Opis:
Solanum dulcamara L. (bittersweet) is a medicinal plant that has been used to treat skin diseases, warts, tumors, felons, arthritis, rheumatism, bronchial congestion, heart ailments, ulcerative colitis, eye inflammations, jaundice and pneumonia. A reliable in vitro culture protocol for bittersweet was established. Explants (leaf and petiole segments) were cultured on Murashige and Skoog minimal organics (MSMO) medium with various plant growth regulator combinations. Leaf explants formed more shoots than petiole explants. Plant regeneration was observed through indirect organogenesis with both explants. Best shoot proliferation was obtained from leaf explants with 3 mg/l BA (benzyladenine) and 0.5 mg/l IAA (indole-3-acetic acid). Regenerated shoots were transferred to rooting media containing different levels of IAA (indole-3-acetic acid), IBA (indole-3-butyric acid), NAA (naphthalene acetic acid) or 2,4-D (2,4 dichlorophenoxyacetic acid). Most shoots developed roots on medium with 0.5 mg/l IBA. Rooted explants were transferred to vermiculate in Magenta containers for acclimatization and after 2 weeks, they were planted in plastic pots containing potting soil and maintained in the plant growth room.

We aimed to produce tissue cultures and plant regeneration from endangered Crocus species: C. scepusiensis, C. tommasinianus, C. vittatus (“Verni” series of the genus) and C. banaticus. For initiation of cultures we used a plant growth regulator (PGR) combination used for in vitro culture of saffron and its relatives: 10 mg L-1 α-naphthaleneacetic acid (NAA) and 1 mg L-1 6-benzyladenine (BA). Shoot tips of young seedlings (C. scepusiensis) and corms (for the rest of species) were used as explants. C. scepusiensis explants developed into organogenic calli. On media with decreased NAA and with or without increased BA concentration, calli produced stigma-like structures and/or shoots and whole plants. In the other species, callus initiation medium induced callus formation with abundant somatic embryos. In C. tommasinianus, embryos developed shoots when auxin content of medium was decreased. In C. banaticus, a decrease of auxin with or without an increase in cytokinin content led to shoot or whole plant regeneration, as in C. scepusiensis. In the case of C. vittatus and C. banaticus, initiation and/or maintenance of cultures on indole-3-butyric acid (IBA) and increased sucrose concentration stimulated whole plant regeneration and in vitro cormlet development. C. scepusiensis and the rest of cultures (organogenic vs. embryogenic) differed at the biochemical level: C. scepusiensis cultures had higher (yet still low) enzymatic antioxidant (catalase, peroxidase) activities. With respect to catalase isoenzyme patterns, C. banaticus was different from the rest of cultures, demonstrating its distinct taxonomical position. Besides germplasm preservation use of the present cultures, they have a potential biotechnological value.

Opis:
Isatis concstricta Davis (an endemic plant of Turkey) suffers from low propagation rates under natural conditions and is threatened due to fast unplanned urbanisation. The study compared the effects of variants of BA + NAA on shoot regeneration on leaf and petiole explants excised from one week old in vitro regenerated seedlings. MS medium containing 1 mg L–1 BA + 1 mg L–1 NAA induced maximum proliferation on petiole and leaf explants with 13.33 and 12.75 shoots per explant repectively. However, leaf explant induced shoots were sturdy and healthier compared to petiole explant induced shoots. These shoots were rooted on MS medium containing 0.5 mg L–1 IBA and the plants were acclimatized in peat moss and sand (v/v). They grew to flowering under ex vitro conditions. This system of regeneration is advantageous for conventional propagation and the results will help in establishment of a powerful and meaningful micropropagation system for I. constricta.

Anthers of twenty triticale genotypes were cultured on three different media: 1 - PII (Chuang et al. 1978) with increased 2,4-D to 2 mg L⁻¹ and agarose 6 g L⁻¹ , 2 - Macro-, micronutrients and vitamines like in MN6 (Chu, Hill 1988) + 2 mg L⁻¹ 2.4-D + 0.5 mg L⁻¹ KIN + 5 mg L⁻¹ FeEDTA + 90 g L⁻¹ sucrose; 3 - Macro-, micronutrients and vitamines like in MN6 (Chu, Hill, 1988) + 2 mg L⁻¹ 2.4-D + 0.5 mg L⁻¹ KIN + 5 mg L⁻¹ FeEDTA + 120 g L⁻¹ sucrose. Embryoid induction and plant regeneration were influenced by donor plant genotype and induction medium. Medium 1 was the best for embryoid induction, while for green plant regeneration the best were media 1 and 2. Out of 300 anthers from each genotype plated on each of the three media, 64-1250, 12-486 and 6-212 somatic embryos and 8-86, 3-136 and 1-26 green plants were recorded, on media 1, 2 and 3, respectively.

The list of 46 self-sowing alien woody plant taxa in Rogów Arboretum is presented, which is an expantion of the list from 1992 when the first article on spontaneous regeneration of alien tree and shrub taxa in the Arboretum was published. Most of species described are rather infrequently found in Polish collections and are known to produce self-seedlings not capable of sexual reproduction, however Abies grandis can be included into group of plants that could produce seedlings capable of sexual reproduction.

In the preceding research, stevia has been typically cloned in vitro using two media, on which the shoots were formed (3–6 weeks), and on the other they were rooted (3–5 weeks). This study aimed at finding the possibility for rapid stevia propagation from large nodal explants using the MS basal medium [Murashige and Skoog 1962], with low auxin concentrations (0.5, 1 and 2 mg.dm–3). The plants were obtained as soon as after three weeks. The best results were obtained from media with various concentrations of the indole-3-acetic acid (IAA) and the highest concentration of phenylacetic acid (PAA). Plants were formed by 83.9–86.0% of explants, they had high weight (234–253 mg), two shoots measuring 2.07–2.37 cm and 5.8–8.3 roots measuring 1.00–1.24 cm. Mean plant weight was the lowest on media with indole-3-butyric acid (IBA) (185–192 mg). Both explant buds formed single shoots, but their development was typically uneven. The differences in the length and weight of shoots were the lowest on media with IAA and at lower PAA concentrations. Plants from the media with IAA and the control medium were distinguished by higher number of nodes. The percentage share of shoots in the total plant weight was the highest on media with PAA (62.1–62.7%), and the lowest at higher concentrations of α-naphthaleneacetic acid (NAA) (47.9 and 48.9%). Parts of explants immersed in media developed callus, and the highest amounts of this tissue were found in the media with NAA. 92.3% of plants survived the acclimatization. The applied procedure may be used for rapid in vitro cloning of selected stevia genotypes. The use of one medium enables reduction of seedling production costs. Moreover, cyclical cloning and extending the production scale is possible.

The influences of genetic and environmental factors on the anther culture responses of wheat were investigated. Significant differences for callus induction, plant regeneration, and green plant percentages were observed when the nucleus of Triticum acstivum L. cv. Selkirk was transferred to ten alien cytoplasms by substitution backcrosses. In most cases, the alien cytoplasms decreased anther culture responses, but sometimes they were as good as or better than the T. aestivum cytoplasm. Significant within-genotype variation for anther culture responses were observed for wheat varieties Chris, Yecora Rojo, WA7176 and Edwall, indicating genetic heterogeneity in the present commercial cultivars, and potential for improving anther culture responses by in vitro prescreening. When five genotypes (Chris, Pavon 76, Butte 86, WA6916, and Edwall) were cultured across three (potato-4 liquid, 100 g L⁻¹ ficoll-supplemented, and 6 g L⁻¹ agar-solidified) induction media, the liquid and ficoll-containing media were 10 to 15 times more productive than the agar-solidified medium. Whereas, the ficoll medium was not significantly different from the liquid medium. Several low concentration starch media appeared promising to replace current induction media. The starch media sustained the highcallus-induction properties of the liquid medium, while improving callus aeration similar to that observed on solid media, resulting in markedly higher plant regeneration and green plant percentages.

In vitro organogenesis was studied using Citrus limon L. Burm cv. ‘Primofiore’ leaf explants. The purpose of the present study was to optimize conditions for callogenesis and organogenesis of C. limon. Explants of C. limon were cultured on 16 different media supplemented with various combinations of plant growth regulators, both auxins and cytokinins, such as 6-benzylaminopurine (BAP), naphthalene acetic acid (NAA), 2,4-dichlorophenoxyacetic acid (2,4-D), and kinetin. The best shoot induction was obtained when the leaf explants were cultured on Murashige & Tucker media supplemented with 3.5 mg·L-1 BAP. Histological investigation revealed most likely the initial phase of development of leaf explants during in vitro regeneration of C. limon. t
Organogenezę in vitro badano u Citrus limon L. Burm cv. ‘Primofiore’, wykorzystując eksplantaty z liści. Celem badań była optymalizacja warunków do indukcji tkanki kalusowej oraz regeneracji roślin C. limon. Eksplantaty C. limon wykładano na 16 pożywek różnego rodzaju, które dodatkowo uzupełniano kombinacjami roślinnych regulatorów wzrostu, zarówno auksyn, jak i cytokinin, takimi jak 6-benzylaminopuryna (BAP), kwas naftalenooctowy (NAA), kwas 2,4-dichlorofenoksyoctowy (2,4-D) i kinetyna. Regenerację pędową uzyskano, gdy eksplantaty liściowe wykładano na pożywkę Murashige and Tuckera uzupełnioną 3,5 mg·L-1 BAP. Badania histologiczne pozwoliły na przedstawienie początkowej fazy rozwoju eksplantatów z liści w trakcie regeneracji in vitro C. limon.
