Opis:
Uncoupling proteins (UCPs) mediate fatty acid-induced proton cycling in mitochondria, which is stimulated by superoxide and inhibited by GDP. Fatty acid anions can also be transported by adenine nucleotide translocase (ANT), thus resulting in the uncoupling of oxidative phosphorylation. In the present work, an attempt was made to distinguish between the protonophoric activity of UCP3 and that of ANT using inhibition analysis. This study was carried out using mitochondria from skeletal muscles of hibernating Yakut ground squirrel, which have a significant level of UCP3 mRNA. We found that millimolar concentrations of GDP, which is considered to be a specific inhibitor of UCPs, slightly recoupled the mitochondrial respiration and restored the membrane potential. Addition of the specific ANT inhibitor CAT (carboxyatractylate), in micromolar concentration, prior to GDP prevented its recoupling effect. Moreover, GDP and ADP exhibited a competitive kinetic behavior with respect to ANT. In brown adipose tissue, CAT did not prevent the UCP1-iduced increase in chloride permeability and the inhibitory effect of GDP, thus confirming the inability of CAT to affect UCP1. These results allow us to conclude that the recoupling effect of purine nucleotides on skeletal muscle mitochondria of hibernating ground squirrels can be explained by interaction of the nucleotides with ANT, whereas UCP3 is not involved in the process.

ObjectivesThe proper porthole angle contributes to relieving the operation fatigue and improving the efficiency of oceanauts. In this study, the authors explored the effect of 3 different porthole longitudinal positions on the oceanauts’ back muscles using surface electromyography (sEMG) analysis, and the characteristics of the perceived body comfort was obtained.Material and MethodsOverall, 40 healthy participants were recruited to perform tasks in a simulated cabin environment. Electromyographic (EMG) signals were recorded from the trapezius medius, lower trapezius, and erector spinal muscles for porthole angles of –5°, –15°, and +15°, relative to the horizontal line of sight during a 21-minute experiment. The subject comfort scores were collected at 7, 14 and 21 min. The integrated electromyogram (iEMG) and the root mean square (RMS) of EMG signals, as well as the mean power frequency (MPF), and the mean frequency (MF) were calculated.ResultsThe subjective scores of the +15° porthole at each stage of work are higher than those of the –15° and –5° portholes. The results of iEMG, RMS, MF and MPF all indicated that the +15° porthole design was more conducive to lowering the rate of muscle fatigue, while the –5° and –15° portholes increased the muscle fatigue rate and led to greater fatigue. It was found that the lower trapezius was more prone to fatigue than the trapezius medius and erector spinal muscles. The height, weight and body mass index of the participants were found to negatively correlate with muscles at the +15° porthole, which is highly consistent with the actual situation.ConclusionsThe findings suggested that the +15° position was optimal for delaying the muscle fatigue of the participants and for improving the work efficiency of oceanauts.

Opis:
Activation of the nuclear enzyme poly(ADP-ribose) synthethase (PARS) is important in the cellular response to oxidative stress. During ischemia and reperfusion (I/R) increased free radical production leads to DNA breakage that stimulates PARS which in turn results in an energy-consuming metabolic cycle and initiation of the apoptotic process. Previous studies have reported that PARS inhibition confers protection in various models of I/R-induced cardiovascular damage. The purpose of this study was to determine the role of PARS inhibition in I/R-induced injury of smooth muscle cells and endothelium in the coronary circulation of the isolated guinea-pig heart. Control hearts and those treated with a PARS inhibitor - benzamide (100 µmol L-1), were subjected to 30 min of subglobal ischemia and subsequent reperfusion (90 min). To analyze the functional integrity of smooth muscle cells and endothelium, one-minute intracoronary infusions of endothelium-independent (sodium nitroprusside, NaNP; 3 µmol L-1) and endothelium-dependent (substance P, SP; 10 nmol L-1) vasodilators were used before ischemia and at the reperfusion time. The degree of the injury of coronary smooth muscle and endothelial cells induced by I/R was estimated in terms of diminished vasodilator responses to NaNP (at 55 min and 85 min of reperfusion) and to SP (at 70 min of reperfusion), respectively, and expressed as the percentage of preischemic response. I/R reduced vasorelaxant responses to both vasodilators by half (to 54.1 ± 5.1% and to 53.6 ± 4.9% of preischemic value for NaNP at 55 min and 85 min of reperfusion, respectively and to 45.9 ± 6.5% for SP at 70 min of reperfusion). PARS inhibition provided complete restoration of vasorelaxation induced by NaNP (107.6 ± 13.3% and 104 ± 14.4% of preischemic response at the two time points of reperfusion, respectively). However, there was no effect on the SP-induced response (48+12.1% of preischemic response). We conclude that pharmacological PARS inhibition with benzamide protects coronary smooth muscle cells but not endothelium against I/R-induced reperfusion injury in the coronary circulation of the guinea-pig heart.

Background: Cardiovascular proteomics investigation reveals the characterization and elucidation of the novel therapeutic targets and strategies to prevent the development of heart failure associated diabetic complication by using 2DE and MS. Methods: The experimental animals were made diabetic with a single intraperitoneal injection of alloxan (150 mg/kg of bw). Albino rats were randomly divided into four individual groups: Group-I control (n=6), group-II alloxan-induced diabetic rats, untreated (n=6), group-III (n=6) and group-IV (n=6) alloxan-induced diabetic rats were treated with aqueous and ethanolic extracts of Cynodon dactylon for 15 days, respectively. Animals were euthanized to collect the heart tissues and blood samples. 2DE sample preparation, gel running and staining (n=6: each groups) were performed at the same time to avoid variation. The result of six gel images from each group were analyzed and evaluated as one match set with 2D software (P<0.05). Results: The above experiment revealed two up-regulated proteins in group-II i.e. NTF4 and ETFB. Conclusions: NTF4 is a neuro-protective agent for neuro-degenerative diseases. It will prevent diabetic secondary complications, such as diabetic polyneuropathy and cardiomyopathy. ETFB is active in the mitochondria, the energy-producing centres in cells. It is clear from the experiment that because of up-regulation of ETFB more energy is availabile and the electron transfer for heart during diabetes is possible, what leads to reduce the oxidative stress and free-radical formation. The up-regulated proteins reduced CVD that occurred just before overt hyperglycaemia due to administration of C. dactylon. This approach established the preliminary reference map for decoding cellular mechanisms linked between pathogenesis CVD and diabetes.

Background. To investigate the effect of adding r-ESWT to a standard exercise program of chronic non-specific LBP on electrical muscle activity (EMG), pain and function. Materials and methods. Our single-blind randomized controlled trial enrolled 30 patients with chronic non-specific LBP randomly allocated to an r-ESWT (n=15) group and a control group (n=15). All patients received a standard exercise program, while r-ESWT was additionally administered in the r-ESWT group. EMG activity, pain and function were assessed before and after 6 weeks of treatment. Results. After treatment, all outcome measures were significantly different (p < 0.05). The addition of r-ESWT produced a significant increase in EMG activity (of all muscles tested) and a reduction in pain intensity and functional disability scores (p < 0.05) compared to the control group. Conclusions. 1. A standard intervention offered either alone or with r-ESWT increased EMG activities, reduced pain, and enhanced function in patients with chronic non-specific LBP. 2. Adding r-ESWT to the standard intervention program might produce better results.

The aim of this study was to analyse the electromyographic (EMG) power spectrum indicating differences in the frequency bands of the spectrum resulting from differences in muscle type and force level for the extensor digitorum (ED), palmaris longus (PL) and biceps brachii (BB) muscles. The study was performed on 12 young men as 2 tests different in terms of the type of exerted force (30, 15 and 5% MVC, maximum voluntary contraction). The parameters of the EMG power spectrum were obtained as a result of a division of the EMG power spectrum into ranges determined by border frequencies. The results suggest that (a) frequency bands contained in the 30–80 Hz range and over the frequency of 300 Hz depend on muscle force, and (b) the power spectrum in the frequency range of up to 40 Hz and 60–300 Hz indicates muscle type.

Opis:
Owing to its wholesomeness and good taste, meat of goats and lambs is an increasingly more important in human nutrition. It is characterized by excellent nutritive value and high digestibility. It is also relatively rich in mineral compounds. The aim of this study has been to determine the content of some elements in muscles, liver and kidneys of male and felame kid goats, fattened to 150 day of life. The animals received full-ration mixture containing 169 g total protein and 6.0 MJ net energy. The mixture was given ad libitum and supplemented with meadow hay as bulk feed. During the whole period of fattening, the animals were kept under the conventional housing system. Samples for analysis were taken from adductor femoris muscles, liver and kidneys. The muscle tissue of the analysed male goats contained less Cd (0.002 mg kg-1 ± 0.004) than that of the female goats (0.003 mg kg-1 ± 0.001). In turn, the level Pb was lower in the female goats (0.019 mg kg-1 versus 0.026 mg kg-1 in male goats), although the difference was statistically non-significant. In the internal organs, more Cd occurred in kidneys than in the liver, unlike Pb. The concentration Pb was higher in kidneys of the male than female goats (0.073 mg kg-1 ± 0.02), whereas the level of this element the liver was identical in both groups (0.084 mg kg-1 ). In the muscule tissue, the sex differentiated the content of Fe (p = 0.01) as well as Mg and Zn (p = 0.05). The liver accumulated not only Pb and Cd, but also Cu and Zn. The concentration Cu in the liver of female goats was higher (146.79 mg kg-1 at p = 0.01) than in male goats by about 50 mg kg-1. High disproportions between the sexes in the content of Fe in the liver were observed: 36.30 mg kg-1 for female goats and 21.99 mg kg-1 for male goats (p = 0,01). In kidneys, however, the concentration of Ca was very high, particularly for female goats particularly (81.01 ± 26.55 mg kg-1).
Ze względu na walory zdrowotne i smakowe mięso kozie i jagnięce nabiera coraz większego znaczenia w żywieniu człowieka. Charakteryzuje się doskonałymi właściwościami pod względem wartości odżywczej i wysoką strawnością. Jest stosunkowo bogate w związki mineralne. Celem badań było określenie zawartości wybranych pierwiastków w mięśniach, wątrobie i nerkach koziołków i kózek żywionych do 150. dnia życia. Materiał doświadczalny stanowiły koziołki i kózki rasy białej uszlachetnionej oraz tuczone do 150. dnia życia. Zwierzęta żywiono pełnoporcjową mieszanką o wartości pokarmowej 168 g białka ogólnego i 6,0 MJ energii netto. Mieszankę podawano ad libitum i uzupełniano dodatkiem strukturalnym siana łąkowego. Przez cały okres tuczu zwierzęta utrzymywano w chowie alkierzowym, w pomieszczeniach zamkniętych. Próby do analizy pobrano z mięśnia przywodziciela uda (m. adductor), wątroby i nerek. W tkance mięśniowej analizowanych koziołków doświadczalnych stwierdzono niższą zawartość Cd (0,002 mg kg-1 ± 0, 0,004) ni¿ u kózek (0,003 mg kg-1 ± 0,001). Natomiast poziom Pb był niższy w grupie kózek i wyniósł 0,019 mg kg-1 wobec 0,026 mg kg-1 – różnice okazały się nieistotne statystycznie. W narządach wewnętrznych wyższy poziom Cd stwierdzono w nerkach niż w wątrobie, a w przypadku Pb odwrotnie. Stężenie Pb było wyższe w nerkach koziołków i wyniosło 0,073 mg kg-1 ± 0,02. W wątrobach poziom tego pierwiastka był na tym samym poziomie (0,084 mg kg-1). Płeć różnicowała istotnie zawartość Fe i Ca w tkance mięśniowej (p ≤ 0,01) oraz Mg i Zn (p ≤ 0,05). Wątroba była narządem koncentracji nie tylko Pb i Cd, ale Cu i Zn. Stwierdzono, że stężenie Cu w wątrobie kózek było wyższe (p ≤ 0,01, 146,79 mg kg-1) w porównaniu z koziołkami o ok. 50 mg kg-1. Zaobserwowano wysokie dysproporcje w zawartości Fe w wątrobie, ponieważ u kózek stwierdzono 36,30 mg kg-1, a u koziołków 21,99 mg kg-1 (p ≤ 0,01). W nerkach wykazano zdecydowanie większe stężenie Ca, szczególnie u kózek (81,01 ± 26,55 mg kg-1).

Introduction and objectives. Since the paravertebral muscles promote dynamic stability to protect the spine, the aim of this study was to verify the association between the acute effect of the stretching support time for the multifidus muscle, and changes in the static and dynamic stability of the lumbopelvic region. Materials and method. A total of 46 volunteers were cross-submitted to three different stretching interventions for the multifidus muscle, being the manutention of stretching time the variation between interventions: 10, 30 and 60 seconds. Each volunteer was submitted to the three interventions with a minimum interval of 7 days and a maximum of 10 days. The order of the interventions was determined by lot. First, pre-intervention values for static and dynamic stability were collected using a biofeedback pressure unit. Subsequently, the sample was forwarded for intervention according to its group; after the completion of the same, data were collected on post-intervention stability. The data were analyzed using SPSS 20.0 software and Chi-square test, with a significance level of 5% (α = 0.05). Results. There was no significant association between the manutention of stretching time and static stability, being χ2(4)=0.812; p=0.949. Similarly, there was no significant association between the stretching support time and dynamic stability, and χ2(4)=1.517; p=0.827. Conclusion. In conclusion, there was no significant association between stretching time of the paravertebral muscles and static and dynamic stability.

In this paper the rehabilitation device for upper arm based on artificial muscles is introduced. Presented automated rehabilitation device has three degrees of freedom: 2 DOF in arm and 1 DOF in elbow that provides almost all basic rehabilitation exercises. Artificial pneumatics muscles will be tested in connection with spring and antagonistic connection. This system provides lifting and falling of arm construction based on patient force. There is possibility to generate help force during rehabilitation or opposite load. Artificial muscles are controlled by pneumatic valves terminal from micro computer based on MCU. Higher level control system provides artificial intelligence implementation based on neural network for prediction and change of load according sensor values history (incremental sensor, pressure sensor). For prototype testing there is described usability of industrial robot to test precision of load and trajectories during rehabilitation. This automatic rehabilitation device will help to reduce therapeutics work with patient, automate and improve rehabilitation process.

Previously we have shown that aldolase (ALD; EC 4.1.2.13) is present in cardiomyocyte nuclei. Now, we focused our attention on ALD localization in smooth muscle cells. Immunocytochemical methods were used to study the subcellular localization of ALD. Aldolase was localized in the cytoplasm as well as in the nuclei. Within the nuclei ALD was located in the heterochromatin region. Native polyacrylamide gel electrophoresis followed by aldolase activity staining in gel was used to study the ALD isoenzyme pattern in porcine smooth muscle cells. Two ALD isoenzymes, A and C, were found in these cells but in the nuclei only the muscle isoenzyme was detected. To support the nuclear localization of ALD, measurement of aldolase activity in the smooth muscle cell nuclei isolated from porcine stomach was performed. The ALD activity in the isolated nuclei was detectable only after preincubation of the nuclear fraction with Triton X-100 and high concentration of KCl.

Mythology plays an important part of the role of the American automobile, less so in terms of its primary function that is transportation, more so in terms of an ancillary purpose: its metaphorical significance to both owner or operator and the onlooking public. Across much of the 20th century and continuing now into the third decade of the 21st century, the American automobile has undergone many design changes that have buttressed its metaphorical significance: become streamlined, gained then lost then partially regained size together with a colorful exterior, and in the 21st century become focused on an array of interior gadgets, some cast into hibernation because of an electronic chip scarcity resulting from trade wars and the Covid-19 pandemic. Many Americans seem to have almost become besotted by automobiles, including their own and those driven by others, because in some respects the American automobile has come to define its driver. Automobiles in the United States that are visually appealing symbolize affluence, material success, preoccupation with speed, including the rapid pace of social change, as well as, at least arguably, a lesser regard for protecting the environment. On balance, in the mindset of many Americans, the automobile is larger than life, “a mode of signification, a form” in contrast to a mere machine. Change in automotive design has been heralded as the talisman of a new generation of drivers. However, what is cause and what is effect? American automobiles conflate myth and reality; that which is together with that which might be sometime temporal frustrations with the American Dream.

AMP-deaminase from hen stomach smooth muscle was isolated and physico-chemical properties of the purified enzyme were investigated. The enzyme had an activity optimum at pH 6.5, and poorly deaminated the substrate analogues tested. At optimum pH (6.5), in the absence of regulatory ligands (control conditions), the enzyme manifested hyperbolic substrate-saturation kinetics with half-saturation constant (S0.5) of about 4.5 mM. Additions of adenine nucleotide effectors (ATP, ADP) activated the enzyme strongly at all the concentrations tested, diminishing significantly the value of S0.5 constant. In contrast, the regulatory effect of orthophosphate was variable, and depended on the orthophosphate concentration used. The molecular mass of the enzyme subunit determined in SDS/PAG electrophoresis was about of 37 kDa. The obtained results suggest that in different types of hen muscle, similarly as in humans and rats, expression of AMP-deaminase is under the control of independent genes.
