The grape seeds, which are the post-production waste of juices and wine production, are a rich source of biologically active compounds. The polyphenol compounds present in seeds can be used in the technology of new products of animal origin as a source of natural antioxidants. The effect of three levels (0.1, 0.2 and 0.5%) 40% (v/v) ethanol extract of grape seed (GSE) quality of dry-cured pork neck was tested. Meat product colour, pH value, water activity, free fatty acid (FFA) content and TBARS index were evaluated. The obtained results indicate that the addition of the extract decreased oxidation intensity dry-cured neck during ripening. The treatment with higher concentrations of extracts showed greater lipid stability than that of the lot with 0.1%. The three levels of GSE extract did not influence pH, water activity and colour significantly. This study indicates that the GSE extract was an effective inhibitor of lipid hydrolysis, and at an additive concentration of at least 0.2% limited the oxidative processes occurring in meats during ripening. Therefore, it may be used in dry-cured pork neck to improve the oxidative stability of meat products for the consumers.

Both seed and root extracts of the medicinal plant, Ashwagandha, Withania somnifera exhibit insect growth regulatory activity against the polyphagous pest, Pericallia ricini. Topical administration of W. somnifera seed and root extracts to last instar larvae of P. ricini disrupted moulting and metamorphosis, leading to a number of developmental abnormalities such as delay in larval-pupal and pupal-adult ecdysis, formation of larval-pupal, pupal- -adult and larval-pupal-adult mosaics/chimeras, ecdysial failure, suppression of pupation and adult emergence and formation of abnormal pupae and adultoids. The treatment with seed extracts was more severe than that of root extracts as it completely suppressed the pupation and adult emergence. The results clearly suggest that the medicinal plant, W. somnifera acts as a potential insect growth regulatory (IGR) disrupting the moulting and metamorphosis as a consequence of interference with the endocrine system.

Opis:
Additional approaches to control malignancies are needed due to the emerging trends in the incidence of cancer of different organ sites. Due to the high frequency of hepatocellular carcinoma (HCC) and its poor prognosis, preventing HCC is an urgent priority. To explore the antioxidant and apoptotic pathways of grape seed extract (GSE) we induce HCC experimentally by diethylnitrosoamine (DEN) and treated the animals with low and high doses of GSE. The results indicate good therapeutic possibilities for GSE use in treatment of HCC., This was evidenced via regression of liver enzymes' function (ALT&AST), the HCC markers; α-fucosidase, α-fetoprotein and carcinoembrionic antigen (CEA) in HCC groups treated with the grape seed extract. Also, tumor necrosis factor (TNF-α) showed a significant decrease using GSE in HCC bearing animals. Regarding the apoptotic pathways of GSE, we found a significant down regulation of apoptosis enhancing nuclease (Aen), Bcl2-associated X protein (Bax), B-cell translocation gene 2(Btg2), Cyclin G1 (Ccng1) and Cyclin-dependent kinase inhibitor 1A (Cdkn1a) gene expression in HCC+GSE groups as compared to HCC bearing group. In the same trend, the necrotic/apoptotic rates were significantly higher in the HCC groups treated with GSE vs. the HCC bearing group. Finally, the 8-OHdG/2-dG generation decreased by 73.8% and 52.9% in HCC+GSE at low and high doses, respectively. Based on these encouraging observations, grape seed extract could be a promising natural remedy for attenuating hepatocellular carcinoma that has a great future in approaches directed towards control of HCC.

The tomato leaf miner, Tuta absoluta Meyrick (Lepidoptera: Gelechiidae), is one of the most destructive pest of solanaceae and it prefers tomato (Solanum lycopersicum L.). The aim of the current study was to investigate the effects of a wide range of seed proteinaceous extracts from different plant families against T. absoluta α-amylase activity. The effect of pH on the inhibitory activity of seed extracts showed that seed extracts of amaranth along with a wheat cultivar (Alvand, Aflak, Sarvdasht, Alborz, and Kavir) produced more than a 50% inhibition of the insect amylase. Aflak wheat seed extract at 10 μg, inhibited 81% of the insect amylase. This percent was the highest inhibition achieved. The other proteinaceous seed extracts had a lower effect on the enzymatic activity. Probit analysis showed that Aflak, Kavir, Alborz, Alvand, Sarvdasht, and amaranth inhibited the amylase activity with an I50 of 1.94, 3.24, 3.46, 3.31, 4.97, and 15.39 μg, respectively. The effect of pH on the inhibition of the α-amylase showed the highest inhibition of Amaranth and wheat, at a pH value of 8.0, which corresponds to the pH of the insect’s gut. Gel electrophoresis assays confirmed the spectrophotometric assays showing that the α-amylase of the insect gut was affected by the presence of the seed extracts. In the gel assay, a high concentration (14 μg protein) of amaranth proteinaceous seed extract greatly decreased the intensity of the α-amylase band. A high concentration of the Aflak wheat cultivar (10 μg protein) caused the disappearance of the amylase band in the gel. Thus, it is concluded that the physiochemical environment of the insect gut affects the interaction between digestive α-amylase and the metabolites. The experiments showed that seed proteinaceous extracts from non-host plant species, produced more inhibition of the insect amylase when compared to the host plant species. It appears that with evolution, adaptation took place so that insect/s could overcome the plant metabolites.

Ceruloplasmin (Cp) is the major antioxidant in plasma, a protein that carries about 95% of the total copper. Multiple biochemical activities of ceruloplasmin have been described, including copper transport or oxidation of Fe(II) to Fe(III) for subsequent uptake by transferrin and ferritin. Reduction of the pool of free Fe(II) ions by Cp prevents the generation of free radicals and reactive oxygen species by oxidation of Fe(II), thus inhibiting the Haber-Weiss reaction. The removal of both free Fe(II) and Fe(III) ions from blood plasma by polyphenols enhances the antioxidant system of the living organism. However, the mechanism of interactions between exo- and endogenous antioxidants is still under consideration. The effect of raspberry seed extract (RSE) on the ferroxidase activity of ceruloplasmin isolated from plasma of patients with chronic arterial occlusion of the lower limbs due to atherosclerosis (CpAO) was investigated. Moreover, the effect of RSE on the ferroxidase activity of Cp isolated from healthy volunteers (CpC) was also estimated. The ferroxidase activity of Cp, expressed as ΔFe(II), was determined by spectrophotometry with the use of the Fe(II) – histidine complex and ferrozine as a chromogenic reagent. The addition of RSE to samples with the same amount of both CpAO or CpC in each caused an increase in ΔFe(II). The polyphenol-rich RSE may assist Cp in the fight against free radicals and reactive oxygen species when the disease occurs due to an excessive use or reduced production of endogenous antioxidants.

Chociaż chemiczne czynniki wzrostu są skutecznie stosowane do polepszenia kiełkowania nasion, wschodów i jakości sadzonek u różnych gatunków roślin, to nie są one jeszcze używane w uprawie organicznej. Aksamitka jest rośliną o wielu zastosowaniach: jest jednocześnie źródłem luteiny i kwasu galusowego i jako taką warto badać ją jako organiczny czynnik wzrostu. Celem niniejszego badania było zidentyfikowanie organicznego czynnika wzrostu do uprawy organicznej oraz opracowanie właściwych metod poprzez zastosowanie dwóch różnych gatunków aksamitki (Tagetes patula L. i T. erecta L.). Herbatka ziołowa przygotowana z suszonych kwiatów aksamitki została użyta jako czynnik wzrostu nasion bakłażana (Solanum melongena L) odmiany Pala. Porównano hydrokondycjonowanie oraz nasiona kontrolne z pobudzaniem organicznym (T. patula L. i T. erecta L.). pod względem procentu kiełkowania, średniego czasu kiełkowania, wskażnika kiełkowania, dwóch testów wschodów, wysokości sadzonek oraz świeżej i suchej masy sadzonek. Dane uzyskane po zastosowaniu organicznego pobudzania wzrostu (T. patula L. i T. erecta L.) wykazały różnicę zarówno w kiełkowaniu (82 i 80%), jak i wschodach (73 i 76%) pochodzących z hydrokondycjonowania i kontroli. Nasiona wzeszły, odpowiednio, w 52 i 39%, natomiast gdy były pobudzane za pomocą T. Patula, wzeszły, odpowiednio, w 73 i 56%. ĝredni czas kiełkowania oraz średni czas wschodów były krótsze w porównaniu z kontrolą i hydrokondycjonowaniem. MET zmniejszył się na głębokości siewu 1 cm i 2,5 cm odpowiednio z 14 i 16,1 dni do 10,3 i 11,9 dni dla wschodów oraz z 10,4 do 10,2 dni. Zbadano jakość sadzonek i ich wysokość, świeża i sucha masa T. patula L. i T. erecta L. były także statystycznie istotne. Niniejsze badanie jest pierwszym, którego wyniki wykazały, że gatunki aksamitki moga być stosowane jako potencjalny czynnik organiczny pobudzający wzrost.
Although chemical priming agents have been successfully used to improve seed germination, emergence and seedling quality in different plant species, still they are not yet useful for organic cultivation. Marigold is a multipurpose plant and rich source of lutein and gallic acid, to be explored as an organic priming agent. The purpose of this study was to identify an organic priming agent for organic growing and to develop appropriate methods by using two different species of marigold (Tagetes patula L. and T. erecta L.). Herbal tea obtained from the dried flower of marigold was used as a priming agent for eggplant (Solanum melongena L) cv. Pala seeds. Hydropriming and control seeds were compared with organic priming (T. patula and T. erecta) for germination percentage, mean germination time, germination rate, two different emergence tests, seedling height, seedling fresh and seedling dry weights. Data obtained from the application of organic priming (T. patula and T. erecta) showed a difference in both the germination (82 and 80%), and the emergence (73 and 76%) from Controls and Hydropriming. Seeds emerged in, which unprimed in 52 and 39% respectively, when primed with T. patula, emerged in 73 and 56% respectively. The mean germination time and the mean emergence time were shortened compared with Controls and Hydropriming. MET decreased in 1 cm and 2.5 cm depth sowing from 14.0 and 16.1 days than 10.3 and 11.9 days in Emergence-1 and 10.4 and 10.2 days in Emergence 2, respectively. Seedling quality characteristics were examined in the seedling height, the seedling fresh and dry weights of T. patula and T. erecta applications were also statistically significant. This is the first report which results showed that marigold species can be used as a potential organic priming agent.

Oxidative stress has been recognized as a critical pathogenetic mechanism for the initiation and the progression of hepatic injury in a variety of liver disorders. Antioxidants, including many natural compounds or extracts, have been used to cope with liver disorders. The present study was designed to investigate the hepatoprotective effects of cassia seed ethanol extract (CSE) in carbon tetrachloride (CCl4)-induced liver injury in mice. The animals were pre-treated with different doses of CSE (0.5, 1.0, 2.0 g/kg body weight) or distilled water for 5 days, then were injected intraperitoneally with CCl4 (0.1% in corn oil, v/v, 20 ml/kg body weight), and sacrificed at 16 hours after CCl4 exposure. The serum aminotransferase activities, histopathological changes, hepatic and mitochondrial antioxidant indexes, and cytochrome P450 2E1 (CYP2E1) activities were examined. Consistent with previous studies, acute CCl4 administration caused great lesion to the liver, shown by the elevation of the serum aminotransferase activities, mitochondria membrane permeability transition (MPT), and the ballooning degeneration of hepatocytes. However, these adverse effects were all significantly inhibited by CSE pretreatment. CCl4-induced decrease of the CYP2E1 activity was dose-dependently inhibited by CSE pretreatment. Furthermore, CSE dramatically decreased the hepatic and mitochondrial malondialdehyde (MDA) levels, increased the hepatic and mitochondrial glutathione (GSH) levels, and restored the activities of superoxide dismutase (SOD), glutathione reductase (GR), and glutathione S-transferase (GST). These results suggested that CSE could protect mice against CCl4-induced liver injury via enhancement of the antioxidant capacity.

The present experiment was carried out in the period 2006–2008. The aim of this study was to determine the effect of aqueous soil extracts from the soil of a spring wheat monoculture on seed germination energy and capacity, the length of the first leaf and of the longest radicle as well as the number of radicles. Moreover, the content of 0-dihydroxyphenols in the soil was compared in the last year of the study. The soil used to prepare the solutions came from a field experiment established on medium heavy mixed rendzina soil. Spring wheat, cv. Zebra, was grown using plough tillage and two conservation tillage methods in the presence of undersown crops (red clover, Westerwolds ryegrass) and stubble crops (lacy phacelia, white mustard). Germination energy of the seeds watered with the soil extracts from the ploughed plots was significantly higher than this trait in the seeds watered with the extracts from the conservation tillage treatments with spring disking of the catch crops. Germination energy and capacity of spring wheat in the control treatment watered with distilled water were significantly higher compared to the other treatments under evaluation. Spring wheat watered with the aqueous extract prepared from the soil obtained from the plough tillage treatment produced a significantly longer first leaf compared to the treatments in which both conservation tillage methods had been used. The shortest leaf and the lowest number of radicles were produced by the seedlings watered with the soil extract from the treatment with the white clover stubble crop. Radicle length was not significantly differentiated by the soil extracts under consideration. The content of 0-dihydroxyphenols in the rendzina soil determined during the spring period was higher than that determined in the autumn. The content of 0-dihydroxyphenols in the soil was lower in the conservation tillage treatments with autumn incorporation of the catch crops than in the plots in which plough tillage and conservation tillage with spring disking of the catch crops had been used. The type of catch crop used did not have a significant effect on the soil content of these compounds. At the same time, it was found that the treatments in which the catch crops had been sown tended to have higher contents of these compounds compared to the plots without catch crops.

Ions of irons, especially ferrous ions may be harmful for living organisms, because they generate reactive oxygen species like O2 •- or •OH. Probability of the risk rises especially in pathological conditions, in which high level of iron is observed. For this reason scientists try to establish new methods that can support organism in eliminating reactive ferrous ions. Nowadays, attention focuses on substances present in plants, especially polyphenols, whose administration prevents oxidative damages in iron overloading. This new approach requires some research on behavior of plant-derived compounds in human organism, within a system containing other biomolecules involved in iron metabolism. The aim of this study has been to investigate the influence of black currant (Ribes nigrum) seed extract, a source of polyphenols, on the activity of ceruloplasmin, an enzyme participating in Fe(II) elimination from blood plasma in human organism. Depletion of Fe(II) caused by ceruloplasmin isolated from healthy blood donors was compared to its decrease in a system containing both ceruloplasmin and the extract. The results have shown that addition of a particular amount of the extract elevates the effectiveness of ceruloplasmin in eliminating Fe(II) from the sample but only under physiological condition (pH 7.4; T 37°C). In a weak acidic solution, addition of the extract does not lead to any change in Fe(II) concentration.
Jony żelaza, szczególnie jony żelazawe Fe(II), ze względu na wysoki potencjał oksydoredukcyjny mają zdolność generowania wolnych rodników, takich jak O2 •- i •OH. W pewnych stanach patologicznych, którym towarzyszy przeładowanie organizmu żelazem, ryzyko pojawienia się wolnych jonów Fe(II) inicjujących wiele procesów wolnorodnikowych jest bardzo prawdopodobne. W tym celu dąży się do opracowania terapii, które w chorobach żelazozależnych wspomogą naturalne mechanizmy usuwania reaktywnych jonów żelaza z organizmu. Ostatnio dużą uwagę poświęca się aktywnym substancjom obecnym w roślinach, szczególnie związkom polifenolowym, ze względu na ich silne właściwości chelatujące. Ten nowy kierunek badań wymaga jednak wnikliwej analizy, która wyjaśni mechanizm współdziałania substancji roślinnych z innymi biomolekułami zaangażowanymi w prawidłowy metabolizm żelaza w organizmie oraz rozstrzygnie o bezpieczeństwie ich stosowania. Celem badań było ustalenie wpływu ekstraktu z pestek porzeczki czarnej (Ribus nigrum), bogatego w związki polifenolowe, na aktywność ludzkiej ceruloplazminy, enzymu uczestniczącego w usuwaniu Fe(II) w organizmie ludzkim. Ubytek Fe(II) obserwowany w obecności ceruloplazminy porównywano z jego ubytkiem w mieszaninie ceruloplazmina-ekstrakt. Stwierdzono, że dodatek ekstraktu podnosi skuteczność ceruloplazminy w eliminowaniu Fe(II) ze środowiska. Istnieje więc przypuszczenie, że związki pochodzenia roślinnego takie jak polifenole mogą wspomagać naturalne mechanizmy eliminowania Fe(II) w organizmie.
