starting hypothesis of the article is that the discursive operators of Spanish have not been enabled by grammaticalization processes identical to those of other grammatical components (such as verbal or nominal morphemes or prepositions and conjunctions), since there has been no reduction in the functional scope nor a total bleaching, so that its link with the original lexical items has not disappeared. The consequence is that markers are not integrated into paradigms as closed as grammatical paradigms and both their obsolescence and their renewal, therefore, follow similar patterns to what happens in the evolution of the lexicon. This hypothesis is tried to verify in the article by means of the examination of some discursive markers that, having been used diachronically in Spanish, have been abandoned ina relatively short time.

Introduction. Inflammatory response in Diabetes Mellitus (DM) begins with chronic sub-clinical inflammations as a result of insulin resistance and activation of both innate and adaptive immune system as the disease progresses to complicated diabetes. Hence, the present study investigated the neutrophil phagocytic, complement function (CH50), and some cytokine profiles among diabetic and non-diabetic patients attending the National Hospital in Abuja, Nigeria. Aim. To evaluate the neutrophil phagocytic, complement function (CH50), and some cytokine profiles among post-operative septic diabetic and post-operative septic non-diabetic patients at the National Hospital in Abuja, Nigeria. Material and methods. Subjects were recruited by convenient sampling technique through interviewer-administered questionnaires. Subsequently, blood samples were collected. Fasting blood sugar (FBS) (mmol/L) was determined using glucose oxidase method. Neutrophil function test (Fmol/phag) was assayed using nitroblue tetrazolium reduction test (NBT). Hemolytic complement function (CH 50) test was conducted using serum harvested from sheep sensitized with human group (ORh D +ve) red blood cells. While serum Interleukin-4, -6, -10 and TNF- α were determined using Enzyme Linked Immunosorbent Assay (ELISA). Results. Mean ± Standard deviation (SD) of FBS concentration of 10.5 ± 1.3 (mmol/|L) among diabetic and 4.7 ± 0.9 (mmol/L) among non-diabetics was recorded. There is a decrease in neutrophil phagocytic function with a mean ± SD of 5.4 ± 2.1 (Fmol/ phag) in diabetics compared to 9.2 ± 2.1 (Fmol/phag) in non-diabetics. Similarly, complement (CH 50) function and C-reactive protein were significantly lower in diabetics when compared to non-diabetics (p<0.001). There was a significant difference in IL-6 concentration between diabetics and non-diabetics groups, but no significant difference was observed in TNF-α, IL-4 and IL-10 concentrations between study groups (p>0.05). TNF-α and IL-6 was significantly higher in diabetics with cardiovascular disorders compared to non-diabetics subjects with cardiovascular disorders (p<0.001). Conclusion. Findings from this study revealed the association of complement, neutrophil phagocytic function, CRP and IL-6 among septic diabetic patients,. In addition TNF-α and IL-6 expression was higher in DM patients with cardiovascular disorders.

Keratoconus (KC) is a corneal disease associated with structural abnormalities in the corneal epithelium, Bowman's layer and stroma and altered concentration of tear components. KC corneas show a different pattern of collagen lamellae than their normal counterparts. Also, a reduction of several collagen types in KC epithelium and stroma was observed. Altered expression and/or activity of lysyl oxidase, a critical enzyme of the biogenesis of connective tissue detected in KC corneas, may weaken covalent bonds between collagen and elastin fibrils, what may lead to biomechanical deterioration of the cornea. Increased activity of matrix metalloproteinases observed in KC may induce the degradation of the extracellular matrix causing damage to the cornea. Oxidative and nitrative stress play an important role in KC pathogenesis and KC corneas are characterized by the disturbed lipid peroxidation and nitric oxide pathways. Malfunctioning of these pathways may lead to accumulation of their toxic by-products inducing several detrimental effects, along with apoptosis of the corneal cells, which may result from the loss of β-actin or increased levels of cytokines, including interleukin-1 and -6. Change in the expression of genes associated with wound healing, including the nerve growth factor and the visual system homeobox 1, may contribute to increased susceptibility of KC corneas to injury. Consequently, biochemical changes may play an important role in KC pathophysiology and, therefore, can be considered in prevention, diagnosis, prognosis and in the therapy of this disease as well.

Advanced oxidation protein products (AOPPs) are protein markers of oxidative stress with pro-inflammatory properties that accumulated in liver cirrhosis. In the present study, we investigated the association between chronic inflammatory response triggered by AOPPs and the severity of liver disease as assessed by the Child-Pugh score. Plasma concentrations of AOPPs and inflammatory markers such as C-reactive protein, tumor necrosis factor-α, and interleukin-6 were measured in 41 patients with HCV-related cirrhosis, 43 patients with alcohol-related liver cirrhosis (ALC), and in 30 age and sex matched controls. In comparison with controls, AOPPs were increased in HCV-related compensated (Child-Pugh A) and decompensated (Child-Pugh B-C) cirrhosis and in alcohol-related compensated cirrhosis. AOPPs level positively correlated with Child-Pugh score in alcohol-related cirrhosis but not in HCV-related cirrhosis and the correlation with the indices of chronic inflammation was stronger in ALC. In turn, AOPPs in HCV-related cirrhosis was related to inflammation to a lesser extent, but a significant correlation with antioxidant defense could be noted. In summary, liver cirrhosis was associated with increased formation of AOPPs, which differed between alcohol-related and HCV-related cirrhosis with respect to the relationship between AOPPs and antioxidant defense, stage of liver cirrhosis, and inflammatory response. The significant correlation between AOPPs accumulation and indices of chronic inflammation, more specifically TNF-α, suggests that oxidative stress may be a mediator of chronic inflammatory state in the early stage of alcohol-related cirrhosis.

Sequence tagged site (STS) markers are valuable tools for genetic and physical mapping that can be successfully used in comparative analyses among related species. Current challenges for molecular markers genotyping in plants include the lack of fast, sensitive and inexpensive methods suitable for sequence variant detection. In contrast, high resolution melting (HRM) is a simple and high-throughput assay, which has been widely applied in sequence polymorphism identification as well as in the studies of genetic variability and genotyping. The present study is the first attempt to use the HRM analysis to genotype STS markers in narrow-leafed lupin (Lupinus angustifolius L.). The sensitivity and utility of this method was confirmed by the sequence polymorphism detection based on melting curve profiles in the parental genotypes and progeny of the narrow-leafed lupin mapping population. Application of different approaches, including amplicon size and a simulated heterozygote analysis, has allowed for successful genetic mapping of 16 new STS markers in the narrow-leafed lupin genome.

Due to late diagnosis, ovarian cancer is the most deadly gynecologic malignancy. Inflammation is one of the risk factors of ovarian cancer and the inflammatory response implicates all stages of tumorigenesis. The purpose of this study was to analyze the concentration of molecules, which can take part in malignant processes. We analyzed patients with ovarian cancer, with endometriosis and healthy controls. Thirty-seven analytes were measured in serum using BioPlex Pro Human Inflammation Panel. We were able to detect 28 of the proteins among the studied groups. We found a significant increase in 22 of the tested molecules (BAFF, Chitinase3-like 1, IFN-alpha2, IFN-beta, IFN-gamma, IFN-lambda2, IFN-lambda1, gp130, IL-2, IL-12 (p40), IL-11, IL-32, IL-35, MMP3, Osteocalcin, Pantraxin-3, sCD163, TNFRSF8, sIL-6Ralpha, STNF-R1, STNF-R2, and TSLP) in the ovarian cancer group in comparison to the healthy controls. Two of them (IL-20, MMP1) did not show significant differences between groups. Moreover, we identified decreased concentrations of APRIL and osteopontin in ovarian cancer vs. healthy controls. While this study is a preliminary report, we hope this will encourage a further use of multiplex analysis in ovarian cancer biomarker research.

Opis:
Quercetin and curcumin are plant polyphenolic antioxidants with proven pharmacological efficacy. Their use is, however, limited due to low bioavailability and oral administration route. The encapsulation of lipophilic compounds in liposomes enables to increase their bioavailability and to create an injectable form. The present study aimed to comparatively investigate the antioxidant activity of a complex liposomal preparation containing two lipophilic antioxidants (quercetin and curcumin) and their monopreparations in liposomal forms. This study was conducted on Wistar line rats with experimental model of ischemic heart disease. Oxidative stress markers such as total antioxidant activity, malondialdehyde, and peroxidized proteins were analyzed in blood serum and cardiac tissue. Ischemic heart disease is accompanied with lipid peroxidation and changes in the activity of the antioxidant system. The obtained results demonstrated the antioxidant activity of monopreparations of curcumin and quercetin and their complex in liposomal forms. Quercetin and curcumin showed different antioxidant activities in terms of oxidative stress markers. The complex of the two antioxidants showed the synergistic effect of their lipophilic compounds in liposomal forms, which led to the normalization of test parameters according to the level of the control sample.
