Opis:
Ten to 16 ethanol-soluble carbohydrate components were identified in the seeds of six Mexican wild lupins. The analysed carbohydrates included: monosaccharides, disaccharides, cyclitols, galactosyl cyclitols and raffinose family oligosaccharides. Stachyose and sucrose were the main carbohydrate component in the Lupinus montanus, L. rotundiflorus, L. exaltatus, L. mexicanus and L. elegans seeds. Only trace quantities of verbascose were detected in Lupinus mexicanus seeds. The analysed seeds accumulated 38 to 78 mg/g d.m. carbohydrates. The raffinose family oligosaccharides constituted 41 to 85.2% of the identified carbohydrate component pool. The analysed Lupinus seeds contained 3 to 8 unidentified carbohydrate components.

Yellow lupin seeds cv. Juno were stored under laboratory conditions for 2 month, 4, 6 and 8 years. Eighteen soluble carbohydrates were identified in embryonic axes and cotyledons of different age seeds. The concentration of soluble carbohydrates in analysed seeds ranged from 25 to 34% of dry mass. Axes contained more carbohydrates than cotyledons. Stachyose dominated in axes, and verbascose - in cotyledons. Other detected galactosides were: galactinol, galactosyl pinitols and galactosyl chiro-inositols (fagopyritols), but their content was several-fold lower than that of RFOs (in both axes and cotyledons tissues). The concentration of soluble carbohydrates indicated, that sucrose to RFOs mass ratio, or other changes in sugars composition are not indicators of seed storage.

The pollen grain germination and the growth of pollen tubes after intra- and interspecific reciprocal pollination of Lupinus albus L., L. mutabilis Sweet, and L. angustifolius L. were examined. Results of microscopic observations of pollen grain germination and pollen tube growth enable to deny the existence of certain barriers that prevent fertilization after pollination of L. albus flowers with pollen of L. angustifolius and vice versa, as well as after pollination of L. mutabilis flowers with pollen of L. angustifolius and vice versa. Some delay in pollen grain germination and a slower growth of pollen tubes were observed in all combinations, as related to intraspecific ones, though in the case of each interspecific combination pollen tubes reached the ovary. Moreover, a different level of pollination efficiency (the number of growing pollen tubes) and various rates of pollen grain germination and pollen tube growth, depending on the direction of pollination, were found. The noted pollination efficiency may generally be considered to be low.

Sequence tagged site (STS) markers are valuable tools for genetic and physical mapping that can be successfully used in comparative analyses among related species. Current challenges for molecular markers genotyping in plants include the lack of fast, sensitive and inexpensive methods suitable for sequence variant detection. In contrast, high resolution melting (HRM) is a simple and high-throughput assay, which has been widely applied in sequence polymorphism identification as well as in the studies of genetic variability and genotyping. The present study is the first attempt to use the HRM analysis to genotype STS markers in narrow-leafed lupin (Lupinus angustifolius L.). The sensitivity and utility of this method was confirmed by the sequence polymorphism detection based on melting curve profiles in the parental genotypes and progeny of the narrow-leafed lupin mapping population. Application of different approaches, including amplicon size and a simulated heterozygote analysis, has allowed for successful genetic mapping of 16 new STS markers in the narrow-leafed lupin genome.

The aim of the study was to determine the effect of mechanical harvest on the seed quality of yellow lupin. Two effects were studied: the cultivar of yellow lupin (the indeterminate cultivar - Mister and the determinate cultivar - Perkoz) and harvest methods: hand-picked plants with manual shelling of seeds as a control and mechanical shelling with a plot harvester. In comparison with manual shelling of seeds, the mechanical harvest reduced the seed germination and increased the number of abnormal seeds both cultivars. Determinate cultivar was more sensitive, because the loss of its quality was higher (germination of 10%) than indeterminate cultivar (6%). Perkoz had also higher electrical conductivity, with the mean value of 34.3 μS × cm-1 × g-1 . Manual shelled seeds were characterized by a significantly lower leakage of exudates (24.9 μS × cm-1 × g-1), and hence, it exhibited greater vigour than mechanical harvested seeds. Most relations of Pearson correlation coefficient between vigour tests and germination were strong or practically functional.

The inheritance of light-cream coloration of vegetative parts in yellow lupin plants was studied. It was found that this trait is inherited monomerically. In homozygous system the gene responsible for light-cream coloration has a plieotropic action. Among others it causes a lower fertility and reduction of seed yield per plant. In heterozygous system symptoms of heterosis are observed. In many aspects the action of dilutus gene is similar to that of dwarf gene in yellow lupin previously described by the authors of this paper.

Introduction. Lupinus spp. are herbaceous perennial flowering plants included in the Fabaceae family. Among the approximately 200 species belonging to this genre, Lupinus albus L., also known as white lupin, Lupinus angustifolius L., and narrow-leafed lupin or blue lupin, represent two of the most studied species due to their intense culinary use and potential biological activity. The intention of the study was to characterize total phenolic content, perform FTIR analysis, and antiproliferative effects against A375 human melanoma cells extracts obtained from germinated and ungerminated seeds from Lupinus albus L. and Lupinus angustifolius L. Materials and method. Total phenolic content was assessed using the Folin-Ciocalteu colorimetric method. FTIR spectra were carried out by a Shimadzu Prestige-21 spectrometer in the range 400–4000 cm-1, using KBr pellets and resolution of 4 cm-1. Antiproliferative capacity was screened by employing the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and scratch assay methods. Results. The study showed increased values corresponding to total phenolic content for the germinated extracts. FTIR spectra confirmed the presence of genistein and main cinnamic acids derivatives (ferulic, caffeic, rosmarinic, and coumaric acids). All tested extracts showed weak antiproliferative potential against A375 human melanoma cells. Conclusions. Germinat on increased the amount of bioactive compounds in the seeds of the two studied species of lupin. FTIR analyses provided an important fingerprint of the chemical composition.

Nixtamalization is usually performed on grains by cooking in an alkaline solution to improve the final product characteristics. White bitter lupin (Lupinus albus) seeds were nixtamalized at various concentration of calcium hydroxide in the range of 0.16–3.33% at 50, 70, and 90°C for 35 min and steeped for 0, 8, 16, and 24 h, and the moisture uptake was determined to model seed hydration kinetics. Moisture uptake increased with increasing nixtamalization temperature regardless of calcium hydroxide concentration. The Page and Weibull models adequately described white bitter lupin hydration kinetics during nixtamalization. Model parameters Kp (Page model) and α (Weibull model) ranged from 80.2 to 410.03 and from 88.21 and 93.96, respectively, for nixtamalization at different calcium hydroxide concentrations, and from 58.55 to 662.88 and from 68.74 and 132.99, respectively, for nixtamalization at different temperatures. The alkaloid content of raw lupin flour was 1.08 g/100 g and it gradually decreased as a result of nixtamalization in increasingly concentrated calcium hydroxide solutions and higher temperatures. The cracks were visible in the microstructure of nixtamalized seed coats. Their number and size increased with the increase of processing temperature, calcium hydroxide concentration, and steeping duration. Overall, the presented results may be useful in optimizing the industrial nixtamalization of lupin seeds and increasing the possibility of their use as a valuable food ingredient
