To downregulate expression of the β1 integrin subunit in endothelial cells, plasmid encoding the ribozyme cassette containing hammerhead ribozyme flanked at the 5' terminus by tRNAVal and at the 3' terminus by constitutive transport element sequences was constructed. When used to transfect immortalized human endothelial cell line EA.hy 926, it selectively blocked the synthesis of the β1 integrin subunit and thus inhibited migration and proliferation of the cells. Thus, this construct may be a valuable tool to control the proangiogenic phenotype of stimulated endothelial cells.

Chemical deglycosylation (dg) of sturgeon Acipenser gueldenstaedti Br. (αGTH) resulted in the loss of 83% of its initial carbohydrate content. It altered also recombinant dgαGTH + βGTH dimer molecule, reducing its immunoreactivity by 30%, and fully blocking the hormonal function. CD spectroscopy showed that deglycosylation led to changes in the secondary structure of dgαGTH and in the α-β recombinant. The sugar moiety of sturgeon αGTH is suggested to play an important role in maintaining the biological function of the hormone dimer molecule.

The protein encoded by the IRR1/SCC3 gene is an element of the cohesin complex of Saccharomyces cerevisiae, responsible for establishing and maintaining sister chromatid cohesion during mitotic cell division. We noticed previously that lowering the level of expression of IRR1/SCC3 affects colony formation on solid support. Here we describe two dosage suppressors (IST2, NOG2) overcoming the inability to form colonies of an Irr1p-deficient strain. Ist2 is probably involved in osmotolerance, Nog2p is a putative GTPase required for 60S ribosomal subunit maturation, but may also participate in mRNA splicing.

Paraphysomonas vestita is a unicellular, colorless, silica-scaled chrysophyte that plays an important ecological role in freshwater microbial communities as a consumer of prokaryotic and eukaryotic prey. There is little biogeographical information for this minute protist despite its significant role in aquatic food webs. In addition, the phylogenetic relationship of P. vestita to other taxa is unclear as P. vestita may be polyphyletic or a cryptic species complex. In this study, a clonal isolate from a freshwater sample of Laguna de Bay, Philippines was subjected to morphological study by electron microscopy and its small subunit ribosomal RNA (SSU rRNA) gene was sequenced. Morphological studies showed that the isolate possesses two unequal flagella emerging from the anterior part of the cell. Negative staining revealed the structure of the scales which consist of a baseplate with slightly thickened rim. The narrowing spine arises from the center of the baseplate. These results agree with previously studied isolates of P. vestita. The 18S rRNA gene sequence of the isolate had a very high similarity (99%) to P. vestita strain PV10. Phylogenetic analysis also showed that the isolate clustered with other Paraphysomonas sequences with high bootstrap support. Phylogenetic studies confirmed that P. vestita may be polyphyletic. No studies on the ultrastructure and phylogeny of a silica-scaled chrysophyte isolated in the Philippines have been reported so far. Results from this study may contribute to further ultrastructural and phylogenetic studies on aquatic flagellates and specifically to a revision of this potentially polyphyletic species.

Regulators of G-protein signaling (RGS) proteins are known for the RGS domain that is composed of a conserved stretch of 120 amino acids, which binds directly to activated G-protein α subunits and acts as a GTPase-activating protein (GAP), leading to their deactivation and termination of downstream signals. In this study, a novel human RGS cDNA (RGS21), 1795 bp long and encoding a 152-amino acid polypeptide, was isolated by large-scale sequencing analysis of a human fetal brain cDNA library. Unlike other RGS family members, RGS21 gene has no additional domain/motif and may represent the smallest known member of RGS family. It may belong to the B/R4 subfamily, which suggests that it may serve exclusively as a negative regulator of αi/o family members and/or αq/11. PCR analysis showed that RGS21 mRNA was expressed ubiquitously in the 16 tissues examined, implying general physiological roles.

Service in a special unit of the Police is extremely tedious, both in physical and mental terms, with very high expectations for their policemen. It cannot arise only from official duties, but – above all – specific lifestyle, which should be the primary stimulus for taking them. It is confirmed by research conducted among policemen-antiterrorists, relating to their motivation in taking service in anti-terrorist subunit of the Police. Studies have also shown that such service is not fully in line with the expectations of the officers, and their priorities change over its course. A lot of factors have impact on it, negatively affecting the attitude of policemen, which were revealed in the study. The results can be used to create such conditions of service, which will reduce or eliminate loss of idealistic motives for taking service in the special unit of the Police.

The calcareous nannoplankton and foraminifera from the Gładyszów Beds, regarded as the youngest deposits in the northern marginal Siary Subunit of the Magura Nappe in Poland, are characterized. The material investigated comes from outcrops in the vicinity of Gładyszów and from the Gładyszów PIG-1 borehole. The analysis allowed the establishment of the age of these deposits as not older than late Rupelian–early Chattian (the calcareous nannoplankton NP24 Zone). The taxonomic affiliations, environmental associations and preservation of the microfossils were analyzed to document the sedimentary processes and environmental conditions during the geotectonic transformation of the Magura Basin in the Oligocene. These microfossils also were compared with those reported from the terminal deposits of the Outer Carpathians and the Podhale Basin.

The Magura Nappe in the Polish sector of the Outer Carpathians consists of four tectonic subunits characterized by differing development of facies. From the south to the north, they include the Siary, Rača, Bystrica and Krynica subunits. The sedimentary succession in the Rača Subunit in the vicinity of the village of Osielec is composed of Campanian–Palaeogene flysch deposited in the Magura Basin. In this succession, the Middle Eocene Pasierbiec Sandstone Fm consists of thick-bedded sandstones and conglomerates with occasional intercalations of thin-bedded shale-sandstone flysch. Within the Pasierbiec Sandstone Fm at Osielec there is an olistostrome, rich in pebbles and cobbles of exotic rocks. In addition, large blocks of Neoproterozoic metabasites and boulders of Palaeogene organogenic limestones were found. The discovery of metabasites raised the possibility that the rocks in question could be evidence of supposed oceanic crust in the basement of the Magura sedimentary basin, because of the suggestion that they represent the Alpine orogenic cycle. This concept was abandoned when investigations of the absolute age of the metabasites gave a date of ca. 600 Ma. In the Osielec area, there are two tectonic thrust sheets in the Rača Subunit, namely the Osielczyk Thrust Sheet in the north and the Bystra Thrust Sheet in the south; they are folded and cut by a transverse system of strike-slip and oblique faults. The Osielczyk Thrust Sheet was overthrust northwards on to the Siary Subunit.

The ribosome is a macromolecular assembly that is responsible for protein biosynthesis in all organisms. It is composed of two-subunit, ribonucleoprotein particles that translate the genetic material into an encoded polypeptides. The small subunit is the site of codon-anticodon interaction between the messenger RNA (mRNA) and transfer RNA (tRNA) substrates, and the large subunit catalyses peptide bond formation. The peptidyltransferase activity is fulfilled by 23S rRNA, which means that ribosome is a ribozyme. 5S rRNA is a conserved component of the large ribosomal subunit that is thought to enhance protein synthesis by stabilizing ribosome structure. This paper shortly summarises new results obtained on the structure and function of 5S rRNA.

A new soil ciliate, Pseudouroleptus jejuensis, was discovered from Jeju Island, South Korea and described based on live observation, protargol impregnation, and SSU rRNA gene sequence analyses. Pseudouroleptus jejuensis differs from other congeneric species mainly by number of dorsal kineties (5 vs. 4). Based on our observation of late dividers, we confirm that the dorsal kinety anlage 3 forms 3 kineties (i.e., dorsal kineties 3–5), and the dorsal kinety anlagen 1–3 form 3–5/1–2/0 caudal cirri, respectively. Our gene trees support the assignment of this new species in Pseudouroleptus to full supporting values.

The family Gobiidae is the largest teleost family, consisting of more than 2,000 species in more than 200 genera. They have adapted to a wide range of habitats, inhabiting mostly marine and brackish waters. Recently invasive Ponto-Caspian gobies have caused significant changes to the structure of fish assemblage throughout the European waters. The aim of the study was to verify the possibility of identifying three species of the genus Gobiidae based on the analysis of the mitochondrial region of the cytochrome oxidase subunit 1 (COI) gene. 339 samples originated from seven countries and 17 different sites were investigated. Qualitative and quantitative assessment of the extracted DNA was conducted by measuring light absorbance and electrophoretic separation. Approximately 650-bp (base pairs) fragments were amplified from the 5' region of the mitochondrial COI with the PCR (polymerase chain reaction) reaction. Sequences of 652 bp were obtained and compared with reference sequences in the National Center for Biotechnology Information (NCBI). Based on the sequence analysis, we determined that in the analysed geographical regions, the goby preliminary identified morphologically as the round goby was represented by three species: round goby (Neogobius melanostomus), tubenose goby (Proterorhinus marmoratus), and monkey goby (Neogobius fluviatilis). The latter two are newly reported for their new habitats. Genetic characterisation of such populations offers the opportunity to assess their genetic stability, which provides agencies managing the aquatic environment in EU countries with a tool for revising the legislation, including regulations relevant for updating the status of native and invasive species in European waters.

Recombinant subunit vaccines based on hemagglutinin proteins produced in bacteria (bacterial HAs) are promising candidates for enhancing the supply of vaccines against influenza, especially for a pandemic. Over 20 years after the failure to obtain the antigen with native HA characteristics in the early 1980’s, there are increasing data on successful production of HA proteins in bacteria. The vast majority of bacterial HAs have been based on the HA1 subunit of HA expressed separately or as a component of conjugate vaccines, but those based on the ectodomain and the HA2 subunit have also been reported. The most of HAs have been efficiently expressed as insoluble aggregates called inclusion bodies. Refolded and purified proteins were extensively studied for structure, the ability to bind to sialic acid-containing receptors, antigenicity, immunogenicity and efficacy. The results from these studies contradict the view that glycosylation determines the correct structure of the hemagglutinin, as they proved that bacterial HAs can be valuable vaccine antigens when appropriate folding and purification methods are applied to rationally designed proteins. The best evidence for success in bacterial production of protective HA is that vaccines based on proprietary Toll-like Receptor (VaxInnate) and bacteriophage Qβ-VLPs (Cytos Biotechnology) technologies have been advanced to clinical studies.
