COVID-19-associated ARDS (cARDS) results from multiple pathogenetic mechanisms involving both parenchyma and circulation of the lungs. Despite authors described two pneumonia phenotypes according to lung elastance values, reported as Low and High phenotype, the evidence suggests that patients with cARDS have similar respiratory mechanics to patients with ARDS from other causes. Consequently, the proper management of patients with cARDS is the same as ARDS,consisting of protective mechanical ventilation strategy and prone position ventilation.However, the hypothesis that impaired alveolar perfusion could have a role in cARDS is interesting. Nitric oxide (NO) is a biatomic gaseous moleculeables to induce smooth muscle relaxation and vasodilation. So, inhaled nitric oxide (iNO) acts as a selective vasodilator because it only dilates vessels in areas participating in gas exchange, preserving hypoxic pulmonary vasoconstriction reflex and reducing ventilation/perfusion mismatch. With this narrative review we summarised the role of iNO for cARDS treatment to improve gas exchange. Furthermore, we briefly described its activity as an antiviral agent. In conclusion, although iNO administration can represent a valid theoretical therapeutic choice for the treatment of severe unresponsive hypoxemia in COVID-19 patients, data presented in the literature are scares and inconclusive, and several clinical trials are still ongoing. Further studies are needed to elucidate the pathogenetic mechanism of respiratory failure related to SARS-CoV-2 infection and inhaled NO role in patients with severe disease.

Objectives: For the purpose of evaluation of exhaled NO as an index of airway inflammation, we assessed changes in fractional exhaled NO (FeNO) across a work shift and its relationship with respiratory complaints. Material and Methods: Chronic and work-aggravated respiratory complaints were assessed using a questionnaire in 89 male textile workers. FeNO and spirometry were performed before and after a work shift and all the changes were registered. Results: A significant increase in FeNO after a work shift was observed. Post-shift FeNO was significantly higher among the subjects with chronic respiratory complaints. There was an obvious decrease in FVC, and FEV₁ after a work shift; however, we couldn’t find a significant relationship between changes in respiratory parameters and concentration of inhalable dusts. Conclusions: FeNO increase after a work shift along with pulmonary function decrement and higher post-shift FeNO among subjects with respiratory complaints makes across-shift FeNO a non-invasive test for assessment of airway hyper-responsiveness in textile workers.

INTRODUCTION: Coronary heart disease (CHD) is the most common heart diseases in Europe. The aim of this study was to determine the intensity of L-arginine metabolism by two alternative pathways (oxidative by NOS and nonoxid-ative by arginase) in the blood plasma of patients with CHD associated with hypertension (HT) of different age groups. MATERIALS AND METHODS: 50 patients with isolated CHD and 42 patients with CHD associated with HT were enrolled in this study. NOS activity was determined by nitrite anion formed in the reaction. Arginase activity was tested by the formation of urea. RESULTS: In middle-aged patients with isolated CHD, the total NOS activity statistically significantly increased by 2.2 fold in comparison with healthy subjects of the same age group. In patients with CHD associated with HT, the total NOS activity statistically significantly increased in both middle-aged and older persons 2.3-fold than in healthy subjects of the same age groups. In patients with isolated CHD, the arginase activity increases 1.5-fold in middle-aged patients and 1.7-fold in older patients compared with the healthy participants. In the middle-aged and older patients with CHD associated with HT, the arginase activity statistically significantly increased 1.7- and 1.8-fold than in the healthy subjects of the same age groups. CONCLUSIONS: In patients with isolated CHD and patients with CHD associated with HT, an increased total NO-synthase and arginase activity in comparison with healthy individuals was found. It was shown that the increase in NO-synthase and arginase activity is more expressed in older patients than middle-aged patients.
WSTĘP: Choroba wieńcowa jest najpowszechniejszą z chorób serca występujących na terenie Europy. Celem pracy było zbadanie intensywności metabolizmu L-argininy dwoma alternatywnymi sposobami (utleniającym przez NOS i nieutleniającym poprzez arginazę) w osoczu krwi pacjentów cierpiących na chorobę wieńcową (ChW) serca, współistniejącą z nadciśnieniem tętniczym (NT), w grupie pacjentów w różnym wieku. MATERIAŁ I METODY: Do badania włączono 50 pacjentów z izolowaną postacią ChW i 42 z ChW współistniejącą z NT. Aktywność NOS została wyznaczona przez anion azotynowy utworzony w reakcji. Aktywność arginazy sprawdzono przez formowanie mocznika. WYNIKI: Pacjenci w średnim wieku, z izolowaną postacią ChW wykazywali 2,2 razy wyższą całkowitą aktywność NOS w porównaniu ze zdrowymi badanymi w tej samej grupie wiekowej. W przypadku pacjentów z ChW współistniejącą z NT całkowita aktywność NOS była znacząco (2,3 razy) zwiększona w obu grupach wiekowych (w średnim oraz starszym wieku) w porównaniu z osobami zdrowymi w tym samym wieku. U pacjentów z izolowaną postacią ChW aktywność arginazy była 1,5 razy podwyższona u osób w średnim wieku oraz 1,7 razy u starszych niż u osób zdrowych. W wieku średnim i starszym pacjenci ze współistniejącą postacią ChW wraz z NT wykazywali statystycznie znacznie podwyższoną aktywność arginazy – odpowiednio 1,7 oraz 1,8 razy większą niż u zdrowych w tym samym wieku. WNIOSKI: Wykazano, że w grupie pacjentów z izolowaną postacią ChW oraz w przypadku współistnienienia ChW i NT aktywność NO-syntazy i arginazy jest wyższa niż u osób zdrowych. Zwiększona aktywność NO-syntazy i arginazy jest bardziej znacząca u pacjentów starszych.

Opis:
Investigation of the mechanisms of phenotypic realization of allelic polymorphism of the eNOS gene has shown that the level of eNOS mRNA and activity of this enzyme in platelets depends from genotype. We identified a T-786→C polymorphism in the promoter region, a variable number of tandem repeats (4a/4b) in intron 4 and the G894→T polymorphism in exon 7 of the eNOS gene in isolated human platelets. We measured eNOS mRNA in isolated platelets by reverse transcription-PCR and eNOS enzyme activity by fluorimetric detection system FCANOS-1 using diaminofluorescein diacetate (DAF-2A). It was shown that the level of eNOS mRNA is the lowest for the -786C/C promoter genotype. In exon 7 homozygotes (894T/T) the level of RNA is lower than in normal homozygotes (894G/G), but higher than in heterozygotes (894G/T). The eNOS activity in platelets is lower in carriers of the 786C/C promoter genotype than in normal homozygotes (2.1 × P=0.03), and lower comparing to heterozygotes (2.9 × P>0.05). The eNOS activity accompanying the 894T/T variant of exon 7 is also lower than in normal homozygotes (P>0.05). Regarding the polymorphism in intron 4 - the enzyme's activity is lower in carriers of the 4a/4a genotype comparing to normal homozygotes (1.7 × P>0.05) and lower than in heterozygotes (1.9 × P>0.05). These results allow one to conclude that the T-786→C polymorphism of the eNOS gene promoter most significantly affects the gene expression and eNOS activity.

Cytosolic phospholipase A2 (cPLA2) preferentially liberates arachidonic acid (AA), which is known to be elevated in Alzheimer's disease (AD). The aim of this study was to investigate the possible relationship between enhanced nitric oxide (NO) generation observed in AD and cPLA2 protein level, phosphorylation, and AA release in rat pheochromocytoma cell lines (PC12) differing in amyloid beta secretion. PC12 control cells, PC12 cells bearing the Swedish double mutation in amyloid beta precursor protein (APPsw), and PC12 cells transfected with human APP (APPwt) were used. The transfected APPwt and APPsw PC12 cells showed an about 2.8- and 4.8-fold increase of amyloid β (Aβ) secretion comparing to control PC12 cells. An increase of NO synthase activity, cGMP and free radical levels in APPsw and APPwt PC12 cells was observed. cPLA2 protein level was higher in APPsw and APPwt PC12 cells comparing to PC12 cells. Moreover, phosphorylated cPLA2 protein level and [3H]AA release were also higher in APP-transfected PC12 cells than in the control PC12 cells. An NO donor, sodium nitroprusside, stimulated [3H]AA release from prelabeled cells. The highest NO-induced AA release was observed in control PC12 cells, the effect in the other cell lines being statistically insignificant. Inhibition of cPLA2 by AACOCF3 significantly decreased the AA release. Inhibitors of nNOS and γ-secretase reduced AA release in APPsw and APPwt PC12 cells. The basal cytosolic [Ca2+]i and mitochondrial Ca2+ concentration was not changed in all investigated cell lines. Stimulation with thapsigargin increased the cytosolic and mitochondrial Ca2+ level, activated NOS and stimulated AA release in APP-transfected PC12 cells. These results indicate that Aβ peptides enhance the protein level and phosphorylation of cPLA2 and AA release by the NO signaling pathway.

Opis:
The influence of oxygenated diesel fuel containing n-butanol on a passenger car exhaust emissions was tested over the NEDC transient cycle. The tests showed that a diesel fuel/butanol blend, containing 10% of n-butanol, caused a significant decrease in PM and smoke emissions, had no effect on NOx and CO2 emissions and brought about higher CO and HC emissions. The most favourable fact resulting from the butanol application as diesel fuel component is that it produces desirable changes in PM/NOx trade-off.

Badania przeprowadzono na jednym piecu, z palnikami na olej i ciekły gaz po ustawieniu zużycia paliwa na stałą moc 75 kW. Zawartość tlenków azotu dla biodiesla SETA Diesel równa 80 mg/m3, była o 20% niższa, aniżeli dla oleju EKOTERM Plus. Temperatura spalin była wyższa o ok.10°C. Straty kominowe dla olejów były niskie, jednak dla biopaliwa nieco wyższe, 7,5%, aniżeli dla oleju opałowego 7%. Ilości tlenku węgla dla wszystkich paliw były jedynie śladowe.
Studies were done on one heating furnace with 2 burners for furnace oil, biofuel and liquid propane gas. The consumption was fixed for a thermal power of 75 kW. The temperature of exhaust from biodiesel was by 10°C higher, than from fuel oil. Amount of nitric oxide was by 20% lower for biodiesel than for fuel oil. Chimney losses for LPG were low (6%) and for biodiesel 0.5% higher than for fuel oil (7%). There were only very small amounts of carbon monoxide found.

Skonstruowano model analizy kowariancji dla poziomu emisji tlenków azotu NOx w spalinach silników S-4002 zasilanych mieszankami paliwowymi mineralnego oleju napędowego ON z dodatkiem estru oleju rzepakowego RME. Jako zmienną niezależną (grupującą) obrano udział procentowy RME w paliwie. Za zmienną towarzyszącą obrano moc efektywną Ne. Badano wzajemne położenie odpowiednich krzywych opisujących zmienną zależną - poziom NOx. Dodanie estru oleju rzepakowego RME przy prędkości obrotowej 2000 obr*min-1 powoduje wzrost poziomu emisji tlenków azotu o około 45,6 jednostek ppm. Emisja wzrasta wraz ze wzrostem mocy efektywnej Ne silnika.
The article presents an analysis of NOx emission levels in the fumes of S-4002 engines fed with fuel blends composed of the mineral engine oil ON with an addition of rape oil ester RME in various proportions. As the independent (grouping) variable the percentage of RME in the fuel was chosen. The associated variable was the effective power Ne. The mutual position of respective curves describing the dependent variable NOx level was examined. The addition of the rape oil ester RME at the rotational speed of 2000 rpm results in the increase of nitric oxides emission level by approximately 45.6 units ppm. The emission increases together with the increase of the engine effective power Ne.

The novel MAO-B inhibitor PF9601N, its cytochrome P450-dependent metabolite FA72 and l-deprenyl were studied as potential peroxynitrite (ONOO-) scavengers and nitric oxide synthase (NOS) inhibitors. The scavenging activity of these compounds was evaluated by measuring the oxygen consumption through peroxynitrite-mediated oxidation of both linoleic acid and brain homogenate. FA72, PF9601N and l-deprenyl caused a concentration-dependent inhibition of ONOO--induced linoleic acid oxidation with an IC50 value of 60.2 µM, 82.8 µM and 235.8 µM, respectively. FA72 was the most potent also in inhibiting ONOO--induced brain homogenate oxidation with an IC50 value of 99.4 µM, while PF9601N and l-deprenyl resulted weaker inhibitors in the same experimental model, showing an IC50 value of 164.8 and 112.0 µM, respectively. Furthermore, both the novel MAO-B inhibitor as well as its metabolite were able to strongly inhibit rat brain neuronal NOS (IC50 of 183 µM and 192 µM, respectively), while l-deprenyl at the highest concentration used (3 mM), caused only a slight decrease of the enzyme activity. Moreover, inducible NOS was strongly inhibited by FA72 only. All these results suggest that PF9601N could be a promising therapeutic agent in neurodegenerative disorders such as Parkinson's disease.

Opis:
Carbon monoxide (CO) is an odorless, tasteless and colorless gas which is generated by heme oxygenase enzymes (HOs). HOs degrade heme releasing equimolar amounts of CO, iron and biliverdin, which is subsequently reduced to bilirubin. CO shares many properties with nitric oxide (NO), an established cellular messenger. Both CO and NO are involved in neural transmission and modulation of blood vessel function, including their relaxation and inhibition of platelet aggregation. CO, like NO, binds to heme proteins, although CO binds only ferrous (FeII) heme, whereas NO binds both ferrous and ferric (FeIII). CO enhances the activity of guanylate cyclase although it is less potent than NO. In contrast, CO inhibits other heme proteins, such as catalase or cytochrome P450. The effects of CO on gene expression can be thus varied, depending on the cellular microenvironment and the metabolic pathway being influenced. In this review the regulation of gene expression by HO/CO in the cardiovascular system is discussed. Recent data, derived also from our studies, indicate that HO/CO are significant modulators of inflammatory reactions, influencing the underlying processes such as cell proliferation and production of cytokines and growth factors.

Opis:
A nitric acid plant in Devnya, Bulgaria has implemented a project for catalytic decomposition of nitrous oxide which resulted in reducing N2O emissions from the plant and the overall N2O emissions from the industrial sector in Bulgaria. After implementing the catalyst in September 2005 till the end of 2011, a total amount of 3133 Mg N2O emissions has been measured while the supposed value of N2O emissions without the catalyst being installed would have been 9747 Mg which means a total reduction of 6614 Mg. The results from the research prove the emission reduction potential of the catalyst.

Opis:
HCCI (Homogenous Charge Compression Ignition) combustion system is now one of the most promising solutions used in piston engines. The paper presents the results of three-dimensional modeling of combustion in the single cylinder HCCI engine powered with Diesel fuel. 3D modeling was performed in AVL Fire code. The basic combustion parameters including start of the ignition (SOI), burn duration (BD), indicated pressure (pi) and nitric oxide (NO) and soot (Soot) emissions were analyzed. The modeling results show that combustion process in HCCI engine compared to a conventional engine with compression ignition is characterized by an earlier ignition (SOI) and shorter burn duration (BD). The impossibility of controlling HCCI combustion process leads to deterioration of engine performance and increased emissions of harmful exhaust gas components. Calculations showed that for the same equivalence ratio of burn mixture, uncontrolled HCCI combustion compared to a controlled combustion in engine with fuel injection operated is characterized by higher NO emission and reduced Soot emission.
