The effect of methyl jasmonate (JA-Me) and phenolic acids: trans-cinnamic acid (t-CA), p-coumaric acid (p-CA), salicylic acid (SA) as well as naringenine (NAR) on growth of seedlings and accumulation of anthocyanins in common buckwheat (Fagopyrum esculentum Moench) were studied. JA-Me and phenolics were applied to growth medium of 4-days etiolated buckwheat seedlings before their exposition to day/night (16h/8h) conditions. The increase of primary roots and hypocotyls length were measured after 3 days of seedling growth in such conditions. At the end of experiment the total anthocyanins contents were measured as well. Methyl jasmonate (JA-Me) and trans-cinnamic acid (t-CA) inhibited growth of the primary root in young buckwheat seedlings, while naringenine (NAR) had a stimulatory influence, and p-coumaric acid had no effect at all. None of investigated phenolics or JA-Me had an effect on the growth of buckwheat hypocotyls, except the mixture of JA-Me and p-coumarcic acid. JA-Me significantly decreased the anthocyanins level in buckwheat hypocototyls, but not in cotyledons. trans-Cinnamic acid, p-coumaric acid and naringenine had no significant influence on the anthocyanin level in hypocotyls and cotyledons of buckwheat seedlings. Simultaneous treatment of buckwheat seedlings with JA-Me and t-CA or p-CA did not change the inhibition of anthocyanins accumulation in buckwheat hypocotyls by JA-Me. In the hypocotyls of buckwheat treated with a mixture of JA-Me and NAR, or SA, a synergistic reduction of anthocyanins was observed.
Badano wpływ jasmonianu metylu (JA-Me) i kwasów fenolowych: trans-cynamonowego (t-CA), p-kumarowego (p-CA), salicylowego (SA) oraz naryngeniny (NAR) na wzrost siewek i akumulację antocyjanów w gryce zwyczajnej (Fagopyrum esculentum Moench). JA-Me i związki fenolowe zastosowano na 4-dniowe siewki wyrosłe w ciemności przed ich wystawieniem na warunki dnia/nocy (16h/8h). Badano także przyrost długości korzeni głównych oraz hypokotyli po 3-dniowym okresie wegetacji w takich warunkach. Wówczas też oznaczono sumaryczne zawartości antocyjanów. JA-Me i t-CA hamowały, podczas gdy NAR stymulowała wzrost korzeni głównych, zaś p-CA nie miał wpływu na ich wzrost. Badane związki fenolowe i JA-Me nie wpływały na wzrost hipokotyli siewek gryki zwyczajnej, za wyjątkiem jednocześnie zastosowanych JA-Me i kwasu p-kumarowego, które działały hamująco. JA-Me istotnie obniżał poziom antocyjanów w hipokotylach gryki, ale nie wpływał na ich zawartość w liścieniach. Kwas trans-cynamonowy, p-kumarowy i naryngenina nie miały wpływu na zawartość antocyjanów w hypokotylach i liścieniach siewek gryki. Jednoczesne zastosowanie JA-Me i t-CA lub p-CA nie wpłynęło na akumulację, podczas gdy użycie JA-Me wraz NAR lub SA spowodowało synergistyczne obniżenie zawartości antocyjanów w hipokotylach siewek gryki zwyczajnej.

Opis:
Kwasy fenolowe należą do istotnych składników roślinnych surowców żywnościowych ze względu na ich przeciwutleniające właściwości. W pracy oznaczono ogólną zawartość związków fenolowych oraz zidentyfikowano i dokonano ilościowej analizy następujących fenolokwasów: ferulowego, kawowego, sinapinowego i p-kumarowego w słodzie wyprodukowanym z dwóch odmian jęczmienia. W największych ilościach występował kwas ferulowy. Ponad 90% kwasów fenolowych występowało w postaci związanej. Dokonano również oceny właściwości przeciwrodnikowych kwasów fenolowych zidentyfikowanych w słodzie. Największą aktywność przeciwrodnikową w teście z DPPH' wykazał kwas sinapinowy.
Phenolic acids are interesting constituents of foodstuff of plant origin due to their antioxidant activity. Total phenolic compounds were determined and four phenolic acids (ferulic, caffeic, sinapic and p - coumaric) were identified and quantified in malt from two barley varieties. Ferulic acid has been identified as the most important quantitatively. Insoluble bound phenolic acids covered over 90% of total phenolic acids content. The antiradical efficiency of phenolic acids of malt was investigated using method based on the reaction with DPPH·. Sinapic acid appeared as the most effective radical scavenger.

We examined whether allelochemical stress leads to increased lipoxygenase activity in roots of sweet maize (Zea mays L. ssp. saccharata), pea (Pisum sativum L.) and radish (Raphanus sativum L. var. radicula). The lipoxygenase activity of soluble and membrane-bound fractions was assessed in roots after exposure to ferulic and p-coumaric acids. Lipid peroxidation and membrane injury were determined as indicators of stress. Increased lipoxygenase activity of both studied fractions was followed by lipid peroxidation and plasma membrane injury. The results suggest the key role of lipoxygenase in plasma membrane injury during allelochemical stress caused by administration of hydroxycinnamic acids.

Introduction and aim. Stored erythrocytes develop lesions involving changes in their structure and function reducing their efficacy. Oxidative Stress (OS) being one of the main causes of storage lesion, can be attenuated by antioxidants as additives in the storage solution. This study aims to evaluate the effect of p-Coumaric acid (CA) on erythrocytes during whole blood storage. Material and methods. Blood collected from Male Wistar rats was stored at 4°C in CPDA-1 solution for 21 days. Blood samples were stored with and without 1mM CA (CA 1) and 10 mM CA (CA 10). The erythrocytes were isolated every week during storage and the biomarkers for OS and antioxidant status were analysed. Results. Superoxide dismutase and catalase elevated on day 14. Conjugate dienes decreased in CA 10 on day 14. Thiobarbituric acid reactive substances increased on day 7 and decreased on day 14 in CA groups. Protein sulfhydryls decreased in controls and CA 1 on day 14 whereas, it was maintained in CA 10. Conclusion. Coumaric acid upregulated the antioxidant enzymes and protected the cells from oxidative damage. Thus, coumaric acid can be employed as a potent additive during storage and opens new avenues of employing it in similar OS situations in erythrocytes

Natural phenolic acids are commonly present in plants consumed in the diet. Recently we have observed that different natural phenolic acids exert differential effects on the body mass gain in ovariectomized and non-ovariectomized female rats. The aim of the present study was to investigate the effects of ferulic, caffeic, p-coumaric and chlorogenic acids on serum estradiol and total cholesterol levels in ovariectomized and non-ovariectomized rats. The experiments were carried out on 3-month old female Wistar Cmd:(WI)WU rats, divided into following groups (n=8 in each group): non-ovariectomized control rats and non-ovariectomized rats receiving ferulic, caffeic, p-coumaric or chlorogenic acids, sham-operated control rats, ovariectomized control rats and ovariectomized rats receiving the same phenolic acids. The phenolic acids were administered at a dose of 10 mg/kg p.o. daily for 4 weeks. Serum estradiol and total cholesterol levels on the next day after the last administration of the phenolic acids were examined. The phenolic acids did not affect serum estradiol or total cholesterol levels in non-ovariectomized rats. In ovariectomized rats, caffeic acid and to a lesser extent p-coumaric acid increased serum estradiol level, which effect correlated with a decreased body mass gain. All the phenolic acids decreased serum cholesterol level in ovariectomized rats. Concluding, the anti-obesity activity of some phenolic acids may be, at least partially, connected with estrogenic pathways.

In this work an attempt is made to analysis of the possible different conformers of p-coumaric acid (PCA) by using density functional method. The total energy of four possible conformers were calculated by using B3LYP/6-311G(d,p) method. Computational result identifies that the most stable conformer of PCA is C2. The formation of inter- and intra-molecular hydrogen bonding between -OH and -COOH group gave the evidence for dimer formation for PCA molecule. The highest occupied-lowest unoccupied molecular orbital analysis shows that the negative electrostatic region situated over the -COOH group and positive electrostatic potential region are localized on ring system and all hydrogen. The PCA has been screened to anti-microbial activity and found to exhibit anti-bacterial effects. Molecular docking results suggest that PCA may exhibit inhibitory activity against lung cancer protein and may act as potential against lung cancer.

Shoot-differentiating callus culture of Ruta graveolens ssp. divaricata (Tenore) Gams was maintained on four variants of Linsmaier-Skoog (L-S) medium containing different concentrations of plant growth regulators, 1-naphthaleneacetic acid and 6-benzylaminopurine, ranging from 0.1 do 3.0 mg/l. Methanolic extracts of biomass cultured in vitro were used to determine the contents of free phenolic acids with an HPLC method. Out of eight compounds under analysis, six were shown to be present in the samples: caffeic acid, chlorogenic acid, p-coumaric acid, p-hydroxybenzoic acid, protocatechuic acid and syringic acid. Total contents of the compounds under study ranged from 124.9 to 138.7 mg/100g d.w., depending on the L-S medium variant. p-Coumaric acid was the main compound in the extracts from biomass cultured on all tested medium variants and its amounts were interesting from practical perspective (97.3-112.9 mg/100g d.w.). This compound was isolated, purified and its identity was confirmed by spectral methods. The contents of other compounds did not exceed 11.1 mg/100g d.w. Extracts of the above-ground parts of plants growing in vivo (stems, leaves, herb), analyzed for comparison, contained three phenolic acids: protocatechuic acid (52.0-88.2 mg/100g d.w.), vanillic acid (2.6 – 17.8 mg/100g d.w.) and trace amounts of syringic acid. This is the first report on the effect of growth regulators on the accumulation of free phenolic acids in biomass of R.graveolens cultured in vitro. This is also the first report documenting the isolation of p-coumaric acid from biomass of this subspecies cultured in vitro and also from any plant in vitro culture. In addition, for the first time in the above-ground parts of the plants growing in vivo phenolic acids were analyzed.
