Background . Melanin finds enormous applications in different industries for its unique photoprotective and anti-oxidant properties. Due to its emerging demand, scientific researchers are putting efforts to unravel more microorganisms with a potential of producing melanin on large scale. Hence, the present study was aimed at the isolation of extracellular melanin producing microorganisms from lime quarries of Karnataka, India. Besides this, the tyrosinase gene governing melanin synthesis in different organisms were compared in silico to understand its evolutionary aspects. Material and methods . Melanin producing microorganisms were screened on tyrosine gelatin beef extract agar medium. Potential isolate was explored for submerged production of melanin in broth containing L-tyrosine. Melanin was characterized by UV-Vis spectroscopy, thin layer and high performance liquid chromatographic techniques. Antibacterial activity of melanin was performed by agar well assay. Comparative tyrosinase gene sequence analysis was performed by using Geneious 2021.1 trial version software. Results . Pseudomonas otitidis DDB2 was found to be potential for melanin production. No antibacterial activity was exerted by the melanin against tested pathogens. The in silico studies showed that the common central domain of tyrosinase protein sequence of selected Pseudomonas sps. exhibited 100% identity with the common central domain of Homo sapiens tyrosinase (NP_000363.1). Conclusions . Our study shows the production of melanin in good quantities by the isolate Pseudomonas otitidis DDB2 which can be explored for scale-up process. Since the melanin formed is of eumelanin type and the tyrosinase gene sequence of several Pseudomonas sp. showed relatedness to humans, this molecule may be further developed for sunscreen formulations.

Opis:
Consumer expectations toward food products are changing due to the development of the food industry and new technologies. The growing knowledge in the field of health impact of natural and artificial colorants added to food results in an awaking interest of natural dyes among researchers, producers and consumers. Food colour plays a fundamental role in identifying the quality of food, so it is associated with the sensory quality of food products. In this paper the different types of colorants, their production methods and the evolution of these methods were characterized.
Oczekiwania konsumentów wobec produktów spożywczych zmieniają się z powodu rozwoju przemysłu spożywczego i nowych technologii. Rosnąca wiedza w dziedzinie wpływu na zdrowie naturalnych i sztucznych barwników dodawanych do żywności skutkuje wzbudzeniem zainteresowania naturalnymi barwnikami wśród badaczy, producentów i konsumentów. Kolor żywności ma zasadniczą rolę w identyfikacji jakości żywności, gdyż jest wiązany z jakością sensoryczną produktów spożywczych. W niniejszym artykule opisano różne rodzaje barwników, metody ich produkcji oraz ewolucję tych metod.

Influence of photo- and chromatic acclimation on pigment composition in the sea

Opis:
The aim of this work was to find statistical relationships between the concentrations of accessory pigments in natural populations of marine phytoplankton and the absolute levels and spectral distributions of underwater irradiance. To this end, empirical data sets from some 600 stations in different parts of the seas and oceans were analysed. These data were obtained from the authors’ own research and from the Internet’s bio-optical data base. They included the vertical distributions of the concentrations of various pigments (identified chromatographically) and the vertical and spectral distributions of the underwater irradiance measured in situ or determined indirectly from bio-optical models. The analysis covered a total of some 4000 points illustrating the dependence of pigment concentration on underwater irradiance characteristics, corresponding to different depths in the sea. The analysis showed that the factor governing the occurrence of photoprotecting carotenoids (PPC) is short-wave radiation λ < 480 nm. A mathematical relationship was established between the relative PPC concentration (relative with respect to the chlorophyll a concentration) and the magnitude of the absorbed radiative energy per unit mass of chlorophyll a from the spectral interval λ < 480 nm, averaged in the water layers Δz = 60 m (or less near the surface) to account for vertical mixing. This absorbed short-wave radiation (λ < 480 nm) was given the name of Potentially Destructive Radiation (PDR∗(z)). Analysis of the relationships between the concentrations of particular photosynthetic pigments (PSP), i.e. chlorophyll b, chlorophyll c, photosynthetic carotenoids (PSC), and the underwater irradiance characteristics indicated that these concentrations were only slightly dependent on the absolute level of irradiance E0(λ), but that they depended strongly on the relative spectral distribution of this irradiance f(λ) = E0(λ)/PAR0. The relevant approximate statistical relationships between the relative concentrations of particular PSP and the function of spectral fitting Fj , averaged in the layer Δz, were derived. Certain statistical relationships between the pigment composition of the phtyoplankton and the irradiance field characteristics are due to the photo- and chromatic acclimation of natural populations of marine phytoplankton. These relationships can be applied in models of the coefficients of light absorption by phytoplankton.
